4. Viral Haemorrhagic Septicemia Virus (VHSV) Isolated from Atlantic Herring, Clupea harengus, Causes Mortality in Bath Challenge on Juvenile Herring

- Abstract
Viral hemorrhagic septicaemia virus (VHSV) has been demonstrated to cause high mortalities in a wide range of teleosts, farmed as well as wild. In Europe, VHSV of genotypes Ib, Id, II, and III have been detected in wild fish, including Atlantic herring Clupea harengus, but disease outbreaks have not been observed in Atlantic herring and the effects on wild stocks are not well documented. Here, we have tested two VHSV isolates from herring (genotypes Ib and III, from the western coasts of Norway and Denmark, respectively) in a challenge experiment with herring (mean weight 2.59 g, SD 0.71 g) caught on the west coast of Denmark. The Norwegian genotype Ib isolate (NO-F-CH/2009) showed an accumulated mortality of 47% compared to 6% mortality with the Danish genotype III isolate 4p168 and zero in the unchallenged control group. In both groups, we found positive rt-RT-PCR and positive immunohistochemistry of VHSV from days 6 and 8 onward. With both isolates, the organs mainly affected were the heart and kidney. The results demonstrate the susceptibility of Atlantic herring to VHSV, and both genotypes gave pathological findings in several organs. Genotype III showed a low mortality rate, and the importance of this genotype for herring is therefore not determined. Genotype Ib showed both high prevalence and mortality, and this genotype is therefore likely to have a negative effect on wild Atlantic herring stocks. Further examinations to determine how VHSV can affect wild Atlantic herring stocks are needed.

- Abstract
Oppdrett i åpne merder gjør at smittestoffer fra oppdrettsanleggene vil kunne drive fra merdene og ut i miljøet. Lakselus som slippes fra hunnlus som sitter på oppdrettsfisk antas, i oppdrettsintensive områder, å være den største kilden for lakselus også på vill laksefisk (Fjørtoft mfl. 2019; Dempster mfl. 2021). Økte nivåer av lakselus er vist å ha negative effekter på vertsfisken, som igjen kan medføre en populasjonseffekt. Effekten lakselus har på ville bestander av laks langs hele landet vurderes i denne rapporten opp mot antatte tålegrenser for dødelighet for individer. Derfra estimeres effekt på populasjoner. Hvor mange lus den ville fisken har fått (nivå av infestasjon) vurderes basert på overvåking av lakselus på vill laksefisk, samt på estimert påvirkning av lakselus fra koblede fysisk-biologiske modeller. Tålegrenser for individer og effekten på populasjonsnivå er basert på best tilgjengelig kunnskap fra laboratorie- og feltundersøkelser.

