From the establishment of a seat of government by Kublai Khan, Emperor Shizu of the Yuan Dynasty, at his residence at Jinlianchuan Prairie to the construction of the city of Kaiping, and from thence to the establishment of Kaiping as Xanadu, the northern capital of the Yuan Dynasty, a series of pivotal historical events influencing trends within the Yuan Empire took place in this steppe city. Shizu ultimately chose to implement a two-capital system, traveling back and forth between Xanadu and Khanbaliq to handle court administration and attend simultaneously to the needs of the Central Plains and the grasslands. By means of this structure, the Mongolian rulers declared their ownership of the territories of the Central Plains as well as the steppe, establishing their dominance and control by means of a ceremonial symbol. Analyzing the political and historical significance of this geographical space from Kaiping to Xanadu makes it clear that this region was an important historical artifact for the evolution of the Chinese people as an ethnic community with pluralistic unity. That the Yuan Dynasty achieved the integration of ethnic minorities and Han Chinese as one family as well as territorial unification represents a great step forward in Chinese history. Extinguishing the previously acquired fear is critical for the adaptation of an organism to the ever-changing environment, a process requiring the engagement of GABAA receptors (GABAARs). GABAARs consist of tens of structurally, pharmacologically, and functionally heterogeneous subtypes. However, the specific roles of these subtypes in fear extinction remain largely unexplored. Here, we observed that in the medial prefrontal cortex (mPFC), a core region for mood regulation, the extrasynaptically situated, δ-subunit-containing GABAARs [GABAA(δ)Rs], had a permissive role in tuning fear extinction in male mice, an effect sharply contrasting to the established but suppressive role by the whole GABAAR family. First, the fear extinction in individual mice was positively correlated with the level of GABAA(δ)R expression and function in their mPFC. Second, knockdown of GABAA(δ)R in mPFC, specifically in its infralimbic (IL) subregion, sufficed to impair the fear extinction in mice. Third, GABAA(δ)R-deficient mice also showed fear extinction deficits, and re-expressing GABAA(δ)Rs in the IL of these mice rescued the impaired extinction. Further mechanistic studies demonstrated that the permissive effect of GABAA(δ)R was associated with its role in enabling the extinction-evoked plastic regulation of neuronal excitability in IL projection neurons. By contrast, GABAA(δ)R had little influence on the extinction-evoked plasticity of glutamatergic transmission in these cells. Infectious bursal disease virus (IBDV), which targets bursa B lymphocytes, causes severe immunosuppressive disease in chickens, inducing huge economic losses for the poultry industry. To date, the functional receptor for IBDV binding and entry into host cells remains unclear. This study used mass spectrometry to screen host proteins of chicken bursal lymphocytes interacting with VP2. The chicken transmembrane protein cluster of differentiation 44 (chCD44) was identified and evaluated for its interaction with IBDV VP2, the major capsid protein. Overexpression and knockdown experiments showed that chCD44 promotes replication of IBDV. Furthermore, soluble chCD44 and the anti-chCD44 antibody blocked virus binding. The results of receptor reconstitution indicated that chCD44 overexpression conferred viral binding capability in nonpermissive cells. More important, although we found that IBDV could not replicate in the chCD44-overexpressed nonpermissive cells, the virus could enter nonpermissive cells using chCD44. In omics studies, different sources of information about the same set of genes are often available. When the group structure (e.g., gene pathways) within the genes are of interests, we combine the normal hierarchical model with the stochastic block model, through an integrative clustering framework, to model gene expression and gene networks jointly. The integrative framework provides higher accuracy in extensive simulation studies when one or both of the data sources contain noises or when different data sources provide complementary information. An empirical guideline in the choice between integrative versus separate clustering models is proposed. The integrative clustering method is illustrated on the mouse embryo single cell RNAseq and bulk cell microarray data, which identified not only the gene sets shared by both data sources but also the gene sets unique in one data source. Infectious bursal disease virus (IBDV) is an important member of the Birnaviridae family, causing severe immunosuppressive disease in chickens. The major capsid protein VP2 is responsible for the binding of IBDV to the host cell and its cellular tropism. In order to find proteins that potentially interact with IBDV VP2, a liquid chromatography-mass spectrometry (LC-MS) assay was conducted, and the host chicken CD74 protein was identified. Here, we investigate the role of chicken CD74 in IBDV attachment. Coimmunoprecipitation assays indicated that the extracellular domain of CD74 interacted with the VP2 proteins of multiple IBDV strains. Knockdown and overexpression experiments showed that CD74 promotes viral infectivity. Confocal assays showed that CD74 overexpression allows the attachment of IBDV and subvirus-like particles (SVPs) to the cell surface of nonpermissive cells, and quantitative PCR (qPCR) analysis further confirmed the attachment function of CD74. Anti-CD74 antibody, soluble CD74, depletion of CD74 by small interfering RNA (siRNA), and CD74 knockdown in the IBDV-susceptible DT40 cell line significantly inhibited IBDV binding, suggesting a pivotal role of this protein in virus attachment. Objective: Regression from prediabetes to normal glucose regulation (NGR) was associated with reduced incidence of diabetes by 56% over 10 years in participants in the Diabetes Prevention Program Outcomes Study (DPPOS). In an observational analysis, we examined whether regression to NGR also reduced risk for microvascular disease (MVD).Research Design and Methods: Generalized estimating equations were used to examine the prevalence of aggregate MVD at DPPOS year 11 in people who regressed to NGR at least once (vs. never) during the Diabetes Prevention Program (DPP). Logistic regression assessed the relationship of NGR with retinopathy, nephropathy, and neuropathy, individually. Generalized additive models fit smoothing splines to describe the relationship between average A1C during follow-up and MVD (and its subtypes) at the end of follow-up.Results: Regression to NGR was associated with lower prevalence of aggregate MVD in models adjusted for age, sex, race/ethnicity, baseline A1C, and treatment arm (odds ratio [OR] 0.78, 95% CI 0.65-0.78, P = 0.011). However, this association was lost in models that included average A1C during follow-up (OR 0.95, 95% CI 0.78-1.16, P = 0.63) or diabetes status at the end of follow-up (OR 0.92, 95% CI 0.75-1.12, P = 0.40). Similar results were observed in examination of the association between regression to NGR and prevalence of nephropathy and retinopathy, individually. Risk for aggregate MVD, nephropathy, and retinopathy increased across the A1C range.Conclusions: Regression to NGR is associated with a lower prevalence of aggregate MVD, nephropathy, and retinopathy, primarily due to lower glycemic exposure over time. Since 2015, severe hydropericardium-hepatitis syndrome (HHS), caused by a highly pathogenic fowl adenovirus 4 (FAdV-4), emerged in China. In our previous study, the FAdV-4 has been identified as a novel genotype with a unique 1966-bp nucleotide deletion (1966Del) between open reading frame 42 and 43. In this study, the