1. The challenge of adults with phenylketonuria who have been lost to care; a single center's attempt to reach those diagnosed with PKU over 60 years of newborn screening

Abstract

Background: Those diagnosed with PKU in the early years of newborn screening (NBS) were often discharged from clinic in childhood. Long-term lost to clinic patients may be impacted by untreated PKU and uninformed about current recommendations. We aimed to contact adults away from clinic for 5–50+ years, share current recommendations, offer clinical care, and elicit factors underlying not returning to clinic. Methods: Former patients were identified and offered a virtual meeting with a physician and dietitian for structured interview and education about current guidelines and treatments. Results: We identified 53 eligible patients who had PKU and had not returned to clinic in ≥5 years. Of those 53, 27 were successfully contacted, 16 completed the educational intervention, and 5/16 returned to clinic. Reasons for having been away from clinic included discharge from clinic in childhood and inadequate insurance coverage. Experiences varied and some denied negative impacts after diet discontinuation. Individuals expressed a desire for convenient treatments that aligned with overall health goals. Most participants who completed the educational intervention expressed interest in returning to clinic; however, most did not return within the timeframe of the project. All 27 individuals successfully contacted agreed to be re-contacted with future updates or research opportunities. Discussion: We successfully contacted half of individuals identified as having been lost to clinic follow-up long-term. Limitations included inability to make initial contact, and unwillingness to re-engage by some we reached. Those who agreed to participation desired ongoing PKU clinic and community connection. This experience will inform our process to engage current patients and re-engage those currently lost to care.

Abstract

Abstract Desertification is a major threat to biodiversity in arid areas of the world, partly because many organisms in these regions already exist at or near the limits of their movement and physiology. Here, we used molecular data to investigate patterns of persistence and dispersal in an ecologically and economically important carpenter bee (Xylocopa grisescens Lepeletier) found throughout the semiarid Caatinga region of Brazil. We used a genome‐wide approach (double digest restriction‐site associated DNA, ddRAD) to gather genetic data from bees sampled from eight sites within a semiarid region subject to desertification in Northeastern Brazil. Across all populations, we observed a consistent heterozygosity and effective population size deficit along with low genetic differentiation. We did not find strong evidence of dispersal limitations caused by desertification in this study system despite data collection from sites up to 300 km distant. Thus, our data suggest that human‐mediated changes in the Caatinga, such as habitat loss, have impacted the population genetic patterns of X. grisescens. However, these impacts have also been softened by the species' biological characteristics, such as its relatively high capacity for movement. This study provides insights into how habitat changes might impact the long‐term survival of large solitary bees.

Abstract

Resume Introduction Les concentrations seriques de proteine C reactive (CRP) en deca d’un seuil predefini entre le 3e et le 5e jour postoperatoire sont un marqueur fiable de l’absence de complication infectieuse apres chirurgie colorectale. Cependant, la strategie a mettre en place en cas de CRP elevee n’est pas codifiee. Le but de ce travail etait d’analyser l’utilite du scanner dans cette situation au sein d’une cohorte prospective de chirurgie colorectale reglee. Methodes Entre novembre 2011 et avril 2015, les patients ayant une CRP > 125 mg/L au 4e jour postoperatoire d’une resection colorectale reglee dans l’un des 2 centres participants ont ete inclus prospectivement dans une base. Toutes les complications survenues pendant les 30 j suivant l’intervention, leur mode de diagnostic, prise en charge et evolution ont ete recueillies. La realisation d’un scanner entre le 4e et le 6e jour guide par la seule elevation de la CRP, en l’absence de tout autre signe clinique, a ete etudiee pour evaluer ainsi sa rentabilite diagnostique. Des analyses uni- et multivariees ont ete realisees pour identifier les facteurs de risque d’infection postoperatoire. Resultats Au total, 174 patients ont ete inclus : 56 (32,1 %) ont eu un scanner entre le 4e et le 6e jour, qui a permis de detecter une complication postoperatoire dans 55,4 % des cas. Les patients n’ayant pas eu de scanner avaient des valeurs de CRP plus basses (163 vs 185,5, p = 0,02). Parmi les 118 patients n’ayant pas eu de scanner, 50,8 % ont presente une complication infectieuse a terme. La sensibilite du scanner guide par la CRP etait de 76,7 % et la valeur predictive negative de 78,8 %. Conclusion En presence d’une CRP elevee au 4e jour postoperatoire un scanner abdomino-pelvien doit etre realise. Un resultat normal n’elimine pas formellement l’existence d’une complication intra-abdominale. En cas de persistance de l’inflammation dans les deux jours suivants, une reprise chirurgicale systematique doit etre comparee a un nouveau scanner dans le cadre d’un protocole.

