With its awkward generic shifts, odd repetitions, confusing spatial dislocations, unstable characters and inconclusive supernatural horrors, I here argue, bilingual Danish author Isak Dinesen’s The Angelic Avengers weirds our experience of living in a familiar, predictable and rule-governed universe. In my analysis, I especially foreground Dinesen’s use of West Indian voodoo, which is a prominent weirding device largely overlooked by the novel’s relatively few critics. Dinesen, I argue, wrote her novel amidst a widespread international voodoo and zombie vogue, tapping into popular representations of Caribbean witchcraft in nonfiction, pulp fiction and film. In The Angelic Avengers, I argue, Dinesen appropriates voodoo themes and characters, to conjure the presence of ominous agencies that trouble enlightened reason’s ability to explain and master the world.

Examples of photoluminescence (PL) are being reported with increasing frequency in a wide range of organisms from diverse ecosystems. However, the chemical basis of this PL remains poorly defined, and our understanding of its potential ecological function is still superficial. Amongst mammals, recent analyses have identified free-base porphyrins as the compounds responsible for the reddish ultraviolet-induced photoluminescence (UV-PL) observed in the pelage of springhares and hedgehogs. However, the localization of the pigments within the hair largely remains to be determined. Here we use photoluminescence multispectral imaging emission and excitation spectroscopy to detect, map and characterize porphyrinic compounds in skin appendages in situ. We also document new cases of mammalian UV-PL caused by free-base porphyrins in distantly related species. Spatial distribution of the UV-PL is strongly suggestive of an endogenous origin of the porphyrinic compounds. We argue that reddish UV-PL is predominantly observed in crepuscular and nocturnal mammals because porphyrins are photodegradable. Consequently, this phenomenon may not have a specific function in intra- or interspecific communication but rather represents a byproduct of potentially widespread physiological processes.Co-first authors: Séverine Toussaint and Jasper Ponstein

Even though the central role of mechanics in cardiovascular system is widely recognized, estimating mechanical deformation and strains in-vivo remains an ongoing practical challenge. Herein, we present a semi-automated framework to estimate strains from four-dimensional (4D) echocardiography images and apply it to the aortic roots of patients with normal trileaflet aortic valves (TAV) and congenital bicuspid aortic valves (BAV). The method is based on fully nonlinear shell-based kinematics, which divides the strains into in-plane (shear and dilatational) and out-of-plane components. The results indicate that, even for size-matched non-aneurysmal aortic roots, BAV patients experience larger regional shear strains in their aortic roots. This elevated strains might be a contributing factor to the higher risk of aneurysm development in BAV patients. The proposed framework is openly available and applicable to any tubular structures.

Rapid and cost-effective retrieval of endogenous DNA from ancient specimens remains a limiting factor in palaeogenomic research. Many methods have been developed to increase ancient DNA yield, but modifications to existing protocols are often based on personal experience rather than systematic testing. Here, we present a new silica column-based extraction protocol, where optimizations were tested in controlled experiments. Using relatively well-preserved permafrost samples, we tested the efficiency of pretreatment of bone and tooth powder with a bleach wash and a predigestion step. We also tested the recovery efficiency of MinElute and QIAquick columns, as well as Vivaspin columns with two molecular weight cut-off values. Finally, we tested the effect of uracil-treatment with two different USER enzyme concentrations. We find that neither bleach wash combined with a predigestion step, nor predigestion by itself, significantly increased sequencing efficiency. Initial results, however, suggest that MinElute columns are more efficient for ancient DNA extractions than QIAquick columns, whereas different molecular weight cut-off values in centrifugal concentrator columns did not have an effect. Uracil treatments are effective at removing DNA damage even at concentrations of 0.15 U/µL (as compared to 0.3 U/µL) of ancient DNA extracts.

