A method is designed to quickly form protein hydrogels, based on the self-assembly of highly concentrated lysozyme solutions in acidic conditions. Their properties can be easily modulated by selecting the curing temperature. Molecular insights on the gelation pathway, derived by in situ FTIR spectroscopy, are related to calorimetric and rheological results, providing a consistent picture on structure-property correlations. In these self-crowded samples, the thermal unfolding induces the rapid formation of amyloid aggregates, leading to temperature-dependent quasi-stationary levels of antiparallel cross β-sheet links, attributed to kinetically trapped oligomers. Upon subsequent cooling, thermoreversible hydrogels develop by the formation of interoligomer contacts. Through heating/cooling cycles, the starting solutions can be largely recovered back, due to oligomer-to-monomer dissociation and refolding. Overall, transparent protein hydrogels can be easily formed in self-crowding conditions and their properties explained, considering the formation of interconnected amyloid oligomers. This type of biomaterial might be relevant in different fields, along with analogous systems of a fibrillar nature more commonly considered.

Two symmetric quadrupolar cationic push-pull compounds with a central electron-acceptor (N+-methylpyrydinium, A+) and different lateral electron-donors, (N,N-dimethylamino and N,N-diphenylamino, D) in a D-π-A+-π-D arrangement, were investigated together with their dipolar counterparts (D-π-A+) for their excited-state dynamics and NLO properties. As for the quadrupolar compounds, attention was focused on excited-state symmetry breaking (ESSB), which leads to a relaxed dipolar excited state. Both electron charge displacements and structural rearrangements were recognized in the excited-state dynamics of these molecules by resorting to femtosecond-resolved broadband fluorescence up-conversion experiments and advanced data analysis, used as a valuable alternative approach for fluorescent molecules compared to time-resolved IR spectroscopy, only suitable for compounds bearing IR markers. Specifically, intramolecular charge transfer (ICT) was found to be guided by ultrafast inertial solvation, while diffusive solvation can drive the twisting of lateral groups to originate twisted-ICT (TICT) states on a picosecond time scale. Yet still, only the bis-N,N-diphenylamino-substituted compound undergoes ESSB, in both highly and sparingly polar solvents, provided that it can experience large amplitude motions to a fully symmetry-broken TICT state. Besides well-known solvation effects, this structural requirement proved to be a necessary condition for these quadrupolar cations to undergo ESSB. In fact, a more efficient uncoupling between the out-of-plane D and A+ groups in the TICT state allows a greater stabilization gained through solvation, relative to the bis-N,N-dimethylamino-substituted derivative, which instead maintains its symmetry. This different behavior parallels the two-photon absorption (TPA) ability, which is greatly enhanced in the case of the bis-N,N-diphenylamino-substituted compound, paving the way for cutting-edge bio-imaging applications.

Attaching stable radicals to organic chromophores is an effective method to enhance the intersystem crossing (ISC) of the chromophores. Herein we prepared perylene-oxoverdazyl dyads either by directly connecting the two units or using an intervening phenyl spacer. We investigated the effect of the radical on the photophysical properties of perylene and observed strong fluorescence quenching due to radical enhanced ISC (REISC). Compared with a previously reported perylene-fused nitroxide radical compound (triplet lifetime, ?T=0.1 ?s), these new adducts show a longer-lived triplet excited state (?T=9.5 ?s). Based on the singlet oxygen quantum yield (??=7 %) and study of the triplet state, we propose that the radical enhanced internal conversion also plays a role in the relaxation of the excited state. Femtosecond fluorescence up-conversion indicates a fast decay of the excited state (Q1) at ~80 ps time scale. Time-resolved electron paramagnetic resonance (TREPR) spectral study confirmed the formation of the quartet sate. We observed triplet and quartet states simultaneously with weights of 0.7 and 0.3, respectively. This is attributed to two different conformations of the molecule at excited state. DFT computations showed that the interaction between the radical and the chromophore is ferromagnetic (J>0, 0.05~0.10 eV).

Although the photophysics of BODIPY compounds has been widely investigated in the last few years, their analogue N-BODIPY compounds, with nitrogen substitution at the Boron center, did not receive comparable attention. In this work we report about the systhesis and photochemical characterization of a substituted N-BODIPY compound, by means of a combined theoretical and spectroscopic approach. Compared to a standard BODIPY, the compound under investigation presents a lower fluorescence quantum yield (QY) in the visible region. The excited state relaxation dynamics of the dye was studied in different solvents, showing further fluorescence quenching in polar solvents, and a excited state decay rates strongly dependent on the environment polarity. The role of the pendant moieties and the involvement of charge transfer states in the excited state dynamics was experimentally addressed by transient absorption spectroscopy, and further analyzed with TD-DFT calculations, which allowed precise assignment of the transient signals to the correspondent electronic configuration. The complete picture of the N-BODIPY behavior shows the presence of both charge transfer and localized states, influencing the observed photophysics to different amounts, depending on the excitation conditions and the surrounding environment.

Self assembled ordered structures, such as H- or J- type molecular aggregates of organic chromophores, are extremely interesting materials for different optoelectronic applications. In this work we present a novel characterization of light harvesting nanotubes formed by self-assembling of amphiphilic cyanine dyes in water, through a combined ultrafast spectroscopic and theoretical approach. In the condition of low inhomogeneous disorder at low temperature, broadband transient absorption spectroscopy revealed the presence of an unusual ultrafast behavior of the aggregate, manifested through intense and peculiar oscillations of the kinetic traces, lasting tens of picoseconds. Theoretical simulations were performed by adapting a model which grasps vibronically coherent effects in the double wall nanotube system experiencing inter-wall energy transfer. Good agreement between model predictions and experimental observations were obtained under the assumption of coupling between bright and dark electronic states. The model clarified the vibronic origin of the observed oscillations, evidencing new important pieces of information about transport mechanisms and excitonic interactions in these complex molecular systems.

