ABSTRACT: Rabies is a fatal viral disease that affects animals and humans and is widespread among national ecological systems. We describe the anatomopathological and immunohistochemical findings of a case of rabies in a jaguarundi (Herpailurus yagouaroundi) found in a rural peri-domestic area after showing aggression and attacking two animals. Histopathology of brain tissue showed lymphoplasmacytic meningoencephalitis associated with intracytoplasmic eosinophilic inclusion bodies in neurons, gliosis and neuronophagia. The diagnosis of rabies was based on the lesions and confirmed by immunohistochemistry and qPCR. In Brazil, rabies in wild felines of the genus Puma has only been described in animals through serological examinations. The pathological findings are essential for the diagnosis and elucidation of the participation of this species in the wild cycle of the disease, as well as in the maintenance and conservation of the balance of wild fauna.

Abstract Samples of brain cortex of 12 free-range healthy wild boars from Southern Brazil were tested for RABV by dFAT and qPCR, all negative. The negative results in seropositive healthy wild boars may be associated with exposure of non-viable virus particles, virus proteins by bat saliva or consumption of contaminated carcasses.

Opossums are considered resistant to rabies. Nonhematophagous bats are reservoirs of rabies in urban areas of South America. We analyzed bats and opossums tested for rabies during 2021 in a highly urbanized city in Brazil to understand spillover in an urban setting. Wildlife surveillance is necessary to prevent rabies in humans and domestic animals.

Abstract Background Neonatal calf diarrhea (NCD) is the leading cause of calf morbidity and mortality in beef cattle. Cow’s vaccination in last stage of pregnancy is one of the most important measures to mitigate the risk of NCD outbreaks. The aim of this study was to evaluate the efficacy of prepartum single dose vaccination against NCD, especially Bovine Rotavirus type A (BoRVA) and Bovine Coronavirus (BCoV), in Nelore dams and offspring. A total of 117 pregnant cows (n = 81) and heifers (n = 36) were distributed in two groups, vaccinated (VAC: cows = 40; heifers = 19) and non-vaccinated (NVAC: cows = 41; heifers = 17). Vaccination occurred between 60 to 50 days before the expected calving date with a single dose of a water-in-oil (W/O) vaccine, and NVAC group received a dose of saline solution 0.9%. Blood samples were collected before vaccination and 30 days after to evaluate the antibody (Ab) response. Specific IgG1 Abs against BoRVA and BCoV were measured by using an Enzyme Linked Immuno Sorbent Assay (ELISA). Calves’ births were monitored, and the transference of passive immunity was evaluated. Diarrhea was monitored in the first 30 days of age, and fecal samples were collected for identification of the etiological agent. This review of human and cat rabies from 1986 to 2022 has shown mostly AgV3 variant in human cases with 29/45 (64.4%) reports including 23 from bats, four from cats, and two from unknown species, followed by 8/45 (17.8%) of AgV2 variant (all from dogs), 4/45 from marmoset variant (all from Callithrix jacchus), 2/45 samples compatible with wild canid variant (both from Cerdocyon thous), and one/45 of AgV1 variant from a domestic dog. Only one sample of human rabies was not typified, related to bat aggression. In addition, surveillance conducted in the state of São Paulo confirmed the presence of rabies in 7/23,839 cats (0.031%) and 3/106,637 dogs (0.003%) between 2003 and 2013, with a 10:1 overall cat-to-dog positivity ratio. This 10-fold higher infection rate for cat rabies may be explained by cats’ hunting habits and predation. In addition, after 28 years of rabies-free status, a new cat rabies case was reported in the city of São Paulo in 2011. The rabid cat lived, along with other pets, in a household located near the largest downtown city park, whose owners presented animal hoarding behavior. Thus, animal hoarders and rescuers, public health agents, animal health professionals, and the general population with contact need to be aware of the risk of bat-borne rabies followed by spillover from cats to humans. In conclusion, cat rabies cases are becoming increasingly important in Brazil. This poses a One Health concern, given the overlapping of human, bat and cat populations within the same predisposed environment.

