The Geoengineering Model Intercomparison Project (GeoMIP) has proposed multiple model experiments during phases 5 and 6 of the Climate Model Intercomparison Project (CMIP), with the latest set of model experiments proposed in 2015. With phase 7 of CMIP in preparation and with multiple efforts ongoing to better explore the potential space of outcomes for different solar radiation modifications (SRMs) both in terms of deployment strategies and scenarios and in terms of potential impacts, the GeoMIP community has identified the need to propose and conduct a new experiment that could serve as a bridge between past iterations and future CMIP7 experiments. Here we report the details of such a proposed experiment, named G6-1.5K-SAI, to be conducted with the current generation of scenarios and models from CMIP6 and clarify the reasoning behind many of the new choices introduced. Namely, compared to the CMIP6 GeoMIP scenario G6sulfur, we decided on (1) an intermediate emission scenario as a baseline (the Shared Socioeconomic Pathway 2-4.5), (2) a start date set in the future that includes both considerations for the likelihood of exceeding 1.5 °C above preindustrial levels and some considerations for a likely start date for an SRM implementation, and (3) a deployment strategy for stratospheric aerosol injection that does not inject in the tropical pipe in order to obtain a more latitudinally uniform aerosol distribution. We also offer more details regarding the preferred experiment length and number of ensemble members and include potential options for second-tier experiments that some modeling groups might want to run. The specifics of the proposed experiment will further allow for a more direct comparison between results obtained from CMIP6 models and those obtained from future scenarios for CMIP7. Sulfate aerosol in the stratosphere is an important climate driver, causing solar dimming in the years after major volcanic eruptions. Hence, a growing number of general circulation models are adapting interactive sulfur and aerosol schemes to improve the representation of relevant chemical processes and associated feedbacks. However, uncertainties of these schemes are not well constrained. Stratospheric sulfate is modulated by natural emissions of sulfur-containing species, including volcanic eruptive, and anthropogenic emissions. Model intercomparisons have examined the effects of volcanic eruptions, whereas the background conditions of the sulfur cycle have not been addressed in a global model intercomparison project. Assessing background conditions in global models allows us to identify model discrepancies as they are masked by large perturbations such as volcanic eruptions, yet may still matter in the aftermath of such a disturbance. Here, we analyze the atmospheric burden, seasonal cycle, and vertical and meridional distribution of the main sulfur species among nine global atmospheric aerosol models that are widely used in the stratospheric aerosol research community. We use observational and reanalysis data to evaluate model results. Overall, models agree that the three dominant sulfur species in terms of burdens (sulfate aerosol, OCS, and SO2) make up about 98 % of stratospheric sulfur and 95 % of tropospheric sulfur. However, models vary considerably in the partitioning between these species. Models agree that anthropogenic emission of SO2 strongly affects the sulfate aerosol burden in the Northern Hemispheric troposphere, while its importance is very uncertain in other regions. The total deposition of sulfur varies among models, deviating by a factor of two, but models agree that sulfate aerosol is the main form in which sulfur is deposited. Additionally, the partitioning between wet and dry deposition fluxes is highly model dependent. We investigate the areas of greatest variability in the sulfur species burdens and find that inter-model variability is low in the tropics and increases towards the poles. Seasonality in the southern hemisphere is depicted very similar among models. Differences are largest in the dynamically active northern hemispheric extratropical region, hence some of the differences could be attributed to the differences in the representation of the stratospheric circulation among underlying general circulation models. This study highlights that the differences in the atmospheric sulfur budget among the models arise from the representation of both chemical and dynamical processes, whose interplay complicates the bias attribution. Several problematic points identified for individual models are related to the specifics of the chemistry schemes, model resolution, and representation of cross-tropopause transport in the extratropics. Further model intercomparison research is needed focusing on the clarification of the reasons for biases, given also the importance of this topic for the stratospheric aerosol injection studies. Recent model inter-comparison studies highlighted model discrepancies in reproducing the climatic impacts of large explosive volcanic eruptions, calling into question the reliability of global aerosol model simulations for future scenarios. Here, we analyse the simulated evolution of the stratospheric aerosol plume following the well observed June 1991 Mt. Pinatubo eruption by six interactive stratospheric aerosol microphysics models in comparison to a range of observational data sets. Our primary focus 5 is on the uncertainties regarding initial SO2 emission following the Pinatubo eruption in 1991, as prescribed in the Historical Eruptions SO2 Emission Assessment experiments (HErSEA), in the framework of the Interactive