Luminescent solar concentrators (LSCs) are devices designed to efficiently collect both direct and diffuse solar radiation and concentrate it on photovoltaic cells to foster their use in building-integrated photovoltaics (BIPV). The optimization of LSC performances involves the adjustment of both the fluorophore and the guest polymer matrix. On this account, we investigated a series of high quantum yield, donor–acceptor–donor (D–A–D) photostable fluorophores (DQ1–5), presenting a central quinoxalinic acceptor core, not previously employed in LSCs, and triarylamines or phenothiazine as donor groups. The molecules were also decorated with alkyl chains on the central core and/or the donor groups, to explore their compatibility with the poly(methyl methacrylate) (PMMA) and poly(cyclohexyl methacrylate) (PCMA) matrices utilized in this study. The PMMA and PCMA films (25 μm thick), containing 0.2–2.2 wt% of DQ1–5, absorbed in the 370–550 nm range and presented emission maxima at 550–600 nm, with fluorescence quantum yields higher than 40% even at the highest doping contents. Notably, the DQ1/PMMA thin-films showed enhanced phase compatibility and excellent quantum yields, i.e., >95%. Accordingly, they were designed to obtain 25 cm2 area LSCs with remarkable internal (ηint) and external (ηext) photon efficiencies of 42.9% and 6.2%, respectively, higher than those observed from state-of-the-art devices based on the Lumogen Red 305 (LR305) as the reference fluorophore. Overall, these were the best results ever achieved in our laboratory for thin-film LSCs built with organic fluorescent emitters.

Oxoverdazyl (Vz) radical units were covalently linked to the naphthalenediimide (NDI) chromophore to study the effect of the radical on the photophysical properties, especially the radical enhanced intersystem crossing (REISC), which is a promising approach to develop heavy-atom-free triplet photosensitizers. Rigid phenyl or ethynylphenyl linkers between the two moieties were used, thus REISC and formation of doublet (D1 , total spin quantum number S=1/2) and quartet states (Q1 , S=3/2) are anticipated. Academic, industry, regulatory leaders and patient advocates in cancer clinical research met in November 2018 at the Innovation and Biomarkers in Cancer Drug Development meeting in Brussels to address the existing dichotomy between increasing calls for personalised oncology approaches based on individual molecular profiles and the need to make resource and regulatory decisions at the societal level in differing health-care delivery systems around the globe. Novel clinical trial designs, the utility and limitations of real-world evidence (RWE) and emerging technologies for profiling patient tumours and tumour-derived DNA in plasma were discussed. While randomised clinical trials remain the gold standard approach to defining clinical utility of local and systemic therapeutic interventions, the broader adoption of comprehensive tumour profiling and novel trial designs coupled with RWE may allow patient and physician autonomy to be appropriately balanced with broader assessments of safety and overall societal benefit. Flavohemoglobins have the particular capability of binding unsaturated and cyclopropanated fatty acids as free acids or phospholipids. Fatty acid binding to the ferric heme results in a weak but direct bonding interaction. Ferrous and ferric protein, in presence or absence of a bound lipid molecule, have been characterized by transient absorption spectroscopy. Measurements have been also carried out both on the ferrous deoxygenated and on the CO bound protein to investigate possible long-range interaction between the lipid acyl chain moiety and the ferrous heme. After excitation of the deoxygenated derivatives the relaxation process reveals a slow dynamics (350 ps) in lipid-bound protein but is not observed in the lipid-free protein. The latter feature and the presence of an extra contribution in the absorption spectrum, indicates that the interaction of iron heme with the acyl chain moiety occurs only in the excited electronic state and not in the ground electronic state. Data analysis highlights the formation of a charge-transfer complex in which the iron ion of the lipid-bound protein in the expanded electronic excited state, possibly represented by a high spin Fe III intermediate, is able to bind to the sixth coordination ligand placed at a distance of at 3.5 A from the iron. A very small nanosecond geminate rebinding is observed for CO adduct in lipid-free but not in the lipid-bound protein. The presence of the lipid thus appears to inhibit the mobility of CO in the heme pocket.

The introduction of tyrosine kinase inhibitors (TKIs) has improved the overall survival of chronic myeloid leukemia patients in chronic phase (CP-CML) and reduced the rate of disease-related mortality. Conflicting results have been however reported between data emerged from sponsored clinical trials and from population-based registries. Moreover, no data are so far available for patients treated with frontline second-generation TKIs, excluding those from sponsored studies. We analyzed the mortality rate of 2315 CP-CML patients treated with frontline second-generation TKIs through the Italian Medicines Agency (AIFA) registries and compared it with the ISTAT mortality rate of the general population. The estimated differences show that the increased rate of mortality in CP-CML patients is less than 1% for the class 0–29 years, stable around 2% for the intervals 30–44 years and 45–59 years, and 1.4% for the interval 60–74 years; interestingly this rate is reduced for patients aged 75 years and more as compared to the general population (− 0.65%). The difference between potential and estimated deaths is higher among women in the age classes between 30 and 74 years.