Parasitic salmon lice cause great economic losses in the aquaculture of Atlantic salmon and rainbow trout. It also has a significant impact on wild populations of salmonids, particularly sea trout in areas with aquaculture activity. Several pharmaceuticals have been used for treatment of salmon lice infestations, but over time the lice have developed resistance to these treatments, and there is a growing concern regarding the environmental impact of chemical and pharmaceutical treatments on non-target organisms. Consequently, there has been a strong incentive to find alternative methods for de-lousing. The use of cleaner fish, which pick lice from the skin of infested salmonids, has become an important tool to fight lice. Lumpfish and several species of wrasses are used as cleaner fish. There are extensive fisheries for wrasses in the UK, along the Swedish west coast and in Norway. In addition, there is aquaculture of ballan wrasse and lumpfish in Norway, and lumpfish in the UK and Iceland. The fisheries and use of cleaner fish have evolved relatively fast, whereas scientific data collection, as well as the development of a regulatory framework, has lagged behind. Challenges linked to poor welfare, transmission of diseases and changes in the genetic structure of local populations as a result of translocations have raised questions regarding the sustainability of the current cleaner fish practice. Both ballan wrasse and lumpfish are relatively new as farmed species, and as new species in aquaculture, there are knowledge gaps that must be filled. Research on different aspects for each of the two species has recently increased to address problems and prepare the fish for a life as cleaner fish in sea cages. There is also an increase in research that aims to increase survival and welfare of cleaner fish. There is still a way to go, but information from farmers who have good results show that it should be possible to achieve improvement. A wide range of research projects and studies have been initiated in Scandinavia and the UK over the last decade. In order to facilitate a more rapid spread and implementation of good management solutions that are based on knowledge of the species, it is therefore important that scientists and regulating authorities in different institutions and countries have a common knowledge-base and are collaborating efficiently. This report presents the state-of-knowledge on the biology of cleaner fish, the challenges regarding environmental impacts of fishing, translocation and how management have dealt with the fisheries in the Nordic countries. The development and challenges related to the use and welfare of cleaner fish in captivity are presented and discussed. Based on our shared expert knowledge, we hopefully provide management recommendations on how the fisheries and use of cleaner fish can become more sustainable. A sustainable fishery should allow the target species to replenish and have no long-term or permanent negative changes on ecosystem diversity, function, or productivity. A sustainable practice should minimize the harm and death of cleaner fish, as well as escapement and the associated risks; including transfer of new diseases to new species and areas, and irreversible genetic changes in the wild populations that reduce fitness or adaptability. The report also identifies data gaps that may be filled with future, hopefully collaborative, research or monitoring activities. Although there is an active collaboration between Swedish and Norwegian cleaner fish researchers there is still a considerable potential for improvement through exchange of knowledge and experiences between scientist and managers in Norway, Sweden and the UK, as well as with those in other countries with developing or emerging fisheries for wrasses, like Denmark.

This report presents the state-of-knowledge on biology, fisheries and how management have dealt with the fisheries and use.

