This thesis investigates relationships between clinical, neuroimaging and neuropathological features in autosomal dominant familial Alzheimer’s disease (FAD), with the aim of studying phenotypic heterogeneity and preclinical change. Chapters 1 and 2 introduce the background to the problem to be addressed in this thesis with an emphasis on current understanding of clinical and imaging changes in AD, and specifically in FAD. The FAD phenotype can be highly variable and, although it shares many clinical features with sporadic AD, it also possesses important differences. The clinical spectrum of FAD is first investigated, through analysis of all symptomatic cases studied at our research centre over the past twenty-five years (Chapter 3). Associations between phenotypic and pathological heterogeneity are then explored, with a study investigating genetic determinants of white matter hyperintensities and cerebral amyloid angiopathy (CAA) in FAD (Chapter 4). CAA is a common but variable feature of AD that appears to be an important factor in amyloid-modifying therapy and the term ‘ARIA’ has been coined to describe amyloid-related imaging abnormalities, thought to relate to vascular amyloid, that have been observed in a variety of amyloid-modifying therapy trials. Spontaneous changes of ARIA in FAD and the genetic risk factors that may provoke them are then described (Chapter 5). The recent launch of preclinical treatment trials for FAD necessitates better understanding of the trajectory of biomarker changes early in the disease. Observations from amyloid imaging studies, of presymptomatic amyloid deposition in the thalamus and striatum, motivated the final study, which examines changes in volume and diffusivity of these subcortical structures and their connecting white matter tracts in symptomatic and presymptomatic FAD mutation carriers (Chapter 6). Together, these studies demonstrate that exploring phenotypic heterogeneity and preclinical imaging changes can illuminate aspects of the underlying disease process, informing our understanding of FAD and potential effects of treatment.

25 páginas, 6 figuras, 2 tablas, Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele., This work was funded by a grant (EADB) from the EU Joint Programme – Neurodegenerative Disease Research. A database containing information on over 35,000 coins from sites in southern Britain was established. This was used to investigate chronological and geographical distributions of fourth century Roman coinage in Britain, and the role of coins in archaeological dating. The regularity of finds supports a view of official supply policy as the principal determinant of the coins used, deposited and subsequently recovered. Throughout the fourth century, Britain received supplies of bronze from up to three mints of which one was always the primary source. One or two secondary sources supplemented this, particularly at times of major new issues. The few finds from other mints represent material that arrived through circulation and exchange. The only clear geographical variation Was after 388AD when new issues failed to circulate extensively beyond the towns. Variations between sites are related to differences in coin using and depositing practices. Three groups of sites were recognised: towns and larger settlements, villas and rural buildings, and temples. Differences between these are most apparent after 350AD when circulation and use underwent significant changes in the rural areas. Typical excavations of rural buildings produce few coins, probably representing accidental losses. On some sites casual loss accounts for only a small proportion of the recovered material. Here, votive deposition, rubbish disposal and non-recovery of hoards are the major sources of finds. A study of the stratified material reinforces the need for caution in using coins for dating, and has important implications for the use of coins in archaeological dating. Residuality and lengthy circulation severely limit inferences about the dates of deposition of the contexts in which coins are found. Throughout the fourth century, and probably also earlier, most coins were deposited within a few decades of their issue. Unfortunately for the archaeologist in search of a date for a deposit, examples of primary deposition are greatly outnumbered by residual and re-deposited material.

