The intrinsic redox status of cancer cells limits the efficacy of chemotherapeutic drugs. Auranofin, a Food and Drug Administration‐approved gold‐containing compound, documented with effective pharmacokinetics and safety profiles in humans, has recently been repurposed for anticancer activity. This study examined the paclitaxel‐sensitizing effect of auranofin by targeting redox balance in the MDA‐MB‐231 and MCF‐7 breast cancer cell lines. Auranofin treatment depletes the activities of superoxide dismutase, catalase, and glutathione peroxidase and alters the redox ratio in the breast cancer cell lines. Furthermore, it has been noticed that auranofin augmented paclitaxel‐mediated cytotoxicity in a concentration‐dependent manner in both MDA‐MB‐231 and MCF‐7 cell lines. Moreover, auranofin increased the levels of intracellular reactive oxygen species (observed using 2, 7‐diacetyl dichlorofluorescein diacetate staining) and subsequently altered the mitochondrial membrane potential (rhodamine‐123 staining) in a concentration‐dependent manner. Further, the expression of apoptotic marker p21 was found to be higher in auranofin plus paclitaxel‐treated breast cancer cells compared to paclitaxel‐alone treatment. Thus, the present results illustrate the chemosensitizing property of auranofin in MDA‐MB‐231 and MCF‐7 breast cancer cell lines via oxidative metabolism. Therefore, auranofin could be considered a chemosensitizing agent during cancer chemotherapy. The gram-positive bacterium Deinococcus radiodurans can survive under extreme ionizing radiation environment. This study aims to rationalize the role of redox balance, antioxidant status, and metabolite content on the radiation survival of D. radiodurans. We found that the TrxR inhibitors, i.e., ebselen, auranofin, and epigallocatechin gallate (EGCG) (10 µM) treatment affects the radiation survival of D. radiodurans. The TrxR inhibitors treatment affects the redox status, activities of antioxidant enzymes, increases the intracellular ROS levels and protein carbonylation upon 4 kGy ionizing radiation treatments. Moreover, the alteration in cellular redox status affects the metabolites content of the organism. In addition, we noticed differential metabolomic profiles in sham control, radiation control (4 kGy), and TrxR inhibitors plus radiation-treated D. radiodurans. The TrxR inhibitors plus radiation treated groups exhibit more variation compare to sham control and 4 kGy radiation-exposed D. radiodurans. Further, some novel metabolites can possess the high antioxidant property and involved in vital cellular metabolism were found in sham control and radiation treated cells of D. radiodurans. Cancer is a hyperproliferative disorder that involves transformation, dysregulation of apoptosis, proliferation, invasion, angiogenesis and metastasis. During the last 30 years, extensive research has revealed much about the biology of cancer. Chemotherapy and radiotherapy are the mainstays of cancer treatment, particularly for patients who do not respond to surgical resection. However, cancer treatment with drugs or radiation is seriously limited by chemoresistance and radioresistance. Various approaches and strategies are employed to overcome resistance to chemotherapy and radiation treatment. Many plant-derived phytochemicals have been investigated for their chemo- and radio-sensitizing properties. The peoples of South Asian countries such as India, Pakistan, Sri Lanka, Nepal, Bangladesh and Bhutan have a large number of medicinal plants from which they produce various pharmacologically potent secondary metabolites. The medicinal properties of these compounds have been extensively investigated and many of them have been found to sensitize cancer cells to chemo- and radio-therapy. This review focuses on the role of South Asian medicinal compounds in chemo- and radio-sensitizing properties in drug- and radio-resistant cancer cells. Also discussed is the role of South Asian medicinal plants in protecting normal cells from radiation, which may be useful during radiotherapy of tumors to spare surrounding normal cells. The aim of this study was to evaluate the photochemopreventive effects of ferulic acid (FA) against chronic ultraviolet-B (290–320 nm) induced oxidative stress, inflammation and angiogenesis in the skin of Swiss albino mice. Chronic UVB exposure (180 mJ/cm 2 for 30 weeks; thrice in a week) induced tumor formation in the mice skin that showed increased expression of carcinogenic and inflammatory markers when compared with the