The accurate assessment of forest biomass is vital to climate change mitigation. Based on forest survey data, stand biomass models can effectively assess forest biomass carbon at large scales. However, traditional stand biomass models have ignored the potential effects of the climate on stand biomass estimation. There is still a lack of research on whether or not and in what ways the effects of the climate reduce uncertainty in biomass estimation and carbon accounting. Therefore, two types of stand biomass models, including basic stand biomass models (BBMs) and climate-sensitive stand biomass models (CBMs), were developed and tested using 311 plantation plots of Korean pine (Pinus koraiensis Siebold & Zucc.), Korean larch (Larix olgensisi A. Henry), and Mongolian pine (Pinus sylvestris var. mongolica Litv.) in Northeast China. The two types of models were developed by applying simultaneous equations based on nonlinear, seemingly unrelated, regression (NSUR) to ensure additivity of the stand total and components biomass (root, stem, branch, and needle). The results of fitting and leave-one-out cross-validation (LOOCV) indicated that the CBMs performed better than the corresponding BBMs. The RMSEs of the stand total biomass decreased by 3.5% to 10.6% for the three conifer species. The influence of temperature-related climate variables on the biomass of stand components was greater than that of precipitation-related climate variables. The sensitivity of the three conifer species to climate variables was ranked as Korean pine > Mongolian pine > Korean larch. This study emphasizes the importance of combining climate variables in stand biomass models to reduce the uncertainty and climate effects in forest biomass estimation, which will play a role in carbon accounting for forest ecosystems. Abstract: Horqin Sandy Land is a seriously desertified and an ecologically fragile region of China. Soil degradation and desertification in this region are the result of several decades of overgrazing, non-manure cropping with short fallow, and arbitrary land use and management. We investigated whether the afforestation of active sand dunes with Mongolian pine (Pinus sylvestris var. mongolica Litv.) enhances the carbon (C) and nitrogen (N) storages and the overall soil quality. We compared soil physico-chemical properties, and C and N storages in the total and light fraction soil organic matter of active sand dunes, and of 25- and 35-year-old plantations. Soil water holding capacity, fine particle content and nutrients were significantly higher in plantations than in active sand dunes, with greater improvements occurring in the top 5cm than in the 5–15cm layer of the soil profile. Soil C and total N storages were 6.1 and 3.7 folds in 25-year-old, and 10.2 and 5.3 folds in 35-year-old plantations compared to active sand dunes, respectively. Carbon and N storages in the light fraction were 20.9 and 15.8 folds in 25-year-old, and 36.1 and 25.0 folds in 35-year-old plantations, respectively, relative to active sand dunes. The light fraction played an important role in soil C sequestration and its amount is an indicator of the effects of afforestation on C storage in sandy soil. The upper 15cm of the soil profile in the 35-year-old plantations of Mongolian pine has the capacity to sequester significant amount of C in the region, potentially offsetting part of the carbon that has been lost due to desertification over the past century in the Horqin Sandy Land. Our results conclude that afforestation with Mongolian pine has had positive impacts on soil quality and has increased the capacity for soil C sequestration in semi-arid degraded areas. Abstract: In order to understand the changes of surface soil carbon (C) storage following the afforestation of sandy grasslands, we used physical fractionation procedures to quantify C concentrations and sucrase enzyme activity in bulk soil and different particle fractions along two replicate chronosequences of Mongolian pine (Pinus sylvestris var. mongolica Litv.) plantations in the southeastern Keerqin Sand Lands, Northeast China. Carbon concentration in bulk topsoil (0–15cm) initially decreased following afforestation of grassland and subsequently increased as the forest matured. In general, this pattern of C concentration changes was associated with all particle-size fractions (except clays) and both macro- and microaggregates. The patterns of topsoil C were also influenced by wind erosion and deposition, with marked increases in the relative mass of silt and fine sand fractions occurring during forest development. The loss of aggregates immediately following afforestation was counteracted by formation of aggregates as the forests developed, contributing to the stabilization of carbon. To enhance soil C storage during afforestation of sandy soils in such semi-arid regions it is recommended to minimize disruption of grassland vegetation during the planting stage. The effects of soil animals on soil nitrogen (N) mineralization and its availability were studied by investigating soil animal groups and their amounts of macro-faunas sorted by hand, and middle and microfaunas distinguished with Tullgren and Baermann methods under three Pinus sylvestris var. mongolica Litv. plantations in Zhanggutai sandy land, China. In addition, soil N mineralization rate was also measured with PVC closed-top tube in situ incubation method. The soil animals collected during growing season belonged to 13 orders, 5 groups, 4 phyla, whose average density was 86 249.17 individuals·m−2. There were significant differences in soil animal species, densities, diversities and evenness among three plantations. Permanent grazing resulted in decrease of soil animal species and diversity. The average ammonification, nitrification and mineralization rates were 0.48 g·m−2·a−1, 3.68 g·m−2·a−1 and 4.16 g·m−2·a−1, respectively. The ammonification rate in near-mature forest was higher than that in middle-age forests, while the order of nitrification and net mineralization rates was: middle-age forest without grazing < middle-age forest with grazing < near-mature forest with grazing ( P<0.05). Soil N mineralization rate increased with soil animal amounts, but no significant relationship with diversity. The contribution of soil animals to N mineralization was different for different ecosystems due to influences of complex factors including grazing, soil characteristics, the quality and amount of litter on N mineralization. Pinus sylvestris var. mongolica Litv. (Pinales: Pinaceae) is an excellent tree for soil and water conservation in Northeast China. The Honghua'erji area in Inner Mongolia is the "hometown of P. sylvestris var. mongolica", however, in recent years, coniferous diseases of P. sylvestris var. mongolica have frequently occurred here. During the investigation, it was found that some black spot needle blight had been observed in addition to the common blight caused by Sphaeropsis sapinea. From May to September 2020, black spot needle blight was found on hundreds of P. sylvestris var. mongolica trees in four forest farms, and the infection rate among the forests was 24.58 % (n=240). This disease first appeared on the upper part of the needles, and the needles then became withered and gradually showed light black spots, although they remained green. As the disease progressed, the needles eventually died and turned gray with many dark black spots. Fungal isolate named YJ-1 was obtained from infected needles of symptomatic pine trees, and a voucher specimen was deposited in Heilongjiang Province Key Laboratory of Forest Protection. Microscopic observation showed the conidia were 3-septate (4 cells) clavate spindles that measured 23.9 μm (20.8-25.9) × 5.9 μm (4.5-8.2) (n=50). The middle two cells were dark brown, and the septa were darker than the cells. Both apical and basal cells were hyaline. The apical cell had 2-4 appendages (mostly 3), and the basal cell had a truncate base (n=50). The cultural characteristics on potato dextrose agar medium were flat off-white and dense in 3-5 d. At approximately 5-7 d, the reverse side of the colony turned pale to slightly luteous. Superficial black acervuli were distributed in the center of the mature colonies after 10 d. Morphological, cultural and microscopic characteristics observed were similar of Heterotruncatella spartii (basionym: Truncatella spartii) reported by Hlaiem et al (2019). To further identify, total DNA was extracted and the internal transcribed spacer region (ITS-rDNA) was amplified by PCR using the primers ITS1/ITS4 and sequenced for BLASTn analysis and phylogenetic tree construction. The resulting 564 bp sequence (GenBank Accession No. OL662864) had 99.24% (521/525) to H. spartii MFLUCC 15-0537, with bootstrap support of at least 94% using the Neighbor-Joining algorithm by MEGA-X (Felsenstein, 1985). The fungus was identified as H. spartii based on morphology and molecular methods. A pathogenicity test was conducted by preparing a conidial suspension of 2.0 × 107 conidia/mL. The suspension was sprayed onto the needles of 20 pots of annual P. sylvestris ar. mongolica seedlings, and the control was sprayed with sterile water. Then the seedlings were placed in a constant temperature room at 25 °C. After 30 d, typical symptoms appeared on 11 inoculated needles, while the control needles remained symptomless. After 50 d, the re-isolation infection rate reached 66.7 %. The fungus present on the inoculated seedlings was morphologically identical to that originally observed on diseased pines, fulfilling Koch's postulates. The fungus was isolated from Spartium junceum for the first time and designated Truncatella spartii (Senanayake et al, 2015). It was then renamed H. spartii (Liu et al, 2019) and has been reported to infect P. pinea in Tunisia (Hlaiem et al, 2019). To our knowledge, this is the first report of H. spartii causing black spot needle blight on P. sylvestris var. mongolica in China and worldwide.
