Mollusks are very sensitive to aquatic environmental alterations and then, are important bio-indicators for monitoring the contamination of water bodies. Iodinated X-ray contrast media (ICMs) are ubiquitously present in the aquatic environment, primarily due to their high consumption for diagnosis purposes, high injection levels, low biodegradability, and low removal rates by wastewater treatment plants. Although these compounds are assumed to be of low toxicity, aquatic organisms are continuously exposed to these agents, which may result in adverse effects as ICMs can act as iodine source and disrupt the endocrine system. Thus, the evaluation of their environmental risk, especially on aquatic fauna is of great interest. To this end, we first compared the accumulation behavior, based on iodine analysis, of two ICM exhibiting different osmolality, diatrizoic acid and iohexol in Dreissena polymorpha bivalves exposed under laboratory conditions at concentrations of 0, 100, and 1000 μg/L during 4 and 7 days. This study was the first to provide information on iodine concentration in whole soft tissues and several organs in control zebra mussels. Moreover, it showed, after exposure, an increase of iodine content mainly in the digestive glands, followed by gills and gonads, highlighting that ICMs actually enter the organisms. Thus, bioaccumulation of ICMs studies were then performed, by liquid chromatography coupled to tandem mass spectrometry, on entire mollusks and digestive glands of organisms exposed at 0, 10, 100, and 1000 μg/L of both ICMs during 21 days, followed by 4 days of depuration. These first data on ICMs concentrations in zebra mussels, showed a clear accumulation of ICMs in mussels as a function of relative exposure level, as well as a rapid depuration. Osmolality did not seem to have a significant impact on the accumulation level, but a slight difference was observed on the accumulation pattern between both ICMs.

International audience; This study aimed to determine uranium (U) pollution over time using otoliths as a marker of fish U contamination. Experiments were performed in field contamination (~20 μg L−1: encaged fish: 15d, 50d and collected wild fish) and in laboratory exposure conditions (20 and 250 μg L−1, 20d). We reported the U seasonal concentrations in field waterborne exposed roach fish (Rutilus rutilus), in organs and otoliths. Otoliths were analyzed by ICPMS and LA-ICP SF MS of the entire growth zone.Concentrations were measured on transects from nucleus to the edge of otoliths to characterize environmental variations of metal accumulation. Results showed a spatial and temporal variation of U contamination in water (from 51 to 9.4 μg L−1 at the surface of the water column), a high and seasonal accumulation in fish organs, mainly the digestive tract (from 1000 to 30,000 ng g−1, fw), the gills (from 1600 to 3200 ng g−1, fw) and the muscle (from 144 to 1054 ng g−1, fw). U was detected throughout the otolith and accumulation varied over the season from 70 to 350 ng g−1, close to the values measured (310 ng g−1) after high exposure levels in laboratory conditions. U in otoliths of encaged fish showed rapid and high U accumulation from 20 to 150 ng g−1. The U accumulation signal was mainly detected on the edge of the otolith, showing two U accumulation peaks, probably correlated to fish age, i.e. 2 years old. Surprisingly, elemental U and Zn signatures followed the same pattern therefore using the same uptake pathways. Laboratory, caging and field experiments indicated that otoliths were able to quickly accumulate U on the surface even for low levels and to store high levels of U. This study is an encouraging first step in using otoliths as a marker of U exposure.

Pressurized intraperitoneal aerosol chemotherapy (PIPAC) is a technique to directly deliver chemotherapeutic drugs in the abdomen for the treatment of peritoneal metastases. Pressurization improves the treatment efficacy but increases the risk of exposure for the medical/non-medical staff who can be exposed by dermal or ocular contact, or inhalation of aerosols containing the cytotoxic drugs. The aim of this study was to evaluate the risk of exposure for the medical/non-medical staff (nurses, surgeons, anaesthesiologists and cleaning personnel; n = 13) during PIPAC with oxaliplatin performed according to the protocol recommended in France. Blood samples were collected 1 h before and immediately after PIPAC, and urine samples 1 h before, and then 3 h and the morning after PIPAC. In the control, non-exposed group (n = 7), only one urine and blood sample were collected. Surface contamination in the operating room was assessed in water- and Surfanios-impregnated wipe samples. The total elemental platinum in each sample was quantified by inductively coupled plasma mass spectrometry, using a method adapted to quantify trace amounts (ng.L−1) in very low volumes (100 μl). No surface contamination was detected. Although 25% of urine samples in the exposed group contained platinum, no statistical difference was observed in urine and plasma samples collected before and after PIPAC and with the control group samples. These findings suggest that the French PIPAC protocol does not increase the risk of exposure to platinum in all staff categories involved. This protocol could be considered in future occupational policies and consensus statements. Trial registration: NCT04014426

International audience; Asphaltenes are considered to be the most problematic components of heavy oils because they can self-aggregate which leads to precipitation and causes various problems during oil recovery, transportation, and refining. The contribution of the porphyrins present in asphaltenes to the aggregation was previously studied by gel permeation chromatography inductively coupled plasma-mass spectrometry (GPC-ICP MS). The molecular weight of asphaltene aggregates was shown to be increased by free metal-containing porphyrins (corresponding to the lower molecular weight fraction) interacting with the aggregate’s surfaces by weak forces. The characterization of free porphyrins within the asphaltenes is therefore for the understanding of the mechanism of the aggregation, coprecipitation, and demetalation processes. Here, we developed a method for the separation of free porphyrins from asphaltenes on the basis of their polarity using high-performance thin-layer chromatography (HPTLC). This technique, using disposable plates, is particularly well suited for asphaltene analysis since it eliminates the risk of clogging typical of column chromatography. Cellulose plates were used in this study. The lower polarity of their hydroxyl groups limit the irreversible adsorption and improves the detection limit by the ICP-MS. Two well-separated peaks were obtained from purified asphaltene (Asphaltene 2017; Asphaltene Characterization Interlaboratory Study for PetroPhase 2017. In Proceedings of the 18th International Conference on Pretroleum Phase Behavior and Fouling, Le Havre, France, June 11–15, 2017; Total, the University of Pau, and the University of Rouen-Normandy: Le Havre, France, 2017.) and its corresponding whole crude oil and C5 and C7 fractions. The distribution of vanadium due to migration was determined by laser ablation (LA) ICP MS. The eluted fraction contained the free porphyrins, whereas the major fraction did not migrate and corresponded to trapped porphyrins. A comparison with the signal obtained by UV densitometry allowed the ratio between the inorganic and organic material to be measured.

