International audience; With over 32,000 species, teleost fi sh have adapted to a tremendous range of aquatic habitats. At least 200 of them are used for aquaculture all over the world and even more are handled and transported for the ornamental fi sh market, as well as for bait or research purposes, including cytogenetic studies.

The effects of fishing on life history traits and life history strategies of teleost fishes are analysed by a new comparative method that splits traits into an allometric part (size effect), an autoregressive phylogenetic component, and an environmental component (fishing effect). Both intra- and inter-specific variation of age and size at maturity, fecundity, adult size and egg size are analysed by comparing 84 populations of 49 species submitted to various fishing pressures. Two axes of life history diversification are found among teleosts. One is the well-known slow-fast continuum separating short-lived and early maturing species (like Clupeiformes) From longer-lived species that mature late relative to their size and spawn larger eggs (like salmonids or Scorpaeniformes). An additional strategy involves the schedule of resource allocation to growth and reproduction. Indeterminate growth allows higher teleosts (e.g. Gadiformes) to reach a large size while maturing early and laying small eggs. Increasing fishing pressure decreases age at maturity and egg size, and increases Fecundity at maturity, the slope of the fecundity-length relationship and relative size at maturity. These compensations for higher adult mortality differ among life history strategies. Indeterminate growth is associated with a greater flexibility in resource allocation to growth and reproduction that facilitates greater resilience to fishing mortality.

In order to better assess the risk associated with nonylphenol, a lipophilic contaminant of the aquatic environment having a strong tendency to bioaccumulate in organisms and exerting estrogenic effects in fish and invertebrates, we have investigated the capability of three types of freshwater species to metabolize labelled 4-n-nonylphenol. Reverse-phase radio-HPLC was used to isolate and quantify the metabolites. The identification of residues was based on co-chromatography with available standards or authenfified metabolites and when possible by electrospray ionization LC-MS analyses. The residues found in rainbow trout (Oncorhynchus mykiss), pond snails (Lymnea stagnalis) and duckweeds (Lemna minor) were identified. These freshwater organisms are able to extensively biotransform 4-n-nonylphenol. Our attempts to draw the metabolic pathways of nonlyphenol evidenced oxidative attack on the alkyl chain with the production of several hydroxylated compounds and related carboxylic acids. In addition, trout and duck-weeds were able to conjugate the phenol moiety to glucuronic acid and carbohydrates respectively. Traces of sulphate conjugates were also detected in trout tissues whereas no conjugate was detected in pond snails.

Recently, in addition to IGF-1 and IGF-2 the existence of a third form of IGF, termed IGF-3, limited to fishes, to be present only in the gonads and encoded by a separate gene has been reported. However, no further data have been presented on IGF-3. The present study on tilapia (Oreochromis niloticus) uses quantitative real-time PCR specific for tilapia IGF-1 and IGF-3. The organ distribution of IGF-3 mRNA in adult fish and the early ontogeny of IGF-3 in male and female gonads were studied. The potential sensitivity of IGF-3 to GH was revealed by intraperitoneal injections of bream GH using IGF-1 as control gene. The effects of 17alpha-ethinylestradiol (EE2) exerted after feeding of high EE2 doses and exposure to low environmentally relevant EE2 doses on IGF-3 expression in testis and ovary during early development were determined. Low IGF-3 mRNA expression levels were detected in most organs studied, with the highest extra-gonadal amount in the pituitary. During development, the IGF-3 gene was significantly upregulated in male but downregulated in female gonad. Injections of GH elevated IGF-1 mRNA in male and female liver and ovary. IGF-3 did not respond to GH treatment neither in ovary nor in testis. Both EE2 treatments resulted in significant downregulations of IGF-3 mRNA in testis while ovarian IGF-3 mRNA did not respond. Thus, IGF-3 may be involved in reproduction of fishes most likely in the male gonad only. Whether IGF-3 also has some physiological significance in ovary or other organs should be the topic of further studies.

Insulin-like growth factor (IGF)-I, which is crucially involved in fish growth, differentiation, and reproduction, occurs in tilapia pituitary. IGF-I peptide, which is probably produced in hypothalamic perikarya, is present in axons of the neurohypophysis, and IGF-I mRNA and peptide are present in the adenohypophysis in adrenocorticotrophic hormone cells, melanocyte-stimulating hormone cells, with interindividual differences in growth hormone cells, and, starting with puberty, in gonadotrophic hormone (GTH) cells. Subordinate males showed a high IGF-I but a lower beta-luteinizing hormone expression, while in dominant males the opposite was found. IGF-I from the GTH cells may act as auto/paracrine regulators of GTH cell proliferation and enhance GTH synthesis and release during puberty and reproduction, depending on the social status.