In this essay, I use the recent convergence of queer, feminist, and ecocritical perspectives under the heading of “queer ecology” to analyse how Danish bilingual author Isak Dinesen (real name: Karen Blixen) deploys breastfeeding in her Gothic story “The Caryatids: An Unfinished Story.” Queer ecology entails critiquing the assumption (or myth) that heterosexual identities, monogamous relationships, consanguineous kinship networks, and reproductive nuclear families are better, healthier, or more “natural” than other formations. I situate Dinesen in opposition to the tradition of Enlightenment “lactivism,” whose proponents (chief among them Jean-Jacques Rousseau) celebrated exclusive maternal breastfeeding within hetero-reproductive nuclear family structures. In my reading of “The Caryatids,” I place special emphasis on three Gothic-related female characters—the incestuous wife, the adulterous mother, and the “gypsy” witch—who breastfeed queerly, knowingly or unknowingly thwarting the demand that women nurse the monogamous and patriarchal bio-family order into existence. When removed from its sanctioned familial context, I argue, the lactating breast in Dinesen’s narrative proves a disorderly signifier of errancy and dissent, pointing towards the possibility of alternative, emergent, contingent, and potentially more sustainable configurations of gender, relationship, and community.

Key points Spatiotemporal contractility analysis of human induced pluripotent stem cell derived cardiomyocyte (hiPSC-CM) monolayers seeded on conventional, rigid surfaces (glass or plastic) revealed the presence of multiphasic contraction patterns across the monolayer with a high variability, despite action potentials recorded in the same areas being identical. These multiphasic patterns are not present in single cells, in detached monolayers or in monolayers seeded on soft substrates such as a hydrogel, where only 'twitch'-like transients are observed. HiPSC-CM monolayers that display a high percentage of regions with multiphasic contraction have significantly increased contractile duration and a decreased lusotropic drug response. There is no indication that the multiphasic contraction patterns are associated with significant activation of the stress-activated NPPA or NPPB signalling pathways. A computational model of cell clusters supports the biological findings that the rigid surface and the differential cell-substrate adhesion underly multiphasic contractile behaviour of hiPSC-CMs. Abstract Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CM) in monolayers interact mechanically via cell-cell and cell-substrate adhesion. Spatiotemporal features of contraction were analysed in hiPSC-CM monolayers 1) attached to glass or plastic (Young's modulus (E) >1 GPa), 2) detached (substrate-free) and 3) attached to a flexible collagen hydrogel (E = 22 kPa). The effects of isoprenaline on contraction were compared between rigid and flexible substrates. To clarify underlying mechanisms, further gene expression and computational studies were performed. HiPSC-CM monolayers exhibited multi-phasic contractile profiles on rigid surfaces in contrast to hydrogels, substrate-free cultures or single cells where only simple twitch-like time-courses were observed. Isoprenaline did not change the contraction profile on either surface, but its lusitropic and chronotropic effects were greater in hydrogel compared to glass. There was no significant difference between stiff and flexible substrates in regard to expression of the stress activated genes NPPA and NPPB. A computational model of cell clusters demonstrated similar complex contractile interactions on stiff substrates as a consequence of cell-to-cell functional heterogeneity. Rigid biomaterial surfaces give rise to unphysiological, multi-phasic contractions in hiPSC-CM monolayers. Here we used state-of-the-art spectroscopy and multispectral imaging techniques to document new cases of mammalian ultraviolet-induced PL (UV-PL) and to identify free-base porphyrins and natural derivatives as the organic compounds responsible for the reddish luminescence observed in the hairs and spines of distantly related species. All patients underwent a comprehensive eye examination at no cost, with referral to a university eye clinic if necessary. Demographic characteristics, past ocular history, reasons for attendance, and ophthalmic diagnoses were recorded. Attendance rates and treatment outcomes of patients referred to the university eye clinic were also reviewed.Results: We reviewed records of 360 GES patients (mean age 43 years, age range 1–79 years; 56% [200] male; 37% [133] non-English speakers). The most common reasons for attending were blurry vision (28% [101]), need for new glasses (22% [80]), and referral for a diabetic eye exam (18% [63]). The most common diagnosis made was refractive error (59% [214]), and vouchers for free spectacles were provided. One-third of diabetic patients had retinopathy (32% [38]). Glaucoma suspect (11% [40]), narrow angles (4% [13]), treatment-requiring diabetic eye disease (4% [14]), and visually-significant cataract (3% [11]) were diagnoses that most often prompted referral to the university clinic. In all, 114 patients were referred (32%), of whom 82 (72%) attended the follow-up visit. Other patients continued to receive longitudinal care through GES.Conclusion: Medical student-led outreach programs under the supervision of an attending ophthalmologist can deliver regular eye care to underserved communities while providing referrals to a university clinic for those with advanced disease. The immature electrophysiology of human-induced pluripotent stem cell-derived cardiomyocytes (hiCMs) complicates their use for therapeutic and pharmacological purposes. An insufficient inward rectifying current (IK1) and the presence of a funny current (if) cause spontaneous electrical activity. This study tests the hypothesis that the co-culturing of hiCMs with a human embryonic kidney (HEK) cell-line expressing the Kir2.1 channel (HEK-IK1) can generate an electrical syncytium with an adult-like cardiac electrophysiology. The mechanical activity of co-cultures using different HEK-IK1:hiCM ratios was compared with co-cultures using wildtype (HEK–WT:hiCM) or hiCM alone on days 3–8 after plating. Only ratios of 1:3 and 1:1 showed a significant reduction in spontaneous rate at days 4 and 6, suggesting that IK1 was influencing the electrophysiology. Detailed analysis at day 4 revealed an increased incidence of quiescent wells or sub-areas. Electrical activity showed a decreased action potential duration (APD) at 20% and 50%, but not at 90%, alongside a reduced amplitude of the aggregate AP signal. A computational model of the 1:1 co-culture replicates the electrophysiological effects of HEK–WT. The addition of the IK1 conductance reduced the spontaneous rate and APD20, 50 and 90, and minor variation in the intercellular conductance caused quiescence. In conclusion, a 1:1 co-culture HEK-IK1:hiCM caused changes in electrophysiology and spontaneous activity consistent with the integration of IK1 into the electrical syncytium. However, the additional electrical effects of the HEK cell at 1:1 increased the possibility of electrical quiescence before sufficient IK1 was integrated into the syncytium.

