ABSTRACT: The bacterium Lactobacillus kefiranofaciens OSU-BDGOA1 and yeast Kluyveromyces marxianus bdgo-ym6 were previously isolated from kefir grains and have shown probiotic traits in mono- and coculture. This research evaluates the effect of introducing probiotic kefir microorganisms in monoculture and in coculture alongside yogurt starter cultures on the physicochemical and rheological properties, volatile flavor compounds, survival of the microorganisms during simulated digestion, and sensory attributes of the final fermented products. The incorporation of L. kefiranofaciens OSU-BDGOA1 in monoculture showed promising outcomes, resulting in a final product showing more solid-like characteristics and potentially improving the texture of the product. There was also a significant increase in the concentration of desirable volatile flavor compounds in the yogurt with the monoculture, particularly 2,3-butanedione, displaying a positive correlation with buttery flavor in the sensory analysis. The inclusion of L. kefiranofaciens in monoculture also promoted better sensory attributes and was significantly better than the yogurt with the coculture with the yeast, showing promising results for the incorporation of this probiotic bacterium into functional fermented dairy products.

ABSTRACT: Four dairy foods processing by-products (acid whey permeate [AWP], buttermilk [BM], sweet whey permeate [SWP], and sweet whey permeate with added milk fat globule ingredient [SWP+MFGM]) were fermented for 4 wk and compared with traditional kefir milks for production of novel kefir-like dairy products. Sweet whey permeates and SWP supplemented with 1.5% milk fat globule membrane (MFGM) proved to be the most viable by-products for kefir grain fermentation, exhibiting diverse abundance of traditional kefir microorganisms and positive indicators of bioactive properties. Grain viability was assessed with shotgun metagenomics, texture profile analysis, live cell counts, and scanning electron microscopy. Assessed bioactivities of the kefir-like products included antibacterial, antioxidant, potential anticancerogenic properties, and membrane barrier effects on human colorectal adenocarcinoma Caco-2 cells. All kefir grains were most abundant in Lactobacillus kefiranofaciens when analyzed with shotgun metagenomics. When analyzed with live cell counts on selective media, AWP kefir-like product had no countable Lactococcus spp., indicating suboptimal conditions for kefir grain microbiota survival and application for fermented dairy starter culture bacterium. Live cell counts were affirmed with kefir grain surface scanning electron microscopy images. The SWP treatment had the most adhesive kefir grain surface, and SWP+MFGM had the largest exopolysaccharide yield from grain extraction. All kefir and kefir-like products were able to achieve a 6-log reduction against Listeria innocua and Escherichia coli. Traditional milk kefirs had the highest antioxidant capacity for 2,2-diphenyl-1-picrylhydrazyl (DPPH) and the 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid; ABTS) assay. The AWP formulation had a significantly higher DPPH antioxidant activity compared with the other kefir and kefir-like products, and SWP had the lowest Trolox equivalence concentration in the ABTS assay. Sweet whey and supplemented milk fat sweet whey had upregulation of Cldn-1 and Ocln-1 gene expression, which correspond with a significant increase in transepithelial electrical resistance.

Abstract Mammalian sex determination is controlled by antagonistic gene cascades operating in embryonic undifferentiated gonads. The expression of the Y-linked gene SRY is sufficient to trigger the testicular pathway, whereas its absence in XX embryos leads to ovarian differentiation. Yet, the potential involvement of non-coding regulation in this process remains unclear. Here we show that the deletion of a single microRNA cluster, miR-17~92, induces complete primary male-to-female sex reversal in XY mice. Sry expression is delayed in XY knockout gonads, which develop as ovaries. Sertoli cell differentiation is reduced, delayed and unable to sustain testicular development. Pre-supporting cells in mutant gonads undergo a transient state of sex ambiguity which is subsequently resolved towards the ovarian fate. The miR-17~92 predicted target genes are upregulated, affecting the fine regulation of gene networks controlling gonad development. Thus, microRNAs emerge as key components for mammalian sex determination, controlling Sry expression timing and Sertoli cell differentiation.

Summary: Background: The clinical relevance of recurrent venous thromboembolism (VTE) after discontinuing anticoagulation in patients with COVID-19-associated VTE remains uncertain. We estimated the incidence rates and mortality of VTE recurrences developing after discontinuing anticoagulation in patients with COVID-19-associated VTE. Methods: A prospective, multicenter, non-interventional study was conducted between March 25, 2020, and July 26, 2023, including patients who had discontinued anticoagulation after at least 3 months of therapy. All patients from the registry were analyzed during the study period to verify inclusion criteria. Patients with superficial vein thrombosis, those who did not receive at least 3 months of anticoagulant therapy, and those who were followed for less than 15 days after discontinuing anticoagulation were excluded. Outcomes were: 1) Incidence rates of symptomatic VTE recurrences, and 2) fatal PE. The rate of VTE recurrences was defined as the number of patients with recurrent VTE divided by the patient-years at risk of recurrent VTE during the period when anticoagulation was discontinued. Findings: Among 1106 patients with COVID-19-associated VTE (age 62.3 ± 14.4 years; 62.9% male) followed-up for 12.5 months (p25-75, 6.3–20.1) after discontinuing anticoagulation, there were 38 VTE recurrences (3.5%, 95% confidence interval [CI]: 2.5–4.7%), with a rate of 3.1 per 100 patient-years (95% CI: 2.2–4.2). No patient died of recurrent PE (0%, 95% CI: 0–7.6%). Subgroup analyses showed that patients with diagnosis in 2021–2022 (vs. 2020) (Hazard ratio [HR] 2.86; 95% CI 1.45–5.68) or those with isolated deep vein thrombosis (vs. pulmonary embolism) (HR 2.31; 95% CI 1.19–4.49) had significantly higher rates of VTE recurrences. Interpretation: In patients with COVID-19-associated VTE who discontinued anticoagulation after at least 3 months of treatment, the incidence rate of recurrent VTE and the case-fatality rate was low. Therefore, it conceivable that long-term anticoagulation may not be required for many patients with COVID-19-associated VTE, although further research is needed to confirm these findings. This article aims to carry out an analysis of the new regulatory framework regarding medical devices derived from the approval and entry into force of Royal Decree 192/2023, on medical devices, which adapts and develops the European Union Regulation 2017/745, which meant a new paradigm shift in the European regulation of this matter. It is, therefore, a matter of carrying out a competence analysis and, likewise, of studying which novelties this manifestation of state regulatory power offers in comparison with the regulation provided for in the state regulations of 2009, also focusing on some enforcement problems and identifying possible solutions also.