In relation to the decrease of selenium (Se) content in aquafeeds, the impact of level and form of parental and dietary Se supplementation was investigated in rainbow trout fry using laser ablation-inductively coupled plasma mass spectrometry (LA-ICP MS) bioimaging. The offspring of rainbow trout broodstock, fed either a control diet without any Se supplementation (0.3 mg Se/kg diet) or a diet supplemented with Se (0.6 mg Se/kg diet) either as sodium selenite or hydroxy-selenomethionine, were sampled at swim-up fry stage or after 11 weeks of cross-feeding. Total body Se levels were influenced by parental Se nutrition in swim-up fry and by direct Se feeding in 11-week fry with higher levels in the Se-supplemented groups compared with the control and the highest levels in the hydroxy-selenomethionine treatment. The Se retention was lower for dietary sodium selenite. Selenomethionine levels increased when Se was provided as hydroxy-selenomethionine. LA-ICP MS maps revealed yolk in swim-up fry and intestine, liver, and kidney in 11-week fed fry as tissues with high Se abundance. In swim-up fry, muscle Se was the highest abundant when parents were fed hydroxy-selenomethionine. In 11-week fed fry, muscle Se abundance was higher in the head part of fry fed both Se-supplemented diets, but only in the tail part of fry fed hydroxy-selenomethionine. Liver Se abundance was higher in fry fed sodium selenite compared with the control diet supporting the hypothesis that tissue Se distribution can be influenced by parental and dietary Se forms and levels.

Objectives Platinum salts are commonly used in hyperthermic intraperitoneal chemotherapy (HIPEC) for digestive tract cancer treatment. During HIPEC with oxaliplatin for peritoneal metastases (PMs) treatment, the ovaries are directly exposed to the drug, questioning about ovarian resection and the potential impact of the drug on ovarian functionality, especially in young women of childbearing age. The goal of this work is to understand unwanted damages to the ovaries during HIPEC therapy by the determination of the concentration and distribution of platinum in ovaries in order to address its potential toxicity. Methods Mass spectrometry imaging techniques, matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) and laser ablation inductively coupled plasma mass spectrometry (LA-ICP MS), were used to study the penetration of oxaliplatin in ovaries after HIPEC treatment. Results MALDI-MS allowed the localization of an oxaliplatin-derivative (m/z 456.2) at the periphery of the ovaries. The quantitative LA-ICP MS maps confirmed the localization of elemental platinum as well as in the central part of ovaries from patients who received a previous platinum salt-based chemotherapy. Conclusions LA-ICP MS images showed that platinum diffusion was extended in cases of previous systemic treatment, questioning about platinum derivatives gonado-toxicity when combining the two treatments.

International audience; Asphaltenes are among the most challenging components in petroleum processing because they contain high amounts of heteroatoms (i.e., S, N, O, V, and Ni) thought to be responsible for strong aggregation tendencies, precipitation, and fouling problems. The role of vanadium- and nickel-containing petroleum compounds (i.e., petroporphyrins) in aggregation and fouling is not completely understood because asphaltene composition and structure is still a subject of debate in the petroleum chemistry community. Characterization of asphaltenes, namely, molecular analysis that employs no chromatographic separation, often fails to reveal their comprehensive composition. The work herein presents asphaltene fractionation by (1) solid/liquid extraction, which allows for separation of single-core (“island”) and multicore (“archipelago”) structural motifs and by (2) high-performance thin layer chromatography (HPTLC) with cellulose as the stationary phase and DCM/MeOH as the eluent, which facilitates access to petroporphyrins. Characterization is performed by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and matrix-assisted laser desorption ionization Fourier transform ion cyclotron resonance mass spectrometry (MALDI FT-ICR MS). The results demonstrate that even with multiple separation steps, a large quantity of vanadyl porphyrins remains inaccessible for molecular analysis by MALDI FT-ICR MS, which raises the question of what portion of a complex sample of asphaltene can be revealed by ultrahigh resolution mass spectrometry. Furthermore, the results show that easily accessible porphyrins migrate with the solvent front in HPTLC. Thus, HPTLC can be used to isolate and identify “free” porphyrins not locked into asphaltene aggregates; however, further development of separation methods is required to access the most difficult and problematic asphaltene fractions, which do not migrate and impose analytical challenges due to their stronger aggregation tendency.

International audience; Concentration of uranium (U), a naturally encountered radioactive element in earth’s crust, can be enhanced in freshwater ecosystems (µg.L-1 – mg.L-1) due to various anthropogenic activities. The consequent aquatic organism exposure to U leads to its accumulation in all organs, particularly in the gonad, and in subcellular fractions (mainly the cytosol); then it is known to affect fish at several biological levels, and more particularly, at a reproduction endpoint, with a decrease in the total number of eggs, spawn events and larvae survival. The understanding of U reprotoxicity requires the fine knowledge of its speciation at molecular level, i.e., its interaction with cytosolic biomolecules. In this study, we focus on the U-protein interactions in gonads. A non-denaturating extraction protocol combined with size exclusion chromatography (SEC) allowed the separation of metal-protein complexes in ovaries of U-contaminated wild roaches before their elemental detection (ICP MS). This enables unprecedented information to be obtained about U distribution in ovaries of autochthonous fish, Rutilus rutilus, which is different in some points from that obtained in the model species, Danio rerio under controlled laboratory conditions at a similar concentration level. Finally, the ability to transpose results from model to autochthonous fish was briefly discussed.

