On 16 April 2022, China successfully launched the world's first spaceborne high-spectral-resolution lidar (HSRL), which is called the Aerosol and Carbon Detection Lidar (ACDL), on board the Atmospheric Environment Monitoring Satellite known as Daqi-1 (DQ-1). The ACDL is expected to precisely detect the three-dimensional distribution of aerosol and cloud globally with high spatial–temporal resolutions. To assess the performance of the newly launched satellite lidar, the ACDL-retrieved observations were compared with ground-based lidar measurements of atmospheric aerosol and cloud over northwest China from May to July 2022 using the Belt and Road lidar network (BR-lidarnet) initiated by Lanzhou University in China and the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observation (CALIPSO) lidar observations. A total of six cases in the daytime and nighttime, including clear days, dust events, and cloudy conditions, were selected for further analysis. Moreover, profiles of the total attenuated backscatter coefficient (TABC) and the volume depolarization ratio (VDR) at 532 nm measured by the ACDL, the CALIPSO lidar, and ground-based lidar are compared in detail. Comparison is made between the 532 nm extinction coefficient and lidar ratio obtained from ACDL HSRL retrieval and the Raman retrieval results obtained from BR-lidarnet. The achieved results revealed that the ACDL observations were in good agreement with the ground-based lidar measurements during dust events with a relative deviation of about −10.5 ± 25.4 % for the TABC and −6.0 ± 38.5 % for the VDR. Additionally, the heights of the cloud top and bottom from these two measurements were well matched and comparable. Compared with the observation of CALIPSO, the ACDL also shows high consistency. This study proves that the ACDL provides reliable observations of aerosol and cloud in the presence of various climatic conditions, which helps to further evaluate the impacts of aerosol on climate and the environment, as well as on the ecosystem in the future.

The effective design of metallic metamaterials, characterized by interconnected struts or 'lattices,' hinges on the ability to predict strut and strut intersection ('node') responses. This is critical for predicting the macroscopic properties of structures comprised of thousands of struts. Computationally efficient beam descriptions, defined by strut properties like cross-sectional area, modulus, and yield stress, can significantly expedite the prediction of lattice structures and ultimately enable topology optimization. This paper provides a comprehensive examination of the properties of electron-beam melted three-dimensional printed struts and their 'nodes'—four intersecting struts. The findings elucidate the efficacy of various strategies for defining effective properties that accurately capture mechanical response. The study reveals that using a single set of effective properties can introduce inconsistencies between strut stiffness, peak load, and critical displacement. Stiffness correlates with averaged cross-sectional areas, while the peak load capacity correlates more closely with minimum inscribed cross-sectional areas. Analysis of nodes indicates that the interaction of surface defects and heterogeneity within the node strongly influences multi-strut response. Data from CT scans, EBSD scans, and nanoindentation maps highlight spatial variations comparable to the strut diameter, posing a significant challenge in defining effective homogenized properties. This study emphasizes the need for future efforts to integrate statistical property distributions with high throughput simulations to overcome the difficulty in defining a representative volume element (RVE) at the strut scale.

The first source catalog of the Large High Altitude Air Shower Observatory (LHAASO) reported the detection of a very high energy gamma-ray source, 1LHAASO J1219+2915. This Letter presents a further detailed study of the spectral and temporal behavior of this pointlike source. The best-fit position of the TeV source (R.A. = 185.°05 ± 0.°04, decl. = 29.°25 ± 0.°03) is compatible with NGC 4278 within ∼0.°03. Variation analysis shows an indication of variability on a timescale of a few months in the TeV band, which is consistent with low-frequency observations. Based on these observations, we report the detection of TeV γ -ray emissions from this low-luminosity active galactic nucleus. The observation by LHAASO's Water Cherenkov Detector Array during the active period has a significance level of 8.8 σ with a best-fit photon spectral index Γ = 2.56 ± 0.14 and a flux f _1–10 TeV = (7.0 ± 1.1 _sta ± 0.35 _syst ) × 10 ^−13 photons cm ^−2 s ^−1 , or approximately 5% of the Crab Nebula. The discovery of VHE gamma-ray emission from NGC 4278 indicates that compact, weak radio jets can efficiently accelerate particles and emit TeV photons.

We present the first catalog of very-high-energy and ultra-high-energy gamma-ray sources detected by the Large High Altitude Air Shower Observatory. The catalog was compiled using 508 days of data collected by the Water Cherenkov Detector Array from 2021 March to 2022 September and 933 days of data recorded by the Kilometer Squared Array from 2020 January to 2022 September. This catalog represents the main result from the most sensitive large coverage gamma-ray survey of the sky above 1 TeV, covering decl. from −20° to 80°. In total, the catalog contains 90 sources with an extended size smaller than 2° and a significance of detection at >5 σ . Based on our source association criteria, 32 new TeV sources are proposed in this study. Among the 90 sources, 43 sources are detected with ultra-high energy ( E > 100 TeV) emission at >4 σ significance level. We provide the position, extension, and spectral characteristics of all the sources in this catalog.

