BACKGROUND: Suprahyoid muscles behavior during the tongue lifting movement has not yet been elucidated. OBJECTIVE: The purpose of this study was to investigate the potential of elastography imaging to examine developmental oral dysfunction in children and oral hypofunction in older adults using sonography. METHODS: Tongue pressure was measured using a manometer with a probe. The tongue pressure was measured with simultaneously scanning the geniohyoid muscle (GHM) and the anterior belly of the digastric muscle (DGM) using sonographic elastography. Sagittal images of the GHM and coronal images of the DGM were used for the strain ratio measurement. The strain ratio of the muscles was measured three times for each subject with the tongue pressure values of 0–30 kPa. RESULTS: The strain ratio of the GHM were higher than those of the DGM at tongue pressure of 10, 20 and 30 kPa. The strain ratio of the GHM increased as the tongue pressure increased in all participants. In contrast, the strain ratio of the DGM tended to slowly decrease as tongue pressure increased in female participants. CONCLUSION: Sonographic elastography is useful for visual and quantitative evaluation of elastic properties in suprahyoid muscles during tongue lifting movements.

Saccharomyces cerevisiae has 6 synonymous tRNATrpCCA genes encoding the identical sequence, including their intronic region. They are supposed to express tRNATrpCCA in the same quality and quantity. Here, we generated single to quintuple deletion strains with all the possible combinations of the synonymous tRNATrpCCA genes to analyze whether those individual genes equally contribute cell viability and tRNA production. The quintuple deletion strains that only harbor tW(CCA)J, tW(CCA)M, or tW(CCA)P were viable but almost lethal while the other quintuple deletions showed moderately impaired growth. These growth differences were not obvious among the quadruple deletion strains, which expressed almost one third of mature tRNATrpCCA in the wild type. Therefore, no dosage compensation operates for tRNATrpCCA amount, and growth variations among the quintuple deletion strains may not simply reflect differences in tRNATrpCCA shortage. Yeast may retain the redundancy of tRNATrpCCA genes for a noncanonical function(s) beyond the supply of the tRNA to translation.

Yeasts generally grow with a highly glycolytic metabolism and restrain mitochondrial biogenesis except for some Fe/S proteins. Respiratory mitochondrial functions and biosynthesis pathways are well studied, however how cells coordinate basal fermentative mitochondrial functions is not fully understood. We show that a part of nuclear-encoded mitochondrial mRNAs, which preferentially translated in monosomes, are regulated by Puf3p upon glucose-rich media. Especially, of those monosome-enriched nuclear-encoded mitochondrial mRNAs, CAT5/COQ7 mRNA has a variant of the canonical Puf3p binding site on its 3’-UTR. Western blot analysis showed that Puf3p represses the translation of Cat5p regardless of fermentable or respiratory media. In vitro binding assay revealed that Puf3p directly binds to CAT5 mRNA via the non-canonical Puf3p binding site.Mutants harboring the substitution of the non-canonical Puf3p binding site in CAT5 mRNA grew normally but impaired Cat5p expressions apparently, indicating CAT5 mRNA is a bona fide Puf3p target. Overall, Puf3p, a general key modulator for nuclear-encoded mitochondrial mRNAs, fine-tunes translation of a subset of nuclear-encoded mitochondrial mRNAs including mRNAs with non-canonical Puf3p binding sites under the fermentation. This may be required to keeping the fundamental functions of yeast mitochondria at proper levels.

In eukaryotes and archaea, tRNA genes frequently contain introns, which are removed during maturation. However, biological roles of tRNA introns remain elusive. Here, we constructed a complete set of Saccharomyces cerevisiae strains in which the introns were removed from all the synonymous genes encoding 10 different tRNA species. All the intronless strains were viable, but the tRNAPheGAA and tRNATyrGUA intronless strains displayed slow growth, cold sensitivity and defective growth under respiratory conditions, indicating physiological importance of certain tRNA introns. Northern analyses revealed that removal of the introns from genes encoding three tRNAs reduced the amounts of the corresponding mature tRNAs, while it did not affect aminoacylation. Unexpectedly, the tRNALeuCAA intronless strain showed reduced 5.8S rRNA levels and abnormal nucleolar morphology. Because pseudouridine (Ψ) occurs at position 34 of the tRNAIleUAU anticodon in an intron-dependent manner, tRNAIleUAU in the intronless strain lost Ψ34. However, in a portion of tRNAIleUAU population, position 34 was converted into 5-carbamoylmethyluridine (ncm5U), which could reduce decoding fidelity. In summary, our results demonstrate that, while introns are dispensable for cell viability, some introns have diverse roles, such as ensuring proper growth under various conditions and controlling the appropriate anticodon modifications for accurate pairing with the codon.