Background The hormonal control of oocyte maturation and ovulation as well as the molecular mechanisms of nuclear maturation have been thoroughly studied in fish. In contrast, the other molecular events occurring in the ovary during post-vitellogenesis have received far less attention. Methods Nylon microarrays displaying 9152 rainbow trout cDNAs were hybridized using RNA samples originating from ovarian tissue collected during late vitellogenesis, post-vitellogenesis and oocyte maturation. Differentially expressed genes were identified using a statistical analysis. A supervised clustering analysis was performed using only differentially expressed genes in order to identify gene clusters exhibiting similar expression profiles. In addition, specific genes were selected and their preovulatory ovarian expression was analyzed using real-time PCR. Results From the statistical analysis, 310 differentially expressed genes were identified. Among those genes, 90 were up-regulated at the time of oocyte maturation while 220 exhibited an opposite pattern. After clustering analysis, 90 clones belonging to 3 gene clusters exhibiting the most remarkable expression patterns were kept for further analysis. Using real-time PCR analysis, we observed a strong up-regulation of ion and water transport genes such as aquaporin 4 (aqp4) and pendrin (slc26). In addition, a dramatic up-regulation of vasotocin (avt) gene was observed. Furthermore, angiotensin-converting-enzyme 2 (ace2), coagulation factor V (cf5), adam 22, and the chemokine cxcl14 genes exhibited a sharp up-regulation at the time of oocyte maturation. Finally, ovarian aromatase (cyp19a1) exhibited a dramatic down-regulation over the post-vitellogenic period while a down-regulation of Cytidine monophosphate-N-acetylneuraminic acid hydroxylase (cmah) was observed at the time of oocyte maturation. Conclusion We showed the over or under expression of more that 300 genes, most of them being previously unstudied or unknown in the fish preovulatory ovary. Our data confirmed the down-regulation of estrogen synthesis genes during the preovulatory period. In addition, the strong up-regulation of aqp4 and slc26 genes prior to ovulation suggests their participation in the oocyte hydration process occurring at that time. Furthermore, among the most up-regulated clones, several genes such as cxcl14, ace2, adam22, cf5 have pro-inflammatory, vasodilatory, proteolytics and coagulatory functions. The identity and expression patterns of those genes support the theory comparing ovulation to an inflammatory-like reaction.

Evaluating the ecological impact of an oil spill is a complex issue requiring coherently articulated examination of the sequence of interactions that link the cell, where contaminants exert their effects, to the ecosystem, where interactions with human activities arise. This sequence of interactions traverses the frontiers between scientific disciplines (chemistry, toxicology, physiology, and fisheries ecology). Using the common sole (Solea solea L.) as a model species for the coastal habitats polluted by the "Erika" oil spill, our research project attempted to define indices of functional integrity that characterised the consequences of fuel exposure at the different biological levels. The coupling of field observations with experimental laboratory work revealed how functional alterations which are readily observable within individuals and their organs are progressively obscured as investigation progresses towards more complex organisational levels. Some of the approaches and indices are proposed as instruments for evaluating the impact of contamination by hydrocarbons., L'évaluation des conséquences écologiques d'une marée noire est un problème complexe qui requiert un examen articulé et cohérent de la chaîne d'interactions qui relie l'échelle cellulaire, à laquelle agit le contaminant, à l'échelle écosystémique, où se situent les interactions avec les activités humaines. Cette chaîne d'interactions traverse les frontières des disciplines scientifiques (chimie, toxicologie, physiologie, écologie halieutique). En prenant la sole (Solea solea L.) comme espèce cible - espèce caractéristique des habitats côtiers pollués par l'"Erika" - le programme de recherche ECTOPHY est une première tentative de mise en place d'une approche multidisciplinaire visant à définir, valider et mettre en relation des indicateurs d'intégrité fonctionnelle caractérisant les différents niveaux biologiques. Le couplage d'observations in situ avec des expositions en milieu contrôlé a montré que des pertes de fonctionnalité évidentes et durables à l'échelle de l'individu sont masquées à l'échelle de la population. Certaines des approches (in situ ou expérimentale) et certains des indicateurs testés peuvent être proposés comme instruments utilisables dans le cadre d'évaluation d'impact de pollutions par des hydrocarbures.