ABSTRACT: β-galactosidase (enzymatic class 3.2.1.23) is one of the dairy industry's most important and widely used enzymes. The enzyme is part of a large family known to catalyze hydrolysis and transglycosylation reactions. Its hydrolytic activity is commonly used to decrease lactose content in dairy products, while its transglycosylase activity has recently been used to synthesize galacto-oligosaccharides (GOS). During the past couple of years, researchers have focused on studying β-galactosidase isolated and purified from lactic acid bacteria. This review will focus on β-galactosidase purified and characterized from what used to be the Lactobacillus genera. Furthermore, particular emphasis is given to its kinetics, biochemical characteristics, GOS production, market, and utilization by Lactobacilllaceae species.

This work presents a review of the onshore wind turbines foundations, especially about the different foundation design concepts available in the onshore business, the analysis of some of them and the trends.

Objectives: The aim of the study was to develop and validate a questionnaire to assess attitudes, knowledge and difficulties in the management of social problems by paediatric care providers. Methods: The development of the tool started with a conceptualization phase, followed by the design, pilot testing and psychometric evaluation of the questionnaire based on the responses obtained from 407 professionals. We performed exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) to assess the construct validity of the questionnaire. We used the normed fit index (NFI), the root mean square error of approximation (RMSEA), the χ2 test and the comparative fit index (CFI) to test the goodness of fit. We assessed reliability through the Cronbach’s α coefficient of internal consistency. All quantitative analyses were performed with the Stata/SE software, version16.1. Results: The EFA identified twelve factors. All factors exhibited a good internal consistency (Cronbach’s α, 0.8434). The CFA showed the model was a good fit (RMSEA = 0.037). The NAI and CAI values were 0.742 and 0.797, respectively. Conclusions: This questionnaire comprising 138 items distributed in twelve factors is a reliable and valid instrument to analyse the attitudes, knowledge and difficulties in the approach to social problems in children by paediatricians, which will allow the design of interventions according to the needs and deficiencies identified through it. Resumen: Objetivos: Este estudio tiene como objetivo desarrollar y validar un cuestionario para evaluar actitudes, conocimientos y dificultades en el manejo de problemas sociales por parte de los profesionales de pediatría. Métodos: El desarrollo de la herramienta comenzó con una fase de conceptualización, seguida del diseño, prueba piloto y evaluación de las propiedades psicométrica del cuestionario tras obtener respuestas de 407 profesionales. Se realizaron análisis factoriales exploratorios (AFE) y análisis factoriales confirmatorios (AFC) para explorar la validez de constructo del cuestionario. El índice de ajuste normado (IAN), el error cuadrático medio de aproximación (ECMA), la prueba de Chi-cuadrado y el índice de ajuste comparativo (IAC) se utilizaron para probar la bondad del ajuste. La confiabilidad se exploró a través del alfa de Cronbach para la consistencia interna. Resultados: La AFE identificó doce factores. Todos los factores mostraron buena consistencia interna (alfa de Cronbach 0.8434). El AFC mostró un buen ajuste al modelo (ECMA = 0.037). Los valores de IAN y IAC fueron 0.742 y 0.797 respectivamente. Todos los análisis cuantitativos se llevaron a cabo con el software STATA/SE v.16.1. Conclusiones: Este cuestionario de 138 items distribuidos en doce factores es un instrumento fiable y válido para analizar actitudes, conocimientos y dificultades en el abordaje de problemas sociales en la infancia por parte de los pediatras, que permitirá diseñar intervenciones, de acuerdo con las necesidades y carencias identificadas a través del mismo.

Abstract FBXW7 is a member of the F-box protein family, which functions as the substrate recognition component of the SCF E3 ubiquitin ligase. FBXW7 is a main tumor suppressor due to its ability to control proteasome-mediated degradation of several oncoproteins such as c-Jun, c-Myc, Cyclin E1, mTOR, and Notch1-IC. FBXW7 inactivation in human cancers results from a somatic mutation or downregulation of its protein levels. This work describes a novel regulatory mechanism for FBXW7 dependent on the serine/threonine protein kinase DYRK2. We show that DYRK2 interacts with and phosphorylates FBXW7 resulting in its proteasome-mediated degradation. DYRK2-dependent FBXW7 destabilization is independent of its ubiquitin ligase activity. The functional analysis demonstrates the existence of DYRK2-dependent regulatory mechanisms for key FBXW7 substrates. Finally, we provide evidence indicating that DYRK2-dependent regulation of FBXW7 protein accumulation contributes to cytotoxic effects in response to chemotherapy agents such as Doxorubicin or Paclitaxel in colorectal cancer cell lines and to BET inhibitors in T-cell acute lymphoblastic leukemia cell lines. Altogether, this work reveals a new regulatory axis, DYRK2/FBXW7, which provides an understanding of the role of these two proteins in tumor progression and DNA damage responses.

The goblet cells of the gastrointestinal tract (GIT) produce glycoproteins called mucins that form a protective barrier from digestive contents and external stimuli. Recent evidence suggests that the milk fat globule membrane (MFGM) and its milk phospholipid component (MPL) can benefit the GIT through improving barrier function. Our objective was to compare the effects of two digested MFGM ingredients with or without dextran sodium sulfate (DSS)-induced barrier stress on mucin proteins. Co-cultured Caco-2/HT29-MTX intestinal cells were treated with in vitro digests of 2%, 5%, and 10% (w/v) MFGM or MPL alone for 6 h or followed by challenge with 2.5% DSS (6 h). Transepithelial electrical resistance and fluorescein isothiocyanate (FITC)-dextran (FD4) permeability measurements were used to measure changes in barrier integrity. Mucin characterization was performed using a combination of slot blotting techniques for secreted (MUC5AC, MUC2) and transmembrane (MUC3A, MUC1) mucins, scanning electron microscopy (SEM), and periodic acid Schiff (PAS)/Alcian blue staining. Digested MFGM and MPL prevented a DSS-induced reduction in secreted mucins, which corresponded to the prevention of DSS-induced increases in FD4 permeability. SEM and PAS/Alcian blue staining showed similar visual trends for secreted mucin production. A predictive bioinformatic approach was also used to identify potential KEGG pathways involved in MFGM-mediated mucosal maintenance under colitis conditions. This preliminary in silico evidence, combined with our in vitro findings, suggests the role of MFGM in inducing repair and maintenance of the mucosal barrier.