Early detection of oral disease is important to reduce its severity and increase the likelihood of successful treatment. This study aimed to perform a quantitative assessment of the saliva components as a first stage of the research to screen oral homeostasis. Here, saliva secretions collected from children were evaluated, and their constituents were analyzed to investigate the potential correlations between the buffering capacity and a range of salivary factors. Subjects aged 3-16 years in the primary, mixed, or permanent dentition stage, were selected for this study. The following salivary factors were analyzed: flow rate, total protein, total sugar quantifications, and constituent analyses using RT-PCR and western blotting. The associations between each factor and the buffering capacity were then analyzed using multiple regression analysis. Flow rate, BPIFA2 RNA level, histatin 1 and BPIFB1 protein levels as well as female sex were positively associated with buffering capacity. In contrast, total sugar concentration and MUC7 RNA levels showed a negative relationship with the buffering capacity. Some of these constituents may indicate oral homeostasis and are therefore potential biomarkers of oral health status. These results suggest that the analyses of the correlations between oral homeostasis and salivary factors are an effective strategy for identifying the susceptibility to oral diseases.

The present study was performed to investigate the mineral density distribution in enamel and dentin for both permanent and primary teeth and to establish the standard density per tooth type using micro-computed tomography (CT). Fifty-seven extracted human teeth (37 permanent, 20 primary) were evaluated in the present study. The enamel and dentin mineral densities in the extracted teeth were measured using micro-CT. Cubic regression curves were used to determine the mineral density distribution in the enamel and dentin for each tooth type. The mean values, distributions, and regression equations of the mineral densities were obtained. The mean mineral density values for permanent enamel and dentin were significantly higher than those for their primary counterparts for each tooth type. In the present study, we demonstrated the distribution of mineral density in sound enamel and dentin and attempted to determine the standard mineral density for each tooth type using micro-CT. The mineral density distributions found in this study contribute to our understanding of the mechanical properties of enamel and dentin. A positive correlation suggests that the systemic bone mineral density could be predicted based on the analysis of exfoliated teeth, such as in patients with hypophosphatasia. The present results may be useful in establishing a numerical standard for the mechanism involved in root fracture and for early detection of root fracture risk.

We previously showed that mRNA expression of BPIFB1 (Bpifb1), an antibacterial protein in the palate, lung, and nasal epithelium clone protein family, was increased in parotid acinar cells in non-obese diabetic (NOD, NOD/ShiJcl) mice, which is an animal model for Sjögren's syndrome. However, we did not previously assess the protein levels. In this report, we confirmed the expression of BPIFB1 protein in the parotid glands of NOD mice. Immunoblotting of subcellular fractions revealed that BPIBB1 was localised in secretory granules in parotid glands from NOD mice, and was almost not in parotid glands from the control mice. BPIFB1 had N-linked glycan that reacted with Aleuria aurantia lectin, which caused two types of spots with a slightly different pI and molecular weight. The expression of BPIFB1 protein was also demonstrated by immunohistochemistry. BPIFB1 was detected in the saliva from NOD mice but not in the saliva from the control mice, indicating individual constitution. BPIFB1 in saliva may be applied to other research as a diagnostic marker.