We investigate the static and dynamic states of water network during the phase transitions from double gyroid ((Formula presented.)) to double diamond ((Formula presented.)) bicontinuous cubic phases and from the latter to the reverse hexagonal (HII) phase in monolinolein based lipidic mesophases by combining FTIR and broadband dielectric spectroscopy (BDS). In both cubic(s) and HII phase, two dynamically different fractions of water are detected and attributed to bound and interstitial free water. The dynamics of the two water fractions are all slower than bulk water due to the hydrogen-bonds between water molecules and the lipid's polar headgroups and to nanoconfinement. Both FTIR and BDS results suggest that a larger fraction of water is hydrogen-bonded to the headgroup of lipids in the HII phase at higher temperature than in the cubic phase at lower temperature via H-bonds, which is different from the common expectation that the number of H-bonds should decrease with increase of temperature. Three new fluorophores (BBT2-4) with a D-A-D structure, characterised by the common benzo[1,2-d:4,5-d′]bisthiazole (BBT) as acceptor core (A) and different donor groups (D), were designed and synthesised for use in thin-film luminescent solar concentrators (LSC). The optical and spectroscopic properties of the new dyes were analysed both in solution (toluene) and in polymeric films, using static and time-resolved techniques. All the prepared molecules showed intense emissions between 460 and 550 nm with large Stokes shift (>50 nm), moderate-to-good fluorescence quantum yields (Φf) both in toluene solution (18–73%) and in poly(methyl methacrylate) (PMMA) films (10–52%) and good optical efficiencies (5.8–7.5%) as LSC. Transient absorption spectroscopy (TAS) studies highlighted that higher Φf were related to long excited-state lifetimes and that such properties were critically dependent on the polarity of the surrounding environment. Notably, homogeneous BBT4 dispersions into the less polar poly(benzyl methacrylate) (PBzMA) and poly(cyclohexyl methacrylate) (PCMA) polymeric matrices showed higher Φf (52.6% and 65.6%, respectively) than those gathered from PMMA films. Accordingly, BBT4/PCMA films revealed significant optical efficiency of 9.0% as LSC, which was comparable to that provided by the state-of-art fluorophore Lumogen Red 305 (10.1%).

Donor–acceptor Stenhouse adducts (DASAs) are negative photochromes that switch with visible light and are highly promising for applications ranging from smart materials to biological systems. However, the strong solvent dependence of the photoswitching kinetics limits their application. The nature of the photoswitching mechanism in different solvents is key for addressing the solvatochromism of DASAs, but as yet has remained elusive. Here, we employ spectroscopic analyses and TD‐DFT calculations to reveal changing solvatochromic shifts and energies of the species involved in DASA photoswitching. Time‐resolved visible pump‐probe spectroscopy suggests that the primary photochemical step remains the same, irrespective of the polarity and protic nature of the solvent. Disentangling the different factors determining the solvent‐dependence of DASA photoswitching, presented here, is crucial for the rational development of applications in a wide range of different media.

Donor–acceptor Stenhouse adducts (DASAs) are a rapidly emerging class of visible light-activatable negative photochromes. They are closely related to (mero)cyanine dyes with the sole difference being a hydroxy group in the polyene chain. The presence or absence of the hydroxy group has far-reaching consequences for the photochemistry of the compound: cyanine dyes are widely used as fluorescent probes, whereas DASAs hold great promise for visible light-triggered photoswitching. Here we analyze the photophysical properties of a DASA lacking the hydroxy group. Ultrafast time-resolved pump–probe spectroscopy in both the visible and IR region show the occurrence of E–Z photoisomerization on a 20 ps time scale, similar to the photochemical behavior of DASAs, but on a slower time scale. In contrast to the parent DASA compounds, where the initial photoisomerization is constrained to a single position (next to the hydroxy group), 1H NMR in situ-irradiation studies at 213 K reveal that for nonhydroxy DASAs E–Z photoisomerization can take place at two different bonds, yielding two distinct isomers. These observations are supported by TD-DFT calculations, showing that in the excited state the hydroxy group (pre)selects the neighboring C2–C3 bond for isomerization. The TD-DFT analysis also explains the larger solvatochromic shift observed for the parent DASAs as compared to the nonhydroxy analogue, in terms of the dipole moment changes evoked upon excitation. Furthermore, computations provide helpful insights into the photoswitching energetics, indicating that without the hydroxy group the 4π-electrocyclization step is energetically forbidden. Our results establish the central role of the hydroxy group for DASA photoswitching and suggest that its introduction allows for tailoring photoisomerization pathways, presumably both through (steric) fixation via a hydrogen bond with the adjacent carbonyl group of the acceptor moiety, as well as through electronic effects on the polyene backbone. These insights are essential for the rational design of novel, improved DASA photoswitches and for a better understanding of the properties of both DASAs and cyanine dyes.

The hydrogen bonding of water and water/salt mixtures around the proline-based tripeptide model glycyl-l-prolyl-glycinamide·HCl (GPG-NH2) is investigated here by multi-wavelength UV resonance Raman spectroscopy (UVRR) to clarify the role of ion–peptide interactions in affecting the conformational stability of this peptide. The unique sensitivity and selectivity of the UVRR technique allow us to efficiently probe the hydrogen bond interaction between water molecules and proline residues in different solvation conditions, along with its influence on trans to cis isomerism in the hydrated tripeptide. The spectroscopic data suggest a relevant role played by the cations in altering the solvation shell at the carbonyl site of proline., while the fluoride and chloride anions were found to promote the establishment of the strongest interactions on the C=O site of proline. This latter effect is reflected in the greater stabilization of the trans conformers of the tripeptide in the presence of these specific ions. The molecular view provided by UVRR experiments was complemented by the results of circular dichroism (CD) measurements that show a strong structural stabilizing effect on the β-turn motif of GPG-NH2 observed in the presence of KF as a co-solute.

