Objective: To determine whether anatomical thigh muscle cross-sectional areas (MCSAs) and strength differ between osteoarthritis (OA) knees with frequent pain compared with contra-lateral knees without pain, and to examine the correlation between MCSAs and strength in painful vs painless knees.Methods: Forty-eight subjects (31 women; 17 men; age 45-78 years) were drawn from 4,796 Osteoarthritis Initiative (OAI) participants, in whom both knees displayed the same radiographic stage (KLG2 or 3), one with frequent pain (most days of the month within the past 12 months) and the contra-lateral one without pain. Axial MR images were used to determine MCSAs of extensors, flexors and adductors at 35% femoral length (distal to proximal) and in two adjacent 5 mm images. Maximal isometric extensor and flexor forces were used as provided from the OAI database.Results: Painful knees showed 5.2% lower extensor MCSAs (P=0.00003; paired t-test), and 7.8% lower maximal extensor muscle forces (P=0.003) than contra-lateral painless knees. There were no significant differences in flexor forces, or flexor and adductor MCSAs (P>0.39). Correlations between force and MCSAs were similar in painful and painless OA knees (0.44Conclusions: Knees with frequent pain demonstrate lower MCSAs and force of the quadriceps (but not of other thigh muscles) compared with contra-lateral knees without knee pain with the same radiographic stage. Frequent pain does not appear to affect the correlations between MCSAs and strength in OA knees. Objective: To validate a semi-automated method for thigh muscle and adipose tissue cross-sectional area (CSA) segmentation from MRI., Materials and Methods: An active shape model (ASM) was trained using 113 MRI CSAs from the Osteoarthritis Initiative (OAI) and combined with an active contour model and thresholding-based post-processing steps. This method was applied to 20 other MRIs from the OAI and to baseline and follow-up MRIs from a 12-week lower-limb strengthening or endurance training intervention (n = 35 females). The agreement of semi-automated vs. previous manual segmentation was assessed using the Dice similarity coefficient and Bland-Altman analyses. Longitudinal changes observed in the training intervention were compared between semi-automated and manual segmentations., Results: High agreement was observed between manual and semi-automated segmentations for subcutaneous fat, quadriceps and hamstring CSAs. With strength training, both the semi-automated and manual segmentation method detected a significant reduction in adipose tissue CSA and a significant gain in quadriceps, hamstring and adductor CSAs. With endurance training, a significant reduction in adipose tissue CSAs was observed with both methods., Conclusion: The semi-automated approach showed high agreement with manual segmentation of thigh muscle and adipose tissue CSAs and showed longitudinal training effects similar to that observed using manual segmentation.

Awareness of postmortem degradation processes in a human body is fundamental to develop methods for forensic time since death estimation (TDE). Currently, applied approaches are all more or less limited to certain postmortem phases, or have restrictions on behalf of circumstances of death. Novel techniques, however, rarely exceed basic research phases due to various reasons. We report the first application of a novel method, based on decay of muscle proteins, in a recent case of murder-suicide, where other TDE methods failed to obtain data. We detected considerably different protein degradation profiles in both individuals involved and compared the data to our presently available database. We obtained statistical evidence for un-simultaneous death and therefore received valuable information to trace the progression of events based on protein degradation. Although we could not sensibly convert the data to respective times of death, this case highlights the potential for future application and elucidates the necessary further steps to develop a viable TDE method.

Background: Muscle injuries are among the most common sports-related lesions in athletes; however, optimal treatment remains obscure. Extracorporeal shock wave therapy (ESWT) may be a promising approach in this context, because it has gained increasing importance in tissue regeneration in various medical fields., Hypothesis: ESWT stimulates and accelerates regenerative processes of acute muscle injuries., Study Design: Controlled laboratory study., Methods: Adult Sprague-Dawley rats were divided into 4 experimental groups (2 ESWT+ groups and 2 ESWT- groups) as well as an uninjured control group (n ≥ 6 in each group). An acute cardiotoxin-induced injury was set into the quadriceps femoris muscle of rats in the experimental groups. A single ESWT session was administered to injured muscles of the ESWT+ groups 1 day after injury, whereas ESWT- groups received no further treatment. At 4 and 7 days after injury, 1 each of the ESWT+ and ESWT- groups was euthanized. Regenerating lesions were excised and analyzed by histomorphometry and immunohistochemistry to assess fiber size, myonuclear content, and recruitment of satellite cells., Results: The size and myonuclear content of regenerating fibers in ESWT+ muscle was significantly increased compared with ESWT- muscle fibers at both 4 and 7 days after injury. Similarly, at both time points, ESWT+ muscles exhibited significantly higher contents of pax7-positive satellite cells, mitotically active H3P + cells, and, of cells expressing the myogenic regulatory factors, myoD and myogenin, indicating enhanced proliferation and differentiation rates of satellite cells after ESWT. Mitotic activity at 4 days after injury was doubled in ESWT+ compared with ESWT- muscles., Conclusion: ESWT stimulates regeneration of skeletal muscle tissue and accelerates repair processes., Clinical Relevance: We provide evidence for accelerated regeneration of damaged skeletal muscle after ESWT. Although further studies are necessary, our findings support the view that ESWT is an effective method to improve muscle healing, with special relevance to sports injuries.

