Osteoporosis isa systemic disease which is common in society, characterized by bone micro-architecturedeterioration and low bone mass. The aim of our study was to determine whether paraoxonase1 (PON1) gene correlates with584A>G (Gln192Arg) and 172T>A (Leu55Met) polymorphisms withpostmenopausal osteoporosis. A total of 143 postmenopausal osteoporosis and 102healthy women who were followed up in Eskişehir Osmangazi University Faculty ofMedicine, Department of Physical Medicine and Rehabilitation were included inthe study. The study was planned retrospectively. Genotypes were identified onthe qPCR (The Applied Biosystems® StepOne ™) using the allele-specific DNAprimer and fluorescent-specific DNA probe. There was no statisticallysignificant difference between the groups in the genotype and allelefrequencies of PON1 584A>Gpolymorphism (p=0.362 and p= 0.318). Genotipler alel-spesifikDNA primeri ve floresan özellikli DNA probu kullanılarak qPCR cihazında (TheApplied Biosystems® StepOne™) belirlendi.PON1 584A>G polimorfizminin genotip ve alelfrekanslarında gruplar arasında istatistiksel olarak anlamlı bir farkbulunmamıştır (p=0.362 ve p=0.318). PON1 172T>A polimorfizminin genotip ve alel frekanslarıkarşılaştırıldığında ise, sağlıklı bireylere göre postmenopozal osteoporozuolanlarda T alelinin istatistikselolarak anlamlı derecede yüksek bulunduğu (sırasıyla pA(Leu55Met) gen polimorfizmi ile postmenopozal osteoporoz arasındailişki olduğu gösterildi.

Amaç: Akciğer kanseri, akciğer doku hücrelerinin kontrolsüz çoğalmasıyla oluşan ölümcül bir hastalıktır. Çevresel faktörlerle birlikte genetik ve epigenetik değişikliklerin kanser gelişimine neden olduğu bilinmektedir. DNA metilasyonu en önemli epigenetik değişimlerden biri olup, DNA metiltransferaz (DNMT) enzimleri tarafından katalizlenir ve metile-CpG'ye bağlanan proteinler (MBD1, MBD2, MeCP2 vb) yardımıyla gerçekleşir. Çalışmamızda metilasyon ilişkili proteinlerin plazma seviyeleri ile akciğer kanseri riski arasındaki ilişkinin araştırılması amaçlanmıştır. Yöntemler: Araştırmamızda DNMT1, DNMT2 (TRDMT1), DNMT3A, DNMT3B, MeCP2, MBD1 ve MBD2 seviyeleri, 90 akciğer kanseri hastası (küçük hücreli dışı: 78, küçük hücreli:12) ve 90 kontrol bireyden elde edilen plazma örneklerinde Sandwich-ELISA yöntemi ile ölçüldü. Sonuçlar uygun istatistiksel yöntemler ile değerlendirildi. Bulgular: Kontrol ve akciğer kanserli hastaların DNMT3a, MBD2 ve MeCP2 seviyeleri arasında fark bulunmamıştır (p>0.05). DNMT1, TRDMT1, DNMT3b ve MBD1 seviyeleri ise kontrol ile karşılaştırıldığında akciğer kanserli bireylerde anlamlı oranda yüksek bulunmuştur (p, Objective: Lung cancer is a fatal disease caused by uncontrolled growth of lung tissue cells. It has been known that environmental factors, genetic and epigenetic changes are lead to tumorigenesis. DNA methylation is one of the important epigenetic changes, catalyzed by DNA methyltransferases (DNMTs) and performed with methyl-CpG binding proteins (MBD1, MBD2, MeCP2 etc.). In this study we aimed to determine the association between plasma levels of methylation associated proteins and lung cancer risk. Methods: The plasma levels of DNMT1, DNMT2 (TRDMT1), DNMT3A, DNMT3B, MeCP2, MBD1 and MBD2 measured with Sandwich-ELISA method in 90 of lung cancer patients (non-small cell: 78, small cell: 12) and 90 of healthy controls. Statistical analysis was performed with appropriate statistical methods. Results: When comparing the DNMT3a, MBD2 and MeCP2 levels, no significant differences were found between lung cancer patients and controls (p>0.05). MicroRNAs (MiRNAs) are highly conserved, non-protein-coding, approximately 18-24 nucleotides, small RNA mol­ecules. These non-protein-coding RNAs bind target mR­NAs which are complementary with their squences and post-transcriptionally regulate gene expression through translation repression or mRNA degradation. Using this pathway, miRNAs play important roles in homeostatic process including cell proliferation, differentiation and cell death. Recent studies based on gene expression have re­vealed that miRNA expressions are deregulated in cancer cells which have uncontrolled grow. According to charac­ter of target mRNA, miRNAs act like tumor supressor or oncogene in cancer initiation and progression. In this re­view, miRNA biogenesis and their roles in cancer progres­sion are described and studies setting out the relationship between miRNAs and cancer are reviewed., MikroRNA(miRNA)\'lar, yüksek seviyede korunan DNA bölgelerinden kodlanan fakat proteine translasyonu ger­çekleşmeyen, yaklaşık 18-24 nükleotid uzunluğunda, küçük RNA molekülleridir. Bu protein kodlamayan RNA molekülleri kendi nükleotid dizilerinin tamamlayıcısı olan hedef mRNA\'lara bağlanıp translasyonel baskılama veya mRNA yıkımı ile transkripsiyon sonrası gen ekspresyo­nunun düzenlenmesini gerçekleştirirler. miRNA\'lar bu yo­lağı kullanarak hücre proliferasyonu, hücre farklılaşması ve hücre ölümü gibi homeostatik süreçlerde önemli roller oynamaktadır. Yakın zamanda yapılan gen ekspresyon çalışmaları, kontrolsüz hücre bölünmesinin gerçekleştiği kanser hücrelerinde değişikliğe uğramış miRNA ekspres­yonlarını gözler önüne sermiştir. Kanserin başlamasında ve ilerlemesinde, miRNA\'lar hedefledikleri genin karakte­rine göre tümör süpresörler veya onkogenler gibi fonksi­yon göstermektedirler. Bu derlemede miRNA oluşumu ve miRNA\'ların kanser gelişimindeki rolleri anlatılmış olup, miRNA\'ların kanser ile olan ilişkisini ortaya koyan çalış­malar gözden geçirilmiştir.

