Postmenopozal Osteoporozda Paraoksonaz 1 L55M ve Q192R Polimorfizminin İlişkisinin Değerlendirilmesi

Osteoporosis isa systemic disease which is common in society, characterized by bone micro-architecturedeterioration and low bone mass. The aim of our study was to determine whether paraoxonase1 (PON1) gene correlates with584A>G (Gln192Arg) and 172T>A (Leu55Met) polymorphisms withpostmenopausal osteoporosis. A total of 143 postmenopausal osteoporosis and 102healthy women who were followed up in Eskişehir Osmangazi University Faculty ofMedicine, Department of Physical Medicine and Rehabilitation were included inthe study. The study was planned retrospectively. Genotypes were identified onthe qPCR (The Applied Biosystems® StepOne ™) using the allele-specific DNAprimer and fluorescent-specific DNA probe. There was no statisticallysignificant difference between the groups in the genotype and allelefrequencies of PON1 584A>Gpolymorphism (p=0.362 and p= 0.318). When the genotype and allele frequenciesof PON1 172T>A polymorphism werecompared, it was observed that T allele was significantly higher inpostmenopausal osteoporosis than healthy subjects (p A (Leu55Met) genepolymorphism was associated with postmenopausal osteoporosis in our study.
Osteoporoz,toplumda yaygın görünen, kemik mikro mimarisinde bozulma ve düşük kemik kütlesiile karakterize sistemik bir hastalıktır. Çalışmamızınamacı paraoksonaz (PON1) geninin584A>G(Gln192Arg) ve 172T>A(Leu55Met) polimorfizmleri ile postmenopozalosteoporoz ile ilişkisinin olup olmadığını saptamaktır. Eskişehir OsmangaziÜniversitesi Tıp Fakültesi Fiziksel Tıp ve Rehabilitasyon Anabilim DalıKliniği’nde takipli 143 postmenopozal osteoporozu olan ve 102 sağlıklı kadınçalışmaya dahil edildi. Çalışma retrospektif olarak planlandı. Genotipler alel-spesifikDNA primeri ve floresan özellikli DNA probu kullanılarak qPCR cihazında (TheApplied Biosystems® StepOne™) belirlendi.PON1 584A>G polimorfizminin genotip ve alelfrekanslarında gruplar arasında istatistiksel olarak anlamlı bir farkbulunmamıştır (p=0.362 ve p=0.318). PON1 172T>A polimorfizminin genotip ve alel frekanslarıkarşılaştırıldığında ise, sağlıklı bireylere göre postmenopozal osteoporozuolanlarda T alelinin istatistikselolarak anlamlı derecede yüksek bulunduğu (sırasıyla pA(Leu55Met) gen polimorfizmi ile postmenopozal osteoporoz arasındailişki olduğu gösterildi.
