Abstract IODP Site U1437 is located in the Izu rear‐arc region, approximately 330 km west of the Izu‐Bonin trench axis. The oldest four units (Units IV through Unit VII) include volcaniclastic sediment and in situ hyaloclastites. They have ages of about 6–15 Ma, shortly after cessation of Shikoku backarc basin opening. Three magma types are identified by their distinct geochemistry; they are similar types to those found in the modern Izu arc (Rear Arc Seamount Chain [RASC]‐type, Rift‐type, and volcanic front [VF]‐type). RASC‐type has the most enriched Nd and Hf isotope and fluid‐immobile trace element ratios and dominates from 9 to 6 Ma. Rift‐type, dominant from 15 to 9 Ma, is similar to VF‐type in Nd‐Hf isotopes but has the least radiogenic Sr and Pb, and intermediate La/Yb and Nb/Yb, indicating a more fertile mantle source and less hydrous slab component than VF‐type. Less common and randomly distributed VF‐type sediments have the most radiogenic Sr and Pb, and the highest Ba/(Th, LREE [light rare earth element]) ratios, and are interpreted to be distally derived. The genesis of mafic Unit VII samples (~15 Ma) was modeled using the Arc Basalt Simulator. Results are most similar to those for basalts in the modern rift environment indicating the addition of ~1% of a melt‐rich slab component generated at ~125 km, to a Philippine Sea Plate ambient mantle that was more depleted than DMM (depleted MORB mantle). The initial post‐Shikoku basin magmatism in the Izu rear‐arc generated Rift‐type magmas for about 6 million years before the distinctive RASC‐type magmatism began, which then became increasingly enriched.

Both enantiomers of 2-ethylquinazolin-4-ones bearing ortho -CH 3 O/CD 3 O and CH 3 O/ 13 CH 3 O phenyl groups at the N3 position were prepared. These are isotopic atropisomeric compounds based on a remote and conformationally flexible H/D and 12 C/ 13 C discrimination, and it was found that a CHCl 3 solution of ortho -CH 3 O/CD 3 O derivative shows a slight specific optical rotation. Furthermore, diastereomeric quinazolinone derivatives bearing a chiral carbon were prepared, and their stereochemical purities and rotational stability as well as the isotopic atropisomerism were verified by 1 H NMR and chiral high-performance liquid chromatography (HPLC) analyses.

An outbreak of influenza A(H1N1)pdm09 viruses exhibiting cross-resistance to oseltamivir and peramivir occurred in Yokohama, Japan, in September 2024. Among 24 students in a class, 11 were diagnosed with influenza or influenza-like illness, and viruses harbouring the NA H275Y and HA Q210H substitutions were isolated from four. Deep sequencing analysis confirmed the clonal spread of these mutants. Antigenic analysis revealed differences from the vaccine strain. Continued monitoring is crucial to assess the potential for further spread of these mutant viruses.

Background: Influenza viruses can cause zoonotic infections that pose public health risks. Surveillance of influenza A and B viruses is conducted globally; however, information on influenza C and D viruses is limited. Longitudinal monitoring of influenza C virus in humans has been conducted in several countries, but there has been no long-term monitoring of influenza D virus in humans. The public health risks associated with the influenza D virus therefore remain unknown., Methods: We established a duplex real-time RT-PCR to detect influenza C and D viruses and analyzed respiratory specimens collected from 2144 patients in Japan with respiratory diseases between January 2018 and March 2023. We isolated viruses and conducted hemagglutination inhibition tests to examine antigenicity and focus reduction assays to determine susceptibility to the cap-dependent endonuclease inhibitor baloxavir marboxil., Results: We detected three influenza C viruses belonging to the C/Kanagawa- or C/Sao Paulo-lineages, which recently circulated globally. None of the specimens was positive for the influenza D virus. The C/Yokohama/1/2022 strain, isolated from the specimen with the highest viral RNA load and belonging to the C/Kanagawa-lineage, showed similar antigenicity to the reference C/Kanagawa-lineage strain and was susceptible to baloxavir., Conclusions: Our duplex real-time RT-PCR is useful for the simultaneous detection of influenza C and D viruses from the same specimen. Adding the influenza D virus to the monitoring of the influenza C virus would help in assessing the public health risks posed by this virus., (© 2024 The Authors. Quinazolin-4-one derivatives possessing an isotopic atropisomerism (isotopic N-C axial chirality) based on ortho - 12 CH 3 / 13 CH 3 discrimination were prepared. The diastereomeric quinazolin-4-ones bearing an asymmetric carbon as well as an isotopic atropisomerism were clearly discriminated by 1 H and 13 C NMR spectra and revealed to possess high rotational stability and stereochemical purity.

