Abstract Background Atrial Fibrillation (AF) is known to be associated with a negative emotional state. Patient-reported outcomes (PROs) are important tools for evaluating the endpoints of AF management. This study aims to examine the correlation between personality types and PROs in patients with AF. Methods All included subjects were newly diagnosed with AF fewer than one month, and their personality types were assessed using the Eysenck Personality Questionnaire (EPQ). Quality of life (QoL) was measured using the Atrial Fibrillation Effect on Quality of Life (AFEQT) questionnaire. Anxiety and depression were assessed using the General Anxiety Scale (GAD-7) and the 9-item Patient Health Questionnaire (PHQ-9), respectively. We constructed stepwise linear regression analyses for factors related to the QoL and emotional state in patients with AF. Results A total of 531 AF patients completed the survey and were categorized into four groups based on their personality types. Of these patients (mean age: 67.12 ± 10.93 years, 50.28% male), 357 (67.23%) had paroxysmal AF, and 16.95% (n = 90) had a sanguine personality. 3. Mapping of spastic muscle activity after stroke: difference between passive stretch and active contraction

Abstract Background Investigating the spatial distribution of muscle activity would facilitate understanding the underlying mechanism of spasticity. The purpose of this study is to investigate the characteristics of spastic muscles during passive stretch and active contraction by high-density surface electromyography (HD-sEMG). Methods Fourteen spastic hemiparetic subjects and ten healthy subjects were recruited. The biceps brachii (BB) muscle activity of each subject was recorded by HD-sEMG during passive stretch at four stretch velocities (10, 60, 120, 180˚/s) and active contraction at three submaximal contraction levels (20, 50, 80%MVC). The intensity and spatial distribution of the BB activity were compared by the means of two-way analysis of variance, independent sample t-test, and paired sample t-test. Results Compared with healthy subjects, spastic hemiparetic subjects showed significantly higher intensity with velocity-dependent heterogeneous activation during passive stretch and more lateral and proximal activation distribution during active contraction. In addition, spastic hemiparetic subjects displayed almost non-overlapping activation areas during passive stretch and active contraction. The activation distribution of passive stretch was more distal when compared with the active contraction. Conclusions These alterations of the BB activity could be the consequence of deficits in the descending central control after stroke. The complementary spatial distribution of spastic BB activity reflected their opposite motor units (MUs) recruitment patterns between passive stretch and active contraction. This HD-sEMG study provides new neurophysiological evidence for the spatial relationship of spastic BB activity between passive stretch and active contraction, advancing our knowledge on the mechanism of spasticity. 4. A comparative plastome approach enhances the assessment of genetic variation in the Melilotus genus

Abstract Background Melilotus, a member of the Fabaceae family, is a pivotal forage crop that is extensively cultivated in livestock regions globally due to its notable productivity and ability to withstand abiotic stress. However, the genetic attributes of the chloroplast genome and the evolutionary connections among different Melilotus species remain unresolved. Results In this study, we compiled the chloroplast genomes of 18 Melilotus species and performed a comprehensive comparative analysis. Through the examination of protein-coding genes, we successfully established a robust phylogenetic tree for these species. This conclusion is further supported by the phylogeny derived from single-nucleotide polymorphisms (SNPs) across the entire chloroplast genome. Notably, our findings revealed that M. infestus, M. siculus, M. sulcatus, and M. speciosus formed a distinct subgroup within the phylogenetic tree. Additionally, the chloroplast genomes of these four species exhibit two shared inversions. Moreover, inverted repeats were observed to have reemerged in six species within the IRLC. The distribution patterns of single-nucleotide polymorphisms (SNPs) and insertions/deletions (InDels) within protein-coding genes indicated that ycf1 and ycf2 accumulated nonconservative alterations during evolutionary development. Furthermore, an examination of the evolutionary rate of protein-coding genes revealed that rps18, rps7, and rpl16 underwent positive selection specifically in Melilotus. Conclusions We present a comparative analysis of the complete chloroplast genomes of Melilotus species. This study represents the most thorough and detailed exploration of the evolution and variability within the genus Melilotus to date. Our study provides valuable chloroplast genomic information for improving phylogenetic reconstructions and making biogeographic inferences about Melilotus and other Papilionoideae species.

5. Efficacy and safety of intracoronary pro-urokinase combined with low-pressure balloon pre-dilatation during percutaneous coronary intervention in patients with anterior ST-segment elevation myocardial infarction