- Abstract
Årets Risikorapport norsk fiskeoppdrett omfatter effekter på vill laksefisk, utslipp fra anlegg, fiske og bruk av leppefisk i lakseoppdrett, samt velferd hos oppdrettsfisk i merd i sjø. Akvakultur har et svært komplekst risikobilde, og det er varierende kunnskapsnivå og grad av tilgjengelige overvåkningsdata knyttet til de ulike faktorene. For å kunne gi en mer ensartet framstilling av temaene, og for å kommunisere dynamikken og kompleksiteten av risiko knyttet til akvakultur bedre, har vi valgt å implementere en ny metode for risikovurdering i årets rapport. Bayesianske nettverk er valgt som et kvalitativt verktøy for å visualisere risiko i form av årsaks- sammenhenger, enkeltfaktorers grad av påvirkning og kunnskapsstyrke. Hensikten er at de grafiske strukturene (kalt risikokart) og tilhørende argumentasjon skal bidra til hurtig og intuitiv forståelse av risikobildet, også blant dem som ikke har bakgrunn fra oppdrettsnæringen. I tråd med forskningsfronten innen risikofaget er ikke formålet med denne risikovurderingen å beregne risiko nøyaktig, men heller å legge til rette for risikoforståelse og risikoerkjennelse hos forvaltning og andre brukere som utgangspunkt for prioriteringer og beslutninger om veivalg og tiltak. Ytterligere genetisk endring hos villaks som følge av rømt oppdrettslaks Hvert år rømmer det titusenvis av laks fra norske oppdrettsanlegg, og i enkelte år har antall rapporterte rømt oppdrettslaks vært høyere enn antall voksen villaks som returnerer til elvene for å gyte. De fleste oppdrettslaks som rømmer, forsvinner i havet. Det er sannsynlig at de dør av sult, sykdom, eller blir spist av predatorer. Likevel overlever noen etter rømming, og flere tusen vandrer opp i elvene hvert år og utgjør en risiko for innkryssing og genetisk påvirkning av villaksen. Over tid vil innkryssing av rømt oppdrettslaks kunne forandre egenskapene til de ville laksebestandene, redusere antall villaks som produseres og svekke bestandenes evne til å tilpasse seg endringer i miljøet. Risikovurderingen viser at kun de to sørligste (produksjonsområdene 1 og 2), samt det nordøstligste produksjonsområdet (område 13) vurderes til å ha lav risiko for ytterligere genetisk endring (ytterligere innkryssing) som følge av rømt oppdrettslaks. Tre av de andre produksjonsområdene (5, 6 og 12) vurderes til å ha moderat risiko for ytterligere genetisk endring, mens sju produksjonsområder (3, 4 og 7–11) vurderes til å ha høy risiko for ytterligere genetisk endring som følge av rømt oppdrettslaks. Vi konkluderer derfor med at det også i de kommende år vil være moderat til høy risiko for ytterligere genetisk endring som følge av rømt oppdrettslaks i ville bestander i store deler av landet. Kunnskapsstyrken i produksjonsområdene 3 og 12 vurderes som god, mens for de andre produksjonsområdene vurderes den som moderat. Miljøeffekter som følge av utslipp av løste næringssalter fra fiskeoppdrett Overgjødsling (eutrofiering) grunnet økte konsentrasjoner av næringssalter i kystvannet, med påfølgende økt produksjon av planteplankton, kan medføre store negative endringer i økosystemet. Økt produksjon av planteplankton fører til økt mengde dyreplankton, økt nedfall til bunn med konsekvenser for oksygenkonsentrasjoner i bunnvann og effekter på dyresamfunn i sedimentene og makroalgesamfunn i fjæresonen. Overgjødsling av kystvann er kjent tidligere fra skagerrakområdet og fra mange deler av verden, mens mesteparten av norskekysten er vist å ha relativt lavt innhold av næringssalter. Norske kystvannsområder er i hovedsak nitrogenbegrensede, det vil si at i sommerhalvåret er det lite nitrogen i vannet. Produksjonsområde 3, Karmøy til Sotra, er det området i landet som har høyest utslipp av løste næringssalter per sjøareal. Vi har imidlertid gode miljødata fra dette området, og tilstanden i PO3 vurderes som god, med god kunnskapsstyrke. Vår vurdering av de andre produksjonsområdene støtter på erfaringer fra dette området, og risikoen for regionale miljøeffekter (eutrofiering) som følge av økt næringssalttilførsel fra fiskeoppdrett, vurderes derfor som lav i alle produksjonsområder langs kysten. For produksjonsområder som mangler miljødata, er kunnskapsstyrken satt til moderat der utslippene er så lave at de med stor sannsynlighet vil ha neglisjerbar effekt, og kunnskapsstyrken er vurdert som dårlig der utslippene er høyere. Miljøpåvirkning på bunn som følge av partikulære organiske utslipp fra fiskeoppdrett Oppdrettsfisk i Norge produseres i all hovedsak i åpne merdanlegg, og det slippes ut organiske partikler direkte til miljøet i form av fekalier fra fisken, og fôr som ikke spises. Utslippene spres eller akkumuleres på eller i sedimentet, og vil påvirke miljøet i større eller mindre grad rundt oppdrettsanlegget. Utslippene av organiske partikler fra fiskeoppdrett er høye, og påvirkningen på bunnen kan bli stor under produksjonen. Utslippene består imidlertid hovedsakelig av lett nedbrytbare forbindelser, påvirkningen er reversibel, og regenerering av bunnen vil kunne vare fra noen måneder til noen år. For bløtbunn er der satt grenser for hvor stor bunnpåvirkning som aksepteres fra partikulære organiske utslipp både under anleggene og i områdene rundt. For blandingsbunn og hardbunn finnes ennå ingen standardisert overvåkning med grenseverdier. Basert på rapporteringen gjennom dagens overvåkningssystem, vurderes tilstanden til bløtbunnslokaliteter å være god i alle produksjonsområder, og at risiko for uakseptable miljøpåvirkninger som følge av partikulært organisk utslipp er lav. Det kan imidlertid være stor variasjon i bunntype over