The effects of gamma-ray-induced radicals on the activities of viruses and membrane proteins were studied with Sendai virus in aqueous suspensions with or without additives including OH scavengers. The activities measured were hemagglutination and hemolysis located in the viral membrane. The changes in the protein components of viruses were analyzed at the same time by polyacrylamide gel electrophoresis (PAGE). 2-Mercaptoethanol (70 mM) and p-aminobenzoic acid (1 mM), both known as OH scavengers, fully protected the viral activities from gamma-ray-induced inactivation. Deaeration of virus suspensions by bubbling argon through the suspension did not affect the inactivation curve of the activities of the virus. Conspicuous changes in the membrane-associated glycoprotein bands found by PAGE analysis generally coincided with the findings in the activities of the virus. These findings suggest that OH radicals are the major damaging species acting on the glycoproteins in the membrane, and result in the inactivation of viral functions. In spite of strong OH scavenging ability, t-butanol resulted in the enhancement of inactivation and a decrease in the intensity of glycoprotein bands in the PAGE pattern. This finding, which is seemingly contradictory to the above conclusion, is discussed in terms of the action of alcohol radicals generated after OH scavenging. In particular, the possibility is pointed out that the membrane structure is affected by the reaction with the alcohol radicals, causing a crucial alteration of embedded proteins.

Taken altogether, the EM evidence we have obtained indicates that the induced (both viral and PEG) and spontaneous (entrance of a splenocyte into a cell) fusion of mammalian somatic cells are associated with alterations in the structure of fusing cells. For example there are alterations in the structure of not only the surfaces of fusing cells but also in the nucleus envelopes and cytoplasmic organelles after PEG treatment. Also, there is long retention of cellular plasma membrane remnants in virally-induced heterokaryons. In short, for each case the alterations were unquestionably specific, in response to the imposed challenge. These specific features not only determine the efficiency and rate of fusion, but also the mode of which the hybrid nucleus is formed. This mode directly determines the fate of the synkaryon and the stability of the so formed hybrid genome. It might be thought that an increase in the inner nuclear envelope observed in some hybrids would counteract the consequences of the disproportion arising between the increase in cell volume and nuclear surface. The finger-like invaginations of the hybrid nuclei nuclear envelope, surrounded by replicatively and transcriptionally active chromatin, appear to be EM demonstrations of such counteracting mechanisms. These invaginations, by augmenting the available inner layer, most likely increase the anchorage sites for chromatin. It is noteworthy that the invaginations occur mainly in multichromosomal hybrids with little chromosome loss. It appears possible that some of the hybrids may contain particular chromosomes from the more differentiated parent cell.

Protection of mice from lethal Sendai virus (HVJ) infections by a temperature-sensitive mutant, HVJpi, which was isolated from a carrier culture, was studied. HVJpi had a strong interfering capacity with the replication of virulent wild-type virus in LLCMK2 cells. When a high dose of HVJpi (3.0 x 10(7) CIU) was inoculated intranasally into mice, the mice showed neither illness nor lung lesions but gained significant resistance against the challenge of virulent wild-type virus (18 LD50) immediately after inoculation. In contrast, the mice inoculated with a lower dose of HVJpi (8.2 x 10(5) CIU) did not show the immediate resistance but became immune several days after inoculation. Time courses of the virus replication in the lung revealed that the replication of wild-type virus was strongly suppressed to about 1/1000 by the simultaneous infection with a high dose of HVJpi, thus resulting in minimizing the lung lesions and survival of all the mice infected. Neither interferon nor natural killer cells appeared to play a major role in the immediate immune status by HVJpi, since no difference was observed in protection of mice simultaneously infected with wild-type virus and HVJpi in spite of pretreatment of the mice with anti-interferon and anti-asialo GM1 antibodies as compared with that of the untreated doubly infected mice. On the other hand, it was suggested by analysis of viral polypeptides synthesized in the lung of infected mice by Western blotting that the early stage of replication of wild-type virus in the lung was inhibited mainly by the interfering capacity of HVJpi. These results indicate that HVJpi is an unique virus mutant which is capable of protecting mice from lethal Sendai virus infections by its interfering capacity immediately after inoculation and then by the induction of virus-specific immune responses.