Abstract Background The efficacy and safety of low-pressure balloon pre-dilatation before intracoronary pro-urokinase (pro-UK) in preventing no-reflow during percutaneous coronary intervention (PCI) remains unknown. This study aimed to evaluate the clinical outcomes of intracoronary pro-UK combined with low-pressure balloon pre-dilatation in patients with anterior ST-segment-elevation myocardial infarction (STEMI). Methods This was a randomized, single-blind, investigator-initiated trial that included 179 patients diagnosed with acute anterior STEMI. All patients were eligible for PCI and were randomized into two groups: intracoronary pro-UK combined with (ICPpD group, n = 90) or without (ICP group, n = 89) low-pressure balloon pre-dilatation. The main efficacy endpoint was complete epicardial and myocardial reperfusion. The safety endpoints were major adverse cardiovascular events (MACEs), which were analyzed at 12 months follow-up. Results Patients in the ICPpD group presented significantly higher TIMI myocardial perfusion grade 3 (TMPG3) compared to those in the ICP group (77.78% versus 68.54%, P = 0.013), and STR ≥ 70% after PCI 30 min (34.44% versus 26.97%, P = 0.047) or after PCI 90 min (40.0% versus 31.46%, P = 0.044). MACEs occurred in 23 patients (25.56%) in the ICPpD group and in 32 patients (35.96%) in the ICP group. There was no difference in hemorrhagic complications during hospitalization between the groups. Conclusion Patients with acute anterior STEMI presented more complete epicardial and myocardial reperfusion with adjunctive low-pressure balloon pre-dilatation before intracoronary pro-UK during PCI. Herba Epimedii’s leaves are highly valued in traditional Chinese medicine for their substantial concentration of flavonoids, which play a crucial role in manifesting the plant’s therapeutic properties. This study investigated the metabolomic, transcriptomic and proteomic profiles of leaves from two Herba Epimedii cultivars, Epipremnum sagittatum (J) and Epipremnum pubescens (R), at three different developmental stages. Metabolite identification and analysis revealed a total of 1,412 and 1,421 metabolites with known structures were found. Flavonoids made up of 33%, including 10 significant accumulated icariin analogues. Transcriptomic analysis unveiled totally 41,644 differentially expressed genes (DEGs) containing five encoded genes participated in icariin biosynthesis pathways. Totally, 9,745 differentially expressed proteins (DEPs) were found, including Cluster-47248.2.p1 (UDP-glucuronosy/UDP-glucosyltransferase), Cluster-30441.2.p1 (O-glucosyltransferase), and Cluster-28344.9.p1 (anthocyanidin 3-O-glucoside 2 “-O-glucosyltransferase-like) through proteomics analysis which are involved to icariin biosynthesis. Background: Neutrophils play important roles in atherosclerosis and atherothrombosis. Bactericidal/permeability-increasing protein (BPI) is mainly expressed in the granules of human neutrophils in response to inflammatory stress. This observational, cross-sectional study investigated the plasma level of BPI in patients with acute coronary syndrome (ACS) and its correlation with blood neutrophil counts and circulating inflammatory biomarkers. Methods: A total of 367 patients who had acute chest pain and who were admitted to our hospital for coronary angiography (CAG) and/or percutaneous coronary intervention (PCI) from May 1, 2020 to August 31, 2020 were recruited. Among them, 256 had a cardiac troponin value above the 99th percentile upper reference limit and were diagnosed with ACS. The remaining patients (n = 111) were classified as non-ACS. The TIMI and GRACE scores were calculated at admission. The Gensini score based on CAG was used to determine atherosclerotic burden. Plasma levels of interleukin (IL)-1β, myeloperoxidase-DNA (MPO-DNA), high sensitivity C-reactive protein (hs-CRP), S100A8/A9, and BPI were measured using enzyme-linked immunosorbent assays. Correlations of plasma BPI levels with examination scores and levels of circulating inflammatory biomarkers were explored. Receiver operating characteristic (ROC) curve analysis was used to determine the diagnostic efficacy of BPI for ACS and myocardial infarction. ElectricalImpedance Myography (EIM) is an innovative, non-invasive technique offering a convenient means of localized exogenous electrophysiological recording. By measuring muscle impedance parameters, this method characterizes the physiological state of muscles, functioning as a biomarker for muscle contractility, injuries, and the progression of neuromuscular diseases. This paper provides an overview of the current state of EIM technology development, along with modeling and data analysis methods, focusing on their application requirements. It further highlights the advancements in EIM research within the realm of sports health, emphasizing its efficacy in identifying injuries and monitoring wound healing, and discusses existing technological limitations. Additionally, the paper explores future research directions. Serving as a transient biosensor during physical activity, EIM holds significant potential in sports health. It presents a promising alternative to invasive and costly clinical assessment methods, positioning itself as a viable personal monitoring tool for both professional athletes and fitness enthusiasts. Nevertheless, the resolution of technical challenges and the establishment of industry-standard implementation programs are essential prerequisites for EIM to evolve into a standard clinical assessment tool.

Abstract Rechargeable lithium batteries using 5 V positive electrode materials can deliver considerably higher energy density as compared to state-of-the-art lithium-ion batteries. However, their development remains plagued by the lack of electrolytes with concurrent anodic stability and Li metal compatibility. Here we report a new electrolyte based on dimethyl 2,5-dioxahexanedioate solvent for 5 V-class batteries. Benefiting from the particular chemical structure, weak interaction with lithium cation and resultant peculiar solvation structure, the resulting electrolyte not only enables stable, dendrite-free lithium plating-stripping, but also displays anodic stability up to 5.2 V (vs. Li/Li+), in additive or co-solvent-free formulation, and at low salt concentration of 1 M. Consequently, the Li | |LiNi0.5Mn1.5O4 cells using the 1 M LiPF6 in 2,5-dioxahexanedioate based electrolyte retain >97% of the initial capacity after 250 cycles, outperforming the conventional carbonate-based electrolyte formulations, making this, and potentially other dicarbonate solvents promising for future Lithium-based battery practical explorations.

10. Synthesis of micron-particle-size HZSM-5 zeolite with different Si/Al ratio and its catalytic activity for toluene methanol alkylation reaction

HZSM-5 molecular sieve is one of the commonly used catalysts for the alkylation of toluene with methanol to para-xylene (PX). This study introduces a one-step method to directly prepare microscale HZSM-5 catalysts with suitable pore structure and acidity. Specifically, this paper combined crystal seed regulation of the morphology of HZSM-5 and adjusted the Si/Al ratios of the precursor solution for HZSM-5 synthesis to regulate the acidic phase of the HZSM-5. It analyzed the phase composition, pore structure parameters, microstructure, and acidity of HZSM-5 molecular sieve through X-ray diffraction, N2 adsorption desorption, SEM, NH3 temperature programmed desorption, pyridine infrared spectroscopy, etc. This study investigated the catalytic performance of HZSM-5 in the alkylation of toluene with methanol to PX, and explored the effects of pore structure and acidity on the catalytic performance. Results indicate that in the hydrothermal crystallization method, the crystal seeds and aluminum content in the synthesis solution have a significant impact on the acidity and morphology structure of the HZSM-5 molecular sieve. The HZSM-5 (150)molecular sieve catalyst with 5μm grain size exhibits 60% PX selectivity and 11% toluene conversion rate in the evaluation reaction of toluene methanol alkylation due to its longer diffusion path and more suitable acidic sites. By fine-tuning the pores and acidic sites of HZSM-5 molecular sieve, the selectivity for PX has been effectively improved, providing new methods and approaches for the design of shape-selective catalysts.

A theoretical model for the calculation of thermal elastohydrodynamic lubrication performance of the flow distribution pair of piston pumps is established, which is composed of the oil film pressure governing equation and energy equation, and solved by means of numerical solution and simulation. We carry out quantitative analysis of the influence of various parameters on the thermal elastohydrodynamic lubrication characteristics of the flow distribution pair. The results indicate that both the oil film thickness and the cylinder tilt angle of the flow distribution pair vary in a periodic manner. The increase in the rotational speed of the cylinder block will increase the film thickness of the oil film and reduce the fluctuation, and the inclination angle of the cylinder block and its fluctuation amplitude will decrease. An increase in working pressure will lead to a decrease in the average oil film thickness, an increase in fluctuations, and an elevation in both the tilt angle of the cylinder block and its fluctuation amplitude. The increase in the rotational speed of the cylinder block and the increase in the working pressure will lead to the increase in the viscous friction dissipation of the flow distribution pair, the increase in the oil film temperature and the increase in the leakage. The reduction in the sealing belt will lead to the reduction in oil film friction torque and leakage.

