One-step oxidative cleavage of cyclohexene to adipic acid using H2O2 as oxidizing agent and heteropolysalts as heterogeneous catalysts was successfully performed, without the use of additives, such as organic or inorganic acids. The K3PW12O40, K3PMo12O40, Cs3PW12O40 and Cs3PMo12O40 heteropolysalts were synthesized by ion exchange of the respective heteropolyacids using cesium and potassium carbonates. The catalysts were thermally treated at 200 or 600 °C and characterized by Raman spectroscopy, thermogravimetric analysis, X-ray fluorescence (XRF) and photoelectronic X-ray spectroscopy (XPS). Raman results showed that the Keggin structure of the polyoxometalates was successfully obtained for both heteropolyacids and heteropolysalts and remained stable even after thermal treatment at 600 °C. XRF analysis confirmed the insertion of the cations (Cs+ and K+) into the heteropolysalt structures. All catalysts completely converted cyclohexene within 24 h and they were selective to adipic acid. The best catalyst, K3PW12O40 calcined at 600 °C (KPW-600), achieved the highest adipic acid yield (83 %). In this work, we associate the best activity of the KPW-600 heteropolysalt not only to its greater acidity, but also to the higher concentration of (W-O-W)/W species (identified by XPS). Both properties are fundamental for the oxidation reaction of cyclohexene to adipic acid under the experimental conditions used. The results showed that the presence of peroxide species is essential, mainly to favor oxidation reactions, such as the Baeyer-Villiger rearrangement reaction. A reaction scheme for the oxidation of cyclohexene to adipic acid with the best catalyst (KPW-600), was proposed based on our kinetic studies and literature results. In addition to the main route of adipic acid formation from the direct oxidation of cyclohexene, undesirable parallel reactions of the rearrangement of 1,2 - cyclohexanediol and allyl oxidation were also observed. The influence of reaction conditions was also investigated, as well as the reuse of the most promising catalyst.

Synthetic studies on an ABC-ring model of tubiferal A, a triterpenoid isolated from the fruit bodies of the Tubifera dimorphotheca myxomycete, are described. The stereogenic centers at the angular positions were constructed through the stereoselective addition of a C-ring allylborane followed by an Eschenmoser–Claisen rearrangement reaction prior to the formation of the AB-ring system by a double intramolecular alkylation reaction of a dichloro nitrile intermediate.

ConspectusThe pursuit for the discovery of new and powerful synthetic methods to access high-value N-heterocycles has been at the forefront of organic chemistry research for more than a century. Considering the importance of N-scaffolds in modern science, over the past few decades, great research efforts have been made to develop efficient synthetic methods for the construction of nitrogen-rich molecules. Among many efforts, transition metal catalyzed denitrogenative annulation reaction has emerged as a cornerstone due to its innate versatility and wider scope of application.The denitrogenative annulation approach offers clear advantages over many existing methods, as it enables effective, single-step interconversion of easily available feedstocks into a variety of other important N-containing heterocyclic frameworks. Recently, transition metal catalyzed denitrogenative annulation reaction of the 1,2,3-triazole via a metal carbene intermediate sparked significant interest in the application of various important heterocycle syntheses. Denitrogenative annulation reaction of 1,2,3-triazoles proceeds via an ionic mechanism. Recently, we demonstrated a new concept for the denitrogenative reaction of triazoles with alkenes and alkynes via in situ generated 2-(diazomethyl)pyridines. The method takes advantage of the inherent properties of a Co(III)-carbene radical intermediate and is the first report of the denitrogenative annulation/cyclopropanation by a radical-activation mechanism.On the other hand, in contrast to the denitrogenative annulation of 1,2,3-triazole, annulation reaction of 1,2,3,4-tetrazole (a surrogate of azide having an important pyridyl unit) via metal nitrene remains a big challenge. Previously, flash vacuum pyrolysis studies had been used for nitrene-nitrene rearrangement of 1,2,3,4-tetrazole at high temperature. This Account summarizes our recent efforts in developing transition metal catalyzed denitrogenative annulation of 1,2,3-triazoles via a radical mechanism and 1,2,3,4-tetrazoles via metal nitrene to access important nitrogen-rich molecules. We demonstrated that the 1,2,3,4-tetrazole under Ir-catalyzed reaction conditions can produce a productive Ir-nitrene intermediate that can successfully be employed for the construction of a wide number of α-carbolines and 7-azaindoles. Moreover, we developed an iron-based unique strategy for the intermolecular denitrogenative annulation reaction between tetrazoles and alkynes. The reaction overcomes the traditional click reaction and proceeds via an unprecedented metalloradical activation mechanism. Furthermore, we used our understanding of tetrazole reactivity to design an iron-catalyzed intramolecular denitrogenative C(sp3)-H amination reaction of primary, secondary, and tertiary centers by using a metalloradical activation concept. At the same time, we also developed a general catalytic method to enable two distinct reactions (1,3-cycloaddition and denitrogenative annulation) using Mn(TPP)Cl that afforded two different classes of nitrogen heterocycles. Mechanistic studies showed that although the click reaction likely proceeds through an ionic mechanism and the denitrogenative annulation reaction likely proceeds via an electrophilic metallonitrene intermediate rather than a metallonitrene radical intermediate. Finally, we report an iron-catalyzed rearrangement reaction (ring expansion/migration) that proceeded with an unprecedented level of selectivity, reactivity, and functional group tolerance offering rapid access to numerous complex N-heterocycles. We believe that our continuous efforts in this field would be beneficial for pharmaceutical industries, drug discovery, and other fields of medicinal chemistry.

