2. No Differences between Adults with and without Autism in Audiovisual Synchrony Perception

- Abstract
To facilitate multisensory processing, the brain binds multisensory information when presented within a certain maximum time lag (temporal binding window). In addition, and in audiovisual perception specifically, the brain adapts rapidly to asynchronies within a single trial and shifts the point of subjective simultaneity. Both processes, temporal binding and rapid recalibration, have been found to be altered in individuals with an autism spectrum disorder diagnosis. Here, we used a large adult sample (autism spectrum disorder: n = 75, no autism spectrum disorder: n = 85) to replicate these earlier findings. In this study, audiovisual stimuli were presented in a random order across a range of stimulus onset asynchronies, and participants indicated whether they were perceived simultaneously. Based on the synchrony distribution, their individual temporal binding window and point of subjective simultaneity were calculated. Contrary to previous findings, we found that the temporal binding window was not significantly different between both groups. Rapid recalibration was observed for both groups but did not differ significantly between groups. Evidence of an age effect was found which might explain discrepancies to previous studies. In addition, neither temporal binding window nor rapid recalibration was correlated with self-reported autistic symptoms or sensory sensitivity.

7. Adults with Autism Spectrum Disorder Show Atypical Patterns of Thoughts and Feelings during Rest

- Abstract
Mind wandering constitutes a major part of everyday experience and is inherently related to how we feel and identify ourselves. Thus, probing the character and content of thoughts and feelings experienced during mind-wandering episodes could lead to a better understanding of the human mind in health and disease. How mind wandering and spontaneous thought processes are affected in disorders such as autism is poorly understood. Here, we used the eyes-closed rest condition to stimulate mind wandering and quantified the subjective experiences using the Amsterdam Resting-State Questionnaire--which quantifies subjective psychological states of resting-state cognition across 10 domains--in 88 adults with autism spectrum disorder and 90 controls. We observed an atypical pattern of both thoughts and feelings in the autism spectrum disorder cohort, specifically in the domains of Theory of Mind, Comfort, and Discontinuity of Mind. We propose that the use of the Amsterdam Resting-State Questionnaire as a standardized cognitive instrument could advance our understanding of thoughts and feelings in autism spectrum disorder as well as in a wide variety of other brain disorders and how these may change due to therapeutic interventions.

8. The Dutch Sensory Perception Quotient-Short in Adults with and without Autism

- Abstract
Sensory symptoms were recently added to the diagnostic criteria of autism spectrum disorder and may be a mechanism underlying the broad phenotype of autism spectrum disorder. To measure sensory symptoms based on perceptual rather than affective, regulative, or attention components, the Sensory Perception Quotient (SPQ) measuring five modalities of sensory sensitivity has been developed. In this study, the Dutch translation of the abridged SPQ-Short was investigated in a large sample of adults with (n = 657) and without autism spectrum disorder (n = 585). Its hypothesized factor structure, combining modality specific and one modality-independent factor, was assessed in a hierarchical model. Results show that modality-specific subscales are indeed present in the short version. Furthermore, its reliability is high and comparable to the original English version. The autism spectrum disorder group reported higher sensory sensitivities than the comparison group, and women with autism spectrum disorder reported higher sensitivities compared with men with autism spectrum disorder. The SPQ-Short correlates with all Autism Quotient (AQ)-Short subscales, except for the "imagination" subscale. The SPQ-Short seems suitable to further explore the relationship between basic sensory sensitivities in autism spectrum disorder and their related symptoms such as over- and under-responsivity to sensory stimulation.

- Abstract
BACKGROUND: The COVID-19 pandemic has had an acute impact on child mental and social health, but long-term effects are still unclear. We examined how child mental health has developed since the start of the COVID-19 pandemic up to 2 years into the pandemic (April 2022).METHODS: We included children (age 8-18) from two general population samples ( N = 222-1333 per measurement and N = 2401-13,362 for pre-covid data) and one clinical sample receiving psychiatric care ( N = 334-748). Behavioral questionnaire data were assessed five times from April 2020 till April 2022 and pre-pandemic data were available for both general population samples. We collected parent-reported data on internalizing and externalizing problems with the Brief Problem Monitor and self-reported data on Anxiety, Depressive symptoms, Sleep-related impairments, Anger, Global health, and Peer relations with the Patient-Reported Outcomes Measurement Information System (PROMIS®). RESULTS: In all samples, parents reported overall increased internalizing problems, but no increases in externalizing problems, in their children. Children from the general population self-reported increased mental health problems from before to during the pandemic on all six PROMIS domains, with generally worst scores in April 2021, and scores improving toward April 2022 but not to pre-pandemic norms. Children from the clinical sample reported increased mental health problems throughout the pandemic, with generally worst scores in April 2021 or April 2022 and no improvement. We found evidence of minor age effects and no sex effects.CONCLUSIONS: Child mental health in the general population has deteriorated during the first phase of the COVID-19 pandemic, has improved since April 2021, but has not yet returned to pre-pandemic levels. Dit rapport biedt een overzicht van onderzoeken naar de belangrijkste gevolgen van de coronacrisis en daaraan gerelateerde maatregelen op het mentaal welbevinden van kinderen, jongeren en jongvolwassenen in Nederland. Doel is ook om de bevindingen uit alle beschikbare onderzoeken door jongeren en onderzoekers gezamenlijk te laten duiden. We kijken in dit rapport naar verschillende aspecten van mentaal welbevinden. Zoals kwaliteit van leven, depressie, angst en psychosomatische klachten, waaronder stress. Ook kijken we naar wat er tijdens de coronacrisis gebeurde in de sociale omgeving van jongeren. Bijvoorbeeld in het gezin, onder leeftijdsgenoten, hun vrijetijdsbesteding, op school en werk. Daarnaast beschouwen we hoe het toekomstperspectief van jongeren zich heeft ontwikkeld tijdens de coronacrisis en hoe jongeren zelf aankijken tegen het coronabeleid