Abstract

IntroductionBee conservation in the US is currently hindered by challenges associated with assessing the status and trends of a diverse group of >3000 species, many of which are rare, endemic to small areas, and/or exhibit high inter-annual variationin population size. Fundamental information about the distribution of most species across space and time, thus, is lacking yet urgently needed to assess population status, guide conservation plans, and prioritize actions among species and geographies.MethodsUsing wild bee data from two public data repositories representing the contiguous US, we evaluated the availability and sufficiency of data for use in species assessments of wild bees. We also examined the number of bee species recorded in each US state and the proportion of species with recent records (2012–2021).ResultsAlthough efforts to monitor bees continue to grow, there remains a massive paucity of data. Exceedingly few records (0.04%)reported both sampling protocol and effort, greatly limiting the usefulness of the data. Few species or locations have adequate publicly available data to support analyses of population status or trends, and fewer than half of species have sufficient data to delineate geographic range. Despite an exponential increase in data submissions since the 2000s, only 47% of species were reported within the last decade, which may be driven by how data are collected, reported, and shared, or may reflect troubling patterns of local or large-scale declines and extirpations.DiscussionBased on our analysis, we provide recommendations to improve the quality and quantity of data that can be used to detect, understand, and respond to changes in wild bee populations.

Abstract

Resume Introduction La perfusion sur foie isole de la circulation est une methode de delivrance de la chimiotherapie permettant d’augmenter fortement les doses en diminuant la toxicite extrahepatique. Elle s’adresse a des patients porteurs de tumeurs hepatiques non resecables chirurgicalement. Principes Une exclusion vasculaire du foie est realisee apres verification de la bonne tolerance hemodynamique et de l’eventuel recours a un shunt cavo-cave et/ou porto-systemique. L’entree de perfusion peut etre arterielle et/ou portale, et la sortie portale ou cave. Le circuit de perfusion peut etre ouvert ou ferme, avec recours a une pompe de circulation avec ou sans oxygenation. Les differentes chimiotherapies utilisees sont le melphalan (avec ou sans TNF-alpha), l’oxaliplatine, le cisplatine et la mitomycine, a fortes doses, parfois associees a une hyperthermie moderee. La duree de perfusion oscille entre 30 et 90 minutes suivant les protocoles. Une technique percutanee avec exclusion incomplete est possible. Resultats Les series les plus importantes montrent un taux de reponse (partielle ou stabilisation) de 60 a 80 %, avec environ 5 % de reponses completes morphologiques. La morbidite et la mortalite sont autour de 40 % et 5 %, respectivement, incluant la morbidite liee a la procedure de perfusion et la morbidite specifique de la chimiotherapie. Conclusion La perfusion de chimiotherapie sur foie isole est une therapeutique recente, encore en voie de developpement, proposee aux patients atteints de tumeurs primitives ou secondaires du foie non resecables dans le cadre d’essais cliniques. Ses resultats semblent prometteurs, mais associes a une morbidite non negligeable.

Abstract

Summary Introduction Isolated hepatic perfusion allows the delivery of high dose chemotherapy while decreasing extra-hepatic toxicity, and is used mainly for patients with surgically unresectable liver tumors. Principles Vascular exclusion of the liver is performed after obtaining satisfactory hemodynamic tolerance, occasionally after cavocaval shunt and/or porto-systemic shunt. Perfusion entry can be arterial and/or portal while the exit is portal or caval. The perfusion circuit can be open or closed, using a circulation pump with or without oxygenation. The chemotherapy regimens used are high dose melphalan (with or without TNF-alpha), oxaliplatin, cisplatin and mitomycin, sometimes associated with moderate hyperthermia. The duration of perfusion ranges between 30 and 90 minutes according to the different protocols used. A percutaneous technique with incomplete liver vascular exclusion is also possible. Results The larger series in the literature show a response rate (partial or complete stabilization) between 60 and 80%, with approximately 5% complete morphologic responses. Morbidity and mortality are 40 and 5%, respectively, including specific morbidity related to the perfusion procedure as well as to chemotherapy. Conclusion Chemotherapy delivered through isolated hepatic perfusion is a new therapeutic alternative, still under development, and can be proposed to patients with surgically unresectable primary or secondary liver tumors within clinical trials. These results seem to be promising, but are still associated with a non-negligible morbidity rate.