12. Improving Hydrolytic Activity and Enantioselectivity of Epoxide Hydrolase from Phanerochaete chrysosporium by Directed Evolution

Epoxide hydrolases (EHs) catalyze the conversion of epoxides into vicinal diols. The epoxide hydrolase gene from P. chrysosporium was previously cloned and subjected to site-directed mutation to study its enzyme activity, but the results were unsatisfactory. This study used error prone PCR and DNA shuffling to construct a PchEHA mutation library. We performed mutation-site combinations on PchEHA based on enzyme activity measurement results combined with directed evolution technology. More than 15,000 mutants were randomly selected for the preliminary screening of PchEHA enzyme activity alongside 38 mutant strains with increased enzyme activity or enantioselectivity. Protein expression and purification were conducted to determine the hydrolytic activity of PchEHA, and three mutants increased their activity by more than 95% compared with that of the wt. After multiple rounds of screening and site-specific mutagenesis, we found that F3 offers the best enzyme activity and enantioselectivity; furthermore, the molecular docking results confirmed this result. Overall, this study uncovered novel mutants with potential value as industrial biocatalysts.

During the COVID-19 pandemic, forecasting COVID-19 trends to support planning and response was a priority for scientists and decision makers alike. In the United States, COVID-19 forecasting was coordinated by a large group of universities, companies, and government entities led by the Centers for Disease Control and Prevention and the US COVID-19 Forecast Hub (https://covid19forecasthub.org). We evaluated approximately 9.7 million forecasts of weekly state-level COVID-19 cases for predictions 1-4 weeks into the future submitted by 24 teams from August 2020 to December 2021. We assessed coverage of central prediction intervals and weighted interval scores (WIS), adjusting for missing forecasts relative to a baseline forecast, and used a Gaussian generalized estimating equation (GEE) model to evaluate differences in skill across epidemic phases that were defined by the effective reproduction number. Overall, we found high variation in skill across individual models, with ensemble-based forecasts outperforming other approaches. Forecast skill relative to the baseline was generally higher for larger jurisdictions (e.g., states compared to counties). Over time, forecasts generally performed worst in periods of rapid changes in reported cases (either in increasing or decreasing epidemic phases) with 95% prediction interval coverage dropping below 50% during the growth phases of the winter 2020, Delta, and Omicron waves. Ideally, case forecasts could serve as a leading indicator of changes in transmission dynamics. However, while most COVID-19 case forecasts outperformed a naïve baseline model, even the most accurate case forecasts were unreliable in key phases. Further research could improve forecasts of leading indicators, like COVID-19 cases, by leveraging additional real-time data, addressing performance across phases, improving the characterization of forecast confidence, and ensuring that forecasts were coherent across spatial scales. In the meantime, it is critical for forecast users to appreciate current limitations and use a broad set of indicators to inform pandemic-related decision making.

IntroductionSound waves are refracted along the direction of their propagation owing to spatial and temporal fluctuations in the speed of sound in seawater. Errors are compounded when sound speed profiles (SSPs) with low precision are used to detect and locate distant underwater targets because an accurate SSP is critical for the identification of underwater objects based on acoustic data. Only sparse historical spatiotemporal data on the SSP of the South China Sea are available owing to political issues, its complex atmospheric system, and the unique topography of its seabed, because of which frequent oceanic movements at the mesoscale affect the accuracy of inversion of its SSP.MethodIn this study, we propose a method for the inversion of the SSP of the South China Sea based on a long short-term memory model. We use continuous-time data on the SSP of the South China Sea as well as satellite observations of the height and temperature of the sea surface to make use of the long-term and short-term memory-related capacities of the proposed model.ResultIt can achieve highly accurate results while using a small number of samples by virtue of the unique structure of its memory. Compared with the single empirical orthogonal function regression method, the inversion accuracy of this model is improved by 24.5%, and it performed exceptionally well in regions with frequent mesoscale movements.DiscussionThis enables it to effectively address the challenges posed by the sparse sample distribution and the frequent mesoscale movements of the South China Sea.

BackgroundOur previous multicenter case-control study showed that aging, up-regulation of platelet glycogen synthase kinase-3β (GSK-3β), impaired olfactory function, and ApoE ϵ4 genotype were associated with cognitive decline in type 2 diabetes mellitus (T2DM) patients. However, the causal relationship between these biomarkers and the development of cognitive decline in T2DM patients remains unclear.MethodsTo further investigate this potential relationship, we designed a 6-year follow-up study in 273 T2DM patients with normal cognitive in our previous study. Baseline characteristics of the study population were compared between T2DM patients with and without incident mild cognitive impairment (MCI). We utilized Cox proportional hazard regression models to assess the risk of cognitive impairment associated with various baseline biomarkers. Receiver operating characteristic curves (ROC) were performed to evaluate the diagnostic accuracy of these biomarkers in predicting cognitive impairment.ResultsDuring a median follow-up time of 6 years (with a range of 4 to 9 years), 40 patients (16.13%) with T2DM developed MCI. 16. Potential selection for lipid kinase activity and spermatogenesis in Henan native pig breeds and growth shaping by introgression of European genes

Abstract Background China has one third of the worldwide indigenous pig breeds. The Henan province is one of the earliest pig domestication centers of China (about 8000 years ago). However, the precise genetic characteristics of the Henan local pig breeds are still obscure. To understand the origin and the effects of selection on these breeds, we performed various analyses on lineage composition, genetic structure, and detection of selection sweeps and introgression in three of these breeds (Queshan, Nanyang and Huainan) using genotyping data on 125 Queshan, 75 Nanyang, 16 Huainan pigs and 878 individuals from 43 Eurasian pig breeds. Results We found no clear evidence of ancestral domestic pig DNA lineage in the Henan local breeds, which have an extremely complicated genetic background. Not only do they share genes with some northern Chinese pig breeds, such as Erhualian, Hetaodaer, and Laiwu, but they also have a high admixture of genes from foreign pig breeds (33–40%). Two striking selection sweeps in small regions of chromosomes 2 and 14 common to the Queshan and Nanyang breeds were identified. The most significant enrichment was for lipid kinase activity (GO:0043550) with the genes FII, AMBRA1, and PIK3IP1. Another interesting 636.35-kb region on chromosome 14 contained a cluster of spermatogenesis genes (OSBP2, GAL3ST1, PLA2G3, LIMK2, and PATZ1), a bisexual sterility gene MORC2, and a fat deposition gene SELENOM. Reproduction and growth genes LRP4, FII, and ARHGAP1 were present in a 238.05-kb region on SSC2 under selection. We also identified five loci associated with body length (P = 0.004) on chromosomes 1 and 12 that were introgressed from foreign pig breeds into the Henan breeds. In addition, the Chinese indigenous pig breeds fell into four main types instead of the previously reported six, among which the Eastern type could be divided into two subgroups. Conclusions Admixture of North China, East China and foreign pigs contributed to high genetic diversity of Henan local pigs. Ontology terms associated with lipid kinase activity and spermatogenesis and growth shaping by introgression of European genes in Henan pigs were identified through selective sweep analyses.