The screening of flu-like syndrome is difficult due to nonspecific symptoms or even oligosymptomatic presentation and became even more complex during the Covid-19 pandemic. However, an efficient screening tool plays an important role in the control of highly contagious diseases, allowing more efficient medical-epidemiological approaches and rational management of global health resources. Infrared thermography is a technique sensitive to small alterations in the skin temperature which may be related to early signs of inflammation and thus being relevant in the detection of infectious diseases. Thus, the objective of this study was to evaluate the potential of facial thermal profiles as a risk evaluator of symptoms and signs of SARs diseases, using COVID-19 as background disease. A total of 136 patients were inquired about the most common symptoms of COVID-19 infection and were submitted to an infrared image scanning, where the temperatures of 10 parameters from different regions of the face were captured. We used RT-qPCR as the ground truth to compare with the thermal parameters, in order to evaluate the performance of infrared imaging in COVID-19 screening. Only 16% of infected patients had fever at the hospital admission, and most infrared thermal variables presented values of temperature significantly higher in infected patients. The maximum eye temperature (MaxE) showed the highest predictive value at a cut-off of >35.9°C (sn = 71.87%, sp = 86.11%, LR+ = 5.18, LR- = 0.33, AUC = 0.850, p < 0.001). Our predictive model reached an accuracy of 86% for disease detection, indicating that facial infrared thermal scanning, based on the combination of different facial regions and the thermal profile of the face, has potential to act as a more accurate diagnostic support method for early COVID-19 screening, when compared to classical infrared methods, based on a single spot with the maximum skin temperature of the face.

Canine Distemper is a disease caused by Canine morbillivirus (CM), a pantropic virus that can affect the central nervous system (CNS), causing demyelination. However, the pathogenesis of this lesion remains to be clarified. Brain samples of 14 naturally infected dogs by CM were analyzed to evaluate the presence of oxidative stress and demyelination. RT-PCR assay was performed to confirm a diagnosis of canine distemper in the brain, immunohistochemistry anti-CM was used to localize the viral proteins in the tissue, and anti-4-hydroxy-2 nonenal (4-HNE) was a marker of a product of lipid peroxidation. The results showed the presence of viral proteins in the demyelinated area with the presence of 4-HNE. Our results suggest that the CM virus infection causes oxidative stress leading to lipid peroxidation, which causes tissue damage and demyelination. In conclusion, oxidative stress plays a significant role in canine distemper pathogenesis in the CNS.

Lyssavirus rabies (RABV) is an RNA virus and, therefore, is subject to mutations due to low RNA polymerase replication fidelity, forming a population structure known as a viral quasispecies, which is the core of RNA viruses’ adaptive strategy. Under new microenvironmental conditions, the fittest populations are selected, and the study of this process on the molecular level can help determine molecular signatures related to virulence. Our aim was to survey gene signatures on nucleoprotein and glycoprotein genes that might be involved in virulence modulation during the in vitro evolution of RABV lineages after serial passages in a neuronal cell system with or without the presence of neutralizing antibodies based on replicative fitness, in vivo neurotropism and protein structure and dynamics. The experiments revealed that amino acids at positions 186 and 188 of the glycoprotein are virulence factors of Lyssavirus rabies, and site 186 specifically might allow the attachment to heparan as a secondary cell receptor, while polymorphism at position 333 might allow the selection of escape mutants under suboptimal neutralizing antibodies titers.

Enteropathogenic Escherichia coli (EPEC) constitutes one of the main causes of mortality in children in low- to medium-income countries. Diverse animal species have been linked as reservoirs, including birds. The aim of this study was to describe the genomic and phylogenetic features of an EPEC recovered from a pet macaw and further characterizing the macro and microscopic lesion in a rabbit ileal loop experimental model. The isolate was whole-genome sequenced (WGS) obtaining its genotypic and phenotypic in silico characteristics and inoculated in a rabbit experimental model with subsequently evaluating the strain’s pathogenicity by scanning electron microscopy (SEM) and histopathology. The isolate was characterized as O109:H21-B1-ST40 typical EPEC, harboring several virulence factors of diarrheagenic E. coli. The macaw EPEC genome was located in a monophyletic clade of human and animal ST40 EPEC sequences. In vivo inoculation demonstrated severe hemorrhage with SEM and histopathological analysis confirming these lesions to be associated with intra-epithelial lymphocytes. Therefore, the isolate not only shared several genotypic and phylogenetic similarities with EPEC that affects humans and animals, but was able to induce severe tissue injury in a mammal model. These findings highlight the underrated role of pet birds as zoonotic reservoirs and the diversity in virulence factors being unraveled by new WGS studies.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), responsible for the coronavirus disease 2019 (COVID-19), is the causative infectious agent of the current pandemic. As researchers and health professionals are still learning the capabilities of this virus, public health concerns arise regarding the zoonotic potential of SARS-CoV-2. With millions of people detected with SARS-CoV-2 worldwide, reports of companion animals possibly infected with the virus started to emerge. Therefore, our aim is to review reported cases of animals naturally infected with SARS-CoV-2, particularly companion pets, shedding light on the role of these animals in the epidemiology of COVID-19.