natural 1966Del was frequently identified among 17 clinical isolates and other reported Chinese clinical strains. To investigate the relationship between 1966Del and the increased virulence of the novel FAdV-4, a CRISPR/Cas9 operating platform for FAdV-4 was developed for the first time in this study. Based on this platform, a Re1966 strain was rescued, inserted the relative 1966Del sequence of a nonpathogenic strain KR5. In the pathogenicity study, the Re1966 strain retained high virulence for specific-pathogen-free chickens, similar to the parental wild-type HLJFAd15, although the survival time of chickens infected with Re1966 was much longer. Therefore, the natural 1966Del was identified as a non-essential site for the increased virulence of the emerged novel FAdV-4. Although further research on the virulence-determining region or point within the genome of the novel FAdV-4 is needed, the CRISPR/Cas9 operating platform for the novel FAdV-4 was developed and successfully applied to edit the genomic DNA for the first time, and it provides a novel powerful tool for both basic virology studies and vaccine vector development of FAdVs. Background • Diabetic retinopathy (DR) is the most prevalent microvascular complication of diabetes. Panretinal photocoagulation (PRP) is the established treatment for mitigating severe visual impairment resulting from proliferative DR. Objective • This study aims to investigate the impact of PRP on the macular region in patients with DR, utilizing optical coherence tomography (OCT) for assessment. Design • An experimental study was meticulously designed, implementing PRP as the primary intervention. Setting • The investigation was conducted within the Department of Ophthalmology at the Affiliated Huaian No.1 People’s Hospital, Huai’an, Jiangsu, China. Participants • A total of 120 participants diagnosed with DR and undergoing treatment at our hospital were enrolled in the study. Interventions • The participants were randomly assigned to either the control group (CG, n=60) or the study group (SG, n=60). The CG received conventional drug treatment involving oral iodized lecithin, while the SG received PRP. OCT was employed to monitor changes in macular fovea volume and macular retinal thickness. Primary Outcome Measures • Evaluation criteria encompassed clinical efficacy, macular fovea volume, macular retinal thickness, IL-6 and VEGF levels, incidence of adverse reactions, and quality of life in both groups. Results • The study resulted in a higher total effective rate in the SG (96.67%) compared to the CG (80.00%) (χ2 = 8.09, P < .05). Post-treatment, reductions were observed in macular fovea volume and macular retinal thickness, with significantly lower SG values than CG values (P < .05). Both serum IL-6 and VEGF levels exhibited reductions in both groups after treatment, with the SG displaying a more significant decrease compared to the CG (P < .05). The occurrence of adverse reactions significantly decreased in the SG relative to the CG (P < .05). Quality of life scores for the SG was notably elevated compared to the CG (P<.05). Conclusions • PRP emerges as a highly valuable approach in the management of DR. It contributes to retinal thickness improvement within the macular region and inflammation reduction, and also enhances therapeutic outcomes, minimizes adverse reactions, and optimizes patients’ quality of life. Objective: Both lifestyle and metformin interventions can delay or prevent progression to type 2 diabetes mellitus (DM) in people with impaired glucose regulation, but there is considerable interindividual variation in the likelihood of receiving benefit. Understanding an individual's 3-year risk of progressing to DM and regressing to normal glucose regulation (NGR) might facilitate benefit-based tailored treatment.Research Design and Methods: We used the values of 19 clinical variables measured at the Diabetes Prevention Program (DPP) baseline evaluation and Cox proportional hazards models to assess the 3-year risk of progression to DM and regression to NGR separately for DPP lifestyle, metformin, and placebo participants who were adherent to the interventions. Lifestyle participants who lost ≥5% of their initial body weight at 6 months and metformin and placebo participants who reported taking ≥80% of their prescribed medication at the 6-month follow-up were defined as adherent.Results: Eleven of 19 clinical variables measured at baseline predicted progression to DM, and 6 of 19 predicted regression to NGR. Compared with adherent placebo participants at lowest risk of developing diabetes, participants at lowest risk of developing diabetes who adhered to a lifestyle intervention had an 8% absolute risk reduction (ARR) of developing diabetes and a 35% greater absolute likelihood of reverting to NGR. Participants at lowest risk of developing diabetes who adhered to a metformin intervention had no reduction in their risk of developing diabetes and a 17% greater absolute likelihood of reverting to NGR. Participants at highest risk of developing DM who adhered to a lifestyle intervention had a 39% ARR of developing diabetes and a 24% greater absolute likelihood of reverting to NGR, whereas those who adhered to the metformin intervention had a 25% ARR of developing diabetes and an 11% greater absolute likelihood of reverting to NGR.Conclusions: Unlike our previous analyses that sought to explain population risk, these analyses evaluate individual risk. The models can be used by overweight and obese adults with fasting hyperglycemia and impaired glucose tolerance to facilitate personalized decision-making by allowing them to explicitly weigh the benefits and feasibility of the lifestyle and metformin interventions. Since 2015, China has experienced outbreaks of severe hydropericardium syndrome (HPS), associated with a novel genotype and hypervirulent fowl adenovirus serotype 4 (FAdV-4) infection, with a prevalence in various provinces of the country. This has resulted in huge economic losses in the poultry industry. The novel FAdV-4 showed new genome characters, such as the natural deletion of open reading frame (ORF) 19 and ORF 27 (1966 bp), and high pathogenicity toward chickens. These are coupled with severe hydropericardium, inclusion body hepatitis, and mortality rates ranging from 30% to 90%. Although several inactivated and subunit vaccines against the traditional FAdV-4 have been developed, no commercial vaccine against the emerged disease caused by the novel strain has been available until now. The potential risks of infection with this novel hypervirulent FAdV-4 urgently require an effective vaccine. Thus, an inactivated oil-emulsion FAdV-4 vaccine formulated with the novel genotype virus was developed in this study. The vaccine provided a high level of antibody, preferential T helper 2 (Th2) (interleukin-4 secretion) not Th1 (interferon-secretion) response, and full protection against a lethal dose of the novel hypervirulent FAdV-4. Silkworm pathogens have been heavily impeding the development of sericultural industry and play important roles in lepidopteran ecology, and some of which are used as biological insecticides. Rapid advances in studies on the omics of silkworm pathogens have produced a large amount of data, which need to be brought together centrally in a coherent and systematic manner. This will facilitate the reuse of these data for further analysis. We have collected genomic data for 86 silkwormpathogens from 4 taxa (fungi, microsporidia, bacteria and viruses) and from 4 lepidopteran hosts, and developed the open-access Silkworm Pathogen Database (SilkPathDB) to make this information readily available. The implementation of SilkPathDB involves integrating Drupal and GBrowse as a graphic interface for a Chado relational database which houses all of the datasets involved. The genomes have