6. QUARTERLY MONITORING OF SUSPECTED ILLEGAL HUMAN ACTIVITIES IN NATIONAL NATURE RESERVES OF CHINA WITH SENTINEL-2 AND HIGH-RESOLUTION IMAGES ON THE CLOUD PLATFORM

Nature reserves play an important role in protecting biodiversity, preserving natural heritage, and maintaining national ecological security. Satellite remote sensing technology has become an essential way to capture the impact of human activities on nature reserves by monitoring the land cover change. However, current monitoring frequency and scale were inadequate to detect the land change related to illegal human activities rapidly and accurately. To solve the above problems, this study proposed an operational suspected illegal change detection method which combined the temporal advantage of Sentinel-2 double satellites with the spatial advantage of high-resolution images from SiweiEarth and Google Earth cloud platforms, together with high-precision high-resolution images from cloud platforms, historical land cover data from Geoinformation Survey Data of China, and the thematic information of study area. Eight typical national nature reserves were selected as study areas. January 1st, 2019 was taken as the time point of the base period. May 31st, August 31st, and November 30th were taken as the time point of the first period, the second period, and the third period respectively. Results showed that four national nature reserves had nine suspected illegal land change parcels, with a good ecological protection situation of the other four nature reserves. Deforestation and illegal construction were the two main suspected illegal types. In the next step, the study results will be provided to the Law Enforcement Administration of China to evaluate the applicability and generalization of this method, and a more rapid and intelligent method needs to be improved.

Urban ecological space is a significant factor for sustainable urban development and ecological civilization construction. Traditional urban ecological space analysis mainly used medium-resolution image data at large scales and used high-resolution images at a typical urban scale. Few studies focused on the high-precision ecological space analysis at a national urban scale. In this study, high-precision urban boundary and land cover data were utilized to analyze the urban ecological space change and its reason from 2016 to 2017. Ecological space was extracted and merged from high-precision land cover data, which came from the National Geoinformation Survey data of China. Results showed that in 2017, the total urban ecological space area of 338 prefecture-level cities in China was 8514.2 square kilometers, accounting for 22.4% of the total urban area, which was far below the threshold of 40% for evaluating the urban green coverage in China. Urban ecological space of 184 cities declined. There were four principal reasons for the decrease of urban ecological space. First, the green space supporting the buildings was occupied. Second, the green landscape space near rivers and lakes was occupied. Third, blocks of woodland were occupied. Fourth, the water area was occupied. The reduced urban ecological space was mainly changed into construction sites, structures, and buildings. Urban ecological space of China was seriously insufficient, and it was heavily occupied. Adequate ecological space should be preserved not only in new urban development areas but also in the old urban areas to ensure people a more comfortable living environment.

The virtual restoration was one of the important protection approaches for the Great Wall. With the help of this type of technology, the damaged Great Wall can be restored to the previous state of the Great Wall at a specific time node so that the tourists can visit the Great Wall before the damaged in the computer. This this paper presented a virtual restoration framework for the “Moon Gate” located on the XinGuangWu Great Wall so that virtual recovered results can guide the researcher to repair the Great Wall in the real world without the secondary damage. This method can be divided into 4 parts: (1) collection of evidence based on the modified scale of evidence; (2) the fusion of DIM point clouds; (3) excavation of the reconstruction technology from the collected evidence; (4) the generation of 3D model. The experimental results showed that the maximum error of the reconstructed “Moon Gate” was 1.088 cm without considering the brick seam and our proposed virtual restoration framework of the Great Wall was robust and accuracy, and it has certain reference significance for guiding the restoration of the damaged Great Wall.

A simultaneous high-resolution x-ray backlighting and self-emission imaging method for laser-produced plasma diagnostics is developed in which two Kirkpatrick–Baez imaging channels for high-energy and low-energy diagnostics are constructed using a combination of multilayer mirrors in near-coaxial form. By using a streak or framing camera placed on the image plane, both backlit and self-emission images of a laser-produced plasma with high spatial and temporal resolution can be obtained simultaneously in a single shot. This paper describes the details of the method with regard to its optical and multilayer design, assembly, and alignment method. In addition, x-ray imaging results with a spatial resolution better than 5 µm in the laboratory and experimental results with imploding capsules in the SG-III prototype laser facility are presented.