Cardiac resynchronization therapy (CRT) has been increasingly performed in patients having heart failure with dyssynchrony. We report a successful case of total thoracoscopic left ventricular (LV) lead implantation in CRT. A 77-year-old man with marked dyssynchrony of the LV wall motion and a low ejection fraction (EF17%) due to pacemaker-mediated cardiomyopathy was referred to us. CRT was planned, but percutaneous LV lead implantation proved difficult owing to anatomical variations. The LV lead was placed in the post-lateral wall of the LV base using a total thoracoscopic procedure. Preoperative dyspnea and dyssynchrony were clearly improved. In CRT, the LV wall stimulation site is important. The LV lead should be implanted in the latest activation area, which can be detected using speckle tracking echocardiography. Surgical lead implantation can be performed in the ideal area, and this procedure may play a new role as a hybrid CRT.

The use of agar gel electrophoresis and bioautography for the identification of small amounts of seven aminoglycoside antibiotics (streptomycin, dihydrostreptomycin, kanamycin, bekanamycin, gentamicin, fradiomycin and paromomycin) was studied. The procedure used for the electrophoretic separation of these components is described. The antibiotics were detected with high sensitivity using Bacillus subtilis ATCC 6633 as a test organism, Tris buffer at pH 8.0, and Bacto-Agar as the supporting medium for each of the components. The limits of detection obtained for the various antibiotics ranged between 0.078 and 0.313 μg/ml and the standard deviation ranged from 0.05 and 0.34. Recoveries from extracts of bovine kidney tissue containing each drug ranged from 59.0% to 90.2%. The sensitivity of this method can be adjusted through minor modifications, thus allowing it to be used for routine residual analysis of aminoglycosides in biological samples.

RNA localization is an important event that is essential for the polarization and differentiation of a cell. Although several methods are currently used to detect localized RNAs, a simplified detection system has not yet been developed for Schizosaccharomyces pombe. In the present study, we describe a new vector system for the visualization of localized RNAs in S. pombe using a U1A-tag-GFP system. A pREP1-U1A-tag vector plasmid to express U1A-tagged RNA and a pREP2-U1AGFP plasmid to produce a U1A-GFP fusion protein were constructed for this system. Since the U1A-GFP protein binds U1A-tagged RNA, fluorescence is observed at the location of U1A-tagged RNA in cells expressing both of these. The nucleolar localization of U3 snoRNA was successfully detected using this system, and a novel RNA localized at the DNA region of the nucleus was found by screening localized RNAs. This system will accelerate the study of localized RNAs in S. pombe.

A thoracic aortic pseudoaneurysm is a life-threatening complication following thoracic aortic surgery. We describe a surgical strategy for this pseudoaneurysm with a high risk for rupture during median sternotomy. The pseudoaneurysm was distended and widely adherent to the posterior sternum. Elective cardiopulmonary bypass and moderate hypothermia were established, and sternotomy was performed without left ventricle distention or brain ischemia. Total arch replacement was successful and the patient was discharged on post operative day (POD) 18. A key surgical strategy was to avoid ventricular fibrillation before sternotomy. Appropriate sternotomy timing and perfusion strategy are crucial for successful treatment.

Objective: Most cases of hypophosphatasia (HPP) exhibit early loss of primary teeth. Results of micro-computed tomography (micro-CT) analysis of teeth with HPP have not yet been reported. The purpose of the present study was to describe the size and mineral density distribution and mapping of exfoliated teeth with HPP using micro CT. Study design: Seven exfoliated teeth were obtained from a patient with HPP. Exfoliated teeth sizes were measured on micro CT images and mineral densities of the mandibular primary central incisors were determined. Results: Partial dentures were fabricated for the patient to replace the eight primary teeth which had exfoliated. Most primary teeth sizes were within the normal range. The mean values of enamel and dentin mineral densities in teeth with HPP were 1.35 and 0.88 g/cm3, respectively, in the mandibular primary central incisors. Conclusion: Mineral density distribution and mapping revealed that the values in teeth with HPP were lower than the homonymous teeth controls in all regions from the crown to apex. Furthermore, it was demonstrated that the differences between HPP and controls were larger on the crown side and the differences tended to converge on the apex side. These results suggested that the present patient showed mild hypomineralization in the primary dentition.