been assembled and annotated for comparative purposes and allow the search and analysis of homologous sequences, transposable elements, protein subcellular locations, including secreted proteins, and gene ontology. We believe that the SilkPathDB will aid researchers in the identification of silkworm parasites, understanding the mechanisms of silkworm infections, and the developmental ecology of silkworm parasites (gene expression) and their hosts. Clinically relevant weight loss is achievable through lifestyle modification, but unintentional weight regain is common. We investigated whether recently discovered genetic variants affect weight loss and/or weight regain during behavioral intervention. Participants at high-risk of type 2 diabetes (Diabetes Prevention Program [DPP]; N = 917/907 intervention/comparison) or with type 2 diabetes (Look AHEAD [Action for Health in Diabetes]; N = 2,014/1,892 intervention/comparison) were from two parallel arm (lifestyle vs. comparison) randomized controlled trials. The associations of 91 established obesity-predisposing loci with weight loss across 4 years and with weight regain across years 2-4 after a minimum of 3% weight loss were tested. Each copy of the minor G allele of MTIF3 rs1885988 was consistently associated with greater weight loss following lifestyle intervention over 4 years across the DPP and Look AHEAD. No such effect was observed across comparison arms, leading to a nominally significant single nucleotide polymorphism×treatment interaction (P = 4.3 × 10(-3)). However, this effect was not significant at a study-wise significance level (Bonferroni threshold P < 5.8 × 10(-4)). Background: Inflammatory cytokines and transforming growth factor-β (TGF-β) are mutually inhibitory. However, hyperactivation of nuclear factor-κB (NF-κB) and TGF-β signaling both emerge in glioblastoma. Here, we report microRNA-148a (miR-148a) overexpression in glioblastoma and that miR-148a directly suppressed Quaking (QKI), a negative regulator of TGF-β signaling. Methods: We determined NF-κB and TGF-β/Smad signaling activity using pNF-κB-luc, pSMAD-luc, and control plasmids. The association between an RNA-induced silencing complex and QKI, mitogen-inducible gene 6 (MIG6), S-phase kinase-associated protein 1 (SKP1), and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA was tested with microribonucleoprotein immunoprecipitation and real-time PCR. Xenograft tumors were established in the brains of nude mice. Results: QKI suppression induced an aggressive phenotype of glioblastoma cells both in vitro and in vivo. Interestingly, we found that NF-κB induced miR-148a expression, leading to enhanced-strength and prolonged-duration TGF-β/Smad signaling. Notably, these findings were consistent with the significant correlation between miR-148a levels with NF-κB hyperactivation and activated TGF-β/Smad signaling in a cohort of human glioblastoma specimens. Conclusions: These findings uncover a plausible mechanism for NF-κB-sustained TGF-β/Smad activation via miR-148a in glioblastoma, and may suggest a new target for clinical intervention in human cancer. PURPOSE: To assess the results of iris suture fixation of subluxated intraocular lenses. DESIGN: Retrospective study. METHODS: This was a nonrandomized chart review of eyes with subluxated intraocular lenses that underwent iris suture fixation at an academic institutional care center. Seventy-two eyes of 67 consecutive patients were included. The following cases were excluded: posterior dislocations necessitating pars plana vitrectomy; secondary implantations for aphakia; and iris suture fixation at primary cataract extraction. Main outcome measures included visual acuity improvement, surgically induced astigmatism, and postsurgical complications. RESULTS: The mean follow-up duration was 16.64 ± 24.37 months (median = 4.03 months). All patients had preoperative monocular diplopia or unstable vision attributable to the subluxated intraocular lenses, and 40.3% of them required aphakic correction. There was an overall improvement in best-corrected visual acuity from a mean preoperative logMAR 0.35 ± 0.32 (Snellen equivalent~20/45) to logMAR 0.21 ± 0.25 (20/32, P = .001). There was no significant change in astigmatism secondary to the surgery. The mean difference in preoperative keratometry readings was 1.6 ± 1.07 diopter (D), whereas the mean postoperative manifest refraction astigmatic error (vertexed to the corneal surface) was 1.29 ± 0.92 D (P < .02). Re-subluxations occurred in 7 eyes during follow-up; the majority of these eyes underwent repeat fixation. Most (93.55%) intraocular lenses were stable and centered at the final follow-up. Glaucoma developed in 2 eyes postoperatively. CONCLUSIONS: Iris suture fixation of subluxated intraocular lenses was efficacious for the eyes included in this study, and it led to long-term stability of the intraocular lenses in 93.55% of cases. Estimation of the boundary condition is a critical problem in simulating hemodynamics in microvascular networks. This paper proposed a boundary estimation strategy based on a particle swarm optimization (PSO) algorithm, which aims to minimize the number of vessels with inverted flow direction in comparison to the experimental observation. The algorithm took boundary values as the particle swarm and updated the position of the particles iteratively to approach the optimization target. The method was tested in a real rat mesenteric network. With random initial boundary values, the method achieved a minimized 9 segments with an inverted flow direction in the network with 546 vessels. Compared with reported literature, the current work has the advantage of a better fit with experimental observations and is more suitable for the boundary estimation problem in pulsatile hemodynamic models due to the experiment-based optimization target selection. OBJECTIVE This study examined specific measures of weight loss in relation to incident diabetes and improvement in cardiometabolic risk factors. RESEARCH DESIGN AND METHODS This prospective, observational study analyzed nine weight measures, characterizing baseline weight, short- versus long-term weight loss, short- versus long-term weight regain, and weight cycling, within the Diabetes Prevention Program (DPP) lifestyle intervention arm (n = 1,000) for predictors of incident diabetes and improvement in cardiometabolic risk factors over 2 years. RESULTS Although weight loss in the first 6 months was protective of diabetes (hazard ratio [HR] 0.94 per kg, 95% CI 0.90, 0.98; P < 0.01) and cardiometabolic risk factors (P < 0.01), weight loss from 0 to 2 years was the strongest predictor of reduced diabetes incidence (HR 0.90 per kg, 95% CI 0.87, 0.93; P < 0.01) and cardiometabolic risk factor improvement (e.g., fasting glucose: β = -0.57 mg/dL per kg, 95% CI -0.66, -0.48; P < 0.01). Weight cycling (defined as number of 5-lb [2.25-kg] weight cycles) ranged 0-6 times per participant and was positively associated with incident diabetes (HR 1.33, 95% CI 1.12, 1.58; P < 0.01), fasting glucose (β = 0.91 mg/dL per cycle; P = 0.02), HOMA-IR (β = 0.25 units per cycle; P = 0.04), and systolic blood pressure (β = 0.94 mmHg per cycle; P = 0.01). After adjustment for baseline weight, the effect of weight cycling remained statistically significant for diabetes risk (HR 1.22, 95% CI 1.02, 1.47; P = 0.03) but not for cardiometabolic traits. In the present study, we aimed to determine whether mice with coronavirus-induced encephalomyelitis (CIE) develop neurogenic bladder dysfunction that is comparable with the neurogenic detrusor overactivity observed in patients with multiple sclerosis. Adult mice (C57BL/6J, 8 wk of age, n = 146) were inoculated with a neurotropic strain of mouse hepatitis virus (A59 strain) and followed for 4 wk. Inoculation with the virus caused a significant neural deficit