Penetration of human immunodeficiency virus (HIV) into the cells of lymphoblastoid T-cell line H9 was studied using 35S-methionine-labeled virus by demonstration of virus-specific proteins in the cytoplasm of the infected cells. Purification of the virus by ultracentrifugation through 30% glycerol was shown to lead to virus aggregation and its partial destruction manifested by the loss of gp120 protein, therefore unlabeled concentrated virus was used mainly with subsequent determination of virus-specific proteins by immune blotting. The addition of Sendai virus inactivated with UV rays to HIV increased the amount of HIV associated with cells as well as the amount of virus-specific proteins in the cytoplasm of the infected cells.

Mice inoculated with ultraviolet light-inactivated Sendai virus mount a cell- mediated immune response to the virus. Cytotoxic T cells specific for Sendai virus can be obtained by in vitro secondary stimulation of primed spleen cells with syngeneic stimulator cells coated with UV-inactivated Sendai virus. Neither in vivo nor in vitro stimulation alone is sufficient to generate specific cytotoxic T cells. Sharing of the H-2 haplotype between cytotoxic T cells and target cells is required for the Sendai virus-specific lysis to occur. The fusion (F) glycoprotein of Sendai virus has been implicated in target antigen formation (20). Ethanol treatment of Sendai virus causes complete inactivation of the cell-fusion and hemolytic activities of the envelope, but does not affect the antigenicity of the F glycoprotein; furthermore, hemagglutinin and neuraminidase activities of the envelope HANA glycoprotein are also left intact after ethanol treatment. Target cells can be prepared by coating them with various numbers of UV-inactivated Sendai virus that have been treated with ethanol or, as a control, phosphate-buffered saline (PBS). The amount of virus adsorbed to target cells during the cytotoxicity reaction time using either ethanol-treated or untreated (PBS "treated") virions is essentially identical, but target cells coated with ethanol-treated Sendai virus fail to serve as targets for cytotoxic T cells. These results indicate that fusion activity of the Sendai virus envelope is essential to the formation of the target antigen and that virus adsorption to cell surfaces without fusion of the envelope with cell membranes is not sufficient to allow killing by virus-specific cytotoxic T cells.

Cytotoxic T cells specific for Sendai virus were generated by culturing murine spleen cells in vitro together with UV-inactivated Sendai virus. In vivo immunization of donor mice with UV-inactivated Sendai virus resulted in an in vitro secondary response of increased magnitude. Cytotoxic activity was demonstrated in a short-term 51Cr-release assay, using syngeneic tumor cells which had been coated with inactivated Sendai virus by incubation at 4 degrees C for 30 min. The lysis of Sendai virus-coated target cells was restricted by the H-2 haplotype of the target cells, suggesting that the H-2 genes of the target cell contributed to the specificity of the lysis. Kinetic experiments showed that susceptibility to lysis by cytotoxic T cells specific for Sendai virus appeared within 30 min after coating target cells with inactivated virus. Furthermore, there was no detectable synthesis of new proteins in cells treated with UV-inactivated Sendai virus. For these reasons, we suggest that neither viral replication nor the synthesis of new proteins are necessary for the production of the antigen recognized by cytotoxic cells specific for Sendai virus. We infer that the virus-specific component on the target cells is probably a preformed virion antigen adsorbed onto or integrated into the cell membrane. These results imply that, if the cytotoxic T cell recognizes a single antigenic determinant specified both by viral and H-2 genes, this determinant is formed by the physical association of H-2 and Sendai virus antigens rather than by their alteration during the processes of synthesis.

