Increased temperatures associated with urbanization (the "urban heat island" effect) have been shown to impact a wide range of traits across diverse taxa. At the same time, climatic conditions vary at fine spatial scales within habitats due to factors including shade from shrubs, trees, and built structures. Patches of shade may function as microclimate refugia that allow species to occur in habitats where high temperatures and/or exposure to ultraviolet radiation would otherwise be prohibitive. However, the importance of shaded microhabitats for interactions between species across urbanized landscapes remains poorly understood. Weedy plants and their foliar pathogens are a tractable system for studying how multiple scales of climatic variation influence infection prevalence. Powdery mildew pathogens are particularly well suited to this work, as these fungi can be visibly diagnosed on leaf surfaces. We studied the effects of shaded microclimates on rates of powdery mildew infection on Plantago host species in (1) "pandemic pivot" surveys in which undergraduate students recorded shade and infection status of thousands of plants along road verges in urban and suburban residential neighborhoods, (2) monthly surveys of plant populations in 22 parks along an urbanization gradient, and (3) a manipulative field experiment directly testing the effects of shade on the growth and transmission of powdery mildew. Together, our field survey results show strong positive effects of shade on mildew infection in wild Plantago populations across urban, suburban, and rural habitats. Our experiment suggests that this relationship is causal, where microclimate conditions associated with shade promote pathogen growth. Overall, infection prevalence increased with urbanization despite a negative association between urbanization and tree cover at the landscape scale. These findings highlight the importance of taking microclimate heterogeneity into account when establishing links between macroclimate or land use context and prevalence of disease. We present a case of spontaneous nontraumatic retrobulbar hemorrhage associated with anti-coagulation therapy and a new diagnosis of scurvy. A 68-year-old male on chronic anti-coagulation therapy presented with a retrobulbar hemorrhage requiring urgent canthotomy and cantholysis. Despite the absence of a supratherapeutic INR and normal clotting factors, the patient continued to have spontaneous hemorrhages within the orbit and elsewhere. Workup revealed a severe vitamin C deficiency consistent with scurvy. Further investigation of dietary history demonstrated an avoidance of all citrus fruit upon starting warfarin due to misunderstanding in medication counseling on avoidance of grapefruit. With repletion of vitamin C and further medication counseling, the patient had no further episodes of spontaneous hemorrhage. [ABSTRACT FROM AUTHOR]

Productivity in the Arctic is expected to increase as temperatures and the number of open water days rise. With this increased productivity, the coastal shelves of the Arctic Ocean may act as a sink for atmospheric carbon. However, this storage is dependent on a sufficient nitrogen (N) supply and current literature on biogeochemical rates of N uptake in this region is severely limited. Here, we report the spatial extent and rate at which the aquatic microbial community utilizes inorganic and organic N substrates in the Alaskan Arctic during late summer. Uptake rates (> 0.3 μm) were measured in 2016 and 2017 using isotopically labeled ammonium (NH4+), nitrate (NO3−), urea, and mixed algal amino acids. Rates of NH4+ regeneration were also measured to investigate the contribution of remineralized N to primary production. Primary production was estimated using isotopically labeled bicarbonate. We found that N species uptake varied by location. Although NH4+ was the form of N taken up at the greatest rate at most sites, we also found that uptake rates of urea could be greater than NO3− and that amino acid uptake was widespread. Supporting previous studies, NO3− uptake rates were correlated with primary production rates. Variability in nutrient reservoirs and sampling conditions in the Chukchi Sea between the 2 yr were responsible for some of the variances observed in estimated NH4+ recycling rates and primary production. A man with early non-fluctuating Parkinson's disease developed disabling camptocormia. The patient was treated with posterior thoracolumbar fixation, which subsequently had to be augmented with anterior interbody fusion. Although the patient ultimately achieved excellent sagittal correction, his postoperative course was complicated and prolonged. Therefore, although this case demonstrates that spinal fixation surgery can be successful, it should probably only be offered after subthalamic nucleus deep brain stimulation has been unsuccessful, or for well motivated patients who express a strong wish for this major reconstructive surgery. In this paper, we study the propagation of quantum correlations in open quantum systems using quantum discord as their measure. The role of system-environment correlations in discord dynamics and some operational interpretations of discord are discussed, in particular, activation of correlations into entanglement. The