Two triads (BDP-SQ and Styryl-BDP-SQ) were prepared with Bodipy, styrylBodipy and Squaraine (SQ) units. SQ shows unexpected efficient intersystem crossing (ISC. ΦT = 50%), which is attributed to S1→T1 transition. In the two triads, the Forster Resonance Energy Transfer (FRET) direction, as well as the spatial localization of the T1 state, was judiciously tuned. The cascade photophysical properties of the triads were studied with steady-state and time-resolved optical spectroscopies, as well as with electrochemical characterization and theoretical computations. We show that triplet state was produced in triad BDP-SQ upon photoexcitation, but in Styryl-BDP-SQ the fast FRET and the charge separation (CS) processes compete with the ISC of the SQ unit, and no triplet state was formed upon photoexcitation. The singlet energy transfer kinetics were found to be 1.6 and 0.6 ps, respectively and are solvent polarity dependent. Charge transfer was confirmed with ultrafast transient absorption spectroscopy.

Donor-acceptor Stenhouse adducts (DASAs) are negative photochromes that hold great promise for a variety of applications. Key to optimizing their switching properties is a detailed understanding of the photoswitching mechanism, which, as yet, is absent. Here we characterize the actinic step of DASA-photoswitching and its key intermediate, which was studied using a combination of ultrafast visible and IR pump-probe spectroscopies and TD-DFT calculations. Comparison of the time-resolved IR spectra with DFT computations allowed to unambiguously identify the structure of the intermediate, confirming that light absorption induces a sequential reaction path in which a Z-E photoisomerization of C2-C3 is followed by a rotation around C3-C4 and a subsequent thermal cyclization step. First and second-generation DASAs share a common photoisomerization mechanism in chlorinated solvents with notable differences in kinetics and lifetimes of the excited states. The photogenerated intermediate of the second-generation DASA was photo-accumulated at low temperature and probed with time-resolved spectroscopy, demonstrating the photoreversibility of the isomerization process. Taken together, these results provide a detailed picture of the DASA isomerization pathway on a molecular level.

The understanding of charge carrier dynamics in hybrid materials based on colloidal semiconductor nanocrystals (or quantum dots) and organic moieties is fundamental for the design of efficient photonic and photovoltaic devices. In the present work, we investigate the interactions occurring between CdTe quantum dots, capped with a strong capping agent such as 3-mercaptopropionic acid, and a well known electron acceptor such as methylviologen molecule. The nature of the interactions and of exciton dynamics is investigated by stationary and time-resolved spectroscopies. Luminescence data recorded in presence of increasing methylviologen concentrations, indicate that the organic molecule is able to statically interact with the surface sites of CdTe quantum dots; a biphasic interaction behavior is evidenced by determining the apparent association constants. These latter are obtained through the analysis of luminescence data, and values in the range 10(3)-10(4) are determined. The nature of the interactions is characterized by nanosecond and femtosecond transient absorption spectroscopies, to clarify the dynamics and the conditions able to foster charge mobility. Nanosecond flash photolysis measurements, carried out upon quantum dots excitation, shows the absorption of methylviologen radical cation specie at 605 nm, suggesting the occurrence of electron transfer from CdTe nanocrystals to the organic acceptor; the relatively long decay time of the transient signal (10.3 mu s) indicates that back electron transfer processes are negligible. Ultrafast transient absorption measurements confirm the occurrence of an ultrafast electron transfer process; spectral and kinetic analysis of the transient data show that methylviologen radical cation is formed almost instantaneously on the ps-time scale but mainly when the samples are pumped in the energy continuum at 400 nm. This finding suggests that electron mobility from the nanocrystals to the organic units is achieved mainly when the excitonic states possess an excess of energy. The comparison of the kinetic behaviour of the signals at increasing methylviologen concentrations indicates that the electron transfer process competes with the radiative exciton recombination. In addition, the kinetic data suggests that surface trapping and Auger recombination processes might slow down the charge mobility. (C) 2017 Elsevier B.V. All rights reserved.

Spin-orbit charge-transfer intersystem crossing (SOCT-ISC) is useful for the preparation of heavy atom-free triplet photosensitisers (PSs). Herein, a series of perylene-Bodipy compact electron donor/acceptor dyads showing efficient SOCT-ISC is prepared. The photophysical properties of the dyads were studied with steady-state and time-resolved spectroscopies. Efficient triplet state formation (quantum yield phi(T)=60 %) was observed, with a triplet state lifetime (tau(T)=436 mu s) much longer than that accessed with the conventional heavy atom effect (tau(T)=62 mu s). The SOCT-ISC mechanism was unambiguously confirmed by direct excitation of the charge transfer (CT) absorption band by using nanosecond transient absorption spectroscopy and time-resolved electron paramagnetic resonance (TREPR) spectroscopy. The factors affecting the SOCT-ISC efficiency include the geometry, the potential energy surface of the torsion, the spin density for the atoms of the linker, solvent polarity, and the energy matching of the (CT)-C-1/(LE)-L-3 states. Remarkably, these heavy atom-free triplet PSs were demonstrated as a new type of efficient photodynamic therapy (PDT) reagents (phototoxicity, EC50=75 nm), with a negligible dark toxicity (EC50=78.1 mu m) compared with the conventional heavy atom PSs (dark toxicity, EC50=6.0 mu m, light toxicity, EC50=4.0 nm). This study provides in-depth understanding of the SOCT-ISC, unveils the design principles of triplet PSs based on SOCT-ISC, and underlines their application as a new generation of potent PDT reagents.