Avian coronavirus (AvCoV) infects a range of tissues in chickens and several other avian species. Although the virus can be isolated in chicken embryos, only a few strains of the 6 genotypes/33 lineages can grow in cell lines, with the Beaudette strain (GI-1 lineage) being the most used for in vitro studies. Considering the differences between cell lines and chicken embryos as habitats for AvCoV, this study aimed to assess the diversity of the genes coding for the nonstructural protein 3 (nsp3) and spike envelope protein (S) after serial passages in BHK-21 and Vero cells. After 14 passages of an embryo-adapted Beaudette strain, the virus loads fluctuated in both cell lines, with the highest loads being 8.72 log genome copies/µL for Vero and 6.36 log genome copies/µL for BHK-21 cells. No polymorphisms were found for nsp3; regarding S, not only aa substitutions (Vero: 8th passage A150S, and 14th S150A; BHK-21: 4th S53F, 8th F53Y, and 8th S95R), but also minor variants could be detected on chromatograms with fluctuating intensities. As the regions of these aa substitutions are within the receptor-binding domain of S, it can be speculated that differences in cell receptors between Vero and BHK-21 cells and the speed of cell death led to the selection of different dominant strains, while the stability of nsp3 supports its function as a protease involved in AvCoV replication. In conclusion, AvCoV quasispecies evolution is influenced by the biological model under consideration, and a gradual transition is seen for minor and major variants.

Mast cell tumors (MCTs) are common neoplasms in dogs, and treatments for these diseases include surgery, polychemotherapy and targeted therapy with tyrosine kinase inhibitors. This study aimed to evaluate the response and the adverse events of treatment with imatinib mesylate (IM) compared to conventional therapy using vinblastine and prednisolone (VP) in canine cutaneous MCTs. Twenty-four dogs were included in the study; 13 animals were treated with IM and 11 with VP. Tumor tissue samples were submitted for histological diagnosis, grading and KIT immunostaining. The response to treatment was assessed by tomographic measurements according to VCOG criteria. Adverse events were classified according to VCOG-CTCAE criteria. The IM and VP groups had dogs with similar breeds, gender, ages, MCT localization, WHO stages and lymph node metastasis profiles. Most MCTs were grade 2/low and had KIT- patterns 2 and 3. The objective response rate (ORR) was significantly higher (30.79%) in the IM group then in VP group (9.09%). Adverse events (AE) in IM group were all grade 1, significantly different from VP. In conclusion, IM presented better ORR and less severe adverse events when compared to VP, representing a suitable option for the treatment of low-grade canine MCTs.

Abstract This paper reports on an investigation of the role of codon usage evolution on the suggested bovine-to-human spillover of Bovine coronavirus (BCoV), an enteric/respiratory virus of cattle, resulting in the emergence of the exclusively respiratory Human coronavirus OC43 (HCoV-OC43). Analyses based on full genomes of BCoV and HCoV-OC43 and on both human and bovine mRNAs sequences of cholecystokinin (CCK) and surfactant protein 1 A (SFTP1-A), representing the enteric and respiratory tract codon usage, respectively, have shown natural selection leading to optimization or deoptimization of viral codon usage to the human enteric and respiratory tracts depending on the virus genes under consideration. A higher correlation was found for the nucleotide distance at the 3rd nucleotide position of codons and codon usage optimization to the human respiratory tract when BCoV and HCoV-OC43 were compared. An MCC tree based on relative synonymous codon usage (RSCU) data integrating data from both viruses and hosts into a same analysis indicated three putative host/virus contact dates ranging from 1.54E8 to 2.44E5 years ago, suggesting that an ancestor coronavirus might have followed human evolution.

Objectives The objectives of this study were to confirm the prevalence of feline immunodeficiency virus (FIV) infection in domestic cats in the region north of Ceará, Brazil, and to determine the factors associated with infection and the major circulating subtypes of the virus in this area. Methods Samples from 148 cats were collected and tested using anti-FIV antibody screening, with confirmation of positive results by PCR. Univariate analysis was performed considering the epidemiological characteristics and FIV status. Sequencing and phylogenetic analysis of the gag and pol genes were performed to confirm the FIV subtype. Results Nine cats (6.1%) tested positive for FIV – one female (0.7%) and eight males (5.4%). Introduction: The most frequent demands on microscopic food analysis are allegations of consumers finding macroscopic foreign matter or suspecting the presence of undeclared ingredients on products labels. The byproducts and foreign matters detection are fundamental practice for indirectly verifying the conditions of food production. Objective: This study reports the processes of microscopic and molecular identification (PCR) of a foreign matter found in a meat pie after a consumer complaint, occurred in the city of Itapira, state of São Paulo, Brazil. Method: Two distinct procedures were used to identify foreign matter: macroscopic examination, following FDA standards, and polymerase chain reaction (PCR) technique to identify DNA extracted from foreign materials. Results: The macroscopic analysis identified animal taste buds composing the pie fillings, and the PCR test confirmed that they were of bovine origin. Conclusions: Macroscopic analysis and the PCR test allowed the identification of the type of foreign matters and confirmed its bovine origin, what was enough to characterize it as a fraud by the improper use of inferior tissues in the preparation of ready-to-eat pastry.