Aimed at the problem of insufficient heat resistance and poor ablation resistance of phenolic resin (RF), and the compatibility of SiO2 particles with phenolic resin, the nano-scale SiO2/RF hybrid aerogel was prepared by the co-gel method. The interpenetrated gel network was constructed to increase the compatibility of two phases. The microstructure, chemical structure and thermophysical properties of the SiO2/RF hybrid aerogel were explored. The silica-modified phenolic/carbon fiber composite material was prepared. The ablation properties of the composite material before and after modification were compared. The results show that the hybrid aerogel possesses a bi-continuous structure of skeleton and pores. The density of the hybrid aerogel fluctuates in the range of 0.145-0.160 g/cm3. As the silica content increases, the residual carbon ratio of the hybrid aerogel increases, and the Si—O bond absorption vibration peak is more obvious, but XRD has no diffraction peak. Considering the pore size distribution and thermal physical properties, the silica-modified phenolic/carbon fiber composite material was prepared with the best performance hybrid aerogel. The mass ablation rate of the modified composite material is 0.046 g/s, and the linear ablation rate is 0.074 mm/s. Compared with the unmodified composite material, the mass ablation rate is reduced by 20.7%, the linear ablation rate is reduced by 21.3%. The oxidation resistance and the residual carbon ratio of the modified material are significantly improved.

Research on ocean ambient noise is highly important for environment monitoring, marine mammal protection, underwater communication and navigation. In this paper, we present the long-term statistics and wind dependence of near-bottom and deep-sea ambient noise in the northwest South China Sea, at a depth of 1240 m. The data were collected from 11th July 2022 to 31st December 2022 together with local wind speeds ranging from 1 to 58 knots (two typhoons involved), and the processing frequency band is between 20 and 2000 Hz. The long-term mean noise level is calculated along with its skewness, kurtosis and percentile distributions. Diurnal and monthly average of noise levels are analyzed, and the large fluctuations in lower (≤100 Hz) and higher (≥400 Hz) frequencies are respectively caused by the variation of the number of nearby and distant ships and the diverse distributions of the windspeeds in individual months. We find that the noise level in winter (Dec.) is 10~11 dB higher than that in summer (Jul.) at higher frequencies. The probability densities of noise levels in the situation of a fixed wind speed are likely to obey the Burr distributions in low frequencies (50 and 100 Hz) and the Weibull distributions in high frequencies (400 and 1000 Hz). In addition, the mean noise levels for different Beaufort scales match well with the 5-dB-addtion Wenz curves, and a mathematic relationship is acquired between the noise level and wind speed in the experimental site. The results are of great representativeness, and are significant to data-driven noise modelling, evaluation and improvement of sonar performance in the region of South China Sea with an incomplete deep-water sound channel.

In this paper, we present a scientometric approach to explore the trends of the association between TRPV1 channel and inflammation and our goal is to provide creative directions for future research. The related literature was retrieved from Web of Science Core Collection and then analyzed by CiteSpace and VOSviewer. A total of 1533 documents were screened. The most productive country, institution, journal, author, cited journal, cited author, and references were the United States, University of California, San Francisco, Pain, Lu-yuan Lee, Nature, Michael J. Caterina, and Caterina MJ (Science, 2000), respectively. The most influential country and institution were Switzerland and University of California, San Francisco, respectively. The cooperation among countries or institutions was extensive. Amounts of documents were distributed in molecular, biology, genetics. TRPV1-associated neurons, neuropeptides, neuropathic pain, neuroinflammation, and neurogenic inflammation were mainly hotspots in this field. The research has presented valuable data about previous studies in the link of TRPV1 channel and inflammation.

The practical applications of lithium metal batteries are limited by uncontrolled dendrite growth during cycling. Herein, we propose a simple and scalable approach to stabilize lithium metal anodes using laser scribing technology to integratively design and construct a laser-induced graphene (LIG) with lithiophilic metal oxide nanoparticles. The porous LIG and lithiophilic MnOx nanoparticles effectively reduce the nucleation overpotential of Li and regulate uniform Li plating, while the array structure offers continuous and ultra-fast ion/electron transport channels, accelerating Li+ transport kinetics at high rate and high capacity. Consequently, the Li@MnOx@LIG-a anode exhibits superior rate capability of up to 40 mA cm−2 with low nucleation overpotential. It also can withstand ultra-high Li capacity to 20 mAh cm−2 without dendrite growth and stably cycle for 3000 h with 100% depth of discharge at 40 mA cm−2. More importantly, this technology can be expanded to other metal oxides for various metal batteries.

Lycii Fructus (LF) is a medicinal-edible homology material frequently used for treating age-related macular degeneration (AMD) in Chinese medicine. Abundant of literatures verified the retina protection effect of LF on AMD-like animal models. However, previous studies on LF attenuating retinal damage mainly focused on its protection effect on pigment epithelial cells and optic ganglion cells while few reports on Müller cells. In the current study, we found that LF extracts attenuated oxidation damages and inhibited cell apoptosis of retina on a mouse retinal injury model induced by sodium iodate. LF extracts also increased expressions of Müller cell biomarker glutamine synthetase of eyeballs of model mice, which indicated that LF extract protected Müller cell from oxidation injury induced by sodium iodate. In Müller cell MIO-M1 cultures, LF also significantly inhibited cellular apoptosis induced by NaIO3 by regulating Nrf2/HO-1 signaling pathway and the aqueous extracts exerted better active tendency than ethanol extracts. These data implied that the antioxidant effect of LF on protecting Müller cell might contribute to its retina repair function, thereby providing more scientific evidences for the development of LF as an alternative therapy or preventive measures for AMD.

Abstract Advanced mRNA vaccines play vital roles against SARS-CoV-2. However, most current mRNA delivery platforms need to be stored at −20 °C or −70 °C due to their poor stability, which severely restricts their availability. Herein, we develop a lyophilization technique to prepare SARS-CoV-2 mRNA-lipid nanoparticle vaccines with long-term thermostability. The physiochemical properties and bioactivities of lyophilized vaccines showed no change at 25 °C over 6 months, and the lyophilized SARS-CoV-2 mRNA vaccines could elicit potent humoral and cellular immunity whether in mice, rabbits, or rhesus macaques. Furthermore, in the human trial, administration of lyophilized Omicron mRNA vaccine as a booster shot also engendered strong immunity without severe adverse events, where the titers of neutralizing antibodies against Omicron BA.1/BA.2/BA.4 were increased by at least 253-fold after a booster shot following two doses of the commercial inactivated vaccine, CoronaVac. This lyophilization platform overcomes the instability of mRNA vaccines without affecting their bioactivity and significantly improves their accessibility, particularly in remote regions.