quantum nature of correlations is studied in systems of optical modes, that is, Gaussian quantum states. A counter-intuitive scheme of entanglement distribution by an auxiliary mode, which remains separable at all times, is analyzed to unveil the synergy of coherence and dissipation in quantum protocols with mixed states. When cut scores for classifications occur on the total score scale, popular methods for estimating classification accuracy (CA) and classification consistency (CC) require assumptions about a parametric form of the test scores or about a parametric response model, such as item response theory (IRT). This article develops an approach to estimate CA and CC nonparametrically by replacing the role of the parametric IRT model in Lee's classification indices with a modified version of Ramsay's kernel-smoothed item response functions. The performance of the nonparametric CA and CC indices are tested in simulation studies in various conditions with different generating IRT models, test lengths, and ability distributions. The nonparametric approach to CA often outperforms Lee's method and Livingston and Lewis's method, showing robustness to nonnormality in the simulated ability. The nonparametric CC index performs similarly to Lee's method and outperforms Livingston and Lewis's method when the ability distributions are nonnormal. [ABSTRACT FROM AUTHOR]

Within the framework of item response theory (IRT), there are two recent lines of work on the estimation of classification accuracy (CA) rate. One approach estimates CA when decisions are made based on total sum scores, the other based on latent trait estimates. The former is referred to as the Lee approach, and the latter, the Rudner approach, each after its representative contributor. In this article, the two approaches are delineated in the same framework to highlight their similarities and differences. In addition, a simulation study manipulating IRT model, sample size, test length, and cut score location was conducted. The study investigated the empirical CA that can be achieved using either the total scores or the latent trait estimates. It also evaluated the performances of the two approaches in estimating their respective empirical CAs. Results on the empirical CA suggest that when the model fits, classifications made with the latent trait estimate shall be equally or more accurate than classifications made with total score. The magnitude of difference was governed by divergence from the one-parameter logistic (1PL) model. Both Lee and Rudner approaches provided good estimates of their respective empirical CAs for every condition that was simulated. Practical implications of the simulation results are discussed. Quinn N, Shulman A, Knifton L, Byrne P. The impact of a national mental health arts and film festival on stigma and recovery. This study aims to evaluate the impact of a national mental health arts festival for the general public, encompassing a wide variety of art forms and themes. An evaluation was undertaken with 415 attendees from 20 different events, combining qualitative and quantitative approaches. The findings demonstrate positive impact on the relationship between arts and mental health. Events increased positive attitudes, including positive representations of people's contributions, capabilities and potential to recover. They did not decrease negative attitudes. Intended behaviour change was modest and one film event increased audience perceptions of dangerousness. The paper argues that the arts can change stigma by constructing shared meanings and engaging audiences on an emotional level. Carefully programmed, collaborative, community-based arts festivals should form an integral part of national programmes to address stigma and to promote mental health and wellbeing, alongside traditional social marketing and public education approaches. The goal of this study was to investigate three potential ways that the soluble organic nitrogen (N) fraction of wastewater treatment plant (WWTP) effluents, termed effluent organic N (EON), could contribute to coastal eutrophication - direct biological removal, photochemical release of labile compounds, and salinity-mediated release of ammonium (NH4+). Effluents from two WWTPs were used in the experiments. For the bioassays, EON was added to water from four salinities (∼0 to 30) collected from the James River (VA) in August 2008, and then concentrations of N and phosphorus compounds were measured periodically over 48 h. Bioassay results, based on changes in DON concentrations, indicate that some fraction of the EON was removed and that the degree of EON removal varied between effluents and with salinity. Further, we caution that bioassay results should be interpreted within a broad context of detailed information on chemical characterization. EON from both WWTPs was also photoreactive, with labile NH4+ and dissolved primary amines released during exposure to sunlight. We also present the first data that demonstrate that when EON is exposed to higher salinities, increasing amounts of NH4+ are released, further facilitating EON use as effluent transits from freshwater through estuaries to the coast. Abstract: The aim of this study is to derive an equation that has the ability