in mice with an average clinical symptom score of 2.6 ± 0.5 at 2 wk. These changes were accompanied by 25 ± 5% weight loss at 1 and 2 wk postinoculation (P ≤ 0.001 vs. baseline) followed by a recovery phase. Histological analysis of spinal cord sections revealed multifocal sites of demyelinated lesions. Assessment of micturition patterns by filter paper assay determined an increase in the number of small and large urine spots in CIE mice starting from the second week after inoculation. Cystometric recordings in unrestrained awake animals confirmed neurogenic bladder overactivity at 4 wk postinoculation. One week after inoculation with the A59 strain of mouse hepatitis virus, mice became increasingly sensitive to von Frey filament testing with responses enhanced by 45% (n = 8, P ≤ 0.05 vs. baseline at 4 g); however, this initial increase in sensitivity was followed by gradual and significant diminution of abdominal sensitivity to mechanical stimulation by 4 wk postinoculation. Our results provide direct evidence showing that coronavirus-induced demyelination of the central nervous system causes the development of a neurogenic bladder that is comparable with neurogenic detrusor overactivity observed in patients with multiple sclerosis. Techniques that model microvascular hemodynamics have been developed for decades. While the physiological significance of pressure pulsatility is acknowledged, most of the microcirculatory models use steady flow approaches. To theoretically study the extent and transmission of pulsatility in microcirculation, dynamic models need to be developed. In this paper, we present a one-dimensional model to describe the dynamic behavior of microvascular blood flow. The model is applied to a microvascular network from a rat mesentery. Intravital microscopy was used to record the morphology and flow velocities in individual vessel segments, and boundaries are defined according to the experimental data. The system of governing equations constituting the model is solved numerically using the discontinuous Galerkin method. An implicit integration scheme is adopted to increase computing efficiency. The model allows the simulation of the dynamic properties of blood flow in microcirculatory networks, including the pressure pulsatility (quantified by a pulsatility index) and pulse wave velocity (PWV). From the main input arteriole to the main output venule, the pulsatility index decreases by 66.7%. PWV obtained along arterioles declines with decreasing diameters, with mean values of 77.16, 25.31, and 8.30 cm/s for diameters of 26.84, 17.46, and 13.33 pm, respectively. Highly dispersive nanospheres of MnFe2O4 are prepared by template free hydrothermal method. The nanospheres have 47.3-nm average diameter, narrow size distribution, and good crystallinity with average crystallite size about 22 nm. The reaction temperature strongly affects the morphology, and high temperature is found to be responsible for growth of uniform nanospheres. Raman spectroscopy reveals high purity of prepared nanospheres. High saturation magnetization (78.3 emu/g), low coercivity (45 Oe, 1 Oe = 79.5775 A·cm−1), low remanence (5.32 emu/g), and high anisotropy constant 2.84 × 104 J/m3 (10 times larger than bulk) are observed at room temperatures. The nearly superparamagnetic behavior is due to comparable size of nanospheres with superparamagnetic critical diameter Dcrspam. The high value of Keff may be due to coupling between the pinned moment in the amorphous shell and the magnetic moment in the core of the nanospheres. The nanospheres show prominent optical absorption in the visible region, and the indirect band gap is estimated to be 0.98 eV from the transmission spectrum. A variational-integral perturbation method (VIPM) has been established by combining the variational perturbation with the integral perturbation. We present the results obtained by the application of VIPM to the ground and excited states associated with the Zeeman effect on a hydrogen atom. We construct correction wave functions and calculate energy corrections for the ground state and some high excited states. These values are compared with results of Smith and Jason et al.; our results are more accurate than those from Smith and Jason, especially in the 4f excited state where we find a very interesting result that when magnetic fields increase, the energy level trap appears. The anaerobic global regulator FNR from Escherichia coli is a [4Fe-4S]2+ duster-containing dimer that is inactivated by O2 through disruption of the Fe-S duster and conversion to the monomeric apoprotein. It was shown that apo-FNR is subject to ClpXP proteolysis, and two recognition sites, amino acids 5-11 and amino acids 249 and 250, are responsible for targeting PNR to the protease. However, how the exposure of these sites is mediated such that only apo-FNR is recognized by the ClpXP protease and is degraded in a regulated manner so that a sufficient and similar FNR level is maintained in both anaerobic and aerobic conditions is unknown. To investigate this, we performed three-alanine scanning on amino acids 2-19 and 236-250 that are in the proximity of the two ClpXP recognition sites, and their functions remain unknown. We found that three-alanine substitution of residues 239-241 (LAQ239-241A3) and 242-244 (LAG242-244A3) caused reduced FNR protein levels, transcription activities, and growth rates under anaerobic conditions. Lu vivo degradation assays demonstrated that these mutants were degraded significantly faster than the wild type (WT), and either deletion of clpXP or blocking the ClpXP recognition Site of amino acids 249 and 250 stabilizes these proteins. Circular dichroism analysis revealed that introduction of LAQ239-241A3 caused conformational changes with a significant loss of secondary structures in both WT and an O2 stable FNR dimer, FNR D 154A. We propose that the region of amino acids 239-244 plays a negative role in the proteolysis of FNR by promoting a structural fold that limits the exposure of the proximal ClpXP site to the protease. OBJECTIVE--We tested genetic associations with weight loss and weight regain in the Diabetes Prevention Program, a randomized controlled trial of weight loss--inducing interventions (lifestyle and metformin) versus placebo. RESEARCH DESIGN AND METHODS--Sixteen obesity-predisposing single nucleotide polymorphisms (SNPs) were tested for association with short-term (baseline to 6 months) and long-term (baseline to 2 years) weight loss and weight regain (6 months to study end). RESULTS--Irrespective of treatment, the Ala12 allele at PPARG associated with short- and long-term weight loss (20.63 and 20.93 kg/allele, P ≤ 0.005, respectively). Gene-treatment interactions were observed for short-term (LYPLAL1 rs2605100, Plifestyle*SNP = 0.032; GNPDA2 rs10938397, Plifestyle*SNP = 0.016; MTCH2 rs10838738, Plifestyle*SNP = 0.022) and long-term (NEGR1 rs2815752, Pmetformin*SNP = 0.028; FTO rs9939609, Plifestyle*SNP = 0.044) weight loss. Three of 16 SNPs were associated with weight regain (NEGR1 rs2815752, BDNF rs6265, PPARG rs1801282), irrespective of treatment. TMEM18 rs6548238 and KTCD15 rs29941 showed treatment-specific effects (Plifestyle*SNP < 0.05). Background: Since 2008, a progressive pneumonia has become prevalent in broilers and laying hens. This disease occurs the first day after hatching and lasts more than 30 days, resulting in approximately 70% morbidity and 30% mortality in broilers. The objective of this study was to isolate and identify the pathogens that are responsible for the progressive pneumonia and establish an animal model for drug screening. Results: 193 serum samples were collected from 8 intensive farms from 5 provinces in China and analysed in the current research. Our clinical survey showed that 65.2% to 100% of breeding broilers, breeding layers, broilers and