Abstract

Introduction/Objectives: To report the findings of extended follow-up and biweekly administration of elranatamab monotherapy in patients (pts) with relapsed/refractory multiple myeloma (RRMM) naïve to BCMA-directed therapies enrolled in Cohort A of MagnetisMM-3. Materials and methods: MagnetisMM-3 (NCT04649359) is an open-label, multicenter, registration phase 2 study evaluating the efficacy and safety of elranatamab monotherapy in pts with RRMM. Eligible pts were refractory to at least 1 proteasome inhibitor, 1 immunomodulatory drug, and 1 anti-CD38 antibody. Pts received subcutaneous elranatamab in 28-d cycles with step-up doses of 12 mg on cycle 1 day 1 (C1D1) and 32 mg on C1D4 followed by 76 mg once-weekly beginning C1D8. Pts treated for 6 cycles and achieving partial response (PR) or better, lasting ≥2 mo, were switched to 76 mg once every two weeks (Q2W). Results: Overall, 123 pts received elranatamab. Median pt age was 68.0 y (range, 36-89); 63.4% of pts had an ECOG PS ≥1. The median number of prior lines of therapy was 5.0 (2-22), with 96.7% and 42.3% of pts having triple-class- and penta-drug refractory disease, respectively. At data cutoff (≍12 mo after last pt initial dose), the median follow up was 12.8 mo (0.2-22.7); 34.1% of pts remained on treatment. The most common reasons for treatment discontinuation were progressive disease (39.0%) and adverse events (AE; 13.8%). Objective response rate per blinded independent central review (BICR) was 61% (95% CI 51.8-69.6), with 39 (31.7%) pts with complete response (CR) or stringent CR (sCR); very good partial response (VGPR) and PR were achieved in 29 (23.6%) and 7 (5.7%) pts, respectively. MRD-negativity (threshold 10−−5) was achieved by 92.0% (n = 23/25) of evaluable pts. Median duration of response (mDOR) has not been reached (95% CI 12.9-NE), and DOR at 12 mo was 74.1% (95% CI 60.5-83.6). In pts with CR/sCR or VGPR, mDOR was not reached by 12 mo; in pts with PR, mDOR was 5.2 mo (95% CI 1.6-NE). There were 46 responders by BICR who switched to Q2W dosing ≥24 wk prior to the data cutoff; among these pts, 80.4% maintained/improved their response ≥24 wk after the switch. Median progression-free and overall survival have not been reached by 12 mo, and the respective rates (95% CI) at 12 mo were 57.1% (47.2-65.9) and 62.0% (52.8-70.0). Most common grade 3/4 treatment emergent AEs were hematologic; grade 3/4 nonhematologic events reported in ≥5% of pts were COVID-pneumonia (10.6%), hypokalemia (9.8%), pneumonia (7.3%), sepsis (6.5%), hypertension (6.5%), ALT increased (5.7 %), and SARS-COV-2 test positive (5.7%). Among pts who switched to Q2W dosing (n = 58), the incidence of grade 3/4 AEs decreased by >10% after the switch. Discussion: Elranatamab remains efficacious and well tolerated in pts with RRMM after >1 y of follow-up. Updated analysis with a median follow-up of ≍15 mo, the longest of all phase 2 BCMA-CD3 bispecific antibody studies, including the outcome of pts who switched to the Q2W dosing, will be presented. Conclusion: These results support continued elranatamab development for pts with MM.