Sarcoidosis is a complex, multifactorial immune disorder with unknown etiology. A single nucleotide polymorphism ( G->A, rs2076530) in the butyrophilin-like 2 (BTNL2) gene results in a truncating protein formation. It has been previously reported that this variation may be a risk factor for sarcoidosis in certain ethnic groups. This study was conducted to determine whether there is any genetic predisposition for the BTNL2 rs2076530 polymorphism in the 329-bp region in Turkish patients with sarcoidosis. Materials and methods: DNA samples were obtained from volunteers including 53 Turkish patients with sarcoidosis and 52 healthy controls. Analysis of the 329-bp region was carried out by polymerase chain reaction and sequencing of genomic DNA. Results: We did not find any genetic variation except the rs2076530 polymorphism in the 329-bp region. The AA genotype was associated with an increased risk of sarcoidosis in a recessive model [P = 0.027, OR 2.56 (95% CI 1.02-6.49)], but it did not include a risk for sarcoidosis in a dominant model (P = 0.885). Conclusion: Our results emphasize the recessive characteristic of the rs2076530 polymorphism in Turkish patients with sarcoidosis. The lack of any genetic variation except rs2076530 in the 329-bp region is another significant finding for Turkish patients.

Klopidogrel direnç mekanizmasını araştırdığımız çalışmamızda özellikle CYP2C19, CYP3A4, CYP2B6, ABCB1, ITGB3 ve PON1 genlerindeki varyasyonların bu mekanizma ile olan ilişkisini belirlemeyi amaçladık. Bu amacımızı gerçekleştirirken klopidogrele dirençli bireylerin ve klopidogrel aktivitesi ile ilişkili varyasyonların sıklığını Türk toplumunda ortaya çıkarmak ulaşmak istediğimiz hedeflerimiz arasındaydı.Çalışmamıza elektif veya acil stent implantasyonlu perkütan koroner girişim yapılan 223 hasta dahil edildi. En az 7 gün süreyle standart doz (75 mg/gün) klopidogrel kullanan hastalardan alınan kan örneklerinde VerifyNow P2Y12 kiti ile trombosit reaktivitesi (PRU) ve % inhibisyon ölçüldü. Farklı tüpe alınan kan örneklerinden izole edilen DNA'ların genetik varyasyon analizleri için Sequenom MassARRAY sistemi ile genotipleme protokolleri yürütüldü. Hastaların genetik varyasyon genotipleri, ilaç kullanım durumu, demografik ve klinik özellikleri trombosit reaktivitesine göre istatistiksel testler ile analiz edildi. PRU değeri >208 olan hastalar klopidogrel tedavisine cevap vermeyen (klopidogrele dirençli) bireyler olarak gruplandırıldığında 69 (%30,9) hastanın klopidogrele dirençli, 154 (%69,1) hastanın ise klopidogrele dirençsiz olduğu belirlendi. Objectives: in this study, the antioxidant activity of tannic acid was investigated experimentally with respect to its protective effects against overdosed MSG-induced oxidative stress in rats. the study took place in Turkey in August 2013. Methods: Four groups (n = 7) of three-to four-month-old Sprague-Dawley female rats were used in this study. the first group was the control, who were administered saline. the second group received tannic acid (50 mg/kg, 3 days) intraperitoneally (i.p.). the third group received MSG (2 g/kg, 7 days) i.p., and the fourth group received both tannic acid (50 mg/kg, 3 days, pretreatment) and MSG (2 g/kg, 7 days) i.p. the animals were euthanized ten days later. Blood was collected for determining the hematological values and blood glucose levels. Superoxide dismutase (SOD) andmalondialdehyde(MDA) levels were determined in the brain, liver, and kidney homogenates, and in the erythrocyte hemolysate. Histopathological examination of the brain, liver, and kidneys was conducted through hematoxylin-eosin staining. Results: the data showed that the tannic acid treatment statistically decreased the MDA levels in the brain tissues of the group administered MSG and tannic acid (P < 0.001) when compared to the corresponding values of the control group. the SOD activities in the blood hemolysates of the MSGandtannic acid group increased when compared to the corresponding values for the MSGgroup (P < 0.01). Additionally, we found that pretreatment with tannic acid reduced blood glucose levels in comparison to the levels of the MSG group (P = 0.029). the results of our study show that tannic acid pretreatment in adult rats decreased blood glucose levels and oxidative stress. Conclusions: in the literature, it was observed that short-term MSG exposure does not cause significant histological changes in the kidneys, liver, or brain cortex. These findings should be re-evaluated in additional long-term studies.
