Abstract

Reported herein is an anti-HIV monochlorinated compound, 1β-acetoxy-3β-chloro-5α,6α-dihydroxycrotocascarin L (1), of the rare crotofolane diterpenoid class. Compound 1, a suspected artifact of extraction, along with the previously undescribed 11β-acetoxycrotocascarin L (2) and a known compound, crotocascarin K (3), were isolated from the bark of Croton megalocarpus, a Kenyan oil-producing seed crop. Compounds 1and 3inhibited HIV-1 replication with IC50values of 28 and 5.5 nM, respectively. Furthermore, both compounds lacked cytotoxicity toward MT-4 cells and FM-55-M1 cells at concentrations of up to 50 μM. Compounds 1and 3were both found to inhibit HIV-1 protease.

Abstract

The subtribe Espeletiinae (Asteraceae) constitutes a recently diversified group endemic to the South American Andes. Due to its impressive morphological diversity and remarkable success in the páramo ecosystem, the subtribe has been the focus of intensive research. However, disagreement persists over the taxonomic subdivisions of Espeletiinae as well as its phylogeny. Chemically, the group is characterized by the presence of ent-kaurane diterpenes, but this information is currently dispersed and fragmented. Considering that chemical markers could aid in developing a deeper understanding of the taxonomy of Espeletiinae, the aim of this study was to (1) review the secondary metabolite chemistry of this subtribe, (2) discuss taxonomic implications and (3) perform phylogenetic analyses using chemical data to assess similarities and differences among the chemical and molecular phylogenies. Interestingly, our phylogenetic analyses using chemical data showed a characteristic geographic structure, with one grade containing mostly Venezuelan taxa and one clade composed mainly of Colombian species, which is in agreement with the molecular phylogeny of the subtribe. Our results suggest that chemical markers can be used to infer chemotaxonomic and chemosystematic relationships in Espeletiinae and offer the first report of the use of Bayesian inference and maximum likelihood methods with chemical data. Abstract

The discovery of antibiotics more than 80 years ago has led to considerable improvements in human and animal health. Although antibiotic resistance in environmental bacteria is ancient, resistance in human pathogens is thought to be a modern phenomenon that is driven by the clinical use of antibiotics1. Here we show that particular lineages of methicillin-resistant Staphylococcus aureus—a notorious human pathogen—appeared in European hedgehogs in the pre-antibiotic era. Subsequently, these lineages spread within the local hedgehog populations and between hedgehogs and secondary hosts, including livestock and humans. We also demonstrate that the hedgehog dermatophyte Trichophyton erinacei produces two β-lactam antibiotics that provide a natural selective environment in which methicillin-resistant S. aureus isolates have an advantage over susceptible isolates. Together, these results suggest that methicillin resistance emerged in the pre-antibiotic era as a co-evolutionary adaptation of S. aureus to the colonization of dermatophyte-infected hedgehogs. The evolution of clinically relevant antibiotic-resistance genes in wild animals and the connectivity of natural, agricultural and human ecosystems demonstrate that the use of a One Health approach is critical for our understanding and management of antibiotic resistance, which is one of the biggest threats to global health, food security and development.