korte avstander, og mange av dagens oppdrettsanlegg ligger over områder med både hardbunn og bløtbunn. Siden det per i dag ikke eksisterer noen god overvåkning av hardbunnslokaliteter, vurderes risikoen for miljøpåvirkning som moderat i alle produksjonsområder for denne bunntypen. Miljøeffekter på non-target-arter ved bruk av legemidler Lakselus tilhører dyregruppen krepsdyr, og medikamenter som dreper lakselusen kan også påvirke andre krepsdyrarter og også andre arter. Andre arter enn lakselus er i denne risikovurderingen gitt en generell betegnelse som «non-target-arter», og omfatter arter som lever fritt i vannmassene, på bunnen og i strandsonen. Det er forskjell i måten bademidler og fôrmidler påvirker non-target-arter. Bademidler gir kortvarig effekt, mens fôrmidler vil kunne påvirke non-target-arter over en lengre tidsperiode. Hvilke avlusningsmiddel som brukes, vil også ha stor betydning, siden de ulike avlusningsmidlene har varierende giftighet og effekt på ulike arter og livsstadier. Av de avlusningsmidlene som vurderes i årets rapport, er det kun azametifos som vurderes å ha lav risiko for miljøeffekter på non-target-arter. De andre midlene, hydrogenperoksid, deltametrin, flubenzuroner og emamektin, vurderes å ha moderat risiko. Kunnskapsstyrken vurderes som moderat for alle midlene, da det fortsatt mangler en del kunnskap om hvilke arter som påvirkes og hvilken effekt en eventuell endring i artssammensetningen vil ha for miljøet. For bademidler som slippes ut fra en merd, vil risikoen for effekt på miljøet være størst nær merdene der konsentrasjonen er høyest, og om våren på grunn av våroppblomstringen (høy forekomst av egg og larver i frie vannmasser). Våren 2018 (første og andre kvartal) var det flest forskrivninger av bademidler som kan medføre økt risiko for effekt på arter i de frie vannmassene i produksjonsområdene 3 og 4. For fôrmidler som spres via organiske partikler til bunnsedimentet, vil effekten i hovedsak være knyttet til langtidspåvirkning av bunnlevende organismer. Miljøeffekter ved bruk av villfanget leppefisk i fiskeoppdrett Leppefisk og rognkjeks brukes som rensefisk for å bekjempe lakselus hos laksefisk i oppdrett, og er regnet som et miljøvennlig alternativ til bruk av legemidler. Rensefisk påfører også oppdrettsfisken mindre stress enn avlusningsmetoder som krever håndtering. De vanligste artene som brukes til avlusning er berggylt, grønngylt, bergnebb, gressgylt og rognkjeks. All rognkjeks som brukes som rensefisk, kommer fra oppdrett. Bruk av oppdrettet rensefisk er ikke vurdert i denne risikovurderingen, heller ikke smitte fra rensefisk til oppdrettsfisk. Det er et mål at fiske og bruk av rensefisk ikke skal ha vedvarende negative miljøeffekter. Dette innebærer ingen permanent endring i genetisk struktur, ikke endret helsestatus og ingen uønskede, langvarige eller permanente økosystemendringer. Risikovurderingen viser at i de sonene hvor det fiskes og benyttes villfanget leppefisk som rensefisk, er det moderat til høy risiko for at det vil forekomme miljøeffekter som følge av denne bruken. Vi konkluderer derfor med at dersom dagens praksis med bruk av villfanget leppefisk opprettholdes, må det forventes at uønskede hendelser som smittespredning og genetisk endring vil forekomme. I tillegg må det forventes uønskede effekter som følge av selve fisket etter leppefisk. Risikovurderingen baserer seg på dagens status og kunnskap om bruk av villfanget leppefisk som rensefisk i oppdrett, og gjelder så lenge dagens praksis videreføres. Velferd hos laks og rensefisk i merder i sjø I norske oppdrettsmerder er det til enhver tid 300–400 millioner oppdrettslaks og flere titalls millioner rensefisk. Dette er individer som etter dyrevelferdsloven har krav på å bli holdt i et miljø som gir god velferd ut fra artstypiske og individuelle behov, og mulighet for stimulerende aktivitet, bevegelse, hvile og annen naturlig atferd. Oppdretterne må også sikre at fôret er av god kvalitet og dekker fiskens behov, at oppdrettsfisken er beskyttet mot skade, sykdom og andre farer. Oppdrettsfisken må være robust nok og ha forutsetninger for å tåle oppdrettsforholdene, og de skal ikke utsettes for unødige påkjenninger og belastninger. Risikovurderingen viser at velferd til laks i merd vurderes som god for produksjonsområdene 6–11, mens den vurderes som moderat for produksjonsområdene 2–5 og 12–13. Utfordringene i nord er først og fremst knyttet til lave temperaturer og bakterielle sårinfeksjoner, mens Vestlandet har store utfordringer med PD og skader i forbindelse med hyppige avlusingsoperasjoner. Oppdrettsmerdene er bygget og lokalitetene er valgt for å være optimale for lakseoppdrett. Risikoen for dårlig velferd hos rognkjeks vurderes som høy, hvor høye temperaturer, sterk vannstrøm og særlig sykdom, er viktige risikofaktorer. På lokaliteter med mindre strøm og lavere temperatur, og med tilstrekkelig skjul og flater hvor rognkjeksen kan feste seg, kan velferden være bedre. På samme måte som for rognkjeks, blir velferden til leppefisk vurdert som dårlig. For leppefisk er sykdom en viktig risikofaktor, men i tillegg har de ofte dårlige forutsetninger for å håndtere miljøforholdene i merdene, og heller ikke noen av de andre påvirkningsfaktorene vurderes som gode. Høy dødelighet kan derfor skyldes at de har dårlige betingelser i merdmiljøet og derfor er mer mottakelige for sykdom. I motsetning til for rognkjeks, er lav temperatur en negativ miljøfaktor. I denne risikovurderingen har vi valgt å vurdere alle leppefiskartene under ett, selv om risikoen for lav velferd er større for noen arter enn andre.