The purpose of the present study was to develop a novel-designed phantom that could be scanned with a sample in the same image, that specialize for quantitative analyses of human dentin mineral density using the X-ray attenuation method. A further attempt was made to demonstrate the intracoronal dentin mineral density using this phantom in mandibular incisors. The phantom prepared with a 15 mm hole in the center of an acrylic resin bar having an outside diameter of 25 mm and 8 small holes (diameter, 3 mm) were made at equal intervals around the center. Liquid dipotassium hydrogen phosphate (K2HPO4) solutions were established at 0.4, 0.6, 0.8 and 1.0 g/cm3, and were arranged to these holes. The mean value of the intracoronal dentin mineral density was 1.486 ± 0.016 g/cm3 in the present study. As the results of the present study corresponded to previous reports, this new phantom was considered to be useful. This phantom enables the analysis of samples that are not readily available by conventional mechanical tests and may facilitate biomechanical investigations using X-ray images. It was suggested that this system is a simple, accurate and novel mineralization measuring system.

The molecular sieving effect of tetrahydrofuran (THF) clathrate hydrate was experimentally investigated. The capture and release of seven gases, i.e., helium (He), hydrogen (H2), neon (Ne), nitrogen (N2), xenon (Xe), methane (CH4), and ethane (C2H6), in THF clathrate hydrate were investigated. For gases with a diameter smaller than the window diameter, the order of the gas uptake rates in the THF clathrate hydrate was in accordance with the order of their van der Waals diameters, except for He. This result indicates that the gas uptake rate is generally controlled by the size of the gas molecules. The apparent diffusion coefficients for these gases in the THF clathrate hydrate were estimated. The van der Waals diameters of CH4 and Xe are larger than the window diameter. However, gas uptake by THF clathrate hydrate was observed for both gases. This can be attributed to the formation of the type I clathrate hydrate of these gases. No significant gas uptake of ethane was observed. Based on the results, H2/N2 and H2/C2H6 separation experiments were conducted. Gas separation of H2/N2 by THF clathrate hydrate was successfully achieved using the difference in their gas uptake rates of the gases, and the separation ratios were in the range 4–16 mol/mol. Gas separation of H2/C2H6 was also achieved utilizing the molecular sieving effect of THF clathrate hydrate.

A rapid and simple determination method of seven fungicides, thiabendazole (TBZ), pyrimethanil (PYR), o-phenylphenol (OPP), fludioxonil (FLD), azoxystrobin (AZX), imazalil (IMZ) and diphenyl (DP) in citrus fruits by LC-MS and HPLC-FL was developed. The seven fungicides were extracted with acetonitrile from citrus fruits and cleaned up with Z-Sep/C18 cartridges. The LC separation was performed on a phenyl-hexyl column with methanol-acetonitrile-10 mmol/L ammonium formate (10 : 35 : 55) as a mobile phase. The recoveries from citrus fruits fortified with the compounds at the MRLs and at 0.1 μg/g ranged from 85.4 to 106.3% and from 75.8 to 99.7%, respectively. The quantitation limits (S/N=10) were 0.03-0.07 μg/g.

Dental cone-beam computed tomography (CBCT) received regulatory approval in Japan in 2000 and has been widely used since being approved for coverage by the National Health Insurance system in 2012. This imaging technique allows dental practitioners to observe and diagnose lesions in the dental hard tissue in three dimensions (3D). When performing routine radiography, the examination must be justified, and optimal protection should be provided according to the ALARA (as low as reasonably achievable) principles laid down by the International Commission on Radiological Protection. Dental CBCT should be performed in such a way that the radiation exposure is minimized and the benefits to the patient are maximized. There is a growing demand for widespread access to cutting-edge health care through Japan’s universal health insurance system. However, at the same time, people want our limited human, material, and financial resources to be used efficiently while providing safe health care at the least possible cost to society. Japan’s aging population is expected to reach a peak in 2025, when most of the baby boomer generation will be aged 75 years or older. Comprehensive health care networks are needed to overcome these challenges. Against this background, we hope that this text will contribute to the nation’s oral health by encouraging efficient use of dental CBCT.

