The Anglo-Arabian horse breed (AA) is the result of crossing between Thoroughbred and Arabian horses renowned for versatility in show-jumping and racing. Southwest France and Sardinia in Italy are considered the historical nucleus of European AA breeding. Since little is known on genetic variability in the AA breed, especially those bred in the Sardinia region, this study investigates: i) inbreeding levels via pedigree and SNP genotyping; ii) runs of homozygosity (ROH) and selection signatures; iii) effective population size; and iv) distinctiveness of AA compared to Arabian and Thoroughbred horses. The ROH revealed an excessive amount of homozygosity, with a high kurtosis value observed in several chromosomes and an average inbreeding based on ROH of 21.1%. This latter value is higher compared to what was found based on pedigree data, highlighting the importance of complete pedigree records when estimating inbreeding based on genealogical data. The genomic inbreeding in AA horses was higher compared to other Italian horse breeds but lower than what was found in pure Thoroughbred and Arabian horses. This result suggests that the selection process for AA horses, may have prioritised racing ability over breed preservation which is the main goal in most local Italian breeds. The reduction in effective population size found in the latest 20 generations corroborates the potential presence of genetic diversity reduction due to selection in the AA breed. Remarkably, a large ROH island was identified on ECA17, which overlapped with two QTLs associated with racing performance.

Abstract Background Goats were domesticated in the Fertile Crescent about 10,000 years before present (YBP) and subsequently spread across Eurasia and Africa. This dispersal is expected to generate a gradient of declining genetic diversity with increasing distance from the areas of early livestock management. Previous studies have reported the existence of such genetic cline in European goat populations, but they were based on a limited number of microsatellite markers. Here, we have analyzed data generated by the AdaptMap project and other studies. More specifically, we have used the geographic coordinates and estimates of the observed (Ho) and expected (He) heterozygosities of 1077 European, 1187 African and 617 Asian goats belonging to 38, 43 and 22 different breeds, respectively, to find out whether genetic diversity and distance to Ganj Dareh, a Neolithic settlement in western Iran for which evidence of an early management of domestic goats has been obtained, are significantly correlated. Results Principal component and ADMIXTURE analyses revealed an incomplete regional differentiation of European breeds, but two genetic clusters representing Northern Europe and the British-Irish Isles were remarkably differentiated from the remaining European populations. In African breeds, we observed five main clusters: (1) North Africa, (2) West Africa, (3) East Africa, (4) South Africa, and (5) Madagascar. Regarding Asian breeds, three well differentiated West Asian, South Asian and East Asian groups were observed. For European and Asian goats, no strong evidence of significant correlations between Ho and He and distance to Ganj Dareh was found. In contrast, in African breeds we detected a significant gradient of diversity, which decreased with distance to Ganj Dareh. Conclusions The detection of a genetic cline associated with distance to the Ganj Dareh in African but not in European or Asian goat breeds might reflect differences in the post-domestication dispersal process and subsequent migratory movements associated with the management of caprine populations from these three continents.

Objectives To investigate pregnancy outcomes in women with autoimmune rheumatic diseases (ARD) in the Italian prospective cohort study P-RHEUM.it.Methods Pregnant women with different ARD were enrolled for up to 20 gestational weeks in 29 Rheumatology Centres for 5 years (2018–2023). Maternal and infant information were collected in a web-based database.Results We analysed 866 pregnancies in 851 patients (systemic lupus erythematosus was the most represented disease, 19.6%). Maternal disease flares were observed in 135 (15.6%) pregnancies. 53 (6.1%) pregnancies were induced by assisted reproduction techniques, 61 (7%) ended in miscarriage and 11 (1.3%) underwent elective termination. Obstetrical complications occurred in 261 (30.1%) pregnancies, including 2.3% pre-eclampsia. Two cases of congenital heart block were observed out of 157 pregnancies (1.3%) with anti-Ro/SSA. Regarding treatments, 244 (28.2%) pregnancies were treated with glucocorticoids, 388 (44.8%) with hydroxychloroquine, 85 (9.8%) with conventional synthetic disease-modifying anti-rheumatic drugs and 122 (14.1%) with biological disease-modifying anti-rheumatic drugs. Live births were 794 (91.7%), mostly at term (84.9%); four perinatal deaths (0.5%) occurred. Among 790 newborns, 31 (3.9%) were small-for-gestational-age and 169 (21.4%) had perinatal complications. Exclusive maternal breast feeding was received by 404 (46.7%) neonates. The Edinburgh Postnatal Depression Scale was compiled by 414 women (52.4%); 89 (21.5%) scored positive for emotional distress.Conclusions Multiple factors including preconception counselling and treat-to-target with pregnancy-compatible medications may have contributed to mitigate disease-related risk factors, yielding limited disease flares, good pregnancy outcomes and frequency of complications which were similar to the Italian general obstetric population. Disease-specific issues need to be further addressed to plan preventative measures.