Viral Haemorrhagic septicaemia viruses (VHSV) are isolated from 82 different fish species and are globally spread in freshwater as well as marine environments. The virus can cause disease outbreaks both in wild and farmed fish. Different genotypes of the virus have been isolated in different parts of the world. Furthermore, most isolates seem to show some degree of species specificity. For instance, the genotype 1a, that is a major killer in farmed rainbow trout, shows low mortality to cod, and vice versa the genotype III from cod show low mortality to rainbow trout. However, the established view was challenged in 2007, when a VHSV of genotype III resulted in a VHS outbreak on several sites with farmed rainbow in one fjord in Norway. The farmed populations were slaughtered, according to legislative regulation. Experimental challenge studies confirmed that this genotype III isolate showed higher morality rates compared to other genotype III isolates. The source of infection is still unknown, but wild reservoirs are suspected. A major aim is to investigate wild fish along the Norwegian cost for detection of VHSV. New isolates will be characterized and further investigated along with the genotype III isolated from 2007. So far we have tested almost 3000 fish from 36 different fish species. Results from the screening and tentative challenge trials will be presented.

En av de syke hummerne hadde egg, og DNAanalyser avslørte at faren er en europeisk hummer. Dette er første gang det er bevist at disse to artene kan parre seg i naturen.

Viral hemorrhagic septicemia virus (VHSV) infects a wide range of marine fish species. To study the occurrence of VHSV in wild marine fish populations in Norwegian coastal waters and fjord systems a total of 1927 fish from 39 different species were sampled through 5 research cruises conducted in 2009 to 2011. In total, VHSV was detected by rRT-PCR in twelve samples originating from Atlantic herring (Clupea harengus), haddock (Melanogrammus aeglefinus), whiting (Merlangius merlangus) and silvery pout (Gadiculus argenteus). All fish tested positive in gills while four herring and one silvery pout also tested positive in internal organs. Successful virus isolation in cell culture was only obtained from one pooled Atlantic herring sample which shows that today's PCR methodology have a much higher sensitivity than cell culture for detection of VHSV. Sequencing revealed that the positive samples belonged to VHSV genotype Ib and phylogenetic analysis shows that the isolate from Atlantic herring and silvery pout are closely related. All positive fish were sampled in the same area in the northern county of Finnmark. This is the first detection of VHSV in Atlantic herring this far north, and to our knowledge the first detection of VHSV in silvery pout. However, low prevalence of VHSV genotype Ib in Atlantic herring and other wild marine fish are well known in other parts of Europe. Earlier there have been a few reports of disease outbreaks in farmed rainbow trout with VHSV of genotype Ib, and our results show that there is a possibility of transfer of VHSV from wild to farmed fish along the Norwegian coast line. The impact of VHSV on wild fish is not well documented.