When a 24-h tube culture of rabbit alveolar macrophages was infected with Sendai virus, the rate of infected cells was found to be limited. Even at a multiplicity of infection (MOI) of 500 plaque-forming units per cell, an average of 63% cells was found to synthesize viral antigens stainable by direct immunofluorescence. When the macrophages obtained from rabbits hyperimmunized by an intravenous injection of Sendai virus were infected under the same in vitro conditions, the rate of antigen synthesis averaged a low as 23%. At the time of infection of alveolar macrophages from immunized rabbits (immune macrophages), cell aggregation at an MOI 50 and cell fusion at an MOI 500 were found 24 h after infection, and these reactions were never encountered after the infection of nonimmune macrophages. When the immune macrophages were either pretreated by trypsin or incubated in medium at pH 4.0, the infection no longer caused the aggregation. The supernatant fluid obtained after incubation at pH 4.0 contained neutralizing antibody to Sendai virus. Conversely, when nonimmune macrophages were incubated in the presence of rabbit anti-Sendai virus serum or purified immunoglobulin G, the same aggregation reaction occurred after virus infection. Ultraviolet light-killed Sendai virus could be used as the counterpart of alive virus in the same aggregation reaction. These results suggest that the aggregation reaction of the immune macrophages could be attributed to the presence of specific cytophilic antibodies on their surface.

Chick-mouse heterokaryons were obtained by UV-Sendai virus-induced fusion of chick erythrocytes with thymidine (dT) kinase-deficient mouse fibroblast [LM(TK(-))] cells. Autoradiographic studies demonstrated that 1 day after fusion, [(3)H]dT was incorporated into both red blood cell and LM(TK(-)) nuclei of 23% of the heterokaryons. Self-fused LM(TK(-)) cells failed to incorporate [(3)H]dT into nuclear DNA. 15 clonal lines of chick-mouse somatic cell hybrids [LM(TK(-))/CRB] were isolated from the heterokaryons by cultivating them in selective hypoxanthine-aminopterin-thymidine-glycine medium. LM(TK(-)) and chick erythrocytes exhibited little, if any, cytosol dT kinase activity. In contrast, all 15 LM(TK(-))/CRB lines contained levels of cytosol dT kinase activity comparable to that found in chick embryo cells. Disk polyacrylamide gel electrophoresis and isoelectric focusing analyses demonstrated that the LM(TK(-))/CRB cells contained chick cytosol, but not mouse cytosol dT kinase. The LM(TK(-))/CRB cells also contained mouse mitochondrial, but not chick mitochondrial dT kinase. Hence, the clonal lines were somatic cell hybrids and not LM(TK(-)) cell revertants. The experiments demonstrate that chick erythrocyte cytosol dT kinase can be activated in heterokaryons and in hybrid cells, most likely as a result of functions supplied by mouse fibroblast cells.

Syngeneic spleen cells (SPC) sensitized in vitro with noninfectious NVJ were shown to effectively stimulate mice to generate the HVJ-specific cell-mediated immunity for second set rejection (SSR) of virus-infected syngeneic leukemia cells. As few as 10(4) live but not disrupted SPC either infected with a temperature-sensitive mutant of HVJ (HVJts) or sensitized passively with ultraviolet (UV)-inactivated HVJts were active as immunogen. Syngeneic SPC as the carrier of virus could be replaced by allogeneic SPC or L cells, a fibroblast cell line, without reduction of the immunogenicity. Further study demonstrated that a special density of antigen on the surface of HVJts-sensitized SPC is required for high immunogenicity. It was suggested that live cells appropriately sensitized with noninfectious virus would serve as an excellent vaccine for virus-specific cell-mediated immunity.