It is important to know the safety and efficacy of vaccination in immunocompromised people living with HIV (PLWH), but currently, there is limited data on the inactivated SARS-CoV-2 vaccines’ safety and immune responses in PLWH. In this prospective observational study, 139 PLWH and 120 healthy controls were enrolled and monitored for 21–105 days after a two-dose vaccination. The safety, anti-receptor binding domain IgG (anti-RBD-IgG) and anti-spike-IgG responses, and RBD-specific memory B cell (MBC) responses were evaluated. Coastal acoustic tomography (CAT) is a remote sensing technique that utilizes acoustic methodologies to measure the dynamic characteristics of the ocean in expansive marine domains. This approach leverages the speed of sound propagation to derive vital ocean parameters such as temperature and salinity by inversely estimating the acoustic ray speed during its traversal through the aquatic medium. Concurrently, analyzing the speed of different acoustic waves in their round-trip propagation enables the inverse estimation of dynamic hydrographic features, including flow velocity and directional attributes. An accurate forecasting of inversion answers in CAT rapidly contributes to a comprehensive analysis of the evolving ocean environment and its inherent characteristics. Graph neural network (GNN) is a new network architecture with strong spatial modeling and extraordinary generalization. We proposed a novel method: employing GraphSAGE to predict inversion answers in OAT, using experimental datasets collected at the Huangcai Reservoir for prediction. The results show an average error 0.01% for sound speed prediction and 0.29% for temperature predictions along each station pairwise. This adequately fulfills the real-time and exigent requirements for practical deployment.

Abstract Polyaryletherketone is a kind of special engineering plastics with excellent comprehensive properties, high strength, good stability, and almost insoluble in any common solvent except concentrated sulfuric acid at room temperature. However, its high processing temperature also hinders its application and development in the foam. Here, a series of polyetheretherketone (PEEK) and diphenyl polyetheretherketone (PEDEK) composite foams are prepared by temperature induced phase separation, using diphenyl sulfone as solvent. The microstructure, thermal and mechanical properties of PEEK and PEDEK foam are compared. Highly porous PEEK and PEDEK foams with densities ranging from 0.13 to 0.25 g cm−3, with compressive strength ranging from 0.68 to 2.6 MPa, respectively, are produced. Meanwhile, the introduction of diphenyl structure can effectively improve the performance of PEEK foams at high temperature. It is found that PEDEK foam has a higher operating temperature than PEEK, and the compressive strength of 0.24 g cm−3 foam is still ≈1 MPa at 200 °C.

Summary: Background: The presence of gross extrathyroidal extension (ETE) in thyroid cancer will affect the prognosis of patients, but imaging examination cannot provide a reliable diagnosis for it. This study was conducted to develop a deep learning (DL) model for localization and evaluation of thyroid cancer nodules in ultrasound images before surgery for the presence of gross ETE. Methods: From January 2016 to December 2021 grayscale ultrasound images of 806 thyroid cancer nodules (4451 images) from 4 medical centers were retrospectively analyzed, including 517 no gross ETE nodules and 289 gross ETE nodules. 283 no gross ETE nodules and 158 gross ETE nodules were randomly selected from the internal dataset to form a training set and validation set (2914 images), and a multitask DL model was constructed for diagnosing gross ETE. In addition, the clinical model and the clinical and DL combined model were constructed. In the internal test set [974 images (139 no gross ETE nodules and 83 gross ETE nodules)] and the external test set [563 images (95 no gross ETE nodules and 48 gross ETE nodules)], the diagnostic performance of DL model was verified based on the pathological results. And then, compared the results with the diagnosis by 2 senior and 2 junior radiologists. The quality of perovskite films plays a crucial role in the performance of perovskite solar cells (Pero‐SCs) in terms of power conversion efficiency (PCE) and stability. Herein, an ionic liquid, tetra‐butylammonium trifluoromethanesulfonate (denoted TBASF), is used to treat perovskite films. The tetra‐butylammonium cation (TBA+) can bind the unreacted PbI2 and form a low‐dimensional material on the surface of the perovskite. This membrane improves the water resistance of the perovskite film. Meanwhile, trifluoromethanesulfonate anion (SF−) can passivate the uncoordinated Pb2+ and stabilize I− on the perovskite surface, which is beneficial to the stability of the devices. After the treatment, the Pero‐SCs achieve a champion PCE of 22.80% and excellent stability in highly humid conditions.

Bactericidal/permeability-increasing protein (BPI) exhibits a number of important characteristics. RNA-seq analysis revealed that the BPI expression was increased in platelets of (non)ST-elevated myocardial infarction (NSTEMI/STEMI) patients. Activated platelets can induce NETosis which may be accompanied by the release of myeloperoxidase-DNA (MPO-DNA) and S100A8/A9. This study investigated the plasma BPI levels in myocardial infarction patients and its correlation with MPO-DNA and S100A8/A9. This prospective study recruited 80 control individuals, as well as 63 NSTEMI and 59 STEMI patients who were admitted to the First Affiliated Hospital of Bengbu Medical College for coronary angiography (CAG) and/or percutaneous coronary intervention (PCI) between May 1, 2020 and August 31, 2020. Demographic and clinical characteristics, clinical indicators, hs-CRP, IL-1β, MPO-DNA (a circulated marker of NETs), circulating levels of S100A8/A9 and BPI were measured from each individual. The severity of coronary lesions was evaluated by the Gensini score, based on the results of the CAG. Pearson’s or spearman’s correlation was used to examine the correlation between BPI and the above-mentioned parameters, as well as the severity of coronary artery disease. Linear regression analysis was applied to identify the independent predictive factors of BPI. Received operating characteristic (ROC) curve analysis was used to evaluate the diagnostic efficacy of plasma BPI for MI. The plasma BPI levels increased by 8.76 times in the STEMI group and 5.38 times in the NSTEMI group compared to the control group. The plasma level of hs-CRP and IL-1β in both STEMI and NSTEMI groups were also significantly higher than the control group. In addition, the plasma levels of MPO-DNA and S100A8/A9 in the STEMI and NSTEMI groups were significantly higher than the control group. Plasma levels of BPI were positively correlated with IL-1β, hs-CRP, MPO-DNA and S100A8/A9. The correlation between BPI and the severity of coronary artery disease was also significant. The optimal cutoff value of plasma BPI was 35.1705 ng/ml for MI patients from the ROC curve analysis. Plasma BPI levels are increased in myocardial infarction patients and positively correlated with MPO-DNA and S100A8/A9. Plasma BPI level may serve as a potential biomarker of myocardial infarction.