International audience; Understanding how essential and toxic elements are distributed in cereal grains is a key to improving the nutritional quality of cereal-based products. The main objective of this work was to characterize the distribution of Cd and of nutrients (notably Cu, Fe, Mn, P, S and Zn) in the durum wheat grain. Laser ablation inductively coupled mass spectrometry and synchrotron micro X-ray fluorescence were used for micro-scale mapping of Cd and nutrients. A dissection approach was used to quantitatively assess the distribution of Cd and nutrients among grain tissues. Micro X-ray absorption near-edge spectroscopy was used to identify the Cd chemical environment in the crease. Cadmium distribution was characterized by strong accumulation in the crease and by non-negligible dissemination in the endosperm. Inside the crease, Cd accumulated most in the pigment strand where it was mainly associated with sulfur ligands. High-resolution maps highlighted very specific accumulation areas of some nutrients in the germ, for instance Mo in the root cortex primordia and Cu in the scutellum. Cadmium loading into the grain appears to be highly restricted. In the grain, Cd co-localized with several nutrients, notably Mn and Zn, which challenges the idea of selectively removing Cd-enriched fractions by dedicated milling process.

International audience; Due to the dramatic quantity of plastic debris released into our environment, one of the biggest challenges of the next decades is to trace and quantify microplastics (MPs) in our environments, especially to better evaluate their capacity to transport other contaminants such as trace metals. In this study, trace elements (Fe, Cu, Zn, As, Cd, Sn, Sb, Pb, and U) were analyzed in the microplastic subsurface (200 μm) using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Microplastics subjected to the marine environment were collected on beaches (Guadeloupe) exposed to the north Atlantic gyre. We established a strategy to discriminate sorbed contaminants from additives based on the metal concentration profiles in MP subsurface using qualitative and quantitative approaches. A spatiotemporal correlation of the sorption pattern was proposed to compare MPs in terms of relative exposure time and time-weighted average concentrations in the exposure media.

A large fraction of the South-American cacao production is affected by new cadmium (Cd) regulations in cacao. This work was set up to characterize the distribution and speciation of Cd within the cacao fruit and to monitor potential Cd redistribution during cacao fermentation. In cacao fruits from four locations, Cd concentrations decreased with testa > nib ~ placenta ~ pod husk > mucilage. The distribution of Cd within cacao beans was successfully visualized using laser ablation inductively coupled plasma spectrometry (LA-ICP-MS) and confirmed higher Cd concentrations in the testa than in the nib. Speciation analysis by X-ray absorption spectroscopy (XANES) of unfermented cacao beans revealed that Cd was bound to O/N-ligands in both nib and testa. Fermentation induced an outward Cd migration from the nibs to the testa, i.e. against the total concentration gradient. International audience; Mass Spectrometry Imaging is an effective technology that allows to determine the in-situ distribution of endogen and/or exogen small molecules. It is a rapidly emerging approach for visualizing drugs and their metabolites within biological tissues. Matrix-Assisted Laser Desorption Ionization (MALDI) Mass Spectrometry Imaging (MSI) coupled to high resolving power analyzer (e.g. TOF) was already investigated for metallodrug localization and metabolization studies, but was proved to suffer from a lack of sensitivity and resolution, leading to poor coverage and assignment. To counter these technological limitations, the use of ultra-high resolving power analyzer such as Fourier Transform Ion Cyclotron Resonance (FTICR) could be revealed as a technique of choice. The high field FTICR MS provides ultra-high resolving power and mass accuracy that allows exhaustive molecule coverage and non-ambiguous molecular formula assignments. Platinum derivatives, such as oxaliplatin, are widely used as therapeutic agents for cancer treatment. The assessment of their intake, distribution and metabolism within the organs is important to know the risks associated with their use. In this study, MALDI FTICR MSI analyses were performed to better understand the penetration and metabolization of platinum derivatives in ovaries of women treated by Hyperthermic Intraperitoneal Chemotherapy (HIPEC) for peritoneal metastasis of colorectal or appendicular origin. International audience; We studied the effects of supplementing a plant-ingredient based rainbow trout broodstock diet with different forms of selenium (Se) on reproductive performance, parental Se transfer and antioxidant metabolism in their progeny before first-feeding. Three groups of rainbow trout were fed a diet either without selenium supplementation (NC, basal Se level: 0.3 ppm) or supplemented with 0.3 ppm Se either in the form of sodium selenite (SS) or as hydroxy-selenomethionine (SO, dietary Se level: 0.7 ppm) over a 6-month period prior to spawning. In Se-supplemented groups, the total number of females spawning was significantly higher compared to the negative control group and the females fed SO began to spawn earlier compared to females fed SS or NC. Total Se concentrations were significantly higher in female muscle of SO group. Higher Se concentrations in the oocytes of both Se-supplemented groups confirmed a maternal transfer of Se, while total Se concentrations in milt samples were not significantly different between dietary groups. There was no effect on glutathione peroxidase (GPX) or other antioxidant enzyme activity in the liver of female or male broodstock, whereas in the swim-up fry, GPX activity was significantly higher in both Se-supplemented treatments with the highest activity in the SO group. Se-supplementation enhanced the expression of hepatic SelPa in broodstock males and females along with MsrB1, GPx1a, GPx4a2, CAT, Gclc, Keap1 and MsrB2 in male liver. In swim-up fry, only organic Se-supplementation led to a higher gene expression for SelPa, GPX1a, GPX1b2, CAT and MsrB2. An improvement of the oxidative status in whole-body of swim-up fry could not be confirmed in this study as the GSH/GSSG was even lower in swim-up fry of Se-supplemented groups, whereas the 8-isoprostane levels were lower in the SS group and no effect on protein carbonyl levels could be detected. Organic Se-supplementation led to a significant increase in the α-tocopherol and vitamin C levels in the progeny. These results show that dietary Se in broodstock nutrition influences spawning occurrence and along with the parental transfer, several traits in the progeny are affected.

