Clinical knowledge, group facilitation skills, and cognitive congruence are considered important factors for the successful tutoring of Problem Based Learning sessions. In addition, the theory of Community of Practice has become an important tool to approach social learning and knowledge integration in medical education and organizational studies. More research is needed to link these two strands of research. We look at novice medical students' experiences and comparative reflections on student-tutors and clinician-tutors as facilitators of PBL sessions in a participatory, randomized cross-over design. Qualitative methodologies were used to probe the experiences of participants. In this study, the main factor for successful PBL sessions for first-year students was the creation of a nonhierarchical learning atmosphere, which starkly differentiated itself from the rigidity of a PBL structure organized around clinician-tutors and their hierarchically-imparted knowledge. In contrast, a more flexible strategy of student tutors and their constructive management of "not-knowing" enabled novice students to take steps on their own--of which they were highly appreciative, stressing how it allowed them to develop earlier autonomy with regard to PBL methodology, manage uncertainty, and create a shared identity as a community of learners.

Communication instructors have long insisted on the importance of audience adaptation. But they have said less about (a) the dimensions along which adaptation might proceed or (b) how a student might learn the art of adapting. In this article, I contribute toward addressing these two deficiencies. I suggest a dimension for adaptation - the value frameworks (or value vocabularies) in which people express evaluations of better and worse. And I propose that instructors teach adaptation by imitation. In addition to elaborating on these ideas, I also offer materials for use in classes.

We gathered data from business practitioners to learn how they describe successful online business presentations. We found that many--but not all--successful examples were described in terms of classical rhetorical concepts (e.g., source credibility and content). We also found that about 20% of the examples were described as successful because of technology deployment, audience interactivity, or both. We conclude that professors of management communication should teach the online presentation, that such instruction should include classical rhetorical concepts (with some appropriate adjustments), and that instruction should be expanded to include technology and interactivity.

In Germany, the debate on preimplantation and prenatal testing is heavily influenced by the idea of an antagonism between prospective parents and their potential children. Underlying this antagonism is the assumption that prospective parents follow an ableist logic and do not regard the lives of children with a chronic illness as “worth living”. Taking the example of two rare genetic illnesses, Marfan Syndrome and Cystic Fibrosis, we investigate how persons who themselves are affected by these conditions view preimplantation or prenatal testing for their condition in the context of reproductive decision-making. Our empirical research shows that chronically ill patients’ appreciation of their own life as a “life worth living” does not always translate into an unequivocal wish and readiness to give birth to a child that is affected by their condition as well and argue that taking their views into consideration adds an additional level of complexity to the debate.

Human induced pluripotent stem cells (hiPSCs) are an invaluable tool to study molecular mechanisms on a human background. Culturing stem cells at an oxygen level different from their microenvironmental niche impacts their viability. To understand this mechanistically, dermal skin fibroblasts of 52 probands were reprogrammed into hiPSCs, followed by either hyperoxic (20 % O2) or physioxic (5 % O2) culture and proteomic profiling. Analysis of chromosomal stability by Giemsa-banding revealed that physioxic -cultured hiPSC clones exhibited less pathological karyotypes than hyperoxic (e.g. 6 % vs. 32 % mosaicism), higher pluripotency as evidenced by higher Stage-Specific Embryonic Antigen 3 positivity, higher glucose consumption and lactate production. Global proteomic analysis demonstrated lower abundance of several subunits of NADH:ubiquinone oxidoreductase (complex I) and an underrepresentation of pathways linked to oxidative phosphorylation and cellular senescence. Accordingly, release of the pro-senescent factor IGFBP3 and β-galactosidase staining were lower in physioxic hiPSCs. RNA- and ATAC-seq profiling revealed a distinct hypoxic transcription factor-binding footprint, amongst others higher expression of the HIF1α-regulated target NDUFA4L2 along with increased chromatin accessibility of the NDUFA4L2 gene locus. While mitochondrial DNA content did not differ between groups, physioxic hiPSCs revealed lower polarized mitochondrial membrane potential, altered mitochondrial network appearance and reduced basal respiration and electron transfer capacity. Blue-native polyacrylamide gel electrophoresis coupled to mass spectrometry of the mitochondrial complexes detected higher abundance of NDUFA4L2 and ATP5IF1 and loss of incorporation into complex IV or V, respectively. Taken together, physioxic culture of hiPSCs improved chromosomal stability, which was associated with downregulation of oxidative phosphorylation and senescence and extensive re-wiring of mitochondrial complex composition.