Abstract

Tiivistelmä. Tässä kandidaatintutkielmassa käsitellään lapsen kielenkehityksen vaikeuksia varhaiskasvatusiässä. Tutkielmassa vastataan kysymykseen, miten varhaiskasvatuksessa voidaan tunnistaa ja tukea lapsen kielenkehityksen vaikeuksia. Kielenkehityksen vaikeudet ovat yleisiä kehitysvaikeuksia varhaiskasvatusikäisillä lapsilla. Kielenkehityksen vaikeudet aiheuttavat lapselle usein ihmissuhdehaasteita sekä tunne- ja käytöshäiriöitä. Jotta lapsi, jolla on kielenkehityksen vaikeuksia, voisi osallistua yhdenvertaisesti muiden lasten kanssa varhaiskasvatukseen, olisi tärkeää saada mahdollisimman varhain käyttöön tukitoimia. Tutkielmani on laadullinen tutkimus, jonka toteutan kuvailevana kirjallisuuskatsauksena. Tutkimustulokseni ovat syntyneet aiemmin tutkitun tiedon pohjalta. Tutkimusaineisto pohjautuu kasvatustieteen teoriaan, mutta myös logopediaan ja esimerkiksi lääketieteeseen, sillä kielenkehityksen vaikeuksia on relevanttia käsitellä poikkitieteellisesti, jotta kielenkehityksen vaikeuksien diagnostisiin kriteereihin voidaan pureutua. Kielenkehityksen vaikeuksia voidaan huomioida varhaiskasvatuksessa oppimisympäristöissä, vuorovaikutustilanteissa sekä vanhempien ja varhaiskasvatuksen ammattilaisten välisessä yhteistyössä. Jos lapsella on kielenkehityksen vaikeuksia, on tärkeää ottaa käyttöön puhetta tukevat ja korvaavat kommunikaatiomenetelmät. Jotta kielenkehityksen vaikeuksia voidaan tunnistaa, on tärkeää tietää, miten lapsen kieli kehittyy tyypillisesti.

Abstract

Synopsis Generation of neutral atomic beam is under development for the ion-neutral merged-beam experiment at the RIKEN Cryogenic Electrostatic ion storage ring (RICE). The beam is produced by a photodetachment of a negative ion beam using a high power laser diode array with a wavelength of 808 nm. In order to increase the detachment efficiency, we have developed an optical cavity built with a planar mirror. In this presentation we will present the current status of the neutral atom beam source.

This is a report of a CMOS image sensor with a sub-pixel architecture having a pixel pitch of 3 um. The aforementioned sensor achieves both ultra-low random noise of 0.68e-rms and high dynamic range of 121 dB in a single exposure, further realizing LED flicker mitigation.

Abstract

Gene mutations were found in acute myeloid leukemia (AML) and their importance has been noted. To clarify the importance and stability of mutations, we examined gene mutations in paired samples at diagnosis and relapse of 34 adult AML patients. Five acquired gene mutations were detected at relapse. Of the 45 gene mutations at diagnosis, 11 of them were lost at relapse. The acquired mutations at relapse were all class I mutations as Fms-like tyrosine kinase 3 (FLT3) and rat sarcoma viral oncogene homolog (RAS) mutations. The disappeared mutations at relapse were 3 of 11 internal tandem duplications of FLT3 (FLT3-ITD) (27.3%), 3 of 3 FLT3 tyrosine kinase domain (FLT3-TKD) (100%), 3 of 13 Nucleophosmin 1 (23.1%) and 2 of 5 CCAAT/enhancer-binding protein-α (40%) mutations. However, epigenetics-modifying gene (DNMT3a, TET2 and IDH1/2) mutations had no change between diagnosis and relapse samples, and may become minimal residual disease marker. The frequency of FLT3-ITD at relapse in patients with DNMT3a mutation at diagnosis is significantly higher than those in patients without them (P=0.001). The clinical development of specific/selective or more potent analogues of somatostatin has made the evaluation of somatostatin receptor subtypes in archival specimens or 10% formalin-fixed and paraffin-embedded specimens of neuroendocrine tumor (NET) critical to conferring the maximum clinical benefits on NET patients. Immunohistochemistry of somatostatin receptor subtypes could contribute to profiling their expression patterns in these patients. Therefore, surgical pathologists are expected to be asked to immunostain and evaluate somatostatin receptor subtypes in NET specimens in the near future by clinicians. However, there are problems associated with immunohistochemistry of somatostatin receptor subtypes, in particular with their evaluation and interpretation. Therefore, both pathologists and clinicians involved in the care or management of NET patients should be aware of the advantages and limitations of immunohistochemical evaluation of somatostatin receptor subtypes in order to achieve the most efficient treatment outcome for patients.