MSA1 mRNA encodes Msa1p, a protein associated with the SCB-binding factor (SBF) and MCB-binding factor (MBF) complex. Msa1p promotes the transcription of G1 phase-specific genes, and is subjected to cell cycle-dependent regulation for its abundance and subcellular localization. MSA1 mRNA and Msa1p levels oscillate in the cell cycle with peaks at the late M/early G1 phase and early G1 phase, respectively. Phosphorylation by CDK1 negatively regulates the nuclear localization of Msa1p. In the present study, we identified MSA1 mRNA as a bud tip-localized mRNA in screening using a Tag-GFP system. A fragmentation analysis revealed a sequence of ~145 bases for the bud tip localization. Endogenous MSA1 mRNA localized at the bud tip in a manner that depended on SHE2. Msa1p levels were also affected by SHE2 in cells constitutively expressing MSA1 mRNA. These results suggest the existence of a regulatory mechanism for Msa1p through the localized control of MSA1 mRNA.

Ribosome stalling is an important incident enabling the cellular quality control machinery to detect aberrant mRNA. Saccharomyces cerevisiae Hbs1-Dom34 and Ski7 are homologs of the canonical release factor eRF3-eRF1, which recognize stalled ribosomes, promote ribosome release, and induce the decay of aberrant mRNA. Polyadenylated nonstop mRNA encodes aberrant proteins containing C-terminal polylysine segments which cause ribosome stalling due to electrostatic interaction with the ribosomal exit tunnel. Here we describe a novel mechanism, termed premature translation termination, which releases C-terminally truncated translation products from ribosomes stalled on polylysine segments. Premature termination during polylysine synthesis was abolished when ribosome stalling was prevented due to the absence of the ribosomal protein Asc1. In contrast, premature termination was enhanced, when the general rate of translation elongation was lowered. The unconventional termination event was independent of Hbs1-Dom34 and Ski7, but it was dependent on eRF3. Moreover, premature termination during polylysine synthesis was strongly increased in the absence of the ribosome-bound chaperones ribosome-associated complex (RAC) and Ssb (Ssb1 and Ssb2). On the basis of the data, we suggest a model in which eRF3-eRF1 can catalyze the release of nascent polypeptides even though the ribosomal A-site contains a sense codon when the rate of translation is abnormally low.

Sinodielide A (SA) is a naturally occurring guaianolide, which is isolated from the root of Sinodielsia yunnanensis. This root, commonly found in Yunnan province, is used in traditional Chinese medicine as an antipyretic, analgesic and diaphoretic agent. A number of studies have reported that agents isolated from a species of Umbelliferae (Apiaceae) have antitumor activities. We previously reported, using combined treatments with this medicinal herb and hyperthermia at various temperatures, an enhanced cytotoxicity in the human prostate cancer androgen‑independent cell lines, PC3 and DU145, and analyzed the related mechanisms. In the present study, we investigated the effects of treatment with SA prior to hyperthermia on the thermosensitivity of DU145 cells, and the mechanisms related to the induction of apoptosis and G(2)/M cell cycle arrest via the activation of extracellular-regulated kinase (ERK)1/2, c-Jun N-terminal kinase (JNK) mitogen-activated protein kinase (MAPK) signaling pathways, as well as the phosphoinositide 3-kinase (PI3K)/Akt signaling pathways. Cells were exposed to hyperthermia alone (40-44˚C) or hyperthermia in combination with SA. Lethal damage to cells treated with mild hyperthermia (40 or 42˚C) for up to 6 h was slight; however, hyperthermia in combination with SA synergistically enhanced thermosensivity. Lethal damage to cells treated with acute hyperthermia (43 or 44˚C) was more severe, but these effects were also enhanced and were more significant by the combined treatment with SA. The kinetics of apoptosis induction and cell cycle distribution were analyzed by flow cytometry. In addition, the levels of ERK1/2, JNK and Akt were determined by western blot analysis. The incidence of apoptotic cells after treatment with SA (20.0 µM) at 37˚C for 4 h, hyperthermia (44˚C) alone for 30 min, and the combination in sequence were examined. The sub-G1 division (%) in the diagram obtained by flow cytometry was applied to that assay. The percentage of apoptotic cells (10.53±5.02%) was higher at 48 h as compared to 0, 12 and 24 h after treatment. The distribution of DU145 cells in the G2/M cell cycle phase was markedly increased after 24 h of heating at 44˚C and after the combined treatment with heating and SA. The phosphorylation of ERK1/2 was reduced following treatment with heating and SA, while the levels of phosphorylated JNK (p-JNK) were markedly increased immediately after heating at 44˚C and when heating was combined with SA. By contrast, the levels of phosphorylated Akt (p-Akt) were immediately increased only after heating at 44˚C. Thus, we concluded that SA exerts its thermosensitizing effects on DU145 cells by inhibiting the activation of the MAPK/ERK1/2 and PI3K/Akt signaling pathways.