Abstract

In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

Abstract

JET, the world’s largest operating tokamak with unique Be/W wall and tritium handling capability, completed a Deuterium-Tritium (D-T) campaign in 2021 (Maggi et al 29th Fusion Energy Conf. ) following a decade of preparatory experiments, dedicated enhancements, technical rehearsals and training (Horton et al 2016 Fusion Eng. Des. 109–111 925). Operation with tritium raises significant technical, safety and scientific challenges not encountered in standard protium or deuterium operation. This contribution describes the tritium operational requirements, pulses and technical preparations, new operating procedures, lessons learned and details on the achieved operational availability and performance. The preparation and execution of the recent JET tritium experiments benefitted from the previous experience in 1991 (Preliminary Tritium Experiment), 1997 (DTE1 campaign) and 2003 (Trace Tritium Campaigns) and consisted of the following five phases: technical rehearsals and scenario preparation, tritium commissioning, 100% tritium campaign, D-T campaign (DTE2), tritium clean-up. Following the clean-up JET resumed normal operation and is currently undertaking a further D-T campaign (DTE3).

Abstract

Summary Introduction Serum concentration of C-reactive protein (CRP) that exceeds a pre-defined threshold between the 3rd and 5th postoperative day is a reliable marker of infectious complications after colorectal surgery. However, the optimal strategy to follow when a high CRP is found has not been defined. The aim of this study was to analyze the usefulness of computed tomography (CT) scan in this situation in a prospective cohort of patients following colorectal surgery. Methods Between November 2011 and April 2015, patients at two surgical centers who had undergone elective colorectal resection with anastomosis and who had a CRP > 12.5 mg/dL on the 4th postoperative day (POD) were prospectively included in a database. Data were collected concerning all complications occurring during the 30 days following surgery, method of diagnosis, management and clinical course. The decision to perform a CT scan between POD 4 and POD 6 day was guided only by the elevation of CRP in the absence of any other clinical signs; results were analyzed to evaluate the diagnostic value of elevated CRP. Uni- and multivariable analyses were performed to identify risk factors for postoperative infection. Results The study included a total of 174 patients: 56 (32.1%) had a CT between POD 4 and 6, and this helped detect a postoperative complication in 55.4% of cases. Patients who did not undergo CT had lower CRP values (16.3 vs. 18.5, P = 0.02). Among the 118 patients who did not undergo CT, 50.8% eventually developed an infectious complication. The sensitivity of CRP-guided CT was 76.7% with a negative predictive value of 78.8%. Conclusion If an elevated CRP is found on POD 4, an abdominopelvic CT should be performed. A normal result does not formally eliminate the existence of intra-abdominal complication. A study protocol should be set up to evaluate whether systematic revisional surgery or repeat CT scan is the appropriate management if CRP in the next two days reveals persistent inflammation.

Abstract

Off-line land surface modeling simulations require accurate meteorological forcing with consistent spatial and temporal resolutions. Although reanalysis products present an attractive data source for these types of applications, bias to many of the reanalysis fields limits their use for hydrological modeling. In this study, we develop a global 0.5° forcing data sets for the time period 1979–1993 on a 6-hourly time step through application of a bias correction scheme to reanalysis products. We then use this forcing data to drive a land surface model for global estimation of soil moisture and other hydrological states and fluxes. The simulated soil moisture estimates are compared to in situ measurements, satellite observations and to a modeled data set of root zone soil moisture produced within a separate land surface model, using a different data set of hydrometeorological forcing. In general, there is good agreement between anomalies in modeled and observed (in situ) root zone soil moisture. Similarly, for the surface soil wetness state, modeled estimates and satellite observations are in general statistical agreement; however, correlations decline with increasing vegetation amount. Comparisons to a modeled data set of soil moisture also demonstrates that both simulations present estimates that are well correlated for the soil moisture in the anomaly time series, despite being derived from different land surface models, using different data sources for meteorological forcing, and with different specifications of the land surfaces properties. Abstract