control animals. The intraperitoneal (FAIP) and topical (FAT) administration of FA significantly reduced the incidence of UVB-induced tumor volume and tumor weight in the mice skin. Histopathological studies revealed that both FAIP and FAT administration prevented the UVB-induced hyperplasia, squamous cell carcinoma (SCC) and dysplastic feature in the mice skin. Further, it has been observed that FA treatment reverted chronic UVB-induced oxidative damage (thiobarbituric acid reactive substances, superoxide dismutase, catalase, glutathione peroxidase) accompanied with modulation of vascular endothelial growth factor ( VEGF ), inducible nitric oxide synthase ( iNOS ), TNF-α and IL-6 in the mice skin tumor. FA treatment also modulates mutated p53 , Bcl-2 and Bax expressions in the UVB-induced mice skin tumor. Abstract: The aim of the present study was to assess the protective effect of apigenin, a dietary flavone, against cytogenetic alterations in human peripheral blood lymphocytes (HPBL) induced by Cobalt-60 radiation (3Gy). Results of MTT [3-(4, 5-dimethyl-2-thiaozolyl)-2,5-diphenyl-2H tetrazolium bromide] assay revealed that 37.2μM of apigenin was found to be non-toxic in HPBL. At this dose (37.2μM) of apigenin, the LD50 radiation dose of HPBL increased from 2.9Gy to 3.4Gy, which resulted in a DMF of 1.17. Apigenin (37.2μM) treatment 1h before irradiation significantly (p <0.05) reduced DNA damage in irradiated HPBL as measured by comet assay (% tail DNA, tail length, tail moment, and olive tail moment). Moreover, apigenin treatment significantly decreased the frequencies of dicentric (DC), acentric fragments (AF), and acentric rings (AR) in irradiated HPBL. Apigenin pretreatment also reduced the radiation-induced CBMN (cytokinesis blocked micronuclei) anomalies such as micronuclei (MNi), nucleoplasmic bridges (NPB) and nuclear buds (NBUD) in HPBL. These results also showed that there was a significant correlation between NPB and DC frequencies and MNi and AF+AR. Treatment with apigenin alone had no significant effect on DNA damage and chromosomal aberrations in HPBL. Abstract: Exposure to ultraviolet B (UVB, 280–320) radiation induces the formation of reactive oxygen species (ROS) in the biological system. In this study, we examined the protective effect of ursolic acid on UVB-induced lipid peroxidation and oxidative DNA damage with reference to alterations in cellular antioxidant status in human lymphocytes. Series of in vitro tests (hydroxyl radical, superoxide, nitric oxide, DPPH and ABTS radical scavenging assays) demonstrates antioxidant property of ursolic acid in our study. Treatment of lymphocytes with ursolic acid alone (at 10μg/mL) gave no significant change in cell viability, thiobarbituric acid reactive substances (TBARSs), lipid hydroperoxides (LHPs), % tail DNA and tail moment when compared with normal lymphocytes. UVB-exposure significantly increased TBARS, LHP and % tail DNA, tail moment; decreased % cell viability and antioxidant status in irradiated lymphocytes. Treatment with ursolic acid 30min prior to UVB-exposure resulted in a significant decline of TBARS, LHP, % tail DNA and tail moment and increased % cell viability as ursolic acid concentration increased. Based on our results we conclude that ursolic acid, a dietary antioxidant, mediates its protective effect through modulation of UVB-induced reactive oxygen species. [Copyright &y& Elsevier]

Abstract: UV-B induces lipid peroxidation and oxidative stress in mammalian cells by producing reactive oxygen species. In this present study, we report the protective effect of ferulic acid (FA; 3-methoxy-4-hydroxycinnamic acid), a dietary antioxidant, on UV-B–induced oxidative stress and lipid peroxidation using normal human lymphocytes. Treatment of the human peripheral lymphocytes with UV-B irradiation caused a significant (P < .05) depletion of reduced glutathione (GSH) levels, superoxide dismutase, catalase, and GSH peroxidase activities, and increased the levels of thiobarbituric acid–reactive substances. Treatment of human lymphocytes with FA before 30 minutes of irradiation inhibited UV-B–induced lipid peroxidation. Ferulic acid also inhibited UV-B–induced depletion of antioxidant defense components, such as GSH peroxidase, catalase, superoxide dismutase, and GSH. The maximum dose of FA (10 μg/mL) normalized the UV-B–induced oxidative stress, indicating the protective effect of FA in human peripheral lymphocytes under in vitro conditions against UV-B exposure. Our observations suggest that