Mechanistic study on the 1,3,2-diazaphospholene (1)-catalyzed reduction reaction of allyl 2-phenylacrylate 4 with HBpin or ammonia borane (AB) was systematically performed by the density functional theory (DFT) method. When HBpin is employed as the reductant, the reductive Ireland-Claisen (IC) rearrangement reaction occurs. First, the active species P-hydrido-1,3,2-diazaphospholene 3 is generated through the metathesis reaction of 1 with HBpin. Next, the terminal C═C double bond of 4 is inserted into the P-H bond of 3 to produce 6a through the 1,2-addition (Markovnikov) step, which is followed by the pinB-H bond activation to afford key boron enolate 8. Then, 8 undergoes the [3,3] rearrangement that is followed by the alcoholysis reaction with methanol leading to the final product γ,δ-unsaturated carboxylic acid. The [3,3] rearrangement step is the rate-determining step with the Gibbs energy barrier (ΔG≠) and Gibbs reaction energy (ΔG) of 23.9 and -27.5 kcal/mol, respectively. When AB is employed as the reductant, the transfer hydrogenation reaction occurs through two comparable pathways, 1,2- and 1,4-transfer hydrogenation pathways. The former pathway directly leads to the hydrogenation product with the ΔG≠ and ΔG values of 22.4 and -27.7 kcal/mol, respectively. The latter pathway produces an enolate intermediate (rate-determining step, ΔG≠/ΔG = 24.1/-0.3 kcal/mol) first, which then prefers to undergo the enol-keto tautomerism instead of the [3,3] rearrangement to afford the hydrogenation product. Obviously, the generation of the boron enolate plays a crucial role in the reductive IC rearrangement reaction because it prevents the enol-keto tautomerism.

[Image: see text] A new synthetic method for the construction of benzoazepine analogues has been developed employing ortho-arylmethylbenzyl azide derivatives as precursors using an azide rearrangement reaction. In this work, 14 benzoazepine compounds were successfully synthesized in moderate to excellent yields. All synthetic benzoazepines were evaluated for their cytotoxicity against normal human kidney cell line (HEK cell). The results showed that compound 18c had the lowest cytotoxicity (IC(50) = 65.68 μM) among tested compounds, which was comparable with the antianxiety drug diazepam (IC(50) = 87.90 μM). Based on the cytotoxicity results, five benzoazepine analogues (compounds 18c, 18h, 18j, 18n, and 18p) were selected to determine the antianxiety effect on stressed rats using elevated plus maze (EPM) and open field test (OFT) methods. Interestingly, compound 18c showed better anxiolytic activity than diazepam without a sedative effect by showing superior hyperlocomotor activity. Therefore, this discovery could pave the way for drug development to treat patients with anxiety disorder.

