5. Physicochemical Characteristics of Commercially Available Greek Yoghurts.

Abstract

In the present study, the physicochemical characteristics of 108 yoghurts purchased from the Greek market have been assessed. Generally, the range of the mean pH values of samples was 3.58–4.64, of fat 0–10.8%, of protein 3.29–10.05%, of total solids 10.75–25.18%, and of ash 0.5–1.17%. Samples were categorized as strained and non-strained (traditional with a crust or plain without a crust). The milk origin was classified as being from sheep, goats, cows, mixture of sheep–goat–cow milk, or cow–donkey milk combination. A significant effect of species origin was determined for yoghurt physicochemical attributes, whereas geographical origin, mainland or island, affected yoghurt acidity only. Discriminant analysis revealed MDA, pH, acidity, syneresis, fat, and protein content and color lightness and redness as the traits responsible for the discrimination of yoghurts into milk-type classes, whereas fat, protein, and ash content, pH, and syneresis attributes were responsible for the discrimination into fat content classes. Yoghurt samples were sufficiently clustered according to their fat content, whereas protein content and species origin discriminated yoghurts to a lesser extent. This first in-depth descriptive research on a wide range of samples of the renowned Greek yoghurt showed that several physicochemical characteristics can be used for sample discrimination. Abstract

Background: The egg donation model offers an opportunity to isolate the male factor and evaluate its impact on IVF–intracytoplasmic sperm injection and pregnancy outcomes. Objective: To study the effect of non‐obstructive azoospermia on intracytoplasmic sperm injection and pregnancy outcomes compared with severe oligozoospermia and mild‐to‐moderate oligozoospermia in egg recipient cycles. Materials and methods: This is a retrospective longitudinal cohort study, including 1594 patients who underwent intracytoplasmic sperm injection in egg recipient cycles with preimplantation genetic testing for aneuploidies. The cohort was divided into three groups: couples with non‐obstructive azoospermia accounting for 479 patients (30%); couples with severe oligozoospermia (sperm number <5 × 106/mL), accounting for 442 patients (27.8%); couples with mild‐to‐moderate oligozoospermia, with sperm number >5 × 106and <15 × 106/mL, accounting for 673 patients (42.2%). Results: The fertilisation rate was significantly reduced in the non‐obstructive azoospermia group as compared with the severe oligozoospermia and the mild‐to‐moderate oligozoospermia group: 30.3% versus 63% and 77.3% (p < 0.05). Logistic regression analysis adjusted for confounders highlighted non‐obstructive azoospermia as a negative predictor of obtaining a euploid blastocyst both per injected oocyte and per obtained blastocyst. The miscarriage rate in the non‐obstructive azoospermia group was 11.8%; higher than the severe oligozoospermia and mild‐to‐moderate oligozoospermia groups (7% and 2.7%) (p < 0.05). The live birth rate per embryo transfer (ET) was significantly lower in the non‐obstructive azoospermia group compared with the severe oligozoospermia and the mild‐to‐moderate oligozoospermia group (20.4% vs. 30.3% and 35.4%, p < 0.05). The risk of preterm labour was significantly higher in the non‐obstructive azoospermia group, compared with the severe oligozoospermia and mild‐to‐moderate oligozoospermia group (55.1% vs. 46.8% and 16.1%, p < 0.001), and this difference was observed in both singleton and twin pregnancies. Discussion and conclusion: In our retrospective comparative study, non‐obstructive azoospermia significantly affects early embryonic potential and live birth rates per cycle and per embryo transfer. It is also associated with higher risk of preterm birth. Future prospective multi‐centre studies are needed to highlight the effect of sperm quality on ART and pregnancy outcomes. 31. Comparative Study between Dietary Nanoelemental, Inorganic, and Organic Selenium in Broiler Chickens: Effects on Meat Fatty Acid Composition and Oxidative Stability.

Abstract

The present study investigated the impact of dietary supplementation with nano-elemental, inorganic, and organic selenium (Se) on the Se content, fatty acid (FA) composition, and oxidative stability of meat in 150 one-day-old broiler chickens. The broiler chickens were allotted into three groups: control (C), SS+SY, and SeNP. The C group received a control diet without any added Se, while the SS+SY and SeNP groups were fed diets containing 0.4 mg Se/kg from a combination of sodium selenite and selenium yeast (SS+SY at a 1:1 ratio) or elemental Se nanoparticles (SeNP), respectively. Breast meat samples were collected from 10 broiler chickens per diet group (2 per replicate) at 42 days of age for the analysis of Se content, FA composition, and oxidative stability. The findings of the study revealed that the Se levels in the breast tissue significantly increased (p < 0.05) and the concentrations of malondialdehyde (MDA), a marker of oxidative stress, decreased (p < 0.05) with the inclusion of SS+SY and SeNP in the diet. Furthermore, the levels of 22:6n − 3 (docosahexaenoic acid) and total n − 3 FA significantly increased (p < 0.05) in the breast meat of broiler chickens supplemented with SeNP compared to the C and SS+SY groups. In conclusion, both dietary supplementation with SeNP and SS+SY had a positive impact on the Se content and oxidative stability of the breast meat. However, SeNP supplementation resulted in a more desirable modification of the FA composition. Abstract

