In fish, gonadotropin hormones FSH-GTH1 and LH-GTH2 are less specific for their cognate receptors than in mammals. The respective reproductive functions of fish LH and FSH are thus difficult to establish. We aimed to study the effect of specific antagonists of the two gonadotropin receptors on trout sexual maturation in both sexes by targeting specific regions of LH and FSH receptors, Lhr and Fshr. Filamentous phages displaying Lhr specific or Fshr specific decapeptides from the extracellular hormone binding domain were engineered. Recombinant phages were used as receptor-specific antagonistic vaccines. Male and female trouts were immunized with anti-LHR, anti-FSHR, anti-FSHR+LHR or adjuvant alone, through multiple injections over 8-24 weeks, starting at different stages of sexual maturation. The consequences of immunization on gonadal development were evaluated by determining gonad growth, by histological analysis of testis and ovaries at the end of the vaccination period and by measuring blood plasma sex steroids using radioimmunoassay. We show for the first time in fish that the anti-receptor vaccinations could have specific antagonistic effects on the development of the reproductive functions; while the anti-FSHR affected the sexual maturation of prepubertal males and delayed sperm production, the anti-LHR blocked vitellogenesis in females. In maturing males, the combined anti-FSHR+LHR vaccine inhibited spermatogenesis and affected steroidogenesis. In that case, the effects of the vaccine on spermatogenesis were transient and reversible when immunization was stopped. Such an immunological strategy to specifically and transiently inhibit a receptor provides a promising approach for discovering their specific functions; it could also lead to a new technology for controlling the onset of puberty in aquaculture species.

The involvement of autoimmune mechanisms in premature ovarian failure has been put forward by numerous investigators. In various other ovarian pathologies, such as idiopathic infertility, polycystic ovary syndrome, or endometriosis, similar mechanisms have been suggested. However, the exact role of autoimmunity in the pathophysiology of these diseases still remains controversial. The diagnosis of autoimmune ovarian disease relies on several clinical, biological and histological findings, but special interest has been focused on antiovarian autoantibodies. The search for these antibodies has been undertaken by several authors and yielded somewhat conflicting results which might be conditioned by methodological differences and by the multiplicity of potential immune targets. These targets, which comprise various steroidogenic enzymes, gonadotrophins and their receptors, the corpus luteum, zona pellucida and oocyte, are reviewed. Further investigation of these targets is required to improve the diagnostic tools that will lead to a precocious and reliable diagnosis of autoimmune ovarian disease, an appropriate clinical surveillance as well as the selection of patients who may benefit from immune-modulating therapy and possibly recover ovarian function and fertility.

The ovary can be the target of an autoimmune disease involving many different autoantigens. The clinical feature of this disease often results in premature ovarian failure or infertility and may be either isolated or associated with other autoimmune pathologies, especially with adrenal autoimmunity. The diagnosis of an autoimmune mechanism relies on the presence of anti-ovarian antibodies, whose prevalence is quite variable according to the different methods used to detect them, and to the different stages of the disease. In addition, their clinical significance is not always clear, as to their pathologic or epiphenomenal nature. However, the study of these autoantibodies has led to the identification of some of their antigenic targets which have to be known for a better understanding of the pathologic mechanisms involved. This paper reviews anti-steroid producing cells, anti-gonadotrophin receptor, anti-gonadotrophin, anti-corpus luteum, anti-zona pellucida and anti-oocyte antibodies.

Fertility management is a global issue of agricultural, medical, economic, and social consequence. Although many methods have been devised to both inhibit and assist reproduction, more acceptable alternatives are needed. Regulation by immune intervention is a promising technology as applied to livestock, pets, wildlife, and human beings. Outcome is dictated by site within the reproductive axis that is targeted. Fertility is suppressed by immunization against gonadotropin-releasing hormone, gonadotropins, prostaglandin F2 alpha, oxytocin, gonadotropin receptors, and gamete/embryonic antigens. It also is possible to lyse gonadal cells with ligand-antibody hybrid molecules. Ovulation rates are enhanced by vaccination with inhibin. Antibodies to sex steroid hormones have yielded mixed results. Perhaps recombinant viral vectors can be used to deliver reproductive immunogens. A new and simple technique to generate sustained autoimmune reactions to hormones and cellular antigens entails direct gene transfer into somatic cells. Evolving advances in reproductive immunology and biotechnology should furnish us with novel nonsurgical contraceptives and profertility agents that can be efficiently and safely implemented.