Chunking is a process to split a file into smaller files called chunks. In some applications, such as remote data compression, data synchronization, and data deduplication, chunking is important because it determines the duplicate detection performance of the system. Content-defined chunking (CDC) is a method to split files into variable length chunks, where the cut points are defined by some internal features of the files. Unlike fixed-length chunks, variable-length chunks are more resistant to byte shifting. Thus, it increases the probability of finding duplicate chunks within a file and between files. However, CDC algorithms require additional computation to find the cut points which might be computationally expensive for some applications. In our previous work (Widodo etal., 2016), the hash-based CDC algorithm used in the system took more process time than other processes in the deduplication system. This paper proposes a high throughput hash-less chunking method called Rapid Asymmetric Maximum (RAM). Instead of using hashes, RAM uses bytes value to declare the cut points. The algorithm utilizes a fix-sized window and a variable-sized window to find a maximum-valued byte which is the cut point. The maximum-valued byte is included in the chunk and located at the boundary of the chunk. This configuration allows RAM to do fewer comparisons while retaining the CDC property. We compared RAM with existing hash-based and hash-less deduplication systems. The experimental results show that our proposed algorithm has higher throughput and bytes saved per second compared to other chunking algorithms. Cloud storages can be used to solve limited storage on mobile devices. The stored data can be reached from anywhere if network access is available. Network access can be costly, especially for mobile devices, which have limited power source. One of the solutions to reduce the amount of data transfer is data deduplication. This paper proposes a deduplication system called Smart Deduplication for Mobiles (SDM) for mobile devices. SDM chooses the best deduplication method without specific configurations for any file type. We compared our system to existing deduplication systems available on mobile devices. Results show that SDM achieves higher deduplication accuracy and faster deduplication speed over existing systems for mobile devices with negligible additional time in most scenarios. The additional time required by our system is negligible when compared to the power saved from uploading fewer data to a cloud storage.

Risk for late-onset Alzheimers disease (LOAD), the most prevalent dementia, is partially driven by genetics. To identify LOAD risk loci, we performed a large genome-wide association meta-analysis of clinically diagnosed LOAD (94,437 individuals). We confirm 20 previous LOAD risk loci and identify five new genome-wide loci (IQCK, ACE, ADAM10, ADAMTS1, and WWOX), two of which (ADAM10, ACE) were identified in a recent genome-wide association (GWAS)-by-familial-proxy of Alzheimers or dementia. Fine-mapping of the human leukocyte antigen (HLA) region confirms the neurological and immune-mediated disease haplotype HLA-DR15 as a risk factor for LOAD. Pathway analysis implicates immunity, lipid metabolism, tau binding proteins, and amyloid precursor protein (APP) metabolism, showing that genetic variants affecting APP and Aβ processing are associated not only with early-onset autosomal dominant Alzheimers disease but also with LOAD. Analyses of risk genes and pathways show enrichment for rare variants (P = 1.32 2009»10−7), indicating that additional rare variants remain to be identified. We also identify important genetic correlations between LOAD and traits such as family history of dementia and education.

This article is a review of the book "From Seminary to University: An Institutional History of the Study of Religion in Canada" by Aaron W. Hughes. The book explores the transformation of Canada's English-speaking seminaries into secular universities. The author argues that this evolution was primarily influenced by non-academic factors such as the secularization of public education, the union of the United Church of Canada, the adoption of multiculturalism as a federal policy, and the introduction of the Charter of Rights and Freedoms. Cloud storages are used to expand the storage on mobile devices. It can be accessed through an internet connection. On mobile devices, network data can be expensive in terms of tariff and power consumption. Deduplication can be used to reduce the amount of data to be transferred. This paper proposes a fast deduplication system for mobile devices named rapid deduplication for mobile cloud storage (RDM). RDM is based on smart deduplication for mobile cloud storage (SDM) and rapid asymmetric extremum (RAM). RDM used RAM as the chunking algorithm instead of Rabin which is used in SDM. Our experimental results show RDM is at least 39.5% to 44.7% faster at the cost of 4.2% to 9.1% higher size after deduplication.