In this investigation, we report the first hyperpolarizabilities and two-photon absorption cross sections of a large series of 12 push–pull cationic chromophores. All of these dyes show a dipolar acceptor+–π–donor structure, where the nature of the donor and acceptor units and π-bridge was synthetically tuned to allow insightful comparisons among the molecules. The hyperpolarizability was obtained through a solvatochromic method, by exploiting the rare negative solvatochromism exhibited by the investigated compounds. The two-photon absorption cross sections were determined through two-photon excited fluorescence measurements by means of a tunable nanosecond laser system for sample excitation. The nonlinear optical properties were discussed relatively to the photoinduced intramolecular charge transfer occurring in these donor–acceptor systems, investigated by femtosecond transient absorption experiments. We found a strong increase in hyperpolarizability upon increasing the molecular conjugation. Unexpectedly, the hyperpolarizability is almost unaffected by an increase in donor/acceptor strength and intramolecular charge transfer degree. Differently, the two-photon absorption cross sections of these dyes are enhanced by an increase in both molecular conjugation and intramolecular charge transfer efficiency. Several recent literature works have reported at the same time scattered information about the hyperpolarizability and two-photon absorption of small organic molecules. Our investigation is, to the best of our knowledge, the first attempt to uncover detailed structure–property relationships for these two nonlinear optical properties. Our results represent a promising route to achieve large hyperpolarizability and two-photon absorption in push–pull dyes and may drive the design of new efficient nonlinear optical materials.

In order to study the relationship between the molecular structures of compact electron donor/acceptor dyads and the spin-orbit charge transfer intersystem crossing (SOCT-ISC) efficiency, we prepared a perylene (Pery)-naphthalimide (NI) dyad, in which the Pery unit is the electron donor and the NI unit is the electron acceptor, where the two units adopt an almost orthogonal geometry. The photophysical properties of the dyad were studied with steady state and femtosecond/nanosecond transient absorption (fs TA and ns TA) spectroscopies, time resolved electron paramagnetic resonance (TREPR) spectroscopy and DFT computations. A very weak charge transfer (CT) absorption band was observed, but the fluorescence of the Pery unit is strongly quenched. Upon selective excitation into the NI unit, the fast intramolecular CS process (10 ps) occurs, followed by ISC and only the 3Pery* state is observed; whereas upon selective photoexcitation into the perylene unit, an ultrafast charge separation (0.66 ps) is observed, followed by SOCT-ISC (8 ns) to populate the 3Pery* state. Moreover, the perylene radical cation is also observed on the ns timescale, presumably formed by intermolecular charge transfer. The lifetime of the 3Pery triplet state was determined to be ?T = 214 ?s with ns TA spectroscopy. The singlet oxygen quantum yield was measured as ?? = 80%, although the potential energy curve of the torsion between the donor and acceptor is shallow. The SOCT-ISC mechanism was confirmed by TREPR spectroscopy. The dyad was used as a triplet photosensitizer for the generation of delayed fluorescence (luminescence lifetime ?DF = 57.3 ?s).

5,10-dihydro-5,10-dimethylphenazine (PZ) and boron dipyrromethene (Bodipy) were linked to prepare compact electron donor/acceptor dyads and triads. The effect of the molecular geometry on the UV-Vis absorption, fluorescence and the spin-orbit charge transfer intersystem crossing (SOCT-ISC) efficiency were studied. Interestingly, a broad near-IR charge transfer (CT) absorption band (centred at 670 nm) was observed for the dyad/triad showing more coplanar geometry. Triplet state formation was observed in low polar solvents for the triad with orthogonal geometry (triplet state quantum yield is 30 % and lifetime is ?T=210 ?s). Femtosecond transient absorption spectra indicated fast charge separation (CS, 2 ps) and slow charge recombination (CR, >1.5 ns). The low-lying 3CT state of the dyads/triads in polar solvent (1.23 eV vs. the 1.65~1.69 eV of the 3LE state) was confirmed by intermolecular triplet photosensitizing experiments and the spin density surface analysis. These results are useful for design of new broadband, near-IR absorbing heavy atom-free triplet photosensitizers.

The comprehensive characterization of Intramolecular Charge Transfer (ICT) stemming in push-pull molecules with a delocalized π-system of electrons is noteworthy for a bespoke design of organic materials, spanning widespread applications from photovoltaics to nanomedicine imaging devices. Photo-induced ICT is characterized by structural reorganizations, which allows the molecule to adapt to the new electronic density distribution. Herein, we discuss recent photophysical advances combined with recent progresses in the computational chemistry of photoactive molecular ensembles. We focus the discussion on femtosecond Transient Absorption Spectroscopy (TAS) enabling us to follow the transition from a Locally Excited (LE) state to the ICT and to understand how the environment polarity influences radiative and non-radiative decay mechanisms. In many cases, the charge transfer transition is accompanied by structural rearrangements, such as the twisting or molecule planarization. The possibility of an accurate prediction of the charge-transfer occurring in complex molecules and molecular materials represents an enormous advantage in guiding new molecular and materials design. We briefly report on recent advances in ultrafast multidimensional spectroscopy, in particular, Two-Dimensional Electronic Spectroscopy (2DES), in unraveling the ICT nature of push-pull molecular systems. A theoretical description at the atomistic level of photo-induced molecular transitions can predict with reasonable accuracy the properties of photoactive molecules. In this framework, the review includes a discussion on the advances from simulation and modeling, which have provided, over the years, significant information on photoexcitation, emission, charge-transport, and decay pathways. Density Functional Theory (DFT) coupled with the Time-Dependent (TD) framework can describe electronic properties and dynamics for a limited system size. More recently, Machine Learning (ML) or deep learning approaches, as well as free-energy simulations containing excited state potentials, can speed up the calculations with transferable accuracy to more complex molecules with extended system size. A perspective on combining ultrafast spectroscopy with molecular simulations is foreseen for optimizing the design of photoactive compounds with tunable properties.