FA reduces UV-B–induced lipid peroxidation and enhances antioxidant status in human lymphocytes exposed to UV-B radiation, and therefore, FA could serve as a protector against the UV-B–induced pathologic changes. Abstract: In this study we examined radioprotective effect of ferulic acid (FA) on gamma radiation-induced dicentric aberration and lipid peroxidation with reference to alterations in cellular antioxidant status in cultured lymphocytes. To establish most effective protective support we used three different concentrations of FA (1, 5 and 10μg/ml) and three different doses of γ-radiation (1, 2 and 4Gy). Treatment of lymphocytes with FA alone (at 10μg/ml) gave no significant change in micronuclei (MN), dicentric aberration (DC), thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT) or glutathione peroxidase (GPx) activities when compared with normal lymphocytes; irradiation at 1, 2 and 4Gy increased the MN and DC frequencies in a dose-dependent manner. Treatment with FA for 30min before radiation exposure resulted in a significant decline of MN and DC yields as FA concentration increased. Compared to 1Gy exposure alone, the extent to which FA (1μg/ml) reduced the MN and DC yields was 75% and 50%, respectively. With 4Gy irradiation, FA (10μg/ml) decreased 45% MN and 25% DC frequencies. FA-pretreated lymphocytes (1, 5 and 10μg/ml) showed progressively decreased TBARS levels after irradiation. Irradiation (1, 2 and 4Gy) significantly decreased GSH levels, SOD, CAT and GPx activities in a dose-dependent manner. Pretreatment with 10μg/ml of FA significantly (p <0.05) prevented the decreases in the radiation-induced GSH, SOD, CAT and GPx activities. Abstract: Sesamol pretreated (1, 5 and 10μg/ml) lymphocytes were exposed to different doses of γ-radiation, i.e., 1, 2 and 4Gray (Gy) and the cellular changes were estimated by using cytokinesis blocked micronucleus assay (MN), dicentric aberration (DC), thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH) and the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Radiation significantly increased MN, DC frequencies, TBARS levels and decreased GSH and antioxidant enzyme levels in a dose dependent manner. The highest damage to lymphocytes was observed at 4Gy irradiation. On the other hand, sesamol pretreatment significantly decreased MN, DC frequencies, TBARS levels and increased GSH levels and SOD, CAT and GPx activities in a concentration dependent manner. At 1Gy irradiation all concentrations of sesamol (1, 5 and 10μg/ml) significantly protects the lymphocytes from radiation damage. At 2Gy irradiation 5 and 10μg/ml of sesamol shows significant radioprotection. Since the highest damage was observed at 4Gy irradiation both 1 and 5μg/ml of sesamol pretreatment were not sufficient to protect the lymphocytes from radiation damage but 10μg/ml of sesamol significantly (p &#60;0.05) protects the lymphocytes from radiation effect. Thus, sesamol pretreatment gives significant protection to cultured human lymphocytes against γ-radiation induced cellular damage. Multidrug resistance (MDR) is a major obstacle in cancer chemotherapy. P‐glycoprotein (P‐gp) one of the ATP‐binding cassette (ABC) transporters plays an important role in MDR. In this study, we examined the sensitizing property of andrographolide (Andro) to reverse MDR in the drug‐resistant KBChR 8‐5 cells. Andro exhibited increased cytotoxicity in a concentration‐dependent manner in the P‐gp overexpressing KBChR 8‐5 cells. Furthermore, Andro showed synergistic interactions with PTX and DOX in this drug‐resistant cells. Andro co‐administration enhanced PTX‐ and DOX‐induced cytotoxicity and reduced cell proliferation in the MDR cancer cells. Moreover, reactive oxygen species (ROS) were elevated with a decrease in the mitochondrial membrane potential (MMP) during Andro and chemotherapeutic drugs combination treatment in the drug‐resistant cells. Furthermore, Andro and PTX‐induced cell cycle arrest was observed in the drug‐resistant cell. We also noticed that the expression of ABCB1 and AKT were downregulated during Andro (4 µM) treatment. Furthermore, Andro treatment enhanced the expression of caspase 3 and caspase 9 in the combinational groups that support the enhanced apoptotic cell death in drug‐resistant cancer cells. Therefore, the results reveal that Andro plays a role in the reversal of P‐gp‐mediated MDR in KBChR 8‐5 cells which might be due to regulating ABCB1/AKT signaling pathway. Significance statement: ATP‐binding cassette (ABC) transporters‐mediated