A Bi(OTf)3 -catalyed reaction of 3-aryl propargyl alcohols with sulfonamide and halogen source was firstly investigated, which provided a facile route for the synthesis of a large variety of α-halo-β-amino ketones. The key intermediates, β-amino ketones, were obtained through tandem Meyer-Schuster rearrangement reaction of propargyl alcohols and intermolecular Michael addition of α, β-unsaturated ketones and sulfonamide. Then the in situ generated α-halo-β-amino ketones underwent the base-promoted intramolecular cyclization to give diverse acyl aziridines in a one-pot fashion. These transformations are reliable on a large scale. The high yields and convenient experimental operations make it a valuable method for the construction of α-halo-β-amino ketones and acyl aziridine derivatives.

A new and efficient cascade electrophilic trifluoromethylthiolation/radical rearrangement reaction of ketoximes is reported using N-trifluoromethylthio-dibenzenesulfonimide as the SCF3 source without any additives. This simple one-pot reaction provides the corresponding N-trifluoromethanesulfinyl ketimine products in good yields with high E selectivity under mild reaction conditions.

The ligand scrambling reaction of gold(I) complexes is a phenomenon occurring primarily in L–AuI–X (L = phosphine, N-heterocyclic carbene (NHC), and thiol; X = halide and thiol) complexes and has been observed among others for e.g., the bromido[3-ethyl-4-(4-methoxyphenyl)-5-(2-methoxypyridin-5-yl)-1-propyl-1,3-dihydro-2H-imidazol-2-ylidene]gold(I) complex (7a), which underwent ligand rearrangement in aqueous solutions. In this study, we investigated the influence of substituents on the 4-aryl ring of the related (NHC)AuIBr complexes (1a–9a) in terms of the conversion to the [(NHC)2AuI]+ (1b–9b) and [(NHC)2AuIIIBr2]+ (1c–9c) species. Furthermore, the influence of external factors such as solvent, temperature, concentration, and presence of halides (Cl–, Br–, and I–) or hydroxyl ions was studied to gain a deeper understanding of the ligand rearrangement reaction. The substituent on the 4-aryl ring has a marginal impact on the scrambling reaction. Out of the investigated organic solvents (dimethylformamide (DMF), dimethyl sulfoxide (DMSO), ethanol (EtOH), methanol (MeOH), and acetonitrile (ACN)), only ACN separates single complex molecules. In all other solvents, relatively stable ((NHC)AuIBr)2 dimers are present. The addition of water to ACN solutions forces the formation of such dimeric units, starting the transformation to [(NHC)2AuI]+ and [(NHC)2AuIIIBr2]+. The rate-determining step is the release of Br– from a T-shape intermediate because an excess of KBr terminates this reaction. Furthermore, it is obvious that only single molecules react with halides. The aurophilic interactions between two (NHC)AuIBr molecules are too strong in the presence of water and largely impeded reaction with halides. As a single molecule, the reaction with Cl– (e.g., in a 0.9% NaCl solution) is notable, while I– even leads to a fast and quantitative conversion to (NHC)AuII and finally to [(NHC)2AuI]+., Internal and external factors of the ligand scrambling reaction in (NHC)AuIBr complexes were investigated by the means of HPLC. The data represent the impact of substituents, temperature, solvent, concentration and addition of halides on the conversion to the respective [(NHC)2AuI]+ and the oxidation to the [(NHC)2AuIIIBr2]+ complexes.

Improved methods of convergent synthesis for peptidomimetic utilizing a chloroalkene dipeptide isostere (CADI) are reported. In this synthesis, Fmoc- or Boc-protected carboxylic acids can be produced from N- and C-terminal analogues corresponding to each amino acid starting material via an Evans syn aldol reaction, followed by a [3.3] sigmatropic rearrangement utilizing the Ichikawa allylcyanate rearrangement reaction. With this strategy, an Fmoc-protected CADI can be directly applied for solid-phase peptide synthesis. Using this approach, we have also identified the CADI-containing cyclo[-Lys-Leu-Val-Phe-Phe-] peptidomimetic, which is a superior inhibitor of amyloid-β aggregation than the parent peptide.