Canine leproid granuloma or “canine leprosy” is a mycobacteriosis that has a dermatopathic framework, which includes aspects that are still unclear, especially regarding its mode of transmission and etiological agent. In order to verify the genetic characterization of the mycobacteria that is involved in the condition, the present study analyzed histological cuts from animals with an established diagnosis of such disorder. The study samples were taken from of 13 dogs that were assisted at the HOVET / USP Dermatology Service. This material was subsequently subjected to “nested” and real-time polymerase chain reaction (PCR). The nested PCR confirmed the presence of the agent in 18.7% of the animals, whereas the real-time PCR detected 69.2% of samples with DNA of the mycobacterium, thus proving that such method presents higher sensitivity. Furthermore, it was observed that the species of mycobacterium that causes Canine Leproid Granulnoma in dogs from Brazil presents a 100% genetic homology when compared to samples obtained from animals in studies abroad.

Canine coronavirus (CCoV) exists in types I and II and infects dogs leading mainly to enteritis, though type II has already been associated with generalized and highly lethal infection. A CCoV-type II inactivated vaccine produced in A72 canine cells is available worldwide and largely used, though the molecular stability after serial passages of vaccine seeds is unknown. This article reports the evolution of the CCoV-II vaccine strain 1-71 in A72 cells based on partial S gene sequencing, showing the predominance of neutral evolution and the occurrence of four sites under purifying selection. Thus, cell-adapted strains of CCoV-II may be genetically stable after serial passages in a same cell line due to a stable virus-host relationship.

Rotavirus is the causative pathogen of diarrhea in humans and in several animal species. Eight pairs of primers were developed and used for Sanger sequencing of the coding region of the NSP1, NSP3, and VP6 genes based on the conserved regions of the genome of the group A porcine rotavirus. Three samples previously screened as positive for group A rotaviruses were subjected to gene amplification and sequencing to characterize the pathogen. The information generated from this study is crucial for the understanding of the epidemiology of the disease.

In this study, we describe the viral composition of adult Antricola delacruzi ticks collected in a hot bat cave in the state of Rondônia, Western Amazonia, Brazil. A. delacruzi ticks, are special, compared to many other ticks, in that they feed on both bats (larval blood feeding) and bat guano (nymphal and adult feeding) instead of feeding exclusively on vertebrate hosts (blood feeding). Considering this unique life-cycle it is potentially possible that these ticks can pick up/be infected by viruses not only present in the blood of viremic bats but also by virus shed through the bat guano. The viral metagenomic investigation of adult ticks showed that single-stranded negative-sense RNA viruses were the dominant group of viruses identified in the investigated ticks. Out of these, members of the Nairoviridae family were in clear majority constituting 88% of all viral reads in the data set. Genetic and phylogenetic analyses indicate the presence of several different orthonairoviruses in the investigated ticks with only distant relationship to previously described ones. In addition, identification of viral sequences belonging to Orthomyxoviridae, Iflaviridae, Dicistroviridae, Polycipiviridae, Reoviridae and different unclassified RNA viruses showed the presence of viruses with low sequence similarity to previously described viruses.

Different herpesviruses have been associated with respiratory and enteric disease and mortality among seabirds and waterfowl. In 2011, a respiratory disease outbreak affected 58.3% (98/168) of the Magellanic penguins undergoing rehabilitation due to an oil spill off the southern Brazilian coast. Etiology was attributed to a novel herpesvirus identified by histopathology, immunohistochemistry, electron microscopy and molecular studies with partial DNA sequencing. Since migration, rehabilitation and translocation may facilitate the spread of pathogens between populations and trigger the onset of clinical disease in animals with latent infections, investigation of herpesvirus occurrence in asymptomatic seabirds was performed. Samples from free-ranging seabirds were collected in Argentinian Patagonia (Magellanic penguins) and the Abrolhos Archipelago in Brazil (Brown boobies, Masked boobies, Red-billed tropicbirds, White-tailed tropicbirds and South American tern). Furthermore, asymptomatic seabirds housed at the facility where the outbreak occurred were also sampled. In total, 354 samples from eight seabird species were analyzed by PCR for herpesvirus. Four different sequences of herpesviruses were identified, one in Yellow-nosed Albatross, one in Boobies and Tropicbirds and two in Magellanic penguins. Magellanic penguin herpesvirus 1 was identified during the penguin outbreak at the rehabilitation facility in Brazil, while Magellanic penguin herpesvirus 2 was recovered from free-ranging penguins at four reproduction sites in Argentina. Phylogenic analysis of the herpesviruses sequences tentatively identified suggested that the one found in Suliformes and the one associated with the outbreak are related to sequences of viruses that have previously caused seabird die-offs. These findings reinforce the necessity for seabird disease surveillance programs overall, and particularly highlight the importance of quarantine, good hygiene, stress management and pre-release health exams in seabirds undergoing rehabilitation.