In this paper, a new tuning strategy is proposed by incorporating a pneumatically actuated metamaterial to achieve on-demand polarization manipulation at THz frequencies. Through controlling the actuation pressure, the device function can be flexibly switched among three types of polarization conversion capabilities within the same operation frequency band, from 1.3 THz to 1.5 THz, in which the mutual conversion between linear polarization and circular polarization, such as a quarter-wave plate, and handedness inversion between circular polarizations as a helicity inverter as well as a helicity keeper, have been successfully achieved between the incidence and reflection. Moreover, the intrinsic tuning mechanism for the polarization manipulation is also discussed.

Abstract Objectives We aimed to verify the role of signal peptide-CUB-EGF-like domain-containing protein3 (SCUBE3) in the hepatocellular carcinoma (HCC) progression. Methods The role of SCUBE3 in HCC cell proliferation, apoptosis, and cell cycle in vitro were detected using MTT assay, colony formation assay, 5-ethynyl-2´-deoxyuridine assay (EDU), Celigo cell counting assay, Caspase3/7 activity assay, and flow cytometry. The effect of SCUBE3 on HCC cell proliferation in vivo was inspected by a xenograft tumour model in nude mice. The related mechanisms were further studied. Results The level of SCUBE3 was upregulated in HCC tissues and cell lines. Knockdown of SCUBE3 inhibited proliferation, promoted apoptosis, and induced cell cycle arrest in HCC cell lines in vitro and in vivo. Screening of cell cycle-related proteins revealed that CCNL2, CDK6, CCNE1, and CCND1 exhibited a significantly different expression profile. We found that SCUBE3 may promote the proliferation of HCC cells by regulating CCNE1 expression. The pathway enrichment analysis showed that the TGFβ signalling pathway and the PI3K/AKT signalling pathway were significantly altered. Co-immunoprecipitation results showed that SCUBE3 binds to the TGFβRII receptor. SCUBE3 knockdown inhibited the PI3K/AKT signalling pathway and the phosphorylation of GSK3β to inhibit its kinase activity. Conclusions SCUBE3 promotes HCC development by regulating CCNE1 via TGFβ/PI3K/AKT/GSK3β pathway. In addition, SCUBE3 may be a new molecular target for the clinical diagnosis and treatment of HCC.

Paeonia lactiflora Pall. (Paeonia) has aroused many concerns due to its extensive medicinal value, in which monoterpene glucoside paeoniflorin and its derivatives are the active chemical components. However, little is known in the molecular mechanism of monoterpenoids biosynthesis, and the regulation network between small RNAs and mRNAs in monoterpenoids biosynthesis has not been investigated yet. Herein, we attempted to reveal the tissue-specific regulation network of miRNAs and their targets related to paeoniaflorin and monoterpenoids biosynthesis in Paeonia by combining mRNA and miRNA expression data with degradome analysis. In all, 289 miRNAs and 30177 unigenes were identified, of which nine miRNAs from seven miRNA families including miR396, miR393, miR835, miR1144, miR3638, miR5794 and miR9555 were verified as monoterpenoids biosynthesis-related miRNAs by degradome sequencing. Moreover, the co-expression network analysis showed that four monoterpenoids-regulating TFs, namely AP2, MYBC1, SPL12 and TCP2, were putatively regulated by five miRNAs including miR172, miR828, miR858, miR156 and miR319, respectively. The present study will improve our knowledge of the molecular mechanisms of the paeoniaflorin and monoterpenoids biosynthesis mediated by miRNA to a new level, and provide a valuable resource for further study on Paeonia.

Underwater environment observation with underwater acoustic tomography has been considerably developed in recent years. Moving sound transmission can obtain the observation of entire spatial area with sound station moving. Various internal structures, unique surface and submarine boundaries and changing environment constitutes a complex acoustic propagation channel. This paper focus on the inversion method and signal resampling for sound moving transmission. Also, the current field in three-dimensional (3D) scale is also studied. A five-station sound transmission experiment with four moored station and one moving station that conducted in range of 500m×500m at Huangcai reservoir, Changsha, China is presented. Signal resampling is performed to get correlation of received acoustic data. The vertical layer-averaged flow current results between moving station and moored station are inversed with 2D grid method. 3D flow current field result is composed by grid-averaged inversion current of vertical profile via moving station at different moment. The received results of reciprocal signal transmission between two moored stations and one moving station were used for layer-averaged current variations at vertical scale and grid-averaged current. The feasibility of the method in underwater moving acoustic tomography research is proved and its applicability is discussed. The proposed underwater acoustic tomography technology develops an innovative idea for the further development of temporal- spatial grided tomography observation.

Abstract Transforming acidic coiled-coil containing protein1 (TACC1) is closely related to transcription, translation and centrosome dynamics. Dysregulation of TACC1 is associated with multiple malignancies. Alternative splicing (AS) of TACC1 produces multiple variants, which are of great significance in cancer biology. However, the expression and biological functions of TACC1 variants in head and neck squamous cell carcinoma (HNSCC) remain unclear. In this study, we found for the first time that TACC1 variants exhibited a characteristic expression pattern and that TACC1 variant25 (TACC1v25) was downregulated in HNSCC tissues and cell lines. Overexpression of TACC1v25 in Cal27 and Fadu cells significantly inhibited proliferation and promoted autophagy. Moreover, expression levels of nuclear pERK and p-mTOR were significantly decreased, while the expression of Beclin-1 and the LC3II/LC3I ratio were increased in TACC1v25-overexpressed Cal27 and Fadu cells. After the addition of AKT activator SC79 to TACC1v25-overexpressed Cal27 and Fadu cells, the autophagy levels were remarkably rescued. In conclusion, TACC1v25 inhibits HNSCC progression through the ERK and AKT/mTOR pathways by inhibiting proliferation and increasing autophagy. TACC1v25 might have potential use as a tumour suppressor in HNSCC.

A two-dimensional optical fiber grating with multi sub-gratings based on a 2D spatially encoded phase mask is designed in this study. The 2D fiber Bragg grating (FBG) is composed of two non-overlapping sub-FBGs, which are laterally separated along the radial direction of the fiber core. Unlike traditional FBGs, the refractive index of the 2D FBG is modulated both on the axial and radial directions of the fiber core, which are realized by spatially encoded diffraction based on a 2D spatially encoded phase mask. Compared with the overlapping grating, the 2D FBG can be fabricated at one time to achieve multi-wavelength output, and its compact structure provides a new idea for multi-wavelength multiplexing. As examples, a single-core multi-channel moiré fiber grating and 2D FBG with three subgratings are designed and fabricated on a single-mode fiber and LMA fiber, respectively. Using a 2D spatially encoded mask can improve the optical fiber refractive index modulation from one- to two- or even three-dimensions, which is helpful for precise manipulation of the complex optical field of optical fiber.