Abstract

Systemic lupus erythematosus (SLE) patients are at high risk of developing osteonecrosis. This study utilized MRI to document the long-term natural history of asymptomatic osteonecrosis associated with corticosteroid therapy in SLE patients. Two hundred and one SLE patients treated with high-dose corticosteroids were prospectively observed from 1986 to 1997. The inclusion criterion was that patients had received periodic MRI examinations of all their hip and knee joints for ≥10 years. Joints that were already collapsed and symptomatic at the first examination were excluded. Five hundred and thirty-seven joints (251 hips and 286 knees) were identified in 144 patients, with a mean follow-up period of 13.6 years (range, 10—20 years) and a follow-up rate of 73%. Mean age of SLE onset was 26 years, and the mean highest oral corticosteroid dosage was 57 mg/day. Osteonecrosis developed in 238 (44%) of 537 joints. At final follow-up, 117 (49%) of these 238 joints demonstrated spontaneous repair in the necrotic area. Osteonecrosis completely disappeared in 21 joints. Enlargement of osteonecrosis was noted in 35 joints (15%) following increased steroid dosage because of SLE recurrence. Finally, 52 joints (22%) were collapsed. Spontaneous repair of asymptomatic osteonecrosis was observed, whereas enlargement occurred only after corticosteroid dosage increases. Lupus (2010) 19, 1307—1314.

Limiting the ingestion of protein is the fundamental idea in the diet therapy for patients with chronic renal failure. Two mutations involved in the content of major rice storage proteins useful for developing low easy-to-digest protein rice variety have been isolated. The glb1 mutation causes the deficiency of alpha-globulin, and the Lgc1 mutation reduces the glutelin content. By combining the glb1 and the Lgc1 mutations, it is possible to reduce the easy-to-digest protein content by approximately 50%. The Lgc1 mutation has been shown to be caused by a 3.5-kb deletion between the glutelin structural genes, GluB4 and GluB5, while the molecular basis of glb1 mutation has been less understood. PCR analysis of the glb1 mutation revealed a 62.8-kb deletion, including the structural gene of alpha-globulin. Based on these lines of information, we generated PCR markers that make it possible to detect the glb1 and Lgc1 mutations. Using those PCR markers, we genotyped F(2) plants segregating for the glb1 mutation and the Lgc1 mutation and confirmed the consistency of genotype and phenotype. Because the PCR marker sets can distinguish heterozygotes, they will be very useful in developing new varieties of low easy-to-digest protein rice.

長伐期施業等，人工林の高齢級化による下層植生現存量の変動を明らかにするために，施業履歴の明確なトドマツ林を21林分選択し，林内の下層植生現存量（樹高3 m以下）と光環境条件を調査した。下層植生直上の相対光強度（ISF）は5.2∼22.3%であり，ISFは上層林冠構成木の立木密度の逆数（Dsi）を用いて推定することができた。下層植生現存量は0.01∼4.3 Mg ha－1であり，また最大の葉面積指数（3.37 m2m－2）は落葉広葉樹林のそれに匹敵した。下層植生現存量は光環境条件，Dsiおよび除間伐作業後の経過年数に影響されていた。ISF 20%以下の下層植生地上部現存量は7 Mg ha－1に満たないと推定されたが，これは国内の既存の報告と一致した。このことは長期間施業が行われない場合でも下層植生現存量は上層林冠木現存量の0.01∼4.1%に過ぎず，下層植生の炭素貯留機能には大きな期待を持てないことが示唆された。しかしながら，既存の研究を概観すると，土砂流出防備等の水土保全機能に対しては十分に機能を発揮できると考えられた。

Abstract

The results of the application of the spectroscopic phase modulated ellipsometry (SPME) to a series of novel ternary compounds from anisotropic to polycrystalline are presented to show the high effectiveness of this optical technique with regard to the dielectric function of the new materials. Besides, SPME in specular reflection mode to study the polarization degree of the light reflected at specular angle is shown to be very informative in determination of the best conditions for optical measurements on rough surfaces such as, for example, surfaces of the polycrystalline ternary thiogallate compounds studied in this work. An incoherent approach relating the measured depolarization with the surface variance and optical constants of a material or thin film is proposed.