In Brazil, bats have been assigned an increasing importance in public health as they are important rabies reservoirs. Phylogenetic studies have shown that rabies virus (RABV) strains from frugivorous bats Artibeus spp. are closely associated to those from the vampire bat Desmodus rotundus, but little is known about the molecular diversity of RABV in Artibeus spp. The N and G genes of RABV isolated from Artibeus spp. and cattle infected by D. rotundus were sequenced, and phylogenetic trees were constructed. The N gene nucleotides tree showed three clusters: one for D. rotundus and two for Artibeus spp. Regarding putative N amino acid-trees, two clusters were formed, one for D. rotundus and another for Artibeus spp. RABV G gene phylogeny supported the distinction between D. rotundus and Artibeus spp. strains. These results show the intricate host relationship of RABV's evolutionary history, and are invaluable for the determination of RABV infection sources.

Descreve-se um surto de disenteria de inverno que afetou 10 vacas leiteiras de uma propriedade localizada em Viamão, Rio Grande do Sul. O quadro clínico caracterizou-se por uma diarréia inicialmente líquida esverdeada com estrias de sangue e muco, evoluindo, em alguns animais, para uma diarréia de coloração marrom escura à sanguinolenta, que persistiu, em média, cinco dias. Drástica diminuição na produção de leite e no consumo de alimentos, além de graus variados de depressão também foram observados. Apenas um dos 10 animais afetados morreu. Durante a necropsia, observaram-se mucosas pálidas, conteúdo sanguinolento com presença de grande quantidade de coágulos, principalmente no cólon espiral e petéquias na mucosa do cólon. Os principais achados histológicos foram encontradas no cólon espiral, onde havia criptas dilatadas, sem epitélio de revestimento ou revestidas por epitélio pavimentoso e/ou cuboidal, por vezes com núcleos grandes e nucléolos proeminentes. Algumas criptas eram preenchidas por debris necróticos e polimorfonucleares. Na imuno-histoquímica com anticorpo monoclonal para coronavírus bovino (8F2) em cortes do cólon espiral, havia marcações positivas no citoplasma de enterócitos das criptas, nos debris necróticos destas e em macrófagos na lâmina própria.The report describes a winter dysentery outbreak that affected 10 dairy cows from a ranch located in the county of Viamão, Rio Grande do Sul. The most significant clinical sign was profuse and watery diarrhea, which ranged from a greenish to a brownish coloration and from occasional blood streaks with mucus to a bloody diarrhea. Most cases persisted for 5 days and also included depression, drastic decrease in milk production and in food consumption. Only one of the affected animals died. The necropsy revealed pale mucosa and sanguineous content with high quantity of blood clots, particularly within the spiral colon, and petechiae on the colonic mucosa. Histopathological lesions were predominant in the spiral colon, and consisted of a high number of dilated crypts without epithelium or with the replacement of a pavement epithelium with occasional immature cuboidal cells, which sometimes showed enlarged nucleus and proeminent nucleolus. Some crypts were filled with epithelial desquamation and polymorphnuclear cells. Bovine coronavirus (8F2) monoclonal antibody was used for immunohistochemistry on sections of the spiral colon that showed positive reactions in the cytoplasm of the infected crypt epithelium, sloughed necrotic cells, and within macrophages in the lamina propria of both.

Salmonella spp. é um dos principais agentes envolvidos em casos de doenças de origem alimentar em humanos, responsável por perdas significativas na avicultura. O objetivo deste estudo foi investigar a presença de Salmonella spp. em fezes de perus comerciais no Brasil. Foram colhidos suabes fecais de 14 lotes de perus comerciais (pool de seis aves/lote). Os suabes foram submetidos aos procedimentos de isolamento bacteriológico convencionais e a detecção de DNA do agente foi realizada através da técnica de PCR. Salmonella spp. foi detectada em um total de nove lotes dos 14 avaliados. As amostras foram negativas na identificação molecular dos sorovares Enteritidis e Typhimurium. Os isolados foram encaminhados ao laboratório de referência para sorotipagem e identificados como S. Agona; um patógeno considerado emergente em vários países.

