Abstract

Les marais de Brière (Loire-Atlantique) font l’objet d’une forte colonisation par la Grande Jussie (Ludwigia grandiflora ssp. hexapetala) depuis 1994. Autrefois connue comme plante strictement aquatique, elle commence à envahir les prairies humides exondées en période estivale. La Grande Jussie régresse dans les endroits où elle est gérée annuellement, mais les contraintes économiques ne permettent pas d’envisager actuellement l’extension des moyens de lutte de manière à gérer l’ensemble des colonisations aquatiques. C’est pourquoi, les gestionnaires locaux orientent leurs réflexions vers la recherche de solutions globale, reposant sur une gestion des milieux défavorables à la Jussie. L’un des axes de travail développé est la restauration d’une eau salée provenant de l’estuaire de la Loire, dans des espaces naturellement soumis à l’influence des marées dynamiques. Cette étude a donc deux objectifs : - apporter des compléments quant à l’impact du sel en milieu contrôlé sur les phénotypes aquatiques et terrestre de la Grande Jussie ; - évaluer l’impact de l’envoi d’eau saumâtre au niveau du Canal du Priory sur la population de Jussie du lieu-dit « Caloyau » situé sur la commune de Montoir de Bretagne (44).

Abstract

Although most telomere repeat sequences are found at the ends of chromosomes, some telomeric repeat sequences are also found at intrachromosomal locations in mammalian cells. Several studies have found that these interstitial telomeric repeat sequences can promote chromosome instability in rodent cells, either spontaneously or following ionizing radiation. In the present study we describe the extensive cytogenetic analysis of three different human cell lines with plasmids containing telomeric repeat sequences integrated at interstitial sites. In two of these cell lines, Q18 and P8SX, instability has been detected in the chromosome containing the integrated plasmid, involving breakage/fusion/bridge cycles or amplification of the plasmid DNA, respectively. However, the data suggest that the instability observed is characteristic of the general instability in these cell lines and that the telomeric repeat sequences themselves are not responsible. Consistent with this interpretation, the chromosome containing an integrated plasmid with 500 bp of telomeric repeat sequences is highly stable in the third cell line, SNG28, which has a relatively stable genome. The stability of the chromosome containing the integrated plasmid sequences in SNG28 makes this an excellent cell line to study the effect of ionizing radiation on the stability of interstitial telomeric sequences in human cells. Abstract

Performing fish survey protocols with different electrofishing equipment and settings can lead to difficulties in comparing the data obtained. In this study, we captured two fish taxa (i.e. the European eel and salmonids: Atlantic salmon and brown trout) in the field using four models of electrofishers that provided different waveforms: direct current (DC), pulsed direct current (PDC) and pulsed exponential current (PEC). This study aimed to assess effects of using different waveforms and frequencies, while considering environmental variables, on i) attraction, represented by occurrence and catch-per-unit-effort (CPUE) of fishes, and ii) spinal damage, based on ultrasound of the spinal column. For all species, DC and PDC yielded equivalent occurrences and CPUEs, regardless of the frequency. In contrast, PEC induced a significant decrease in occurrences and CPUEs of both eels and salmonids. The percentage of injured fishes increased as the frequency or length of the salmonids increased while waveform had no effect. Ultrasound was determined to be a good non-invasive method to detect injuries in fishes.

Spinability is a rheological parameter which characterizes the property of fluids to be drawn into threads when stretched. It has been shown that this property is of particular importance in the mucus transport process. This paper describes an automatic apparatus which measures the spinability of a calibrated volume (20 microliters) of sample at a stretching rate of 10 mm.sec.-1. The reproducibility of the measurements is analyzed on mucus simulants and on different types of bronchial mucus. Using this technique, we observed that the spinability of bronchial secretions is significantly correlated (r = 0.69, p less than 0.001) with their transport rate measured on the depleted frog palate. This device is suitable for measuring the spinability of all conductive types of mucus and other biological fluids.