The immunoprophylactic effects of nasal vaccination with 13 different kinds of inactivated Sendai virus vaccines were compared by contact exposure to infector mice. Efficacies of the vaccines were evaluated on the basis of the presence of virus-infected cells by immunofluorescent examination of the entire respiratory tract, including the nasal mucosa. A single or double inoculations of B-propiolactone (0.5%)-vaccine promoted the infection in the respiratory tract, particularly in the nasal mucosa, whereas three inoculations of B-propiolactone (0.2%)-vaccine provided considerable protection throughout the respiratory tract with only slight development of serum HI titer. Formalin (0.1%)-vaccine and UV irradiated-vaccine strongly protected the nasal mucosa from infection, but did not sufficiently safeguard the lower respiratory tract even with three vaccinations despite adequate development of serum antibody. Nearly complete protection of the entire respiratory tract was induced with six to eight inoculations of a vaccine treated excessively with both UV rays and 1% formalin, without significant development of serum antibody. Out of eight thermal vaccines, five (inactivated at 23 C, 30 C, 37 C and 7 C, and 30 C and 7 C) provided strong protection against infection when inoculated three times. The others inactivated at higher temperatures (37 C, 50 C, or 60 C) were not so protective. High serum HI titers developed, on the whole, with the drop in the temperature required for inactivating the virus. In eight immune mouse groups in which infection was strongly suppressed in the entire respiratory tract, most of the mice harbored less than 50 viral antigen-positive cells in their nasal mucosa in the postexposure period. The number of the cells was assumed to be a useful criterion for evaluation of vaccine efficacy.

The method of transfection of eukaryotic cells by plasmid DNA and production of recombinant vaccinia virus is described. The method is based on DNA encapsulation into reverse evaporation vesicles containing gangliosides, with a subsequent incubation of liposomes with vaccinia-infected cells in the presence of UV-inactivated Sendai virus. The recombinant DNA of vaccinia virus containing part of env gene of HTLV-3/LAV virus was obtained.

The target sizes associated with fusion and hemolysis carried out by Sendai virus envelope glycoproteins were determined by radiation inactivation analysis. The target size for influenza virus mediated fusion with erythrocyte ghosts at pH 5.0 was also determined for comparison; a value of 57 +/- 15 kDa was found, indistinguishable from that reported previously for influenza-mediated fusion of cardiolipin liposomes [Gibson, S., Jung, C. Y., Takahashi, M., & Lenard, J. (1986) Biochemistry 25, 6264-6268]. Sendai-mediated fusion with erythrocyte ghosts at pH 7.0 was likewise inactivated exponentially with increasing radiation dose, yielding a target size of 60 +/- 6 kDa, a value consistent with the molecular weight of a single F-protein molecule. The inactivation curve for Sendai-mediated fusion with cardiolipin liposomes at pH 7.0, however, was more complex. Assuming a "multiple target-single hit" model, the target consisted of 2-3 units of ca. 60 kDa each. A similar target was seen if the liposomes contained 10% gangliosides or if the reaction was measured at pH 5.0, suggesting that fusion occurred by the same mechanism at high and low pH. A target size of 261 +/- 48 kDa was found for Sendai-induced hemolysis, in contrast with influenza, which had a more complex target size for this activity (Gibson et al., 1986). Sendai virus fusion thus occurs by different mechanisms depending upon the nature of the target membrane, since it is mediated by different functional units. Hemolysis is mediated by a functional unit different from that associated with erythrocyte ghost fusion or with cardiolipin liposome fusion.

The steady-state fluorescence anisotropy measurements on influenza and parainfluenza viruses, showed no changes in the microviscosity of the viral membranes after exposure to UV-irradiation, when a fluorescent probe was used, but the intrinsic fluorescence of viral proteins presented, under the same experimental conditions, a significant difference of anisotropy behaviour in the two viruses used.