The aim of the study was to investigate the influence of growth hormone (GH) on Vitamin D3 metabolism and the subsequent effects on calcium (Ca) homeostasis and skeletal growth in growing dogs. A group of Miniature Poodles received supraphysiological doses of GH (GH group; n = 6; 0.5 IU GH per kg body per day) from 12 to 21 weeks of age and was compared with a control placebo-treated group (n = 8). Biologic activity of GH in the GH compared to the control group was indicated by (a) the 2.5- to 3.5-fold increase in the plasma concentrations of insulin-like growth factor I (IGF-I), (b) the increased production of 1,25-dihydroxycholecalciferol as indicated by the significantly increased plasma 1,25-dihydroxycholecalciferol concentrations and the 12.9-fold increase in renal 1alpha-hydroxylase gene expression, and (c) the inhibited production of 24,25-dihydroxycholecalciferol as indicated by the significantly lower plasma 24,25-dihydroxycholecalciferol concentrations and the similar levels of renal 24-hydroxylase gene expression. Despite the distinct effects on Vitamin D(3) metabolism in the GH group, there were only moderate effects on the intestine, i.e. at 20 weeks of age there was a significant increase of 14.4 and 5.6% in fractional absorption of Ca and phosphate (Pi), respectively, compared to the control group. GH administration resulted in significantly elevated glomerular filtration rate, with no differences in Pi urine excretion as a result of a concomitant increase in the tubular reabsorption of Pi. GH had only limited disturbing effects on endochondral ossification as indicated by the maintenance of the regularity of the growth plates. However, GH had specific anabolic effects on bone formation without concomitant effect on bone resorption that may result in disorders of skeletal remodeling and manifestation of enostosis.

A degenerate ground state spin pattern is found for the type II antiferromagnet EuTe in the bulk limit. The model system is EuTe with ferromagnetic nearest neighbor (NN,J1=0.06K) and antiferromagnetic next nearest neighbor (NNN,J2=−0.16K) exchange interaction. For the non-trivial ground state the spins within a single (1 1 1) plane are no longer all parallel. Instead a quarter of the spins are parallel to the NN spins in the adjacent layer. This state becomes metastable if dipolar interaction is taken into account, or for low-dimensional structures of few (1 1 1)-monolayer systems of EuTe.

The magnetic properties depending on the temperature and magnetic field of few (1 1 1) monolayers of antiferromagnetic EuTe are analyzed by means of Monte Carlo simulations. We apply a classical Heisenberg as well as a XY model Hamiltonian with ferromagnetic nearest neighbor ( J 1 =0.06 K) and antiferromagnetic next-nearest-neighbor ( J 2 =−0.16 K) exchange constants. Furthermore, dipolar interaction is included which links the spin space to real space and therefore introduces the easy plane anisotropy. Structures consisting of incomplete sets of monolayers, e.g. two perfect monolayers and a finite island as the third monolayer, are investigated. We analyze the atomic spin patterns and compare these results with total magnetization and neutron diffraction experiments. It is emphasized that EuTe in the reduced dimensional limit of few odd numbers of monolayers exhibits a field-induced uniaxial anisotropy, even in the frame of isotropic Heisenberg exchange interaction. A concomitant field-parallel ferrimagnetic order is sustained up to a field H ap .

Abstract

The magnetic properties of short-period superlattices consisting of zincblende MnTe (type-III antiferromagnet with Neel temperature of 67 K in bulk-like films) and CdTe are investigated by coherent Raman scattering (CRS) experiments as well as SQUID-magnetometry as a function of temperature (1.6–150 K) and magnetic field (0–7 T). In CRS the magnetization is probed by observing the paramagnetic (PMR) and antiferromagnetic resonances (AFMR) of localized d-electrons of Mn-ions and the spin-splitting of delocalized band carriers, respectively. Resonant CRS is particularly sensitive to Mn-ions located in regions with high expectation propabilities of electrons, i.e. near the interfaces between the CdTe quantum wells and MnTe barriers. Simultaneous observation of PMR and AFMR demonstrates that besides the antiferromagnetically coupled, also uncoupled Mn-spins exist in the interface regions. Their number is determined quantitatively by magnetization experiments. The observed free carrier spin-splitting energy is directly related to the local magnetization of magnetic ions within regions of high expectation probabilities of electrons. The data are analyzed within the framework of a mean-field analysis (MFA) which incorporates the electron–Mn-ion exchange as well as the Mn–Mn interion exchange interaction. The latter is represented by the individual nearest and next nearest neighbor exchange interactions, the coordination number of spins and their respective orientation. The phase transition temperature is substantially decreased due to the reduced dimensionality (less nearest neighbors) and due to strain which modifies the exchange integrals and bond angles.