International audience; Quantitative localization of metals in biological tissue sections is critical to obtain insight into metal toxicity mechanisms or their beneficial characteristics. This study presents the development of a quantitative LA-ICP MS bioimaging methodology based on the polymer film strategy and internal standardization. To maximize the number of elements mapped, an aqueous soluble polymer (dextran) was selected. Among the elements studied, the great majority (eight out eleven), i.e., Co, Ni, Cu, Zn, Se, Mo, Cd and Pt, exhibited linear regression after LA-ICP MS analysis of metal-spiked polymer standards. Methodology performances were carefully assessed as a function of the three internal standards (In, Rh and Ir) considered, the analytical operational conditions (ICP power, addition of O 2 to ICP, and laser fluency) and the thickness of the biological tissue section. The results indicated that three groups (Co, Mo; Ni, Cu, Pt; and Zn, Se, Cd) of elements could be distinguished from their analytical response as a function of analytical conditions and the internal standard. These different element behaviors appeared to be mainly First Ionization Potential dependent (FIP). For elements with lower FIP (Co, Ni, Cu, Mo and Pt), differential responses due to carbon load in the ICP MS plasma could be efficiently corrected as a function of analytical conditions. Matrix effects were more pronounced for higher FIP elements (i.e., Zn, Cd and Se), and analysis of

International audience; Selenium (Se) is an essential component of genetically encoded selenoproteins, in the form of a rare amino acid, namely the selenocysteine (Sec). Radioactive (75)Se has been widely used to trace selenoproteins in vitro and in vivo (cell models and animals). Alternatively, its unique isotopic pattern can be used to detect and characterize nonradioactive Se-compounds in cellular extracts using molecular or elemental mass spectrometry at ppm levels. However, when studying trace levels of Se-compounds, such as selenoproteins (ppt levels), the distribution of the signal between its six naturally abundant isotopes reduces its sensitivity. Here, we describe the use of isotopically enriched forms of Se as an alternative strategy to radioactive (75)Se, for the labeling and tracing of selenoproteins in cultured cell lines.

International audience; Limited knowledge about in vivo non-covalent uranium (U)-protein complexes is largely due to the lack of appropriate analytical methodology. Here, a method for screening and identifying the molecular targets of U was developed. The approach was based on non-denaturing 1D and 2D gel electrophoresis (ND-PAGE and ND-2D-PAGE (using ND-IEF as first dimension previously described)) in conjunction with laser ablation inductively coupled plasma mass spectrometry (LA-ICP MS) for the detection of U-containing proteins. The proteins were then identified by μbore HPLC-Orbitrap MS/MS. The method was applied to the analysis of cytosol of hepatopancreas (HP) of a model U-bioaccumulating organism (Procambarus clarkii). The imaging of uranium in 2D gels revealed the presence of 11 U-containing protein spots. Six protein candidates (i.e. ferritin, glyceraldehyde-3-phosphate dehydrogenase, triosephosphate isomerase, cytosolic manganese superoxide dismutase (Mn-SOD), glutathione S transferase D1 and H3 histone family protein) were then identified by matching with the data base of crustacea Decapoda species (e.g. crayfish). International audience; A comprehensive study of the bioavailability of orally administered silver nanoparticles (AgNPs) was carried out using a rat model. The silver uptake was monitored in liver and kidney tissues, as well as in urine and in feces. Significant accumulation of silver was found in both organs, the liver being the principal target of AgNPs. A significant (∼50%) fraction of silver was found in feces whereas the fraction excreted via urine was negligible (\textless0.01%). Intact silver nanoparticles were found in feces by asymmetric flow field-flow fractionation (AsFlFFF) coupled with UV-Vis analysis. Laser ablation-ICP MS imaging showed that AgNPs were able to penetrate into the liver, in contrast to kidneys where they were retained in the cortex. Silver speciation analysis in cytosols from kidneys showed the metallothionein complex as the major species whereas in the liver the majority of silver was bound to high-molecular (70-25 kDa) proteins. These findings demonstrate the presence of Ag(i), released by the oxidation of AgNPs in the biological environment.

International audience; As methylmercury (MeHg) can be bioaccumulated and biomagnified in the trophic web, its toxicity for marine mammals is of major concern. Mercury speciation in marine biota has been widely studied, mainly focused on the discrimination and quantification of inorganic Hg and MeHg. Less attention has been paid to the interactions of Hg with biomolecules and the characterization of its specific binding, which play a key role in metabolic pathways controlling its uptake, transformation, and toxicity. In the studied white-sided dolphin (Lagenorhynchus acutus) liver homogenate (QC04LH4) sample, approximately 60 % of the total MeHg was found in the water soluble fraction, specifically associated with high molecular weight biomolecules. The identity of the involved proteins was investigated (after tryptic digestion of the fraction) by μRPLC with parallel detection by ICP-MS and ESI-MS/MS. Molecular mass spectrometry experiments were carried out at high resolution (100000) to ensure accurate protein identification and determination of the MeHg binding sites. Cysteine residue on the dolphin hemoglobin β chain was found to be the main MeHg binding site, suggesting that hemoglobin is a major MeHg binding protein in this marine mammal and could be a potential carrier of this MeHg from blood to liver prior to its degradation in this organ. In parallel, a significant proportion of selenium was found to be present as selenoneine and a potential role for this compound in Hg detoxification is discussed.