Mutations in the scaffolding domain of Receptor Interacting Protein kinases (RIP) underlie the recently described human autoimmune syndrome, CRIA, characterized by lymphadenopathy, splenomegaly, and autoantibody production. While disease mechanisms for CRIA remain undescribed, RIP kinases work together with caspase-8 to regulate cell death, which is critical for normal differentiation of many cell types. Here, we describe a key role for RIP1 in facilitating innate B cell differentiation and subsequent activation. By comparing RIP1, RIP3, and caspase-8 triple deficient and RIP3, caspase-8 double deficient mice, we identified selective contributions of RIP1 to an accumulation of murine splenic Marginal Zone (MZ) B cells and B1-b cells. We used mixed bone-marrow chimeras to determine that innate B cell commitment required B cell-intrinsic RIP1, RIP3, and caspase-8 sufficiency. RIP1 regulated MZ B cell development rather than differentiation and RIP1 mediates its innate immune effects independent of the RIP1 kinase domain. NP-KLH/alum and NP-Ficoll vaccination of mice doubly deficient in both caspase-8 and RIP3 or deficient in all three proteins (RIP3, caspase-8, and RIP1) revealed uniquely delayed T-dependent and T-independent IgG responses, abnormal splenic germinal center architecture, and reduced extrafollicular plasmablast formation compared to WT mice. Thus, RIP kinases and caspase-8 jointly orchestrate B cell fate and delayed effector function through a B cell-intrinsic mechanism.

Prevention of bacterial infection and reduction of hemorrhage, the primary challenges posed by trauma before hospitalization, are essential steps in prolonging the patient's life until they have been transported to a trauma center. Extracellular matrix (ECM) hydrogel is a promising biocompatible material for accelerating wound closure. However, due to the lack of antibacterial properties, this hydrogel is difficult to be applied to acute contaminated wounds. This study formulates an injectable dermal extracellular matrix hydrogel (porcine acellular dermal matrix (ADM)) as a scaffold for skin defect repair. The hydrogel combines vancomycin, an antimicrobial agent for inducing hemostasis, expediting antimicrobial activity, and promoting tissue repair. The hydrogel possesses a porous structure beneficial for the adsorption of vancomycin. The antimicrobial agent can be timely released from the hydrogel within an hour, which is less than the time taken by bacteria to infest an injury, with a cumulative release rate of approximately 80%, and thus enables a relatively fast bactericidal effect. The cytotoxicity investigation demonstrates the biocompatibility of the ADM hydrogel. Dynamic coagulation experiments reveal accelerated blood coagulation by the hydrogel. In vivo antibacterial and hemostatic experiments on a rat model indicate the healing of infected tissue and effective control of hemorrhaging by the hydrogel. Therefore, the vancomycin-loaded ADM hydrogel will be a viable biomaterial for controlling hemorrhage and preventing bacterial infections in trauma patients.

17. Aerosol pH and its driving factors in Beijing

Aerosol acidity plays a key role in secondary aerosol formation. The high-temporal-resolution PM2.5 pH and size-resolved aerosol pH in Beijing were calculated with ISORROPIA II. In 2016–2017, the mean PM2.5 pH (at relative humidity (RH) > 30 %) over four seasons was 4.5±0.7 (winter) > 4.4±1.2 (spring) > 4.3±0.8 (autumn) > 3.8±1.2 (summer), showing moderate acidity. In coarse-mode aerosols, Ca2+ played an important role in aerosol pH. Under heavily polluted conditions, more secondary ions accumulated in the coarse mode, leading to the acidity of the coarse-mode aerosols shifting from neutral to weakly acidic. Sensitivity tests also demonstrated the significant contribution of crustal ions to PM2.5 pH. In the North China Plain (NCP), the common driving factors affecting PM2.5 pH variation in all four seasons were SO42-, TNH3 (total ammonium (gas + aerosol)), and temperature, while unique factors were Ca2+ in spring and RH in summer. The decreasing SO42- and increasing NO3- mass fractions in PM2.5 as well as excessive NH3 in the atmosphere in the NCP in recent years are the reasons why aerosol acidity in China is lower than that in Europe and the United States. The nonlinear relationship between PM2.5 pH and TNH3 indicated that although NH3 in the NCP was abundant, the PM2.5 pH was still acidic because of the thermodynamic equilibrium between NH4+ and NH3. To reduce nitrate by controlling ammonia, the amount of ammonia must be greatly reduced below excessive quantities.