Neutron diffraction and magnetization studies of short-period antiferromagnetic (AF) semiconducting (1 1 1) EuTe/PbTe superlattices are presented. Due to the symmetry-breaking lattice mismatch strain, only a single AF configuration forms in the EuTe layers, instead of four allowed by symmetry in bulk EuTe. The strain and the finite layer thickness also lead to a strong shift in the Neel temperature. Neutron-diffraction spectra exhibit pronounced patterns of satellites, indicating spin correlations between successive EuTe layers separated by PbTe layers, and the transfer of magnetic interactions across the diamagnetic spaces. Experiments on samples with doped PbTe layers and in external magnetic fields rule out that the coupling is caused by any of the mechanisms known to occur in metallic superlattices. Hence, our data strongly suggest the existence of a new interlayer coupling mechanism.

Abstract

Systems for animal disease mitigation involve both surveillance activities and interventions to control the disease. They are complex organizations that are described by partial or imprecise data, making it difficult to evaluate them or make decisions to improve them. A mathematical method, called loop analysis, can be used to model qualitatively the structure and the behavior of dynamic systems; it relies on the study of the sign of the interactions between the components of the system. This method, currently widely used by ecologists, has to our knowledge never been applied in the context of animal disease mitigation systems. The objective of the study was to assess whether loop analysis could be applied to this new context. We first developed a generic model that restricted the applicability of the method to event-based surveillance systems of endemic diseases, excluding the emergence and eradication phases. Then we chose the mitigation system of highly pathogenic avian influenza (HPAI) H5N1 in Cambodia as an example of such system to study the application of loop analysis to a real disease mitigation system., Breaking down the generic model, we constructed a 6-variables model to represent the HPAI H5N1 mitigation system in Cambodia. This construction work improved our understanding of this system, highlighting the link between surveillance and control which is unclear in traditional representations of this system. Then we analyzed the effect of the perturbations to this HPAI H5N1 mitigation system that we interpreted in terms of investment in a given compartment. This study suggested that increasing intervention at a local level can optimize the system's efficiency. Indeed, this perturbation both decreases surveillance and intervention costs and reduces the disease's occurrence., Loop analysis can be applied to disease mitigation systems. Its main strength is that it is easy to design, focusing on the signs of the interactions. It is a simple and flexible tool that could be used as a precursor to large-scale quantitative studies, to support reflection about disease mitigation systems structure and functioning.

Abstract

In the scope of the German D 2 mission, samples of a nickel super alloy CMSX 6 were remelted and solidified. The sintered sample material contained a strengthening dispersion of Al 2 O 3 particles with a grain size of approximately 50 nm. Reference samples consisted of the cast material without particles. The samples shape was similar to small turbine blades. In order to preserve their geometry the samples were provided with a thin ceramic mold skin. The main objectives of the experiments were the shape preservation by means of the skin and informations on the particles behaviour. The local and the chronological temperature profile was evaluated by computer simulation additionally to the ground lab runs. Both results showed a good consistency. The results of the flight experiments were influenced by the formation of gas cavities in the samples processed from the powdery condition. Corresponding to the calculated results the melting boundary was detected in all samples exactly in the predicted position. The particle showed a distinct tendency to agglomeration. The ceramic mold skin fulfilled its task, e.g. the reliable enclosure of the melt, in spite of the considerably bulged samples.

Abstract

The computer-assisted simulation of solidification processes influenced by gravity has gained increased importance during the previous years regarding ground-based as well as microgravity research. Depending on the specific needs of the investigator, the simulation model ideally covers a broad spectrum of applications. These primarily include the optimization of furnace design in interaction with selected process parameters to meet the desired crystallization conditions. Different approaches concerning the complexity of the simulation models as well as their dedicated applications will be discussed in this paper. Special emphasis will be put on the potential of software tools to increase the scientific quality and cost-efficiency of microgravity experimentation. The results gained so far in the context of TEXUS, FSLP, D-1 and D-2 (preparatory program) experiments, highlighting their simulation-supported preparation and evaluation will be discussed. An outlook will then be given on the possibilities to enhance the efficiency of pre-industrial research in the Columbus era through the incorporation of suitable simulation methods and tools.

We investigate the crossover from antiferromagnetic to ferromagnetic behaviour in semimagnetic semiconductor structures. Systems with two dimensional magnetic layers of (1 0 0)-MnTe equispatially inserted four times in a CdTe quantum well, embedded between Cd 0.75 Mg 0.25 Te barriers are studied in the ideal paramagnetic limit. The interaction between localized magnetic spins and quantum confined electronic states is shown to give rise to the formation of free magnetic polarons.