Background Various tumors are insensitive to immune checkpoint blockade (ICB) therapy. Toll-like receptors (TLRs) establish the link between innate and adaptive immunity, which can assist T-cell activation and serve as promising targets for combination to enhance ICB therapy. Here, we aimed to improve efficacy for anti-programmed death ligand 1 (PD-L1) therapy by developing a PD-L1/TLR7 dual-targeting nanobody-drug conjugate (NDC), based on the PD-L1 nanobodies and TLR7 agonist we developed.Methods PD-L1 nanobodies were obtained by phage display screening and identified through T-cell activation bioassay, in vivo imaging and quantitative biodistribution study. Immune activation and PD-L1-inducing of TLR7 agonists were evaluated in diverse innate cell models. We constructed PD-L1/TLR7 dual-targeting NDCs by chemically coupling PD-L1 nanobodies and TLR7 agonists. The antitumor effect was evaluated via several murine or humanized solid tumor models. Immunophenotyping, immune cell depletion, tumor rechallenge, RNA sequencing and PD-L1-deficient models were combined to determine the mechanism for NDCs function. The dynamics of the in vivo behaviors of NDCs were assessed based on multiorgan changes in PD-L1 levels.Results The screened PD-L1 nanobodies were characterized as tumor-targeting and alleviated T-cell immunosuppression. The TLR7 agonists induced broad innate immune responses and intratumoral PD-L1 expression on antigen-presenting cells (APCs), and its antitumor effect was dependent on intratumoral delivery. The combination of TLR7 agonists and PD-L1 nanobodies activated both innate and adaptive immunity and upregulated PD-L1-related signaling pathways. After coupling to form dual-targeting NDCs, TLR7 agonists and PD-L1 nanobodies exerted synergistic antitumor effects and safety in either ‘hot’ or ‘cold’ tumor and early or advanced tumor models, reshaped the tumor immune microenvironment and induced antitumor immune memory. CD8+ T cells and natural killer cells were the main effector cells for NDCs to function. NDCs can promote PD-L1 expression on intratumoral APCs and tumor cells, and subsequently achieve targeted enrichment in tumors. Moreover, the efficacy of NDCs is biased toward dependence on host expression of PD-L1.Conclusions The novel PD-L1/TLR7 dual-targeting NDC exhibited potent efficacy against heterogeneous tumors through orchestrating innate and adaptive immunity, which could act as a promising strategy to improve ICB therapy and shows prospects for clinical development.

This work designed a dual-wavelength 2D fiber Bragg grating (FBG) engraved on the single-mode fiber to measure the temperature and strain. The FBG is composed of two sub-gratings that are not overlapped spatially at the same location of the fiber core. Experiments showed that the temperature and strain sensitivities of this grating were separately measured to be 10.64 p.m./°C and 0.882,731 p.m./μɛ at the central wavelength of 1,548 nm, and 10.74 p.m./°C and 0.916,080 p.m./μɛ at the central wavelength of 1,550 nm. These coefficients constitute a coefficient matrix that can solve the problem of cross sensitivity between temperature and strain, which has been verified by varying central wavelengths caused by the synchronous change of temperature and strain.

Acute myeloid leukemia (AML) is a heterogeneous disease characterized by malignant proliferation of myeloid hematopoietic stem/progenitor cells. NPM1 represents the most frequently mutated gene in AML and approximately 30% of AML cases carry NPM1 mutations. Mutated NPM1 result in the cytoplasmic localization of NPM1 (NPM1c). NPM1c interacts with other proteins to block myeloid differentiation, promote cell proliferation and impair DNA damage repair. NPM1 is a good prognostic marker, but some patients ultimately relapse or fail to respond to therapy. It is urgent for us to find optimal therapies for NPM1-mutated AML. Efficacy of multiple drugs is under investigation in NPM1-mutated AML, and several clinical trials have been registered. In this review, we summarize the present knowledge of therapy and focus on the possible therapeutic interventions for NPM1-mutated AML.

44. Effects of Four Extraction Methods on Structure and In Vitro Fermentation Characteristics of Soluble Dietary Fiber from Rape Bee Pollen

In this study, soluble dietary fibers (SDFs) were extracted from rape bee pollen using four methods including acid extraction (AC), alkali extraction (AL), cellulase extraction (CL) and complex enzyme extraction (CE). The effects of different extraction methods on the structure of SDFs and in vitro fermentation characteristics were further investigated. The results showed that the four extraction methods significantly affected the monosaccharide composition molar ratio, molecular weight, surface microstructure and phenolic compounds content, but showed little effect on the typical functional groups and crystal structure. In addition, all SDFs decreased the Firmicutes/Bacteroidota ratio, promoted the growth of beneficial bacteria such as Bacteroides, Parabacteroides and Phascolarctobacterium, inhibited the growth of pathogenic bacteria such as Escherichia-Shigella, and increased the total short-chain fatty acids (SCFAs) concentrations by 1.63–2.45 times, suggesting that the bee pollen SDFs had a positive regulation on gut microbiota. Notably, the SDF obtained by CE exhibited the largest molecular weight, a relatively loose structure, higher extraction yield and phenolic compounds content and the highest SCFA concentration. Overall, our results indicated that CE was an appropriate extraction method of high-quality bee pollen SDF.

Movement variability reflects the adaptation of the neuromuscular control system to internal or external perturbations, but its relationship to stroke-induced injury is still unclear. Eight healthy subjects and ten patients after stroke participated in the experiment, performing a treadmill-walking task at a comfortable speed. The kinematics data of the lower limbs were collected by the motion-capture system, and two indicators, the degree of multifractality (α) and degree of correlation [h(2)], were used to investigate the mechanisms underlying neuromuscular control. The results showed that the knee and ankle joint angles were multifractal and persistent at various scales, and there was a significant difference in the degree of multifractality and the degree of correlation at the knee and ankle joint angles among the three groups, with the values being ranked in the following order: healthy subjects < non-paretic limb < paretic limb. These observations highlighted increased movement variability and multifractal strength in patients after stroke due to neuromotor defects. This study provided evidence that multifractal detrended analysis of the angles of the knee and ankle joints is useful to investigate the changes in movement variability and multifractal after stroke. Further research is needed to verify and promote the clinical applications.