Considering the critical role of trajectory data in Big Data era for dynamic geographical processes, human behaviour analysis and meteorological prediction, trajectory clustering has attracted growing attention. Many literatures have discussed the spatiot emporal clustering method of simple trajectories (i.e., has no branches, e.g. vehicle trajectories), yet there are few researches for clustering complex trajectories (i.e., has at least one split and/or merger and/or split -merger branch, e.g. ocean eddy trajectories, rainstorm trajectories). For addressing this issue, we propose a Process-Oriented Spatiotemporal Clustering Method (POSCM) for clustering complex trajectory data. The POSCM includes three parts: the first uses the semantic of process-sequence-state to represent the complex trajectories; the second proposes a Hierarchical Similarity Measurement Method (HSMM) to get the similarity between any two complex trajectories; in the last step, the complex trajectories clustering pattern is extracted through density-based clustering algorithm. Experiments on simulated trajectories are used to evaluate the POSCM and demonstrate the advantage by comparing against that of the VF2 algorithm. The POSCM is applied to the sea surface temperature abnormal variations trajectories from January 1950 to December 2017 in the Pacific Ocean. As shown in this case study, some new mined spatiotemporal patterns can provide new references for understanding the behaviours of marine abnormal variations under the background of the global change.

Background: The mitochondrial associated endoplasmic reticulum (ER) membrane (MAM) provides a platform for communication between the mitochondria and ER, and it plays a vital role in many biological functions. Disulphide-bond A oxidoreductase-like protein (DsbA-L), expressed in the MAM, serves as an antioxidant and reduces ER stress. However, the role of DsbA-L and MAM in kidney pathobiology remains unclear. Methods: Molecular biology techniques, transmission electron microscopy (TEM), in situ proximity ligation assays (PLAs), confocal microscopy, TUNEL staining and flow cytometry were utilized to analyse apoptosis and status of MAM in DsbA-L mutant mice. Findings: We showed that MAM was significantly reduced in the kidneys of streptozotocin-induced diabetic mice, which correlated with the extent of renal injury. We also observed a correlation between the loss of MAM integrity and increased apoptosis and renal injury in diabetic nephropathy (DN). These alterations were further exacerbated in diabetic DsbA-L gene-deficient mice (DsbA-L−/−). In vitro, overexpression of DsbA-L in HK-2 cells restored MAM integrity and reduced apoptosis induced by high-glucose ambience. These beneficial effects were partially blocked by overexpression of FATE-1, a MAM uncoupling protein. Finally, the expression of DsbA-L was positively correlated with MAM integrity in the kidneys of DN patients but negatively correlated with apoptosis and renal injury. Interpretation: Our results indicate that DsbA-L exerts an antiapoptotic effect by maintaining MAM integrity, which is apparently disrupted in DN. To identify factors that affect the likelihood of follow-up after emergency department (ED) visit for ophthalmic complaints and to evaluate a protocol to improve compliance.Prospective interventional study with historical controls.This study was conducted at Jamaica Hospital Medical Center in Jamaica, New York. The study population included 962 patients who presented to the ED and who required ophthalmology consultation. Participants in the control group were given only verbal follow-up instructions. Participants in the intervention group were given verbal instructions, written instructions, telephone calls, and, if not responding to calls, a mailed letter. The primary outcome was the overall follow-up rate. Secondary outcomes were follow-up rate by demographic subgroup.Patients in the intervention group were significantly more likely to follow up (68.8% vs 42.9%, P.001). Nearly all subgroups exhibited significantly improved follow-up with the intervention, with the exception of patients 18 to 29 years of age, patients with diagnosis severity class III, patients with no insurance, patients with hospital financial aid, patients paying with workers' compensation, and patients with an unknown employment status.Before the intervention, most patients receiving ophthalmology consultation in the ED did not return for follow-up care. These patients tended to be young, unemployed, uninsured or use hospital financial aid, were in the control group, had good visual acuity, reported no change in vision, and had a condition that was not vision-threatening. Follow-up rates were improved in nearly all subgroups by providing written instructions, telephone calls, and mailed letters. Such instructions should be considered in similar populations.

Currently, there is no reproducible, widely accepted gold standard to classify osteoporotic vertebral body fractures (OVFs). The purpose of this study is to refine a method with clear rules to classify OVFs for machine learning purposes. The method was found to have moderate interobserver agreement that improved with training.The current methods to classify osteoporotic vertebral body fractures are considered ambiguous; there is no reproducible, accepted gold standard. The purpose of this study is to refine classification methodology by introducing clear, unambiguous rules and a refined flowchart to allow consistent classification of osteoporotic vertebral body fractures.We developed a set of rules and refinements that we called m2ABQ to classify vertebrae into five categories. A fracture-enriched database of thoracic and lumbar spine radiographs of patients 65 years of age and older was retrospectively obtained from clinical institutional radiology records using natural language processing. Five raters independently classified each vertebral body using the m2ABQ system. After each annotation round, consensus sessions that included all raters were held to discuss and finalize a consensus annotation for each vertebral body where individual raters' evaluations differed. This process led to further refinement and development of the rules.Each annotation round showed increase in Fleiss kappa both for presence vs absence of fracture 0.62 (0.56-0.68) to 0.70 (0.65-0.75), as well as for the whole m2ABQ scale 0.29 (0.25-0.33) to 0.54 (0.51-0.58).The m2ABQ system demonstrates moderate interobserver agreement and practical feasibility for classifying osteoporotic vertebral body fractures. Future studies to compare the method to existing studies are warranted, as well as further development of its use in machine learning purposes.