Although the inclusion of polyunsaturated fatty acids (PUFAs) in ruminants' diets appears to be a well-documented strategy to enrich milk with PUFAs, several gene networks that regulate milk synthesis and mammary gland homeostasis could be impaired. The objective of this literature review is to assess the effects of nutritional strategies focused on enriching milk with PUFAs on gene networks regulating mammary gland function and lipogenesis, as well as the impact of feed additives and bioactive compounds with prominent antioxidant potential on immune-oxidative transcriptional profiling, as a part of mammary gland homeostasis and health. The findings support the conclusion that PUFAs' inclusion in ruminants' diets more strongly downregulate the stearoyl-CoA desaturase (SCD) gene compared to other key genes involved in de novo fatty acid synthesis in the mammary gland. Additionally, it was revealed that seed oils rich in linoleic and linolenic acids have no such strong impact on networks that regulate lipogenic homeostasis compared to marine oils rich in eicosapentaenoic and docosahexaenoic acids. Furthermore, ample evidence supports that cows and sheep are more prone to the suppression of lipogenesis pathways compared to goats under the impact of dietary marine PUFAs. On the other hand, the inclusion of feed additives and bioactive compounds with prominent antioxidant potential in ruminants' diets can strengthen mammary gland immune-oxidative status. Considering that PUFA's high propensity to oxidation can induce a cascade of pro-oxidant incidences, the simultaneous supplementation of antioxidant compounds and especially polyphenols may alleviate any side effects caused by PUFA overload in the mammary gland. In conclusion, future studies should deeply investigate the effects of PUFAs on mammary gland gene networks in an effort to holistically understand their impact on both milk fat depression syndrome and homeostatic disturbance. Abstract

Bladder Cancer (BC) is11th most common malignancy. In terms of pathology, the vast majority of patients suffer from transitional cell carcinoma. Apart from this common type of BC, there are many morphological subtypes with different oncological characteristics. Plasmacytoid BC is a well-recognized subtype of BC with great diagnostic importance as it usually presents in locally advanced or even metastatic stage.The objective of this study was to evaluate our experience in diagnosing and treating patients with this rare BC subtype.A retrospective analysis of 5 patients diagnosed with plasmacytoid BC in our department during they ears 2014-2016 was performed. Transurethral resection of the tumors was performed in all patients and pathology diagnosis of plasmacytoid variant was based on several morphologic and immunohistochemical parameters. Staging included abdominal and thoracic CT.3 of 5 patients were diagnosed with metastatic disease. These patients were referred to the oncology department. 2 patients presented with non-metastatic BC after initial staging and thus a radical cystectomy was performed. Follow up of all patients was carried out and their survival was recorded.We concluded that despite the fact that the plasmacytoid variant of BC is rare, it is important to take into account the pathologic and clinical features of this tumor in order to manage the optimal treatment of this poor prognosis cancer.INTRODUCCIÓN: El cáncer de vejiga es el 11o cáncer mas común. Del punto de vista de la patología, la mayoría de pacientes tienen carcinoma urotelial de vejiga.A parte de la forma mas común, el cáncer de vejiga plasmocitoide es un subtipo ampliamente reconocido de cáncer de vejiga con una gran importancia diagnóstica ya que habitualmente se presenta como localmente avanzadoo en estadio metastático.OBJETIVO: El objetivo de este estudio fue evaluar nuestra experiencia en el diagnóstico y tratamiento en pacientes con esta entidad.MÉTODOS: Análisis retrospectivo de 5 pacientes diagnosticados con cáncer de vejiga plasmocitoide en nuestro departamento entre 2014 y 2016. Se realizó una resección transuretral de vejiga en todos los pacientes y un análisis histopatológico que demostró la variante plasmocitoide del punto de vista morfológico e immunohistoquímico. El estadiaje incluyó TAC toracoabdominopélvico.RESULTADOS: 3 de los 5 pacientes fueron diagnosticado sen estadio metastático. Estos pacientes pasaron a cargo del departamento de oncología medica. 2 pacientes se diagnosticaron en estadio localizado por lo que se realizó una cistectomía radical. Se realizó el seguimiento y se determinó la supervivencia de estos pacientes. CONCLUSIONES: Se concluye que todo y que el diagnóstico de cáncer de vejiga plasmocitoide es raro; es importante reconocer esta variedad histológica para poderdar un manejo optimo a esta variedad con muy mal pronostico.

