The introduction of Zika virus (ZIKV) to the Americas raised concern that the virus would spill back from human transmission, perpetuated by Aedes aegypti, into a sylvatic cycle maintained in wildlife and forest-living mosquitoes. In the Americas, Sabethes species are vectors of sylvatic yellow fever virus (YFV) and are therefore candidate vectors of a sylvatic ZIKV cycle. To test the potential of Sabethes cyaneus to transmit ZIKV, Sa. cyaneus and Ae. aegypti were fed on A129 mice one or two days post-infection (dpi) with a ZIKV isolate from Mexico. Sa. cyaneus were sampled at 3, 4, 5, 7, 14, and 21 days post-feeding (dpf) and Ae. aegypti were sampled at 14 and 21 dpf. ZIKV was quantified in mosquito bodies, legs, and saliva to measure infection, dissemination, and potential transmission, respectively. Of 69 Sa. cyaneus that fed, ZIKV was detected in only one, in all body compartments, at 21 dpf. In contrast, at 14 dpf 100% of 20 Ae. aegypti that fed on mice at 2 dpi were infected and 70% had virus in saliva. These data demonstrate that Sa. cyaneus is a competent vector for ZIKV, albeit much less competent than Ae. aegypti.

Mosquitoes of the Neotropical genus Sabethes, some species of which are yellow fever vectors, most often develop through the immature stages in tree holes. Sabethes eggs have not been previously characterized using scanning electron microscopy. Eggs of Sabethes cyaneus (length: 349.6 +/- 2.7 microm; width: 172.6 +/- 1.14 microm; n = 10) are almost biconical when examined from the top. From a lateral perspective 2 surfaces can be seen. One surface is smooth and more convex, whereas the other is less convex and partially covered by a network from which many fungiform tubercles arise. The micropyle is situated on the smooth surface of the pointed anterior tip and is surrounded by an irregular row of tubercles, some of which are leaf shaped. No structures possibly involved in adhesion to surfaces are visible. When hatching, the egg splits dorsoventrally approximately two-thirds of the length from the anterior end. The tubercles appear to be water repellent, and the more convex/smoother surface is downturned, and this position on water was confirmed by direct observation. The eggs float free on the water surface.

The evolution of elaborate male ornaments via sexual selection is well-understood while the selective pressures acting on female ornaments remains unresolved. Female ornaments in species with strong sexual selection on the male homologue of the ornament were originally thought to result from an intersexual genetic correlation. My thesis explores the evolution of ornaments in females due to direct selection by developing theoretical models and examining the biology of a neotropical mosquito (Sabethes cyaneus) with sexually homologous ornaments coinciding with male-male competition. I began by exploring the morphology of the ornaments in both sexes of S. cyaneus. Sexual dimorphism in the size and shape of the ornaments was slight and both male and female ornaments showed classic hallmarks of sexually selected traits. I then tested for direct selection on S. cyaneus male and female ornaments via mutual mate choice. I found evidence of male, but surprisingly not female, preferences for ornaments. I then further considered the evolution of male mate choice in polygynous species. First, I investigated whether male investment in courtship by S. cyaneus may result in a lower operational sex ratio and thereby reduce the costs associated with male mate choice. Male courtship did pose a significant longevity cost to male S. cyaneus. Second, I explored the possibility that a female preference for male courtship effort may contribute to the benefits of male mate choice in a series of population genetic models. The spread of a male preference gene can be driven by female preferences for male courtship when males court preferred females more. Finally, I found that female S. cyaneus are not benefitting from signalling to increase their mating rate as they are monandrous. My thesis therefore challenges standing sexual selection theory and suggests that sexual selection on females may be more widespread than previously thought. Unlike any other mosquito reported, Sabethes cyaneus(Fabricius) displays an elaborate courtship before and during copulation. A male approaches a female suspended from a horizontal stick, suspends himself in front of her as he grasps her folded wings, and proceeds with a series of discrete stereotyped behaviors that involve proboscis vibration and movement of iridescent blue paddles on his midlegs. The sequence of these behaviors is as follows: freeleg waving, swinging, copulation attempt, superficial coupling, waving, genital shift, waggling, and release. Insemination occurs after genital shift. The only overt reciprocation by the female is abdomen lowering during the male's swinging. Courtship is often unsuccessful, and males are usually rejected during freeleg waving. The relation between male performance and mating success remains obscure.

In 2 forested areas near Belem (Para State, Brazil), 2 Haemagogus and 6 Sabethes species were marked released and recaptured in May 1989 and in April 1993. The recapture rates were high, 4.9 and 13.1% for Haemagogus and Sabethes spp., respectively. For Haemagogus janthinomys Dyar, females were recaptured until 27 d after release. The duration of the gonotrophic cycle was between 5.0 and 9.5 d and the survival rate was 0.90-0.92. Haemagogus leucocelaenus (Dyar & Shannon) was recaptured once, 21 d after release. Twelve Sabethes chloropterus (Von Humboldt) were recaptured, with a peak at 15-18 d; 1 female was recaptured at 44 d, indicating extended survival. Seven Sabethes amazonicus Gordon & Evans and 7 Sabethes cyaneus (F.) were recaptured, mostly at 14-39 d. These results indicate that Haemagogus and Sabethes spp. have a gonotrophic cycle in nature longer than inferred from laboratory studies, and that cycle length varies seasonally. The capacity of these species to sustain epizootics or epidemics of arboviruses may depend on local weather, with risk greatest at the end of the rainy season.