Abstract Background The developmental fates of offspring have the potential to be influenced by the identity of their care-givers and by the nature of the care that they receive. In animals that exhibit both parental and alloparental care, such as the annually eusocial insects, the influence of care-giver identity can be directly assessed to yield mechanistic and evolutionary insights into the origins and elaboration of brood care. Here, we performed a comparative investigation of maternal and worker brood care in bumble bees, a pollinator group where mothers (queens) rear the first offspring in the nest, and then daughters (workers) assume this role upon their emergence. Specifically, we compared the effects of queen and worker brood care on offspring development and also offspring performance, for a set of traits related to sensory biology, learning, and stress resistance. Results We found that queen-reared workers were smaller-bodied than worker-reared offspring, suggesting that bumble bee queens influence body size determination in their offspring. We also found that queen-reared workers were more resistant to starvation, which might be beneficial for early nesting success. These maternal influences could not be explained by feeding rate, given that we detected a similar offspring feeding frequency in both queens and workers. Conclusion Bumble bee queens have a unique influence on the development of the first offspring in the nest, which they rear, relative to worker-reared workers. We propose that bumble bee brood care has been shaped by a suite of evolutionary and ecological factors, which might include a maternal influence on traits that promote survival of incipient colonies.

Abstract

Evapotranspiration is integral to studies of the Earth system, yet it is difficult to measure on regional scales. One estimation technique is a terrestrial water budget, i.e., total precipitation minus the sum of evapotranspiration and net runoff equals the change in water storage. Gravity Recovery and Climate Experiment (GRACE) satellite gravity observations are now enabling closure of this equation by providing the terrestrial water storage change. Equations are presented here for estimating evapotranspiration using observation based information, taking into account the unique nature of GRACE observations. GRACE water storage changes are first substantiated by comparing with results from a land surface model and a combined atmospheric-terrestrial water budget approach. Evapotranspiration is then estimated for 14 time periods over the Mississippi River basin and compared with output from three modeling systems. The GRACE estimates generally lay in the middle of the models and may provide skill in evaluating modeled evapotranspiration.

Abstract

The magnetic properties of antiferromagnetic (AFM) EuTe epitaxial layers and short period EuTe/PbTe superlattices (SLs), grown by molecular beam epitaxy on (111) BaF$_2$ substrates, were studied by magnetization and neutron diffraction measurements. Considerable changes of the N\'eel temperature as a function of the EuTe layer thickness as well as of the strain state were found. A mean field model, taking into account the variation of the exchange constants with the strain-induced lattice distortions, and the nearest neighbor environment of a Eu atoms, was developed to explain the observed $T_{\text N}$ changes in wide range of samples. Pronounced interlayer magnetic correlations have been revealed by neutron diffraction in EuTe/PbTe SLs with PbTe spacer thickness up to 60 \AA. The observed diffraction spectra were analyzed, in a kinematical approximation, assuming partial interlayer correlations characterized by an appropriate correlation parameter. The formation of interlayer correlations between the AFM EuTe layers across the nonmagnetic PbTe spacer was explained within a framework of a tight-binding model. In this model, the interlayer coupling stems from the dependence of the total electronic energy of the EuTe/PbTe SL on the spin configurations in adjacent EuTe layers. Abstract