Rabies virus samples (n = 17) isolated from bovines (n = 11), equines (n = 4) and buffalo (n = 2) from Pará State (n = 7), Tocantins (n = 6) and Rondônia (n = 4) were submitted to RT-PCR in order to obtain partial sequences of nucleoprotein (N) and glycoprotein gene (G). Nucleotide sequences were analyzed using Neighbor-Joining model, Kimura 2-parameters evolutionary model. All the 17 samples analyzed were related to cluster A, lineage associated with the hematophagous bat Desmodus rotundus. The phylogenetic analysis based on the N and G genes, suggests the presence of five sub-lineages (A1-A5), while G gene showed seven sub-lineages (A1-A7). In both phylogenies, sublineages A1 to A3 exhibit a similar composition and geographic distribution. Diverse composition of remaining groups of N and G gene is attributable to different sequences used in the alignments for each genomic region. Glycoprotein amino acid sequence showed molecular markers in sub-lineages A2, A3, A4 and A7. This information provides a better comprehension of molecular epidemiology of rabies, starting with the knowledge of viral lineages circulating in the Brazilian Amazon.

Chlamydophila felis is associated with upper respiratory tract infections. In the present study, 31 cats from a noncommercial shelter located in Osasco, SP, Brazil, were examined. The cats presented with clinical manifestations, which were classified from grade 1 to 4, with 4 indicating severe manifestations. In total, 16.13% of the cats presented with grade 1 severity of clinical manifestations, 25.81% presented with grade 2, 38.71% presented with grade 3, and 19.35% presented with grade 4. PCR was used to detect C. felis in samples taken from the oral mucosa and ocular conjunctiva of both eyes using sterile, dry cotton swabs. Overall, 58% of the samples were positive for C. felis. Of these animals, none showed clinical manifestations that were classified as grade 1, whereas 5.56% of cats were classified as grade 2, 61.11% were classified as grade 3, and 33.33% were classified as grade 4. The median clinical manifestation intensity score for the first group was 3 and ranged from 2 to 4. In the second group not positive for C. felis, 38.45% of the animals (5/13) presented with manifestations classified as grade 1, 53.85% (7/13) were classified as grade 2, 7.69% (1/13) were classified as grade 3, and no animals were classified as grade 4. The median clinical manifestation intensity score for the second group was 2 and ranged from 1 to 3. In this study, there was a high occurrence of C. felis in animals with clinical manifestations.

This study investigated the occurrence of rotavirus in porcine and Rattus norvegicus, at the same time, on a pig farm in the city of Jaguariúna, São Paulo, Brazil. Swine () and rat () fecal samples were analyzed by nested RT-PCR assay. Rotavirus occurred in seven porcine and two rat samples. A total of three pig and one rat samples were further submitted to genetic sequencing. The partial NSP5 gene phylogeny showed that all strains were segregated in the genotype H1. These results point toward a cross-species transmission between rats and pigs on the surveyed farm and represent the first detection of rotavirus in Rattus norvegicus in Brazil.

O vírus da bronquite infecciosa das galinhas (IBV) (Nidovirales: Coronaviridae) é um Gammacoronavirus com a maior taxa evolutiva no gênero e, apesar de uma recentemente relatada atividade corretiva de sua polimerase, a diversidade intra e inter-hospedeiros é um fenômeno bem documentado. Este estudo objetivou avaliar a variação genética após passagens seriais de uma amostra de IBV variante. A amostra CRG-BETA, propagada em embriões de galinhas, foi inoculada em monocamadas de células VERO até a quarta passagem e cada passagem foi monitorada com uma RTPCR para a região S1 do gene S (nt 705 a 1094) e uma RT-PCR para o mRNA da proteína 5a do vírus. Todas as passagens foram positivas para S1 e as passagens 1 a 3 para mRNA 5a; sequências de S1 não apresentaram polimorfismos. O encontro de mRNA de IBV nos cultivos celulares demonstra que a amostra CRG-BETA está replicando nas células VERO e, em relação à análise de S1, a ausência de mutações de nucleotídeos demonstra que a amostra CRG-BETA pode ter atingido um estado fixo. Como conclusão, diferentes genótipos de IBV apresentam diferentes padrões evolutivos não apenas in vivo, como previamente conhecido, mas também in vitro, como aqui relatado.