Abstract Soil organic carbon (SOC) is a soil health indicator and understanding dynamics changing SOC stocks will help achieving net zero goals. Here we present four datasets featuring 11,750 data points covering co-located aboveground and below-ground metrics for exploring ecosystem SOC dynamics. Five sites across England with an established land use contrast, grassland and woodland next to each other, were rigorously sampled for aboveground (n = 109), surface (n = 33 soil water release curves), topsoil, and subsoil metrics. Commonly measured soil metrics were analysed in five soil increments for 0–1 metre (n = 4550). Less commonly measured soil metrics which were assumed to change across the soil profile were measured on a subset of samples only (n = 3762). Additionally, we developed a simple method for soil organic matter fractionation using density fractionation which is part of the less common metrics. Finally, soil metrics which may impact SOC dynamics, but with less confidence as to their importance across the soil profile were only measured on topsoil (~5–15 cm = mineral soil) and subsoil (below 50 cm) samples (n = 2567).

Abstract This bioequivalence research aims to evaluate the relative bioavailability and pharmacokinetic characteristics of ethinyl estradiol and drospirenone in the test preparation in comparison to the reference preparation during fasting conditions. A liquid chromatography method with tandem mass spectrometry was used to determine the concentrations of drospirenone and ethinyl estradiol in plasma. The pharmacokinetic parameters that were analyzed were the maximum plasma concentration (Cmax), time to achieve Cmax (tmax), elimination half life, and area under the concentration time curve of plasma (AUC0‐t, AUC0‐∞ for ethinyl estradiol, and AUC0‐72h for drospirenone). Both the AUC and Cmax parameters were determined to be between 80.00% and 125.00% (90% confidence intervals), which is the acceptable range. Based on the study findings, it was concluded that the test formulation, which includes 3 mg of drospirenone and 0.03 mg of ethinyl estradiol, demonstrated bioequivalence when compared to the reference formulation.

The summer drought in the United Kingdom (UK) in 2022 produced significant speculation concerning how its termination may impact and interact with the soil resource. Whilst knowledge regarding soils and droughts exists in the scientific literature, a coherent understanding of the wider range of impacts on soil properties and functions has not been compiled for temperate soils. Here, we draw together knowledge from studies in the UK and other temperate countries to understand how soils respond to drought, and importantly what and where our knowledge gaps are. First, we define the different types of droughts and their frequency in the UK and provide a brief overview on the likely societal impacts that droughts place on the soil and related ecosystems. Our focus is on ‘agricultural and ecosystem drought’, as this is when soils experience dry periods affecting crops and ecosystem function, followed by rewetting. The behaviour of moisture in soils and the key processes that contribute to its storage and transport are examined. The principal changes in the physical, chemical, and biological properties of soils resulting from drought, and rewetting (i.e., drought termination) are discussed and their extensive interactions are demonstrated. Processes that are involved in the rewetting of soils are explored for soil and catchment-scale soil responses. Lastly, soils’ recovery after drought is considered, knowledge gaps are identified, and areas to improve understanding are highlighted.