- Abstract
Objective: To explain the differential course of anxiety and depression in individuals from childhood to adulthood by examining age-related changes in the genetic and environmental etiology of anxious and depressive symptoms. Method: A sample of 1470, 1839, and 2023 Dutch twins aged 12, 14, and 16 years reported on symptoms of anxious depression (AD) and withdrawn behavior (WB), using the Youth Self Report (YSR). AD and WB were analyzed with bivariate cross-sectional genetic models for each age group to obtain estimates of the relative influence of genes (A), shared (C), and nonshared (E) environment. Results: The best-fitting models revealed no difference between heritability estimates in boys and girls. Familial clustering at age 12 years was explained by genetic and shared environmental factors. At ages 14 and 16 years, genetic factors were sufficient to explain familial clustering, shared environmental effects were absent. Genetic influences on AD and WB correlated highly. Conclusions: These findings are in agreement with earlier studies on age-specific effects of genes and shared environment on anxiety, depression, and withdrawn behavior in childhood and adolescence. The current study demonstrated that the decrease in the role of shared environment occurs after age 12. Hormonal changes accompanying the onset of puberty do not seem to explain the change in risk factors, as in 90% of the subjects puberty had already started. More knowledge on age-specific risk factors may offer opportunities for therapeutic interventions. (Contains 2 figures and 3 tables.)

Sex differences on the Dutch WISC-R were examined in Dutch children (350 boys, 387 girls, age 11-13 years) and Belgian children (370 boys, 391 girls, age 9.5-13 years). Multi-group covariance and means structure analysis was used to establish whether the WISC-R was measurement invariant across sex, and whether sex differences on the level of the subtests were indicative of sex differences in general intelligence (g). In both samples, girls outperformed boys on the subtest Coding, while boys outperformed girls on the subtests Information and Arithmetic. The sex differences in the means of these three subtests could not be accounted for by the first-order factors Verbal, Performance, and Memory. Measurement invariance with respect to sex was however established for the remaining 9 subtest. Based on these subtests, no significant sex differences were observed in the means of the first-order factors, or the second-order g-factor. In conclusion, the cognitive differences between boys and girls concern subtest-specific abilities, and these sizeable differences are not attributable to differences in first-order factors, or the second-order factor g.

31. Across the Continuum of Attention Skills: A Twin Study of the SWAN ADHD Rating Scale

- Abstract
Introduction: Most behavior checklists for attention problems or attention deficit/hyperactivity disorder (ADHD) such as the Child Behavior Checklist (CBCL) have a narrow range of scores, focusing on the extent to which problems are present. It has been proposed that measuring attention on a continuum, from positive attention skills to attention problems, will add value to our understanding of ADHD and related problems. The Strengths and Weaknesses of ADHD symptoms and Normal behavior scale (SWAN) is such a scale. Items of the SWAN are scored on a seven-point scale, with in the middle "average behavior" and on the extremes "far below average" and "far above average". Method: The SWAN and the CBCL were completed by mothers of respectively 560 and 469 12-year-old twin pairs. The SWAN consists of nine DSM-IV items for Attention Deficit (AD) and nine DSM-IV items for Hyperactivity/Impulsivity (HI). The CBCL Attention Problem (AP) scale consists of 11 items, which are rated on a three-point scale. Results: Children who had a score of zero on the CBCL AP scale can be further differentiated using the SWAN, with variation seen between the average behavior and far above average range. In addition, SWAN scores were normally distributed, rather than kurtotic or skewed as is often seen with other behavioral checklists. The CBCL AP scale and the SWAN-HI and AD scale were strongly influenced by genetic factors (73%, 90% and 82%, respectively). However, there were striking differences in genetic architecture: variation in CBCL AP scores is in large part explained by non-additive genetic influences. Variation in SWAN scores is explained by additive genetic influences only. Conclusion: Ratings on the SWAN cover the continuum from positive attention skills to attention and hyperactivity problems that define ADHD. Instruments such as the SWAN offer clinicians and researchers the opportunity to examine variation in both strengths and weaknesses in attention skills.