Abstract

The measurement of wave forces acting on marine structures is a complicated task, both during physical experiments and, even more so, in the field. Force transducers adopted in laboratory experiments require a minimum level of structural movement, thus violating the main assumption of fully rigid structure and introducing a dynamic response of the system. Sometimes the induced vibrations are so intense that they completely nullify the reliability of the experiments. On-site, it is even more complex, since there are no force transducers of the size and capacity able to measure such massive force intensity acting over the very large domain of a marine structure. To this end, this investigation proposes a Bayesian methodology aimed to remove the undesired effects from the directly (laboratory applications) or indirectly (field applications) measured wave forces. The paper presents three applications of the method: i) a theoretical application on a synthetic signal for which MATLAB® procedures are provided, ii) an experimental application on laboratory data collected during experiments aimed to model broken wave loading on a cylinder upon a shoal and iii) a field application designed to reconstruct the wave force that generated recorded vibrations on the Wolf Rock lighthouse during Hurricane Ophelia. The proposed methodology allows the inclusion of existing information on breaking and broken wave forces through the process-based informative prior distributions, while it also provides the formal framework for uncertainty quantification of the results through the posterior distribution. Notable findings are that the broken wave loading shows similar features for both laboratory and field data. Abstract

The survivability analysis of offshore rock lighthouses requires several assumptions of the pressure distribution due to the breaking wave loading (Raby et al. (2019), Antonini et al. (2019). Due to the peculiar bathymetries and topographies of rock pinnacles, there is no dedicated formula to properly quantify the loads induced by the breaking waves on offshore rock lighthouses. Wienke’s formula (Wienke and Oumeraci (2005) was used in this study to estimate the loads, even though it was not derived for breaking waves on offshore rock lighthouses, but rather for the breaking wave loading on offshore monopiles. However, a thorough sensitivity analysis of the effects of the assumed pressure distribution has never been performed. In this paper, by means of the Wolf Rock lighthouse distinct element model, we quantified the influence of the pressure distributions on the dynamic response of the lighthouse structure. Different pressure distributions were tested, while keeping the initial wave impact area and pressure integrated force unchanged, in order to quantify the effect of different pressure distribution patterns. The pressure distributions considered in this paper showed subtle differences in the overall dynamic structure responses; however, pressure distribution #3, based on published experimental data such as Tanimoto et al. (1986) and Zhou et al. (1991) gave the largest displacements. This scenario has a triangular pressure distribution with a peak at the centroid of the impact area, which then linearly decreases to zero at the top and bottom boundaries of the impact area. The azimuthal horizontal distribution was adopted from Wienke and Oumeraci’s work (2005). 49. Rocking of offshore lighthouses under extreme wave impacts: Limit analysis, analytic formulations and distinct element method

Abstract

This study describes the structural response of historic lighthouses to extreme wave impacts. Located offshore on exposed rocks, 19th Century lighthouses were built with large interlocked granite blocks and have survived weathering for nearly two centuries. Under extreme wave impacts, lighthouses of this structural typology may uplift and rock, whereas sliding is prevented by the vertical interlocking. The uplift and sliding thresholds calculated with the limit analysis method reveal why this structural system is capable of bearing extreme wave impacts without failure. The ingenious vertical keying is proven to be a major characteristic that contributes to the resilience of these lighthouses. The structural response is explained with the use of analytic formulations of the rocking motion. Detailed analysis of the response to wave impact is conducted with reference to Wolf Rock lighthouse. The impact wave corresponding to a 250-year effective return period is identified using non-stationary Bayesian extreme analysis. Moreover, wave flume tests on a scaled cylindrical structure were performed to identify the wave impact force time-history shapes. Based on two waves: a theoretical time-history based on existing models in the literature and the measured time-histories from small-scale experiments, a series of synthetic force time-history sequences are generated for the purposes of a parametric analysis. This parametric analysis, with the Distinct Element Method, using the commercial software 3DEC, reveals the influence of the duration and shape of the force time-history function. For impacts with the same impulse values, shorter time impacts produce the most intense opening of joints, despite causing smaller horizontal displacements.