Forensic estimation of time since death relies on diverse approaches, including measurement and comparison of environmental and body core temperature and analysis of insect colonization on a dead body. However, most of the applied methods have practical limitations or provide insufficient results under certain circumstances. Thus, new methods that can easily be implemented into forensic routine work are required to deliver more and discrete information about the postmortem interval (PMI). Following a previous work on skeletal muscle degradation in the porcine model, we analyzed human postmortem skeletal muscle samples of 40 forensic cases by Western blotting and casein zymography. Our results demonstrate predictable protein degradation processes in human muscle that are distinctly associated with temperature and the PMI. We provide information on promising degradation markers for certain periods of time postmortem, which can be useful tools for time since death delimitation. In addition, we discuss external influencing factors such as age, body mass index, sex, and cause of death that need to be considered in future routine application of the method in humans., Competing Interests: Compliance with ethical standards This study was approved by the ethics commission of the University of Salzburg (EK-GZ 39/2015).

Estimating the time since death is a very important aspect in forensic sciences which is pursued by a variety of methods. The most precise method to determine the postmortem interval (PMI) is the temperature method which is based on the decrease of the body core temperature from 37 °C. However, this method is only useful in the early postmortem phase (~0-36 h). The aim of the present work is to develop an accurate method for PMI determination beyond this present limit. For this purpose, we used sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), Western blotting, and casein zymography to analyze the time course of degradation of selected proteins and calpain activity in porcine biceps femoris muscle until 240 h postmortem (hpm). Our results demonstrate that titin, nebulin, desmin, cardiac troponin T, and SERCA1 degraded in a regular and predictable fashion in all samples investigated. Similarly, both the native calpain 1 and calpain 2 bands disintegrate into two bands subsequently. This degradation behavior identifies muscular proteins and enzymes as promising substrates for future molecular-based PMI determination technologies.

PGC-1α (peroxisome proliferator-activated receptor γ co-activator 1α) is an important regulator of mitochondrial biogenesis and a master regulator of enzymes involved in oxidative phosphorylation. Recent evidence demonstrated that the Gly482Ser single nucleotide polymorphism (SNP) in the PGC-1α gene affects insulin sensitivity, blood lipid metabolism and binding to myocyte enhancer factor 2 (MEF2). Individuals carrying this SNP were shown to have a reduced cardiorespiratory fitness and a higher risk to develop type 2 diabetes. Here, we investigated the responses of untrained men with the Gly482Ser SNP to a 10 week programme of endurance training (cycling, 3 x 60 min/week, heart rate at 70-90% VO2peak). Quantitative data from analysis of biopsies from vastus lateralis muscle revealed that the SNP group, in contrast to the control group, lacked a training-induced increase in content of slow contracting oxidative fibres. Capillary supply, mitochondrial density, mitochondrial enzyme activities and intramyocellular lipid content increased similarly in both groups. These results indicate that the impaired binding of MEF2 to PGC-1α in humans with this SNP impedes exercise-induced fast-to-slow muscle fibre transformation.