Silicate spherules have been identified from the ca. 3.4 Ga-old Strelley Pool Formation (SPF) in the Pilbara Craton, Western Australia. Their origins and geochemical characteristics, including the Re and platinum-group elements of their host clastic layer and the overlying and underlying microfossil-bearing finely laminated carbonaceous cherts, were examined. The spherules have various morphologies (completely spherical to angular), sizes (∼20 to >500 μm), textures (layered, non-layered, and fibrous), mineralogy (various proportions of microcrystalline quartz, sericite, anatase and Fe-oxides), and chemistry (enriched in Ni and/or Cr), commonly with thin anatase-rich walls. Their host clastic layer is characterized by rip-up clasts, suggesting a suddenly occurring high-energy depositional environment, such as tsunamis. Although various origins other than asteroid impact were considered, none could unequivocally explain the features of the spherules. In contrast, non-layered spherical spherules that occur as individual framework grains or collectively comprise angular-shaped rock fragments appear to be more consistent with the asteroid impact origin. The calculated Re-Os age of the cherts (3331 ± 220 Ma) was consistent with the established age of the SPF (3426-3350 Ma), suggesting that the Re-Os system was not significantly disturbed by later metamorphic and weathering events.

N-C axially chiral 3-(2-trideuteriomethyl-4,6-dimethylphenyl)-2-ethylquinazolin-4-ones and 3-(2-trideuteriomethyl-4,6-dimethylphenyl)-2-(1-phenylpropan-2-yl)quinazolin-4-ones were prepared in high enantio- and diastereomeric purities (98% ee). These quinazolinone derivatives are isotopic atropisomers based on ortho -CH 3 /CD 3 discrimination and were revealed to possess a slight optical rotation and high rotational stability.

Amino acids contribute to the taste of foods. Previous studies on the taste of amino acids focused mainly on α-amino acids, and therefore, the taste characteristics of amino acid derivatives remain unclear. In the present study, we targeted 6 different amino acid derivatives, β-alanine, citrulline, creatine, γ-aminobutyric acid, taurine, and ornithine, and evaluated their taste characteristics in a human sensory study. All tested amino acid derivatives showed multiple taste qualities; no derivatives had only a single taste quality. However, their taste intensities were relatively weak even at high concentrations. Given that the interactions between amino acid derivatives and nucleotide result in taste enhancements, we investigated the effect of inosine 5'-monophosphate (IMP) on the taste characteristics and found that the taste intensity of ornithine increased in the presence of IMP. This finding will be useful for understanding the role of amino acid derivatives as taste substances in daily foods.