New data on the nature of the protein targets of U within the zebrafish gills were collected after waterborne exposure to better understand uranium (U) toxicity mechanisms. This study evidenced some common characteristics of the U protein target binding properties such as their role in other essential metals regulation and their phosphorus content. International audience; We characterized the biomolecules involved in handling cytosolic metals in larvae of the phantom midge (Chaoborus) collected from five mining-impacted lakes by determining the distribution of Ag, Cd, Cu, Ni, Tl, and Zn among pools of various molecular weights (HMW: high molecular weight, \textgreater670-40 kDa; MMW: medium molecular weight, 40-\textless1.3 kDa; LMW: low molecular weight, \textless1.3 kDa). Appreciable concentrations of nonessential metals were found in the potentially metal-sensitive HMW (Ag and Ni) and LMW (Tl) pools, whereas the MMW pool, which includes metallothioneins (MTs) and metallothionein-like proteins and peptides (MTLPs), appears to be involved in Ag and Cd detoxification. Higher-resolution fractionation of the heat-stable protein (HSP) fraction revealed further differences in the partitioning of nonessential metals (i.e., Ag = Cd ≠ Ni ≠ Tl). International audience; Laser ablation-ICP MS was optimized for the sensitive detection of selenoproteins in polyacrylamide gel and PVDF membrane after blotting. For this purpose, two interlaboratory reference samples were prepared: glutathione peroxidase band in the gel and on the membrane, respectively. The optimisation was carried out using two systems: 213 nm laser (Newwave) - Agilent 7500ce ICP MS, and a 1030 nm high repetition rate femtosecond laser with galvanometric optics (Novalase) - PE/SCIEX DRCII. Sensitivity and signal-to-noise ratio were benchmarked to those obtained for the same sample by a recently published method in a reference lab. The optimization allowed a 12-fold gain of the S/N ratio during ablation of gels and a 3.5-fold gain in the ablation of blots in comparison with the method using an essentially similar system published by the reference lab. The gain of S/N by increasing ablation surface using the high repetition rate laser was not as spectacular as expected (2.5-fold for the gels and 1.5-fold for the blots) as the background noise increased considerably when a larger surface is ablated due to selenoproteins peak tailing. The study allowed for the first time LA-ICP MS detection of selenoproteins (separated by gel electrophoresis) in human cell extracts with the selenium concentration at the 10 ng ml -1 level.

Hg stable isotopically labeled species were used to assist the investigation of Hg species (i.e. IHg and MeHg) binding biomolecules in the aqueous soluble fraction of white-sided dolphin (Lagenorhynchus acutus) liver homogenate (QC04LH4). By using stable enriched isotopic tracers (199IHg and 201MeHg), the transformation yields of both mercury species (i.e. methylation and demethylation) were evaluated during the different sample preparation steps, such as the tissue lyophilization and the extraction by ultrasonication. Hg species partitioning after fractionation of the sample (cytosolic fraction and residual fraction) was also studied and showed that MeHg is mainly present in the cytosol (60%), while IHg is preferentially associated to the solid residue (88%). The analysis of the water-soluble fraction by size exclusion chromatography-ICP MS revealed the association of Hg species with biomolecules in a wide molecular weight range. The ratio of IHg and MeHg was successively determined in each SEC fraction by GC-ICP-MS. Furthermore the use of isotopic tracers allowed demonstration of specific Hg species affinity with different biomolecule molecular weight fractions. The results indicate a specific affinity of the IHg with a biomolecule at an elution time corresponding to a metallothionein whereas MeHg is associated to a biomolecule of larger molecular weight.

International audience; Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) have become established as very efficient and sensitive biopolymer and elemental mass spectrometric techniques for studying metal-binding proteins (metalloproteins) in life sciences. Protein complexes present in rat tissues (liver and kidney) were separated in their native state in the first dimension by blue native gel electrophoresis (BN-PAGE). Essential and toxic metals, such as zinc, copper, iron, nickel, chromium, cadmium and lead, were detected by scanning the gel bands using quadrupole LA-ICP-MS with and without collision cell as a microanalytical technique. Several proteins were identified by using MALDI-TOF-MS together with a database search. For example, on one protein band cut from the BN-PAGE gel and digested with the enzyme trypsin, two different proteins - protein FAM44B and cathepsin B precursor - were identified. By combining biomolecular and elemental mass spectrometry, it was possible to characterize and identify selected metal-binding rat liver and kidney tissue proteins. © 2008 Elsevier B.V. International audience; This text presents a novel method for the separation and detection of phosphorothioate oligonucleotides with the use of ion pair ultra high performance liquid chromatography coupled with inductively coupled plasma mass spectrometry The research showed that hexafluoroisopropanol/triethylamine based mobile phases may be successfully used when liquid chromatography is coupled with such elemental detection. However, the concentration of both HFIP and TEA influences the final result. The lower concentration of HFIP, the lower the background in ICP-MS and the greater the sensitivity. The method applied for the analysis of serum samples was based on high resolution inductively coupled plasma mass spectrometry. Utilization of this method allows determination of fifty times lower quantity of phosphorothioate oligonucleotides than in the case of quadrupole mass analyzer. Monitoring of 31P may be used to quantify these compounds at the level of 80μgL-1, while simultaneous determination of sulfur is very useful for qualitative analysis. Moreover, the results presented in this paper demonstrate the practical applicability of coupling LC with ICP-MS in determining phosphorothioate oligonucleotides and their metabolites in serum within 7min with a very good sensitivity. The method was linear in the concentration range between 0.2 and 3mgL-1. The limit of detection was in the range of 0.07 and 0.13mgL-1. Accuracy varied with concentration, but was in the range of 3%. © 2014 Elsevier B.V.