Abstract

Ce-doped CaGa2S4 and SrGa2S4 thin film electroluminescent (TFEL) devices were prepared for the first time on the basis of films deposited by flash evaporation method. Significant crystallization, stoichiometry improvement of the films and increase of photoluminescence intensity were found after annealing in H2S and O2 gas stream. EL spectra of the cells exhibited the characteristic double-band emission similar to that seen for Ce3+ activated CaGa2S4 and SrGa2S4 films under photon excitation. Applied voltage and frequency dependences of the electroluminescence were studied. Low voltage operation as low as 20 V was observed for these cells. Luminance of about 4 cd/m2 at 100 V operating voltage with 2.5 kHz frequency was achieved for the TFEL cell with films annealed in O2 gas stream. (© 2006 WILEY-VCH Verlag GmbH & Co. KGaA, Weinheim)

Abstract

Thin films were deposited on Si and fused quartz substrates by rf sputtering with EuGa2S4 target. The deposited films were annealed in the mixed atmosphere of S and He, which led crystallization of the film from amorphous phase. Photoluminescence of the annealed films, characteristic to the Eu2+ ion, was observed with room temperature quantum efficiency of 17%. Decay time constants at room temperature and liquid nitrogen temperature were measured to be 140 ns, and 430 ns, respectively. The latter value is close to the reported radiative lifetime of the EuGa2S4 crystal. Construction possibility of a surface-emitting laser is discussed with data on behaviour of excitation intensity dependent time-resolved spectra under pulsed laser excitation and on surface roughness of the film. Abstract

Although a Si(100) surface forms dimers to produce a stable structure, it contains defects such as steps which are thought to affect surface resistance. We measured the surface resistance of step-containing Si manufactured by an ordinary process, and discovered that Si exhibits angular characteristics (anisotropy). After performing measurements at 10° intervals over two revolutions of 360°, we found that the results are reproducible between the first and second turns, that the resistance is periodic with periods of 180° and 360°, and that the sheet resistances measured by the four-point-probe method are 7–13-fold greater than those calculated from the bulk resistance and thickness of the samples. Similar measurements were performed on step-free Si surfaces, which were found to exhibit almost no periodicity and had a sheet resistance is of 4–10-fold the bulk value. It was suggested that the periodicity of resistance is caused by the presence of many steps on the Si surface.

Abstract

Polycrystalline La1-xCaxMnO3-δ thin film was formed on a Si substrate coated with SiO2 by rf-sputtering in a 100% oxygen atmosphere. A sintered target with almost the same composition as the objective film was used for sputtering. In this study, we analyzed the deposited film's composition, crystallization, specific resistance and temperature coefficient of resistance at room temperature. Results show that the film's composition could be controlled by the target component : when the substrate temperature was high, it was possible to obtain a film with superior crystallization and lower specific resistance. Results also indicate that the temperature coefficient of resistance was influenced little by the substrate temperature.

Abstract

A thin Ag film was formed on an Si(1 0 0) substrate at 200 °C in UHV, and the Ag film resistance was measured up to a 15-nm thickness by the four-terminal method. The same measurement was performed using an H terminated Si(1 0 0) substrate, where measurements were carried out at 200 °C. In the case of the Si/Ag system, the resistance change showed a regular oscillation period for every 0.56 nm of Ag film thickness up to 2 nm. This oscillation period corresponds to that of an Ag dimer’s length, and the same oscillation period was obtained for different substrates. This phenomenon suggests that electron conduction is mainly limited to very thin surfaces with an Si/Ag interface and to each Ag island. An Ag island works as a quantum dot (QD) whose conduction electron density changes periodically depending on the island’s dimensions. In the case of the Si/H/Ag system, H appears to block the electron coupling between Ag and Si. Here, oscillation in resistance change also occurs, but for every 0.25 nm of Ag film thickness up to 1 nm.