46. An imputation-based genome-wide association study for growth and fatness traits in Sujiang pigs

Sujiang pigs are a synthetic breed derived from Jiangquhai, Fengjing, and Duroc pigs. In this study, we sequenced the genome of 62 pigs with a coverage depth of 10× to 20×, including 27 Sujiang and 35 founder breed pigs, and we collected 360 global pigs’ genome sequence data from public databases including 39 Duroc pigs. We obtained a high-quality variant dataset of 365 Sujiang pigs by imputing the porcine 80 K single nucleotide polymorphism (SNP) Beadchip to the whole-genome scale with a total of 422 pigs as a reference panel. A dataset of 365 imputated Sujiang pigs was used to perform single-trait genome-wide association study (GWAS) and meta-analyses for growth and fatness traits. Single-trait GWAS identified 1 907, 18, and 14 SNPs surpassing the suggestively significant threshold for backfat thickness, chest circumference, and chest width, respectively. Meta-analyses identified 2 400 genome-wide significant SNPs and 520 suggestively significant SNPs for backfat thickness and chest circumference, and 719 genome-wide significant SNPs and 1 225 suggestively significant SNPs for all seven traits. According to the meta-analysis of backfat thickness and chest circumference, a remarkable region of 2.69 Mb on Sus scrofa chromosome 4 containing FAM110B, IMPAD1, LYN, MOS, PENK, PLAG1, SDR16C5 and XKR4 was identified as a candidate region. The haplotype heat map of the 2.69 Mb region verified that Sujiang pigs were derived from Duroc and Chinese indigenous pigs, especially Jiangquhai pigs. The Kruskal-Wallis test showed that haplotypes of the 2.69 Mb region significantly affected backfat thickness and chest circumference traits. We then focused on PLAG1, an important growth-related gene, and identified two synonymous SNPs with obvious differences among different breeds in the PLAG1 gene. We then performed genotyping of 365 Sujiang, 150 Duroc, 95 Jiangquhai, and 100 Fengjing pigs to confirm the above result and verified that the two variants significantly affected phenotypes of growth and fatness traits. Our findings not only provide insights into the genetic architecture of porcine growth and fatness traits but also provide potential markers for selective breeding of these traits in Sujiang pigs.

Abstract Background Sweetpotato (Ipomoea batatas [L.] Lam.) is an important food crop. However, the genetic information of the nuclear genome of this species is difficult to determine accurately because of its large genome and complex genetic background. This drawback has limited studies on the origin, evolution, genetic diversity and other relevant studies on sweetpotato. Results The chloroplast genomes of 107 sweetpotato cultivars were sequenced, assembled and annotated. The resulting chloroplast genomes were comparatively analysed with the published chloroplast genomes of wild species of sweetpotato. High similarity and certain specificity were found among the chloroplast genomes of Ipomoea spp. Phylogenetic analysis could clearly distinguish wild species from cultivars. Ipomoea trifida and Ipomoea tabascana showed the closest relationship with the cultivars, and different haplotypes of ycf1 could be used to distinguish the cultivars from their wild relatives. The genetic structure was analyzed using variations in the chloroplast genome. Compared with traditional nuclear markers, the chloroplast markers designed based on the InDels on the chloroplast genome showed significant advantages. Conclusions Comparative analysis of chloroplast genomes of 107 cultivars and several wild species of sweetpotato was performed to help analyze the evolution, genetic structure and the development of chloroplast DNA markers of sweetpotato.

Underwater acoustic tomography is an advanced technology in water environment observation. Sound propagation duration between transceivers is used for underwater parameter distribution profile reconstruction in the inverse problem. The key points of acoustic tomography are accurate station distance and time synchronization. Two methods are introduced in this study for sound station position correction. The direct signal transmission correction (DSC) method corrects the multi-peak (expect direct ray) travel time via the travel time difference between different sound rays and reference direct ray. The ray-model position correction (RMC) method calculates exact station position by the station drift distance obtained from transceiver depth variations to correct direct ray travel time; the other multi-peak travel time is revised by the corrected direct ray travel time. A water temperature observation experiment was carried out in a reservoir using coastal acoustic tomography (CAT) systems to verify the flexibility of these two methods. Multi-ray arrival peaks are corrected using DSC and RMC methods; water temperature inversion results in a 2D vertical profile are obtained. The reliability of the method is proved by comparison with temperature depth sensor (TD) data. The methods improve the quality of initial data and can be attempted for further water environment observation in acoustic tomography observation studies.

The tuberous root of Tetrastigma hemsleyanum Diels et Gilg (T. hemsleyanum) is a traditional Chinese medicine with a wide range of clinical applications. However, the scarcity of its wild resources, its low yield, and the variable quality that results from its artificial cultivation leads to expensive market prices that are not conducive to the further industrial development of T. hemsleyanum. In this study, transcriptomic and non-targeted metabolomic analyses were integrated to explore the underlying molecular mechanisms and metabolite biosynthesis that occur during its root development. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis revealed that differentially expressed genes (DEGs) were predominantly enriched for processes associated with flavonoid and phenylpropanoid biosynthesis, starch and sucrose metabolism, and plant hormone signal transduction. Genes related to lignin were downregulated in tuberous roots (TRs), resulting in a decrease in lignification and the downregulation of metabolites related to flavonoids and phenylpropanoid biosynthesis. In addition, the expression levels of starch- and sucrose-related genes were upregulated in TRs. The root development of SYQ is also related to IAA, GA, ABA, and JA signaling pathways. Collectively, this study lays the foundation for analyzing the root development and quality-modulating mechanisms employed by T. hemsleyanum; this will be beneficial in conducting molecular-assisted breeding and controlling its secondary metabolite production.

Abstract Laiwu pigs are a Chinese indigenous breed that is renowned for its exceptionally high intramuscular fat content (average greater than 6%), providing an excellent genetic resource for the genetic improvement of meat quality of modern commercial pigs. To uncover genetic diversity, population structure, signature of selection, and potential exotic introgression in this breed, we sampled 238 Laiwu pigs from a state‐supported conservation population and genotyped these individuals using GeneSeek 80K SNP BeadChip. We then conducted in‐depth population genetics analyses for the Laiwu pig in a context of 1,116 pigs from 42 Eurasian diverse breeds. First, we show that the current Laiwu population has more abundant genetic diversity than the population of 18 years ago likely due to gene flow from European commercial breeds. Both neighbor‐joining (NJ) and principal component analyses indicate the introgression of European haplotypes into Laiwu pigs. The admixture analysis reveals that an average 26.66% of Laiwu genetic components are of European origin. Then, we assigned the tested individuals to different families according to their clustering patterns in the NJ tree and proposed a family‐based conservation strategy to reduce the risk of inbreeding depression in Laiwu pigs. Next, we explored three statistics (ROH and iHS and EigenGWAS) to identify a list of candidate genes for fat deposition, reproduction, and growth in Laiwu pigs. Last, we detected a strong signature of introgression from European pigs into Laiwu pigs at the GPC6 locus that regulates the growth of developing long bones. Further association analyses indicate that the introgressed GPC6 haplotype likely contributed to the improvement of growth performance in Laiwu pigs. Altogether, this study not only benefits the better conservation of the Laiwu pig, but also advances our knowledge of the poorly understood effect of human‐mediated introgression on phenotypic traits in Chinese indigenous pigs.