ABSTRACT: The objectives of this study were to explore the use of Fourier-transform infrared (FTIR) spectroscopy on individual sheep milk samples for predicting cheese-making traits, and to test the effect of the farm variability on their prediction accuracy. For each of 121 ewes from 4 farms, a laboratory model cheese was produced, and 3 actual cheese yield traits (fresh cheese, cheese solids, and cheese water) and 4 milk nutrient recovery traits (fat, protein, total solids, and energy) in the curd were measured. Calibration equations were developed using a Bayesian approach with 2 different scenarios: (1) a random cross-validation (80% calibration; 20% validation set), and (2) a leave-one-out validation (3 farms used as calibration, and the remaining one as validation set) to assess the accuracy of prediction of samples from external farms, not included in calibration set. The best performance was obtained for predicting the yield and recovery of total solids, justifying for the practical application of the method at sheep population and dairy industry levels. Performances for the remaining traits were lower, but still useful for the monitoring of the milk processing in the case of fresh curd and recovery of energy. Insufficient accuracies were found for the recovery of protein and fat, highlighting the complex nature of the relationships among the milk nutrients and their recovery in the curd. The leave-one-out validation procedure, as expected, showed lower prediction accuracies, as a result of the characteristics of the farming systems, which were different between calibration and validation sets. In this regard, the inclusion of information related to the farm could help to improve the prediction accuracy of these traits. Overall, a large contribution to the prediction of the cheese-making traits came from the areas known as “water” and “fingerprint” regions. These findings suggest that, according to the traits studied, the inclusion of water regions for the development of the prediction equation models is fundamental to maintain a high prediction accuracy. However, further studies are necessary to better understand the role of specific absorbance peaks and their contribution to the prediction of cheese-making traits, to offer reliable tools applicable along the dairy ovine chain.

This work aimed to assess the variability of casein genes in a population of 153 bucks and 825 lactating does of the Sarda breed, and to perform association analysis between polymorphic sites and milk yield and composition traits. To genotype the casein genes, we chose an SNP panel including 44 SNPs mapping to the four casein genes CSN1S1, CSN2, CSN1S2, and CSN3. Genotyping (made by KASP™ genotyping assay, based on competitive allele-specific PCR) revealed the high variability of the Sarda goat, and haplotype analysis revealed linkage disequilibrium (LD) between CSN1S1 and CSN2 genes, in addition to two LD blocks within the CSN1S2 and two LD blocks within the CSN3 gene, in bucks and does. Association analysis revealed that variability at all four casein genes was associated with milk protein content, total solids, and milk energy. The three Ca-sensitive casein genes were associated with lipid content, and CSN1S2 showed a unique pattern, with intron variants associated with milk yield, in addition to milk pH, NaCl, and SCS (Somatic Cell Score). This information might prove useful in selection schemes and in future investigations aiming to better understand the biology of lactation, and the direct link between genotype and phenotype.

