Abstract

A significant challenge for all biosensor systems is to achieve high assay sensitivity and specificity while minimizing sample preparation requirements, operational complexity, and sample-to-answer time. We have achieved multiplexed, unamplified, femtomolar detection of both DNA and proteins in complex matrices "including whole blood, serum, plasma, and milk" in minutes using as few as two reagents by labeling conventional assay schemes with micrometerscale magnetic beads, and applying fluidic force discrimination "FFD". In FFD assays, analytes captured onto a microarray surface are labeled with microbeads, and a controlled laminar flow is then used to apply microfluidic forces sufficient to preferentially remove only nonspecifically bound bead labels. The density of beads that remain bound is proportional to the analyte concentration and can be determined with either optical counting or magnetoelectronic detection of the magnetic labels. Combining FFD assays with chip-based magnetoelectronic detection enables a simple, potentially handheld, platform capable of both nucleic acid hybridization assays and immunoassays, including orthogonal detection and identification of bacterial and viral pathogens, and therefore suitable for a wide range of biosensing applications., Biosensors and Bioelectronics, Volume 23, Issue 2, 30 September 2007, Pages 191-200

A unique solid-state giant magnetoresistive (GMR) biosensor is the centerpiece of an integrated sample processing system for the detection of target DNA molecules through hybridization. Abstract

The nature of optical constants and their measurement by reflection or absorption techniques in the extreme ultraviolet and soft X-ray spectral region from 30 to 3000 eV is discussed with emphasis on mirror design. Sources of optical constant data are mentioned and reflectance measurements for SiC and Kanigen between 40 and 200 eV are reported.

36. The density of helium in bubbles in implanted materials: Theoretical interpretation of VUV absorption and EELS spectroscopy

Abstract

The shape of the He resonance absorption line of He bubbles in metals is discussed in terms of He-metal and He-He interactions. For bubble radii larger than 10 Å the metal matrix effects are found to be negligible. For small vacancy clusters there is as yet no quantitative calculation of the role of the metal conduction electrons in determining the position and width of the He resonance excitation.The He-He interactions are treated within static line broadening theory which assumes pairwise additivity of the interactions. These are taken from ab initio quantum mechanical calculations of 2p excimer potential energy curves due to Guberman and Goddard and to Gupta and Matsen. The radial pair distribution function required by our statistical line shape calculation is computed from two theoretical models of the dense fluid: either the exact solution of Percus-Yevick's equation for hard spheres or a Molecular Dynamics computer simulation based on an accurate pair potential. The two approaches give very similar results. The blue shift of the resonance line is found to depend nearly linearly on density but our value for the slope differs substantially from the prediction of a model band structure calculation performed by the Jülich group in the framework of density functional theory.

37. The density of helium in bubbles in implanted materials: Results from VUV absorption and eel spectroscopy

Abstract

The novel application of vacuum ultra-violet absorption spectroscopy and electron energy loss spectroscopy to helium bubbles in metals is presented. These measurements, carried out on thin aluminium films containing different concentrations of helium and various bubble size distributions, were aimed at determining the density (and thus pressure) of helium in bubbles by observing the shift and broadening of the IS-2P transition in the helium. The data coupled with a theoretical model developed by the authors (see following paper) indicate densities as high as 1023 He cm-3 for specimens containing small bubbles. Data are also presented on the effect that annealing and cooling have on these spectra. The annealing experiments give rise to fairly complex changes in absorption peak structure but with a general shift towards the unperturbed resonance line. The cooling experiment gives rise to a further shift and a narrowing of the absorption spectrum on cooling to 77 K which is tentatively identified as the liquid/solid transition in the helium. Finally, fluorescence spectrum of an Al/He specimen excited with low energy electrons is presented.

38. Higher-order suppression in an on-blaze plane-grating monochromator

Abstract

The transmittance of an on-blaze plane-grating monochromator has been calculated as a function of wavelength for different spectral orders. The calculations show that higher, undesirable orders can be suppressed by a judicious selection of grating blaze angle and coating material. However, for intense sources, such as a storage ring operating at high power and current, higher-order suppression due to blaze angle and coating material may have to be augmented by using transmitting filters or spectrally selective detectors or both.

Abstract

Potential multilayer grating applications for the wavelength region from 6 to 350 A are discussed. Multilayer bandwidth and efficiency as affected by stack construction as well as the effects of roughness and corrections to Bragg's law are considered. Grating performance is reviewed with attention to the limitations to high resolving powers and spectral purity in fabrication and instrument design. Combined multilayer grating performance is discussed for both broad and narrow band coatings. Designs for: (a) 10-40 A 1,000,000 resolution, broad-band spectrograph/monochromator are explored.