laying hens were seropositive for ORT antibodies. From 8 intensive farms, six ORT isolates were identified by PCR and biochemical assays, and two H9N2 viruses were isolated. Newcastle Disease Virus (NDV) and Infectious Bronchitis Virus (IBV) were excluded. Typical pneumonia and airsacculitis were observed both in broilers inoculated intraperitoneally with an ORT isolate alone and in those co-infected with ORT and H9N2 virus isolates. Specifically, the survival rate was 30%, 20%, 70%, 50% and 90% in birds inoculated with ORT+H9N2 virus, ORT followed by H9N2 virus, H9N2 virus followed by ORT, and ORT or H9N2 virus alone, respectively. Conclusions: The results of this study suggest that ORT infections of domestic poultry have been occurring frequently in China. ORT infection can induce higher economic losses and mortality if H9N2 AIV is also present. Although the isolation of ORT and H9N2 virus has been reported previously, there have been no reported co-infections of poultry with these two pathogens. This is the first report of co-infection of broilers with ORT and H9N2 virus, and this co-infection is probably associated with the outbreak of broiler airsacculitis in China, which has caused extensive economic losses. To assess the fairness of a jury system, courts compare data on the race–gender–ethnic composition of a number of jury venires (pools) to that of the jury-eligible population of the jurisdiction. The recent U.S. Supreme Court opinion in the Berghuis v. Smith case discussed the criteria a defendant's evidence must meet to establish a prima facie case that the jury pools were not representative. The decision relied on the three criteria set previously in Duren v. Missouri: name a distinct subgroup of the population, demonstrate their underrepresentation with statistics and identify the aspect of the jury system, which produced the disparity. In addition to formal statistical testing, courts have considered the absolute and comparative differences between the percentages of minorities in the age-eligible population and on the jury venires. In Berghuis, data for the period when the defendant's trial was held indicate that 6.03% of the potential jurors in the circuit (felony) courts were African American but they formed 7.28% of the age-eligible residents of the County. The 1.25% absolute difference seems small. The comparative difference or percentage shortfall is 18%, suggesting underrepresentation. The defendant argued that the disparity in the circuit courts arose because, at the time of his trial, jurors were first assigned to local courts and African Americans were concentrated in Grand Rapids. Shortly after the trial, the County changed the system, however, the comparative disparity (CD) only declined by 3%. This small change failed to convince the Court that the juror assignment system caused the underrepresentation. The available data in Berghuis v. Smith differed from most jury discrimination cases as the racial identification of the venire members were unavailable. The defendant's expert estimated the number of African Americans on the venires from their addresses and compared it to their expected number derived from their fraction of the county's age-eligible population. He did not test whether the difference was statistically significant. The two analyses available to the Court treated the estimated number of minority jurors on the venires as though it was the actual number, which is statistically incorrect. This article derives the appropriate standard deviation analysis. Applying it to the data in Berghuis, it is shown that there was a statistically significant disparity between the percentages of African Americans on the venires and in the age-eligible population. On the other hand, the difference between the CDs in the two periods was not near significance. Unlabelled: Stratifin plays an important role in cancer biology by interfering with intracellular signalling pathways and cell-cycle checkpoints. Decreased expression of stratifin gene has been reported to be a poor prognostic indicator in a variety of human malignant tumors.Aim: To clarify the role and prognostic significance of stratifin in esophageal squamous cell carcinoma (ESCC).Methods: The alteration of stratifin messenger RNA (mRNA) and protein was analyzed by reverse-transcription and quantitative real-time polymerase chain reaction (QRT-PCR) and Western blotting in 20 paired ESCC and nonneoplastic esophageal mucosa tissues, respectively. Then, immunohistochemistry (IHC) was used to evaluate expression of stratifin in tissues of 148 ESCC patients (including the former 20 pairs of tissues) and correlate it with clinicopathological parameters and prognosis of ESCC patients.Results: The stratifin level of mRNA and protein was markedly downregulated in ESCC tissue compared with in corresponding nonneoplastic esophageal epithelium (P<0.05). Similarly, the positive rate of stratifin protein expression was lower in the esophageal cancer than in paired nonneoplastic esophageal epithelium as detected by IHC (P=0.007). Statistically, the downregulation of stratifin expression was correlated with tumor infiltration depth (P=0.003), lymph node metastasis (P=0.008), distant metastasis (P=0.013), and lymphovascular invasion (P=0.007) of ESCC. Furthermore, the reduced stratifin expression was associated with shorter 5-year survival rate of ESCC patients after curative surgery (P<0.0001). On the basis of univariate and multivariate Cox regression analysis, we found that reduced stratifin expression, T4 stage, lymph node metastasis, and distant metastasis were independent risk factors for worse prognosis in ESCC patients.Conclusion: The present report indicates that stratifin could be a useful indicator for prognosis of this disease, as well as a potential target for more effective therapy. Deceleration capacity (DC) of heart rate is a novel indicator of autonomic nervous system (ANS) activity. In this paper, we proposed a modified DC index based on improved phase-rectified signal averaging (PRSA) algorithm. Sinusoidal analysis is applied to elucidate the rationality of the improved PRSA. Then the validity of the modified DC is verified by the databases of chronic heart failure (CHF) patients and control group. Both the conventional and modified DCs are significantly lower in CHF patients than that in the control group (2.12 ± 2.98 vs. 6.34 ± 1.92 ms, P < 0.0001 and 5.45 ± 2.48 vs. 10.64 ± 1.76 ms, P < 0.0001, respectively). And the modified DC provides higher accuracy in distinguishing CHF than the conventional one (87.4 vs. 82.1%). The results indicate that the suggested technique enhances the performance of PRSA and improves the efficiency of DC in assessing ANS activity in CHF patients. [ABSTRACT FROM AUTHOR]