Dissertation for the degree philosophiae doctor (PhD) at the University of Bergen The cultivation of marine larvae is often associated with high rates of mortality and is regarded as a bottle-neck in maintaining stable levels of production of juveniles for on-growing. In the course of the past few decades a great deal of effort has been put into increasing the production of farmed marine species. In spite of this, production is still low, compared to the production of salmonids, partly due to problems that arise during the early stages of life. The mortality problem is thought to be caused by bacteria that originate either from larval or live feed cultures. These are systems with high densities of biological waste and debris which may contribute to the growth of opportunistic bacteria. Vibrio spp. have often been suggested as causal agents. Because of the complex microbiological composition involved in larval and live feed cultures, and the small size of the larvae and feed organisms, isolating bacteria and describing their roles may be demanding. In order to identify pathogenic bacteria, reliable infection models are needed. The aim of this thesis is to enhance the knowledge of the bacteria associated with the cultivation of marine species. Experimental model organisms were great scallop Pecten maximus, cod Gadus morhua, halibut Hippoglossus hipposlossus, and turbot Scophthalmus maximus. The present work utilizes the multi-dish system as a model for bath challenge experiments using several species of marine larvae (Papers I-III, V). The method is reliable and efficient both as means of minimizing the number of larvae needed and for saving time and work space. It also provides individual control. In Paper I, six candidates of bacteria pathogenic to great scallop larvae were tested. In a similar experiment with cod larvae, 53 bacterial strains were tested for virulence (Paper II). Both studies included two controls i.e. unchallenged larvae as negative control and one larval group challenged with a known pathogen Vibrio pectenicida (Paper I) and Vibrio anguillarum serotype O2α (Paper II) as positive control. In addition to the positive controls, Vibrio splendidus (Paper I), four different strains of V. anguillarum and Carnobacterium sp. strain were verified as highly pathogenic (Paper II). The attempt to serotype the V. anguillarum strains produced inconclusive results (Paper 12 II), and further studies are needed. The study described in Paper II also show that most bacteria associated with diseased fish is not a primary cause to disease. Studies of the susceptibility of cod, halibut and turbot larvae to various serotypes of V. anguillarum and Vibrio spp. produced high mortality in all groups challenged with the O2α serotype. Cod and halibut larvae also suffered high mortalities when challenged with O1 (Paper V). The immunohistochemical examinations of larvae challenged with serotype O2α showed little or no pathology, leading to the hypothesis that extracellular products or toxins were killing the larvae (Paper III, V). Serotype O1 caused severe pathology (Paper V).

A multidish system was used to perform the bath challenge experiments. Results of the immunohistochemical examinations indicated a variety of histopathology. The Vibriosis pathogens affecting aquaculture are identified including V. anguillarum, V. ordalii, and V. salmonicida.

Heart and skeletal muscle inflammation (HSMI) is a disease that affects farmed Atlantic salmon Salmo salar L. several months after the fish have been transferred to seawater. Recently, a new virus called piscine reovirus (PRV) was identified in Atlantic salmon from an outbreak of HSMI and in experimentally challenged fish. PRV is associated with the development of HSMI, and has until now only been detected in Atlantic salmon. This study investigates whether the virus is also present in wild fish populations that may serve as vectors for the virus. The virus was found in few of the analyzed samples so there is probably a more complex relationship that involves several carriers and virus -reservoirs.