Although many filter algorithms have been presented over past decades, these algorithms are usually designed for the Lidar point clouds and can’t separate the ground points from the DIM (dense image matching, DIM) point clouds derived from the oblique aerial images owing to the high density and variation of the DIM point clouds completely. To solve this problem, a new automatic filter algorithm is developed on the basis of adaptive TIN models. At first, the differences between Lidar and DIM point clouds which influence the filtering results are analysed in this paper. To avoid the influences of the plants which can’t be penetrated by the DIM point clouds in the searching seed pointes process, the algorithm makes use of the facades of buildings to get ground points located on the roads as seed points and construct the initial TIN. Then a new densification strategy is applied to deal with the problem that the densification thresholds do not change as described in other methods in each iterative process. Finally, we use the DIM point clouds located in Potsdam produced by Photo-Scan to evaluate the method proposed in this paper. The experiment results show that the method proposed in this paper can not only separate the ground points from the DIM point clouds completely but also obtain the better filter results compared with TerraSolid. 1.

For a multi-vectored propeller aerostat with actuator faults, this study presents a fault-tolerant tracking control strategy, which includes fault modeling, observer, force estimation and tracking controller. Fault modeling considers the four types of faults of vectored propellers, namely, thrust offset, thrust efficiency loss, vectored angle offset and vectored angle stuck. Actuator faults can be determined from the fault observer, which identifies the thrust offset from the acceleration difference of the faulty aerostat with the ideal model. For tracking positions, a traditional PID controller is constructed with virtual control, compensated with the estimated fault force. The control allocation scheme is proposed to redistribute the available actuators in case faults occur. Simulation results of position tracking prove the effectiveness of the proposed strategy.

Objective This study aimed to investigate the clinical characteristics and long-term prognosis of mycoplasma pneumoniae pneumonia (MPP)-associated thrombosis and to gain a better understanding of the diagnosis and treatment of the disease. Methods The medical records of 14 children with MPP-associated thrombosis between January 2016 and April 2020 were retrospectively reviewed at the Tianjin Children’s Hospital. Results The ages of the patients ranged from 3 to 12 years old. Among the 14 cases, there were five cases of pulmonary embolism, two cases of cerebral infarction, one case of splenic infarction, one case of cardiac embolism, two cases of cardiac embolism with comorbid pulmonary embolism, one case of internal carotid artery and pulmonary embolism, one case of combined internal carotid artery and the cerebral infarction, and one case combined cardiac embolism and lower limb artery embolism. All cases had elevated D-dimer levels. After thrombolysis and anticoagulation therapy, three cases with cerebral embolism still suffered from neurological sequelae. In contrast, the remaining cases did not develop complications. Conclusion MPP-associated thrombosis can occur in any vessel of the body. Thrombosis-associated symptoms may be complex and non-specific. Elevated D-dimer levels in a child with refractory mycoplasma pneumoniae pneumonia should raise suspicion of thrombosis. The long-term prognosis of thrombosis was favorable after the timely administration of anticoagulant therapy.

Chronic refractory wounds and scars caused by abnormal wound repair seriously damage the health of patients and affect their quality of life. At present, there is a lack of simple but effective and economical treatment methods. Adipose-derived stem cells (ASCs), as a kind of mesenchymal stem cells with multi-directional differentiation potential, have been confirmed by several in vivo and in vitro studies to promote wound healing by promoting epithelialization, angiogenesis, immunoregulation, antioxidant properties, and other mechanisms. ASCs and their derivatives have been used in the treatment of refractory wounds caused by burns, diabetic, and radiation injuries with good results achieved. Their potential to become new materials for wound repair has also been confirmed. This paper reviewed the mechanism and clinical application of ASCs in promoting wound repair, and looked into its research direction and prospects.创面修复异常所导致的慢性难愈性创面及瘢痕严重损害了患者身体健康,影响了患者生活质量,且目前缺乏简单有效、经济的治疗手段。脂肪干细胞(ASC)作为一种拥有多向分化潜能的间充质干细胞,已经被多项体内、体外研究证实可以通过促上皮化、促血管生成、免疫调节和抗氧化等机制促进创面愈合。ASC及其衍生物已被用于治疗由烧伤、糖尿病和放射性损伤等导致的难愈性创面并取得良好效果,且其成为创面修复新材料的潜力也得到了证实。该文就ASC促进创面修复的机制和临床应用进行综述,并对其研究方向与前景进行展望。.