In this study, selenium species from Se containing proteins in mushrooms (Agaricus bisporus and Lentinula edodes) were investigated with size-exclusion liquid chromatography coupled to UV and inductively coupled plasma mass spectrometry (ICP-MS). Different protein extraction protocols were investigated. Variability of the fractionation patterns with three extraction media (0.1 M NaOH, 30 mM Tris–HCl, and enzymatic digestions) was evaluated for both mushroom types. A 24 h Tris–HCl extraction followed by acetone addition was found to be optimal for protein precipitation. Presumably protein bound selenoamino acids were released using enzymes (proteinase K, protease XIV and trypsin). The selenium speciation of the proteolytic extract of the water soluble proteins fraction was carried out by using reversed-phase ion-pairing high performance liquid chromatography (RP-HPIPC) coupled on-line to ICP-MS for selenium specific detection. Selenocystine, selenomethionine, methylselenocysteine and inorganic selenium were established in both samples utilizing retention time standards and standard additions to the sample.

Strong cation-exchange chromatography (SCX-HPLC) was used in conjunction with inductively coupled plasma mass spectrometry (ICP-MS) to investigate cationic selenium species present in leaf extract of wild-type Brassica juncea supplemented with selenite. Total amount of Se accumulated by the leaves was found to be 352 μg g −1 . Cation-exchange solid-phase extraction (SCX-SPE) was used to pre-concentrate the cationic species present in the leaf extract. Methylselenomethionine (MeSeMet) and dimethylselenoniumproprionate (DMSeP) were synthesized and characterized by electrospray quadrupole time-of-flight MS (ESI-QTOF-MS). Laboratory synthesized and commercially available standards were used in chromatographic studies to identify the Se species in the leaf extract through retention time comparisons and standard addition method. Major cationic selenium species identified in the present study were MeSeMet and methylselenocysteine (MeSeCys) while selenomethionine (SeMet) was found in minor quantities.

International audience; An in vitro model simulating enzymatic activity in the gastrointestinal tract was developed for the assessment of the potential bioaccessibility of Cd and Pb in cocoa powder and liquor. The model was based on the sequential extraction with simulated gastric and intestinal juices; the residue after the latter extraction was further investigated by using, in parallel, solutions of phytase and cellulase. The solubility of Cd and Pb in the corresponding enzymatic extracts was measured by ICP MS. The bioaccessibility of Cd in cocoa varied from 10 to 50% in gastrointestinal conditions. An additional 20 or 30% of Cd could be recovered by phytase and cellulase, respectively. The bioaccessibility of Pb in gastrointestinal conditions did not exceed 5-10%. Only a few percent more of this metal could be recovered by extraction with phytase and cellulase.

International audience; Fifteen extraction methods were investigated for the recovery of different classes of Cd and Pb species in 8 different cocoa powder samples. The procedures targeted water-soluble compounds, polypeptide and polysaccharide complexes and compounds soluble in simulated gastrointestinal conditions. The extracts were analysed by size-exclusion fast-flow liquid chromatography with ICP-MS detection. The detection limit was 0.5 ?g l-1 and the RSD was less than 7.5%. Cd and Pb were very firmly bound to the insoluble matrix components, of which the binding capacity exceeded about 1000 times the naturally present metal levels. Cocoa powder may show possible detoxifying properties for Pb and Cd by binding them into stable complexes, which are resistant in gastrointestinal conditions. The maximum average recovery for Cd and Pb was, respectively, 15% and 5% of the total metal present.

International audience; An off-gel non-denaturing isoelectric focusing (IEF) method was developed to separate uranium-biomolecule complexes from biological samples as a first step in a multidimensional metalloproteomic approach. Analysis of a synthetic uranium-bovine serum albumin complex demonstrated the focusing ability of the liquid-phase IEF method and the preservation of most of the uranium-protein interactions. The developed method was applied to gill cytosol prepared from zebrafish (Danio rerio) exposed to depleted uranium. The pacific oyster (Crassostrea gigas), a good model for studying biomineralization and coastal environmental change, was used to study the expression of cuproproteins after waterborne Cu exposure (3 μg Cu L(-1)) similarly to the Arcachon Bay aquatic environment. Cuproproteins in oyster cytosols were analyzed by gel electrophoresis (non-denaturing PAGE and SDS 2-DE), laser ablation inductively coupled plasma mass spectrometry (LA-ICP MS) and electrospray ionization mass spectrometry (ESI-MS). The high molecular weight cuproproteins (identified as extracellular superoxide dismutase [Cu-Zn] (EC Cu/Zn-SOD) and L-ascorbate oxidase) in digestive glands of oysters were differentially expressed after Cu exposure. About 1.7 and 1.2-fold increase of the Cu level in EC Cu/Zn-SOD and L-ascorbate oxidase were, respectively, determined, although EC Cu/Zn-SOD was 1.6-1.9-fold down-regulated in the digestive gland of oysters after Cu exposure. At last, the role of EC Cu/Zn-SOD in protection against Cu exposure was discussed to propose possible mechanisms to better understand Cu homeostasis and/or detoxification mechanisms.

International audience; Cadmium (Cd) is a potent selective toxicant that preferentially accumulates in the kidneys where it is known to induce cellular injury. The cellular mechanisms explaining toxicity have been well documented by in vivo and in vitro studies. However, experiments have mainly been performed on cadmium chloride (CdCl2), while several other forms, such as cadmium oxide (CdO) and cadmium sulfide (CdS), are of interest regarding occupational exposure. CdS is mainly used in nanotechnology, primarily to construct particles known as quantum dots (QDs). The present study focuses on cadmium forms that differ compared to their solubility and their particle size (CdS micro- and uncoated nanoparticles (NPs)), and investigates their effects on mesangial and proximal tubular kidney cells. Cellular toxicity was evaluated by using Neutral Red, MTT and WST-1 assays. Intracellular Cd content was measured using inductively coupled optical emission spectrometry (ICP-OES) and cellular stress was investigated via the effects of Cd complexing bioligands using size exclusion chromatography ICP-MS (SEC-ICP-MS). Data indicated the variability of cytotoxicity responses after CdCl2, CdO, and CdS exposure that were strongly correlated to the cellular Cd content. Release of Cd2+ linked to solubility and particle degradation were considered to be the cause of these toxicities. The bioinduction of the Cd-MT also evidenced release of Cd2+. Our in vitro data identified heterogeneity of Cd toxicity that was dependent on the physico-chemical properties of the form studied and, when considering particle size, constitute an additional step toward the determination of nanoparticle effects in renal cells.