multidrug resistance (MDR) is a major hindrance during cancer treatment. Efflux mechanism of P‐glycoprotein (P‐gp) decreases bioavailability and efficacy of chemotherapeutic drugs. Already known inhibitors of P‐gp exhibit higher toxicity to the untargeted cells. Phytochemicals have great potential to reverse the MDR mechanism in cancer cells with less toxicity. Andrographolide (Andro), a phytochemical of Andrographis paniculata, enhanced the efficacy of the chemotherapeutic drugs and reverses drug resistance in MDR cancer cells. Background: The use of percutaneous coronary intervention (PCI) in patients with ST‐segment elevation myocardial infarction (STEMI) is associated with a mortality rate of 5%–7%. It is clear that there is an urgent need to develop new drugs that can effectively prevent cardiac reperfusion injury. ATP‐sensitive K+ (KATP) channel openers (KCOs) can be classified as such drugs. Results: KCOs prevent irreversible ischemia and reperfusion injury of the heart. KATP channel opening promotes inhibition of apoptosis, necroptosis, pyroptosis, and stimulation of autophagy. KCOs prevent the development of cardiac adverse remodeling and improve cardiac contractility in reperfusion. KCOs exhibit antiarrhythmic properties and prevent the appearance of the no‐reflow phenomenon in animals with coronary artery occlusion and reperfusion. Diabetes mellitus and a cholesterol‐enriched diet abolish the cardioprotective effect of KCOs. Nicorandil, a KCO, attenuates major adverse cardiovascular event and the no‐reflow phenomenon, reduces infarct size, and decreases the incidence of ventricular arrhythmias in patients with acute myocardial infarction. Conclusion: The cardioprotective effect of KCOs is mediated by the opening of mitochondrial KATP (mitoKATP) and sarcolemmal KATP (sarcKATP) channels, triggered free radicals' production, and kinase activation. Ultraviolet radiation induces oxidative photoaging in the skin cells. In this study, we investigated the ability of andrographolide (ADP) to protect human dermal fibroblasts (HDFa) from UVB radiation‐induced oxidative stress and apoptosis. The HDFa cells were exposed to UVB (19.8 mJ/cm2) radiation in the presence or absence of ADP (7 μM) and then oxidative stress and apoptotic protein expression were analyzed. UVB exposure resulted in a significant decline in the activity of antioxidant enzymes and altered mitochondrial membrane potential (MMP). Furthermore, UVB‐irradiation causes increased intracellular reactive oxygen species (ROS) production, apoptotic morphological changes, and lipid peroxidation levels in the HDFa. Moreover, the pretreatment with ADP reduced the UVB‐induced cytotoxicity, ROS production, and increased antioxidant enzymes activity. Further, the ADP pretreatment prevents the UVB‐induced loss of MMP and apoptotic signaling in HDFa cells. Therefore, the present results suggest that ADP protects HDFa cells from UVB‐induced oxidative stress and apoptotic damage. Significance statement: Andrographolide (ADP) exhibits potent antioxidant in the in vitro free radical scavenging activity and it prevents UVB‐induced oxidative stress and apoptotic signaling in human dermal fibroblasts. Further, we suggest that the ADP pretreatment predominantly protects against UVB‐induced cytotoxicity, lipid peroxidation, antioxidant depletion, and apoptotic molecules. Introduction: Say Meyer syndrome is rare X linked condition characterized by developmental delay, short stature and metopic suture synostosis. We are reporting a case of Say Meyer syndrome presented to our hospital for short stature and developmental delay at age 3 1/2 years. Case Report: A 3 1/2-year-old boy presented to our hospital for decreased growth velocity from the age of 1 year. History revealed the boy had a birth weight of 2.3 kg, had an episode of seizures in the neonatal period. He was born to non-consanguineous marriage. He had global developmental delay and there was a lack of bowel and bladder control. History did not reveal any hearing or visual impairment. No history of any chronic systemic illnesses. Magnetic resonance imaging (MRI) brain revealed mild diffuse frontotemporal atrophy with multiple irregular gliotic areas in bilateral frontal lobes. Diffuse white matter volume loss in bilateral cerebral hemispheres. Diffuse thinning of corpus callosum. Diffuse periventricular hyper intensity on T2W and fluid attenuated inversion recovery sequences. Conclusion: Say Meyer syndrome is rare X linked condition characterized by developmental