Background: Triple-negative breast cancer (TNBC) is a complex and molecularly heterogeneous entity, with the poorest outcome compared with other breast cancer subtypes. Previously, we developed a TNBC 3-gene score with a significant prognostic capability. This study aims to test the 3-gene score in the different TNBC subtypes. Methods: Data from 204 TNBC patients treated with neoadjuvant chemotherapy were retrieved from public datasets and pooled (GSE25066, GSE58812, and GSE16446). After removing batch effects, cases were classified into Lehman’s TNBC subtypes and then the TNBC 3-gene score was used to evaluate the risk of distant recurrence in each subgroup. In addition, the association with tumor-infiltrating lymphocyte (TILs) levels was evaluated in a retrospective group of 72 TNBC cases. Results: The TNBC 3-gene score was able to discriminate patients with different risks within the pooled cohort (HR = 2.41 for high vs. low risk; 95%CI: 1.50–3.86). The score showed predictive capability in the immunomodulatory subtype (HR = 4.16; 95%CI: 1.63–10.60) and in the mesenchymal stem-like subtype (HR = 18.76; 95%CI: 1.68–208.97). In the basal-like 1, basal-like-2, and mesenchymal subtypes, the observed differential risk patterns showed no statistical significance. The score had poor predictive capability in the luminal androgen receptor subtype (p = 0.765). In addition, a low TNBC 3-gene score was related to a high level of TIL infiltration (p < 0.001). Conclusions: The TNBC 3-gene score is able to predict the risk of distant recurrence in TNBC patients, specifically in the immunomodulatory and mesenchymal stem-like subtype. Despite a small sample size in each subgroup, an improved prognostic capability was seen in TNBC subtypes with tumor-infiltrating components.

Objective: To describe the results of transperineal prostate biopsy by fusion of cognitive magnetic resonance imaging and ultrasound in the detection of cancer. Materials and methods: A retrospective case series study conducted at the Delgado Clinic between July 2019 and October 2021. Patients with digital prostate examination and/or abnormal Prostatic Specific Antigen and prostate MRI images with category lesions were included. PI-RADS 4 or 5. Excluding patients with coagulation disorders, rectal injuries, impossibility for abduction, comorbidities for anesthesia. We recorded clinical and demographic characteristics, prostate-specific antigen data, prostate volume, PI-RADS lesions, cancer rates, and complications. Results: 18 patients were evaluated, with a mean age of 69.33 ± 7.67 years. The mean prostate specific antigen was 14.94 ± 12.42 ng/mL. The mean prostate volume was 57.72 ± 28.68 cc. At the digital examination of the prostate, 16/18 (88.88%) patients had suspected cancer. Biopsies in patients with PI-RADS lesions 4 in 6/18 (33.33%) and 5 in 12/18 (66.67%). Biopsy with 19 cores in 6/18 (33.33%) and 16 cores in 12/18 (66.67%) patients. Cancer patients 15/18 (83.33%), all acinar adenocarcinoma, with Gleason 6 in 2/18 (11.11%) and Gleason ≥ 7 in 13/18 (72.22%) patients. Mild complications such as hematuria 1/18 (5.6%), terminal dysuria 8/18 (44.4%) and perineal discomfort 7/18 (38.8%) and none infectious. Conclusions: transperineal prostate biopsy by fusion of cognitive magnetic resonance imaging and ultrasound is feasible, safe, with high positivity rates and without infections. Objetivo: Describir los resultados de la biopsia prostática transperineal por fusión de la resonancia magnética cognitiva y ultrasonido en la detección del cáncer. Materiales y métodos: Estudio de serie de casos, retrospectivo, realizado en Clínica Delgado entre julio del 2019 y octubre del 2021. Se incluyó pacientes con examen digital de próstata y/o Antígeno Prostático Específico anormal e imágenes de Resonancia Magnética de próstata con lesiones categoría PI-RADS 4 o 5. Excluyendo a pacientes con trastorno de coagulación, lesiones rectales, imposibilidad para abducción, comorbilidades para anestesia. Registramos las características clínicas, demográficas, datos de antígeno prostático específico, volumen de la próstata, lesiones PI-RADS, tasas de cáncer, complicaciones. Resultados: Se evaluaron18 pacientes, con edad media de 69,33 ± 7,67 años. La media del antígeno prostático específico de 14,94 ± 12,42 ng/mL. La media del volumen de próstata de 57,72 ± 28,68 cc.Al examen digital de la próstata 16/18 (88.88 %) pacientes tenían sospecha de cáncer. Se hicieron biopsias con 19 núcleos en pacientes cuya RMNmP tenían lesiones con categoría de PI-RADS 4 en 6/18 (33,33 %) de los cuales 3 resultaron positivas a Adenocarcinoma y otras 3 negativas a cáncer. Biopsia con 16 núcleos en PIRADS 5 en 12/18 (66,67 %) pacientes, resultando todas positivas a cáncer. Adenocarcinoma Acinar en 15/18 (83,33 %) con Gleason 6 en 2/18 (11,11 %) y Gleason ≥ 7 en 13/18 (72,22 %) pacientes. Complicaciones leves como hematuria 1/18 (5,6 %), disuria terminal 8/18 (44,4 %) y molestia perineal 7/18 (38,8 %) y ninguna infecciosa. Conclusiones: la biopsia prostática transperineal por fusión de imágenes de resonancia magnética cognitiva y ultrasonido es factible, segura, con tasas importantes de positividad y sin infecciones.