An iron-catalyzed denitrogenative rearrangement of 1,2,3,4-tetrazole is developed over the competitive C(sp3 )-H amination. This catalytic rearrangement reaction follows an unprecedented metalloradical activation mechanism. Employing the developed method, a wide number of complex-N-heterocyclic product classes have been accessed. The synthetic utility of this radical activation method is showcased with the short synthesis of a bioactive molecule. Collectively, this discovery underlines the progress of radical activation strategy that should find wide application in the perspective of medicinal chemistry, drug discovery and natural product synthesis research.

We report the first enantioselective total syntheses of the hasubanan alkaloid (-)-metaphanine and the norhasubanan alkaloid (+)-stephadiamine. Key features of these syntheses include diastereoselective oxidative phenolic coupling reaction and subsequent regioselective intramolecular aza-Michael reaction, which efficiently construct the hasubanan skeleton with the all-carbon quaternary stereogenic center at C13. Based on our hypothesis regarding the biosynthetic pathway of (+)-stephadiamine, we found that (-)-metaphanine is easily converted to (+)-stephadiamine via aza-benzilic acid type rearrangement reaction.

The first total syntheses of hericenones C-H and "putative 3-hydroxyhericenone F" were achieved. Highlights of the synthesis include the straightforward construction of the resorcinol core and geranyl side chain, assembly of the natural product skeleton by sequential O-geranylation and a clay/zeolite-mediated O → C rearrangement reaction, and a biomimetic cyclization to form a variety of bicyclic natural hericenones and their congeners. The structure of the "putative 3-hydroxyhericenone F" was revised as the 5-exo cyclization product (named: hericenone Z) of epoxyhericenone C through in-depth analyses of the cyclization modes in addition to NMR spectroscopic studies. To gain insights into the biological functions of geranyl-resorcinols in Hericium erinaceus, potential neuroprotective effects against endoplasmic reticulum (ER) stress-dependent cell death were evaluated systematically to clarify a fundamental structure-activity relationship. Among the compounds assayed, the linoleate-containing hericenone analogue, i.e., the regioisomer of hericene D, was found to possess the most potent neuroprotective effect against tunicamycin and thapsigargin-induced ER stress-dependent cell death.

A general synthesis of 2-aryl benzazepines has been developed through palladium-catalyzed ring-expansion reactions of cyclobutanols with 2-haloanilines; the further oxidative rearrangement reaction of benzazepines provided an efficient synthesis of 2-acyl quinolines. These transformations feature the efficient construction of six- and seven-membered N-containing heterocycles from easily obtained materials with excellent functional group tolerance.

The reaction of the titanium imido complex 1b with 2-butyne leads to the formation of the titanium azadiene complex 2a at ambient temperature instead of yielding the archetypical [2 + 2] cycloaddition product (titanaazacyclobutene) which is usually obtained by combining titanium imido complexes and internal alkynes. The formation of 2a is presumably caused by an initial propargylic C(sp3)–H activation step and quantum chemical calculations suggest that the outcome of this unexpected reactivity is thermodynamically favored. The previously reported titanaazacyclobutene I (which is obtained by reacting 1b with 1-phenyl-1-propyne) undergoes a rearrangement reaction at elevated temperature to give the corresponding five-membered titanium azadiene complex 2b.

In our previous study, an unusual rearrangement reaction was discovered whereby dinaphthyl ketones with three hydroxy groups at restricted positions were transformed into a phenalenone ring and a benzene ring. Using the rearrangement as a key reaction, the first total syntheses of FR-901235 and auxarthrones A-D from an unstable triketone common intermediate are described. Furthermore, lamellicolic anhydride was synthesized from the triketone. This conversion is part of the putative biosynthetic pathway and was achieved experimentally for the first time.

Rational conversion of CO2 into valuable fuels would partially or fully close the carbon loop and can have significant environmental benefits. The elemental steps of CO2 conversion into hydrocarbon fuels such as methanol, methane and a series intermediates at the edge of graphitic carbon nitride were investigated using first-principle calculations. Herein, we reported the detailed mechanism of CO2 reduction at the edge of g-C3N4 sheet including two possible pathways based on H-shuttling model. The participation of hydrogen within NH group at the edge of g-C3N4 sheet as another hydrogen source could promote the hydrogenation of intermediate through the rearrangement reaction. Methanol is a quite favorable product than methane during CO2 reduction reaction. Carbon monoxide (CO) and formaldehyde as the products would limit the conversion of CO2 to hydrocarbon fuels.