Stratospheric Aerosol Model Intercomparison Project (ISA-MIP). Six global models with interactive aerosol microphysics took part in this study: ECHAM6-SALSA, EMAC, ECHAM5-HAM, SOCOL-AERv2, ULAQ-CCM and UM-UKCA. Model simulations are performed by varying SO2 injection amount (ranging between 5 and 10 Tg-S), and the altitude of injection (between 18-25 km). We find that the common and main weakness among all the models is that they can not reproduce the persistence of the sulfate aerosols in the stratosphere. Most models show a stronger transport towards the extratropics in the northern hemisphere, at the expense of the observed tropical confinement, suggesting a much weaker subtropical barrier in all the models, that results in a shorter e-folding time compared to the observations. Moreover, the simulations in which more than 5 Tg-S of SO2 are injected show a large surface area density a few months after the eruption compared to the values measured in the tropics and the in-situ measurements over Laramie. This results in an overestimation of the number of particles globally during the build-up phase and an underestimation in the Southern Hemisphere, which draws attention to the importance of including processes as the ash injection and the eruption of Cerro Hudson. Sulfate geoengineering (SG) methods based on lower stratospheric tropical injection of sulfur dioxide (SO2) have been widely discussed in recent years, focusing on the direct and indirect effects they would have on the climate system. Here a potential alternative method is discussed, where sulfur emissions are located at the surface in the form of carbonyl sulfide (COS) gas. A time-dependent chemistry-climate model experiment is designed from year 2021 to 2055, assuming a 40 Tg-S/yr artificial global flux of COS, geographically distributed following the present day anthropogenic COS surface emissions. The budget of COS and sulfur species is discussed, as well as the effects of SG-COS on the stratospheric sulfate aerosol optical depth (Δτ=0.080 in years 2046-2055), aerosol effective radius (0.46 µm), surface SOx deposition (+8.7 %) and tropopause radiative forcing (RF) (-2.0 W/m² for clear sky conditions and -1.5 W/m² including the cloud adjustment). Indirect effects on ozone, methane and stratospheric water vapor are also considered, along with the COS direct contribution (with an overall gas phase global radiative forcing of +0.23W/m²). According to our model results, the resulting net RF of this SG-COS experiment is -1.3 W/m² for the year 2050, and it is comparable to the corresponding RF of -1.7 W/m² obtained with a sustained injection of 4 Tg-S/yr in the tropical lower stratosphere in the form of SO2 (SG-SO2, able to produce a comparable increase of the sulfate aerosol optical depth). Significant changes of the stratospheric ozone response are found in SG-COS with respect to SG-SO2 (+4.9 DU versus +1.5 DU, globally). According to the model results, the resulting UVB perturbation at the surface accounts to -4.3% as a global-annual average (versus -2.4% in the SG-SO2 case), with a springtime Antarctic decrease of -2.7% (versus a +5.8% increase in the SG-SO2 experiment). Overall, we find that an increase in COS surface emission may be feasible, and produce a more latitudinally-uniform forcing without the need for the deployment of stratospheric aircrafts. Abstract Background Diabetes is well known risk factor for thrombotic events. The association between diabetes and venous thromboembolism is still matter of debate. However, during diabetes an acquired thrombophilia is present and is due to the non-enzymatic glycosilation of clotting inhibitors as antithrombin thus leading to hypercoagulable state. A possibile relationship between the presence of FVL gene variant in type 1 or type 2 diabetes has been hypothysed by several reports in the Literature with non-univocal findings. Patients and methods Retrospectively we analysed nearly 7000 patients referred to our Thrombosis Center for venous thromboembolism (VTE) then we selected 115 patients underwent to the screening for inherited thrombophilia. All selected patients were divided in 2 groups: the first group (group A) included 64 patients with previous VTE and carriers of factor V Leiden, while the second group (group B) included 51 patients with previous VTE and evetually carriers of thrombophilic defects other than factor V Leiden. Patients of group B acted as control group. 75 g oral glucose tolerance Test (OGTT) recommended by WHO was perfomed to all subjects in the study in order to screen subjects with glucose reduced tolerance or subjects with inducible diabetes. Statistical analysis was performed with STATA 6 http://www.stata.com with Student t test for unpaired data, with χ2 test or with Fisher exact test where appropriated; differences were considered to be significant if p < 0.05. Results We did not find sifferences between glycaemia at baseline and after OGTT between patients with VTE carriers of FVL compared to non-carriers of FVL. We found a relevant increase in the prevalence of IGT and diabetes between patients with VTE carriers of FVL compared to non-carriers of FVL although this increase did not raise statistical significance. Discussion our data pointed out an interesting aspect of the linking between FVL gene variant, diabetes and atherothrombosis and other vascular complications, although data on larger population are needed; this aspect may be another relevant topic of research based because also a link between the pathogenesis of venous thrombosis and atherothrombosis has been recently reported in the Literature.