Introduction: Quadriceps heads are important in biomechanical stabilization and in the pathogenesis osteoarthritis of the knee. This is the first study to explore the relative distribution of quadriceps head anatomical cross-sectional areas (ACSA) and volumes, and their response to pain and to training intervention., Methods: The relative proportions of quadriceps heads were determined in 48 Osteoarthritis Initiative participants with unilateral pain (65% women; age 45-78 years). Quadriceps head volumes were also measured in 35 untrained women (45-55 years) before and after 12-week training intervention. Cross-sectional areas of the vastus medialis (VM), inter-medius (VIM), and lateralis (VL), and of the rectus femoris (RF) were determined from axial T1-weighted MR images., Results: The proportion of the VM on the total quadriceps ACSA increased from proximal to distal. The difference in quadriceps ACSA of painful (vs. pain-free) limbs was -5.4% for the VM (p<0.001), -6.8% for the VL (p<0.01), -2.8% for the VIM (p=0.06), and +3.4% for the RF (p=0.67) but the VM/VL ratio was not significantly altered. The muscle volume increase during training intervention was +4.2% (p<0.05) for VM, +1.3% for VL, +2.0% for VIM (p<0.05) and +1.6% for RF., Conclusion: The proportion of quadriceps head relative to total muscle ACSA and volume depends on the anatomical level studied. The results suggest that there may be a differential response of the quadriceps heads to pain-induced atrophy and to training-related hypertrophy. Studies in larger samples are needed to ascertain whether the observed differences in response to pain and training are statistically and clinically significant.

Temperature is an important factor influencing teleost muscle growth, including a lasting ('imprinted') influence of embryonic thermal experience throughout all further life. However, little is known about the cellular processes behind this phenomenon. The study reported here used digital morphometry and immunolabelling for Pax7, myogenin and H3P to quantitatively examine the effects of thermal history on muscle precursor cell (MPC) behaviour and muscle growth in pearlfish (Rutilus meidingeri) until the adult stage. Fish were reared at three different temperatures (8.5, 13 and 16°C) until hatching and subsequently kept under the same (ambient) thermal conditions. Cellularity data were combined with a quantitative analysis of Pax7+ MPCs including those that were mitotically active (Pax7+/H3P+) or had entered differentiation (Pax7+/myogenin+). The results demonstrate that at hatching, body lengths, fast and slow muscle cross-sectional areas and fast fibre numbers are lower in fish reared at 8.5 and 13°C than at 16°C. During the larval period, this situation changes in the 13°C-fish, so that these fish are finally the largest. The observed effects can be related to divergent cellular mechanisms at the MPC level that are initiated in the embryo during the imprinting period. Embryos of 16°C-fish have reduced MPC proliferation but increased differentiation, and thus give rise to larger hatchlings. However, their limited MPC reserves finally lead to smaller adults. By contrast, embryos of 13°C-fish and, to a lesser extent, 8.5°-fish, show enhanced MPC proliferation but reduced differentiation, thus leading to smaller hatchlings but allowing for a larger MPC pool that can be used for enhanced post-hatching growth, finally resulting in larger adults.

Quantitative data on muscle volume (MV) are important for estimating maximal muscle power. The objective of this study was to determine the correlation between anatomical cross-sectional areas (ACSAs) and the MV in thigh muscles (extensors, flexors, adductors, and sartorius) in perimenopausal women, and to identify at which proximal-to-distal level a single-slice ACSA measurement with magnetic resonance imaging (MRI) displays the highest correlation in each of these. Axial MRIs of the thigh were acquired in 41 perimenopausal women aged 50.8 +/- 3.2 years. Segmentation of the extensors, flexors, adductors, and the sartorius was performed between the femoral neck (0%) and the distal end of the intermediate vastus (100%). MVs were determined by numerical integration, and the ACSA was calculated from three-dimensional reconstructions at 10% intervals from proximal to distal. The extensors contributed 50%, the flexors 19%, the adductors 28%, and the sartorius 3% of the total thigh MV. Maximal correlations between ACSA and MV were observed at the 20-40% proximal-to-distal level in the extensors (R (2) = 0.73), at 30% in the adductors (R (2) = 0.82), and at 70% in the flexors (R (2) = 0.72) and sartorius (R (2) = 0.85), respectively. ACSA at 50% displayed the highest overall correlations (R (2) >or= 0.69) with MV for all muscle groups. Single-slice ACSAs from MRI displayed high correlations with MVs. Although the (proximal to distal) measurement locations with the highest correlation varied between the muscle groups, a single slice at the 50% location achieved the best compromise in terms of correlation between ACSA and MV across extensors, flexors, adductors and the sartorius.