Descreve-se um ensaio de duplex RT-PCR assay para a detecção simultânea do vírus da bronquite infecciosa das galinhas (IBV) e do metapneumovirus aviário (aMPV), agentes etiológicos de doenças de elevada importância em avicultura. A duplex RT-PCR otimizada mostrou um limiar de detecção de 10-3 (101 EID50/50m L para IBV e 100.5 EID50/50m L para aMPV, respectivamente, quando da combinação dos dois vírus e 10-1 para cada um dos vírus em separado(103 EID50/50m L para IBV e 102.5 EID50/50m L para aMPV, respectivamente. O ensaio foi demonstrado como específico, sensível e aplicável à rápida detecção destes vírus em amostras clínicas.

Actinobaculum suis is an important agent related to urinary infection in swine females. Due to its fastidious growth characteristics, the isolation of this anaerobic bacterium is difficult, thus impairing the estimation of its prevalence. The purpose of this study was to develop and test a polymerase chain reaction (PCR) for the detection and identification of A. suis and then compare these results with traditional isolation methods. Bacterial isolation and PCR were performed on one hundred and ninety-two urine samples from sows and forty-five preputial swabs from boars. The results indicate that this PCR was specific for A. suis, presenting a detection limit between 1.0×101 CFU/mL and 1.0×102 CFU/mL. A. suis frequencies, as measured by PCR, were 8.9% (17/192) in sow urine samples and 82.2% (37/45) in preputial swabs. Assessed using conventional culturing techniques, none of the urine samples were positive for A. suis; however, A. suis was detected in 31.1% (14/45) of the swabs. This PCR technique was shown to be an efficient method for the detection of A. suis in urine and preputial swabs.

O perfil patogênico de um vírus da raiva isolado de um morcego insetívoro Lasiurus ega foi comparado com o de vírus fixo de raiva (CVS/32) em hamster e camundongo, determinando os períodos de incubação e clínico, manifestação clínica e mortalidade. Os animais foram desafiados com 10 2,611-4,021 DL50 /0,05 mL do isolado de L. ega e 10 3,7- 4,7 LD50 /0,05 mL do CVS/32, usando as vias: intramuscular (IM), intradermica (ID), intranasal (IN) e abrasão epidermica (AE). A presença do antígeno viral foi confirmada pela prova de imunofluorescência direta. As porcentagens de mortalidade observadas com o isolado de L. ega foram as seguintes em hamster: 3,5% IM, 10,71% IN; em camundongo: 50.0% IM, 30.0% IN. A forma furiosa da doença foi predominante. As porcentagens de mortalidade observadas com o vírus CVS/32 em hamster foram as seguintes: 12.5% IM, 62.5% ID, 12.5% IN; em camundongo 100.0% IM, 70.0% ID, 10.0% IN. Com este vírus foi observada raiva paralitica. A via AE mostrou-se inadequada para induzir doença. O período de incubação foi de 57 dias para o CVS/32 e 11-16 dias para o isolado de L. ega, entre tanto os períodos clínicos oscilaram entre 47 dias para ambos os vírus. Varias substituições foram achadas em domínios antigênicos da glicoproteína: AI (posição 231), AII (34 42 e 198-200), domínio de fusão dependente de baixo pH (102179), domínio da transmembrana (440461) e resíduo 242. Esses vírus mostraram comportamentos biológicos distintos o que poderia estar ligados às substituições nos domínios antigênicos anteriormente descritos.

O coronavírus bovino (BCoV) é um vírus RNA simples fita, de sentido positivo, não segmentado com envelope constituído de uma camada dupla de lipídios com quatro proteínas (HE, S, E e M) que resultam no aspecto de coroa dos vírions. Como a HE (hemaglutinina-esterase) é uma proteína polimórfica com uma função de receptor aglutinante secundária, estudos sobre a diversidade do gene HE podem possibilitar maiores informações sobre a evolução e interação hospedeiro-parasita do BCoV. Uma reação de hemi-nested RT-PCR foi desenvolvida para a amplificação de um produto de 441pb do gene HE do BCoV (nt 543 ao 562). Temperaturas ótimas de hibridização foram testadas em um termociclador com gradiente para a reação de hemi-nested e utilizada no protocolo final. A sensibilidade analítica foi determinada por meio da diluição serial na base 10 do BCoV amostra Kakegawa (título HA: 256) em uma suspensão fecal negativa para BCoV, resultando positiva até a diluição de 10-6, mostrando uma alta sensibilidade analítica sem inibição na PCR. O protocolo final da hemi-nested RT-PCR foi aplicado a 21 amostras fecais de vacas previamente positivas para BCoV e o sequenciamento de DNA do produto de 441pb de 14 amostras resultaram em sequências com elevado escore do gene HE do BCoV após a análise no BLAST/n. Essa hemi-nested RT-PCR é uma ferramenta poderosa para estudos de diversidade filogenética de linhagens de campo de BCoV e para estudos comparativos entre os diferentes genes dos Coronavírus.