A 94-year-old woman with rupture of a thoracic aortic aneurysm (rTAA) was referred to us. She previously underwent thoracic endovascular aortic repair and was considered to be at high risk for a conventional open operation. Therefore an endovascular procedure was planned. The proximal landing zone needed to be placed at the ascending aorta to seal a type 1a endoleak. A hybrid operation consisting of supraaortic total debranching on the common femoral artery and endovascular repair was performed. All debranched bypasses were patent and the aneurysm was excluded. The patient regained sufficient ambulatory strength and showed no symptoms of syncope.

Prevention is better than cure. Public health plays an important role in promoting prevent medicine. To obtain the abilities to provide appropriate nursing services, learning public health is necessary for students who want to become registered nurses. When teachers teach public health to nursing students, it is important to motivate them to learn it. Therefore, we investigated the reasons for the lack of motivation to learn public health by conducting a questionnaire survey. The subjects were female nursing students in 29 vocational schools in Kanagawa and Chiba prefectures of Japan that allow graduation after a 3-year study period. We asked the students whether or not they had completed the subject of public health and analyzed those students who answered affirmatively. We analyzed 1,553 respondents whose average age was 22.6 ± 5.2 years (range, 18 to 45). Using factor analysis, we discovered the 5 reasons that lead to the lack of nursing students' motivation to learn public health: "Difficulties acquiring knowledge of public health," "Inappropriate attitudes of public health teachers," "Thinking lightly about the national examination in the field of public health," "Lack of understanding the importance of learning public health," and "Future plans that do not specialize in public health." Using multiple linear regression analysis, these 5 reasons were significant predictors for the lack of students' motivation. Older students also had significantly less motivation to learn public health than did younger students. When teachers instruct their students, they should teach public health better with the present knowledge.

The purpose of the present study was to examine the fracture toughness and Vickers microhardness number of permanent and primary human enamel using the indentation microfracture method. Crack resistance and a parameter indirectly related to fracture toughness were measured in 48 enamel specimens from 16 permanent teeth and 12 enamel specimens obtained from six primary teeth. The Vickers microhardness number of the middle portion was greater than the upper portion in primary enamel. The fracture toughness was highest in the middle portion of permanent enamel, because fracture toughness greatly depends upon microstructure. These findings suggest that primary teeth are not miniature permanent teeth but have specific and characteristic mechanical properties.

Numerical simulations have recently increased in scale and have often output high dimensional (three-dimensional and time-evolving) datasets. This makes it difficult for users to quickly grasp physical phenomena involved in such datasets. To overcome this difficulty, we propose a spatio-temporal information mapping technique with a map-design capability (spatio-temporal map) by using an information visualization technique. The spatio-temporal map is generated by mapping the values of a three-dimensional and time-evolving physical quantity into a two-dimensional space with spatial and temporal axes. Here, three-dimensional spatial information is condensed into one dimension by subdividing a target model with an octree. By using the map, users can quickly find regions of interest involved in high dimensional datasets. In addition, users can interactively change several aspects of the map such as its resolution and color coding method. Furthermore, users can design the map by changing the tree structure. By applying the spatio-temporal map to a full-scale three-dimensional vibration simulator for an entire nuclear power plant, we confirmed that the map is a useful technique to quickly identify appropriate regions of interest.