to model live weight of Irish dairy cows over lactation. The data set consisted of 6899 cows from 63 herds, of which 428 were from experimental herds and 6471 cows were from commercial herds. An initial examination focussed on time series techniques, as the data are of a time series nature. Splines were also examined to determine the dimensions of a model required to represent the data. As an incomplete gamma function, which was previously used to model milk yield, has been used in other studies to model live weight, various milk yield models were investigated. Finally, live weight changes between two calvings were modelled as a function of age, days in milk and pregnancy. As multicollinearity was evident in this function, the variance inflation factor was examined and principal component analysis was carried out on the variables responsible for multicollinearity. The proposed live weight model has a better fit than previous models, weak multicollinearity and the residuals are homoskedastic, independent and normally distributed. This live weight model therefore provides an acceptable level of accuracy in representing the shape of the live weight curve for Irish dairy cows and can be easily modified for different environmental scenarios. The basal ganglia are intimately connected to the frontal cortex via five fronto-striatal circuits. While the role of the frontal cortex in cognition has been extensively studied, the contribution of the basal ganglia to cognition has remained less clear. In Parkinson's disease, posteroventral pallidotomy (PVP) involves surgical lesioning of the internal section of the globus pallidus (GPi, the final output pathway from the basal ganglia) to relieve the motor symptoms of the disorder. PVP in Parkinson's disease provides a unique opportunity to investigate the impact of disruption of striatal outflow to the frontal cortex on cognition. We assessed executive function and working memory after withdrawal of medication in 13 patients with Parkinson's disease before and 3 months after unilateral PVP compared to 12 age- and IQ-matched normals assessed twice with an interval of 3 months. The tests used were: Wisconsin Card Sorting (WCST), Self-Ordered Random Number Sequences, Missing Digit Test, Paced Visual Serial Addition Test (PVSAT), and Visual Conditional Associative Learning Test (VCALT). After PVP, the patients performed significantly better on the Self-Ordered Random Number Sequences and the WCST, an improvement that was also observed in the normals across the two assessment and is therefore likely to reflect practice effects. Relative to the normals, the patients showed significant differential change following PVP on the Missing Digit Test and PVSAT, on which they performed worse after compared to before surgery, while the controls performed better on the second assessment. For the patients, performance on the VCALT also indicated deterioration after PVP, but the changes approached significance. The side of PVP had no effect on the results. The pattern of change observed 3 months after PVP was maintained at 15-month follow-up. The results suggest that striatal outflow to the frontal cortex may be essential for those aspects of executive function that showed deterioration after PVP. [ABSTRACT FROM AUTHOR]

A growing body of research is emerging regarding the relationship between parental adverse childhood experiences (ACEs) and negative health, well-being and developmental outcomes in their children. This systematic review seeks to understand the relationship between parental ACEs and the health, well-being and developmental outcomes of their children and whether the relationships differ according to the number and type of parental ACEs. Systematic review. The review includes articles published between 2000 and 2021 from studies using quantitative longitudinal methods and multivariate analysis to investigate the relationship between parental ACEs and their offspring's outcomes. Relevant studies were identified through a systematic search of five databases and findings synthesised using a narrative synthesis. This review was registered on PROSPERO (CRD42021274068). Nineteen studies met the inclusion criteria and were included in the review. This resulted in a combined population sample of 124,043 parents and 128,400 children. Diversity in measurement of parental ACE exposure and in the type of ACEs measured within the studies precluded a meta-analysis. Offspring of parents exposed to ACEs had a higher risk of a range of negative health, well-being and developmental outcomes. This relationship differs according to the number and type of parental ACEs, with a positive relationship observed between the number of parental ACEs and the risk of negative health, well-being and development outcomes in their children. These findings indicate that screening for parental ACEs by health visitors, midwives and other health or social care staff may identify an at-risk population of infants, children and adolescents and improve child outcomes. Here, we present the first study to investigate the seasonal importance of amino acid-nitrogen (N) to Arctic near shore microbial communities. We measured primary productivity and the uptake of ammonium, nitrate, urea, and amino acids in two size