Evidence of subtle effects in the dynamic reorganization of a protic solvent in its first- and farther-neighbor shells, in response to the sudden change in the solute's electronic distribution upon excitation, is unveiled by a multilevel computational approach. Through the combination of nonequilibrium molecular dynamics and quantum mechanical calculations, the experimental time evolution of the transient T1 absorption spectra of a heteroleptic Ru(II)-polypyridine complex in ethanol or dimethyl sulfoxide solution is reproduced and rationalized in terms of both fast and slow solvent re-equilibration processes, which are found responsible for the red shift and broadening experimentally observed only in the protic medium. Solvent orientational correlation functions and a time-dependent analysis of the solvation structure confirm that the initial, fast observed red shift can be traced back to the destruction-formation of hydrogen bond networks in the first-neighbor shell, whereas the subsequent shift, evident in the [20-500] ps range and accompanied by a large broadening of the signal, is connected to a collective reorientation of the second and farther solvation shells, which significantly changes the electrostatic embedding felt by the excited solute.

Hydrated ionic liquids (ILs) have been identified as solvent media able to enhance the structural stability of deoxyribonucleic acid (DNA). In this work, we investigate the molecular interaction between imidazolium-based ILs and DNA during its thermal unfolding pathway, by exploiting synchrotron-based UV Resonance Raman scattering (UVRR) experiments. This technique gives a selective focus on the thermal responses of specific nucleobases in the structure of DNA, providing the experimental sensitivity to both cooperative and local structural changes occurring along the complex unfolding process of DNA. UVRR measurements probe two distinct temperature-dependent phenomena occurring in the DNA double-helix, i.e. a non-cooperative pre-melting mainly involving adenine bases and a cooperative melting transition primarily localized on guanine tracts. The analysis of Raman spectra reveals that both the cation and anion of the ionic liquids strongly interact with the structure of DNA, thus affecting the melting process but not perturbing the pre-melting transition that precedes the complete separation of the strands of DNA. Overall these results suggest that the dominant interaction occurs between the imidazolium cation and the bases of guanine and thymine in the structure of DNA, in agreement with previous results of molecular dynamics simulations.

Three quadrupolar D−π–A−π–D compounds, bearing alkoxy phenyls as mild electron donors and a benzothiadiazole (A), two benzothiadiazoles (B), or a benzothiadiazole linked to two thiophenes (C) as the acceptor units, are collectively the object of this study. They proved to be efficient yellow/orange/red fluorophores, respectively, with fluorescence being their preferred deactivation pathway. These systems exhibited positive fluorosolvatochromism and a noticeable decrease of the fluorescence quantum yield in the most polar solvent (with the quenching following the trend B > A > C). These findings point to an intramolecular charge transfer (ICT) nature of the emitting state, whose photoinduced dynamics was investigated by femtosecond-resolved transient absorption and fluorescence upconversion. A donor acceptor dyad composed of a BF2-chelated dipyrromethene (BODIPY) and a C-60 fullerene has been newly synthesized and characterized. The two moieties are linked by direct addition of an azido substituted BODIPY on the C-60, producing an imino fullerene BODIPY adduct. The photoinduced charge transfer process in this system was studied by ultrafast transient absorption spectroscopy. Electron transfer toward the fullerene was found to occur selectively exciting both the BODIPY chromophore at 475 nm and the C-60 unit at 266 nm on a time scale of a few picoseconds, but the dynamics of charge separation was different in the two cases. Eletrochemical studies provided information on the redox potentials of the involved species and spectroelectrochemical measurements allowed to unambiguously assign the absorption band of the oxidized BODIPY moiety, which helped in the interpretation of the transient absorption spectra. The experimental studies were complemented by a theoretical analysis based on DFT computations of the excited state energies of the two components and their electronic couplings, which allowed identification of the charge transfer mechanism and rationalization of the different kinetic behavior observed by changing the excitation conditions.

In this work we report the linear and non-linear IR spectral response characterization of the CO bonds of PenicillinG sodium salt in D2O and in DMSO&minus, d6 solutions. In order to better characterize the spectral IR features in the CO stretching region, broadband middle infrared pump-probe spectra are recorded. The role of hydrogen bonds in determining the inhomogeneous broadening and in tuning anharmonicity of the different types of oscillators is exploited. Narrow band pump experiments, at the three central frequencies of &beta, &minus, lactam, amide and carboxylate CO stretching modes, identify the couplings between the different types of CO oscillators opening the possibility to gather structural dynamic information. Our results show that the strongest coupling is between the &beta, lactam and the carboxylate CO vibrational modes.

In this work, we studied the propagation of ultrasonic waves of lysozyme solutions characterized by different degrees of aggregation and networking. The experimental investigation was performed by means of the transient grating (TG) spectroscopy as a function of temperature, which enabled measurement of the ultrasonic acoustic proprieties over a wide time window, ranging from nanoseconds to milliseconds. The fitting of the measured TG signal allowed the extraction of several dynamic properties, here we focused on the speed and the damping rate of sound. The temperature variation induced a series of processes in the lysozyme solutions: Protein folding-unfolding, aggregation and sol–gel transition. Our TG investigation showed how these self-assembling phenomena modulate the sound propagation, affecting both the velocity and the damping rate of the ultrasonic waves. In particular, the damping of ultrasonic acoustic waves proved to be a dynamic property very sensitive to the protein conformational rearrangements and aggregation processes.