Growing giant-breed dogs are more susceptible to developing skeletal disorders than small-breed dogs when raised on diets with deficient or excessive Ca content. Differential hormonal regulation of Ca homeostasis in dogs with different growth rates was investigated in Great Danes (GD, n = 9) and Miniature Poodles (MP, n = 8). All animals were raised on the same balanced diet and under identical conditions. Calciotropic and growth-regulating hormones were measured. Production and clearance of 1,25-dihydroxycholecalciferol (1,25[OH] 2 D 3 ) were investigated with the aid of [ 3 H]-1,25(OH) 2 D 3 and renal messenger RNA abundance of 1α-hydroxylase and 24-hydroxylase. Intestinal, renal, and skeletal Ca handling were evaluated with the aid of 45 Ca balance studies. Skeletal development was evaluated by radiology and histomorphometry. Great Danes had greater (P < 0.001) growth rates than MP, as indicated by the 17-fold greater body weight gain, by increased longitudinal growth reflected in the increased (P < 0.05) gain in length of the radius and ulna, and by increased (P < 0.001) growth plate thickness. These findings were accompanied in GD by greater (P < 0.05) plasma GH and IGF-I concentrations. Effects were observed for vitamin D 3 metabolism, such as greater (P < 0.01) plasma 1,25(OH) 2 D 3 concentrations due to decreased (P < 0.01) clearance rather than increased production of 1,25(OH) 2 D 3 , and decreased (P < 0.01) plasma 24,25-dihydroxycholecalciferol (24,25[OH] 2 D 3 ) concentrations likely due to competitive inhibition of the production of 24,25(OH) 2 D 3 . These findings were accompanied in both breeds by a limited hormonal regulation of Ca and P absorption at the intestinal level, and in GD by increased (P < 0.05) renal reabsorption of inorganic P (P ι ) compared with MP, resulting in greater (P < 0.01) P ι retention and greater (P < 0.01) plasma P ι concentrations. Bone turnover, resorption, and formation were greater (P < 0.01) in GD than in MP. In addition, GD had more irregular (P < 0.01) growth plates than MP, accompanied by disorders of endochondral ossification. It is suggested that in GD, increased calcitonin levels and/or a relative deficiency in 24,25(OH) 2 D 3 at the growth-plate level may both be responsible for the retarded maturation of chondrocytes, resulting in retained cartilage cones and osteochondrosis, and this may be a pathophysiological factor for the increased susceptibility of large breed dogs to developing skeletal disorders.

Abstract

The effects of excessive non-toxic dietary Vitamin D3 supplementation on Ca homeostasis with specific effects on endochondral ossification and skeletal remodeling were investigated in a group of growing Great Dane dogs supplemented with cholecalciferol (Vitamin D3; HVitD) versus a control group (CVitD) (1350 μg versus 11.4 μg Vitamin D3 per kilogram diet) from 6 to 21 weeks of age. There were no differences between groups in plasma concentrations of total Ca, inorganic phosphate, growth hormone, and insulin-like growth factor I and no signs of Vitamin D3 intoxication in HVitD. For the duration of the study in HVitD compared to CVitD, plasma levels of parathyroid hormone (PTH) decreased, calcitonin (CT) increased, 25-hydroxycholecalciferol [25(OH)D3] increased 30- to 75-fold, 24,25-dihydroxycholecalciferol [24,25(OH)2D3] increased 12- to 16-fold, and 1,25-dihydroxycholecalciferol [1,25(OH)2D3] decreased by approximately 40%. The latter was attributed to the two-fold increased metabolic clearance rate in the HVitD versus CVitD accompanied by the absence of the anabolic effect of PTH on the production of 1,25(OH)2D3. Fractional Ca absorption (α) did not differ between groups at 8 and 14 weeks of age, whereas at 20 weeks of age α increased by only 16.4% in HViAtD compared to CVitD. Excessive non-toxic Vitamin D3 supplementation resulted in decreased bone remodeling and focal enlargement of the growth plate with morphology resembling those induced by administration of CT. Hypercalcitoninemia and the imbalanced relationship between 1,25(OH)2D3 and 24,25(OH)2D3 are potent candidates for the disturbed endochondral ossification. © 2003 Elsevier Science Inc. Abstract

The evolution of insect sociality has repeatedly involved changes in developmental events and their timing. Here, we propose the hypothesis that loss of a canonical regulator of moulting and metamorphosis, prothoracicotropic hormone (PTTH), and its receptor, Torso, is associated with the evolution of sociality in bees. Specifically, we posit that the increasing importance of social influences on early developmental timing in social bees has led to their decreased reliance on PTTH, which connects developmental timing with abiotic cues in solitary insects. At present, the evidence to support this hypothesis includes the absence of genes encoding PTTH and Torso from all fully-sequenced social bee genomes and its presence in all available genomes of solitary bees. Based on the bee phylogeny, the most parsimonious reconstruction of evolutionary events is that this hormone and its receptor have been lost multiple times, across independently social bee lineages. These gene losses shed light on possible molecular and cellular mechanisms that are associated with the evolution of social behavior in bees. We outline the available evidence for our hypothesis, and then contextualize it in light of what is known about developmental cues in social and solitary bees, and the multiple precedences of major developmental changes in social insects.