fractions (> 3 μm and approximately 0.7-3 μm), as well as ammonium regeneration and nitrification using 15N and 13C tracer approaches in the near-shore waters of the Chukchi Sea, during January, April, and August for two consecutive years. At discrete depths, nitrate comprised 46-78% of the total dissolved N pool during January and April but only 2-6% during August. Dissolved organic N (DON) concentrations increased between January and August though the carbon (C) : N (mol : mol) of the DON pool declined. Of the substrates tested, amino acids supported the bulk of both N and C nutrition in both size fractions during January and April (ice-covered). Urea generally had the lowest uptake rate under ice-covered conditions; uptake of urea-C was only detectable in August. Though previous Arctic studies focused largely on nitrate, we found nitrate uptake was generally lower than other substrates tested. The sharp decline in nitrate concentration between April and August, however, indicates a drawdown of nitrate during that period. Rates of ammonium uptake were highest in August, when it was the dominant N substrate used. During all sample periods, rates of ammonium regeneration were sufficient to supply ammonium demand. Due to its flexibility, the multiple-indicator, multiple-causes (MIMIC) model has become an increasingly popular method for the detection of differential item functioning (DIF). In this article, we propose the mediated MIMIC model method to uncover the underlying mechanism of DIF. This method extends the usual MIMIC model by including one variable or multiple variables that may completely or partially mediate the DIF effect. If complete mediation effect is found, the DIF effect is fully accounted for. Through our simulation study, we find that the mediated MIMIC model is very successful in detecting the mediation effect that completely or partially accounts for DIF, while keeping the Type I error rate well controlled for both balanced and unbalanced sample sizes between focal and reference groups. Because it is successful in detecting such mediation effects, the mediated MIMIC model may help explain DIF and give guidance in the revision of a DIF item. [ABSTRACT FROM AUTHOR]

The release of fresh water from major impoundments to meet the requirements of estuaries has become a recognized need and methodologies to define appropriate release strategies have become an urgent priority in some parts of the world. A recruitment index for juvenile marine fish is presented, based on information relating to the preferred recruitment periods for 27 species, as well as the extent of dependency of these species on estuaries. The status of the estuary mouth (i.e. open or closed) and the longitudinal salinity differences (i.e. the difference between the salinity at the estuary head and the mouth) in the estuary are obtained from a hydrodynamic model of the estuary. The recruitment index integrates this information to provide an indication of potential recruitment (or recruitment opportunity). This index was applied in a case study of the Great Brak Estuary, South Africa, and a distinct decrease in the recruitment index was associated with a reduction in freshwater inflow. In the estuary modelled, annual run-off could be halved without an appreciable decrease in the fish recruitment index (FRI), but with a potentially sharp decline in fish recruitment thereafter. While the index represented recruitment opportunity rather than actual recruitment, it does at least provide an indication of where ecological risk is likely to increase substantially. Since the index integrates the current understanding with respect to the key processes which regulate recruitment of juvenile marine fish into estuaries, it is suggested that it has the potential to fulfil a vital role in identifying appropriate freshwater release policies for impoundments in South Africa. Background Poor quality cardiopulmonary resuscitation (CPR) predicts adverse outcome. During invasive cardiac procedures automated-CPR (A-CPR) may help maintain effective resuscitation. The use of A-CPR following in-hospital cardiac arrest (IHCA) remains poorly described. Aims & methods Firstly, we aimed to assess the efficiency of healthcare staff using A-CPR in a cardiac arrest scenario at baseline, following re-training and over time (Scenario-based training). Secondly, we studied our clinical experience of A-CPR at our institution over a 2-year period, with particular emphasis on the details of invasive cardiac procedures performed, problems encountered, resuscitation rates and in-hospital outcome (AutoPulse-CPR Registry). Results Scenario-based training: Forty healthcare professionals were assessed. At baseline, time-to-position device was slow (mean 59 (± 24) s (range 15-96 s)), with the majority (57%) unable to mode-switch. Following re-training time-to-position reduced (28 (± 9) s, p < 0.01 vs baseline) with 95% able to mode-switch. This improvement was maintained over time. AutoPulse-CPR Registry: 285 patients suffered IHCA, 25 received A-CPR. Survival to hospital discharge following conventional CPR was 28/260 (11%) and 7/25 (28%) following A-CPR. A-CPR supported invasive procedures in 9 patients, 2 of whom had A-CPR dependant circulation during transfer to