Cet article synthétise l’ensemble des études menées sur la biologie et l’écologie de la truite commune (Salmo trutta L.) sur le bassin du Scorff (Morbihan, Bretagne) de 1972 à 1997. Après une brève présentation de la taxonomie et des caractéristiques écologiques générales de l’espèce, les principaux résultats obtenus sont abordés en termes d’éléments de description et de dynamique de populations. La répartition et l’abondance de la truite sur le bassin varient en fonction de l’âge, du milieu (affluent, rivière aval et amont), du type d’habitat et de l’année. Les densités les plus élevées sont observées dans les ruisseaux et les têtes de bassin où se localisent principalement les juvéniles de l’année tandis que les individus plus âgés (un à six ans) colonisent la rivière principale. La profondeur et le couvert sont les deux paramètres de l’habitat qui modulent la distribution de la truite sur le bassin. Les fluctuations annuelles de densité sont fortes (rapport moyen de 1 à 10). La croissance individuelle varie en fonction du milieu (capacité trophique) et de l’année (température). Les différences s’établissent dès la première année de vie. Chez les géniteurs, le rapport des sexes est toujours en faveur des mâles (maturation sexuelle à deux ans). Chez les femelles, la majorité atteint la maturation sexuelle pour la première fois à trois ans, même si des femelles de deux ans peuvent être fréquentes dans les ruisseaux. La maturation à deux ans dépend de la croissance lors de la première année de vie. Le frai a lieu dans les affluents pour la partie aval du bassin et à la fois dans la rivière principale et les affluents dans la partie amont. Les taux de survie ont pu être évalués sur un ruisseau, confirmant que la grande majorité des mortalités est enregistrée durant les premiers mois de vie et plus particulièrement durant la phase de vie sous graviers (forte influence des sédiments fins sur la mortalité). Chez les géniteurs, la survie annuelle des femelles est meilleure (50 %) que chez les mâles (30 %). Les résultats obtenus permettent de déboucher sur un schéma explicatif des différentes stratégies de recrutement de la truite sur le bassin du Scorff. Ils montrent notamment l’existence d’une génération intermédiaire dans certains affluents, venant se placer entre les géniteurs migrant de la rivière principale et les juvéniles (1+ essentiellement) produits par ces ruisseaux et assurant l’essentiel du recrutement en truites dans la partie aval et moyenne de la rivière principale., This paper synthetizes the studies carried out on the biology and ecology of the brown trout (Sahno trutta L.) in the Scorff basin between 1972 and 1997. After a brief introduction on taxonomy and the general ecological characteristics of the species, the main results are shown in terms of a description and the dynamics components of the population. The repartition and abundance of brown trout varied according to age, environment (brook, downstream and upstream main river), habitat type and year. The highest densities were observed in brooks and headwater streams where the young of the year were mainly localized, while the oldest fishes (one to six years old) colonized the main river. Depth and shelter were the two habitat parameters adjusting brown trout distribution within the basin. Yearly variations of density were high (average ratio of 1 to 10). Individual growth varied depending on the environment (trophic capacity) and year (water temperature). The differences were seen early in the first year of life. In spawners there was always a greater ratio of males (sexual maturation at two years old). Most females matured for the first time at three years old, although two years old females might be frequent in brooks. Maturation at two years old depended on the growth during the first year of life. Spawning took place in the tributaries in the downstream of the basin, and both in the main river and the tributaries in the upstream part of the basin. Survival rates were estimated in a brook, where most mortalities were recorded during the first months of life and more particularly during the under gravel phase (a strong influence of fine sediments). These results allowed to propose an explanatory scheme regarding the different recruitment strategies in the brown trout within the Scorff basin. They notably showed the existence of an intermediate generation in some brooks, which took place itself between migrating spawners from the main river and juveniles (mainly 1+) produced from these brooks and ensured the essential trout recruitment in the middle and downstream part of the main river.

Comparative methods are used to investigate the attributes of present species or higher taxa. Difficulties arise from the phylogenetic heritage: taxa are not independent and neglecting phylogenetic inertia can lead to inaccurate results. Within-species variations in life-history traits are also not negligible, but most comparative methods are not designed to take them into account. Taxa are generally described by a single value for each trait. We have developed a new model which permits the incorporation of both the phylogenetic relationships among populations and within-species variations. This is an extension of classical autoregressive models. This family of models was used to study the effect of fishing on six demographic traits measured on 77 populations of teleost fishes.