Este artigo relata a divergência genética de PCV2 após passagens em células das linhagens VERO e SK-RST. O teste exato de Fisher indicou tendência para seleção positiva no gene cap. Estes resultados permitem inferências relativas ao desenvolvimento de vacinas contra PCV2 e sobre a evolução do gênero Circovirus.

Coronavírus entérico de furões (FECV) é associado à enterite catarral epizoótica (ECE) em furões (Mustela putorius furo). Neste estudo, relatamos a ocorrência deste agente em quatro amostras fecais diarreicas de furões domésticos, analisadas por microscopia eletrônica de transmissão (contrastação negativa) e RT-PCR específica e direcionada ao gene de nucleocapsídeo (N). Estes achados constituem o primeiro relato de FECV no Brasil e remetem para a importância deste vírus na etiologia de quadros entéricos nestes animais.

O rotavírus é um agente etiológico de diarréias de distribuição mundial, responsável por grandes prejuízos econômicos. Pesquisou-se a soroprevalência de anticorpos anti-rotavírus do sorogrupo A em bovinos criados no município de Uruará, utilizando a técnica da contraimunoeletroosmofore com a amostra NCDV como antígeno padrão. A prevalência de propriedades positivas foi de 95.6-100%. Diferenças significativas foram observadas entre as sucessivas faixas etárias, quando a soropositividade aumentou dos grupos mais jovens para os mais velhos, bem como entre fêmeas e machos maiores de 1 ano, quando a soropositividade foi maior no primeiro grupo.

In Brazil, bats have been assigned an increasing importance in public health as they are important rabies reservoirs. Phylogenetic studies have shown that rabies virus (RABV) strains from frugivorous bats Artibeus spp. are closely associated to those from the vampire bat Desmodus rotundus, but little is known about the molecular diversity of RABV in Artibeus spp. The N and G genes of RABV isolated from Artibeus spp. and cattle infected by D. rotundus were sequenced, and phylogenetic trees were constructed. The N gene nucleotides tree showed three clusters: one for D. rotundus and two for Artibeus spp. Regarding putative N amino acid-trees, two clusters were formed, one for D. rotundus and another for Artibeus spp. RABV G gene phylogeny supported the distinction between D. rotundus and Artibeus spp. strains. These results show the intricate host relationship of RABV's evolutionary history, and are invaluable for the determination of RABV infection sources.

Identification of animals that are decomposing or have been run over or burnt and cannot be visually identified is a problem in the surveillance and control of infectious diseases. Many of these animals are wild and represent a valuable source of information for epidemiologic research as they may be carriers of an infectious agent. This article discusses the results obtained using a method for identifying mammals genetically by sequencing their mitochondrial DNA control region. Fourteen species were analyzed and identified. These included the main reservoirs and transmitters of rabies virus, namely, canids, chiroptera and primates. The results prove that this method of genetic identification is both efficient and simple and that it can be used in the surveillance of infectious diseases which includes mammals in their epidemiologic cycle, such as rabies.

Rapid diagnosis of rabies in suspected human cases influences post-exposure prophylaxis for potential contacts of the patient and ensures appropriate patient management. Apart from the central nervous system (CNS), rabies virus (RABV) is usually present in small sensory nerves adjacent to hair follicles of infected humans. We used an RT-PCR, with primers targeted to the 3' terminal portion of the nucleoprotein gene (N), to test neck-skin samples of nine patients who had rabies in order to validate a diagnostic method that could serve as an additional tool for rabies diagnosis, particularly in antemortem samples. Six of eight postmortem samples were found to be positive for rabies by RT-PCR, and one of two samples collected antemortem was positive with this same technique. Results were confirmed by DNA sequencing; this validates RT-PCR and neck-skin as a suitable technique and type of sample, respectively, for use in the diagnosis of human rabies. RT-PCR applied to neck-skin biopsies could allow early diagnosis and lead to more effective rabies treatment.

This article reports on the identification of a group 2 coronavirus (BatCoV DR/2007) in a Desmodus rotundus vampire bat in Brazil. Phylogenetic analysis of ORF1b revealed that BatCoV DR/2007 originates from a unique lineage in the archetypical group 2 coronaviruses, as described for bat species elsewhere with putative importance in Public Health.

This study aimed to test in vitro a RNA-interference based antiviral approach for rabies with short-interfering RNAs (siRNAs) against rabies virus nucleoprotein mRNA. BHK-21 cells were infected with serial dilutions of PV rabies virus strain and transfected with a pool of three siRNAs. Direct immunofluorescence staining showed a 5-time decrease in virus titer when compared to a non-treated plate, showing a promising new approach to the development of antivirals for rabies treatment.