Abstract

A study of the liquid phase epitaxial growth of InxGa1−xAs (x = 0.06) layers on different types of patterned GaAs(100) substrates has been carried out. The dependence of growth morphology on the stripe orientation of the star patterned trench substrate has been observed. Pyramid layers were grown in the stripes oriented along the 001 direction. Broken tent structures formed along the 012 direction. Pyramidal structured layers looked to grow faster than the tent-like and broken tent-like structures. In order to analyse the hollow pyramid structure growth in detail, they were grown on circular trench substrates for different periods of time. Hollow pyramidal structures of InGaAs have been grown on circular patterned trench substrates. Effective defect filtration can be realized in this kind of growth of hollow pyramidal structures. The formation mechanism of the hollow pyramid structured layers has been studied in detail.

High quality In 1− x Ga x As ( x =0.2) layers were grown on composition-converted InAs(1 1 1) patterned substrates by liquid phase epitaxy. Epitaxial lateral overgrown In 1− x Ga x As ( x =0.2) layer with flat and mirror-like surface was obtained on trenchless substrates. Clean bridge layer of In 1− x Ga x As ( x =0.2) was not formed when the trench substrate was used. When the trench substrate with SiN x deposition at the trench bottom was used, In 1− x Ga x As ( x =0.2) bridge layer resulted. The etch pit density of the bridge layer was found to be low. Ga and In contents of the In 1− x Ga x As ( x =0.2) epitaxial layers grown on all types of composition-converted InAs substrates were found to be uniform.

Abstract

When distinguishing whether a face displays a certain emotion, some regions of the face may contain more useful information than others. Here we ask whether people differentially attend to distinct regions of a face when judging different emotions. Experiment 1 measured eye movements while participants discriminated between emotional (joy, anger, fear, sadness, shame, and disgust) and neutral facial expressions. Participant eye movements primarily fell in five distinct regions (eyes, upper nose, lower nose, upper lip, nasion). Distinct fixation patterns emerged for each emotion, such as a focus on the lips for joyful faces and a focus on the eyes for sad faces. These patterns were strongest for emotional faces but were still present when viewers sought evidence of emotion within neutral faces, indicating a goal-driven influence on eye-gaze patterns. Experiment 2 verified that these fixation patterns tended to reflect attention to the most diagnostic regions of the face for each emotion. Eye movements appear to follow both stimulus-driven and goal-driven perceptual strategies when decoding emotional information from a face.

SummaryWhat is known and objective This study aimed to elucidate the pharmacokinetics of erlotinib in Japanese patients with advanced non-small cell lung cancer (NSCLC) and to investigate the relationship between erlotinib exposure and the occurrence of interstitial lung disease (ILD)-like events. Methods Population pharmacokinetics analysis was performed using nonlinear mixed-effects modelling software (NONMEM) based on 348 plasma samples from 97 patients obtained in two phase II clinical studies. Individual empirical Bayesian estimates (EBEs) of apparent oral clearance (CL/F) and Cmax were compared between the patients who developed and did not develop ILD-like events. Results A 1-compartment model with first-order absorption and first-order elimination was used to describe the plasma concentrations of erlotinib. The estimated population pharmacokinetics parameters were as follows: 4·71 L/h for CL/F, 163 L for apparent volume of distribution (Vc/F) and 1·97 h−1 for absorption rate constant (Ka). Total bilirubin (TBIL) and alpha 1-acid glycoprotein (AGP) were identified as statistically significant covariates for CL/F. No differences in CL/F and Cmax were observed between the patients with ILD-like events and those without ILD-like events. What is new and conclusions A population pharmacokinetics model of erlotinib was developed and validated in Japanese patients. There was no relationship between exposure of erlotinib before the occurrence of ILD-like events and the occurrence of ILD-like events when erlotinib was administered at the same dosage. The high plasma concentration of erlotinib reported in patients after the onset of ILD-like events may be explained by CL/F decrease which occurs along with increasing levels of AGP which was identified as a covariate for CL/F.