Calorie restriction (CR) reportedly prevents atherosclerotic diseases. Furthermore, CR induces forkhead box protein-O1 (FOXO-1) expression in the skeletal muscle, altering the character of the skeletal muscle. We previously reported that the change in skeletal muscle character, induced by the overexpression of peroxisome proliferator-activated receptor γ coactivator-1α, suppresses atherosclerotic progression in an atherosclerotic apolipoprotein E-knockout (ApoE-KO) mouse model. Thus, we hypothesized that skeletal muscle alternation induced by FOXO-1 may also have an anti-atherosclerotic effect in ApoE-KO mice. In this study, we investigated whether skeletal muscle-specific FOXO-1 overexpression suppresses the progression of atherosclerosis in ApoE-KO mice. We generated ApoE-KO/FOXO-1 mice, in which an ApoE-KO mouse was crossbred with a mouse presenting skeletal muscle-specific FOXO-1 overexpression (FOXO-1Tg). The mice were sacrificed at 20 weeks of age, and atherosclerotic plaque area and protein expression in the plaque were measured. Additionally, we measured the tumor necrosis factor α (TNFα)- induced mRNA expression in human umbilical vein endothelial cells (HUVECs), using serum collected from the FOXO-1Tg mice. Accordingly, ApoE-KO/FOXO-1 mice showed a 65% reduced atherosclerotic plaque area when compared with the ApoE-KO mice, with concomitantly reduced vascular cell adhesion molecule-1 (VCAM-1) and macrophage infiltration. As compared to serum from wild-type mice, the serum collected from the FOXO-1Tg mice significantly suppressed the mRNA expression of VCAM-1, an atherosclerosis initiation factor, in TNFα-treated HUVECs. Therefore, these data suggest that skeletal muscle-specific FOXO-1 overexpression suppresses the progression of atherosclerosis in ApoE-KO mice. The extreme Sr, Nd, Hf, and Pb isotopic compositions found in Pitcairn Island basalts have been labeled enriched mantle 1 (EM1), characterizing them as one of the isotopic mantle end members. The EM1 origin has been vigorously debated for over 25 years, with interpretations ranging from delaminated subcontinental lithosphere, to recycled lower continental crust, to recycled oceanic crust carrying ancient pelagic sediments, all of which may potentially generate the requisite radiogenic isotopic composition. Here we find that δ 26 Mg ratios in Pitcairn EM1 basalts are significantly lower than in normal mantle and are the lowest values so far recorded in oceanic basalts. A global survey of Mg isotopic compositions of potentially recycled components shows that marine carbonates constitute the most common and typical reservoir invariably characterized by extremely low δ 26 Mg values. We therefore infer that the subnormal δ 26 Mg of the Pitcairn EM1 component originates from subducted marine carbonates. This, combined with previously published evidence showing exceptionally unradiogenic Pb as well as sulfur isotopes affected by mass-independent fractionation, suggests that the Pitcairn EM1 component is most likely derived from late Archean subducted carbonate-bearing sediments. However, the low Ca/Al ratios of Pitcairn lavas are inconsistent with experimental evidence showing high Ca/Al ratios in melts derived from carbonate-bearing mantle sources. We suggest that carbonate-silicate reactions in the late Archean subducted sediments exhausted the carbonates, but the isotopically light magnesium of the carbonate was incorporated in the silicates, which then entered the lower mantle and ultimately became the Pitcairn plume source., Competing Interests: The authors declare no conflict of interest.

The ca 3.4 Ga Strelley Pool Formation (SPF) of the Pilbara Craton, Australia, represents a Paleoarchean sedimentary succession preserving well-described and morphologically diverse biosignatures such as stromatolites and cellularly preserved microfossils. The SPF microfossil assemblage identified from three greenstone belts includes relatively large (20-80 μm in width), acid-resistant, organic-walled lenticular microfossils, which can be extracted using a palynological technique. In this study, we present results of measurements of over 800 palynomorphic specimens of SPF lenticular microfossils from 2 remote (∼80 km apart) localities that represent different depositional environments and thus different habitats, as evidenced by their distinct lithostratigraphic association and trace element geochemistry. We demonstrate statistically that the two populations are distinct in oblateness from a polar view and furthermore that each population comprises subpopulations defined by different areas and oblateness. This study may provide the earliest morphological evidence for speciation of unicellular organisms, which could have been allopatric (geographic) and adaptive. It can also be suggested that SPF lenticular microbes had highly organized cytoskeleton indispensable for strict control of the cell morphology of large and robust microbes, which in turn were likely advantageous to their prosperity and diversification.