Treatment of Sendai virus with p-azidophenyl-6-chloro-6-deoxy-beta-D-glucopyranoside (APG) caused chemical modification of the viral envelope under UV irradiation, which did not affect the hemagglutinin activity of the virus but inhibited the hemolytic activity. Also, the transfer of phospholipid from the viral envelope to chicken erythrocytes was measured using a spinlabel technique by electron spin resonance (ESR). In this experiment, the phospholipid transfer was depressed by the treatment with APG under the conditions which inhibited the hemolytic activity of the virus. These results suggest that APG bound covalently to lipid may disturb the specific interaction between the protein and the lipid of the viral envelope, resulting in the inhibition of the hemolytic activity. The effects of APG on the hemolysis and phospholipid transfer were compared with the results for the concanavalin A- and amphotericin B-treated viruses.

HeLa cells infected with Sendai virus (SV) acquired the ability to activate the alternative pathway (AP) of guinea-pig complement without antibody reaction. For induction of complement activating ability (CAA), at least 2 hr incubation of the infected cells was required. However, ultraviolet (UV)-irradiation (8400 erg/mm2) of SV did not impair the inducibility of CAA and 51Cr-labelled HeLa cells infected with UV-irradiated SV (UV-SV) became readily cytolysed by C4-deficient guinea-pig complement without antigen-antibody reaction. This phenomenon may represent a primary in vivo defence mechanism against SV infection by eliminating the virus-infected cells via activation of complement on the cell surface. Although SV-infected HeLa cells activated the AP of guinea-pig complement, they did not activate the AP of human or mouse complement. The species restriction in the CAA induced by SV infection may result in different pathological manifestations in virus-infected animals and may affect their susceptibility to this type of infection.

Homologous interference between a temperature-sensitive small plaque mutant (HVJ-pB) derived from an HVJ (haemagglutinating virus of Japan - the Sendai strain of parainfluenza I virus) carrier culture of BHK cells and the original wild-type virus (HVJ-W) has been investigated. Prior infection of LLCMK2, HeLa, BHK or mouse L cells with HVJ-pB, both at permissive and non-permissive temperatures, for 24 h resulted in a reduced yield of superinfecting HVJ-W, reflecting a smaller number of cells capable of producing the superinfecting virus. However, HVJ-pB did not interfere with the replication of vesicular stomatitis virus, Sindbis virus or Newcastle disease virus. Interference in this system seems to be due to inhibition of the attachment of superinfecting HVJ-W as a result of intracellular mechanisms operating at a late stage in the replication of the interfering virus. There is also blocking or destruction of cellular receptors by extra-cellular particles of the interfering virus. Protein synthesis coded for by the complete virus genome is required to establish and maintain the interference, and treatment with actinomycin D has no effect on the interference phenomenon.

The specific action of T4 endonuclease V on damaged DNA in xeroderma pigmentosum cells was examined using an in vivo assay system with hemagglutinating virus of Japan (Sendai virus) inactivated by UV light. A clear dose response was observed between the level of UV-induce unscheduled DNA synthesis of xeroderma pigmentosum cells and the amount of T4 endonuclease V activity added. The T4 enzyme was unstable in human cells, and its half-life was 3 hr. Fractions derived from an extract of Escherichia coli infected with T4V1, a mutant defective in the endonuclease V gene, showed no ability to restore the UV-induced unscheduled DNA synthesis of xeroderma pigmentosum cells. However, fractions derived from an extract of T4D-infected E. coli with endonuclease V activity were effective. The T4 enzyme was effective in xeroderma pigmentosum cells on DNA damaged by UV light but not in cells damaged by 4-nitroquinoline 1-oxide. The results of these experiments show that the T4 enzyme has a specific action on human cell DNA in vivo. Treatment with the T4 enzyme increased the survival of group A xeroderma pigmentosum cells after UV irradiation.