In México, the utilization of fast-growing plant species for biomass production presents transportation-related issues such as freshness reduction, the transportation of large volumes with minimal weight due to low density, limited storage availability, and logistical challenges. To enhance understanding in this field, the research aimed to evaluate the densification potential of a variety of organic materials, with a specific emphasis on the biomass derived from eight species of rapidly growing plants, three animal species’ manure, and concentrated feed. After conditioning, 100 g samples underwent particle size analysis and were categorized into seven sizes. Pellets and briquettes (40 g, 10% moisture, 15 MPa pressure, 5 min processing, 80 °C temperature) were produced and evaluated for bulk density (BD) and pellet (PD) and briquette durability (BDU). The predominant particle sizes were 0.850 mm and 0.425 mm. Original biomass bulk density (OBBD) varied notably, with ovine manure (0.50 g cm−3) and cattle feed (0.49 g cm−3) exhibiting the highest values. Caprine manure (0.83 g cm−3) and ovine manure (0.78 g cm−3) yielded the densest pellets at 1.76 and 1.84 g cm−3, respectively. Apple tree pellets achieved premium quality with the highest hardness (97.9%). Cattle manure (1.25 g cm−3) and cattle feed (1.25 g cm−3) had the densest briquettes, with notable BBD/OBBD ratios in pine sawdust (4.6) and corn (4.5). Caprine manure and Acacia biomass briquettes showed premium quality with the highest hardness (99.1%), emphasizing densification variations and the need for tailored approaches based on organic material characteristics.

Bifidobacterium longum subsp. infantis is associated with the gut microbiota of breast-fed infants. Bifidobacterium infantis promotes intestinal barrier and immune function through several proposed mechanisms, including interactions between their surface polysaccharides, the host, and other gut microorganisms. Dairy foods and ingredients are some of the most conspicuous food-based niches for this species and may provide benefits for their delivery and efficacy in the gut. Milk phospholipid (MPL)-rich ingredients have been increasingly recognized for their versatile benefits to health, including interactions with the gut microbiota and intestinal cells. Therefore, our objective was to investigate the capacity for MPL to promote survival of B. infantis during simulated digestion and to modulate bacterial polysaccharide production. To achieve these aims, B. infantis was incubated with or without 0.5% MPL in de Man, Rogosa, and Sharpe (MRS) media at 37°C under anaerobiosis. Survival across the oral, gastric, and intestinal phases using in vitro digestion was measured using plate count, along with adhesion to goblet-like intestinal cells. MPL increased B. infantis survival at the end of the intestinal phase by at least 7% and decreased adhesion to intestinal cells. The bacterial surface characteristics, which may contribute to these effects, were assessed by ζ-potential, changes in surface proteins using comparative proteomics, and production of bound polysaccharides. MPL decreased the surface charge of the bifidobacteria from −17 to −24 mV and increased a 50 kDa protein (3-fold) that appears to be involved in protection from stress. The production of bound polysaccharides was measured using FTIR, HPLC, and TEM imaging. These techniques all suggest an increase in bound polysaccharide production at least 1.7-fold in the presence of MPL. Our results show that MPL treatment increases B. infantis survival during simulated digestion, induces a stress resistance surface protein, and yields greater bound polysaccharide production, suggesting its use as a functional ingredient to enhance probiotic and postbiotic effects.

Este ensayo analiza la necesaria, pero subestimada aclaración de las diferencias –conceptuales y de implicación pedagógica– entre la escuela y la educación, la cual permite encadenar las promesas, estrategias y acciones que el “sistema educativo costarricense” ha desarrollado, pero que merecen ser contrastadas con las condiciones materiales para el cumplimiento de los objetivos ministeriales propuestos. Se toman en consideración las diferencias –abismales– entre lo urbano y lo rural, entre las instituciones públicas, privadas y subvencionadas, con la intención de establecer un primer trazo de una propuesta crítica y propositiva, la cual se asumirá como un compromiso por mejorar el aprendizaje de los niños, niñas y juventudes, en tiempos de cambio y ajuste.

One of the most important challenges of agriculture today is increasing its productivity gains, while controlling its environmental footprint. Because of that plastic covered greenhouses (PCG) mapping via remote sensing is receiving a great attention throughout this century. In this study, a fair comparison was carried out in four PCG study areas around the world to test 14 spectral indices mainly focused on the detection of plastic. To the best knowledge of the authors, this is the first research that fairly compares all these spectral indices in such variable number of study sites. The applied OBIA approach was based on the combined use of very high-resolution satellite data (Deimos-2 pansharpened images) to address the segmentation process and Sentinel-2 time series to compute the spectral indices. When dealing with Sentinel-2 single images, the Plastic GreenHouse Index (PGHI) stood out among all the indices tested in the study areas dedicated to the cultivation of vegetables, such as the cases of Almería (Spain), Agadir (Morocco) and Antalya (Turkey). Better Overall Accuracy (OA) values of 94.09%, 92.27%, 92.77% and 92.17% were achieved for Almería, Agadir, Bari and Antalya study sites, respectively, when using statistical seasonal spectral indices based on Sentinel-2 time series, being the maximum and mean values of PGHI (MAX (PGHI) and MEAN (PGHI)) the best ranked. Meanwhile, the PCG area of Bari (Italy), with a monoculture in vineyards, presented the worst and most irregular results.

IntroductionDue to the increasing consumer demand for the development and improvement of functional foods containing probiotics, new probiotic candidates need to be explored as well as novel means to enhance their beneficial effects. Lactobacillus kefiranofaciens OSU-BDGOA1 is a strain isolated from kefir grains that has demonstrated probiotic traits. This species is the main inhabitant of kefir grains and is responsible for the production of an exopolysaccharide (EPS) whit vast technological applications and potential bioactivities. Research has shown that interkingdom interactions of yeast and lactic acid bacteria can enhance metabolic activities and promote resistance to environmental stressors.MethodsComparative genomic analyses were performed to distinguish OSU-BDGOA1 from other strains of the same species, and the genome was mined to provide molecular evidence for relevant probiotic properties. We further assessed the cumulative effect on the probiotic properties of OSU-BDGOA1 and Kluyveromyces marxianus bdgo-ym6 yeast co-culture compared to monocultures.ResultsSurvival during simulated digestion assessed by the INFOGEST digestion model showed higher survival of OSU-BDGOA1 and bdgo-ym6 in co-culture. The adhesion to intestinal cells assessed with the Caco-2 intestinal cell model revealed enhanced adhesion of OSU-BDGOA1 in co-culture. The observed increase in survival during digestion could be associated with the increased production of EPS during the late exponential and early stationary phases of co-culture that, by enhancing co-aggregation between the yeast and the bacterium, protects the microorganisms from severe gastrointestinal conditions as observed by SEM images. Immune modulation and barrier function for recovery and prevention of flagellin-mediated inflammation by Salmonella Typhimurium heat-killed cells (HKSC) in Caco-2 cells were also measured. OSU-BDGOA1 in mono- and co-culture regulated inflammation through downregulation of pro-inflammatory cytokine expression and increased membrane barrier integrity assessed by TEER, FD4 permeability, and expression of tight junctions.DiscussionThe results of the study warrant further research into the application of co-cultures of yeast and LAB in functional probiotic products and the potential to increase EPS production by co-culture strategies.