Ruptures of the rotator cuff tendons of the human shoulder are a common incidence and lead to functional impairment of the four muscles connected to the cuff, entailing profound changes of their cellular tissue composition. Most importantly, such tendon tears lead to atrophy, fatty degeneration and fibrosis of the corresponding muscles. The muscle most commonly affected with such changes is the M. supraspinatus. The present study uses biopsy samples from the supraspinatus muscle of 12 elderly patients and 6 controls to examine the rupture-induced muscle change at both the cellular and the intracellular (ultrastructural) levels. Amounts of fatty tissue, connective tissue and muscle were assessed by light microscopy-based morphometry and stereology. Stereology of electron micrographs was employed to determine volume densities of muscle fibre mitochondria, myofibrils and intracellular lipid. Results demonstrate that the supraspinatus muscles of patients with a massive rupture contain significantly higher amounts not only of fatty tissue but also of intracellular lipid than those of control subjects. These patients further exhibit a major decrease in relative amounts of myofibrils, thus confirming that change of intracellular composition is a major component of the observed muscle degeneration. Introduction: Epidemiological studies show a positive relationship between physical activity and cartilage volume, suggesting that exercise may protect against osteoarthritis. Cross-sectional experimental studies, however, have failed to show significant differences in knee cartilage morphology between athletes and nonathletic controls. The aim of the study was to test the hypothesis that knee cartilage morphology, specifically regional cartilage thickness and global subchondral bone area, is modified in sedentary, untrained adult women who increased their physical fitness during a 3-month supervised exercise intervention., Materials and Methods: Thirty-eight untrained women, aged 45-55 years, were randomly assigned to: endurance training (n = 18), strength training (n = 15) and autogenic training (control group, n = 5). Patellar and femorotibial knee cartilage morphology was determined before and after the 3-month supervised training intervention, using magnetic resonance imaging and state-of-the-art image analysis., Results: Whereas the endurance training group showed a significant increase in cardiorespiratory fitness and the strength training group a significant increase in the maximal voluntary isometric contraction force of the leg, this study did not show significant differences in knee cartilage thickness and volume, subchondral bone area or regional cartilage thickness between baseline and follow-up acquisitions within any intervention group., Discussion: This randomized longitudinal study provides no evidence that a 3-month exercise intervention in untrained middle-aged women can significantly alter knee joint morphology. Muscle development in teleost embryos has been shown to depend on myogenic cell recruitment from the dermomyotome (DM). However, little is known as to the cellular mechanisms that account for myotome growth after the dissociation of the DM. Here we combine immunolabeling for cell-specific markers with quantitative analysis to determine the sources and patterns of activation of myogenic cells in pearlfish larvae. Results demonstrate that appearance of mitotically active myogenic precursors inside the myotome coincides with the dissociation of the DM. Such cells are preferentially aggregated within the posterior lateral fast muscle. We therefore propose a growth model in which a pool of proliferative DM-derived precursors transferred to the posterior lateral fast muscle functions as an important source of myogenic cell spread to carry forward stratified fast muscle hyperplasia. This indicates that postembryonic teleost muscle growth includes a cellular mechanism that has no direct equivalent in the amniotes.