ABSTRACT: At the global level, the quantity of goat milk produced and its gross production value have increased considerably over the last 2 decades. Although many scientific papers on this topic have been published, few studies have been carried out on bulk goat milk samples. The aim of the present study was to investigate in the field the effects of farming system, breed type, individual flock, and stage of production on the composition, coagulation properties (MCP), curd firming over time parameters (CFt), predicted cheese yield (CY%), and nutrient recovery traits (REC) of 432 bulk milk samples from 161 commercial goat farms in Sardinia, Italy. We found that the variance due to individual flock was of the same order as the residual variance for almost all composition and cheesemaking traits. With regard to the fixed effects, the effect of farming system on bulk milk variability was not highly significant for the majority of traits (it was lower than individual flock), whereas the effects of breed type and stage of production were much higher. More specifically, the intensive farms produced milk with the best concentrations of almost all constituents, whereas extensive farms exhibited faster rennet coagulation times, a slower rate of curd firming, lower potential curd firmness, and lower percentages of fat and energy recoveries in the fresh curd. Farms rearing the local breed, Sarda, alone or together with the Maltese breed, produced milk with the best concentrations of fat and protein, superior curd firmness, and better predicted percentage of fresh curd (CYCURD) and recovery traits. The results show the potential of both types of breed, either for their quantitative (specialized breeds) or their qualitative (local breeds) attributes. As expected, the concentrations of fat, protein fractions, and lactose were influenced by the stage of production, with samples collected in the early stage of production (in February and March) having a greater quantity of the main constituents. Somatic cells reached the highest levels in the late stage of production, which corresponds to the goats' advanced stage of lactation (June–July), although no differences were present in the logarithmic bacterial counts between the early and late stages. Regarding cheesemaking potential, bulk milk samples of the late stage were characterized by delayed rennet coagulation and curd firming times, the lowest values of curd firmness, and a general reduction in CY%, and REC traits. In conclusion, we highlight several issues regarding the effects of the most important sources of variation on bulk goat milk, and point to some critical factors relevant for improving dairy goat farming and milk production.

ABSTRACT: The objective of this study was to develop formulas based on milk composition of individual goat samples for predicting cheese yield (%CY) traits (fresh curd, milk solids, and water retained in the curd). The specific aims were to assess and quantify (1) the contribution of major milk components (fat, protein, and casein) and udder health indicators (lactose, somatic cell count, pH, and bacterial count) on %CY traits (fresh curd, milk solids, and water retained in the curd); (2) the cheese-making method; and (3) goat breed effects on prediction accuracy of the %CY formulas. The %CY traits were analyzed in duplicate from 600 goats, using an individual laboratory cheese-making procedure (9-MilCA method; 9 mL of milk per observation) for a total of 1,200 observations. Goats were reared in 36 herds and belonged to 6 breeds (Saanen, Murciano-Granadina, Camosciata delle Alpi, Maltese, Sarda, and Sarda Primitiva). Fresh %CY (%CYCURD), total solids (%CYSOLIDS), and water retained (%CYWATER) in the curd were used as response variables. Single and multiple linear regression models were tested via different combinations of standard milk components (fat, protein, casein) and indirect udder health indicators (UHI; lactose, somatic cell count, pH, and bacterial count). The 2 %CY observations within animal were averaged, and a cross-validation (CrV) scheme was adopted, in which 80% of observations were randomly assigned to the calibration (CAL) set and 20% to the validation (VAL) set. The procedure was repeated 10 times to account for sampling variability. Further, the model presenting the best prediction accuracy in CrV (i.e., comprehensive formula) was used in a secondary analysis to assess the accuracy of the %CY predictive formulas as part of the laboratory cheese-making procedure (within-animal validation, WAV), in which the first %CY observation within animal was assigned to CAL, and the second to the VAL set. Finally, a stratified CrV (SCrV) was adopted to assess the %CY traits prediction accuracy across goat breeds, again using the best model, in which 5 breeds were included in CAL and the remaining one in the VAL set. Fitting statistics of the formulas were assessed by coefficient of determination of validation (R2VAL) and the root mean square error of validation (RMSEVAL). In CrV, the formula with the best prediction accuracy for all %CY traits included fat, casein, and UHI (R2VAL = 0.65, 0.96, and 0.23 for %CYCURD, %CYSOLIDS, and %CYWATER, respectively). The WAV procedure showed R2VAL higher than those obtained in CrV, evidencing a low effect of the 9-MilCA method and, indirectly, its high repeatability. In the SCrV, large differences for %CYCURD and %CYWATER among breeds evidenced that the breed is a fundamental factor to consider in %CY predictive formulas. These results may be useful to monitor milk composition and quantify the influence of milk traits in the composite selection indices of specific breeds, and for the direct genetic improvement of cheese production.

