The impact of intensive milk production on oocyte quality as well as early embryo development has been established but few analyses have addressed this question at the initiation of implantation, a critical milestone ensuring a successful pregnancy and normal post-natal development. Our study aimed to determine if contrasted maternal metabolism affects the previously described sensory properties of the endometrium to the conceptus in cattle. Following embryo transfer at Day 7 post-oestrus, endometrial caruncular (CAR) and intercaruncular (ICAR) areas were collected at Day 19 from primiparous postpartum Holstein-Friesian cows that were dried-off immediately after parturition (i.e., never milked; DRY) or milked twice daily (LACT). Gene quantification indicated no significant impact of lactation on endometrial expression of transcripts previously reported as conceptus-regulated (PLET1, PTGS2, SOCS6) and interferon-tau stimulated (RSAD2, SOCS1, SOCS3, STAT1) factors or known as female hormone-regulated genes (FOXL2, SCARA5, PTGS2). Compared with LACT cows, DRY cows exhibited mRNA levels with increased expression for FOXL2 transcription factor and decreased expression for oxidative stress-related genes (CAT, SOD1, SOD2). In vivo and in vitro experiments highlighted that neither interferon-tau nor FOXL2 were involved in transcriptional regulation of CAT, SOD1 and SOD2. In addition, our data showed that variations in maternal metabolism had a higher impact on gene expression in ICAR areas. Collectively, our findings prompt the need to fully understand the extent to which modifications in endometrial physiology drive the trajectory of conceptus development from implantation onwards when maternal metabolism is altered.

Abstract

Theme: Early Development and Pregnancy; Extracellular vesicles (EVs) are crucial for intercellular communications and they may play important role in the delivery of molecular messages during the pre-implantation period of pregnancy. EVs have been isolated from uterine luminal fluid (ULF) in human and sheep. Recent data have provided evidence that EVs contained in ovine ULF can penetrate endometrial epithelial cells after a 6-days infusion in vivo. Nevertheless, embryo implantation involves rapid and dynamic changes in molecular interactions with the endometrium. Our present work aims to determine whether EVs collected from ULF interact with endometrial epithelial cells and modify cell physiology after a short time of in vitro and in vivo incubation conditions.Primary cultures of endometrial epithelial cells were derived from ovine uteri on day 12 post-oestrus. EVs were purified from ovine ULFs on Day 14 of pregnancy (2 days before conceptus implantation). The presence of EVs in ULF was confirmed by transmission electron microscopic observation. ULF EVs were labeled with lipophilic PKH26 fluorescent dye and then incubated with primary cultures of epithelial cells during 30 min to 24h. Confocal microscopy analyses revealed an uptake of EVs as early as 30 minutes after incubation, followed by a progressive increase of intracellular fluorescence up to 6 hours.For the in vivo study, ovine ULF EVs isolated on Day 14 of pregnancy were labeled with PKH26 fluorescent dye and infused into the uterine lumen of cyclic ewes on Day 12 post-oestrus. After 24h, numerous epithelial cells from the luminal layer and superficial glands exhibited an intensive fluorescence signal, whereas deep endometrial glands displayed few fluorescent cells. No signal was detectable in the stroma. The impact of EVs on endometrial function was investigated by quantifying transcript expression of a selection of endometrial genes. First data pointed out that expression of two genes, including the Myxovirus-Resistance Protein (MX1) was up-regulated following EVs uptake by the endometrial epithelium. This work provides first evidence that ovine EVs from pregnant ULF can (i) enter in endometrial epithelial cells within 30 min in vitro or 24h in vivo (ii) modulate expression of endometrial gene expression known to be critical for embryo implantation. These results suggest a critical role for EVs in the preparation of endometrium when implantation iniates.

Abstract

International audience; Thyroid-stimulating hormone (TSH) is the main regulator of thyroid gland function, and elevated TSH in humans is known to be associated with infertility. The aim was to establish an ELISA for measurement of bovine TSH and to compare TSH serum concentrations of early pregnant (n=19) and non-pregnant (n=19) Holstein Friesian heifers. For the TSH ELISA, an antibody targeted against bovine TSH (anti-bovine TSH, 1:10 pre-diluted, AFP-642482Rb, obtained from the National Hormone & Peptide Program (NHPP), NIDDK and Dr. Parlow) was diluted (1:2500). The applied standard curve ranged from 0.2–100 ng/ml bovine TSH (AFP-8755B, obtained from the NHPP), and biotin-labeled TSH was used to detect binding with subsequent addition of streptavidin horseradish peroxidase solution. The intraassay CV was determined by measuring one bovine sample 20 times. Serum samples of 19 matched pairs were used to compare pregnant and non-pregnant heifers. Blood was taken from the jugular vein on the Day of ovulation (defined by ultrasound; Day 0) and Days 14 and 18. International audience; It is known that changes of sexual steroid hormones during cycle and early pregnancy affect maternal metabolism. But when do embryo-factors start to take influence? The aim was to investigate metabolic changes in cattle already on Day 18. Blood samples of Holstein Friesian heifers (n=30) were taken on Day 0 (Day of ovulation), 7, 14, 16 and 18 in repeated consecutive pregnant and non-pregnant periods. Abstract