The effect of exercise modalities on determinants of sarcopenia, specifically, lean tissue mass, maximal voluntary isometric contraction (MVIC), muscle quality (MQ), and cardiorespiratory fitness (VO(2)peak) over 12 weeks were studied in 42 (45-55 years, 60 days of amenorrhea, with /=48 h rest, ET(i) = 7; consecutive, 24 h, ET(c) = 9) and autogenic training group (AT = 11). The predominant source of myogenic cells in vertebrates is the dermomyotome (DM). In teleost fish, recent research has provided a useful but limited picture of how myogenic precursors originate from the DM and how they develop into muscle fibers. Here, we combine detailed morphological analysis with examination of molecular markers in trout to describe the cellular mechanisms by which the lateral fast muscle growth zone is created during second phase myogenesis. Results suggest that this occurs by lateral-to-medial immigration of myogenic cells de-epithelializing from the posterior DM lip. These cells then appear to stop proliferation and migrate anteriorly to finally differentiate into muscle fibres. This seems to be a continuation of the rotational cell movement that creates the teleost DM during early somite development. In this article, whole mount in situ hybridization is used to examine early blood vessel and blood cell development in the embryos of the brown trout Salmo trutta lacustris. cDNAs encoding for the angiogenic markers fli1 and flk1, and for the hematopoietic markers gata1 and gata2, were identified from an expressed sequence tag library of rainbow trout. Results show that fli1, flk1 and gata2 are activated in bilateral bands of the lateral trunk mesoderm before the onset of somitogenesis, shortly followed by gata1. These bands then converge toward the ventral midline to form the intermediate cell mass (ICM) (anterior ICM). Subsequent axial vasculogenesis and initial blood cell formation involve a clear spatial separation of fli1 and gata gene expression. Fli1 staining is most intense within the axial vessel (dorsal aorta, posterior cardinal vein) forming and lateral ICM cells, whereas binding of gata1 and gata2 probes becomes confined to the central portion of ICM cells beneath the dorsal aorta. This is followed by a first wave of angiogenesis, indicated by expression of fli1 and flk1. This gives rise to the intersegmental, dorsal longitudinal anastomotic and intestinal vessels. Further angiogenesis and hematopoiesis are activated in the "posterior ICM" of the tail. Here, the absence of gata1 indicates that hematopoiesis in this tissue generates myeloid rather than erythroid cells. The results supplement and validate previous, now historical morphological work in salmonids, thus aiding the elucidation of a comprehensive general scheme of angiogenic and hematopoietic development in the teleost embryo.

The decline of cellular and especially mitochondrial functions with age is, among other causes, held responsible for a decrease in physiological fitness and exercise capacity during lifetime. We investigated size- and age-related changes in the physiology of exercising specimens of the short lived swimming scallop Aequipecten opercularis (maximum life span 8 to 10 years) from the Isle of Man, UK. A. opercularis swim mainly to avoid predators, and a decrease in swimming abilities would increase the risk of capture and lower the rates of survival. Bigger (older) individuals were found to have lower mitochondrial volume density and aerobic capacities (citrate synthase activity and adenylates) as well as less anaerobic capacity deduced from the amount of glycogen stored in muscle tissue. Changes in redox potential, tissue pH and the loss of glutathione in the swimming muscle during the exercise were more pronounced in young compared to older individuals. This indicates that older individuals can more effectively stabilize cellular homeostasis during repeated exercise than younger animals but with a possible fitness cost as the change in physiology with age and size might result in a changed escape response behaviour towards predators.

Muscle cell recruitment (hyperplasia) during myogenesis in the vertebrate embryo is known to occur in three consecutive phases. In teleost fish (including zebrafish), however, information on myogenic precursor cell activation is largely fragmentary, and comprehensive characterization of the myogenic phases has only been fully undertaken in a single slow-growing cyprinid species by examination of MEF2D expression. Here, we use molecular techniques to provide a comprehensive characterization of MyoD and Myogenin expression during myogenic cell activation in embryos and larvae of brown trout, a fast-growing salmonid with exceptionally large embryos. Results confirm the three-phase pattern, but also demonstrate that the second and third phases begin simultaneously and progress vigorously, which is different from the previously described consecutive activation of these phases. Furthermore, we suggest that Pax7 is expressed in myogenic progenitor cells that account for second- and third-phase myogenesis. These findings are discussed in relation to teleost myotome development and to teleost growth strategies.

Present knowledge indicates that fibre recruitment (hyperplasia) in developing teleost fish occurs in three distinct phases. However, the origin and relationship of the myogenic precursors activated during the different phases remains unclear. Here, we address this issue using molecular techniques on embryos and larvae of pearlfish, a large cyprinid species. Results provide comprehensive molecular characterisation of cell recruitment over the three phases of myogenesis, identifying muscle types as they arise. Specifically, we show that the myogenic cells arising during 2nd phase myogenesis are clearly different from the myogenic cells arising during the 3rd phase and that the dermomyotome is a major source of myogenic cells driving 2nd phase hyperplasia. Patterns of initial muscle formation are well documented for teleost fish. Here, attention is focused upon sturgeons, which arose close to the base of the actinopterygian radiation and whose early development has remained largely unresearched. We demonstrate that some features of muscle development are common to both groups of fish, the most important being the origin and form of migration of adaxial cells to establish the superficial slow fibre layer. This, together with information on initial innervation and capillarisation, strongly suggests a common basis for muscle developmental mechanisms among fish. An important feature that is different between sturgeons and teleosts is that sturgeons lack any cellular dorsal-ventral separation of the myotome that involves the insertion of muscle pioneer (MP)-like cells at the site of the future horizontal septum. This, and information from other fish and from sarcopterygians, permits the supposition that such MP-defined dorsal-ventral separation is a teleost apomorphism. These and other findings are discussed in relation to their significance for the evolution of fish muscle developmental patterns.