Abstract Native Factor V is an anticoagulant, but when activated by thrombin, Factor X or platelet proteases, it becomes a procoagulant. Due to these double properties, Factor V plays a crucial role in the regulation of coagulation/anticoagulation balance. Factor V Leiden (FVL) disorder may lead to thrombophilia. Whether a reduction in the activation of Factor V or Factor V Leiden may correct the disposition to thrombophilia is unknown. Therefore we tested SV-IV Peptide 1–16 (i.e. a peptide derived by seminal protein vescicle number IV, SV-IV) to assess its capacity to inhibit the procoagulant activity of normal clotting factor V or Factor V Leiden (FVL). We found that SV-IV protein has potent anti-inflammatory and immunomodulatory properties and also exerts procoagulant activity. In the present work we show that the SV-IV Peptide 1–16, incubated with plasma containing normal Factor V or FVL plasma for 5 minutes reduces the procoagulant capacity of both substances. This is an anticoagulant effect whereas SV-IV protein is a procoagulant. This activity is effective both in terms of the coagulation tests, where coagulation times are increased, and in terms of biochemical tests conducted with purified molecules, where Factor X activation is reduced. Peptide 1–16 was, in the pure molecule system, first incubated for 5 minutes with purified Factor V then it was added to the mix of phosphatidylserine, Ca2+, Factor X and its chromogenic molecule Chromozym X. We observed a more than 50% reduction in lysis of chromogenic molecule Chromozym X by Factor Xa, compared to the sample without Peptide 1–16. Such reduction in Chromozym X lysis, is explained with the reduced activation of Factor X by partial inactivation of Factor V by Peptide 1–16. Thus our study demonstrates that Peptide 1–16 reduces the coagulation capacity of Factor V and Factor V Leiden in vitro, and, in turn, causes factor X reduced activation.

Abstract Background Many available data have suggested that hyperhomocysteinaemia, an established independent risk factor for thrombosis (arterial and venous), may be associated with an increased risk of retinal vein occlusion (RVO). Aim of the study To evaluate homocysteine metabolism in consecutive caucasian patients affected by RVO from Northern Italy. Patients and Methods 69 consecutive patients from Northern Italy (mean age 64.1 ± 14.6 yy) with recent RVO, were tested for plasma levels of homocysteine (tHcy: fasting and after loading with methionine), cyanocobalamine and folic acid levels (CMIA-Abbot) and looking for MTHFR C677T mutation (Light Cycler-Roche) and compared to 50 volunteers, enrolled as a control group. Results Fasting levels of tHcy were significantly higher in patients than in controls: mean value 14.7 ± 7.7 vs 10.2 ± 8 nmol/ml. Post load levels were also significantly higher: mean value 42.7 ± 23.7 vs 30.4 ± 13.3 nmol/ml; Total homocysteine increase was also evaluated (i.e. Δ-tHcy) after methionine load and was also significantly higher in patients compared to control subjects: mean Δ-tHcy 27.8 ± 21.5 vs 21.0 ± 16 nmol/ml (normal value < 25 nmol/ml). Furthermore, patients affected by RVO show low folic acid and/or vitamin B12 levels, although differences with control group did not reach statistical significance. Heterozygous and homozygous MTHFR mutation were respectively in study group 46% and 29% vs control group 56% and 4%. Conclusion our data confirm that hyperhomocysteinaemia is a risk factor for RVO, and also that TT genotype of MTHFR C677T is more frequently associated with RVO: if the mutation per se is a risk factor for RVO remains an open question to be confirmed because another study from US did not reveal this aspect. Hyperomocysteinemia is modifiable risk factor for thrombotic diseases. Therefore, a screening for tHcy plasma levels in patients with recent retinal vein occlusion could allow to identify patients who might benefit from supplementation with vitamins and normalization of homocysteine levels, in fasting and after methionine load.