In this article, we consider the setting where the event of interest can occur repeatedly for the same subject (i.e., a recurrent event; e.g., hospitalization) and may be stopped permanently by a terminating event (e.g., death). Among the different ways to model recurrent/terminal event data, the marginal mean (i.e., averaging over the survival distribution) is of primary interest from a public health or health economics perspective. Often, the difference between treatment-specific recurrent event means will not be constant over time, particularly when treatment-specific differences in survival exist. In such cases, it makes more sense to quantify treatment effect based on the cumulative difference in the recurrent event means, as opposed to the instantaneous difference in the rates. We propose a method that compares treatments by separately estimating the survival probabilities and recurrent event rates given survival, then integrating to get the mean number of events. The proposed method combines an additive model for the conditional recurrent event rate and a proportional hazards model for the terminating event hazard. The treatment effects on survival and on recurrent event rate among survivors are estimated in constructing our measure and explain the mechanism generating the difference under study. The example that motivates this research is the repeated occurrence of hospitalization among kidney transplant recipients, where the effect of expanded criteria donor (ECD) compared to non-ECD kidney transplantation on the mean number of hospitalizations is of interest. The purpose of this note is to demonstrate that courts ignore useful information when they fail to consider formal statistical analyses of small to moderate sized data sets. Data from the Ninth Circuit's reversal of a summary judgment decision against four plaintiffs who complained of age discrimination after being discharged in the context of a seasonal slowdown will be used to illustrate the usefulness of statistical analysis. The appellate decision considered both job performance and other factors specific to each plaintiff and statistical information on 44 employees derived from a chart in evidence. The opinion discounted a difference of 9.5 years between the average age of the 16 discharged employees and the average age of the 16 new hires, because the difference of 9.5 years was less than the court's requirement of a minimum ten-year difference to demonstrate age discrimination. The court also found that the sample sizes were too small to form a reliable basis for analysis. In addition, the court described patterns of some simple measures, such as the decrease in the average age of new hires over time, but the court did not apply formal statistical methods to analyze either the hiring or the termination data. After giving a brief review of the case and the statistical evidence summarized in the decision, this note applies formal statistical tests to reexamine the data pertaining to the issues of whether there was a decrease in the average age of new hires over time and whether the age of an employee increased his probability of being reduced in force (RIFed) during the time period of most relevance to the case. Because the plaintiffs alleged that a newly hired supervisor was responsible for their being laid off, three time periods were considered in the opinion(n1) and turn out to be important in the current analysis: (1) before this supervisor was hired, (2) when he co-supervised the crew, and (3) when he became the crew's sole supervisor. The results of the analysis presented here show that there was a statistically significant decline in the ages of new hires over time. Although only 16 employees were RIFed during the period, both a graphical display of the data and a formal statistical analysis indicate that the probability of an employee older than fifty years of age being discharged increased noticeably after the new supervisor took over. While a formal test of the layoff data does not quite reach statistical significance, when the formal test is combined with the result of the hiring data using Fisher's summary Chi-square method, the overall result provides further support for the appellate decision. The Peters–Belson method has been used as an alternative approach to standard linear regression analysis to examine potential wage discrimination as it also accounts for any differences in the qualifications of the applicants. To apply the Peters–Belson method, one first conducts a regression analysis on the favoured class and applies the resulting model to the non-favoured class to identify whether this class would have received a different rate of pay if they had been treated the same as their favoured counterparts. Since this method was recently extended to explore disparities in the personnel selections via logistic regression, we will examine the general properties of this method as compared to standard regression analysis. The effects of the demographic mix and size of the applicant pool, the difference in the distribution of the qualifications and the employer's evaluation criteria on the estimated disparity will be examined. After plaintiffs prevail in employment discrimination cases, courts need to estimate the compensation they deserve. This issue arose after a defendant was found to have discriminated against job applicants older than 40 years. In order to compensate the successful plaintiffs, the court needed to estimate the length of time they would have worked, had they been hired. The job histories of 127 current employees and 43 employees who recently left were available for analysis. The accelerated failure time model, commonly used for the analysis of survival data, provides an estimate of the mean employment duration, adjusting for covariates. Three legal opinions emphasized that damage awards should be based on data concerning employees with similar qualifications as the plaintiffs so an ability model is proposed. A simulation study illustrates the potential underestimation of the job tenures of the deserving plaintiffs by the method adopted in the case. On the other hand, that method, which did not consider ability, can lead to compensating low-ability plaintiffs who would not be hired even in a non-discriminatory environment. To investigate the biological impact of polycyclic aromatic hydrocarbons (PAHs) on reproductive system, scallops Chlamys farreri were continuously exposed to benzo[a]pyrene (BaP) (0.5, 3, 10μgL−1) during 15 days. DNA damage and histological alterations were examined in female gonad. DNA strand break levels significantly increased after 12h exposure, and remained high and significantly different from control values until the end of the exposure. In the ovaries of the scallops exposed to 10μgL−1 BaP for ten days, histological analysis showed that the cytoplasts of the oocytes in the outer layer of the ovaries became sparse, and the nuclear membranes were obscure. Damaging effects on ovaries and oocytes were more severe after 15 days exposure. Degenerating oocytes increased and the connective tissue of the ovary envelops became loose. Electron microscopic examination revealed ultrastructural aspects of degenerating oocyte and degenerated oocyte after 15 days exposure. [Copyright &y& Elsevier]

Volatile compounds formed by photosensitized oxidation and autoxidation of cod liver oil were compared. A 5-mL portion of oil-in-water emulsion (10% oil, 1% Tween 20 and 0.1 mM of rose bengal or 0.1 mM of ferrous chloride) was sealed into a 50-mL crimp top vial. The vial with added rose bengal was exposed to a 100-watt tungsten bulb (6000 lux) in an oven at 40°C, while the vial with added ferrous chloride was wrapped in aluminum foil to shield light and kept in the same oven. Volatiles in the headspace of the vials were collected by solid phase microextraction technique and analyzed with a gas chromatography-flame ionization detector and a gas chromatography– mass spectrometry. Approximately 51 kinds of volatiles were identified from oxidation of cod liver oil. All the volatiles identified in the sample with added rose bengal were identified in the sample with added ferrous chloride, however, tremendous differences of their relative ratios were found, which probably is useful to explain the flavor differences between sun-dried seafood and mechanical-dried seafood. Food colorants including rose bengal, erythrosine B, phloxine B and cochineal extracts were added to Alaska pollackTheragra chalcogrammasurimi which was previously added with methyl linoleate (MeLe) or ethyl eicosapentaenoate (EPA Et). All the surimi were exposed to a 100-watt light tungsten bulb (4500 lux) in a cold room at 5°C. Four isomeric hydroperoxides, including 13-cis,trans-, 13-trans,trans-, 9-trans,cis- and 9-trans,trans-MeLe hydroperoxides, were generated by oxidation of MeLe added to surimi with and without cochineal extracts after exposure to light