The most virulent human malaria parasite, Plasmodium falciparum, has a complex life cycle between its human host and mosquito vector. Each stage is driven by a specific transcriptional program, but with a relatively high ratio of genes to specific transcription factors, it is unclear how genes are activated or silenced at specific times. The P. falciparum genome is relatively euchromatic compared to the mammalian genome, except for specific genes that are uniquely heterochromatinized via HP1. There seems to be an association between gene activity and spatial organization; however, the molecular mechanisms behind genome organization are unclear. While P. falciparum lacks key genome-organizing proteins found in metazoans, it does have all core components of the cohesin complex. In other eukaryotes, cohesin is involved in sister chromatid cohesion, transcription, and genome organization. To investigate the role of cohesin in P. falciparum, we combined genome editing, mass spectrometry, chromatin immunoprecipitation and sequencing (ChIP-seq), and RNA sequencing to functionally characterize the cohesin subunit Structural Maintenance of Chromosomes protein 3 (SMC3). SMC3 knockdown in early stages of the intraerythrocytic developmental cycle (IDC) resulted in significant upregulation of a subset of genes involved in erythrocyte egress and invasion, which are normally expressed at later stages. ChIP-seq of SMC3 revealed that over the IDC, enrichment at the promoter regions of these genes inversely correlates with their expression and chromatin accessibility levels. These data suggest that SMC3 binding helps to repress specific genes until their appropriate time of expression, revealing a new mode of stage-specific, HP1-independent gene repression in P. falciparum.; Le parasite du paludisme humain le plus virulent, Plasmodium falciparum, a un cycle de vie complexe entre son hôte humain et le moustique vecteur. Chaque étape est pilotée par un programme de transcription spécifique, mais avec un rapport relativement élevé de gènes à des facteurs de transcription spécifiques, on ne sait pas comment les gènes sont activés ou réduits au silence à des moments spécifiques. Le génome de P. falciparum est relativement euchromatique par rapport au génome des mammifères, à l'exception de gènes spécifiques qui sont uniquement hétérochromatinisés via HP1. Il semble y avoir une association entre l'activité des gènes et l'organisation spatiale ; cependant, les mécanismes moléculaires derrière l'organisation du génome ne sont pas clairs. Chez d'autres eucaryotes, la cohésine est impliquée dans la cohésion, la transcription et l'organisation du génome des chromatides soeurs. Pour étudier le rôle de la cohésine dans P. falciparum, nous avons combiné l'édition du génome, la spectrométrie de masse, l'immunoprécipitation et le séquençage de la chromatine (ChIP-seq) et le séquençage de l'ARN pour caractériser fonctionnellement la sous-unité cohésine Structural Maintenance of Chromosomes protein 3 (SMC3). L'inactivation de SMC3 aux premiers stades du cycle de développement intraérythrocytaire (IDC) a entraîné une régulation positive significative d'un sous-ensemble de gènes impliqués dans la sortie et l'invasion des érythrocytes, qui sont normalement exprimés à des stades ultérieurs. ChIP-seq de SMC3 a révélé que sur l'IDC, l'enrichissement au niveau des régions promotrices de ces gènes est inversement corrélé à leurs niveaux d'expression et d'accessibilité à la chromatine. Ces données suggèrent que la liaison de SMC3 aide à réprimer des gènes spécifiques jusqu'à leur moment d'expression approprié, révélant un nouveau mode de répression génique spécifique au stade et indépendant de HP1 chez P. falciparum.

UIDB/00749/2020 UIDP/00749/2020 Ao longo dos séculos XIV e XV, a fiscalidade aplicada pelo poder régio em Lisboa foi um importante fator de articulação entre a Coroa e edilidade lisboeta. Uma articulação, por sinal, detetável a diversos níveis, designadamente: a denúncia dos abusos praticados pelos oficiais régios que intervinham no processo de recolha fiscal; o apoio financeiro dispensado pela instituição camarária à Coroa; e a atribuição por parte do poder real de privilégios de natureza fiscal em retribuição pelos serviços prestados pelo concelho. Neste sentido, o presente texto visa dar a conhecer as dinâmicas de conflito e de cooperação entre o poder régio e o poder concelhio e apresentar um balanço da relação entre os dois poderes a partir do observatório da fiscalidade régia, tendo em conta, por um lado, o protagonismo económico, político e social da cidade de Lisboa no contexto da rede urbana medieval portuguesa, traduzido no seu estatuto de cabeça do reino, incontestável na cronologia aqui privilegiada; e, por outro lado, o apertado controlo exercido pela Coroa sobre a gestão deste núcleo urbano e a forte dependência da elite camarária lisboeta em relação à realeza e ao oficialato régio. Over the fourteenth and fifteenth centuries, taxation applied by the royal power in Lisbon was an important factor in the articulation between the Crown and the city council of Lisbon. This articulation was detectable at different levels, namely: the abuses practiced by the royal officials who intervened in the tax collection process; the financial support provided by the municipality to the Crown; and the attribution of fiscal privileges given by the royal power in return for the services provided by the city. Therefore, this paper aims to explore the dynamics of conflict and cooperation between the royal and the municipal authorities, as well as to offer an insight into the relationship between the two powers from the viewpoint of royal taxation, taking into account, on the one hand, the economic, political and social role of Lisbon in the context of the Portuguese medieval urban network, reflected in its status as head of the kingdom, indisputable in this chronology; and, on the other hand, the control exercised by the Crown over this urban center and the dependence of Lisbon's power elite on the monarchy. publishersversion published