NOBODY LIKES A MORALIST. It is right that Christians have moved beyond moralism if by this we mean the habit of reducing faith to sheer obedience. Moralism is always a diluting of religion, a simplification. It is the faith of Pelagius and Kant, not of Augustine and Bossuet. To fall into the habit of moralizing is to separate what belongs together, in this case, love for the good from love for the beautiful. It is an ancient truism that the virtuous person not only does the good, but delights in doing it ("Catechism of the Catholic Church," 1770) (hereafter CCC). Christians rightly point out that love is at the center of ethics and to love someone is to do them good. How do we know what is good? When philosophers and preachers in the mid-twentieth century proclaimed that we had moved beyond moralism, it was not always clear where they hoped we would land. Remember that when the rich young man asked Jesus how he might obtain eternal life, Christ pointed him to the Ten Commandments. The Gospels certainly deepen our understanding of law, but they do not abrogate it. There is no Christian morality apart from law (CCC, 2055). As in the recent revelations of clerical abuse of minors, the consequences of liberating ourselves from law can be terrifying. In 2010, in the face of gross misconduct, the Irish government initiated two separate judicial reviews to determine the extent and causes of corruption among the Catholic hierarchy. The Murphy Commission, in particular, investigated the bishops' handling of allegations of sexual misconduct among priests in the Diocese of Dublin from 1975-2004, which has led to the resignation of several auxiliary bishops of Dublin. Any breach of trust by clergy is occasion for scandal, sorrow, and restitution. But what has caused outrage in the case of Ireland and several other jurisdictions is the way that bishops failed to discipline clergy even when allegations were substantiated. Often offending priests were simply shuffled from one parish to another. In the case of the Diocese of Dublin, Justice Murphy has pointed to how disregard for canon law among bishops and clergy sheltered repeat offenders and exposed the vulnerable. The report notes how in Dublin "the Church authorities failed to implement most of their own canon law rules on dealing with clerical child-sexual abuse." Not that Church law was inadequate. "The Commission is satisfied that Church law demanded severe penalties for clerics who abused children. In Dublin, from the 1970s onwards, this was ignored." It is not the case, in other words, of the law being under or over specified; laws were simply not heeded. Observing what has now become shamefully plain, the report links the neglect of justice to the disrespect of law within the Church more generally: "Canon law, as an instrument of Church governance, declined hugely during Vatican II and in the decades after it." (1) In countering what may have been an excessive legalism within pre-Vatican II Roman Catholic piety, pastors and theologians from the 1960s on swung wildly in the opposite direction. Some among our leaders did not serve well when they replaced moralism with vague appeals to love and therapeutic approaches to inner healing that disregard the fact of sin and the requirements of justice. Fruit grows only amidst the shade of leaves; so also love requires the protective wrapping of justice if it is to blossom. St. Augustine famously said, "Love ... and then do what you will" (Commentary on the Epistle of John 10.7.1). If God is love, why not just leave it at that and let each to his own way? After all, the primacy of charity is what distinguishes Christ's teaching from that of the Pharisees. Augustine was right to say what he did. But if we wish to mouth his words, then we must also think his thoughts. We must relearn also what he knew about the moral life and the relation between law and love. In short, the problem with appeal to love as an isolated command is that it is easily drained of content and filled with unwholesome substitutes. …

Augustine was aware of objections to the idea of educational progress, and nowhere is this more acute than in his treatment of academic skepticism. While much attention has been paid to Augustine's theory of knowledge within the Contra Academicos, too often overlooked is the specifically moral significance that he attaches to the skeptic's critique of knowledge. I argue that Augustine's chief criticism in this dialogue is not directed against an erroneous epistemology, although he does provide a refutation of that. Rather, it is against the disastrous educational and moral implications that follow from the skeptic's views. Along the way I show how this interpretation helps to explain some of the novel structural features of the dialogue and allows us more securely to situate its argument against Augustine's other key early reflections on the limits of knowledge.

Breast cancer is the second most common form of malignancy diagnosed in Australian women, imposing an enormous social and economic burden on society. If the cancer spreads to secondary locations, patient survival decreases dramatically. Therapeutic strategies for treatment of metastatic disease are desperately needed in breast cancer. Recently we have shown that when Myb is conditionally deleted from the mammary gland of MMTV-Neu and MMTV-PyMT mice, tumour formation is ablated. To provide further insight into the function of Myb in mammary gland development, cre-mediated conditional knock out of c-myb in the mouse mammary gland was examined. The conditional deletion of c-myb led to a reduction in branching and terminal end bud formation. These data indicate that if MYB could be inhibited for breast cancer therapy, there are potentially few side effects on the normal mammary gland. The ability to inhibit DNA binding transcription factors is a long sort after goal in oncology, as their importance in disease initiation and progression is well documented. To target MYB, we have developed a DNA vaccine. Preclinical studies have largely examined the MYB DNA vaccine in the context of colon cancer models using prophylactic vaccination. However, preliminary data indicate that it may also be effective in reducing the metastatic burden in preclinical breast cancer models. This thesis aims to further investigate the role of MYB during mammary gland development and provide insights into its involvement in tumourigenesis. Furthermore, the MYB DNA vaccine will be assessed for its effectiveness as a therapeutic treatment in clinically relevant surgical models of metastatic breast cancer, as well as further development as a therapeutic in the setting of colon cancer.