In intensive aquaculture systems, high concentrations of nutrients and high densities of fish larvae provide favorable conditions for opportunistic pathogenic bacteria to flourish. We screened potentially pathogenic bacterial strains isolated from moribund Atlantic cod Gadus morhua larvae, pollack Pollachius pollachius, coalfish Pollachius virens, Atlantic halibut Hippoglossus hippoglossus, rotifers, algae and water samples from different hatcheries. Three identical challenge experiments tested a total of 53 strains. A multidish system was used: cod eggs were placed in single wells, together with 2 ml of sterile seawater, and exposed to the bacterial cultures. Final bacterial concentrations in the wells were 10(6) and 10(4) CFU ml(-1). Eggs and larvae not exposed to bacteria were used as unchallenged controls. Challenged controls were exposed to Vibrio anguillarum strain 610. Eggs were challenged approximately 48 h prior to hatching and mortality was recorded daily throughout the yolk-sac period. In spite of the high challenge dose of 106 CFU ml(-1), only 5 bacterial strains tested caused higher mortality than the unchallenged controls. Four of these strains were identified by 16S rDNA and gyrase B gene (GyrB) sequencing as resembling V. anguillarum and 1 strain resembled Carnobacterium sp. Most of the larvae exposed to these strains died within 10 d of challenge. Serotyping of the strains resembling V. anguillarum gave inconclusive results. This indicates differences in serology compared to the serotypes O1, O2 and O3, associated with disease. Three bacterial strains seemed to have a slower infection rate, indicating a longer incubation period. The remaining 45 strains did not seem to have a negative effect on larval survival, suggesting that these are not primary pathogens.

Farming of Atlantic cod Gadus morhua is one of the most rapidly growing sectors of Norwegian aquaculture. Classical vibriosis caused by Vibrio anguillarum is a problem in cod aquaculture. To prevent disease outbreaks, a thorough understanding of the infection route and the impact of the bacteria on the host is important. The intestinal tract, skin and gills have all been proposed as routes of entry for bacterial infections such as vibriosis. We aimed to further develop understanding of V anguillarum serotype O2alpha infections in cod larvae by elucidation of a possible route of entry, the pattern of infection and its histopathology. Cod eggs were transferred to a 24-well polystyrene multi-dish with 2 ml of sterile aerated 80% (28 per thousand salinity) seawater. Challenge doses were 10(4) and 10(6) CFU ml(-1). Unchallenged larvae were used as controls. Larvae for immunohistochemical examination were sampled daily from each group. In most of the larvae, either no or very few bacteria were observed. Typical findings were clusters of bacteria in the spaces between the primary gill lamellae. None of these bacteria seemed to have adhered to the gills. Intestines of 3 out of 161 larvae examined contained positively immunostained bacteria. Some bacteria appeared attached to the microvilli, but none was observed inside epithelial cells. Only 2 larvae from the low-challenge dose group showed clear signs of histopathology, which occurred in the intestine. It is not possible to draw any conclusions regarding the portal of entry.

Three challenge experiments were carried out on larvae of the great scallop Pecten maximus. Larvae were bath-challenged with Vibrio pectenicida and 5 strains resembling Vibrio splendidus and one Pseudoalteromonas sp. Unchallenged larvae were used as negative controls. The challenge protocol was based on the use of a multidish system, where the scallop larvae (10, 13 and 15 d post-hatching in the 3 experiments, respectively) were distributed to 2 ml wells with stagnant seawater and exposed to the bacterial cultures by bath challenge. Presence of the challenge bacteria in the wells was verified by polymerase chain reaction (PCR). A significantly increased mortality was found between 24 and 48 h in most groups challenged with V. pectenicida or V. splendidus-like strains. The exception was found in larval groups challenged with a Pseudoalteromonas sp. LT 13, in which the mortality rate fell in 2 of the 3 challenge experiments. Larvae from the challenge experiments were studied by immunohistochemistry protocol. Examinations of larval groups challenged with V. pectenicida revealed no bacterial cells, despite a high degree of positive immunostaining. In contrast, intact bacterial cells were found in larvae challenged with V. splendidus. In the case of larvae challenged with the Pseudoalteromonas sp., positive immuno-staining was limited to visible bacteria inside the digestive area and cells of the mucosa. The experiments confirm that V. splendidus and V. pectenicida are pathogenic to scallop larvae, and that the Pseudoalteromonas strain is probably not a primary pathogen, although it cannot be ruled out as a secondary pathogen.