Se requieren materias primas para elaborar biocombustibles y transitar hacia fuentes sostenibles de energía. El objetivo del estudio fue evaluar la morfología, productividad y calidad del grano en higuerilla (Ricinus communis) cultivada en el Centro-Norte de México. En primavera-verano de 2020, se sembraron cuatro variedades y un h.brido de higuerilla en Durango, Durango, y Pabellón, Aguascalientes, México. En la siembra se consideró una densidad de 15000 plantas ha−1. Se evalu. n.mero de d.as a floraci.n (DF), altura de planta (AP), longitud de la espiga (LE), rendimiento (kg ha−1), peso de 100 semillas (P100S), % de brotación (PB), % de germinación (PG) y el contenido de cenizas (CC), proteína (CP) y grasa (CG). El procesamiento estadístico incluyo estad.stica descriptiva (DF, PB y PG) y un análisis de varianza en un diseño en bloques completamente al azar con seis repeticiones y la comparación de medias se obtuvo con la prueba de Tukey (α ≤ 0.05). Se observaron diferencias altamente significativas (α ≤ 0.01) para altura de planta entre localidades (195.0 cm-282.7 cm) y entre materiales para AP, LE y P100S. En ambas localidades SAB 14 (2,906 kg ha−1 y 2,408 kg ha−1) y K 855 (2,669 kg ha−1 y 2,400 kg ha−1) mostraron el rendimiento m.s alto. En CG Guanajuato Oil (57.1%) y SAB 14 (55.4%) superaron estad.sticamente al resto de los materiales. Se corroboró la adaptación de SAB 14, la cual mostró rendimiento alto de grano y sus semillas, resultaron de calidad aceptable, con base en el contenido de grasa.

Photovoltaic panels are exposed to various external factors that can cause damage, with the formation of cracks in the photovoltaic cells being one of the most recurrent issues affecting their production capacity. Electroluminescence (EL) tests are employed to detect these cracks. In this study, a methodology developed according to the IEC TS 60904-13 standard is presented, allowing for the calculation of the percentage of type C cracks in a PV panel and subsequently estimating the associated power loss. To validate the methodology, it was applied to a polycrystalline silicon module subjected to incremental damage through multiple impacts on its rear surface. After each impact, electroluminescence images and I-V curves were obtained and used to verify power loss estimates. More accurate estimates were achieved by assessing cracks at the PV cell level rather than by substring or considering the entire module. In this context, cell-level analysis becomes indispensable, as the most damaged cell significantly influences the performance of the photovoltaic model. Subsequently, the developed methodology was applied to evaluate the conditions of four photovoltaic panels that had been in operation, exemplifying its application in maintenance tasks. The results assisted in decision making regarding whether to replace or continue using the panels.

The objective of this work was the study of four experimental plastic covers (EPCs) to assess the improvement in microclimatic conditions inside a greenhouse. This experiment was based on the comparison of four trilayer plastics: blue, grey, yellow and colorless ECPs. Radiometric and mechanical properties were studied along with the films’ behavior under semi-field conditions through their microclimate parameters. The results show that the addition of the blue pigment causes a considerable reduction in transmission (32.04%) with a reduction in the maximum temperature (+9.8 °C) compared to the other films (14% for grey, 52% for yellow and 46% for colorless). The anti-thermal additive used in the grey EPC did not achieve the desired effect, since it reduced both the photosynthetic active radiation (PAR) (62.33%) and near-infrared radiation (NIR) (62.83%) transmission equally. None of the EPCs achieved a PAR/NIR ratio greater than 1 (0.52 for blue, 0.99 for grey, 0.98 for yellow and 0.99 for colorless). Hindered amine light stabilizers (HALSs) photo-stabilizing additives block UV-A radiation (36.85 for grey) more efficiently compared to nickel quenchers (38.64 for yellow), as they allow earlier PAR transmission. The tensile test showed that all the EPCs manifested a linear relationship between stress and deformation, which defines Young’s modulus.

The milk fat globule membrane (MFGM) imparts human health benefits ranging from improved immune system, gut, and brain function to improved cardiometabolic health. The industry’s growing interest in introducing MFGM-enriched foods requires scientific evidence that the benefits derived from this compound are not affected by the formulation or processes that may alter its function, such as the digestion process. In this study, the impact of food matrices and supplementation levels on the bioaccessibility and assimilation of MFGM lipids in cell culture was investigated. Three food matrices including a protein-rich jelly, carbohydrate-rich cookie, and a carbohydrate- and fat-rich cookie with sunflower oil (SF-cookie) were supplemented with an MFGM ingredient derived from cottage cheese acid whey at 2, 5, and 10% (w/w). Each formulation underwent simulated digestion consisting of oral, gastric, and intestinal phases, and the micellar fraction was collected for both analysis and lipid assimilation in Caco-2 intestinal cells. The micellar fractions were diluted and applied to the cells for 4 h. A lipidomic approach was used to assess the lipid profiles of micellar fractions and intestinal cells. The micelles from digested jellies, cookies, and SF-cookies containing MFGM showed a distinct separation using partial least squares discriminant analysis (PLS-DA). Both correlation loadings and variable importance in projection (VIP) scores demonstrated a tendency of MFGM polar lipids (ceramides, glucosylceramides) for micelles from digested jelly, whereas micelles from digested cookies were associated with MFGM neutral lipids (free fatty acids, cholesterol, etc.). The effect of supplementation level on the micellar lipid profiles reinforced this pattern. The lipid profiles of intestinal cells after incubation with the micellar fractions differed considerably from the corresponding micellar lipid profiles. Specifically, the SF-cookie-treated cells were associated with a greater abundance of PUFA relative to jelly- and cookie-treated cells; however, increasing MFGM supplementation showed irregular patterns and rearrangement of cellular lipid profiles, suggesting the cells’ role in regulating lipid metabolism in response to nutritional stimuli. The nature of lipid micellarization and assimilation in intestinal cells from MFGM-containing food formulations echoes the complexity of lipids inherent to the MFGM itself, suggesting the need for application-based MFGM supplementation.