delay, short stature and metopic suture synostosis. The acute myocardial infarction (AMI) and sudden cardiac death (SCD), both associated with acute cardiac ischemia, are one of the leading causes of adult death in economically developed countries. The development of new approaches for the treatment and prevention of AMI and SCD remains the highest priority for medicine. A study on the cardiovascular effects of chronic hypoxia (CH) may contribute to the development of these methods. Chronic hypoxia exerts both positive and adverse effects. The positive effects are the infarct-reducing, vasoprotective, and antiarrhythmic effects, which can lead to the improvement of cardiac contractility in reperfusion. The adverse effects are pulmonary hypertension and right ventricular hypertrophy. This review presents a comprehensive overview of how CH enhances cardiac tolerance to ischemia/reperfusion. It is an indepth analysis of the published data on the underlying mechanisms, which can lead to future development of the cardioprotective effect of CH. A better understanding of the CH-activated protective signaling pathways may contribute to new therapeutic approaches in an increase of cardiac tolerance to ischemia/reperfusion. Cucurbiturils are magnificent macrocycles that are used as precise molecules for the fabrication of supramolecular chemotherapeutic agents through host-guest molecular recognition. In the present work, the host-guest complex of cucurbit[6]uril (CB[6]) with 5-fluorouracil (Flu, an anticancer drug) is synthesized and its structural study was carried out using UV, XRD, and TG/DTA techniques. The UV-Vis spectrum of the inclusion complex exhibited maximum absorbance with a blue shift at 302 nm whereas the maximum absorbance for free CB[6] was observed at 318 nm. This notable shift in wavelength proves the formation of the inclusion complex of CB[6] with fluorouracil. The host-guest complex of fluorouracil with CB[6] shows sharp endothermic peaks at 403.30C with a heat flow of -0.5 mW/mg the peak covers partially an area of 528.2 J/g, The large variations observed in the DSC curves of the complex compared with free host CB[6] confirm the formation of a stable host-guest complex. The XRD pattern of the host-guest complex is different from the parent compound and this reaffirms the formation of host-guest complex. The cytotoxic effect of 5-fluorouracil and the host-guest complex of CB[6] with fluorouracil on KB cells was determined by MTT assay and the cytotoxicity of anticancer drug Flu towards cancer cells significantly increases by encapsulating 5-fluorouracil into the hydrophobic cavity of CB[6], The ROS generation study was carried out by measurement of intracellular ROS using fluorescent and chemiluminescent probe. The ROS generation of anticancer drugs against cancer cells is significantly increased by encapsulating 5-fluorouracil into the hydrophobic cavity of CB[6], The inclusion complex of cucurbituril with fluorouracil treatment in KB cells showed higher anticancer potential than fluorouracil treatment in KB cells when compared to the control. The IC50 value of 5-flurouracil was found to be higher compared to the inclusion complex of cucurbituril with fluorouracil. Aim: Structurally hybrid ciprofloxacin based sustain drug delivery systems were synthesized with biologically active β and γ-cyclodextrin. Cyclodextrin core ciprofloxacin based drug delivery systems have been synthesized with in two stages by reaction between ciprofloxacin, cyclodextrin and using chloroacetyl chloride as linking agent. Materials and methods: The sustain drug delivery were evaluated by various pH condition and the quantitative analysis of the ciprofloxacin drug formulations on different mode of drug release is monitored using ultraviolet spectrophotometer. Result and Discussion: The structures of the synthesised β-cyclodextrin giant molecules were characterized by means of MALDI-TOF, infrared, H1 and C13 NMR spectral studies. The results obtained undoubtedly evidenced that the drug ciprofloxacin release is pH dependant. Antibacterial performance of the spherical shaped macromolecule were investigated with Escherichia coli and staphylococcus bacteria and the cell viability reduced value of the designed β and γ-cyclodextrin core drug delivery system. The very long time active of drug is compared with standard ciprofloxacin. The in-vitro studies and ciprofloxacin discharge from the spherical shaped macromolecules were also investigated. Graphical abstract Abstract Solar ultraviolet-B