The intensification of dairy and biogas production in Northwestern Europe has led to the conversion of permanent grasslands to arable land, mainly for silage maize production, resulting in significant soil organic carbon (SOC) losses, highlighting the need for implementing alternative management practices during land use change (LUC) for effective climate change mitigation. This study evaluated the impact of incorporating annual grass-clover leys in 3-year arable rotations and slurry application to mitigate SOC losses during LUC. We compared this approach to a continuous silage maize and a permanent grassland on sandy loam soil in Northern Germany. The experiments were simultaneously established at two adjacent 17-year-old sites of permanent grassland and arable cropping, with different levels of initial SOC when the experiment was established. The SOC dynamics in the upper soil layer (0–30 cm) were evaluated by annual 12-year sampling (2011–2022). The cropping systems were unfertilized (N0) or fertilized (N1) using cattle slurry at a rate of 240 kg N ha-1 year-1. The study reveals substantial SOC losses following the conversion of the permanent grassland to grass-clover (ley) based rotation or continuous silage maize, with reductions of 22% and 31%, respectively, compared to baseline levels of the permanent grassland. However, over the 12-year period, the grass-clover ley-based crop rotation demonstrated a 30% reduction in SOC losses compared to continuous silage maize, without compromising dry matter yield. Conversely, the conversion of arable land to grasslands led to SOC increases ranging from 10% to 30%. This recovery was only half the SOC losses observed in the grassland conversion for the same period, indicating a slow-in, fast-out effect during LUC. However, the transition from ley-containing forage rotation to continuous silage maize incurred significant SOC losses of 11%. Overall, these findings underscore the imperative of integrating ley phases to mitigate SOC losses, particularly in high-biomass-yield cropping systems. As a 1-year ley phase was insufficient to sustain carbon sequestration in arable crop rotations, extended ley residence times should be considered.

ABSTRACT: Increasing the nitrogen-utilization efficiency (NUE) of dairy cows by breeding selection would offer advantages from nutritional, environmental, and economic perspectives. Because data collection of NUE phenotypes is not feasible in large cow cohorts, the cow individual milk urea concentration (MU) has been suggested as an indicator trait. Considering the symbiotic interplay between dairy cows and their rumen microbiome, individual MU was thought to be influenced by host genetics and by the rumen microbiome, the latter in turn being partly attributed to host genetics. To enhance our knowledge of MU as an indicator trait for NUE, we aimed to identify differential abundant rumen microbial genera between Holstein cows with divergent genomic breeding values for MU (GBVMU; GBVHMU vs. GBVLMU, where H and L indicate high and low MU phenotypes, respectively). The microbial genera identified were further investigated for their correlations with MU and 7 additional NUE-associated traits in urine, milk, and feces in 358 lactating Holsteins. Statistical analysis of microbial 16S rRNA amplicon sequencing data revealed significantly higher abundances of the ureolytic genus Succinivibrionaceae UCG-002 in GBVLMU cows, whereas GBVHMU animals hosted higher abundances of Clostridia unclassified and Desulfovibrio. The entire discriminating ruminal signature of 24 microbial taxa included a further 3 genera of the Lachnospiraceae family that revealed significant correlations to MU values and were therefore proposed as considerable players in the GBVMU–microbiome–MU axis. The significant correlations of Prevotellaceae UCG-003, Anaerovibrio, Blautia, and Butyrivibrio abundances with MU measurements, milk nitrogen, and N content in feces suggested their contribution to genetically determined N-utilization in Holstein cows. The microbial genera identified might be considered for future breeding programs to enhance NUE in dairy herds.

Passive seismic analyses are a key technology for the exploration and monitoring of subsurface reservoirs. Searching for alternative resources in the framework of the energy transition is creating a surge for identifying as many potential sites as possible suitable for geothermal exploitation. The Lower Rhine Embayment, at the western border of North Rhine-Westphalia in Germany, is an extensional system with a very high potential for geothermal exploitation. The area experiences moderate but continuous natural seismicity. Here, we report on a passive seismic dataset recorded with 48 seismic stations centred at and around Eschweiler–Weisweiler (https://doi.org/10.14470/MO7576467356, Finger et al., 2022). Background seismic noise levels are high at this site due to high levels of anthropogenic noise and thick unconsolidated sedimentary layers. The final station layout is a compromise between targeted network design and suitably quiet locations. We show that the network design allows for the application of state-of-the-art methods including waveform-based source location methods and ambient-noise velocity imaging methods.