RNA localization is a common mechanism for recruiting proteins to specific regions of a cell, which causes cell polarization and sometimes asymmetric division. We found that EGD1 mRNA accumulates dose-dependently as a cytoplasmic granule in Saccharomyces cerevisiae. EGD1 encodes a β-subunit of the nascent polypeptide-associated complex (NAC). NAC is a heterodimer consisting of α- and β-subunits, associated with ribosomes and thought to be involved in the folding of nascent polypeptide chains. Analysis of deletion constructs showed that the localization of EGD1 mRNA requires both an upstream region and an ORF of EGD1, suggesting that the translation of Egd1p is important for localization. We also showed that Egd1p and P-body components are co-localized with EGD1 mRNA. This granule, named the EGD1 granule, has features similar to cellular inclusions containing aggregated proteins. Disruption of microtubules by treatment with a drug, benomyl, resulted in loss of the EGD1 granule. When the expression level of EGD2 encoding the αNAC increased, the percentage of cells showing the EGD1 granule decreased, suggesting that the granular distribution of EGD1 depends on the quantitative balance between α- and β-subunits of NAC. Taken together, we propose a novel microtubule-dependent mechanism for controlling NAC through RNA localization.

In the Japanese population, the second premolars are the secondmost- frequently impacted teeth in the mandible, after the third molars. Although some papers have reported on the treatment of impacted mandibular second premolars caused by abnormal tilting of the tooth germ, there has been little information regarding bilateral horizontal impaction with distal inclination. The purpose of the present report is to describe the treatment course of a rare and severe case of an 11-year, 8-month-old girl without any obvious etiologies whose bilateral mandibular second premolars were inclined to the distal direction at approximately 90 degree angles, with the root formation developed to an approximately half length. After extraction of the retained primary molars, traction was performed in the treatment of the affected teeth. The affected premolars had fully erupted and achieved occlusion one year after the beginning of intervention. In conclusion, it was considered that the present case could obtain a successful outcome by limited traction since no clinical anomalies or discomfort was detectable during the one year of retention.

The purpose of this report was to describe an unusual severe ectopic eruption of the bilateral maxillary canines in a young patient aged 7 years and 8 months and to discuss the effects of early intervention such as extraction of the predecessors and fenestration of the affected canines. The patient was referred to the author's clinic for a consultation regarding bilateral ectopically erupting maxillary canines with an ectopic maxillary right first molar. After improvement of the first molar, the affected canines were fully erupted and aligned in occlusion 3 years and 7 months after the extraction of primary canines. Before treatment, the findings on the orthopantomogram were as follows, there was no direct cause, both canine tooth germs were situated in very high positions, and the cusp tips of both canines were not overlapped with the roots of the adjacent lateral incisors (Sector 1). The maxillary left canine impacted horizontally in a mesio-distal direction (axial angulation: 90 degrees) could be successfully guided to eruption by early intervention such as the extraction of predecessors and three fenestrations. Even in severe ectopic cases, therefore, it is considered that traction may be postponed until no effect of fenestration can be detected in cases of early discovery and intervention when the crown top is situated in Sector 1.

Center for computational science and e-systems of Japan Atomic Energy Agency is carrying out R&D in the area of extra large-scale simulation technologies for solving nuclear plant structures in its entirety. Specifically, we focus on establishing a virtual plant vibration simulator on inter-connected supercomputers intended for seismic response analysis of a whole nuclear plant. The simulation of a whole plant is a very difficult task because an extremely large dataset must be processed. To overcome this difficulty, we have proposed and implemented a necessary simulation framework and computing platform. The computing platform enables an extra large-scale whole nuclear plant simulation to be carried out on a grid computing platform called ITBL-IS, Information Technology Based Laboratory Infrastructure and AEGIS, Atomic Energy Grid Infrastructure. The simulation framework based on the computing platform has been applied to a linear elastic analysis of the reactor pressure vessel and cooling systems of the nuclear research facility, HTTR. The simulation framework opens a possibility of new simulation technologies for building a whole virtual nuclear plant in computers for virtual experiments.