Abstract

Abstract We review the physics opportunities of the Future Circular Collider, covering its e+e-, pp, ep and heavy ion programmes. We describe the measurement capabilities of each FCC component, addressing the study of electroweak, Higgs and strong interactions, the top quark and flavour, as well as phenomena beyond the Standard Model. We highlight the synergy and complementarity of the different colliders, which will contribute to a uniquely coherent and ambitious research programme, providing an unmatchable combination of precision and sensitivity to new physics.

Abstract

Abstract Background Cancer research has made great progress in the recent years. With the increasing number of options in diagnosis and therapy the implementation of tumorboards (TUBs) has become standard procedure in the treatment of cancer patients. Adherence tests on tumor board decisions are intended to enable quality assurance and enhancement for work in tumor boards in order to continuously optimize treatment options for cancer patients. Methods Subject of this study was the adherence of the recommendations made in three of 14 tumorboards, which take place weekly in the Center for Integrated Oncology (CIO) at the University Hospital Bonn. In total, therapy recommendations of 3815 patient cases were checked on their implementation. A classification into four groups has been made according to the degree of implementation. A second classification followed regarding the reasons for differences between the recommendation and the therapy which the patient actually received. Results The study showed that 80.1% of all recommendations in the three TUBs were implemented. 8.3% of all recommendations showed a deviance. Most important reasons for the deviances were patient wish (36.5%), patient death (26%) and doctoral decision, due to the patient’s comorbidities or side effects of the treatment (24.1%).Interestingly, deviance in all three tumor boards in total significantly decreased over time. Conclusions Aim of the study was to clarify the use of tumor boards and find approaches to make them more efficient. Based on the results efficiency might be optimized by increased consideration of patients` preferences, improved presentation of patient-related data, more detailed documentation and further structuring of the tumor board meetings.

Increasing excitability in the peri-infarct area enhances motor recovery after stroke. Here the authors show that expressing CREB, a transcription factor known for its role in synaptic plasticity, or increasing activity of CREB-expressing cells near the stroke site improves recovery in an effect that is strong enough that it can be used to turn on and off motor recovery after stroke.

Abstract

Diagnostics of hydrological models are pivotal for a better understanding of catchment functioning, and the analysis of dominating model parameters plays a key role for region-specific calibration or parameter transfer. A major challenge in the analysis of parameter sensitivity is the assessment of both temporal and spatial differences of parameter influences on simulated streamflow response. We present a methodological approach for global sensitivity analysis of hydrological models. The multilevel approach is geared towards complementary forms of streamflow response targets, and combines sensitivity analysis directed to hydrological fingerprints, i.e. temporally independent and temporally aggregated characteristics of streamflow (INDPAS), with the conventional analysis of the temporal dynamics of parameter sensitivity (TEDPAS).The approach was tested in 14 mesoscale headwater catchments of the Ruhr River in western Germany using simulations with the spatially distributed hydrological model mHM. The multilevel analysis with diverse response characteristics allowed us to pinpoint parameter sensitivity patterns much more clearly as compared to using TEDPAS alone. It was not only possible to identify two dominating parameters, for soil moisture dynamics and evapotranspiration, but we could also disentangle the role of these and other parameters with reference to different streamflow characteristics. The combination of TEDPAS and INDPAS further allowed us to detect regional differences in parameter sensitivity and in simulated hydrological functioning, despite the rather small differences in the hydroclimatic and topographic setting of the Ruhr headwaters.

Abstract

We report on the investigation of CdTe/Cd0.75Mg0.25Te quantum wells grown by molecular-beam epitaxy and containing a digital distribution of magnetic MnTe fractional monolayers in the well. The influence of electronic confinement and of exciton primary localization on the formation of ferromagnetic ordering on the scale of small spin clusters (polarons) has been investigated by performing selective excitation of the exciton luminescence. The calculated temperature dependence of the free magnetic polarons binding energy F is found to qualitatively agree with the photoluminescence excitation data and the ferromagnetic clusters result to be more stable in these magnetic structures of reduced dimensionality, with respect to the three-dimensional case. The magneto-optical Kerr effect is discussed in the case of reflectance difference spectroscopy (RDS) and from the angular dependence of the RDS spectra in magnetic field, signals due to interband Faraday rotation and signals originating from crystal anisotropy ...

Abstract

A global heat flux model based on a fractional derivative of plasma pressure is proposed for the heat transport in fusion plasmas. The degree of the fractional derivative of the heat flux, α, is defined through the power balance analysis of the steady state. The model was used to obtain the experimental values of α for a large database of the Joint European Torus (JET) carbon-wall as well as ITER like-wall plasmas.