Ozone profile measurements collected at L'Aquila (Italy, 42.4° N) during seventeen years of radio-sounding (2000-2016) are presented here, with an analysis of derived trends. Model results from the SPARC-CCMI exercise are used in parallel to highlight the physical and chemical mechanisms regulating mid-latitude ozone trends. The statistically significant trends highlighted in time series at L'Aquila are those in the mid-upper stratosphere (+5.9 ± 4.2), mid troposphere (+5.9 ± 2.4) and upper troposphere (+2.5 ± 0.9), all in percent/decade. The upper stratospheric positive trend was already well documented in recent WMO assessments and attributed to the starting decline of stratospheric Cly and Bry and to the stratospheric cooling induced by increasing well mixed greenhouse gases, thus slowing down gas-phase reactions that destroy ozone in the upper stratosphere. The ozone increase in the mid-upper troposphere is largely regulated by the increasing strength of the Brewer-Dobson circulation, which moves more ozone from the tropics to the extratropics and enhances the tropospheric influx from the lowermost stratosphere. This climate feedback mechanism on tropospheric ozone is only partially compensated by the increasing chemical ozone loss associated to higher H2O values in response to the tropospheric warming. We also note that ozone trends obtained in the lower stratosphere are negative (-2.2 percent/decade), but do not result to be statistically significant in our analyses. [ABSTRACT FROM AUTHOR]

Hydroxyl radical (OH) plays critical roles within the troposphere, such as determining the lifetime of methane (CH4), yet is challenging to model due to its fast cycling and dependence on a multitude of sources and sinks. As a result, the reasons for variations in OH and the resulting CH4 lifetime (τCH4), both between models and in time, are difficult to diagnose. We apply a neural network (NN) approach to address this issue within a group of models that participated in the Chemistry-Climate Model Initiative (CCMI). Analysis of the historical specified dynamics simulations performed for CCMI indicates that the primary drivers of τCH4 differences among ten models are the flux of UV light to the troposphere (indicated by the photolysis frequency JO¹D) due mostly to clouds, mixing ratio of tropospheric ozone (O3), the abundance of nitrogen oxides (NOx≡NO+NO2), and details of the various chemical mechanisms that drive OH. Water vapor, carbon monoxide (CO), the ratio of NO:NOx, and formaldehyde (HCHO) explain moderate differences in τCH4, while isoprene, CH4, the photolysis frequency of NO2 by visible light (JNO2), overhead O3 column, and temperature account for little-to-no model variation in τCH4. We also apply the NNs to analysis of temporal trends in OH from 1980 to 2015. All models that participated in the specified dynamics historical simulation for CCMI demonstrate a decline in τCH4 during the analysed timeframe. The significant contributors to this trend, in order of importance, are tropospheric O3, JO¹D, NOx, and H2O, with CO also causing substantial interannual variability in OH burden. Finally, the identified trends in τCH4 are compared to calculated trends in the tropospheric mean OH concentration from previous work, based on analysis of observations. The comparison reveals a robust result for the effect of rising water vapor on OH and τCH4, imparting an increasing and decreasing trend of about 0.5 % decade−1, respectively. The responses due to NOx, O3 column, and temperature are also in reasonably good agreement between the two studies, though a discrepancy in the CH4 response highlights a need for further examination of the CH4 feedback on the abundance of OH. Background: A possible interference between lupus anticoagulant (LAC), a well characterized clotting inhibitor, in the International Normalized Ratio (INR) determination during oral anticoagulation (OA) has been reported in the literature. Few data are available about the relationship between this kind of interference and the daily clinical management of oral anticoagulation. The aim of the study is to evaluate the role of two different thromboplastins-RecombiPlasTin 2G and HepatoComplex-in the determination of INR values of several patients' ongoing OA for a previous thrombotic disorder with and without positivity to LAC, and to evaluate possible interferences in the daily therapeutic approach. Patients and methods: We selected 16 patients (13 females and 3 males, mean age 59 ± 16 years) with LAC positivity ongoing OA and 11 control subjects (7 females and 4 males, mean age 58 ± 14.5 years) with similar characteristics (ie, ethnic background and weight) with LAC negativity ongoing OA. 165 assays for INR determination were analyzed from both groups. Statistical analysis was performed using STATA 10 software. P values were considered significant if ,0.05. Results: Mean values of INR for patients with LAC positivity were 3.79 ± 1.63 when tested with RecombiPlasTin 2G vs 3.18 ± 1.15 when tested with HepatoComplex (P < 0.001, s); while mean values of INR for patients with antiphospholipid syndrome (APS) with LAC negativity were 3.54 ± 1.39 when tested with RecombiPlasTin 2G vs 3.23 ± 1.14 when tested with HepatoComplex (P < 0.002, s). An INR value > than 4.5 was found in 31/165 samples in 9 subjects, 8 patients with LAC positivity, and 1 control group subject with LAC negativity. There was a great difference in INR values in these subjects if we use the common thromboplastin (ie, RecombiPlasTin 2G) with a INR range varying from 5.14 ± 0.35 vs 3.79 ± 0.38 if we use another thromboplastin (ie, HepatoComplex) (P < 0.001, s). A change in the therapeutic approach for OA is possible in these cases because different INR values were obtained using different thromboplastins. Discussion: Our data confirm that INR evaluation does not reveal significant changes also if tested with two different thromboplastins, for patients ongoing OA with and without LAC positivity, when the INR value is < than 4. Over this INR value there is a significant difference in patients with LAC positivity if we use a different thromboplastin for the INR determination. For this reason values obtained by RecombiPlasTin 2G need to be confirmed and matched with another thromboplastin (ie, HepatoComplex). This approach may be useful in order to have a good INR testing for the chronic long-term treatment with OA in particular in patients with LAC positivity. [ABSTRACT FROM AUTHOR]