for several hours. For the surimi containing rose bengal, erythrosine B and phloxine B, extracted lipids contained not only the above four hydroperoxide isomers but also 12-cis,trans- as well as 10-trans,cis-MeLe hydroperoxides. EPA Et added to surimi which contained rose bengal, erythrosine B and phloxine B generated characteristic isomeric hydroperoxides, including 6-cis,trans- and 17-cis,trans-EPA Et hydroperoxides due to1O2 mediated oxidation. The pro-oxidative potentials of four food colorants in surimi against added EPA Et or MeLe increased as the order of cochineal extracts < phloxine B < erythrosine B < rose bengal. These results clearly demonstrated that the addition of certain food colorants to surimi accelerated1O2 mediated lipid oxidation to result in the generation of characteristic hydroperoxide isomers. Hepatitis A virus (HAV) is mainly transmitted via contaminated food and water. HAV infection is a major global public health problem. Thus, developing a simple, rapid detection method is crucial for containing HAV epidemics, particularly in developing regions with limited laboratory resources. This study established a feasible HAV detection solution by combining reverse transcription multienzyme isothermal rapid amplification (RT-MIRA) and lateral flow dipstick (LFD) strips. Primers targeting the conserved 5'UTR sequence of HAV were used in the RT-MIRA-LFD assay. RNA extraction was enhanced by obtaining RNA directly from the centrifuged supernatant. Our study found that MIRA amplification could be finished in 12 min at 37℃ and naked-eye observation of the LFD strips in 10min. The detection sensitivity of this method reached 1 copy/μl. RT-MIRA-LFD was compared to conventional RT-PCR using 35 human blood samples. The accuracy of the RT-MIRA-LFD method was 100%. The convenience, sensitivity, and rapidness of this detection method could provide a considerable advantage for diagnosing and controlling HAV infection, especially in regions with limited medical resources. Photorespiration begins with the oxygenation reaction catalyzed by Rubisco and is the second highest metabolic flux in plants after photosynthesis. Although the core biochemical pathway of photorespiration has been well characterized, little is known about the underlying regulatory mechanisms. Some rate-limiting regulation of photorespiration has been suggested to occur at both the transcriptional and posttranslational levels, but experimental evidence is scarce. Here, we found that mitogen-activated protein kinase 2 (MAPK2) interacts with photorespiratory glycolate oxidase and hydroxypyruvate reductase, and the activities of these photorespiratory enzymes were regulated via phosphorylation modifications in rice (Oryza sativa L.). Gas exchange measurements revealed that the photorespiration rate decreased in rice mapk2 mutants under normal growth conditions, without disturbing photosynthesis. Due to decreased photorespiration, the levels of some key photorespiratory metabolites, such as 2-phosphoglycolate, glycine, and glycerate, significantly decreased in mapk2 mutants, but those of photosynthetic metabolites were not altered. Transcriptome assays also revealed that the expression levels of some flux-controlling genes in photorespiration were significantly downregulated in mapk2 mutants. Our findings provide molecular evidence for the association between MAPK2 and photorespiration and suggest that MAPK2 regulates the key enzymes of photorespiration at both the transcriptional and posttranslational phosphorylation levels in rice. Deep mixed oils with secondary alterations have been widely discovered in the Tarim Basin, but current methods based on biomarkers and isotopes to de-convolute mixed oil cannot calculate the exact mixing proportion of different end-member oils, which has seriously hindered further exploration of deep hydrocarbons in the study area. To solve this problem, we constructed a novel method based on the carbon isotope (δ13C) of the group components to de-convolute mixed liquid hydrocarbons under the material balance principle. The results showed that the mixed oil in the Tazhong Uplift was dominantly contributed at an average proportion of 68% by an oil end-member with heavier δ13C that was believed to be generated from the Cambrian-Lower Ordovician source rocks, whereas the mixed oil in the Tabei Uplift was predominantly contributed at an average proportion of 61% by an oil end-member with lighter d13C that was believed to be generated from the Middle-Upper Ordovician source rocks. This indicates that, on the basis of the detailed description of the distribution of effective source rocks, the proposed method will be helpful in realizing differential exploration and further improving the efficiency of deep liquid hydrocarbon exploration in the Tarim Basin. In addition, compared to traditional d13C methods for whole oil and individual n-alkanes in de-convoluted mixed oil, the proposed method has a wider range of applications, including for mixed oils with variations in color and density, indicating potential for promoting the exploration of deep complex mixed oils in the Tarim Basin and even around the world. Background: Camel milk has been reportedly used to treat dropsy, jaundice, tuberculosis, and diabetes while camel urine is used to treat diarrhea and cancer. However, there is no scientific evidence on the antiulcer activity of camel milk and urine. Thus, the present is designed to investigate the gastroprotective and ulcer healing effect of camel milk and urine on experimentally induced gastric ulcer models in rats. Materials and Methods: The gastroprotective effect was investigated in HCl/EtOH, water-restraint stress (WRS) and non-steroidal anti-inflammatory drugs (indomethacin)-induced ulcer models while ulcer healing activity was investigated in indomethacin-induced ulcer model. Cimetidine (100 mg/kg) was used as a standard antiulcer drug. Results: Acute toxicity study done up to a dosage of 10 ml/kg of camel milk and urine showed no signs of toxicity and mortality among the rats, indicating the present dosage of 5 ml/kg is safe to be administered to the rats. In the HCl/EtOH model, oral administration of cimetidine (100 mg/kg), camel urine (5 ml/kg), and camel milk (5 ml/kg) significantly (P < 0.05) inhibited gastric lesions by 83.7, 60.5 and 100%, respectively. In the WRS-induced model, cimetidine, and camel urine showed an ulcer inhibition of 100% while camel milk showed an inhibition of 50%. Similarly, in the indomethacin-induced ulcer model, cimetidine, camel milk, and urine showed an ulcer inhibition of 100, 33.3, and 66.7%, respectively. In addition, camel milk and urine also showed a significant (P < 0.05) ulcer healing effect of 100% in indomethacin-induced ulcer model, with no ulcers observed as compared to that of cimetidine, which offers a healing effect of 60.5%. Efficient depolymerization of C–O linkages is essential for converting lignin into fuels and higher value-added chemicals. In this work, CoRuP/SiO2, an amorphous Ru-Co phosphide composite, was fabricated for the efficient hydrogenolysis of ether linkages. The 4–O–5 and α–O–4 linkages containing lignin-related compounds, such as diphenyl ether, benzyl phenyl ether, 3-methyl diphenyl ether, and dibenzyl ether, are selected as representatives of linkages in lignin. Under mild conditions, Ru-containing metallic phosphides have high-performance for the catalytic depolymerization of C–O linkages. Compared with other catalysts, CoRuP/SiO2 shows an outstanding selectivity for benzene and excellent efficiency in depolymerizing diphenyl ethers, yielding only a small amount of by-products. Furthermore, the total acidity shows a linear relationship with the hydrogenolysis reactivity in cleaving aromatic ether bonds. The mechanisms for the catalytic hydrogenolysis of 4–O–5 and α–O–4 bonds