the catheter lab. Conclusion A-CPR may provide excellent haemodynamic support and facilitate simultaneous invasive cardiac procedures. A significant learning curve exists when integrating A-CPR into clinical practice. The development of psychometrically sound health-related quality of life (Hr-QoL) instruments has made it possible to assess subjectively experienced Hr-QoL quantitatively, and to incorporate Hr-QoL as a measure in medical research and clinical trials. Hr-QoL in patients with progressive supranuclear palsy (PSP) has been measured using generic (not disease-specific) instruments, or Parkinson’s disease-specific scales. We give an overview of the development of a disease-specific Hr-QoL instrument for patients with PSP. Based on interviews with 27 patients with PSP a preliminary Hr-QoL instrument was developed and administered to over 300 people with PSP in the UK and US. Following psychometric analysis a rating scale with a physical and a mental subscale emerged. In this patient sample, both subscales satisfied criteria for scaling assumptions, acceptability, reliability and validity (correlations with other measures consistent with a priori hypotheses). Monthly Maximum Discharge Limits (MMDL) have been established for selenium in irrigation drainage by the State of California and the U.S. Environmental Protection Agency following observations of avian teratogenesis at the Kesterson Reservoir in the San Joaquin Valley of California. As a result of these and other adverse effects, farmers and drainage districts on the western side of the San Joaquin Valley must reduce selenium concentrations in irrigation drainage discharged to the San Joaquin River. Drainage treatment will be required in the near future to meet existing MMDL and future Total Maximum Discharge Limits (TMDL) for the San Joaquin River. A 0.4-hectare Algal Bacterial Selenium Removal (ABSR) Facility was designed and constructed at the Panoche Drainage District in 1995 and 1996 using the Advanced Integrated Wastewater Pond Systems® or AIWPS® Technology. Each of two physically identical systems combined a Reduction Pond (RP) with a shallow, peripheral algal High Rate Pond (HRP). A Dissolved Air Flotation (DAF) unit and a slow sand filter were used to remove particulate selenium from the effluent of each system. The two systems were operated under different modes of operation and the bacterial substrate varied in each system. The rates of nitrate and selenium removal were compared. Microalgae were harvested using DAF and used as a carbon-rich substrate for nitrate- and selenate-reducing bacteria. Mass removals of total soluble selenium of 77% or greater were achieved over a three-year period. Nitrate and selenate were removed by assimiliatory and dissimiliatory bacterial reduction, and nitrate was also removed by algal assimilation. The final removal of particulate selenium is the focus of ongoing investigations. The removal of particulate selenium is expected to increase the overall removal of selenium to greater than 90% and would allow farmers and drainage districts to discharge irrigation drainage in compliance with regulatory discharge limits. The comparatively high cost of laboratory detection methods for Clostridium difficile infection (CDI) coupled to a low prevalence rate has resulted in testing algorithms that use cheaper and relatively sensitive screening methods, followed by more specific confirmatory methods. The aim of this prospectively-conducted study from two centres in the UK, and one in the Republic of Ireland was to determine the efficacy of the EntericBio ® realtime C. difficile Assay (EBCD) for the detection of toxigenic C. difficile in stool samples. The EBCD was compared to the in-use testing methods for Clostridium difficile (CD) detection in each centre. In the two UK centres, the EBCD was compared to the C.diff Quik Chek Complete ® kit (Techlab), and discrepancies were tested further using The Xpert ® C. difficile PCR assay (Cepheid) and PCR ribotyping after cultivation using the spore culture method, respectively. In the Irish centre, EBCD comparison was to an algorithm of C. DIFF CHEK™-60 test (Techlab) for screening followed by C. difficile Premier ™ Toxins A&B assay (Meridian Bioscience ® ) in the case of positive results; discrepancies were tested using the Xpert ® C. difficile PCR assay. In a retrospective analysis of data, a total of 947 stool samples were tested, of which eight (0.8%) proved inhibitory to the EBCD assay. Of the 939 valid tests conducted, reported sensitivities of the EBCD were 94.7%, 100% and 97.9%, respectively; specificities were 99.6%, 100% and 100%, respectively; positive predictive values were 94.7%, 100% and 100%, respectively, and negative predictive values were 99.6%, 100% and 99.8%, respectively. The Arctic is warming at a rate nearly twice the global average, leading to thawing permafrost, increased coastal erosion, and enhanced delivery of riverine terrestrially-derived dissolved organic matter (tDOM) to coastal waters. This humic-rich tDOM has the ability to attenuate light required for photosynthesis and stimulate heterotrophic growth by supplying a source of labile organic carbon. Due to tDOM's high carbon to nitrogen (C : N) ratio, additional nitrogen is required