Sendai and measles viruses were tested for their interferon-induced capacity in human lymphoblast cells. Sendai virus reproducibly induced considerable amounts of interferon (I research reference interferon unit/10(3) cells), but no increase in infectious virus titre was observed. Two Edmonston-Enders strains of measles virus grew very well. The attenuated (A) strain was a good interferon inducer (4 units/10(3) cells), while the virulent (V) strain induced only minimal amounts at a high multiplicity of infection. Pre-treatment of the cells with 5-iododeoxyuridine (IdUrd) had no effect on the growth of Sendai or EE measles virus and on interferon yields induced by Sendai virus. It slightly potentiated interferon induction by EE measles virus. Partial u.v.-light-inactivation of Sendai virus infectivity resulted in a parallel loss in interferon-inducing capacity.

Studies were made on the abilities of Sendai virus envelope components to induce primary and secondary generations of virus-specific cytotoxic mouse T lymphocytes (CTL). The primary CTL response in BALB/c mice was induced by reassembled envelope particles that had fusion activity but not by envelope glycoproteins without fusion activity, although both preparations induced a humoral immune response. Reconstitution of membrane-bound envelope proteins from envelope glycoproteins with lipids restored the fusion activity and the capacity to induce CTL. Target cells susceptible to virus-specific CTL could be induced by reassembled envelope particles, but not by envelope glycoproteins or LLC-MK2 cell-grown Sendai virus, neither of which had fusion activity. On the other hand, all the viruses and envelope components tested were found to stimulate a virus-specific CTL response in the in vitro secondary generation of CTL from virus-primed spleen cells. These results suggest that Senaei virus fusion activity is involved in primary induction of the CTL response as well as in target cell formation, but that it is not essential for secondary stimulation of the CTL response.

Radiation inactivation analysis was used to determine the size of the functional unit responsible for fusion of vesicular stomatitis virus (VSV) with cardiolipin or phosphatidylcholine-phosphatidylethanolamine (1:1) liposomes, and for VSV-induced hemolysis. When radiation-insensitive background values were subtracted, the calculated functional units for all three activities were similar, ranging from 866 to 957 kDa, equivalent to about 15 G protein molecules. This is in striking contrast to results of similar studies with influenza and Sendai viruses, in which the functional unit corresponded in size to a single fusion protein monomer, and suggests that VSV fusion may occur by a different mechanism.

Guinea-pig line 10 tumor (GPL10) cells, which were transplantable in strain 2 guinea-pigs, gained the capacity to activate the homologous alternative complement pathway (ACP) following infection with ultraviolet-irradiated Sendai virus (UV-SV). The ACP activation of homologous serum on the UV-SV-infected GPL10 cells (UV-SV-GPL10 cells) resulted in cytolysis of the tumor cells. This indicates that UV-SV infection induced not only the capacity to activate complement on the GPL10 cells, but also made them sensitive to complement attack, because GPL10 cells have been demonstrated to be resistant to the reaction of homologous complement via the classical pathway in the presence of rabbit antibody. Furthermore, UV-SV-GPL10 cells prepared with as little as 0.1 plaque-forming unit equivalent of UV-SV per cell showed suppressed growth in syngeneic strain 2 guinea-pigs. The essential role of complement in the elimination of UV-SV-GPL10 cells in vivo was confirmed by demonstrating that UV-SV-GPL10 could grow in guinea-pigs whose complement had been depleted by administration of cobra venom factor.

The pathogenicity of a strain of parainfluenza type 1 virus, isolated from the cultured brain cells from multiple sclerosis patients and designated as 6/94 virus, was studied in mouse brain. Selective degeneration of cerebral white matter, preceded by mononuclear cell infiltration, and ependymitis were prominent pathological features of the mouse intracerebrally inoculated with 6/94 virus. After inactivation by ultraviolet light the virus was still capable of producing the inflammatory and degenerative white matter lesions, although the severity of ependymitis was substantially reduced.