The 3.4-Ga Strelley Pool Formation (SPF) at the informally named 'Waterfall Locality' in the Goldsworthy greenstone belt of the Pilbara Craton, Western Australia, provides deeper insights into ancient, shallow subaqueous to possibly subaerial ecosystems. Outcrops at this locality contain a thin (<3 m) unit of carbonaceous and non-carbonaceous cherts and silicified sandstones that were deposited in a shallow-water coastal environment, with hydrothermal activities, consistent with the previous studies. Carbonaceous, sulfide-rich massive black cherts with coniform structures up to 3 cm high are characterized by diverse rare earth elements (REE) signatures including enrichment of light [light rare earth elements (LREE)] or middle rare earth elements and by enrichment of heavy metals represented by Zn. The massive black cherts were likely deposited by mixing of hydrothermal and non-hydrothermal fluids. Coniform structures in the cherts are characterized by diffuse laminae composed of sulfide particles, suggesting that unlike stromatolites, they were formed dominantly through physico-chemical processes related to hydrothermal activity. The cherts yield microfossils identical to previously described carbonaceous films, small and large spheres, and lenticular microfossils. In addition, new morphological types such as clusters composed of large carbonaceous spheroids (20-40 μm across each) with fluffy or foam-like envelope are identified. Finely laminated carbonaceous cherts are devoid of heavy metals and characterized by the enrichment of LREE. This chert locally contains conical to domal structures characterized by truncation of laminae and trapping of detrital grains and is interpreted as siliceous stromatolite formed by very early or contemporaneous silicification of biomats with the contribution of silica-rich hydrothermal fluids. Biological affinities of described microfossils and microbes constructing siliceous stromatolites are under investigation. However, this study emphasizes how diverse the microbial community in Paleoarchean coastal hydrothermal environment was. Early Palaeoproterozoic (2.5-2.0 billion years ago) was a critical phase in Earth's history, characterized by multiple severe glaciations and a rise in atmospheric O(2) (the Great Oxidation Event). Although glaciations occurred at the time of O(2) increase, the relationship between climatic and atmospheric transitions remains poorly understood. Here we report high concentrations of the redox-sensitive element Os with high initial (187)Os/(188)Os values in a sandstone-siltstone interval that spans the transition from glacial diamictite to overlying carbonate in the Huronian Supergroup, Canada. Together with the results of Re, Mo and S analyses of the sediments, we suggest that immediately after the second Palaeoproterozoic glaciation, atmospheric O(2) levels became sufficiently high to deliver radiogenic continental Os to shallow-marine environments, indicating the synchronicity of an episode of increasing O(2) and deglaciation. This result supports the hypothesis that climatic recovery from the glaciations acted to accelerate the Great Oxidation Event.

A series of i-->i + 4 side-chain to side-chain lactam analogs of vasoactive intestinal peptide has been prepared in order to study the effect of cyclization on biological activity. In vitro, on guinea pig tracheal smooth muscle and on human bronchial tissue, approximately half of the cyclic analogs showed increased potency and half were decreased over the linear analogs. Several cyclic compounds were between 10- and 20-fold more potent and one was 290-fold more potent than the linear species. In vivo, in guinea pigs, the cyclic compounds showed increased potency by up to 70-fold and significantly enhanced duration of action as compared to linear compounds.

The effects of bilateral locus ceruleus (LC) lesions on the pericapillary nerve terminals were investigated in the feline brain parenchyma using electron microscopy. LC lesions were induced stereotaxically and the animals were sacrificed after intravenous administration of 5-hydroxydopamine (5-OHDA). The diameter and number of dense-cored vesicles (DCVs) and clear vesicles (CVs) in the pericapillary nerve terminals were measured. The number of DCVs in the nerve terminal was significantly decreased by bilateral LC lesions. The diameters of the DCVs and CVs decreased significantly as compared with those in the non-operated control group. These data suggest that the LC is closely related to the pericapillary nerve terminals in the brain parenchyma and that not only nerve terminals with DCVs but also those with CVs are affected by LC lesions.