Abstract

Acute and chronic rejection impact distinct compartments of cardiac allografts. Intramyocardial mononuclear cell infiltrates define acute rejection, whereas chronic rejection affects large arteries. Hearts transplanted from male to female C57BL/6 mice undergo acute rejection with interstitial infiltrates at 2 weeks that resolve by 6 weeks when large arteries develop arteriopathy. These processes are dependent on T cells because no infiltrates developed in T cell deficient mice and transfer of CD4 T cells restored T cell as well as macrophage infiltrates and ultimately neointima formation. Markers of inflammatory macrophages were upregulated in the interstitium acutely and decreased as markers of wound healing macrophages increased chronically. Programmed cell death protein, a negative costimulator, and its ligand PDL1 were upregulated in the interstitium during resolution of acute rejection. Blocking PDL1:PD1 interactions in the acute phase increased interstitial T cell infiltrates. Toll Like Receptor 4 and its endogenous ligand hyaluronan were increased in arteries with neointimal expansion. Injection of hyaluronan fragments increased intragraft production of chemokines. Our data indicate that negative co-stimulatory pathways are critical for the resolution of acute interstitial infiltrates. In the arterial compartment recognition of endogenous ligands including hyaluronan by the innate toll like recepetors may support the progression of arteriopathy.

Abstract

An attempt has been made to further improve the quality of In x Ga 1− x As ( x =0.06) bridge layers on GaAs (1 1 1)B trench substrates by depositing SiN x on the inner surfaces of the trenches. In x Ga 1− x As ( x =0.06) layers grown in this way were found to exhibit better quality, as evidenced by microscopic photoluminsecence measurements. The trapezoidal SiN x film of large area deposited over the inner surface of the trench was found to be effective in reducing the dislocation content of the bridge layer. The minimum and maximum diameters of the trenches on GaAs(1 1 1)B substrates, keeping other parameters constant, under which the growth of In x Ga 1− x As bridge layers is possible have been determined.

Abstract

Two isomers of Dy@C 82 were separated by high performance liquid chromatography (HPLC), and their UV–VIS absorption spectra were measured to characterize these isomers. The crystalline powder of Dy@C 82 was obtained by removing the solvent (toluene) at 250°C under vacuum. The X-ray diffraction pattern can be indexed with fcc crystal lattice, as that in La@C 82 . The lattice constant a at 298 K, 15.86(1) A, is close to that of La@C 82 , 15.78 A. The distances between Dy and the first and second nearest C atoms are determined to be 2.52(2) and 2.86(2) A, respectively, on the basis of Dy L III -edge EXAFS. The XANES shows that the valence of the Dy atom in Dy@C 82 is +3.

Abstract

InxGa1-xAs (x = 0.06) epitaxial lateral overgrown (ELO) layers were grown on (111)B GaAs patterned substrates covered with SiNx masks by liquid phase epitaxy. The thickness of the ELO layer with a { 111} A growth front was found to decrease as the growth progressed. On the other hand, for the ELO layer with a { 111} B growth front, this kind of decrease of thickness was found to be very small. When the layers with different growth fronts ({ 111} A and { 111} B) coalesced, dislocations were found to be generated. In the case of the coalesced ELO layers which were in constant contact with the basal SiNx mask, the surface became concave due to the decrease of thickness of the layers. This problem of decrease of thickness was not observed when a large enough void structure was present in the grown ELO layers of InGaAs.