for microorganisms to utilize this excess carbon for growth, thus exacerbating competition between autotrophs and heterotrophs for limiting nutrients and potentially reducing primary production. The effect of Arctic tDOM additions on nitrate uptake by two microplankton size fractions in the coastal Chukchi Sea was quantified using 15N tracer methods. To assess the biogeochemical vs. spectral impacts of tDOM, the uptake incubations were amended with either tDOM or light attenuating films that mimic light absorption by the tDOM. Nitrate uptake and primary production rates in the larger, predominantly phytoplankton, size fraction generally decreased with increasing tDOM additions. The change in light attenuation alone accounted for a ∼ 50% reduction in nitrate uptake. Responses in the smaller size fraction varied seasonally with tDOM additions stimulating uptake in spring and suppressing it in summer. The largest variation in summer nitrate uptake can be explained by the shared effect of biogeochemistry and light attenuation. Therefore, large increases in tDOM delivery currently occurring and predicted to increase in the coastal Arctic, could reduce primary production, broadly impact nitrogen and carbon cycling, and affect higher trophic levels. Alfalfa (Medicago sativa) is one of the main forage crops for cattle feeding in Argentina. It is necessary to have information in the area of influence of the Faculty of Veterinary Sciences of the National University of La Pampa on the adaptation and productive behavior of alfalfa varieties offered in the market. Therefore, in the General Pico area, the Experimental, Experimental and Productive Unit of this institution, a trial of two alfalfa commercial cultivars of different degree of latency (intermediate and short) was carried out, applying two seed densities (8 And 16 kg / ha) and two fertilization doses (0 and 60 kg of superphosphate / ha). This publication evaluates the productive and economic results of the first year of production. On average, the intermediate latency variety produced more dry matter than short latency (P <0.05), but no significant differences were observed between seed densities. A tendency was found in favor of fertilized versus unfertilized plots. The economic analysis showed significant differences between variety and densities, but not with respect to fertilization. Mutations in the DJ-1 gene have recently been shown to cause autosomal recessive Parkinson's disease. To estimate the prevalence of this mutation, an analysis was undertaken of 39 index cases of Parkinson's disease in whom a family history suggested autosomal recessive inheritance. No DJ-1 mutations were found in these patients, indicating that this gene is unlikely to be of numerical significance in clinical practice. The hypothesis was also tested that young onset Parkinson's disease patients in whom, despite extensive analysis, only a single heterozygous parkin mutation was found, might harbour a second mutation in the DJ-1 gene--that is, digenic inheritance. No patient was found with a single mutation in both DJ-1 and parkin genes, making this mode of inheritance unlikely. Finally it was confirmed that PARK6 and PARK7 (DJ-1), despite being phenotypically similar and mapping to the same small chromosomal region of 1p36, are caused by mutations in separate genes. Introduction Individual social capital has been recognized as having an important role for health and well-being. We tested the hypothesis that poor social capital increases internalized stigma and, in turn, can reduce empowerment among people with major depressive disorder (MDD). Materials and methods This is a cross-sectional multisite study conducted on a sample of 516 people with MDD in 19 European countries. Structural Equation Models were developed to examine the direct and indirect effects of self-stigma and social capital on empowerment. Results Social capital and self-stigma accounted for 56% of the variability in empowerment. Higher social capital was related to lower self-stigma ( r = –0.72, P < 0.001) which, in turn, partially mediated the relationship between social capital and empowerment ( r = 0.38, P < 0.001). Conclusions Social capital plays a key role in the appraisal of empowerment, both directly and through the indirect effect mediated by self-stigma. In order to improve empowerment of people with MDD, we identify strategies to foster individual social capital, and to overcome the negative consequences related to self-stigma for attainment of life goals. Six extracts were prepared from hawthorn ( Crataegus monogyna ) leaves and flowers (HLF) and berries (HB) using solid–liquid [traditional ( T ) (HLF T , HB T ), sonicated ( S ) (HLF S , HB S )] and supercritical fluid ( C ) extraction (HLF C , HB C ) techniques. The antioxidant activities of HLF and HB extracts were characterised using in vitro antioxidant assays (TPC, DPPH, FRAP) and in 25% bovine muscle ( longissimus lumborum ) homogenates (lipid oxidation (TBARS), oxymyoglobin (% of total myoglobin)) after 24 h storage at 4 °C. Hawthorn extracts exhibited varying degrees of antioxidant potency. In vitro and muscle homogenate (TBARS) antioxidant activity followed the order: HLF S > HLF T and HB T > HB S . In supercritical fluid extracts, HLF