1. Structural geology of the Cadillac Group along the Malartic segment of the Larder Lake Cadillac deformation zone, Quebec, and implications for gold mineralization

Abstract

The gold-endowed Larder Lake Cadillac deformation zone (LLCDZ) in the Abitibi greenstone belt of the Archean Superior Province generally trends east but swings to the southeast along the Malartic segment in Quebec. Immediately north of the LLCDZ along this segment, deformation of the less than ca. 2687 Ma metasedimentary Cadillac Group began with the formation of isoclinal [F.sub.1] folds, which are overprinted by post-2676 [+ ou -] 2.0 Ma regional [F.sub.2] folds with an axial planar [S.sub.2] cleavage oriented parallel to the trend of the Malartic segment. Auriferous quartz veins strike east-west (085[degrees]) anticlockwise to bedding and [S.sub.2]. They are folded into sigmoidal S-shaped folds due to sinistral shear parallel to bedding. Later dextral shearing segmented the veins and folded the [F.sub.2] folds into Z-shaped [F.sub.3] folds. The veins are similar in structural chronology, fluid isotopic composition (calculated [Please download the PDF to view the formula] value between-9.5[per thousand] and-3.4[per thousand] and [Please download the PDF to view the formula] values of 12[per thousand]), and isotopic re-equilibration temperatures (469 and 498 [degrees]C, quartz-biotite oxygen thermometer) to the veins at the nearby Canadian Malartic deposit to the east. They possess comparable Au-As-Sb association and sericite-arsenopyrite alteration halo, as other Cadillac-Group-hosted veins at the Lapa mine to the west. Collectively, they constitute a vein field that extends across the entire length of the Malartic segment. Contrary to recent interpretations of the Malartic segment as an early accretionary structure that controlled the emplacement of gold deposits, its southeast trend is interpreted as the short limb of a broad Z-shaped flexure that formed during later [F.sub.3] folding of the LLCDZ. Immédiatement au nord de la ZDLLC le long de ce tronçon, la déformation du Groupe métasédimentaire de Cadillac, de < 2687 Ma environ, a commencé avec la formation de plis isoclinaux [F.sub.1], sur lesquels se sont surimposés, après 2676 [+ ou -] 2,0 Ma, des plis régionaux [F.sub.2] associés à un clivage de plan axial [S.sub.2] de direction parallèle à l'orientation du tronçon de Malartic. Ils sont déformés en des plis sigmoïdaux en S résultant d'un cisaillement senestre parallèle au litage. Un cisaillement dextre subséquent a segmenté les filons et replié les plis [F.sub.2] en des plis [F.sub.3] en Z. Les filons sont semblables sur le plan de la chronologie structurale, de la composition isotopique des fluides (valeur calculée de [Please download the PDF to view the formula] de-9,5 à-3,4 [per thousand] et valeurs de [Please download the PDF to view the formula] de 12 [per thousand]) et des températures de rééquilibration isotopique (469 et 498 [degrees]C, thermomètre basé sur l'oxygène dans le quartz et la biotite) aux filons dans le gisement Canadian Malartic non loin à l'est. Ils présentent une association Au-As-Sb et un halo d'altération à séricite-arsénopyrite semblables à d'autres filons dans le Groupe de Cadillac observés à la mine Lapa à l'ouest. Collectivement, ils constituent un champ filonien qui s'étend sur toute la longueur du tronçon de Malartic. Contrairement à des interprétations récentes du tronçon de Malartic comme étant une structure accrétionnaire précoce ayant contrôlé la mise en place de gisements aurifères, son orientation sud-est nous amène à l'interpréter comme étant le flan court d'une large flexure en Z qui s'est formée durant le plissement [F.sub.3] tardif de la ZDLLC. Abstract