A 54-month-old female patient presented to the department of ophthalmology with abnormal head posture and facial asymmetry for two years. The patient's facial development was asymmetrical, with the middle 1/3 of the left side shorter than the right side. The left ear is less malformed than the right. There was no obvious abnormality in corneal light reflex and eye movement. Head tilt test ( -). So, paralysis of the superior oblique muscle was excluded. In consultation with the department of maxillofacial surgery, the patient was confirmed as the first and second branchial arch syndrome and torticollis.患儿女性,4岁6个月,因发现视物时头向左歪、面部发育不对称2年余就诊于眼科。患者面部发育不对称,左侧面中1/3高度较右侧短小,左耳畸形,较右耳偏小,眼位正,眼球运动未见明显异常,歪头试验阴性,排除上斜肌麻痹。经颌面外科会诊后明确诊断为鳃弓综合征、侧视症。.

Coronavirus disease 2019 symptom definitions rarely include symptom severity. We collected daily nasal swab samples and symptom diaries from contacts of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) case patients. Requiring ≥1 moderate or severe symptom reduced sensitivity to predict SARS-CoV-2 shedding from 60.0% (95% confidence interval [CI], 52.9%−66.7%) to 31.5% (95% CI, 25.7%− 38.0%) but increased specificity from 77.5% (95% CI, 75.3%−79.5%) to 93.8% (95% CI, 92.7%−94.8%).

Bintrafusp alfa, a first-in-class bifunctional fusion protein composed of the extracellular domain of TGF-βRII (a TGF-β "trap") fused to a human IgG1 mAb blocking PD-L1, is being evaluated for efficacy and safety in solid tumor indications as monotherapy and in combination with small-molecule drugs. We evaluated the perpetrator drug-drug interaction (DDI) potential of bintrafusp alfa via cytochrome P4503A4 (CYP3A4) enzyme modulation, which is responsible for the metabolism of a majority of drugs. The holistic approach included (1) evaluation of longitudinal profiles of cytokines implicated in CYP3A4 modulation and serum 4β-hydroxycholesterol, an endogenous marker of CYP3A4 activity, in a phase I clinical study, and (2) transcriptomics analysis of the CYP3A4 mRNA levels vs the TGFB gene expression signature in normal hepatic tissues. Bintrafusp alfa was confirmed not to cause relevant proinflammatory cytokine modulation or alterations in 4β-hydroxycholesterol serum concentrations in phase I studies. Transcriptomics analyses revealed no meaningful correlations between TGFB gene expression and CYP3A4 mRNA expression, supporting the conclusion that the risk of CYP3A4 enzyme modulation due to TGF-β neutralization by bintrafusp alfa is low. Thus, bintrafusp alfa is not expected to have DDI potential as a perpetrator with co-administered drugs metabolized by CYP3A4; this information is relevant to clinical evaluations of bintrafusp alfa in combination settings.