ABSTRACT: Bovines produce about 83% of the milk and dairy products consumed by humans worldwide, the rest represented by bubaline, caprine, ovine, camelid, and equine species, which are particularly important in areas of extensive pastoralism. Although milk is increasingly used for cheese production, the cheese-making efficiency of milk from the different species is not well known. This study compares the cheese-making ability of milk sampled from lactating females of the 6 dairy species in terms of milk composition, coagulation properties (using lactodynamography), curd-firming modeling, nutrients recovered in the curd, and cheese yield (through laboratory model-cheese production). Equine (donkey) milk had the lowest fat and protein content and did not coagulate after rennet addition. Buffalo and ewe milk yielded more fresh cheese (25.5 and 22.9%, respectively) than cow, goat, and dromedary milk (15.4, 11.9, and 13.8%, respectively). This was due to the greater fat and protein contents of the former species with respect to the latter, but also to the greater recovery of fat in the curd of bubaline (88.2%) than in the curd of camelid milk (55.0%) and consequent differences in the recoveries of milk total solids and energy in the curd; protein recovery, however, was much more similar across species (from 74.7% in dromedaries to 83.7% in bovine milk). Compared with bovine milk, the milk from the other Artiodactyla species coagulated more rapidly, reached curd firmness more quickly (especially ovine milk), had a more pronounced syneresis (especially caprine milk), had a greater potential asymptotical curd firmness (except dromedary and goat milk), and reached earlier maximum curd firmness (especially caprine and ovine milk). The maximum measured curd firmness was greater for bubaline and ovine milk, intermediate for bovine and caprine milk, and lower for camelid milk. The milk of all ruminant species can be used to make cheese, but, to improve efficiency, cheese-making procedures need to be optimized to take into account the large differences in their coagulation, curd-firming, and syneresis properties.

Abstract The aim of this research was to characterize the genetic diversity of the Sarda (Sa, n = 131), Sardo Bruna (SB, n = 44) and Sardo Modicana (SM, n = 26) cattle breeds, reared in the island of Sardinia (Italy). A portion of the mitochondrial DNA hypervariable region was sequenced, in order to identify a potential signature of African introgression. The FST coefficients among populations ranged between 0.056 for Sa vs SB and 0.167 for SB vs SM. AMOVA analysis indicated there was a significant differentiation of the three breeds, although most of diversity was gathered at the within-breed level. The Median Joining Network of the Sardinian sequences showed a potential founder effect signature. A MJ network including Sardinian cattle plus African, Italian, Iberian and Asian sequences, revealed the presence of haplogroup T3, already detected in Sa cattle, and the presence of Hg T1 and Hg T1′2′3, in Sa and SB. The presence of a private haplotype belonging to haplogroup T1, which is characteristic of African taurine breeds, may be due to the introgression of Sardinian breeds with African cattle, either directly (most probable source: North African cattle) or indirectly (through a Mediterranean intermediary already introgressed with African blood).