Las nanopartículas de plata (NPs-Ag) son de gran interés debido a las aplicaciones potenciales en diferentes áreas. En busca de metodologías que no presenten riesgos a la salud y sean amigables con el ambiente, la biosíntesis de nanopartículas ha sido una opción viable. Se sabe que diversas biomoléculas presentes en los microrganismos pueden tener las funciones de agente oxidante, reductor y estabilizador, favoreciendo la formación de nanopartículas. En el presente trabajo se tuvo como objetivo la biosíntesis, caracterización y evaluación de la actividad antibacteriana de NPs-Ag. La biosíntesis se realizó a partir de sobrenadante de un cultivo de Pseudomonas aeruginosa, las nanopartículas se caracterizaron mediante espectroscopía UV-VIS, microscopía electrónica de barrido y difracción de rayos X, y la actividad antibacteriana se evaluó con ensayos de dilución en caldo Mueller Hinton, se determinó la concentración mínima inhibitoria (CMI) y concentración mínima bactericida (CMB) sobre bacterias Gram positivas y negativas. Se encontró que la mejor síntesis fue con una mezcla de 20% de sobrenadante conteniendo 3 mM de AgNO3, observando nanoestructuras con morfologías mixtas. Presentaron actividad contra bacterias Gram positivas y negativas con CMI de 1 a 2 µg/ml y CMB de 2 a 4 µg/ml. En conclusión, el sobrenadante de cultivos de P. aeruginosa es una alternativa viable para la síntesis de NPs-Ag con actividad antibacteriana.

ABSTRACT: Kefir is a fermented dairy product with well recognized probiotic properties. Recently, consumer interest in fermented products with probiotic microorganisms has increased due to the accumulating evidence of the effects of kefir microorganisms on the modulation of gut microbiota and their antimicrobial activity. Although the health properties of kefir have been reviewed in other works, the present review addresses the antimicrobial effects of kefir microbiota and associated compounds. The antimicrobial activity of kefir microorganisms could derive from different mechanisms. The microorganisms' capacity to adhere to the intestinal epithelium, preventing the adhesion of pathogens, and their immunomodulation properties are among the mechanisms suggested. Bacteria and yeast isolated from kefir have been shown to have in vivo and in vitro antimicrobial activity against enteropathogenic bacteria and spoilage fungi. However, most reports have focused their approach on single-strain antimicrobial properties; evaluation of antimicrobial activity of cocultures of kefir microbiota and their potential mechanisms of action has been neglected. Kefir microbiota and associated compounds have shown promising antimicrobial effects; however, more research needs to be done to discern the mechanisms of action.

In the midst of rising consumer health and environmental concerns, pea protein has increased in popularity as an alternative to animal-origin proteins. However, the use of pea protein in food systems is largely hindered by its poor functionality, including low solubility. The objective of this study was to measure the textural, functional, and rheological properties of a mixed plant- and animal-based protein system. Caseins, the major protein in bovine milk, are a known animal-based protein with optimal functional properties and high sensory acceptability. Through cold-temperature homogenization, insoluble pea proteins were incorporated with casein micelles in a stable, mixed, colloidal dispersion. Three blends with various casein-to-pea ratios (90:10, 80:20, 50:50) were prepared and analyzed. We hypothesized that incorporation with casein micelles would improve the poor functional properties of pea protein, and thus increase its potential uses in the food industry as a functional ingredient. The protein blend successfully underwent chymosin coagulation, a key ability of caseins, and formed protein gels with textures similar to commercial queso fresco and hard tofu. The 50% casein micelle:50% pea protein blend had better emulsification properties than pea protein alone. In contrast, this blend had the same foaming properties as pea protein alone. The mixed protein blends had similar rheological properties to skim milk, thus increasing their potential applications in the food industry. These results serve as a starting point to begin fully understanding the interactions between pea protein isolate and casein micelles combined via low-temperature homogenization and the effect on their techno-functional properties.

β-Galactosidase is an enzyme produced by some strains of lactic acid bacteria (LAB) commonly found in dairy products; however, industrial demand for these enzymes is still low. Acid whey (AW), a lactose-rich byproduct, has large output from cottage cheese and remains unexploited. The purpose of this study was to understand the production mechanism of β-galactosidase from LAB using AW as a culture medium. First, bioinformatics analysis was conducted on 15 species of LAB. Then, 24 strains were selected and inoculated in de Man, Rogosa, and Sharpe (MRS) broth and in AW medium to compare the bacterial kinetic growth and β-galactosidase production. Bacterial growth and total protein activity were measured using spectrophotometric techniques. β-Galactosidase activity was determined by 2 methods: following the hydrolysis of o-nitrophenyl-β-d-galactopyranoside and of 5-bromo-4-chloro-3-indoyl-β-d-galactopyranoside (X-gal) in tryptic soy agar plates. The relative expression of the β-galactosidase gene was performed using real-time quantitative PCR. Despite generally lower growth in AW, 18 strains showed higher β-galactosidase activity when grown in AW compared with MRS medium. The highest β-galactosidase activity in AW was in Lactobacillus helveticus strain OSU-PECh-4A, which showed almost 5 times higher activity than average. Analysis of 6 selected strains for expression of the bgal-620 gene found higher overexpression in AW than in MRS, regardless of specific β-galactosidase activity. Strains of LAB such as OSU-PECh-4A could valorize AW through the production of β-galactosidase (as an aid to lactose digestion) and production of prebiotic galactooligosaccharides.

ABSTRACT: The dairy industry struggles to maintain consumer attention in the midst of declining fluid milk sales. Current trends create an opportunity to incorporate plant-based proteins with milk to produce a high-protein, multisourced, functional food product. Plant-based proteins, such as those in peas, can be challenging to use in food systems because of their low solubility and undesirable off-flavors. Casein micelles have unique structural properties that allow for interactions with small ions and larger macromolecules that aid in their noteworthy ability as a nanovehicle for hydrophobic compounds. The objective of this study was to use the inherent structure of the casein micelle along with common dairy processing equipment to create a stable colloidal dispersion of casein micelles with pea protein to improve its solubility in aqueous solutions. We created 3 blends with varying ratios of casein-to-pea protein (90:10, 80:20, 50:50). We subjected the mixtures to 3 cycles of homogenization using a bench-top GEA 2-stage homogenizer at 27,580 kPa maintained at 4°C, followed by pasteurization at 63°C for 30 min. The resulting blends were homogeneous liquids with increased stability due to the lack of protein precipitation. Further protein analysis by HPLC and AA sequencing revealed that vicilin, an insoluble storage protein, was the main pea protein incorporated within the casein micelle structure. These results supported our hypothesis that low-temperature homogenization can successfully be used to create a colloidal dispersion with increased stability, in which insoluble plant-based proteins may be incorporated with casein micelles in an aqueous solution. Additionally, 3-dimensional microscope images of the blends indicated a noticeable difference between the surface roughness upon addition of pea protein to the casein micelle matrix. This research highlights a promising application for other plant-based proteins to be used within the dairy industry to help drive future product innovation while also meeting current processing conditions and consumer demands.