The situation of LGBT+ people has long been characterized as marginal, socially and textually. This is notably true of the history of LGBT+ cinematic representation. However, marginality takes different forms, at the level of characterization (notably in terms of functionality and vividness) and in terms of structure (sometimes really incidental, sometimes serving to create the contours of centrality or to underpin or undermine it). These distinctions are discussed through the case of La dolce vita, a film in which gay characters are apparently extremely marginal but to which they are in fact integral. Comparison is drawn with the representation of ethnicity in the film and of gay characters in a parody of it.

Ambient sound and silence largely function in film to hold narrative space and time in place. They are rarely allowed just to be—they must be made to speak. La Captive (Belgium/France 2000, dir. Chantal Akerman) is an interrogation of this impulse to interrogate aural opacity and absence, in the context of a male need to probe the female. Here, a woman’s silence becomes a refusal to be known by a man and the clacking of high heels becomes maddeningly erotic and elusive, while verbal interrogation leads nowhere and florid music functions as an excess of misleading expressivity. La Captive is structured both thematically and formally by listening, and it suggests the way that the latter, like looking, organizes the gendered epistemological drive of narrative and perception in film.

The pathology has evolved together with scientific and technological advances, perhaps being the oncologydiscipline where these advances have been incorporated more quickly into daily practice. Molecular pathology stems from advances in the understanding of the human genome and the subsequent description of cancer genomes, as well as from the rapid simplification and cheapening of molecular techniques. Molecular pathology offers not only a more comprehensive description of the characteristics of cancer, but also provides the treating physician with a range of cancer weaknesses that are therapeutic targets. El análisis de la proporción y cantidad de células sanguíneas es una forma muy confiable de evaluar el estado de salud en humanos y otras especies. El objetivo fue estandarizar una metodología para la cuantificación rápida de poblaciones de leucocitos sanguíneos de Oncorhynchus mykiss (trucha arcoíris). Se procesaron muestras de leucocitos obtenidos de la sangre de un grupo de truchas juveniles saludables mantenidas en el laboratorio. Las muestras fueron analizadas empleando el minicitómetro Scepter™ y los resultados fueron contrastados con los obtenidos por citometría de flujo. Los histogramas generados por ambos métodos mostraron un máximo correspondiente a la población de linfocitos/trombocitos y otro a la de monocitos/neutrófilos. No se encontraron diferencias significativas entre los resultados de ambos métodos (P > 0,05). El método con el minicitómetro se utilizó para evaluar muestras de truchas procedentes de una piscigranja, el perfil obtenido mostró la disminución significativa de neutrófilos compatible con los signos clínicos de enfermedad observados en los especímenes evaluados. Estos resultados indican que la metodología aplicada es válida y útil para determinar las concentraciones y proporciones celulares en muestras de sangre en trucha arcoíris y permiten el análisis rápido de su estado de salud. The analysis of the ratio and number of blood cells is a very reliable way to assess the health status in humans and other species. The aim was to standardize a methodology for rapid quantification of blood leukocytes in rainbow trout Oncorhynchus mykiss. Samples of blood leukocytes obtained from a group of healthy juvenile trout maintained in the laboratory were processed. The samples were analyzed using a Scepter™ minicytometer, the results were compared with those obtained by flow cytometry. Histograms generated by both methods showed a peak corresponding to the population of lymphocytes/thrombocytes and one of monocyte/neutrophil. Non-significant differences were found between the results of both methods (P > 0.05). The method employing minicytometer was used to evaluate samples of trout from a fish farm, the profile obtained showed a significant decrease in the neutrophils consistent with clinical signs of disease observed in the tested specimens. These results indicate that the methodology is valid and useful in determining the concentrations and cell proportions in blood samples of rainbow trout and allow rapid analysis of its health status.