The photosynthetic Reaction Center (RC) from the purple bacterium Rhodobacter sphaeroides has unique photoconversion capabilities, that can be exploited in assembly biohybrid devices for applications in solar energy conversion. Extending the absorption cross section of isolated RC through covalent functionalization with ad-hoc synthesized artificial antennas is a successful strategy to outperform the efficiency of the pristine photoenzyme under visible light excitation. Here we report a new heptamethine cyanine antenna that, upon covalent binding to RC, forms a biohybrid (hCyN7-RC) which, under white light excitation, has doubled photoconversion efficiency versus the bare photoenzyme. The artificial antenna hCyN7 successfully meets appropriate optical properties, i.e. peak position of absorption and emission maximum in the visible and NIR region respectively, large Stokes shift, and high fluorescence quantum yield, required for improving the efficiency of the biohybrid in the production of the charge-separated state in the RC. In this work, we present for the first time the synthesis and characterization of potential DSSC organic sensitizers whose cyanoacrylic acceptor/anchoring group was modified by replacement of oxygen with one or more sulfur atoms. Using known carboxylic acid dye DF15 as a reference, their TD-DFT computational analysis indicated that oxygen-sulfur substitution should induce a significant red-shift of the corresponding UV/Vis absorption spectra. Whereas synthesis of monothio-carboxylic derivatives of DF15 was successfully carried out, isolation of the analogous dithiocarboxylic acid proved impossible due to its very low stability: despite that, their relative spectroscopic properties could be compared by analyzing the corresponding benzylic esters. Combined computational and transient absorption spectroscopy studies suggested that photoexcited thiocarboxylic acid 1-SO and thioamide 1-SN were able to inject electrons into nanocrystalline TiO2 and indicated a similar charge injection efficiency for 1-SO relative to DF15. Preliminary DSSC studies showed that, when tert-butylpyridine was present in the electrolyte solution, 1-SO was rapidly degraded; however, in the absence of basic additives 1-SO provided a sufficiently stable device, giving a slightly lower efficiency compared to carboxylic sensitizer DF15.

The specific interactions of water molecules with the tripeptide glutathione are investigated by UV Resonance Raman spectroscopy taking advantage of its sensitivity and selectivity. The peptide-solvent interactions at different chemical sites of glutathione, i.e. the peptide backbone and the side groups of the molecule, are investigated. With the aim of gaining new insights on the mechanism regulating the hydration of glutathione, intimately related to its biological functionality, the spectra of the tripeptide solutions are examined as a function of different concentration, pH and temperature. The Amide and the OH stretching spectral regions result very sensitive to the variations of the explored experimental parameters. Our observations provide a picture of the hydrogen-bonding network around glutathione. The number and the strength of hydrogen bonds increase in the deprotonated form of tripeptide that exhibits also a more marked capacity in decreasing the intermolecular order of water in its hydration shell. According to this view, a progressive reduction in the strength of hydrogen bond interactions on amide sites is probed upon the increment of thermal motion, accompanied by conformational changes involving the trans-cis isomerization of glutathione.

We report a combined approach of stationary and time-resolved fluorescence measurements and ultraviolet-visible (UV-vis) transient absorption spectroscopy (TAS) along with ab initio calculations, which provide an overall picture of the dynamics occurring after excitation in a push-pull molecule, namely, 4,7-bis (4,5-dibutylbenzo[1,2-b:4,3-b?]bisthiophene[1,2-b:4,3-b?]bisthiophen-2-yl)-2,1,3-benzothiadiazole. The analysis of the emission spectra in solvents of different polarities reveals the presence of three conformers whose structures differ in the orientation of the 4,5-dibutylbenzo-bisthiophene groups and in their planarity with respect to the benzothiadiazole acceptor group. The Kawski method allows us to estimate the ground- and first-excited state dipole moments (? g and ? e ) for the three conformers. We find values of ? e similar for the three conformers and higher than the relative ? g values as can be expected from a push-pull molecule undergoing a light-induced charge-transfer (CT) transition. UV-vis TAS in different solvents highlights the instantaneous (within our instrumental resolution) formation of a locally excited S 1 state (accompanied by a big change in the dipole moment with respect to S 0 ), which undergoes a rapid intramolecular CT (ICT) assisted by molecule planarization [planar ICT (PICT)]. The strong dipole-dipole interactions with the polarized solvent molecules stabilize the S 1 CT state that decays principally through fluorescence emission. Both PICT and solvation dynamics are responsible for the big Stokes' shift characterizing the molecule, particularly in polar solvents. The fluorescence lifetimes are substantially longer in polar solvents, and also fluorescence quantum yields are higher in polar solvents. We conclude that the radiative relaxation time increases when molecular planarization of the S 1 emissive state takes place, and this condition is favored in polar solvents where local dipole-dipole interactions support the structural stabilization of the CT emissive state. In the poly(methyl methacrylate) matrix, the structural and solvation dynamics are strongly inhibited, leading to reduction of nonradiative processes and to shortening of the fluorescence relaxation time

Photosynthetic antennae and organic electronic materials use topological, structural, and molecular control of delocalized excitons to enhance and direct energy transfer. Interactions between the transition dipoles of individual chromophore units allow for coherent delocalization across multiple molecular sites. This delocalization, for specific geometries, greatly enhances the transition dipole moment of the lowest energy excitonic state relative to the chromophore and increases its radiative rate, a phenomenon known as superradiance. In this study, we show that ordered, self-assembled light-harvesting nanotubes (LHNs) display excitation-induced photobrightening and photodarkening. These changes in quantum yield arise due to changes in energetic disorder, which in turn increases/decreases excitonic superradiance. Through a combination of experiment and modeling, we show that intense illumination induces different types of chemical change in LHNs that reproducibly alter absorption and fluorescence properties, indicating control over excitonic delocalization. We also show that changes in spectral width and shift can be sensitive measures of system dimensionality, illustrating the mixed 1-2D nature of LHN excitons. Our results demonstrate a path forward for mastery of energetic disorder in an excitonic antenna, with implications for fundamental studies of coherent energy transport.