C > HB C ( in vitro antioxidant activity) and HLF C ≈ HB C (TBARS). All extracts (except HB S ) reduced oxymyoglobin oxidation. The HLF S extract had the highest antioxidant activity in all test systems. To investigate nutrient release from decaying fish carcasses as a potential source fueling blooms of the toxic dinoflagellate, Karenia brevis (C.C. Davis) G. Hansen & Ø. Moestrup this study evaluated the type and quantity of nutrients (nitrogen (N), phosphorus (P), and carbon (C)) released from decaying fish using both wet chemistry and compound specific (Fourier transform ion cyclotron resonance mass spectrometry, FT-ICR MS) techniques. A 3 day bioassay experiment was then conducted to examine how eutrophic (Charlotte Harbor, CH) and oligotrophic (Blue Water, BW) microbial communities in the eastern Gulf of Mexico (GoM) respond to fish-derived nutrients (FDN) relative to other in situ N sources. Decaying fish released a suite of compounds, primarily in the form of ammonium (NH 4 + ); release rates ranged from 37.7 ± 12.6 to 102.3 ± 4.7 μmol N L −1 g −1 wet wt. d −1 . Phosphate release was 13.9 ± 2.8 to 21.4 ± 2.9 μmol P L −1 g −1 wet wt. d −1 . A total of 3515 organic compounds were produced, which were dominated by lipid and protein-like compounds. In the bioassay, FDN were utilized by microbial communities collected from both CH and BW, with complete drawdown of fish-derived NH 4 + in the CH treatment in conjunction with a 3-fold increase in Chl a . FDN provided the necessary nutrients to alleviate P limitation in the BW treatment, resulting in a 13-fold increase in Chl a . Results from this experiment indicate FDN are important N and P sources to the microbial community along the eastern GoM and have important implications for maintenance of associated ichthyotoxic K. brevis blooms. Background: We determined the prevalence of urethral Mycoplasma genitalium (MG) infection and whether infection was associated with circumcision status among men enrolled in the randomized trial of medical male circumcision to prevent HIV acquisition in Kisumu, Kenya.Methods: MG and Trichomonas vaginalis were detected in first void urine by APTIMA transcription-mediated amplification assay. first void urine and urethral swabs were assessed for Neisseria gonorrhoeae (NG) and Chlamydia trachomatis (CT) by polymerase chain reaction assay. Herpes simplex virus type 2 antibodies were detected by IgG ELISA. Multivariable logistic regression identified factors associated with MG infection.Results: Specimens were collected between July and September 2010, and 52 (9.9%; 95% confidence interval [CI]: 7.3%-12.4%) MG infections were detected among 526 men. N. gonorrhoeae and T. vaginalis were not associated with MG. CT coinfection was 5.8% in MG-infected men, and 0.8% among MG-uninfected men (P = 0.02). MG infection was predominantly asymptomatic (98%). The prevalence of MG was 13.4% in uncircumcised men versus 8.2% in circumcised men (P = 0.06). Being circumcised nearly halved the odds of MG (adjusted odds ratio [aQR] = 0.54; 95% CI: 0.29-0.99), adjusted for other variables significant at the P < 0.05 level: herpes simplex virus type 2 infection (aOR = 2.05; 95% CI: 1.05-4.00), CT infection (aOR = 2.69; 95% CI: 1.44-5.02), and washing the penis ≤1 hour after sex (aOR = 0.47; 95% CI: 0.24-0.95).Conclusions: MG infection was reduced among men who were circumcised, adding to the benefits of male circumcision in preventing several sexually transmitted infections. Patients with Parkinson's disease (PwPD) have a slow, shuffling gait, marked by sporadic freezing of gait (FoG) during which effective stepping ceases temporarily. As these gait problems are not commonly improved by medical and surgical treatments, alternative approaches to manage these problems have been adopted. The aim of this study was to evaluate the effect of real and virtual visual cues on walking in PD. We assessed 26 mid-stage PwPD, on and off medication, on a laboratory-based walking task which simulated real world challenges by incorporating FoG triggers and using appropriate placebo conditions. Cueing interventions were presented via virtual reality glasses (VRG rhythmic, visual flow and static placebo cues), and as transverse lines (TL) on the walkway. Objective measures of gait (task completion time; velocity, cadence, stride length; FoG frequency) and self-rated fear of falling (FoF) were recorded. Cueing intervention affected task completion time only off medication. Whereas placebo VRG cues provided no improvement in walking, visual flow VRG cues marginally reduced the task completion time. TL on the floor elicited more substantial improvements in gait with reduced cadence, increased stride length and reduced FoG frequency. VRG rhythmic cueing impaired overall walking. Notably, a final no-intervention condition yielded quicker task completion, greater walking velocity, increased stride length and less frequent FoG. Although the VRG produced modest improvements only in the visual flow condition, their flexibility is an advantage. Patients with Parkinson's disease experience motor and perceptual timing difficulties, which are ameliorated by dopaminergic medication. We investigated the neural correlates of motor timing in Parkinson's disease, including the