International audience; The follow-up of the Heated Intraoperative Chemotherapy (HIPEC) of peritoneal carcinomatosis would benefit from the monitoring of the penetration, distribution and metabolism of the drug within the tumor. As tumor nodules can be resected during the therapy, mass spectrometry imaging is a suitable tool for the evaluation of treatment efficacy, and, as a result, the therapy can be re-optimized. In this work we demonstrate the complementarity of laser ablation (LA) ICP mass spectrometry and MALDI imaging to study the penetration and distribution of two Pt-based metallodrugs (cisplatin and oxaliplatin) in human tumor samples removed from patients diagnosed with colorectal or ovarian peritoneal carcinomatosis. LA ICP MS offered sensitive (LOD for 195Pt 4.8 pg s-1) imaging of platinum quasi-independently of the original species and the sample matrix and thus an ultimate way of verifying the penetration of the Pt-containing drug or its moieties into the tumor. MALDI imaging was found to suffer in some cases from signal suppression by the matrix leading to false negatives. In the case of the oxaliplatin metallodrug, the results obtained from ICP and MALDI MS imaging were coherent whereas in the case of cisplatin, species detected by ICP MS imaging could not be validated by MALDI MS. The study is the first application of the dual ICP and MALDI MS imaging to the follow-up of metallodrugs in human tumors

International audience; The toxicity of uranium (U) to aquatic organisms depends notably on its compartmentalization in organs, tissues, cells as well as on its distribution among biomolecules. In order to contribute to the understanding of U accumulation and associated toxicity mechanisms in case of waterborne exposure, this study focused on U fate in the gills epithelia, uptake pathway, of the fish model Danio rerio (zebrafish). U distribution among cytosolic biomolecules was investigated after no addition (0μgL-1 (c0) for 3 and 30d), chronic (20μgL-1 (c20) for 30d) and acute (20μgL-1 (c20) and 250μgL-1 (c250) for 3d) exposures to depleted U. Cytosolic U accounted for an average of 24-32% of gills burden for c20 and c250, respectively. Size Exclusion Chromatography (SEC) coupled with Inductively Coupled Plasma-Sector Field Mass Spectrometry (ICP-SFMS) allowed identification of ecotoxicologically relevant U-containing fractions among cytosolic biomolecules as a function of exposure conditions. The coupling of capillary zone electrophoresis with electrospray mass spectrometry was optimized for the direct determination of phytochelatins (PCs) in extracts obtained from cells and plants that had been exposed to metal stress. Gluthathione and phytochelatins belonging to the different families (gamma Glu-Cys)nGly (n-PC), (gamma Glu-Cys)nSer, (gamma Glu-Cys)n beta Ala and (gamma Glu-Cys)n were separated in an uncoated capillary at pH 4 using a 5 mM ammonium acetate buffer, and detected by electrospray (ES) MS in the full scan mode (300-1100 u). The use of on-line tandem MS detection in the product ion scan mode of putative protonated molecules of PCs allowed the unambiguous confirmation of the identity of the compounds detected by ES MS. The operational conditions were optimized and the figures of merit were evaluated using n-PC2, n-PC3 and n-PC4 standards purified from a mixture obtained after the reaction of glutathione in the presence of Cd2+ and the enzyme PC-synthase. The method was applied to the characterization of bioinduced ligands in cell cultures of soybeans (Glycine max) and in rice (Oryza sativa) roots without the need for a preliminary sample cleanup by size-exclusion and/or reversed phase chromatography.

International audience; A non-denaturating isoelectric focusing (ND-IEF) gel electrophoresis protocol has been developed to study and identify uranium (U)-protein complexes with laser ablation-inductively coupled plasma mass spectrometry (LA-ICP MS) and electrospray ionization mass spectrometry (ESI-MS). The ND-IEF-LA-ICP MS methodology set-up was initiated using in vitro U-protein complex standards (i.e., U-bovine serum albumin and U-transferrin) allowing the assessment of U recovery to 64.4 ± 0.4 %. This methodology enabled the quantification of U-protein complexes at 9.03 ± 0.23, 15.27 ± 0.36, and 177.31 ± 25.51 nmol U L-1 in digestive gland cytosols of the crayfish, Procambarus clarkii, exposed respectively to 0, 0.12, and 2.5 μmol of waterborne depleted U L-1 during 10 days. ND-IEF-LA-ICP MS limit of detection was 19.3 pmol U L-1. Elemental ICP MS signals obtained both in ND-IEF electropherograms and in size exclusion chromatograms of in vivo U-protein complexes revealed interactions between U- and Fe- and Cu-proteins. Moreover, three proteins (hemocyanin, pseudohemocyanin-2, and arginine kinase) out of 42 were identified as potential uranium targets in waterborne-exposed crayfish cytosols by microbore reversed phase chromatography coupled to molecular mass spectrometry (μRPC-ESI-MS/MS) after ND-IEF separation. International audience; Microbial activity is recognized to play an important role on Hg methylation in aquatic ecosystems. However, the mechanism at the cellular level is still poorly understood. In this work subcellular partitioning and transformation of Hg species in two strains: Desulfovibrio sp. BerOc1 and Desulfovibrio desulfuricans G200 (which exhibit different Hg methylation potential) are studied as an approach to the elucidation of Hg methylation/demethylation processes. The incubation with isotopically labeled Hg species (199Hgi and Me201Hg) not only allows the determination of methylation and demethylation rates simultaneously, but also the comparison of the localization of the originally added and resulting species of such metabolic processes. A dissimilar Hg species distribution was observed. In general terms, monomethylmercury (MeHg) is preferentially localized in the extracellular fraction; meanwhile inorganic mercury (Hgi) is associated to the cells. The investigation of Hg binding biomolecules on the cytoplasmatic and extracellular fractions (size exclusion chromatography coupled to ICP-MS) revealed noticeable differences in the pattern corresponding to the Hg methylating and nonmethylating strains.