N-aryl hydroxamic acids, which are best known for their metal-chelating properties in chemical and biomedical research, have been found to markedly detoxify carcinogenic halogenated quinones. However, the exact chemical mechanism underlying such detoxication remains unclear. Here, we show that a very fast reaction took place between N-phenylbenzohydroxamic acid (N-PhBHA) and 2,5-dichloro-1,4-benzoquinone (DCBQ), forming an unexpected new carbon-carbon bonding phenyl-quinone product with high yield. In contrast, no reaction was observed with O-benzoyl N-PhBHA. Analogous results were observed for other N-aryl hydroxamic acids and halogenated quinones, which have an ortho-hydrogen adjacent to the reaction site (DCBQ-type). Interestingly, no free radical intermediates could be detected by both ESR spin-trapping and radical-scavenging methods during the reaction process. Taken together, we proposed that nucleophilic substitution followed by an unusual two-step Claisen-type rearrangement reaction was responsible for the formation of a new C-C bonding compound and the detoxication reaction. This represents the first report of an unusually mild and facile two-step Claisen-type rearrangement, which could take place under normal physiological conditions.

Synthesis of mixed‐substituted tetraphosphetanes (RP−PtBu)2 (R=Ph (4), Py (5); Py=2‐pyridyl) is achieved from the condensation of dipyrazolylphosphanes RPpyr2 (R=Py (1), Ph (3); pyr=3,5‐dimethylpyrazolyl) as P1‐building block (R−P) and tBuPH2 in an equimolar ratio. Compound 5 is of special interest since the presence of two pyridyl‐substituents as well as the P4‐core allows for a rich coordination chemistry with coinage metal salts [Cu(MeCN)4][OTf], Ag[OTf] and in situ formed [Au(tht)][OTf] (tht=tetrahydrothiophene). Both tetraphosphetanes undergo alkylation reaction with MeOTf to give a series of tetraphosphetanium and tetraphosphetanediium triflate salts with additional methylation of the pyridyl‐moiety in case of 5 resulting in interesting novel cyclic trications. Harsh reaction condition and an excess of MeOTf converts 5 into the cyclic trication [‐P(MePy)PMe2P(MePy)PtBu‐]3+ (13 3+; MePy=1‐methylpyridiniumyl) through the elimination of isobutene. This salt undergoes a complicated rearrangement reaction involving a P−P/P−P bond metathesis to form trication [‐P(MePy)3PtBu‐]3+ (17 3+) when reacted with Me2PPMe2., Choose your donor: A convenient route towards mixed‐substituted tetraphosphetanes was established, enabling the synthesis of (PhP‐PtBu)2 and (PyP‐PtBu)2 (Py=2‐pyridyl) in good yields. The coordination chemistry of (PyP‐PtBu)2 was investigated by reacting it with coinage metal triflate salts. Further methylation studies on both tetraphosphetanes revealed an interesting chemistry of multiply charged, cylic polyphosphanes.

Cyanogramide (1) from the marine actinomycete Actinoalloteichus cyanogriseus WH1-2216-6 features a unique spirooxindole skeleton and exhibits significant bioactivity to efficiently reverse drug resistance in tumor cells. The biosynthetic gene cluster of 1 in A. cyanogriseus WH1-2216-6 was identified and refactored by promoter engineering for heterologous expression in Streptomyces coelicolor YF11, thereby enabling the production of 1 and five new derivatives. Interesting, four of them, including 1, were identified as enantiomeric mixtures in different ratios. The functions of tailoring enzymes, including two methyltransferases (CyaEF), and three cytochrome P450 monooxygenases (CyaGHI) were confirmed by gene inactivation and feeding experiments, leading to the elucidation of a concise biosynthetic pathway for 1. Notably, CyaH was biochemically verified to catalyze the formation of the spirooxindole skeleton in 1 through an unusual carbocation-mediated semipinacol-type rearrangement reaction.