The most common etiology of an isolated abducens nerve palsy in an older adult with vasculopathic risk factors is presumed microvascular ischemia. Because the diagnosis of microvascular ischemia in this setting would not alter patient management, there is debate as to whether further workup is warranted under these circumstances. In this report, we describe a fascicular sixth nerve palsy as the initial presenting sign of metastatic ovarian carcinoma, and we highlight the importance of considering additional workup in select cases of isolated abducens nerve palsies.

Introduction Current ophthalmologic training in medical school is inadequate in preparing medical students to handle basic eye complaints as nonophthalmology residents. Most medical students are uncomfortable performing eye examinations, but increased ophthalmology training improves confidence in this area. The University of Pittsburgh School of Medicine (UPSOM) teaches students the basics of ophthalmology with a required 1-week rotation during the 1-month specialty care clerkship (SCC), providing students with skills to perform rudimentary eye examinations as nonophthalmology providers. Methods Within a 1-week ophthalmology rotation, we developed a series of interactive case-based teaching sessions, handouts, and homework that accompanied clinical instruction to familiarize third- and fourth-year medical students with ophthalmic equipment, terminology, diagnosis, and management. Of learners, 67 (roughly 11 per cohort) rotated on six consecutive SCCs beginning in May 2019. All learners completed an in-house exam and received resident clinical evaluations at the end of their rotation. Results Of the 64 participants who responded to the survey, 100% rated the quality of teaching sessions outstanding or good, and 83% of students strongly agreed or agreed with the statement, “I believe the overall teaching in the ophthalmology clinical settings was good quality.” The average clinical and exam score for ophthalmology over 6 months was 4.5 out of 5, and 83% respectively. Discussion Generally positive student feedback as well as high clinical and exam scores suggested that the required UPSOM ophthalmology clerkship was both engaging and effective. This course can be easily adapted to teach students at other medical institutions.