32. The Phenotypic and Genotypic Relation between Working Memory Speed and Capacity

- Abstract
This study examined the phenotypic and genotypic relationship between working memory speed (WMS) and working memory capacity (WMC) in 12-year-old twins and their siblings (N = 409). To asses WMS all children performed a reaction time task with three memory loads from which a basic mental speed measure and the derived slope were used. WMC was measured with two subtests of the WISC-R, namely Arithmetic and Digit Span. The phenotypic correlations among the WMS and WMC indices were around -0.30. Heritabilities for all variables ranged from 43% to 56%. Structural equating modelling revealed that a model with two genetic factors, representing WMS and WMC, which were correlated (-0.54) fitted the data best, indicating that WMS and WMC are partly mediated by the same set of genes and partly by separate sets of genes. When general IQ was simultaneously analysed with the data the correlation between the genetic factors for WMS and WMC decreased (-0.25), but was still significant. This means that approximately 50% of the genetic correlation between WMS and WMC is explained by IQ.

- Abstract
This chapter will first present an overview of the genetic and environmental mechanisms that cause differences in human complex traits and diseases. We focus on findings that demonstrate that all traits are heritable, whether and how heritability of traits and diseases differ between men and women, and the same genetic factors account for why two traits or two diseases correlate. We then present findings on how environmental factors, both measured and unmeasured, augment, moderate, and correlate with genotype to maximize (and minimize) genetic expression of traits and diseases. Here, we give the three most common examples in the behavior genetics literature: cognitive ability, personality, and psychopathology. Finally, we cover the ways in which behavior genetics will be important in future research for clarifying the role of genotype and environment in understanding the etiology of traits and diseases.

Insomnia is the second most prevalent mental disorder, with no sufficient treatment available. Despite substantial heritability, insight into the associated genes and neurobiological pathways remains limited. Here, we use a large genetic association sample (n = 1,331,010) to detect novel loci and gain insight into the pathways, tissue and cell types involved in insomnia complaints. We identify 202 loci implicating 956 genes through positional, expression quantitative trait loci, and chromatin mapping. The meta-analysis explained 2.6% of the variance. We show gene set enrichments for the axonal part of neurons, cortical and subcortical tissues, and specific cell types, including striatal, hypothalamic, and claustrum neurons. We found considerable genetic correlations with psychiatric traits and sleep duration, and modest correlations with other sleep-related traits. Mendelian randomization identified the causal effects of insomnia on depression, diabetes, and cardiovascular disease, and the protective effects of educational attainment and intracranial volume. Our findings highlight key brain areas and cell types implicated in insomnia, and provide new treatment targets.

Genome-wide association meta-analysis (GWAMA) is an effective approach to enlarge sample sizes and empower the discovery of novel associations between genotype and phenotype. Independent replication has been used as a gold-standard for validating genetic associations. However, as current GWAMA often seeks to aggregate all available datasets, it becomes impossible to find a large enough independent dataset to replicate new discoveries. Here we introduce a method, MAMBA (Meta-Analysis Model-based Assessment of replicability), for assessing the “posterior-probability-of-replicability” for identified associations by leveraging the strength and consistency of association signals between contributing studies. We demonstrate using simulations that MAMBA is more powerful and robust than existing methods, and produces more accurate genetic effects estimates. We apply MAMBA to a large-scale meta-analysis of addiction phenotypes with 1.2 million individuals. In addition to accurately identifying replicable common variant associations, MAMBA also pinpoints novel replicable rare variant associations from imputation-based GWAMA and hence greatly expands the set of analyzable variants., In genome-wide association meta-analysis, it is often difficult to find an independent dataset of sufficient size to replicate associations. Here, the authors have developed MAMBA to calculate the probability of replicability based on consistency between datasets within the meta-analysis.

Cortical thickness, surface area and volumes vary with age and cognitive function, and in neurological and psychiatric diseases. Here we report heritability, genetic correlations and genome-wide associations of these cortical measures across the whole cortex, and in 34 anatomically predefined regions. Our discovery sample comprises 22,824 individuals from 20 cohorts within the Cohorts for Heart and Aging Research in Genomic Epidemiology (CHARGE) consortium and the UK Biobank. We identify genetic heterogeneity between cortical measures and brain regions, and 160 genome-wide significant associations pointing to wnt/β-catenin, TGF-β and sonic hedgehog pathways. There is enrichment for genes involved in anthropometric traits, hindbrain development, vascular and neurodegenerative disease and psychiatric conditions. These data are a rich resource for studies of the biological mechanisms behind cortical development and aging., Cortex morphology varies with age, cognitive function, and in neurological and psychiatric diseases. Here the authors report 160 genome-wide significant associations with thickness, surface area and volume of the total cortex and 34 cortical regions from a GWAS meta-analysis in 22,824 adults.

Enhancing NeuroImaging Genetics through Meta-Analysis Consortium (ENIGMA)—Genetics working group., The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity. Variation in cortical structure is genetically correlated with cognitive function, Parkinson's disease, insomnia, depression, neuroticism, and attention deficit hyperactivity disorder.

- Abstract
The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity. Variation in cortical structure is genetically correlated with cognitive function, Parkinson's disease, insomnia, depression, neuroticism, and attention deficit hyperactivity disorder.