ABSTRACT: Monitoring the ripening process by prevalent analytic methods is laborious, expensive, and time consuming. Our objective was to develop a rapid and simple method based on vibrational spectroscopic techniques to understand the biochemical changes occurring during the ripening process of Turkish white cheese and to generate predictive algorithms for the determination of the content of key cheese quality and ripening indicator compounds. Turkish white cheese samples were produced in a pilot plant scale and ripened for 100 d, and samples were analyzed at 20 d intervals during storage. The collected spectra (Fourier-transform infrared, Raman, and near-infrared) correlated with major composition characteristics (fat, protein, and moisture) and primary products of the ripening process and analyzed by pattern recognition to generate prediction (partial least squares regression) and classification (soft independent analysis of class analogy) models. The soft independent analysis of class analogy models classified cheese samples based on the unique biochemical changes taking place during the ripening process. partial least squares regression models showed good correlation (RPre = 0.87 to 0.98) between the predicted values by vibrational spectroscopy and the reference values, giving low standard errors of prediction (0.01 to 0.57). Portable and handheld vibrational spectroscopy units can be used as a rapid, simple, and in situ technique for monitoring the quality of cheese during aging and provide real-time tools for addressing deviations in manufacturing.

Antibiotic resistance is a major threat to global health. Optimizing the use of antibiotics is a key measure to prevent and control this problem. Antimicrobial Stewardship Programs (ASPs) are designed to improve clinical outcomes, minimize adverse effects and protect patients, and to ensure the administration of cost-effective treatments. Inappropriate use of antibiotics also occurs in pediatric clinical practice. For this reason, ASPs should include specific objectives and strategies aimed at pediatricians and families. Implementing these programs requires the involvement of institutions and policy makers, healthcare providers as well as individuals, adapting them to the characteristics of each healthcare setting. Pediatric primary care (PPC) faces specific issues such as high demand and immediacy, scarce specialized professional resources, difficulties to access regular training and to obtain feedback. This requires the design of specific policies and strategies to achieve the objectives, including structural and organizational measures, improvement of the information flow and accessibility to frequent trainings. These programs should reach all health professionals, promoting regular trainings, prescription support tools and supplying diagnostic tests, with adequate coordination between health care levels. Periodic evaluations and surveillance tools are useful to assess the impact of the actions taken and to provide feedback to health providers in order to adapt and improve their clinical practice to meet ASPs objectives. La implementación de estos programas requiere la implicación de instituciones, profesionales y población, adaptándolos a las características de cada ámbito asistencial. This study investigated the impact of ultra-shear technology (UST) processing on dairy-pea protein dispersions with different fat levels. Raw milk, skim milk, and cream, as well as model dispersions with combinations of dairy products and pea protein (i.e., raw milk with pea protein, skim milk with pea protein, and cream with pea protein) were employed as test samples. UST experiments were conducted at a pressure of 400 MPa and 70 °C shear valve exit temperature. The UST treatment increased the viscosity of the dispersions and the increases depended on the fat level. Dairy-pea protein dispersions from raw milk and skim milk were shear thinning and mathematically described by the power-law model defined by the consistency coefficient, K (Pa·sn) and the flow behavior index, n. UST treated cream + pea protein dispersions produced structures with gel-like characteristics. Microstructure and particle size analysis determined by laser scanning microscope revealed a reduction in particle size after UST treatment in raw milk + pea protein and skim milk + pea protein dispersions up to 7.55 and 8.30 μm, respectively. In contrast, the particle mean diameter of cream + pea protein dispersions increased up to 77.20 μm after the UST treatment. Thus, the effect of UST on the particle size and rheological behavior of the dispersions depended on the fat level. UST-treated dispersions were stable with no visible phase separation or sedimentation upon centrifugation at 4000×g for 30 min (4 °C). Heat treatment and freeze–thaw treatment of UST-treated samples showed stable blends immediately after the treatments, but subsequent centrifugation showed solid separation. Results from the study suggest that UST is a potential technology to produce stable dairy + pea protein liquids foods with different rheological characteristics for diverse applications.

El español chocoano (EC) es un vernáculo afrohispánico hablado en las tierras bajas del Pacífico colombiano (Departamento del Chocó). Aunque lingüísticamente similar al español colombiano estándar, las condiciones históricas del Departamento del Chocó parecen haber sido ideales para el desarrollo de una lengua criolla: población blanca reducida, introducción masiva de esclavos, duras condiciones de trabajo, total aislamiento del contexto urbano y de la lengua española. Al respecto, se han propuesto dos hipótesis principales: (1) descriollización y (2) afrogénesis. Analizando la evidencia lingüística y sociohistórica del EC, este trabajo indica que las condiciones no fueron propicias para la formación de un criollo en el Chocó colonial. Por lo tanto, las hipótesis 1 y 2 no reflejan la verdadera naturaleza y evolución de este dialecto. El presente estudio sugiere que el EC puede analizarse como el resultado de procesos avanzados de adquisición de segunda lengua, no implicando necesariamente una etapa criolla previa.

Background: Severe spermatogenic failure (SPGF) represents one of the most relevant causes of male infertility. This pathological condition can lead to extreme abnormalities in the seminal sperm count, such as severe oligozoospermia (SO) or non-obstructive azoospermia (NOA). Most cases of SPGF have an unknown aetiology, and it is known that this idiopathic form of male infertility represents a complex condition. In this study, we aimed to evaluate whether common genetic variation in TEX15, which encodes a key player in spermatogenesis, is involved in the susceptibility to idiopathic SPGF.Materials and Methods: We designed a genetic association study comprising a total of 727 SPGF cases (including 527 NOA and 200 SO) and 1,058 unaffected men from the Iberian Peninsula. Following a tagging strategy, three tag single-nucleotide polymorphisms (SNPs) of TEX15 (rs1362912, rs323342, and rs323346) were selected for genotyping using TaqMan probes. Case-control association tests were then performed by logistic regression models. In silico analyses were also carried out to shed light into the putative functional implications of the studied variants.Results: A significant increase in TEX15-rs1362912 minor allele frequency (MAF) was observed in the group of SO patients (MAF = 0.0842) compared to either the control cohort (MAF = 0.0468, OR = 1.90, p = 7.47E-03) or the NOA group (MAF = 0.0472, OR = 1.83, p = 1.23E-02). The genotype distribution of the SO population was also different from those of both control (p = 1.14E-02) and NOA groups (p = 4.33–02). The analysis of functional annotations of the human genome suggested that the effect of the SO-associated TEX15 variants is likely exerted by alteration of the binding affinity of crucial transcription factors for spermatogenesis.Conclusion: Our results suggest that common variation in TEX15 is involved in the genetic predisposition to SO, thus supporting the notion of idiopathic SPGF as a complex trait.