International audience; Background The identification of patients at high risk of a disabling disease course would be invaluable in guiding initial therapy in Crohn’s disease (CD). Our objective was to evaluate a combination of clinical, serological, and genetic factors to predict complicated disease course in pediatric-onset CD. Methods Data for pediatric-onset CD patients, diagnosed before 17 years of age between 1988 and 2004 and followed more than 5 years, were extracted from the population-based EPIMAD registry. The main outcome was defined by the occurrence of complicated behavior (stricturing or penetrating) and/or intestinal resection within the 5 years following diagnosis. Lasso logistic regression models were used to build a predictive model based on clinical data at diagnosis, serological data (ASCA, pANCA, anti-OmpC, anti-Cbir1, anti-Fla2, anti-Flax), and 369 candidate single nucleotide polymorphisms. Results In total, 156 children with an inflammatory (B1) disease at diagnosis were included. Among them, 35% (n = 54) progressed to a complicated behavior or an intestinal resection within the 5 years following diagnosis. The best predictive model (PREDICT-EPIMAD) included the location at diagnosis, pANCA, and 6 single nucleotide polymorphisms. This model showed good discrimination and good calibration, with an area under the curve of 0.80 after correction for optimism bias (sensitivity, 79%, specificity, 74%, positive predictive value, 61%, negative predictive value, 87%). Decision curve analysis confirmed the clinical utility of the model. Conclusions A combination of clinical, serotypic, and genotypic variables can predict disease progression in this population-based pediatric-onset CD cohort. Independent validation is needed before it can be used in clinical practice.

Abstract

BACKGROUND: The present study evaluated the relative survival benefits associated with enasidenib and current standard of care (SoC) therapies for patients with relapsed/refractory (R/R) acute myeloid leukemia (AML) and an isocitrate dehydrogenase 2 (IDH2) mutation who are ineligible for hematopoietic stem cell transplantation (HSCT). METHODS: Propensity score matching (PSM) analysis compared survival outcomes observed with enasidenib 100 mg daily in the phase I/II AG221-C-001 trial and SoC outcomes obtained from a real-world chart review of patients in France. RESULTS: Before matching, enasidenib (n = 195) was associated with numerically improved overall survival (OS) relative to SoC (n = 80; hazard ratio [HR], 0.82; 95% confidence interval [CI], 0.61-1.11). After matching and adjusting for covariates (n = 78 per group), mortality risk was significantly lower with enasidenib than with SoC (HR, 0.67; 95% CI, 0.47-0.97). The median OS was 9.26 months for enasidenib (95% CI, 7.72-13.24) and 4.76 months for SoC (95% CI, 3.81-8.21). Results remained robust across all sensitivity analyses conducted. CONCLUSIONS: PSM analyses indicate that enasidenib significantly prolongs survival relative to SoC among patients with R/R AML and an IDH2 mutation who are ineligible for HSCT. Future prospective studies are needed to validate these findings using other data sources and to assess the comparative efficacy of enasidenib for other treatment outcomes.

Abstract

The characterization of protein interactions has become essential in many fields of life science, especially drug discovery. Microscale thermophoresis (MST) is a powerful new method for the quantitative analysis of protein-protein interactions (PPIs) with low sample consumption. In addition, one of the major advantages of this technique is that no tedious purification step is necessary to access the protein of interest. Here, we describe a protocol using MST to determine the binding affinity of the PD-1/PD-L1 couple, which is involved in tumour escape processes, without purification of the target protein from cell lysates. The method requires the overexpression of fluorescent proteins in CHO-K1 cells and describes the optimal conditions for determining the dissociation constant. The protocol has a variety of potential applications in studying the interactions of these proteins with small molecules and demonstrates that MST is a valuable method for studying the PD-1/PD-L1 pathway. 7

Abstract

Hypomethylating agents are able to prolong the overall survival of some patients diagnosed with acute myeloid leukemia. The aim of this study was to evaluate the clinical use of azacitidine as front-line therapy in unfit acute myeloid leukemia (AML) patients and to develop a clinical prediction model to identify which patients may benefit more from the drug. One hundred and ten untreated unfit AML patients received front-line azacitidine therapy in Spain, and response and survival were evaluated in them following European LeukemiaNet (ELN) guidelines. A clinical prediction rule was obtained from this population that was validated and refined in 261 patients treated in France, Austria and Italy. ELN response was achieved in 21.0% of the 371 patients (CI95% 17.0-25.5) and did not depend on bone marrow blast cell percentage. Median overall survival was 9.6 months (CI95% 8.5-10.8) and 40.6% of the patients were alive at 1 year (CI95% 35.5-45.7). European ALMA score (E-ALMA), based on performance status, white blood cell counts at azacitidine onset and cytogenetics, discriminated three risk groups with different survival and response rates. Azacitidine seems a reasonable therapeutic option for most unfit AML patients, i.e. those displaying a favorable or intermediate E-ALMA score. © 2014 Elsevier Ltd.