Larval zebrafish (Danio rerio) of two different age classes ("swim-up" larvae, 9 days old; "free-swimming" larvae, 21 days old) were exposed to either an endurance/continuous training or interval training. Control animals were kept in stagnant water. A comparison of cardiac activity of trained (either endurance or interval) and untrained animals at the end of the training regime revealed no differences in heart rate, end-diastolic and end-systolic ventricular volume, and cardiac output. Training also had no influence on the concentration of erythrocytes in the blood. Thus, at the level of total oxygen transport in the blood, training did not provoke any improvement during the first 32 days of development. Significant changes, however, were observed at the tissue level. In free-swimming larvae [i.e., between 21 and 32 days postfertilization (dpf)] endurance training increased the capillarization of both axial muscle caudal to the anus and the tail fin. In addition, mitochondrial density of red and intermediate muscle fibers increased significantly. In contrast to capillarization, even swim-up larvae, trained between 9 and 15 dpf, were affected. The observed increase in mitochondrial content indicates a high demand for oxygen and energy-rich metabolites for oxidative phosphorylation. In older larvae, this is met by the increase in capillarization that improves the blood supply and with it the required oxygen and metabolite supply of muscle tissue. Both of these adaptational changes result in a reduction of diffusion distances (between capillary and muscle fiber as well as mitochondria) and may contribute to a higher resistance toward oxygen deficiency. Furthermore, this study indicates that plasticity of muscle tissue is already established in early stages of development at both the tissue and cellular levels.

Muscle cellularity patterns in teleost fish have normally been investigated using animals reared under constant temperature conditions. In the present study, Danube bleak (Chalcalburnus chalcoides mento) were reared under two different rising temperature regimes (cold, 12-16 degrees C; warm, 18-20 degrees C) designed to mimic the natural conditions experienced by the fish in temperate freshwater environments. Samples were taken from both groups of animals at intervals during their development. Transverse sections at the level of the anal vent were examined using light and electron microscopy, histochemistry and immunohistochemistry techniques. Total cross-sectional area of red and white muscle, as well as fibre numbers and fibre cross-sectional areas of one epaxial quadrant per specimen, were measured. Analysis of fibre numbers and sizes indicated that white and red myotomal muscles each develop in a different manner. In white muscle, the initial growth phase is dominated by fibre hypertrophy, while the later larval growth phase also includes significant hyperplasia. Red muscle growth is mainly due to hypertrophy within the studied developmental period. The temperature regimes applied in the present study may modify the mechanisms of muscle growth in different ways. For white muscle, pre-hatching hyperplasia (i.e. proliferation of somitic white fibre precursor cells) is reduced under the cold regime whereas post-hatching hyperplasia is not. The inverse is true for white fibre hypertrophy. A similar situation is seen with red muscle except that post-hatching hyperplasia is low and refractory to temperature. Rates of increase in relative amount of red muscle appear to depend not only upon species and temperature but also upon whether the fish have been reared under changing or constant thermal regimes. These findings are discussed in relation to 'landmark' events of early ontogeny (hatching, onset of swimming, start of exogeneous feeding) and to their implications for future accurate interpretation of temperature effects on teleost developmental biology and functional ecology.