Handling and transport of contaminated soil from industrial sites in Denmark requires classification based on concentrations of selected metals and organic contaminants. Reliable soil classification is needed for defensible remedial decision-making. Today’s sampling process in Denmark is based on grab sampling of prescribed standard volumes of soil; 30 tons is typically used as the basic sampling Decision Unit. Soil classification follows a number of varying systems, but classification into five classes (class 0 to class 4) based on analytical results from sub-samples of 50 g is the most common. In this study we investigate the sampling uncertainty obtained by sampling of > 1800 samples at a former industrial site in Copenhagen, Denmark. The aim of the study was to conduct a critical assessment of the current sampling strategy by determination of soil classification errors obtained for duplicate primary samples collected from the same truck- load of soil, but with different distances from the original primary sample. It is also discussed which contaminants are the major parameters responsible for final soil classification designations. The original primary samples for the soil classification were taken at the centre of a 7 × 7 m grid for every 33 cm to a depth of 1 m or 3 m. In addition - for approximately half of the positions - samples were taken at a distance of 1, 2 or 3 m from the original sample. The classifications for the original samples were compared to the classifications for the additional samples in the same Decision Unit (7 × 7 × 0.33 m box) and from duplicate original samples. The results show that at least 50 % of all samples were misclassified, 20 % were mis-classified by two or more classes. This study demonstrates that the risk of misclassification is highest for less mobile parameters, metals and PAHs compared to the volatile organic solvents.

Introduction Ophthalmology education during medical school is often very limited. To provide exposure to areas beyond its standard curriculum, the University of Pittsburgh School of Medicine offers mini-elective courses in various disciplines. We developed such a course to provide instruction in the basics of clinical ophthalmology to interested preclinical medical students. Methods First- and second-year medical students electively enrolled in our course (mean number of students per year = 12), which included four sessions combining didactics and hands-on learning. Additionally, each student individually spent time with an ophthalmologist in the operating room. Our course was held each year from 2015 to 2019. Results Participants completed pre- (n = 25) and postsurveys (n = 20), reflecting increased comfort with the ophthalmologic history and physical examination. In 2019, participants also completed pre- and posttests, demonstrating increased knowledge of ophthalmology. Discussion The Ophthalmology Mini-Elective is a unique educational tool that introduces the principles of ophthalmology to preclinical medical students, addressing an area of medicine that is generally minimally included in the required curriculum.

Purpose To compare outcomes between 2 nonvalved glaucoma drainage devices (GDDs) used to treat refractory glaucoma or in patients with neovascular/uveitic glaucoma likely to be poorly responsive to less aggressive therapies. Design Retrospective, nonrandomized, multicenter comparative study. Participants A total of 117 eyes from 117 patients. Methods Retrospective chart review of patients who underwent implantation of the Baerveldt (BGI) (Abbott Medical Optics, Abbott Park, IL) or the Molteno3 glaucoma implant (MGI) (Molteno Ophthalmic Limited, Dunedin, New Zealand). Noninferiority of the MGI versus the BGI was tested with Cox and mixed-effects regression models. Interventions in each group were analyzed with chi-square tests. Main Outcome Measures The primary outcome was time until device failure, defined as intraocular pressure (IOP) >21 mmHg or a reduction Results The MGI could not be deemed noninferior to the BGI with regard to time until device failure (hazard ratio [HR], 0.83; confidence interval [CI], 0.41–1.65). The MGI was noninferior to the BGI when comparing postoperative IOP, a difference of −0.40 mmHg (95% CI, −1.74–0.93). The MGI needed 2% fewer medications (ratio of 0.98, 95% CI, 0.79–1.22), but noninferiority could not be claimed. With regard to VA, the MGI's mean was 0.10 logarithm of the minimum angle of resolution (logMAR) higher (95% CI, −0.01–0.21), but noninferiority testing was again inconclusive. Intraoperative time for the MGI was 15.7 minutes shorter versus the 350 mm2 plate size BGI (P Conclusions The MGI was noninferior to the BGI in lowering IOP. Differences in time until device failure, VA outcomes, and medication use were inconclusive. The MGI required more secondary operative interventions. The MGI required less time to implant than the BGI's 350 mm2 plate size implant. Overall, the use of both GDDs is justifiable to lower IOP when more conservative management has failed.