Sendai mutant ts271 contains less than 5% of the amount of HN glycoprotein found in wild-type Sendai. Fusion of this mutant with cardiolipin liposomes revealed no differences from the wild-type virus with regard to specific activity, pH dependence, or radiation inactivation. Target sizes of both mutant and wild-type viral proteins were determined by the radiation-induced disappearance of each band from an SDS-polyacrylamide gel and no differences were found. Of the viral proteins, only F had a target size corresponding to the monomer molecular weight, ca. 60 kDa, identical to the minimum unit previously determined by functional assay for Sendai virus-erythrocyte membrane fusion (K. Bundo-Morita, S. Gibson, and J. Lenard, Biochemistry 26, 6223-6227 (1987)). This provides additional evidence that F alone is the active protein mediating Sendai-erythrocyte fusion. It is concluded that the HN protein is unlikely to mediate any fusion reactions of the intact virions, either with biological membranes or with cardiolipin liposomes.

Several murine Sendai virus-specific T-cell clones were characterized in vitro and in vivo. All T-cell clones were phenotypically Thy-1.2+, and most clones were Lyt-1+,2-; one T-cell clone was Lyt-1-,2-. Some of the clones proliferated in response to antigen presented on I region-compatible stimulator cells. Proliferation could be inhibited by monoclonal antibodies directed against class II antigens. Clones which proliferated in response to antigen secreted lymphokines which could be identified as Interleukin 2 and Interleukin 3. All of the clones tested in vivo induced a delayed-type hypersensitivity response in syngeneic mice challenged with antigens. Depending on the experimental conditions chosen, Interleukin 2-producing clones as well as non-Interleukin 2-producing clones mediated help for stimulation of cytolytic T lymphocytes.

The effect of treating Sendai Virus with reducing agents as dithiothreitol (DTT) or with ultraviolet (U.V.)--irradiation, upon the induction of an immune response in Balb/c mice, was studied. Humoral immune response was assessed measuring the haemolysis inhibiting (HLI) antibody titre in serum 1 and 2 weeks after injection. The native viral particles elicited an HLI antibody titre proportional to the amount of viral proteins injected in the range of 8-300 micrograms. The U.V.--irradiation of viral inoculum produced a 6-7 fold decrease in HLI antibody titre even raising the injected dose to 740 micrograms. Similarly, dithiothreitol treatment, which abolishes binding and fusogenic properties of Sendai virus particles, affected the HLI antibody titre about 2-6 fold.

The purpose of this review was to focus mainly on the molecular events related to the progression of cells through the G2 period to examine the cause for G2-arrest in mammalian cells after exposure to various anticancer drugs. With few exceptions, most of the eukaryotic cells exhibit a G2 period in their life cycles. The G2 period, which separates S phase from mitosis, represents the time necessary for the synthesis of the various components related to the condensation of chromosomes, assembly of the mitotic spindle, and cytokinesis. Continued synthesis of RNA and protein is necessary for the successful completion of G2 and the initiation of mitosis. Inhibition of RNA and protein synthesis, replacement of phenylalanine by its analog paraversible G2 arrest in cultured cells. Exposure of cells to certain antineoplastic drugs also blocks cells preferentially in G2. This irreversible drug-induced G2 arrest is associated with extensive chromosome damage. The G2-arrested cells were found to be deficient in certain proteins that may be specific for the G2-mitotic transition. These mitotic or chromosome condensation factors synthesized during the G2 period, reach their maximum levels at mitosis. A preliminary characterization of the chromosome condensation factor revealed that it is a heat labile, Ca2+-sensitive, nondialyzable protein with a sedimentation value of 4-5S.

Ultraviolet-inactivated Sendai virus used in conjunction with a selective system reproducibly gives high yields of viable hybrid cell lines. With two different crosses, the frequency of hybrid colonies obtained after virus treatment was found to be 100 times greater than the frequency of spontaneous hybrids, and this increase was found to show little variation between 33 and 1000 hemagglutinating units of virus. No differences have been found between the properties of hybrids obtained after Sendai treatment and those obtained from untreated mixed cultures of parental cells.