International audience; Knowledge of the organ and subcellular distribution of metals in organisms is fundamental for the understanding of their uptake, storage, elimination and toxicity. Detoxification via MTLP and MRG formation and chelation by some proteins are necessary to better assess the metal toxic fraction in aquatic organisms. This work focused on uranium, natural element mainly used in nuclear industry, and its subcellular fractionation and chemical speciation to elucidate its accumulation pattern in gills and hepatopancreas of crayfish Procambarus clarkii, key organs of uptake and detoxification, respectively. Crayfish waterborne exposure was performed during 4 and 10d at 0, 30, 600 and 4000μgUL-1. After tissue dissection, uranium subcellular fractionation was performed by successive ultracentrifugations. SEC-ICP MS was used to study uranium speciation in cytosolic fraction. The uranium subcellular partitioning patterns varied according to the target organ studied and its biological function in the organism. The cytosolic fraction accounted for 13-30% of the total uranium amount in gills and 35-75% in hepatopancreas. The uranium fraction coeluting with MTLPs in gills and hepatopancreas cytosols showed that roughly 55% of uranium remained non-detoxified and thus potentially toxic in the cytosol. Furthermore, the sum of uranium amount in organelle fractions and in the non-detoxified part of cytosol, possibly equivalent to available fraction, accounted for 20% (gills) and 57% (hepatopancreas) of the total uranium. Finally, the SEC-ICP MS analysis provided information on potential competition of U for biomolecules similar than the ones involved in endogenous essential metal (Fe, Cu) chelation. © 2012 Elsevier Ltd.

International audience; A novel analytical procedure for the identification of metal (Hg, Cd, Cu, Zn) complexes with individual metallothionein (MT) isoforms in biological tissues by electrospray MS/MS was developed. The sample preparation was reduced to three rapid steps: the two-fold dilution of the sample cytosol with acetonitrile, the recovery of the supernatant containing MT-complexes by centrifugation and its concentration under nitrogen flow. The replacement of reversed phase HPLC by hydrophilic interaction LC (HILIC) allowed the preservation of the unstable and low abundant metallothionein zinc-mercury mixed complexes (MT-Zn6Hg). The MT complexes eluted were detected by ICP MS and identified in terms of molecular mass by electrospray high resolution (100000) MS. The identification was completed by on line demetallation and the determination of the molecular mass of the apoform, followed by amino acid sequencing in the top-down mode using high energy collision fragmentation (HCD). The method was applied to the identification of MT complexes in a white-sided dolphin (Lagenorhynchus acutus) liver homogenate. The Zn complex of the N-acetylated MT2 isoform was found to be predominant, the presence of mixed complexes with Cd, Cu and, for the first time ever, Hg, was demonstrated. The latter finding has the potential to shed new light on the mercury detoxification mechanism in marine organisms.

Transgenic and non-transgenic soybean seeds were compared in terms of total element concentrations, behavior of elements during sequential extraction fractionation and element bioaccessibility using an in vitro simulated gastrointestinal digestion. The analysis were carried out by ICP-sector field-MS or size-exclusion ICP-MS (25 elements in concentrations varying from ng g−1 to the % level). It seems that transgenic and non-transgenic soybean seeds exhibit statistically significant differences in concentrations of Cu, Fe and Sr, which are also reflected by element contents in water extracts and residues. Additionally, contributions of bioaccessible fractions of Cu, Fe and other elements (Mn, S, Zn) for transgenic soybean seeds appear to be larger than those found in non-transgenic soybean seeds.

cited By 20; International audience; A comprehensive approach to the characterization of metallothionein (MT) isoforms based on microbore HPLC with multimodal detection was developed. MTs were separated as Cd7 complexes, detected by ICP MS and tentatively identified by molecular mass measured with 1-2 ppm accuracy using Orbital ion trap mass spectrometry. The identification was validated by accurate mass of the corresponding apo-MTs after postcolumn acidification and by their sequences acquired online by higher-energy collision dissociation MS/MS. The detection limits down to 10 fmol and 45 fmol could be obtained by ESI MS for apo- and Cd7-isoforms, respectively, and were lower than those obtained by ICP MS (100 fmol). The individual MT isoforms could be sequenced at levels as low as 200 fmol with the sequence coverage exceeding 90%. The approach was successfully applied to the identification of MT isoforms induced in a pig kidney cell line (LLC-PK1) exposed to CdS nanoparticles. © 2010 American Chemical Society.

International audience; The potential of inductively-coupled plasma mass spectrometry (ICP-MS) and its complementarity to soft-ionization MS techniques are discussed in the context of the analysis for biomolecules. ICP-MS offers detection limits in the attomolar range, regardless of the molecular environment of the target element. The sensitivity is hardly affected by the sample matrix, chromatographic mobile phase, or co-eluted compounds. The abundance sensitivity over six decades and the linear dynamic range over nine decades make simultaneous multi-isotopic analysis routinely possible. The manuscript discusses the state-of-the-art of ICP-MS for the detection of proteins in gel electrophoresis and of peptides in 2D high-performance liquid chromatography. The possibilities of quantification to the degree of some post-translational modifications are highlighted. Attention is also paid to the role of ICP-MS in protein quantification via metal-coded labeling and to the use of differentially-labeled antibodies for the multiplexed biomarker analysis. The key role of ICP-MS in the emerging area of metallomics is briefly discussed. © IM Publications LLP 2010.