Milk phospholipids (PL) are valuable dairy components that appear to impart human health benefits, including improved cognitive function in infants and adults. The commercial food industry uses primarily plant-based sources of PL, such as soy lecithin. However, it remains unclear whether different compositions of PL from different dietary sources, such as milk, convey the same benefits. We hypothesized that PL derived from bovine milk or soy have differing physiological effects in terms of inflammation due to their differences in composition. The objectives of this study were to characterize milk and soy liposomes by their physicochemical properties and composition and to evaluate their effects in vitro by means of inflammatory gene expression analyses. Milk and soy phospholipid large unilamellar vesicles (MPL-LUV and SPL-LUV, respectively) prepared using thin-film hydration coupled with extrusion were similar in terms of structure, size, and stability; however, they differed significantly in composition. The 3T3-L1 adipocytes were selected for this work because adipocytes are the main site of uptake, synthesis, modification, and breakdown of lipids and are important inflammatory mediators in mammalian systems. In this work, these cells exposed to both liposome varieties showed high biocompatibility and low cytotoxicity up to concentrations of 0.5 mg/mL as measured by colorimetric MTT and lactate dehydrogenase assays. Furthermore, SPL-LUV showed trends toward stimulating inflammation compared with MPL-LUV as measured by expression of 2 proinflammatory cytokines, monocyte chemoattractant protein-1 (MCP-1) and IL-6. Expression of MCP-1 significantly increased 1.82-fold relative to the control upon SPL-LUV treatment, with similar trends for IL-6 (increased 1.59-fold). The MPL-LUV showed relatively no change in cytokine expression. The results obtained in this work suggest that the methodology used to prepare LUV and the composition and proportion of milk PL are important in measuring cell physiology changes and inflammatory status in mammalian cells.

This paper describes the identification of body function (BF) mentions within the clinical text within a large, national, heterogeneous corpus to highlight structural challenges presented by the clinical text. BF in clinical documents provides information on dysfunction or impairments in the function or structure of organ systems or organs. BF mentions are embedded in highly formatted structures where the formats include implied scoping boundaries that confound existing natural language processing segmentation and document decomposition techniques. This paper describes follow-up work to adapt a rule-based system created using National Institutes of Health records to a larger, more challenging corpus of Social Security Administration data. Results of these systems provide a baseline for future work to improve document decomposition techniques.

One of the largest health problems worldwide is the development of chronic noncommunicable diseases due to the consumption of hypercaloric diets. Among the most common alterations are cardiovascular diseases, and a high correlation between overnutrition and neurodegenerative diseases has also been found. The urgency in the study of specific damage to tissues such as the brain and intestine led us to use Drosophila melanogaster to study the metabolic effects caused by the consumption of fructose and palmitic acid in specific tissues. Thus, third instar larvae (96 ± 4 h) of the wild Canton-S strain of D. melanogaster were used to perform transcriptomic profiling in brain and midgut tissues to test for the potential metabolic effects of a diet supplemented with fructose and palmitic acid. Our data infer that this diet can alter the biosynthesis of proteins at the mRNA level that participate in the synthesis of amino acids, as well as fundamental enzymes for the dopaminergic and GABAergic systems in the midgut and brain. These also demonstrated alterations in the tissues of flies that may help explain the development of various reported human diseases associated with the consumption of fructose and palmitic acid in humans. These studies will not only help to better understand the mechanisms by which the consumption of these alimentary products is related to the development of neuronal diseases but may also contribute to the prevention of these conditions.

In Mexico, pasture degradation is associated with extensive pastures; additionally, under these conditions, livestock activities contribute considerably to greenhouse gas (GHG) emissions. Among the options to improve grazing systems and reduce GHG emissions, silvopastoral systems (SPS) have been recommended. The objectives of this work were to quantify the N outflow in a soil–plant–animal interface, as well as the CH4 emissions and milk production in an SPS with woody legumes (Leucaena leucocephala) that is associated with stargrass (Cynodon nlemfuensis). This was then compared with stargrass in a monoculture system (MS) in the seasons (dry and rainy period) over a two-year period. Dung was collected from the animals of each of the grazing systems and applied fresh to the land plots. Fresh dung and urine were collected from the cows of each grazing system and were applied to the experimental plots. In addition, the soil CH4 and N2O contents were measured to quantify the emissions. Average milk yield by seasons was similar: MS (7.1 kg per animal unit (AU)/day−1) and SPS (6.31 kg per AU/day−1). Cows in the MS had a mean N intake of 171.9 g/UA day−1 without seasonal variation, while the SPS animals’ mean N intake was 215.7 g/UA day−1 for both seasons. For the urine applied to soil, the N2O outflow was higher in the MS (peak value = 1623.9 μg N-N2O m−2 h−1). The peak value for the SPS was 755.9 μg of N-N2O m−2 h−1. The N2O emissions were higher in the rainy season (which promotes denitrification). The values for the feces treatment were 0.05% (MS) and 0.01% (SPS). The urine treatment values were 0.52% (MS) and 0.17% (SPS). The emissions of CH4 showed that the feces of the SPS systems resulted in a higher accumulation of gas in the rainy season (29.8 g C ha−1), followed by the feces of the MS system in the dry season (26.0 g C ha−1). Legumes in the SPS helped to maintain milk production, and the N2O emissions were lower than those produced by the MS (where the pastures were fertilized with N).

In this study, the effects of orange essential oil (OEO) on the rumen fermentation, nutrient utilization, and methane (CH4) emissions of beef heifers fed a diet of bermudagrass (Cynodon dactylon) were examined. In addition, in vitro and in situ experiments were conducted. The in vitro experiment consisted of three treatments: control (CTL, no OEO), OEO1 (0.25% OEO), and OEO2 (0.5% OEO). The forage to concentrate ratio was 70:30 (dry matter [DM] basis) in all treatments. No changes in pH, proportions of volatile fatty acids, and the acetate:propionate ratio were observed (P > 0.05). The addition of 0.25% OEO resulted in a reduction in CH4 production (mL/g) relative to the control (P < 0.05). In the in situ experiment, 5 g of total mixed ration (CTL, OEO1, and OEO2) were incubated for 6, 12, 24, 48, and 72 h. Potential and effective degradability were not affected by OEO supplementation (P > 0.05). In the in vivo study, six crossbred beef heifers (Bos indicus × Bos taurus), fitted with rumen cannulas, were assigned to three different treatments: no additive (CTL), 0.25% OEO (OEO1), and 0.5% OEO (OEO2) in a replicated 3 × 3 Latin square (21-day periods). Heifers were fed at 2.8% body weight. In vivo CH4 production was measured in open-circuit respiration chambers. Reductions in gross energy consumption, apparent total tract digestibility, and rumen valerate concentration were observed for OEO2 compared to the control (P < 0.05). Additionally, decreases in CH4 emissions (g/day; P < 0.05) and CH4 (MJ gross energy intake/day; P < 0.05) were observed in response to supplementation of 0.5% OEO as compared to the CTL treatment. Thus, supplementation of 0.5% OEO reduced CH4 emissions (g/day) by 12% without impacting the DM intake of heifers fed bermudagrass hay as a basal ration.