The aim of the present study was to examine the prevalence rate and treatment outcome of maxillary canine transposition in Japanese children according to the conventional standard and our new additional consideration of the classification. From 140 cases with ectopic canines diagnosed in the Pediatric Dental Clinic of Niigata University Medical and Dental Hospital, we selected 10 transposition cases as the present subjects. Out of 10 cases, 4 were males and 6 females. Six were classified into type A: the mesio-distal positional interchange of the canine and the first premolar, 2 cases into type B: the mesio-distal transposition of the canine and the lateral incisor, and the other 2 cases into type C: the present new classification that showed the vertical transposition of the canine and the first premolar within the developmental bone. The prevalence rate of types A and B resembled the previous papers regarding the ratio of sex and bilateral occurrence. Although 3 cases in type A, one case in type B and one case in type C could finally be aligned in the normal tooth order, every one of the types A and B cases were aligned in the reverse order. For every one case in types A and C, the transposed canines were extracted. It is recommended that pediatric dentists take an orthopantomogram during early mixed dentition as a routine practice in order to detect anomalies of tooth eruption, such as tooth transposition, as early as possible.

The aim of the present study was to examine retrospectively how best to treat severe cases of ectopically erupting maxillary canines with mesial angulation exceeding 50 degrees. From the ectopically erupting canines diagnosed at the Pediatric Dental Clinic of Niigata University Hospital, we selected for our study 14 severe cases in which mesial angulation exceeded 50 degrees. Nine ectopically erupting canines could be aligned within the arch after traction, and two canines are in the course of treatment, while three canines were removed. In the removed canines, the vertical length from the occlusal line of the crown top was shorter than that in the aligned canines with similar initial condition. This means that the vertical length to the occlusal line of the impacted canine influences the treatment complexity. In four similar ectopically erupting canines in which mesial inclination was nearly 80 degrees or greater and vertical length was over 30 millimeters with Sector 1 or 2 positioning distal to the midline of the root of its adjacent lateral incisor, the initial disruptive conditions were improved after extraction of the predecessor and/or fenestration of the affected canine. It is concluded that traction should be postponed until a few months after extraction of the predecessor if the ectopically erupting canine is located far away from the occlusal line (over 30 millimeters) and its mesial inclination is more than nearly 80 degrees; also that this treatment plan should be limited to cases in which the crown top belongs to Sector 1 and 2.

47, XYY is a sex chromosome-number disorder occurring in males. Pediatric dentists seldom report encountering 47, XYY males, probably because there are no overt signs of the disease. The purpose of this report is to present the clinical oro-facial findings of the case of a 47, XYY boy based on cephalometric analysis and study model examinations as well as micro-CT measurement of tooth mineralization. An 11-year-old Japanese boy was referred to our clinic for extraction of primary teeth because of their prolonged retention. He had 47, XYY disorder, pulmonary hypertension, and slight mental retardation. Cephalometric analysis and study model examinations that included a comparison of tooth size and arch assessments were performed. A further attempt was made to demonstrate the degree of dentin mineralization in primary molars. The boy's height and weight were normal but his head circumference had exceeded the 94th percentile since 2 years of age. The cephalometric analysis showed an increase in the lower facial height and bimaxillary protrusion with a longer mandible. Study model examination revealed that the mesio-distal crown widths of all erupted primary and permanent teeth were larger than the standard values, except for the mandibular permanent canines. The degree of dentin mineralization was almost normal in the crown and root areas. A profilogram showed an increase in the lower facial height and bimaxillary protrusion with a longer mandible. The dental arch showed a remarkably contracted width but an extended length. The crown width was larger than the standard values, but the distribution of the degree of dentin mineralization differed little from normal.

Most cases of hypophosphatasia (HPP) exhibit early loss of primary teeth. Results of microcomputed tomography (micro-CT) analysis of teeth with HPP have rarely been reported. The purpose of the present study was to describe the mineral density distribution and mapping of exfoliated teeth from an HPP patient using micro-CT. Four exfoliated teeth were obtained from a patient with HPP. Enamel and dentin mineral densities of exfoliated teeth were measured on micro-CT. The mean values of enamel and dentin mineral densities in mandibular primary central incisors with HPP were 1.61 and 0.98 g/cm3, respectively. The corresponding values in the mandibular primary lateral incisors were 1.60 and 0.98 g/cm3, respectively. Enamel hypoplasia was seen in the remaining teeth, both maxillary and mandibular primary canines and first and second molars. Micro-CT enables nondestructive, noninvasive evaluation and is useful for studying human hard tissues obtained from patients.