Coal is one of the major fuels, which brings huge energy and economic benefits to global industry and daily life. large amounts of coal dust produced in the process of coal mining and transportation, which seriously threatens the health of related workers. Productive coal dust exposure not only directly leads to respiratory diseases, but also may cause health damage to various systems throughout the body. Childhood obesity and metabolic syndrome sequelae associated with early-onset insulin resistance and type 2 diabetes are at epidemic levels globally with increasing evidence for worrisome young adult health sequelae trajectories. Pediatric obesity is often associated with dramatic rise in hyperinsulinemia and acanthosis during puberty, a risk for progression to type 2 diabetes and/or hepatic steatosis. Recently, we reported leucine-rich alpha-2-glycoprotein 1 (LRG1) as an adipokine, which levels in sera are positively associated with BMI in humans. LRG1 binds with multiple metabolic organs in vivo, including the liver and pancreas. Deletion of the Lrg1 gene in mice alleviates high-fat diet (HFD) -induced hyperinsulinemia, hepatosteatosis, insulin resistance, and obesity at a young age. Our current study indicates that treating the MIN6 beta-cells with LRG1 was sufficient to trigger insulin secretion both in the absence and presence of glucose in a dose-dependent manner, specifically enhancing the first phase of insulin secretion. We also identified an LRG1 monoclonal antibody that neutralizes LRG1-related stimulation of insulin secretion in MIN6 beta-cells. Serum levels of LRG1 in young mice fed with HFD led to enhanced levels of LRG1 in sera in these mice, which preceded hyperinsulinemia and subsequent increased weight gain. Together, our results reveal that up-regulated LRG1 in circulation could be a causal factor for persistent hyperinsulinemia by directly targeting pancreatic beta-cells to enhance insulin secretion in childhood obesity and a likely biomarker for early childhood hyperinsulinemia and treatment assessments. Binding of insulin receptor substrate proteins 1 and 2 (IRS1/2) to the insulin receptor (IR) is essential for the regulation of insulin sensitivity and energy homeostasis. However, the mechanism of IRS1/2 recruitment to the IR remains elusive. Here, we identify adaptor protein APPL1 as a critical molecule that promotes IRS1/2-IR interaction. APPL1 forms a complex with IRS1/2 under basal conditions, and this complex is then recruited to the IR in response to insulin or adiponectin stimulation. The interaction between APPL1 and IR depends on insulin- or adiponectin-stimulated APPL1 phosphorylation, which is greatly reduced in insulin target tissues in obese mice. appl1 deletion in mice consistently leads to systemic insulin resistance and a significant reduction in insulin-stimulated IRS1/2, but not IR, tyrosine phosphorylation, indicating that APPL1 sensitizes insulin signaling by acting at a site downstream of the IR. Our study uncovers a mechanism regulating insulin signaling and crosstalk between the insulin and adiponectin pathways.

45. On the consistency of MODIS chlorophyll $a$ products in the northern South China Sea

Chlorophyll a (Chl) concentrations derived from satellite measurements have been used in oceanographic research, for example to interpret eco-responses to environmental changes on global and regional scales. However, it is unclear how existing Chl products compare with each other in terms of accuracy and consistency in revealing temporal and spatial patterns, especially in the optically complex marginal seas. In this study, we examined three MODIS (Moderate Resolution Imaging Spectroradiometer) Chl data products that have been made available to the community by the US National Aeronautics and Space Administration (NASA) using community-accepted algorithms and default parameterization. These included the products derived from the OC3M (ocean chlorophyll three-band algorithm for MODIS), GSM (Garver–Siegel–Maritorena model) and GIOP (generalized inherent optical properties) algorithms. We compared their temporal variations and spatial distributions in the northern South China Sea. We found that the three products appeared to capture general features such as unique winter peaks at the Southeast Asian Time-series Study station (SEATS, 18° N, 116° E) and the Pearl River plume associated blooms in summer. Their absolute magnitudes, however, may be questionable in the coastal zones. Additional error statistics using field measured Chl as the truth demonstrated that the three MODIS Chl products may contain high degree of uncertainties in the study region. Root mean square error (RMSE) of the products from OC3M and GSM (on a log scale) was about 0.4 and average percentage error (ε) was ~ 115% (Chl between 0.05–10.41 mg m−3, n = 114). GIOP with default parameterization led to higher errors (ε = 329%). An attempt to tune the algorithms based on a local coastal-water bio-optical data set led to reduced errors for Chl retrievals, indicating the importance of local tuning of globally-optimized algorithms. Overall, this study points to the need of continuous improvements for algorithm development and parameterization for the coastal zones of the study region, where quantitative interpretation of the current Chl products requires extra caution.

Adiponectin is an adipokine that exerts insulin-sensitizing and anti-inflammatory roles in insulin target tissues including liver. While the insulin-sensitizing function of adiponectin has been extensively investigated, the precise mechanism by which adiponectin alleviates diet-induced hepatic inflammation remains elusive. Here, we report that hepatocyte-specific knockout (KO) of the adaptor protein APPL2 enhanced adiponectin sensitivity and prevented mice from developing high-fat diet–induced inflammation, insulin resistance, and glucose intolerance, although it caused fatty liver. The improved anti-inflammatory and insulin-sensitizing effects in the APPL2 hepatocyte–specific KO mice were largely reversed by knocking out adiponectin. Mechanistically, hepatocyte APPL2 deficiency enhances adiponectin signaling in the liver, which blocks TNF-α–stimulated MCP-1 expression via inhibiting the mTORC1 signaling pathway, leading to reduced macrophage infiltration and thus reduced inflammation in the liver. With results taken together, our study uncovers a mechanism underlying the anti-inflammatory role of adiponectin in the liver and reveals the hepatic APPL2–mTORC1–MCP-1 axis as a potential target for treating overnutrition-induced inflammation in the liver.