radiation (UVB) has severe adverse effects on the structure and functions of the skin. Although, UVB (290–320 nm) represents only 5–10% of UV light reaching earth's surface, its contribution towards photoaging is tremendous. In this present study was investigate the photoprotective effect of methanolic extract of the male flower of J. regia L. (MEJR) against UVB induced photoaging in human epidermal keratinocytes (HaCaT). Cells were exposed to UVB-irradiation at a dose of 20 mJ/cm2, induces the activation of several signaling pathways which are associated with oxidative stress and photoaging. A single dose of UVB irradiation increased the protein and mRNA expression of MAPKs, AP-1, MMPs, Smad7 and decreased expression of TIMP-1/2, TGF-β1, Smad3, procollagen type-1 in HaCaT cells. In contrast, pretreatment of MEJR (80 μg/ml) prior to UVB-irradiation significantly prevented the overexpression of MAPKs, AP-1, MMPs, Smad7 and decreased expression of TIMP-1/2, TGF-β1, Smad3 and procollagen type-1 in HaCaT cells. Moreover, pretreatment of MEJR (80 μg/ml) prior to UVB-irradiation significantly prevents apoptosis in sub G o -phase. Thus, MEJR protects UVB-mediated photoaging in human skin cells, by modulating the expression of photoaging markers. Exposure to solar ultraviolet-B (UVB) radiation leads to the formation of cyclobutane pyrimidine dimers (CPDs). We investigated the protective effect of apigenin against UVB-induced CPDs formation in human dermal fibroblasts cells (HDFa). For this purpose, HDFa cells were treated with apigenin (15 μM) prior to UVB irradiation (20 mJ/cm 2 ); DNA damage and subsequent molecular end points were observed. Exposure to UVB radiation increased significant CPDs formation in HDFa cells and the frequencies of CPDs were reduced by treatment with apigenin (15 μM). UVB-induced CPDs downregulates the expression of nucleotide excision repair (NER) genes such as xeroderma pigmentosum complementation group C, B, G and F (XPC, XPB, XPG and XPF), transcription factor II human (TFIIH) and excision repair cross-complementation group 1 (ERCC1) in HDFa cells. Conversely, apigenin treatment restored UVB-induced loss of NER proteins in HDFa cells, which indicates its preventive effect against CPDs formation. Besides, single low dose UVB-exposure induced nuclear fragmentation, apoptotic frequency and apoptotic proteins expression (Bax and Caspase-3) have been prevented by the apigenin pretreatment. Furthermore, apigenin exhibits strong UV absorbance property and showed 10.08 SPF value. Thus, apigenin can protect skin cells against UVB-induced CPDs formation probably through its sunscreen effect. There are two types of vitamin D dependent rickets (VDDR) that cause rickets in children. VDDR type 1 (VDDR-I) is caused by an inborn error of vitamin D metabolism, which interferes with renal conversion of calcidiol (25OHD) to calcitriol (1,25(OH)2 D) by the enzyme 1-α-hydroxylase. Patients with VDDR-I have mutations of chromosome 12 that affect the gene for the enzyme 1-α-hydroxylase, resulting in decreased levels of 1,25(OH) vitamin D. Clinical features include growth failure, hypotonia, weakness, rachitic rosary, convulsions, tetany, open fontanels and pathologic fractures. We report a case of VDDR-I in 14-month-old male child. Establishing an early diagnosis of these genetic forms of rickets is challenging, especially in developing countries where nutritional rickets is the most common variety of the disease where genetic diagnosis is not always possible because of financial constraints. A prompt diagnosis is necessary to initiate adequate treatment, resolve biochemical features and prevent complications, such as severe deformities that may require surgical intervention. Multidrug resistance (MDR) remains a major obstacle in cancer chemotherapy. The use of the dietary phytochemicals as chemosensitizing agents to enhance the efficacy of conventional cytostatic drugs has recently gained the attention as a plausible approach for overcoming the drug resistance. The aim of this study was to investigate whether a naturally occurring diet-based phenolic acid, ferulic acid, could sensitize paclitaxel efficacy in ABCB1 overexpressing (P-glycoprotein) colchicine selected KB Ch R 8-5 cell line. In vitro drug efflux assays demonstrated that ferulic acid inhibits P-glycoprotein transport function in drug resistant KB Ch R 8-5 cell lines. However, ferulic acid significantly downregulates ABCB1 expression in a concentration dependent manner. Cytotoxicity assay reveals