There is evidence that Murciano Granadina (MG), the most important caprine dairy breed in Spain, has been introgressed by African goats, but the precise geographic origin of such introgression has not been identified yet. Moreover, an accurate estimate of the magnitude of this African introgression is lacking, since current estimates are based on small numbers of sampled individuals. The aim of our work was to tackle these two issues by genotyping 500 MG goats with the Goat SNP50 BeadChip and comparing their genotypes with those of reference populations from Spain (Bermeya), France (Saanen), Morocco (Barcha, Draa, Ghazalia, Noire de Atlas, Nord, Moroccan), Egypt (Barki, Oasis, Saidi), Algeria (Arabia, Makatia, M'Zabite, Kabyle), Tunisia (Tunisian native breeds) and Sudan (Desert, Nilotic, Taggar). The population of 500 MG goats was subdivided into 10 datasets of 50 individuals to ensure that sample sizes of the target (MG) and reference populations are balanced. Performance of an unsupervised ADMIXTURE analysis demonstrated that MG goats have a North African ancestry, with an average proportion of 4.4 ± 2.3%. Next, we did a supervised ADMIXTURE analysis that revealed that the Moroccan genetic component reaches a proportion of 4.01 ± 3.9% in MG goats, while the Algerian (0.001 ± 0.001%), Egyptian (0.2 ± 0.1%), Sudanese (0.1 ± 0.1%) and Tunisian (0.3 ± 0.4%) components are present in extremely small proportions. The historical circumstances of this introgression event are currently unknown, but several plausible scenarios are outlined. Moreover, our results show considerable inter‐individual heterogeneity regarding the magnitude of the Moroccan introgression of MG goats (0%– 12% depending on the MG data set under analysis). This result implies that reliable estimates about the introgression of autochthonous livestock by exotic breeds can only be obtained by extensively sampling target populations. Several local sheep breeds of the Alpine regions are in danger of extinction as a result of mountain farms being abandoned. Three fattening trials were carried out on 4 breeds of the Veneto region (Foza, Lamon, Alpagota, and Brogna) on a total of 115 ewes and suckling lambs, weaned lambs and heavy lambs slaughtered at 1356 ± 267, 104 ± 24, 219 ± 26, and 342 ± 14 d of age, respectively. The results confirmed that the adoption of total mixed diets resulted in good in vivo and post mortem performances of culled ewes (64 kg slaughter weight, 48% dressing percentage), suckling lambs (21 kg, 50%), weaned lambs (30 kg, 42%) and heavy lambs (61 kg, 48%). Carcass compactness and fatness increased with the age of the lambs, whereas the proportions of lean meat and bone decreased. Meat lightness and cooking losses decreased with age, whereas redness and chroma increased. The meat quality of culled ewes was like that of heavy lambs. Carcass conformation and composition (aside from size traits), and the quality of the meat differed little among the four Alpine breeds. These results show that local breeds are potentially valuable sources of lamb and mutton meat across different seasons, paving the way for strategies to develop and promote the meat with the involvement of high-quality restaurants and meat retailers.Highlights Compared to outdoor grazing, the adoption of indoor diets resulted in good in vivo and post-mortem performances of ewes, suckling lambs and heavy lambs. Autochthonous Alpine breeds are potentially valuable sources of high-quality lamb and mutton across different seasons of the year. This potential could be exploited in developing a prestigious product aimed at high quality restaurants and butchers. The subsequent demand for the product would, in turn, contribute to the survival of the local sheep breeds of Alpine regions.

To promote local food products could be a valuable strategy for preserving autochthonous sheep breeds. A total of 115 lambs and ewes of local sheep breeds (Alpagota, Bogna, Foza and Lamon) of the Veneto region were used to evaluate the effect of different feeding systems on the production and quality of lambs and mutton meat. We carried out three fattening trials involving 24 ewes and their 31 suckling lambs, 24 weaned lambs, and 24 heavy lambs, fed indoor diets with or without a rumen-protected conjugated linoleic acids (rp-CLA) supplement (C18:2c9,t11 and C18:2t10,c12) to improve the nutritional value of the meat. Controls were a group of 12 pasture-fed weaned lambs. The weaned lambs at pasture had good fattening performances (33 kg live-weight at slaughter, 45% dressing percentage), whereas lambs fattened indoors on hay and some concentrates had lower weight (28 kg) and dressing percentages (41%), and require improvements to their feed. Feeding the animals on total mixed diets resulted in good in vivo and post mortem performances of culled ewes (64 kg slaughter weight, 48% dressing percentage), suckling lambs (21 kg, 50%) and heavy lambs (61 kg, 48%), showing that valuable supplies of lamb and mutton meat can be obtained across different seasons. Supplementing indoor diets with rp-CLA does not modify the animals’ performances nor the organoleptic traits of meat.Highlights Intensive indoor fattening allows to obtain suckling, weaned and heavy lambs and mutton from culled ewes all-year-round beyond the seasonal supplies of traditional feeding system. The supplementation of indoor diets with rp-CLA does not modify the animals’ performances, slaughter traits or meat organoleptic traits.