UIDB/00749/2020 UIDP/00749/2020 Pelas suas dimensões e peso demográfico, Lisboa era a principal cidade do Portugal medieval e, como tal, aquela cujas necessidades de abastecimento eram mais difíceis de satisfazer. Sobre os bens essenciais de consumo (pão, carne, peixe, legumes, vinho, etc.), importados e/ou transacionados em Lisboa, recaíam encargos fiscais que, ao longo do período medieval, conheceram uma nítida evolução, sendo que o reinado de D. João I foi a este nível um ponto de viragem. De facto, no decurso de quase meio século de governação, D. João I reformou o quadro da fiscalidade régia em Lisboa, quer pela concessão de privilégios de isenção, quer pela institucionalização de novos impostos, designadamente as sisas gerais e a dízima nova do pescado. Para além disso, o reinado deste monarca foi também um período de frequentes carestias, que obrigaram à adaptação do fisco régio de modo a salvaguardar o abastecimento desta cidade. Com efeito, o presente estudo presta-se a dar a conhecer o aparelho fiscal montado pela Coroa em torno do abastecimento de Lisboa, com enfoque sobre os produtos alimentares, e, de resto, presta-se a explicar de que forma o abastecimento desta cidade condicionou a política fiscal régia, ao tempo de D. João I. Given its dimensions and its demographic weight, Lisbon was the main city of Medieval Portugal, and also the one whose supplying needs were harder to accommodate. Essential goods, such as cereal, meat, fish, vegetables, wine, etc., that were either imported or traded in Lisbon were taxed by the Crown and, at the time of D. João I, royal fiscality undertook significant changes, brought about by the fiscal privileges granted to Lisbon’s municipality and by the collection of new taxes. Furthermore, the reign of D. João I was also a period of frequent shortages that forced royal fiscality to adapt to ensure that Lisbon’s supplying needs were met. Therefore, the following text aims to present the fiscal apparatus established by the Crown in order to tax Lisbon’s supply system of essential goods, and also to explain how supplying this city conditioned royal fiscal policy. publishersversion published

Após a conquista da cidade de Lisboa, em 1147, o poder régio procurou assegurar o aproveitamento fiscal deste núcleo urbano, através da organização de um complexo aparato fiscal, cuja constituição teve início no rescaldo da conquista. Todavia, só mais tarde, em 1179, é que se procedeu ao seu enquadramento normativo, sob a forma de uma carta de foral. A partir da análise deste texto é possível fazer uma aproximação ao elenco fiscal estabelecido nesta cidade, que, ao longo das centúrias seguintes, foi objeto de sucessivas alterações, quer por via da incorporação de novas soluções fiscais, quer por via do recurso ao poder de exceção do monarca, traduzido na concessão de privilégios. Neste sentido, a presente dissertação de mestrado, que tem como objeto de estudo a fiscalidade régia na Lisboa do período medieval, com enfoque nos séculos XIV e XV, presta-se justamente a dar a conhecer a constituição e evolução do quadro fiscal aplicado pelo poder régio nesta cidade, atendendo, por um lado, ao seu enquadramento ao nível do sistema fiscal régio, e por outro lado, à sua especificidade, subjacente ao caráter excecional do espaço de observação escolhido. After the conquest of Lisbon, in 1147, the royal power sought to ensure the fiscal use of this city, through the organization of a complex fiscal apparatus, that was first established in the aftermath of the conquest. However, it was only later, in 1179, that Lisbon's first normative framework was put in place, in the form of a charter. From the analysis of this text, it is possible to make an approximation to the tax list established in this city, which, over the following centuries, was subject to successive changes, either through the incorporation of new tax solutions or through the use of the monarch’s power of exception, translated into the granting of privileges. The present master's dissertation, whose object of study is the royal taxation in Medieval Lisbon , focusing on the 14th and 15th centuries, lends itself precisely to make known the organization and evolution of the fiscal framework applied by the monarchy in this city, taking into account, on the one hand, its framing at the level of the royal tax system, and on the other hand, its specificity, underlying the exceptional character of the chosen space of observation.