The key parameter during purification of enzymes is the strength of the intermediates and products. Employing near infrared spectroscopy, enzyme activity of the concentrate can be obtained much faster than by traditional off-line analysis in a central laboratory. This paper describes the development of a monitoring system for the enzyme activity in a recovery plant and provides a comparison between two real-time probe setups (on-line v. in-line) in a full scale application. The focus in the paper will be on the chemometric calibration development plus validation and the application of the models for monitoring purposes. For this investigation, four different types of industrial enzymes have been sampled over a period of ten months. Real-time output of the partial least squares regression models was used along with conventional process data generated to evaluate the pros and cons of the in-line and on-line setup. Both implementations deliver good results in monitoring the ultrafiltration process, but problems (precipitation/phase transition) were occasionally encountered for the on-line arrangement.

Specific strength training is shown to relieve neck pain in office workers. The purpose of this study is to evaluate the effectiveness of specific strength training in women with severe neck pain and to analyze the dose-response relationship between training adherence and pain reduction. One hundred eighteen untrained women with severe neck pain (>30 mm VAS pain) were included from a larger study, in which the subjects were randomized to 20-week specific strength training for the neck/shoulders or to a control group. In the intention-to-treat analysis, the training group experienced greater pain relief than the control group (p < 0.01). Participants who adhered "per protocol" decreased pain by 35 mm VAS (95% confidence interval: -26 to -44) from baseline to follow-up corresponding to a 70% reduction. In the dose-response analyses, participants with medium and high training adherence showed better pain relief than the control group and those with low adherence (p < 0.0001). The decrease from baseline in the medium and high adherence groups was 37 mm VAS (28-46 mm) and 33 mm VAS (24-43 mm), respectively. Specific strength training reduces pain intensity in women with severe neck pain, and 1-2 training sessions per week for 20 weeks (∼30 training sessions) seems sufficient for optimal pain relief.

We studied geographical and temporal body size trends among 169 adult museum specimens of the Eurasian otter (Lutra lutra) collected in Sweden between 1962 and 2008, whose sex, year of collection, and locality were known. Skull size and body mass increased significantly in relation to the year of collection, and skull size (but not body mass) was significantly and negatively related to latitude, contrasting Bergmann’s rule and the trend found for Norwegian otters. Latitudinal differences in body size between the two countries may be due to differences in food availability. The temporal increase in body size among Swedish otters resembled that observed for Norway otters, though Swedish otters are smaller with respect to their Norwegian counterparts. Latitude and year represent a combination of environmental factors, including ambient temperature in the year of collection as well as the number of days of ice coverage. We replaced the above factors with mean annual temperature or the number of days of ice coverage, and found that each of these factors explains a similar proportion of the variation in body size as did latitude and year. We hypothesize that this temporal increase in body size is related to a combination of factors, including reduced energy expenditure resulting from increasing ambient temperature, and increased food availability from longer ice-free periods.

Mass spectrometry-based proteomics critically depends on algorithms for data interpretation. A current bottleneck in the rapid advance of proteomics technology is the closed nature and slow development cycle of vendor-supplied software solutions. We have created an open source software environment, called MSQuant, which allows visualization and validation of peptide identification results directly on the raw mass spectrometric data. MSQuant iteratively recalibrates MS data thereby significantly increasing mass accuracy leading to fewer false positive peptide identifications. Algorithms to increase data quality include an MS(3) score for peptide identification and a post-translational modification (PTM) score that determines the probability that a modification such as phosphorylation is placed at a specific residue in an identified peptide. MSQuant supports relative protein quantitation based on precursor ion intensities, including element labels (e.g., (15)N), residue labels (e.g., SILAC and ICAT), termini labels (e.g., (18)O), functional group labels (e.g., mTRAQ), and label-free ion intensity approaches. MSQuant is available, including an installer and supporting scripts, at http://msquant.sourceforge.net .