Objectives To describe the outcomes of pregnancies in antiphospholipid antibody (aPL)-positive patients since the inception of the AntiPhospholipid Syndrome Alliance for Clinical Trials and InternatiOnal Networking Registry.Methods We identified persistently aPL-positive patients recorded as ‘pregnant’ during prospective follow-up, and defined ‘aPL-related outcome’ as a composite of: (1) Preterm live delivery (PTLD) at or before 37th week due to pre-eclampsia (PEC), eclampsia, small-for-gestational age (SGA) and/or placental insufficiency (PI); or (2) Otherwise unexplained fetal death after the 10th week of gestation. The primary objective was to describe the characteristics of patients with and without aPL-related composite outcomes based on their first observed pregnancies following registry recruitment.Results Of the 55 first pregnancies observed after registry recruitment among nulliparous and multiparous participants, 15 (27%) resulted in early pregnancy loss

Abstract Background Despite conflicting results, considerable evidence suggests the association between single nucleotide polymorphisms in MTHFR, XRCC1 and OGG1 genes and, risk of developing breast cancer. Here a case-control study is reported, including 135 breat cancer patients and 112 healthy women, all representative of Northern Sardinian population. Methods Polymerase chain reaction/restriction fragment length polymorphism method was used to determine the genotypes of five polymorphisms: MTHFR C677T (rs1801133) and A1298C (rs1801131), XRCC1 Arg194Trp (rs1799782) and Arg399Gln (rs25487) and OGG1 Ser326Cys (rs1052133). Allelic, genotypic and haplotype association analyses with disease risk and clinicopathological parameters were performed. Results A nominally significant association with breast cancer risk was observed for MTHFR C677T polymorphism heterozygous genotype in the codominant model (OR: 0.57, 95% CI: 0.32–1.00, p = 0.049) and for Cys/Cys genotype of the OGG1 Ser326Cys polymorphism in the recessive model (OR: 0.23, 95% CI: 0.05–1.11, p = 0.0465). No significant differences were found at genotype-level for A1298C polymorphism of the MTHFR gene and Arg194Trp and Arg399Gln of the XRCC1 gene. Furthermore, the OGG1 and XRCC1 rs25487 polymorphisms were nominally associated with PgR, Her2 status and with sporadic breast cancer, respectively. Conclusions Based on genetic characteristics of individuals included in this study, results suggest that MTHFR CT and OGG1 Cys/Cys genotypes have a protective effect that may have an influence on breast cancer risk in a representative Northern Sardinian population.

The infrared spectrum of bovine milk is used to predict many interesting traits, whereas there have been few studies on goat milk in this regard. The objective of this study was to characterize the major sources of variation in the absorbance of the infrared spectrum in caprine milk samples. A total of 657 goats belonging to 6 breeds and reared on 20 farms under traditional and modern dairy systems were milk-sampled once. Fourier-transform infrared (FTIR) spectra were taken (2 replicates per sample, 1314 spectra), and each spectrum contained absorbance values at 1060 different wavenumbers (5000 to 930 × cm−1), which were treated as a response variable and analyzed one at a time (i.e., 1060 runs). A mixed model, including the random effects of sample/goat, breed, flock, parity, stage of lactation, and the residual, was used. The pattern and variability of the FTIR spectrum of caprine milk was similar to those of bovine milk. The major sources of variation in the entire spectrum were as follows: sample/goat (33% of the total variance); flock (21%); breed (15%); lactation stage (11%); parity (9%); and the residual unexplained variation (10%). The entire spectrum was segmented into five relatively homogeneous regions. Two of them exhibited very large variations, especially the residual variation. These regions are known to be affected by the absorbance of water, although they also exhibited wide variations in the other sources of variation. The average repeatability of these two regions were 45% and 75%, whereas for the other three regions it was about 99%. The FTIR spectrum of caprine milk could probably be used to predict several traits and to authenticate the origin of goat milk.