In this work we present the synthesis, time-resolved spectroscopic characterization and computational analysis of a bichromophore composed of two very well-known naturally occurring dyes: 7-hydroxycoumarin (umbelliferone) and 1,2-dihydroxyanthraquinone (alizarin). The umbelliferone donor (Dn) and alizarin acceptor (Ac) moieties are linked to a triazole ring via ? bonds, providing a flexible structure. By measuring the fluorescence quantum yields and the ultrafast transient absorption spectra we demonstrate the high efficiency (~85%) and the fast nature (~1.5 ps) of the energy transfer in this compound. Quantum chemical calculations, within the density functional theory (DFT) approach, are used to characterize the electronic structure of the bichromophore (Bi) in the ground and excited states. We simulate the absorption and fluorescence spectra using the TD-DFT methods and the vertical gradient approach (VG), and include the solvent effects by adopting the conductor-like polarizable continuum model (CPCM). The calculated electronic structure suggests the occurrence of weak interactions between the electron densities of Dn and Ac in the excited state, indicating that the Förster-type transfer is the appropriate model for describing the energy transfer in this system. The average distance between Dn and Ac moieties calculated from the conformational analysis (12 Å) is in very good agreement with the value estimated from the Förster equation (~11 Å). At the same time, the calculated rate constant for energy transfer, averaged over multiple conformations of the system (3.6 ps), is in reasonable agreement with the experimental value (1.6 ps) estimated by transient absorption spectroscopy. The agreement between experimental results and computational data leads us to conclude that the energy transfer in Bi is well described by the Förster mechanism. © the Partner Organisations 2014.

One of the challenges for achieving efficient exciton transport in solar energy conversion systems is precise structural control of the light-harvesting building blocks. Here, we create a tunable material consisting of a connected chromophore network on an ordered biological virus template. Using genetic engineering, we establish a link between the inter-chromophoric distances and emerging transport properties. The combination of spectroscopy measurements and dynamic modelling enables us to elucidate quantum coherent and classical incoherent energy transport at room temperature. Through genetic modifications, we obtain a significant enhancement of exciton diffusion length of about 68% in an intermediate quantum-classical regime.

The properties of lipid membranes at low temperature are important for a number of biomedical and biotechnological applications, and the success of these applications depends on understanding the effects of temperature changes on intermolecular lipid-lipid and lipid-water interactions. Here, we use Fourier transform infrared spectroscopy to study lipid suspensions in water/dimethyl sulfoxide (DMSO) solutions in the -60 to 30 °C range. DMSO is a cryopreservative agent of cellular systems, and its action is largely related to its interaction with the lipid membrane, especially in the low-temperature regime. In the present work, we analyze the effects of solvent composition on the structural and thermotropic properties of cholesterol (chol)-loaded liposomes of palmitoyl-oleylphosphatidylcholine (POPC) because POPC/chol liposomes are suitable models of the plasmatic membrane. To this extent, we compare the properties of lipid vesicles suspended in water and water/DMSO solution at 0.10 DMSO mole fraction and we observe that the gel phase of the membrane has an increased thermal stability on DMSO addition. We estimate that the amount of unfrozen water at T = -60 °C is much reduced by the presence of DMSO, both in the gel- and the liquid-ordered phase of the membrane. Interestingly, we also evidence a reduced hydration of the lipid heads in the presence of DMSO when the vesicles are dispersed in a liquid solution, whereas the addition of DMSO does not alter the hydration state of phosphate and carbonyl groups in the frozen state of the membrane.

A series of Bodipy dimers with orthogonal conformation were prepared. The photophysical properties were studied with steady-state and time-resolved transient spectroscopies. We found the triplet-state quantum yield is highly dependent on the solvent polarity in the orthogonally linked symmetric Bodipy dimers, and the intersystem crossing (ISC) is efficient in solvents with moderate polarity. The photoinduced symmetry-breaking charge transfer (SBCT) in polar solvents was confirmed by femtosecond transient absorption spectroscopy, with the charge separation (CS) kinetics on the order of a few picoseconds and the charge recombination (CR) process occurring on the nanosecond time scale in dichloromethane. These observations are supported by the calculation of the charge separated state (CSS) energy levels, which are high in nonpolar solvents, and lower in polar solvents, thus the CR-induced ISC has the largest driven force in solvents with moderate polarity. These results clarify the mechanism of SOCT-ISC in the orthogonally symmetric Bodipy dimers. The acquired information, relating molecular structure and ISC property, will be useful for devising new strategies to induce ISC in heavy atom-free organic chromophores.

Phenothiazine (PTZ)-anthracene (An) compact electron donor/acceptor dyads were synthesized. The molecular conformation was constrained by rotation restriction to achieve an orthogonal geometry between the electron donor (PTZ) and the electron acceptor (An), with the aim to enhance the spin-orbit charge-transfer intersystem crossing (SOCT-ISC). The substitution positions on the PTZ and An moieties were varied to attain dyads with different mutual orientations of the donor/acceptor as well as different rotation-steric hindrances. The electronic coupling strengths between the electron donor and the acceptor were quantified with the matrix elements (V-DA, 0.04-0.18 eV); the smallest value was observed for the dyad with orthogonal geometry. Charge-transfer absorption and fluorescence emission bands were observed for the dyads, for which the intensity varied, manifested by the V-DA values. The fluorescence of the An moiety was significantly quenched in the dyads, efficient ISC, and the formation of the triplet state were confirmed with nanosecond transient absorption spectroscopy (Phi(Delta) = 65%, tau(T) = 209 mu s). The rotation-steric hindrance was analyzed with potential energy curves, and PTZ was found to be an ideal electron donor to attain SOCT ISC. Time-resolved electron paramagnetic resonance spectra revealed the electron-spin polarization (ESP) of the triplets of the dyads, which is drastically different from that of An, thus confirming the SOCT ISC mechanism. Moreover, we found that the ESP patterns of the dyads strongly depend on the topological features of the molecules and the structure of the electron donor, thus indicating that the relationship between the molecular conformation and the ESP parameters of the triplet state of the dyads cannot be described solely by the orthogonal geometry, as was previously observed.