The Norwegian novelist Knut Hamsun's erratic behavior during the 1930s and early 1940s, when he publicly endorsed the German war effort, has tarnished the reputation of his Nobel Prize-winning novel Growth of the Soil (1917) and in some circles stigmatized it as "reactionary," "antimodern" and even "proto-fascist" text. Placing Hamsun within an international context and drawing on both critical theory and revisionist scholarship into the back-to-the-land movement, I find instead that contemporary land and life reformers' conflicted attempts to map out "alternative modernities" are mirrored in Growth of the Soil 's fraught negotiation with modern technology, producing a complex dialectic of critique and affirmation that cannot simply be dismissed as backward-looking. Rather than directly prefiguring the political mobilization of retrograde anxieties in the mass movements of the 1930s, Hamsun's "agricultural book" signifies ambiguously within a heterogeneous space of conflicting discourses and practices directed against and towards the modern world.

The Eurasian lynx (Lynx lynx) is a common predator of both roe deer (Capreolus capreolus) and reindeer (Rangifer tarandus) in Sweden. We investigated the influence of prey availability, latitude, sex, and age on body size and body mass variation of the Eurasian lynx in Sweden, using data from 243 specimens whose locality of capture, year of capture, sex, and age were known. We found that both body size and body mass of the lynx in Sweden are mainly affected by the lynx sex and age but also by the availability of prey during the first year of life. Body size and body mass of lynx as well as the density of roe deer increased from Central Sweden to South. Furthermore, body size and body mass of lynx increased from Central Sweden to North (i.e. within the reindeer husbandry area). Lynx body size was slightly smaller within the reindeer husbandry area (approximately north of latitudes 62°–63°N) compared to outside, probably because reindeer are more difficult prey to hunt, as well as being migratory and thus an unpredictable prey for the Eurasian lynx compared to the non-migratory roe deer. Our results support a growing body of evidence showing that food availability at growth has a major effect on body size of animals.

The past quarter century has seen the deployment of ethnographic methods in business grow from a curiosity to a prerequisite for success. But in the process, the outcomes of ethnographic research–customer empathy, strategic directions, lasting market insights that shape design–have not been adopted at the same rate. The hand-off from ethnographers to designers and business decision-makers is the biggest challenge to success. The time has come for ethnographers to again reframe their role within business. Rather than acting as interpreters between the lives of ordinary people and the companies who serve them, ethnographers have the opportunity to instead help the entire business organization to gather a clear sense of its customers' lives. Ethnographers need to switch from being gurus of customer experience to being guides who take everyone in the company into the outside world.

Mice have close genetic/physiological relationships to humans, breed rapidly, and can be genetically modified, making them the most used mammal in biomedical research. Because the red blood cell (RBC) is the sole gas transporter in vertebrates, diseases of the RBC are frequently severe; much research has therefore focused on RBC and cardiovascular disorders of mouse and humans. RBCs also host malaria parasites. Recently we presented an in-depth proteome for the human RBC. Here we present directly comparable data for the mouse RBC as membrane-only, soluble-only, and combined membrane-bound/soluble proteomes (comprising, respectively, 247, 232, and 165 proteins). All proteins were identified, validated, and categorized in terms of subcellular localization, protein family, and function, and in comparison with the human RBC, were classified as orthologs, family-related, or unique. Splice isoforms were identified, and polypeptides migrating with anomalous apparent molecular weights were grouped into putatively ubiquitinated or partially degraded complexes. Overall there was close concordance between mouse and human proteomes, confirming the unexpected RBC complexity. Several novel findings in the human proteome have been confirmed here. This comparison sheds light on several open issues in RBC biology and provides a departure point for more comprehensive understanding of RBC function.

Didactic data sets representing a range of real-world processes are used to illustrate “how to do” representative process sampling and process characterisation. The selected process data lead to diverse variogram expressions with different systematics (no range vs. important ranges; trends and/or periodicity; different nugget effects and process variations ranging from less than one lag to full variogram lag). Variogram data analysis leads to a fundamental decomposition into 0-D sampling vs. 1-D process variances, based on the three principal variogram parameters: range, sill and nugget effect. The influence on the variogram from significant trends and outliers in the original data series receive special attention, due to their critical adverse effects. We highlight problem-dependent interpretation of variographic analysis a.o. the problem-dependent background for periodicities and trends. All presented cases of variography either solved the initial problems or served to understand the reasons and causes behind the specific process structures revealed in the variograms.