that ferulic acid decreased paclitaxel resistance in KBCh R 8-5 and HEK293/ABCB1 cells, which indicates its chemosensitizing potential. Clonogenic cell survival assay and apoptotic morphological staining further confirm the chemosensitizing potential of ferulic acid in drug resistant KB Ch R 8-5 cell lines. Ferulic acid treatment enhances paclitaxel mediated cell cycle arrest and upregulates paclitaxel-induced apoptotic signaling in KB resistant cells. Hence, it has been concluded that downregulation of ABCB1 and subsequent induction of paclitaxel-mediated cell cycle arrest and apoptotic signaling may be the cause for the chemosensitizing potential of ferulic acid in P-gp overexpressing cell lines. Quantum dots (QDs) are gaining widespread recognition for its luminescence behavior and unique photo physical properties as a bio-marker and inorganic fluorophore. In spite of such rampant advantages, its application is clinically hampered depending on the surface coating decreasing its luminescence efficiency. The present study reports preparation of CdTe QDs capped with biologically active thiol based material, mercaptosuccinic acid (MSA) for diagnosis of oral cancer (KB) cells by acting as a fluorophore marking targeted tumor cells and at the same time exhibiting certain cytotoxic effects. Synthesized MSA coated CdTe QDs is spherical in shape with an average particle size of 3-5nm. In vitro, the rapid uptake of MSA CdTe QDs in oral cancer cell lines were assessed through fluorescence microscopy. Further, this study evaluates the therapeutic efficiency of MSA CdTe QDs in human oral cancer cell lines using MTT analysis. MSA CdTe QDs exhibit significant cytotoxicity in oral cancer cells in a dose dependent manner with low IC50 when compared with other raw CdTe QDs. MSA CdTe QDs were also treated with human lymphocytes (normal cells) to assess and compare the toxicity profile of QDs in normal and oral tumors. The results of our present study strengthen our hypothesis of using MSA CdTe QDs as detector for tracking and fluorescence imaging of oral cancer cells and exhibiting sufficient cytotoxicity in them. Introduction: Identification of heterozygous germ-line mutations in the human CDKN1B in patients with multiple endocrine tumors leads to the identification of multiple endocrine neoplasia (MEN) 4 caused due to mutations in p27. Background: We are presenting a case of MEN 4 who presented with pituitary macroadenoma, primary hyperparathyroidism. Case Report: A 37-year-old male patient presented with a history of erectile dysfunction and recurrent headaches on and off, and no history of vomiting, or visual abnormalities. His elder sister died because of brain tumor. Serum prolactin initially was >200 ng/ml and after serial dilutions, it was 6096 ng/ml. MRI brain revealed a lobulated solid mass lesion involving the pituitary stalk extending into the right cavernous sinus and encasing the right internal carotid artery and displacing the optic chiasma. His serum calcium was 10.9 mg/dl and parathyroid hormone was 245.5 pg/ml. Ultrasound thyroid revealed a well-deined hyper echoic lesion posterior to the left lobe of thyroid and ill-deined hyper echoic heterogenous lesion in the inferior polar region of the left lobe of thyroid. His thyroid proile, 8 am cortisol, follicle stimulating hormone (FSH), growth hormone hemodynamics were normal at the initial presentation. He gave history of renal stone disease during the past. On family screening of his three younger sisters one had hyperparathyroidism, one had hyperprolactinemia and one had both. Conclusion: In view of presence only hyperparathyroidism, pituitary tumors without pancreatic tumors, and strong family history possibility of MEN 4 was kept. [ABSTRACT FROM AUTHOR]

In this study, we evaluated the role of linalool in acute ultraviolet-B (UVB; 280–320 nm) radiation-induced inflammation and chronic UVB-mediated photocarcinogenesis in mouse skin. Acute UVB-irradiation (180 mJ cm−2) causes hyperplasia, edema formation, lipid peroxidation, antioxidant depletion, and overexpression of cyclooxygenase-2 (COX-2) and ornithine decarboxylase (ODC) in mouse skin. Topical or intraperitoneal (i.p.) treatment of linalool prevented acute UVB-induced hyperplasia, edema formation, lipid peroxidation, and antioxidant depletion in mouse skin. Further, linalool treatment prevented UVB-induced overexpression of COX-2 and ODC in mouse skin. In the chronic study, mice were subjected to UVB-exposure thrice weekly for 30 weeks. Chronic UVB-exposure