This study was conducted to determine if the main components of the somatotropic axis change during the early phase of pregnancy in the maternal blood system and whether differences exist on day 18 after pregnancy recognition by the maternal organism. Blood samples of pregnant heifers (Holstein Friesian; n = 10 after embryo transfer) were obtained on the day of ovulation (day 0), as well as on days 7, 14, 16 and 18 and during pregnant, non-pregnant and negative control cycles. The concentrations of progesterone (P-4), oestrogen, growth hormone (GH), insulin-like growth factor-1 and -2 (IGF1, -2) and IGF-binding protein-2, -3 and -4 (IGFBP2, -3, -4) were measured. The mRNA expressions of growth hormone recefitor 1A, IGF1, IGF2, IGFBP2, IGFBP3 and IGFBP4 were detected using RT-qPCR in liver biopsy specimens (day 18). In all groups, total serum IGF1 decreased from day 0 to 16. Notably, IGFBP4 maternal blood concentrations were lower during pregnancy than during non-pregnant cycles and synchronized control cycles. It can be speculated that the lower IGFBP4 in maternal blood may result in an increase of free IGF1 for local action. Further studies regarding IGFBP4 concentration and healthy early pregnancy are warranted.

Abstract

Présentée le 27 septembre 2006 Diplôme : Dr. d'Universite; La mise en évidence d’une agueusie spécifique au glutamate monosodique (MSG) chez 3,5% de la population française (Lugaz et al., 2002) nous a conduit à étudier l’implication des récepteurs gustatifs dans ce phénomène. Nous avons utilisé des papilles fongiformes prélevées chez des sujets agueusiques et normogueusiques afin d’explorer et comparer les différents récepteurs au MSG présents dans ces papilles. Ainsi par RT-PCR et immunohistochimie, nous avons montré la présence de T1R1 et T1R3 chez tous les types de sujets. Par ailleurs, il semble que les récepteurs métabotropiques du glutamate de type 2 (mGluR2) et de type 4 (mGluR4) soient présents dans les papilles fongiformes de quelques sujets. L’analyse de la séquence codante des gènes TAS1R1 et TAS1R3 nous a permis d’identifier 3 polymorphismes qui aboutissent à une substitution d’acides aminés dans TAS1R1 : SNP329 (A110V), SNP1039 (K347E) et SNP1114 (A372T), ainsi que 5 polymorphismes faux-sens dans TAS1R3 : SNPs 13 et 15 (A5T), SNP2203 (A735T), SNP2246 (F749S), SNP2269 (R757C). L’allèle 329 est le seul associé à l’agueusie au MSG mais il n’est ni nécessaire ni suffisant. Nous n’avons pas observé de substitutions dans mGluR4 chez les sujets agueusiques testés. Etant donné le manque de corrélation entre polymorphismes et agueusie et en fonction de la multimodalité observée dans les distributions de sensibilités des individus au MSG au niveau du seuil, il semble raisonnable de proposer que la sensibilité au MSG et/ou au GMP implique la contribution de plusieurs allèles des gènes TAS1R1 et TAS1R3 ou d’autres récepteurs gustatifs comme le mGluR2, le mGluR4 ou encore d’autres facteurs non géniques ou encore la combinaison de tous ces facteurs.

Abstract

Background: Before the onset of fetal thyroid hormone production, the transplacental delivery of maternal thyroid hormones is necessary for embryonic and fetal development. Therefore, the adaptation of maternal thyroid hormone metabolism may be important for pregnancy success and embryo survival. The aims of this study were to determine the thyroid hormone levels during the early peri-implantation period until day 18 and on the day of ovulation, to determine whether pregnancy success is dependent on a “normothyroid status” and to determine whether physiological adaptations in maternal thyroid hormone metabolism occur, which may be necessary to provide sufficient amounts of biologically active T3 to support early pregnancy. Therefore, blood samples obtained on the day of ovulation (day 0) and days 14 and 18 of the Holstein–Friesian heifers (n = 10) during the respective pregnant, non-pregnant and negative control cycles were analyzed for thyroid-stimulating-hormone (TSH), thyroxine (T4) and triiodothyronine (T3). Liver biopsies (day 18) from pregnant and respective non-pregnant heifers were analyzed for mRNA expression of the most abundant hepatic thyroid hormone deiodinase (DIO1) by real time qPCR. Results: Although liver DIO1 mRNA expression did not differ between the pregnant and non-pregnant heifers on day 18, the serum concentrations of TSH and T3 on day 18 were higher in non-pregnant heifers compared to pregnant heifers (P < 0.05). Moreover, T3 decreased between day 0 and 18 in pregnant heifers (P < 0.001). Conclusions: In conclusion, no associations between thyroid hormone patterns on day 18 and pregnancy success were detected. During the early peri-implantation period, TSH and T3 may be affected by the pregnancy status because both TSH and T3 were lower on day 18 in pregnant heifers compared to non-pregnant dairy heifers. In further studies, the thyroid hormone axis should be evaluated throughout the entire gestation to confirm these data and identify other possible effects of pregnancy on the thyroid hormone axis in cattle.