This article applies a dialogical analysis to the change processes involved in moving from engagement with to disengagement from an armed militant group, as well as from radicalization to deradicalization. The findings underline the interplay between different push and pull factors at individual, organizational, and societal levels that played a role in the already mentioned processes in three periods of time—engagement with, life within, and disengagement from an armed organization. The dialogical framework conceptualizes the development trajectory as relationships between a variety of positions of the self (I-positions), which generate different personal meanings involved in processes of disengagement and deradicalization.

Este capítulo apresenta os pressupostos teóricos e as aplicações clínicas da terapia narrativa da re-autoria. Teoricamente, a terapia narrativa da re-autoria é um modelo influenciado por autores de diversas áreas, como a análise literária, a antropologia ou a epistemologia. Os pressupostos deste modelo organizam-se em torno do conceito de narrativa, enfatizando como as histórias que contamos sobre nós e o mundo se podem tornar limitadoras e pouco adaptativas e como a psicoterapia pode ajudar os clientes a identificar as fontes de poder dessa narrativa problemática e a construir narrativas alternativas mais satisfatórias. Clinicamente, a terapia narrativa da re-autoria propõe-se devolver ao cliente o papel de autor, e não de mero ator ou de espectador, da sua narrativa de vida. Neste modelo, o processo de psicoterapia compreende três fases: a desconstrução, a reconstrução e a consolidação. Através de diferentes estratégias terapêuticas, como a externalização ou a identificação e expansão de resultados únicos, o cliente e o terapeuta perscrutam as pressuposições da narrativa problemática que trouxe o cliente à terapia, contribuindo para a emergência e enraizamento de uma narrativa alternativa e preferencial na esfera da experiência do cliente, info:eu-repo/semantics/publishedVersion

Enquadramento: Os Cuidados Paliativos, sendo cuidados de saúde exigentes e humanizados, intervêm sobretudo no sofrimento das pessoas com doenças graves e/ou avançadas e irreversíveis, maximizando, quanto possível, a sua qualidade de vida e dignidade. Com o mesmo propósito intervém a enfermagem de reabilitação num cuidar especializado, no sentido de otimizar o conforto e bem-estar da pessoa, porém nem sempre reconhecido pelos seus pares. Assim pretendemos conhecer a perceção dos enfermeiros sobre a intervenção do enfermeiro especialista em enfermagem de reabilitação nas pessoas internadas em Unidades de Cuidados Paliativos. Métodos: Estudo qualitativo, com recurso à análise de conteúdo. Os dados foram colhidos junto de uma amostra de 13 enfermeiros, sendo 12 do sexo feminino e 1 do sexo masculino. Eram todos licenciados em enfermagem e exerciam a sua atividade profissional em Unidades de Cuidados Paliativos. Para a recolha de dados recorreu-se a uma entrevista semiestruturada. Resultados: Observámos que os enfermeiros da nossa amostra têm uma perceção muito positiva acerca da intervenção do enfermeiro especialista em reabilitação nas Unidades de Cuidados Paliativos, considerando-o como agente facilitador na satisfação das necessidades do doente, destacando a sua intervenção sobretudo aos níveis da cinesiterapia respiratória, promoção/preservação da autonomia/independência do doente, mobilização, treino da deglutição/disfagia e massagem. Conclusões: A visão dos enfermeiros sobre a intervenção do enfermeiro especialista em reabilitação nas unidades de Cuidados Paliativos é positiva salientando a importância da sua intervenção na manutenção das capacidades funcionais dos doentes, a prevenção de riscos/complicações e a promoção de intervenções terapêuticas que objetivem a melhoria das funções residuais e reduzam o impacto das incapacidades resultantes da doença. Abstract Background: Palliative Care, being a demanding and humanized health care, mainly involves the suffering of people with serious and / or advanced and irreversible diseases, maximizing, as much as possible, their quality of life and dignity. With the same purpose, rehabilitation nursing intervenes in a specialized care, in order to optimize the comfort and well-being of the person, but not always recognized by their peers. Thus we intend to know the nurses' perception about the intervention of the nurse specialist in rehabilitation nursing in the people hospitalized in Palliative Care Units. Methods: Qualitative study using content analysis. Data were collected from a sample of 13 nurses, 12 females and 1 males. They were all licensed in nursing and exercised their professional activity in Palliative Care Units. For the collection of data a semi-structured interview was used. Results: We observed that the nurses in our sample have a very positive perception about the intervention of the specialist nurse in rehabilitation in the Palliative Care Units, considering it as a facilitating agent in the patient's needs, emphasizing their intervention mainly at the levels of kinesitherapy respiratory therapy, promotion / preservation of autonomy / independence of the patient, mobilization, deglutition / dysphagia training and massage. Conclusions: The nurses' view on the intervention of the nurse specialist in rehabilitation in the Palliative Care units is positive, emphasizing the importance of their intervention in the maintenance of the functional capacities of the patients, the prevention of risks / complications and the promotion of therapeutic interventions that aim to improve and reduce the impact of disability resulting from the disease.