Abstract Background The Italian Renal Cell Cancer Early Access Program was an expanded access program that allowed access to nivolumab, for patients (pts) with metastatic renal cell carcinoma (mRCC) prior to regulatory approval. Methods Pts with previously treated advanced or mRCC were eligible to receive nivolumab 3 mg/kg every 2 weeks. Pts included in the analysis had received ≥1 dose of nivolumab and were monitored for drug-related adverse events (drAEs) using CTCAE v.4.0. Immune-related (ir) AEs were defined as AEs displaying a certain, likely or possible correlation with immunotherapy (cutaneous, endocrine, hepatic, gastro-intestinal and pulmonary). The association between overall survival (OS) and irAEs was assessed, and associations between variables were evaluated with a logistic regression model. Results A total of 389 pts were enrolled between July 2015 and April 2016. Overall, the objective response rate was 23.1%. At a median follow-up of 12 months, the median progression-free survival was 4.5 months (95% CI 3.7–6.2) and the 12-month overall survival rate was 63%. Any grade and grade 3–4 drAEs were reported in 124 (32%) and 27 (7%) of pts, respectively, and there were no treatment-related deaths. Any grade irAEs occurred in 76 (20%) of patients, 8% cutaneous, 4% endocrine, 2% hepatic, 5% gastro-intestinal and 1% pulmonary. Of the 22 drAEs inducing treatment discontinuation, 10 (45%) were irAEs. Pts with drAEs had a significantly longer survival than those without drAEs (median OS 22.5 versus 16.4 months, p = 0.01). Pts with irAEs versus without irAEs had a more significant survival benefit (median OS not reached versus 16.8 months, p = 0.002), confirmed at the landmark analysis at 6 weeks. The occurrence of irAEs displayed a strong association with OS in univariable (HR 0.48, p = 0.003) and multivariable (HR 0.57, p = 0.02) analysis. Conclusions The appearance of irAEs strongly correlates with survival benefit in a real-life population of mRCC pts treated with nivolumab.

Simple Summary: Extensive goat breeding follows principles of sustainable agriculture, and it can be achieved with local breeds because their genome has been shaped by centuries of interaction with the environment in which goats were raised. The Sarda goat is well adapted to the arid environment of Sardinia (Italy). The aim of the present work was to study the variability of the casein genes, the main proteins of milk. We measured milk traits such as protein, fat, lactose, total solids, and milk yield, and performed association analyses with casein gene variants. The variability of the four casein genes was associated with milk protein, fat, total solids, and milk energy. The main finding was that intronic variants of the CSN1S2 gene (encoding the αs2-casein protein) were associated with milk yield and protein and fat content. This information might be used in selection schemes, and in future investigations aiming to disclose the direct link between genotype and phenotype. This work aimed to assess the variability of casein genes in a population of 153 bucks and 825 lactating does of the Sarda breed, and to perform association analysis between polymorphic sites and milk yield and composition traits. To genotype the casein genes, we chose an SNP panel including 44 SNPs mapping to the four casein genes CSN1S1, CSN2, CSN1S2, and CSN3. Genotyping (made by KASP™ genotyping assay, based on competitive allele-specific PCR) revealed the high variability of the Sarda goat, and haplotype analysis revealed linkage disequilibrium (LD) between CSN1S1 and CSN2 genes, in addition to two LD blocks within the CSN1S2 and two LD blocks within the CSN3 gene, in bucks and does. Association analysis revealed that variability at all four casein genes was associated with milk protein content, total solids, and milk energy. The three Ca-sensitive casein genes were associated with lipid content, and CSN1S2 showed a unique pattern, with intron variants associated with milk yield, in addition to milk pH, NaCl, and SCS (Somatic Cell Score). This information might prove useful in selection schemes and in future investigations aiming to better understand the biology of lactation, and the direct link between genotype and phenotype. The aim of the present research was to analyze the variability of 45 SNPs from different genes involved in metabolism and innate immunity to perform an association analysis with the milk yield, composition and milk coagulation traits. A population of 1112 Sarda breed sheep was sampled. Genotyping was generated by a TaqMan Open ArrayTM. Thirty out of the 45 SNPs were polymorphic, and 12 displayed a minor allele frequency higher than 0.05. An association analysis showed that the variability at genes PRKAG3 and CD14 was significantly associated with the daily milk yield. The variability at PRKAG3 was also associated with the protein and casein content, somatic cell score and bacterial score. The variation at the PRKAA2 gene was associated with the milk lactose concentration. The SNPs at CD14 were also associated with the traditional milk coagulation properties, while the SNPs at GHR and GHRHR were associated with kSR, a derived coagulation parameter related to the rate of syneresis. The information provided here is new and increases our knowledge of genotype–phenotype interactions in sheep. Our findings might be useful in appropriate breeding schemes to be set up for the Sarda sheep breed, but these should be confirmed by further studies, possibly performed on independent populations.