β-Lactoglobulin (β-LG) is believed to be a common allergen in bovine milk. Buttermilk (BM) powder has abundant contents of milk fat globule membrane and phospholipid, both of which have been demonstrated to have positive effects on brain and cognitive development during early infancy. This study focused on modifying β-LG in BM via supercritical CO2 (ScCO2) treatment to modify its reactivity to antibodies and thus reduce its antigenicity. Buttermilk powder was treated in a supercritical fluid extraction system with food-grade CO2 at 100, 150, 200, 250, 350, and 400 bar at 2 temperatures, 50 and 75°C. All analyses were completed in a 10% BM suspension (wt/vol). The BM proteins were examined using sodium dodecyl sulfate (SDS)-PAGE, Western blot, ELISA, and periodic acid staining methods. Semi-purified β-LG was used to evaluate the cytotoxicity, viability, and inflammatory response in the Caco-2 cell line by means of the lactate dehydrogenase assay, MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium] assay, and IL-8 production, respectively. The SDS-PAGE showed that the signal intensity of β-LG bands was reduced by up to 50% after being processed at 250 bar and 75°C for 30 min. Lighter and more diffuse signals were found by Western blot, indicating modification of the protein structure. The ELISA demonstrated that ScCO2 treatment could significantly change β-LG antigenicity in BM. Sugar moieties in bands corresponding to β-LG were revealed by periodic acid staining, indicating glycosylation only in samples treated with ScCO2. Caco-2 cells treated with whey proteins had high viability, 24.9% lower inflammation, and no evidence of cytotoxicity compared with untreated cultures. These results showed that reduced antigenicity of β-LG was caused by lactosylation, which has been reported as a possible pathway to reduce the allergenicity in foods. The denaturation of β-LG by supercritical fluid processing is a promising way to address milk allergy, which remains a problem requiring more attention and further research.

The benefits of fermented dairy products, in particular the presence of lactic acid bacteria (LAB) and milk phospholipids (MPL), seem to correlate with positive effects on human health. We hypothesize that one aspect of this benefit is the adhesion of LAB to the milk fat globule membrane via the interaction of LAB and MPL. Our first objective was to present a method to characterize and quantify such adhesion and investigate its association with a physical test. Our second objective was to further analyze the mechanism of interaction by analyzing expression of 3 previously reported surface binding-promoting genes (MapA, Cnb, and CmbA). We categorized adhesion between MPL and LAB by observing the distribution of MPL in corresponding bacterial cultures. Our working hypothesis was that any interaction or adhesion between these 2 components would yield differences in the distribution of MPL. Out of 122 LAB tested, 27% showed what could be characterized as adhesion; 38% of these strains were Limosilactobacillus reuteri. Further characterization of adhesion was carried out using an reverse transcription quantitative-PCR experiment, which demonstrated that the relative expression level of CmbA was positively associated with that adhesion. In addition, supplementation of MPL caused overexpression of MapA and Cnb in L. reuteri OSU-PECh-37A and OSU-PECh-48. This study indicated strain-specific adhesion between MPL and LAB and suggested that CmbA, which encodes a surface protein, is a potential factor involved in that adhesion. A better understanding of interactions between MPL and LAB may contribute to the design of new functional products and improve the delivery of these bioactive ingredients to their target site of action.

Increasing interest in foods beyond their nutritional value has promoted the development of various novel functional foods that could convey multiple health benefits to consumers. The application of lactic acid bacteria (LAB) and milk phospholipids (MPL) in combination has shown some amplifying effects on the health benefits provided by both ingredients. Freezing is commonly used in LAB preservation and storage. However, the freezing/thawing process damages cell membranes and leads to a loss in viability and functionality of LAB. This study aimed to investigate the influence of MPL on growth and cryotolerance of LAB using acid whey-based medium (AWM) supplemented with 0.5% MPL. Fourteen LAB strains were initially screened from 124 LAB using acid whey-based medium (AW). We then evaluated the cell viability and acidification ability, using the plate counting method and skim milk fermentation test, respectively, of the 14 strains cultured in AW and AWM before and after 3 cycles of freezing/thawing. The presence of 0.5% MPL in AWM significantly promoted the growth of LAB. Supplementing the culture and storage medium with 0.5% MPL significantly enhanced the resistance of selected LAB to freeze-thaw cycles in terms of cell viability and acidification ability. These results suggest that supplementing with 0.5% MPL might promote the growth of LAB and enhance the cryotolerance of LAB cultures in fermented dairy products. This finding leads to a better understanding of the synergistic effects contributed by the LAB–MPL combination and promotes the development of new LAB–MPL functional products.

His bundle pacing was developed while seeking a physiological alternative to biventricular cardiac resynchronization therapy. However, His bundle pacing may not be adequate in all patients. In this scenario, left bundle branch pacing has arisen as a new cardiac resynchronization therapy modality to correct left bundle branch block and restore ventricular synchrony. Este estudio evalúa si el sexo, la dependencia a la nicotina y variables emocionales (ansiedad, depresión e ira) permiten describir un perfil de paciente que pueda beneficiarse de una terapia cognitivo-conductual para abandonar la adicción al tabaco. Se analizaron los resultados de una muestra de 120 fumadores adultos que voluntariamente recibieron TCC. Se valoró la adherencia de los pacientes y el éxito del programa a través de árboles de decisión. Los resultados indicaron que la adherencia al tratamiento ya implica alta probabilidad de éxito (86.4%), aumentando hasta el 95.6% cuando los participantes presentaban altos niveles de ira externa. Además, la adherencia al tratamiento fue completa (100%) cuando la ansiedad en contexto de evaluación, la ansiedad fisiológica y la motivación fueron altas. La obtención de este tipo de perfiles diferenciales permite determinar qué personas son más indicadas para cada tratamiento, lo que incrementaría la eficacia de los mismos.