Teleost fish are known to develop small populations of muscle fibres that are assumed to be tonic in nature although their contractile properties and many other characteristics remain unknown. Here we attempt to resolve some of the ambiguity and confusions surrounding the definition and functional role of tonic fibres in teleosts and provide new information on their ontogeny. We investigate the differentiation of tonic muscle fibres in three species of cyprinid fish using electron microscopy, histochemistry, immunohistochemistry and in situ hybridization. The fine structure of the fibres defined as tonic in the larvae used in this study complies with patterns known from studies in teleost adults. This allows formal definition of tonic fibres in cyprinid larvae. The tonic fibres may be recognized by a variety of features: (1) by their characteristic position along the medial confines of the red muscle insertion at the horizontal septum, (2) their fine structure, including solid clusters of irregularly cleaved myofibrils, thick and wavy Z-lines, and T-tubules at the A-band/I-band transitions, (3) their histochemical features, specifically weak but obvious staining for mATPase after alkaline preincubation, and lack of SDH activity in the more advanced larval stages, (4) their unique immunological properties, being the only fibre type in the myotome that reacts with a serum against chicken tonic myosin (anti- T2). Expression of tonic characters usually begins within a few fibres in the dorsal domain of the superficial red muscle insertion at the horizontal septum and hence involves a high degree of dorso-ventral polarity. The present evidence indicates that tonic fibres arise from separate myogenic stem cells rather than by transdifferentiation from existing red fibres. First appearance of tonic fibres during ontogeny correlates closely with the onset of free swimming and exogeneous feeding. We use this fact to argue that tonic fibres are probably a prerequisite for efficient locomotory control during prey capture, shoaling and predator avoidance.

The Straub tail reaction in mice is an S-shaped dorsiflexion of the mouse tail, long known as a sensitive and specific bioassay for morphine (CAS 57-27-2). It is based on a contraction of the sacro-coccygeal dorsalis muscles. In the case of morphine the reaction is thought to be induced by a long-lasting stimulation of the muscle's motor innervation at the level of the lumbosacral spinal cord. A similar reaction caused by epibatidine differs pharmacologically, because in this case the reaction can be specifically inhibited by the ganglionic blocking agent mecamylamine. This points to a site of action of epibatidine (CAS 140111-52-0) at the neuromuscular junction of the sacro-coccygeal dorsalis muscles and requires the presence of muscle fibres which react to depolarization by sustained contraction. The present study provides evidence of the existence of such fibres using histochemical and immunohistochemical methods.

The debate about the pattern of muscle formation in teleost fish has recently been heightened in the literature. Here we examine superficial muscle development in the pearlfish, a cyprinid endemic to a small area of Central Europe, and uninfluenced by economic interest and breeding. Using light and electron microscopy, histochemistry and immunohistochemistry techniques, we report that: (1) Superficial fibre precursors originate close to the notochord, are part of the same cell population as the so-called muscle pioneer cells, and are transferred laterally to end up at the surface of the myotome. (2) Superficial fibre maturation is exceptionally rapid. Structural and enzymatic functionality is attained at a time when prospective deep fibres have not passed beyond the early myotube state. This strong contrast weakens as the embryo develops. (3) Apart from the muscle pioneers, the superficial fibres appear to be capable of functioning before they receive any direct innervation, implying that signals are transferred to these fibres via cell-to-cell junctions. We suggest that the capability of rapid superficial fibre maturation is a rather general feature among teleosts and may aid pre-hatch survival under a variable environment. Our results indicate that muscle formation in teleost fish may follow a common basic pattern that is open to considerable ontogenetic and phylogenetic modification in response to habitat conditions.

This study is based on transmission electron microscopic (TEM) investigations of deep (fast, 'white') teleost fish muscle proliferation in early developmental stages of three European cyprinid species and the rainbow trout. Our fine structural findings provide evidence that early myotomal growth in these animals may utilize different mechanisms that are activated in close succession during early life history. First, initial enlargement of the deep muscle bulk in the embryo seems to be due to hypertrophy of the somite-cell derived stock of muscle fibres. Second, we suggest that deep muscle growth becomes additionally powered by attachment of presumptive myogenic cells that originate from and proliferate within the adjacent mesenchymal tissue lining. Third, mesenchyme-derived muscle cell precursors are thought to enter the myotomes via the myosepta. After migration between the pre-established muscle fibres these cells may function as myosatellite cells, thus at least partly providing the stem cell population for subsequent rapid hyperplastic growth. Finally, there is evidence that presumptive deep muscle satellite cells also proliferate by mitotic division in situ. A similar process of myogenic cell migration and proliferation may foster intermediate fibre differentiation. The model of myogenic cell migration is discussed in view of in vitro and in vivo data on satellite cell migratory power and with respect to temperature-induced and species dependent differences. As for the latter, our results indicate that patterns of muscle differentiation may diverge between a fast growing salmonid species and a moderately growing cyprinid species of similar final size. The model is compatible with the well-established idea that teleost muscle growth may rely on different subclasses of myosatellite cells.