effects of dopaminergic medication on patterns of brain activation. Eight patients with Parkinson's disease and eight healthy controls were scanned with H(15)(2)O positron emission tomography while engaged in three tasks: synchronization (right index finger tapping in synchrony with a tone presented at 1 Hz), continuation (tapping at 1 Hz in the absence of a tone), and a control simple reaction time task. During the first 6 scans, the patients were assessed after overnight withdrawal of medication. Scans 7-12 were completed with the patients in the 'ON' state, after injections of apomorphine, a dopamine receptor agonist. For the healthy controls, relative to the control reaction time task, motor timing (synchronization + continuation) was associated with significantly greater activation in left medial prefrontal cortex (Brodmann area 10, 32), right hippocampus, bilateral angular gyrus (Brodmann area 39), left posterior cingulate (Brodmann area 31) and left nucleus accumbens/caudate. This pattern of brain activation during motor timing was not observed for patients, who showed significantly greater activation in bilateral cerebellum, right thalamus and left midbrain/substantia nigra compared to the control participants. Relative to the externally-paced synchronization task, the internally controlled continuation task was associated with greater activation in the dorsolateral prefrontal cortex (Brodmann area 46/9) in both the control and Parkinson's disease groups. Analysis of medication-related effects indicated that cortical activation was significantly more predominant during motor timing when the patients were 'ON' medication, whereas pallidal and cerebellar activations were evident 'OFF' medication. Effective connectivity analysis established that activity in the left caudate nucleus was associated with increased activity in the right lentiform nucleus and cerebellum 'OFF' medication, and with increased activity in the prefrontal cortex 'ON' medication. These results suggest that in Parkinson's disease, in the 'OFF' medication state, excessive inhibitory pallidal outflow is associated with a lack of adequate frontal activation and reliance on the cerebellum for motor timing. In contrast, our results establish for the first time that administration of dopaminergic medication increases striatal-frontal connectivity between the caudate nucleus and prefrontal cortex during motor timing. Objectives: To determine the effect of electrode contact location on efficacy of bilateral globus pollidus internus (GPi) deep brain stimulation (DBS) for primary generalised dystonia (PGD). Subjects and methods: A consecutive series of 15 patients with PGD (10 females, mean age 42 years, seven DYT1) who underwent bilateral GPi DBS, were assessed using the Burke-Fahn-Marsden (BFM) dystonia scale before and 6 months after surgery. The position of the stimulated electrode contact(s) was determined from the postoperative stereotactic MRI. Contralateral limb and total axial BFM subscores were compared with the location of the stimulated contact(s) within the GPi. Results: The mean total BFM score decreased from 38.9 preoperatively to 11.9 at 6 months, an improvement of 69.5% (p<0.00001). Cluster analysis of the stimulated contact coordinates identified two groups, distributed along an anterodorsal to posteroventral axis. Clinical improvement was greater for posteroventral than anterodorsal stimulation for the arm (86% vs 52%; p<0.05) and trunk (96% vs 65%; p<0.05) and inversely correlated with the y coordinate. For the leg, posteroventral and anterodorsal stimulation were of equivalent efficacy. Overall clinical improvement was maximal with posteroventral stimulation (89% vs 67%; p<0.05) and inversely correlated with the y (A-P) coordinate (r= -0.62, p<0.05). Conclusion: GPi DBS is effective for PGD but outcome is dependent on contact location. Posteroventral GPi stimulation provides the best overall effect and is superior for the arm and trunk. Most patients with Parkinson’s disease (PD) develop dyskinesia and other motor complications after prolonged L-dopa use. We now report on the relationship between L-dopa dose and the duration and severity of dyskinesia in L-dopa-primed MPTP-treated primates with marked nigral degeneration mimicking late stage PD. With increasing doses of L-dopa, locomotor activity increased and motor disability declined. The duration of dyskinesia following L-dopa administration increased dose-dependently, and showed a linear correlation with total locomotor activity. In addition, the time-course of dyskinesia paralleled closely that of locomotor activity in a dose-dependent manner. In contrast, severity of dyskinesia showed a non-linear correlation with total locomotor activity, low doses of L-dopa eliciting severe dyskinesia for short periods of time. The threshold for dyskinesia induction and the antiparkinsonian effects of L-dopa appear very similar in primed MPTP primates mimicking late stage PD. Reducing individual doses of L-dopa to avoid severe dyskinesia can markedly compromise the antiparkinsonian response. Our results extend the relevance of the dyskinetic MPTP-treated primate in studying the genesis of involuntary movements occurring in L-dopa treated patients with PD. [ABSTRACT FROM AUTHOR]
