1. Swimming behaviour and magnetotaxis function of the marine bacterium strain MO-1

Abstract

Summary Magnetotactic bacteria (MTB) have the unique capacity to align and swim along the geomagnetic field lines downward to the oxic–anoxic interface in chemically stratified water columns and sediments. They are most abundant within the first few centimetres of sediments below the water–sediment interface. It is unknown how MTB penetrate into the sediment layer and swim in the pocket water, while their movements are restricted by the alignment along the magnetic field lines. Here we characterized the swimming behaviour of the marine fast-swimming magnetotactic ovoid bacterium MO-1.We found that it rotates around and translates along its short body axis to the magnetic north (northward). MO-1 cells swim forward constantly for a minimum of 1770 μm without apparent stopping. When encountering obstacles, MO-1 cells squeeze through or swim southward to circumvent the obstacles. The distance of southward swimming is short and inversely proportional to the magnetic field strength. Using a magnetic shielding device, we provide direct evidence that magnetotaxis is beneficial to MO-1 growth and becomes essential at low cell density. Environmental implications of the fast-swimming magnetotactic behaviour of magnetococci are discussed.

Abstract

International audience; We propose a simple microfluidic device able to separate submicron particles (critical size ~0.1 µm) from a complex sample with no filter (minimum channel dimension being 5 µm), by hydrodynamic filtration. A model taking into account the actual velocity profile and hydrodynamic resistances enables predicting the chip sorting properties for any geometry. Two design families are studied to obtain (i) small sizes within minutes (low-aspect ratio, two-level chip) and (ii) micron-sized sorting with µL flowrate (3D architecture based on lamination). We obtain quantitative agreement of sorting performances both with experiments and with numerical solving, and determine the limits of the approach. We therefore demonstrate a passive, filter-less sub-micron size sorting with simple, robust, easy to fabricate design.

Abstract

International audience; We demonstrate a new concept of devices, which by combining 3D fluid engineering and localized mag-netic actuation enables the full integration of a cell tagging and magnetic separation device. We used a low cost, commercially available dry film (EMS Inc, Ohio, USA) that fits microfluidic requirements and gives the possibility to build easily 3D microfluidic structures. The labelling of blood monocytes with su-perparamagnetic particles was performed "up stream" with the aim of a microparticles 3D micromixer while cell separation was achieved "downstream" through continuous vertical separation over integrated micro-coils.

Abstract

International audience; We demonstrate the use of a novel dry film photoresist DF-1000 series for the fabrication of multilevel microfluidic devices by combining a standard lithography technique and lamination technology. The optimization of the technological process enables achievement of high aspect ratio structures: 7 : 1 for free standing structures and 5 : 1 for channel structures. We proved that DF films feature a low autofluorescence level, similar to that of the SU-8 resist and compatible with most lab-on-a-chip applications. The chemical stability against aggressive solvents was also investigated. Last but not least, the non-cytotoxic effect according to ISO 10993-5 on the development of L-929 mouse fibroblast cells was established. Ultimately, we showed that this low-cost material combined with multilevel lamination and UV-lithography techniques allowed the fabrication of 3D microfluidic mixers and opened the way to perform microfluidics in three dimensions.

Abstract

Accepté 10th International Conference on the Scientific and Clinical Applications of Magnetic Carriers, 10-14 June 2014 Dresden, Germany; International audience; Super-paramagnetic micro particles (SMPs) are broadly used in medical and biological applications (from cellular to genomic scales), but the handling tools for those particles remain mostly non-integrated. Since a decade, a few solutions for integrated actuation have been proposed and paved the way to the micro-scale control of SMPs. So far, spiral planar coils showed the best compromises between the available magnetic force and the actuator footprint. Albeit the technology to integrate this type of coils with microfluidic structures exists, it is hard to find examples of dual architectures using those two aspects for concrete applications. We therefore developed a SU-8 dry film lamination technique, which can be used with standard micro technology processes. It allows us to manufacture lab on chip integrating miniaturized electromagnets. Using that system, we achieved to separate a solution of SMPs with an 80% efficiency at a 2.5 µL/min flow rate (4.6 mm/s). It is also possible to perform trapping (all the beads stay on the coils) at lower flow rate.

Abstract

International audience; Circulating blood monocytes belong to the first line of defense against pathogens and inflammation. Monocytes can be divided into three populations defined by the expression of the cell surface molecules, CD 14 and CD 16. The CD 14(++) CD 16(-) cells, called "classical" monocytes, represent 85% to 95% of the total monocytes in a healthy person whereas CD 14(-) CD 16(+), called "proinflammatory" monocytes, are found in greater numbers in the blood of patients with acute inflammation and infectious diseases. This increase in the concentration of proinflammatory monocytes can be a good indicator of an infectious state. This study presents an immunosensor based on impedance detection for specific cell trapping of classical and proinflammatory monocytes. The grafting of specific antibodies (CD 14 or CD 16) was based on the use of mixed SAM associated with protein G. Each step of the functionalization was characterized by electrochemical methods, quartz crystal microbalance and atomic force microscopy. Faradaic electrochemical impedance spectroscopy and voltametric analysis confirmed the success of the modification process with a surface coverage reaching 92% for the antibody layer. The increase in the deposited mass at each step of the modification process confirmed this results revealing that one protein G in two was bound to an antibody. The cell trapping capacity, evaluated by the variation in the film resistance using non-faradaic impedance spectroscopy revealed that the cell trapping is selective, depending on the specific antibody grafted and quantitative with the range of detection being 1000 to 30,000 infected cells. This range of detection is consistent with the application targeted.

Abstract

Magnetotactic bacteria (MTB) have the unique capacity to align and swim along the geomagnetic field lines downward to the oxic-anoxic interface in chemically stratified water columns and sediments. They are most abundant within the first few centimetres of sediments below the water-sediment interface. It is unknown how MTB penetrate into the sediment layer and swim in the pocket water, while their movements are restricted by the alignment along the magnetic field lines. Here we characterized the swimming behaviour of the marine fast-swimming magnetotactic ovoid bacterium MO-1.We found that it rotates around and translates along its short body axis to the magnetic north (northward). MO-1 cells swim forward constantly for a minimum of 1770 μm without apparent stopping. When encountering obstacles, MO-1 cells squeeze through or swim southward to circumvent the obstacles. The distance of southward swimming is short and inversely proportional to the magnetic field strength. Using a magnetic shielding device, we provide direct evidence that magnetotaxis is beneficial to MO-1 growth and becomes essential at low cell density. Environmental implications of the fast-swimming magnetotactic behaviour of magnetococci are discussed.

Abstract

International audience; We present the use of a novel dry film photoresist DF-1050 (EMS) for the integration of multilevel microfluidic systems. We showed that high resolution and high aspect ratio (7:1) can be achieved for free standing structures. We proved also that the dry film autofluorescence level is compatible with most of lab-on-a-chip applications and established the biocompatibility of this dry film. Ultimately, we demonstrated that this low-cost material combined with multilevel lamination technology would allow introducing massively 3D microfluidic in lab-on-a-chip devices.