We reported the production of monoclonal antibodies (McAbs) against chorionic gonadotropin hormone (CG) receptor by fusing spleen cells of BALB/c mice which had been immunized by purified bacteria (Pseudomonas maltophilia) CG receptor with mouse myeloma line SP2/0. Four hybridoma cell lines secreting CG receptor McAbs were obtained (ED490 DG390, AB890 and GE590). The titers of specific antibodies of both mice ascites and culture supernatant were 10(-2)-10(-6) and 1-10(-2) respectively, by solid phase ELISA. Double-Immunodiffusion test showed that the McAb GE590 was IgG1, and the McAbs ED490, DG390 and AB890 were IgG2b Immunoprecipitation indicated that 125I-HCG could bind the HCG receptor which had reacted with McAbs ED490, AB890 and DG390, suggesting that they may recognize the receptor with different antigenic determinant. Interaction of McAb GE590 with the receptor showed that the increased concentration of GE590 was in inverse proportion to the amount of 125I-HCG binding receptor, indicating that both the McAb and 125I-HCG could recognize a common site of receptor and that increased concentration of McAb GE590 may induce some change in conformation and structure of the receptor. Our study suggested that these McAbs may be used for studying structure of CG receptor.

Polyclonal antisera to a SDS-denatured and partially renatured rat luteal 90 K LH/CG receptor were raised in rabbits, characterized, and their applicability for immunohistochemical location of the receptor examined. The LH/CG receptor was purified by hCG-affinity chromatography and subjected either to a preparative SDS-PAGE or Western blotting. Gel slices containing the SDS-denatured or nitrocellulose strips containing the renatured 90 K LH/CG receptor were used for immunization. The antisera, termed ARS-2 and ARS-3, respectively, possessed similar antibody titres. Both antisera were able to recognize the native, SDS-denatured, and SDS-denatured and reduced forms of the LH/CG receptor on dot blots, but only ARS-3 contained antibodies to the hormone binding site or a region near to it, as it was able to inhibit the hCG binding to the membrane-bound LH/CG receptor in a dilution-dependent manner. Both antisera recognized the receptor-hCG complex, but ARS-2 stained the complex with about 50% less intensity than the free receptor. ARS-3 located the LH/CG receptor distinctly on the luteal cell surfaces in immunohistochemical staining with peroxidase antiperoxidase complex method, but ARS-2, although it possessed similar antibody titre, revealed negligible staining. Thus, the antisera readily recognize the native receptor, but differ in their capability for inhibiting hormone binding. Only ARS-3, produced against the renatured receptor, contains sufficient amounts of antibodies capable of recognizing free and occupied receptors in immunohistochemistry.

The present study was performed to evaluate the relationship between the antagonist effect and the conformation of deglycosylated human chorionic gonadotropin (DGhCG) on the Leydig cell lutropin receptors. The maximum binding of [125I]DGhCG to anti-hCG beta antibody was decreased by 50%, while its binding profile to anti-hCG, anti-DGhCG and anti-hCG alpha antibodies remained unchanged, suggesting conformational changes in the beta subunit of DGhCG. However, the association of [125I]DGhCG to the binding sites of the receptors was much faster than that of [125I]hCG, and the ligand reached the binding equilibrium at 4 and 37 degrees C for 3 h and 15-30 min, respectively. Thus, the conformational changes in the beta subunit were not accompanied by loss of its receptor binding ability. The agonist properties of DGhCG which was bound to the receptors was fully restored by the addition of anti-hCG beta subunit antibody, while anti-hCG or anti-DGhCG restored only about 30% of the full agonist activity. This was probably due to a change of the conformation of the beta subunit to make it similar to that of the intact hormone. This restoration caused by anti-hCG beta was partially prevented by anti-hCG alpha. These facts indicate that some conformational change only in the beta subunit, not in the alpha subunit, of the deglycosylated hormone bound to the receptors is essential for the restoration of agonist properties.