Adiponectin is an adipokine with anti-insulin resistance and anti-inflammatory functions. It exists in serum predominantly in three multimeric complexes: the trimer, hexamer, and high-molecular-weight forms. Although recent studies indicate that adiponectin promotes wound healing in rodents, its role in the wound healing process in humans is unknown. This study investigated the expression levels of adiponectin in adipose tissue and serum of women who experienced either normal or delayed wound healing after abdominal plastic surgery. We found that obese women with delayed healing had slightly lower total adiponectin levels in their adipose tissue compared with women with normal healing rates. Among the different isoforms of adiponectin, levels of the trimer forms were significantly reduced in adipose tissue, but not the serum, of obese women with delayed healing compared to women who healed normally. This study provides clinical evidence for a potential role of low-molecular-weight oligomers of adiponectin in the wound healing process as well as implications for an autocrine and/or paracrine mechanism of adiponectin action in adipose tissues.

48. Molecular dynamics simulation of the gun barrel material erosion by propellant gas

The mechanisms of propellant gas chemical erosion on gun barrel materials are discussed creatively from a microscopic perspective of molecular diffusion and intermolecular interaction. The influence of high-temperature and high-pressure propellant gas atoms on the shallow surface microstructure of the barrel material is theoretically analyzed using the molecular dynamics simulation method, which can be used as a new idea for further study of the gun barrel erosion mechanism. The results show that the diffusion of the propellant gas atoms into the barrel material can be characterized by the significant longitudinal distribution characteristics of propellant gas atoms and the normal stress distribution characteristics in the surface layer of the barrel. Moreover, the diffusion ability of the propellant gas atoms in the surface of the barrel material decreases gradually with the increasing diffusion depth. Under the action of propellant gas atoms, the dislocation structure dominated by face-centered cubic structure (FCC) forms in the shallow surface of the barrel material. The production of initial erosion in the shallow surface of barrel materials can be predicted by the continual growth of the new structure and the stress distribution characteristics of the material. In addition, the temperature variation of the propellant gas contributes significantly to barrel material erosion.

In this paper, α-olefins with different chain lengths are used as monomers, and metallocene catalysts were used to prepare low-viscosity metallocene-catalysed poly-α-olefin (mPAO) lubricating base oils. Structure and performance tests were carried out. Three molecular structure parameters were selected: relative molecular weight (Mn), branch carbon number (L), and number of branches (n). By mixing and compounding each component oligomer and performing data analysis, the relationships between the molecular structure of mPAO and the key physical properties of the base oil, such as viscosity, viscosity index and pour point (PP), were studied. The results show that in a certain range of Mn, when L (or n) is constant, with an increase in Mn, the viscosity (Kv) and Mn show a power function growth trend, and the growth rate of Kv40°C is greater than that of Kv100°C. The viscosity index (VI) increases with increasing Mn, a greater n results in a more obvious growth trend, and a greater L results in a weaker growth trend. When L is constant, an increase in n results in an initial decrease followed by an increase in PP, and PP is the lowest at n = 2.

Background and purpose Adiponectin (APN) is an adipokine secreted from adipocytes that binds to APN receptors AdipoR1 and AdipoR2 and exerts an anti-inflammatory response through mechanisms not fully understood. There is a need to develop small molecules that activate AdipoR1 and AdipoR2 and to be used to inhibit the inflammatory response in lipopolysaccharide (LPS)-induced endotoxemia and other inflammatory disorders. Experimental approach We designed 10 new structural analogues of an AdipoR agonist, AdipoRon (APR), and assessed their anti-inflammatory properties. Bone marrow-derived macrophages (BMMs) and peritoneal macrophages (PEMs) were isolated from mice. Levels of pro-inflammatory cytokines were measured by reverse transcription and real-time quantitative polymerase chain reaction (qRT-PCR), enzyme-linked immunosorbent assay (ELISA) and microarray in LPS-induced endotoxemia mice and diet-induced obesity (DIO) mice in which systemic inflammation prevails. Western blotting, immunohistochemistry (IHC), siRNA interference and immunoprecipitation were used to detect signalling pathways. Key results A novel APN receptor agonist named adipo anti-inflammation agonist (AdipoAI) strongly suppresses inflammation in DIO and endotoxemia mice, as well as in cultured macrophages. We also found that AdipoAI attenuated the association of AdipoR1 and APPL1 via myeloid differentiation marker 88 (MyD88) signalling, thus inhibiting activation of nuclear factor kappa B (NF-κB), mitogen-activated protein kinase (MAPK) and c-Maf pathways and limiting the production of pro-inflammatory cytokines in LPS-induced macrophages. Conclusion and implications AdipoAI is a promising alternative therapeutic approach to APN and APR to suppress inflammation in LPS-induced endotoxemia and other inflammatory disorders via distinct signalling pathways.