The present work involves an extensive analytical and toxicological description of a recently identified mixed halogenated carbazole found in sediment samples, 1,8-dibromo-3,6-dichloro-9H-carbazole (BCCZ). Concentrations and the relative effect potency (REP) were calculated for the target BCCZ in a set of stream sediments collected in 2008 in Ontario, Canada. The flight control teams of two low Earth orbit missions at EUMETSAT present an overview of the automated tools and methodologies being used to analyse and report on spacecraft health including trend analysis, data mining and outlier detection. A qualitative analysis of the techniques is provided based on in-flight experience, and proposals for future development of such toolsets are presented. This paper focuses on the experiences of the Copernicus Sentinel-3 and EPS MetOp flight control teams in using the EUMETSAT CHART framework, which allows engineers to define automated reports and perform ad hoc analysis on large data sets with multiple input sources. Arguments are also presented regarding whether or not it may be appropriate for future missions to consider applying some of these techniques directly on-board as an extension of the currently in-place FDIR mechanisms.

The nature and management of agricultural soils can influence the forms of legacy P present in affected sediments; however, few studies have specifically characterized P in sediments affected by polder agriculture. In this study, the speciation of P as it flows from the muck soils of the Holland Marsh to the sediments of the West Holland River and Lake Simcoe, Ontario, Canada, was investigated. The distribution of P fractions and the characterization of organic P were analyzed by the sequential fractionation method and solution P nuclear magnetic resonance spectroscopy, respectively. Organic P was the predominant P form (∼58% of total P) in muck soils, whereas the redox-sensitive P fraction was predominant in surface stream sediments rich in organic matter (∼41-48% of total P), despite these sediments exhibiting near-neutral pH and high concentrations of both Ca and P. The proportion of relatively recalcitrant organic P forms was much greater in the muck soils than that exhibited by both stream and lake sediments. The decreasing proportion of recalcitrant organic P forms in sediments downstream from the Holland Marsh indicated the potential for faster organic P cycling. Our findings support the notion that diesters and pyrophosphate should be monitored, in addition to loosely bound inorganic P, due to their potential impact on water quality. The unique environment of the streams and lake area is considered to be particularly vulnerable to excessive fertilizer P use in adjacent croplands.

The analysis of hexavalent chromium, Cr(VI), in soil and sediment samples has been predominantly carried out in materials containing elevated levels. Reliable analysis of trace-level of Cr(VI) in sediment samples remains challenging. Cr(VI) analyses with multipoint calibration and speciated isotope dilution (SID) adapted from U.S. EPA method 6800 were used to measure lower-level Cr(VI) on an ion chromatograph coupled with a tandem mass spectrometer (IC-MS/MS). Lake sediment samples were collected from various locations in Northern Ontario and Cr(VI) was extracted using both alkaline digestion and ethylene diaminetetraacetic acid (EDTA) extraction. Certified reference materials were extracted and analyzed by IC-MS/MS and UV-VIS detection. The SID-MS approach allowed for the quantification of Cr(VI) in samples with concentration levels below 0.5 μg.g-1 wet weight.