induced tumor incidence and expression of proliferative markers such as NF-κB, TNF-α, IL-6, COX-2, VEGF, TGF-β1, Bcl-2 and mutated p53 in mouse skin. Treatment with linalool before each UVB-exposure significantly prevented the expression of these proliferative markers and subsequently decreased the tumor incidence in mice skin. Histopathological studies confirmed the development of dysplasia and squamous cell carcinoma (SCC) in the chronic UVB-exposed mouse skin; and this was prevented by both topical and i.p. linalool treatment. Introduction: Autoimmune hypoglycemia is a rare cause of hyperinsulinemic hypoglycemia. Background: We report a case of 60-year-old male, non-diabetic patient who presented with insulin autoantibody syndrome. Case Report: A 60-year-old male patient admitted to our hospital with a history of recurrent episodes of sweating, giddiness, and seizures since 7 days, which was corrected after taking sugar water. The patient documented sugars of 64 mg% and 24 mg% on 2 separate days, during the attacks. There was no history of diabetes mellitus. He is a known case of hypertension and cerebrovascular accident with right hemiparesis with history of use of telmisartan and amlodepin. History of repeated use of aceclofenac with paracetamol for body aches in the past 6 months. There was no history of hepatic or renal dysfunction. The patient was initiated on 72 h extended fasting test to induce hypoglycemia. Within 4 h after initiation of the fast, patient developed hypoglycemia (25 mg/dl). During hypoglycemia, he had serum insulin of 300 µu/ml, and insulin antibody titer of 256.2 units (Normal range <18 units/ml). The patient was having normal cortisol, euthyroid, and had normal hepatic and renal functions. Conclusion: Insulin autoantibody syndrome is a rare condition characterized by antibodies directed against insulin, bind with insulin, and release insulin inappropriately when substrate (glucose) levels are normal. P-glycoprotein, encoded by ATP-binding cassette transporters B1 gene (ABCB1), renders multidrug resistance (MDR) during cancer chemotherapy. Several synthetic small molecule inhibitors affect P-glycoprotein (P-gp) transport function in MDR tumor cells. However, inhibition of P-gp transport function adversely accumulates chemotherapeutic drugs in non-target normal tissues. Moreover, most small-molecule P-gp inhibitors failed in the clinical trials due to the low therapeutic window at the maximum tolerated dose. Therefore, downregulation of ABCB1 -gene expression (P-gp) in tumor tissues seems to be a novel approach rather than inhibiting its transport function for the reversal of multidrug resistance (MDR). Several plant-derived phytochemicals modulate various signal transduction pathways and inhibit translocation of transcription factors, thereby reverses P-gp mediated MDR in tumor cells. Therefore, phytochemicals may be considered an alternative to synthetic small molecule P-gp inhibitors for the reversal of MDR in cancer cells. This review discussed the role of natural phytochemicals that modulate ABCB1 expression through various signal transduction pathways in MDR cancer cells. • Preparation of self-assembled two opposites charged polysaccharide (fucoidan and chitosan). • Chitosan enhanced intestinal absorption of fuocidan in mice intestine. • Fucoidan are capable of increased protecting the intestinal epithelium from radiation damage. Fucoidan, a sulphated polysaccharide, plays a vital role in reducing cellular oxidative damage by exerting potential antioxidant activity. However, because of the negative surface charges of oligofucoidan, it shows poor oral intestinal absorption. To overcome this drawback, the oligofucoidan polysaccharides self-assembled with opposite charge based polysaccharides (chitosan) to form the chitosan-fucoidan polysaccharides (C 1 -F 3 P) nanoparticles (NPs) of 190–230 nm in size. The oligofucoidan and C 1 -F 3 P NPs were studied for their radioprotective property using mice exposed to 5 Gy radiation. The C 1 -F 3 P NPs prevents radiation induced lipid peroxidation and restores intestinal enzymatic and non-enzymatic antioxidants (p < 0.05) status. In addition, hematoxylin-eosin staining revealed the radioprotective effect of oligofucoidan and C 1 -F 3 P NPs by mitigating the loss of crypt and villi in the small intestine. Thus, the present study demonstrated that C 1 -F 3 P NPs can be considered as a radioprotective agent that can be used for the prevention and treatment of Gy-radiation-induced intestine injury.