over CoRuP/SiO2 are proposed. Moreover, two-dimensional heteronuclear single quantum coherence nuclear magnetic resonance spectroscopic analysis demonstrates that CoRuP/SiO2 could effectively depolymerize C–O bonds of lignin. These dominant hydrogenolysis products from lignin have excellent potential in the production of high value-added drugs or pharmaceutical intermediates. When comparing the success rates of two groups, statisticians often stratify the data into subgroups of individuals with similar levels of other factors related to the response of interest. In the clinical trial context, one might create subgroups according to the number and strength of risk factors the subjects have, while in the equal employment context, one might form strata based on the type of position and seniority. Sometimes the data are naturally stratified, e.g. applicants for different jobs requiring different skills. However, plaintiffs may pool all the data into one large 2 × 2 table, making it easier to find that the pass rates are statistically significantly different. In contrast, defendants may argue that only when statistical significance is reached in many, e.g. at least one-half of the strata, should a court find that the data supports a prima facie case of discrimination. These issues arose in Kerner v. Denver, a case concerning the disparate impact of a pre-employment exam on minority applicants. The statistical presentations of both parties are reviewed and potential issues with themare noted. After demonstrating that the disparities in pass rates in the various jobs are sufficiently similar, the Mantel-Haenszel test is used to combine the data in each stratum into one overall test. Our analysis shows that there is a statistically significant disparity in the odds minority and majority applicants pass the test. Furthermore, the associated estimator of the common odds ratio of the pass rates is 0.40 indicating that the odds a minority applicant had of passing the test were less than one-half those of a Caucasian. The receptor of the subgroup A avian leukosis virus (ALV-A) in chicken is Tva, which is the homologous protein of human CD320 (huCD320), contains a low-density lipoprotein (LDL-A) module and is involved in the uptake of transcobalamin bound vitamin B12/cobalamin (Cbl). To map the functional determinants of Tva responsible for ALV-A receptor activity, a series of chimeric receptors were created by swapping the LDL-A module fragments between huCD320 and Tva. These chimeric receptors were then used for virus entry and binding assays to map the minimal ALVA functional domain of Tva. The results showed that Tva residues 49 to 71 constituted the minimal functional domain that directly interacted with the ALV-A gp85 protein to mediate ALV-A entry. Single-residue substitution analysis revealed that L55 and W69, which were spatially adjacent on the surface of the Tva structure, were key residues that mediate ALV-A entry. Structural alignment results indicated that L55 and W69 substitutions did not affect the Tva protein structure but abolished the interaction force between Tva and gp85. Furthermore, substituting the corresponding residues of huCD320 with L55 and W69 of Tva converted huCD320 into a functional receptor of ALV-A. Importantly, soluble huCD320 harboring Tva L55 and W69 blocked ALV-A entry. Finally, we constructed a Tva gene-edited cell line with L55R and W69L substitutions that could fully resist ALV-A entry, while Cbl uptake was not affected. The photocatalytic generation of hydrogen (H2) from protons by two cyclometalated ruthenium-platinum polypyridyl complexes, [Ru(bpy)2(2,5-bpp)PtIS]2+ (1) and [Ru(dceb)2(2,5-bpp)PtIS]2+ (2) [where bpy = 2,2'-bipyridine, 2,5-bpp = 2,2',5',2"-terpyridine, dceb = 4,4'-di(carboxyethyl)bipyridine, and S = solvent], is reported. Turnover numbers (TONs) for H2 generation were increased by nearly an order of magnitude by the introduction of carboxyethyl ester units, i.e., from 80 for 1P to 650 for 2P after 6 h of irradiation, with an early turnover frequency (TOF) increasing from 15 to 200 h-1. The TON and TOF values for 2P are among the highest reported to date for supramolecular photocatalysts. Due to an error in the computer program used to summon potential jurors for service, the African-American percentage (4.17%) of jury pools was only one-half their percentage (8.25%) of the age-eligible population for about fifteen months. In June 2012, in Ambrose v. Booker, the Sixth Circuit accepted the statistical evidence as sufficient to meet the requirements for a prima facie case of unfair representation the U.S. Supreme Court established in Duren. On the same day, the Michigan S. Ct. in People v. Bryant said that the statistics were insufficient. The Michigan Court accepted a new measure, the disparity of the risk (DR), of under-representation. The DR measure is shown to be extremely stringent and an alternative measure based on the probability that a jury randomly selected from the jury pool will have fewer minorities than a jury selected from the age-eligible population is proposed. The minority fraction of individuals ultimately serving on juries also depends on the fairness of the peremptory challenges made by the parties. A method for detecting unfairness is reviewed; its effectiveness depends on the number of minorities on the venire. A reanalysis of the Michigan data shows that if the proportion of African-Americans in the jury pool equaled their proportion in the age-eligible population, prosecutors could only reduce their proportion on juries to about 80% of their proportion in the population. In contrast, the criteria for adequate representativeness based on the DR measure adopted by the Michigan Court could lead to virtually no minorities serving on actual juries as only three percent of the venires would have a sufficient number of minorities to classify a prosecutor's peremptory challenging all of them as significant. Our results indicate that when assessing statistics on the demographic mix of jury pools for legal significance, courts should consider the possible reduction in minority representation that can occur in the peremptory challenge proceedings. Background: Hepatocellular carcinoma (HCC) is a malignant tumor with complex pathogenesis. In HCC, the possible roles of transcriptional factor WD repeat and HMG-box DNA binding protein 1 (WDHD1) remain unclear. Hence, our study is aimed at verifying the prognosis prediction ability and potential biological mechanisms of WDHD1 in HCC. Results: In this study, a total of 7171 clinical samples were obtained to quantitatively analyze the protein and mRNA expression levels of WDHD1 by using immunohistochemistry, gene microarrays, and highthroughput sequencing technologies. The result of in-house immunohistochemistry assay indicated that WDHD1 protein was remarkably overexpressed in HCC tissues compared with the non-HCC tissues (AUC > 0.99, the single Standardized Mean Difference [SMD] = 4.46). The overexpression trend of WDHD1 was validated by the comprehensive analysis based on a total of 4004 HCC tissues and 3167 controls (SMD = 1.333; AUC = 0.91). Moreover, the higher WDHD1 expression resulted in the poorer prognosis of HCC, as assessed by overall survival and relapse-free survival analyses (pooled hazard ratios > 1). WDHD1-coexpressed genes were screened out for enrichment analyses to enquire the prospective signaling pathways of WDHD1 in HCC and to probe the potential transcriptional targets of WDHD1. The WDHD1-coexpressed genes were mainly involved in the division process of chromosome and cell nucleus in HCC. UBA52 was identified as a crucial target of WDHD1. Conclusions: WDHD1 may act as an oncogene in HCC and it has the potential to become a novel marker for predicting the prognosis of HCC patients, which may benefit from the early intervention of HCC.