A series of multiblock poly­(benzoxazole-co-imide) (PBOI) membranes were prepared via thermal rearrangement of their corresponding multiblock copolyimide (CPI) precursors based on 4,4′-(hexafluoroisopropylidene) diphthalic anhydride (6FDA), 2,2′-bis­(3-amino-4-hydroxy-phenyl) hexafluoropropane (APAF), and 5-amino-2-(4-aminobenzene) benzimidazole (BIA). For ortho-hydroxy-functionalized CPI precursors, the difference in stacking behavior between the 6FDA–APAF and 6FDA–BIA blocks results in a weak microphase separation in CPI membranes, whereas the random precursor exhibits a homogeneous morphology, which has been identified by dynamic mechanical analysis, small-angle X-ray scattering, and atomic force microscopy analysis. After the subsequent thermal rearrangement at 420 °C, the microphase separation was also observed in the multiblock poly­(benzoxazole-co-imide) (TR-PBOI) membranes with an enlarged domain size. The impacts of the sequence structure on the membrane phase-separation behavior and mechanical and gas separation properties were investigated. Comparatively, an obvious increase in the CO2/CH4 gas separation property from the random to TR-PBOI membranes was observed. Specially, the resultant multiblock A40B40-TR-420 membrane possesses a CO2 permeability of 92 Barrer and a CO2/CH4 selectivity of 54.7, which is substantially higher than the values of the random-PBOI-420 membrane (CO2 permeability of 40.2 Barrer and CO2/CH4 of 58) and those of recently reported TR-PBOI membranes. The appealing gas separation performance of the multiblock PBOI membranes can be attributed to their microphase-separated morphology, in which continuous percolating microcavities formed in the thermal rearrangement reaction provide transport channels for gas molecules. The present study demonstrates that the modification of the micromorphological structure of membranes can effectively tune their final gas separation properties.

Incorporation of a non-hexagonal ring into a nanographene framework can lead to new electronic properties. During the attempted synthesis of naphthalene-bridged double [6]helicene and heptagon-containing nanographene by the Scholl reaction, an unexpected azulene-embedded nanographene and its triflyloxylated product were obtained, as confirmed by X-ray crystallographic analysis and 2D NMR spectroscopy. A 5/7/7/5 ring-fused substructure containing two formal azulene units is formed, but only one of them shows an azulene-like electronic structure. The formation of this unique structure is explained by arenium ion mediated 1,2-phenyl migration and a naphthalene to azulene rearrangement reaction according to an in-silico study. This report represents the first experimental example of the thermodynamically unfavorable naphthalene to azulene rearrangement and may lead to new azulene-based molecular materials.

The reaction of 2-(bromomethyl)-1,3-thiaselenole with potassium selenocyanate proceeded via a rearrangement with ring expansion, leading to a six-membered 2,3-dihydro-1,4-thiaselenin-2-yl selenocyanate (kinetic product) which in turn underwent rearrangement with ring contraction to a 1,3-thiaselenol-2-ylmethyl selenocyanate (thermodynamic product). These rearrangements occurred by a nucleophilic attack of the selenocyanate anion at two different carbon atoms of the seleniranium intermediate. The efficient regioselective synthesis of alkyl, allyl, 2-propynyl, benzyl, 4-fluorobenzyl, and 2-pyridinylmethyl 1,3-thiaselenol-2-ylmethyl selenides was developed based on the generation of sodium 1,3-thiaselenol-2-ylmethylselenolate from 1,3-thiaselenol-2-ylmethyl selenocyanate or bis(1,3-thiaselenol-2-ylmethyl) diselenide followed by nucleophilic substitution reactions. Sodium 1,3-thiaselenol-2-ylmethylselenolate underwent nucleophilic addition to alkyl propiolates in a regio- and stereoselective manner affording 1,3-thiaselenol-2-ylmethyl vinyl selenides in high yields predominantly with Z-configuration. Not a single representative of the 1,3-thiaselenol-2-ylmethyl selenide scaffold has been previously described in the literature.

We report the first examples of catalytic enantioselective benzilic ester rearrangement reaction. In the presence of a catalytic amount of Cu(OTf)(2) and a chiral box ligand under mild conditions, reaction of 2,3-diketoesters with alcohols afforded structurally diverse alpha-aryl(alkyl) substituted-alpha-hydroxy malonates (tartronic esters) in good to excellent yields with high enantioselectivities. Preliminary mechanistic studies indicated that hemiketalization, rather than the dynamic kinetic resolution of hemiketal, was the enantiodetermining step under our reaction conditions.