The common dentex (Dentex dentex) is a sparid fish which is considered a suitable candidate for Mediterranean aquaculture. A seven-weeks feeding trial was conducted over the summer period with common dentex, sea bream (Sparus aurata) and sea bass (Dicentrarchus labrax). All three species were fed to visual satiety with two practical diets with varying crude protein and crude fat levels (55 % protein, 9 % lipids; 46 % protein, 17 % lipids). The initial body weights were 8.2, 11.6 and 17.3 g for common dentex, sea bass and sea bream, respectively. In all cases, specific growth rates were not significantly affected by dietary treatment, but they varied among species (sea bass 1.7-1.8 %; sea bream 2.1-2.3 %; common dentex 3.1-3.2 %). When comparisons between fish species were made, we observed that the increase in growth rates was linked to a decrease of the whole body fat content, as a consequence of a greater utilisation of dietary lipids as energy substrates. Furthermore, in common dentex, the 17 % lipid diet was able to improve protein retention, but this diet effect was less significant in sea bream and sea bass. In contrast to sea bream, the 17 % lipid diet did not up-regulate plasma GH levels in common dentex, which provides additional evidence for a more efficient utilisation of dietary lipids., Le dentex est un sparidé reconnu comme ayant un potentiel aquacole en Méditerranée. Une étude comparative de croissance entre le dentex, la daurade et le bar a été réalisée. Les trois espèces (poids moyen initial: 8,2; 11,6 et 17,3 g respectivement pour le dentex, le bar et la daurade) ont été nourries à satiété avec deux régimes expérimentaux (55 % de protéines brutes avec 9 % de lipides ou 46 % de protéines brutes avec 17 % de lipides). Chez chacune des espèces, le taux de croissance spécifique (TCS) n'a pas varié en fonction du régime alimentaire. En revanche, une différence significative interspécifique a été observée (bar 1,7¿1,8 %; daurade 2,1¿2,3 %; dentex 3,1¿3,2 %) avec également une différence significative dans le dépôt lipidique corporel, reflétant des différences dans l'utilisation des apports énergétiques d'origine lipidique. L'effet de rétention de protéines par des lipides a été plus important chez le dentex que chez les deux autres espèces. Par ailleurs, contrairement à ce qui est observé chez la daurade, le régime riche en lipides n'induit pas une régulation de la sécrétion de l'hormone de croissance chez le dentex, indiquant aussi une meilleure utilisation des lipides alimentaires.

The regulation of the Glucocorticoid Receptor (GR) transcript was investigated in the gills of Atlantic salmon (Salmo salar) during the parr-smolt transformation. Sampling of parr and smolt fish was performed between December and July and in particular during the smoltification period occurring in spring. Quantification of GR transcripts revealed differences between the two groups in March and at the beginning of April. During these dates, the amounts of GR mRNA in parr gills were respectively three and two fold lower than those measured in smolts. In order to determine which factors are responsible for these differences, we studied the long-term effects of prolactin and Cortisol treatments on GR transcript in the gills of presmolt fish. The plasma levels of these two hormones respectively drop and rise during smoltification. Contrary to Cortisol long-term treatment which did not modify the amount of gill GR transcript, short-term treatment induced a significant decrease within 12 hours. Prolactin long-term treatment caused a significant increase of GR transcript abundance after 13 days of implant treatment. This result is unexpected with regard to those obtained in the smoltification analysis but is in agreement with previous studies performed in mammary gland revealing a positive control of PRL on GR in epithelial cells. Our data suggest that the regulation of the GR transcript during the parr-smolt transformation probably involves several hormonal factors.; La régulation de l'expression du gène codant pour le récepteur aux glucocorticoïdes (GR) dans les branchies de saumon atlantique (Salmo salar) a été étudiée durant la période de smoltification. Les prélèvements chez les poissons parrs et smolts ont été réalisés entre décembre et juillet et plus particulièrement pendant la phase de smoltification au printemps. La quantification du transcrit GR a révélé des différences significatives entre les deux groupes en mars et début avril. Durant ces périodes, les quantités d'ARNm GR dans les branchies de saumons parrs sont respectivement trois et deux fois plus faibles comparées à celles mesurées chez les saumons smolts. Dans le but de déterminer quels facteurs interviennent dans ces variations, nous avons étudié les effets à long terme de traitements à la prolactine et au Cortisol sur la quantité de transcrits GR dans les branchies de saumons presmolts. Les concentrations plasmatiques de ces deux hormones chutent et augmentent respectivement durant la période de smoltification. Contrairement au traitement à long terme du Cortisol qui n'a pas modifié la quantité d'ARNm GR, un traitement à court terme par cette même hormone induit une baisse significative des taux de transcrits GR à partir de 12 heures de traitement. D'autre part, un traitement à long terme par la prolactine provoque une augmentation significative des taux de transcrits GR à partir de 13 jours de traitement. Ce résultat, allant à rencontre de ceux obtenus dans l'analyse de la phase de smoltification, est en accord avec les résultats obtenus dans des lignées de cellules épithéliales de glandes mammaires qui démontrent un contrôle positif de la prolactine sur la quantité de récepteurs GR. Nos résultats suggèrent donc que de multiples facteurs doivent probablement intervenir dans la régulation survenant sur la quantité d'ARNm GR branchiale durant la smoltification.

Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

International audience; Adult goldfish put under controlled-raising photoperiod were subjected to a progressive rise in rearing temperature between February and May, or to a sudden rise in February when gametogenesis is initiated or in May when it is in progress. Plasma gonadotropin (c-GtH) was measured by radioimmunoassay. When temperature rose from 10 °C to 20 or 30 °C, a significant increase of immunoreactive c-GtH was observed. In a control group maintained at 10 °C from February to May, the cGtH level increased significantly, but remained significantly lower than the level of the group subjected to a progressive rise in rearing temperature and kept under the same photoperiod. This suggests an effect of both temperature and photoperiod on c-GtH release.; Des poissons rouges adultes placés en photopériode artificielle et croissante sont soumis à une élévation progressive de leur température d’élevage entre février et mai. D’autres subissent une élévation de température rapide, soit en février lors de l’initiation de la gamétogenèse ou en mai lorsque les animaux sont en pleine gamétogenèse. La gonadotropine plasmatique (c-GtH) est dosée par une méthode radioimmunologique. Lorsque la température s’élève de 10 à 20 ou 30 °C, il se produit un accroissement significatif de la c-GtH immunoréactive. Dans les groupes d’animaux maintenus sous photopériode croissante et soumis à des températures d’élevage constantes (10°C) ou croissantes (passage de 10 à 20 °C entre février et mai) les niveaux circulant de c-GtH augmentent significativement, mais l’augmentation est plus marquée pour le lot soumis à l’élévation de température (P < 0,005). Ceci suggère la possibilité d’une action combinée de la température et de la photopériode sur la sécrétion de c-GtH.

International audience; A testicular feed back on the hypothalamo-pituitary system was studied in male rainbow trout (Salmo gairdneri) after bilateral castration and steroid or testicular supplementation. Experiments were carried out at various periods of the spermatogenetic cycle ; in March-April (resting stage), June (initiation of spermatogenesis), October (end of spermatogenesis and before spermiation), and December (running stage). In March and April, plasma t-GTH is low in intact males (< 2 ng/mi) and castration induced a threefold rise of GTH. Testosterone (T) given by a silastic capsule inserted in the body cavity of the castrate did not change plasma t-GTH in comparison with the intact control. When T is given in the same manner in intact males GTH is significantly increased. In June, a significant t-GTH rise was observed after castration (P < 0.05), this rise was prevented by injection of T and estradiol ()2.E When 2E and T were given to intact males plasma t-GTH was significantly decreased. In October plasma t-GTH was high (13 ng/ml) and was not modified by castration ; injected T or E2 had no effects on plasma t-GTH either in intacts or castrates. In December, a sevenfold increase of t-GTH is observed after castration. This was partly suppressed by intraperitoneal injection of T and E2 and declined after stereotaxic implantation of 11-ketotestosterone into the pituitary. It was not significantly suppressed by intraperitoneal injection of testicular extracts. It is suggested that at this period steroids are the main gonadal factors involved in the feed back at the hypothalamo-pituitary system level.

International audience; The number of germ cells in fry gonads was determined from histological sections sampled periodically over a 10-week post-hatching period. Several successive types of germ cells were identified: primordial germ cells (PGC), two kinds of gonocytes (G1 and G2) and all the stages of female meiotic prophase. The mean diameters are shown in table 1. The number of germ cells increased regularly from 47 +/- 35 (SD) after hatching (PGC) to 166 +/- 25 at 2 post-hatching weeks (G1). At 5 post-hatching weeks, two groups of fish could be distinguished by the number of their germ cells (G2) - one group had less than 926 cells and the other more than 1 577 - and by their gonadal morphology (Filiform or with an enlarged anterior part due to germ cell concentration). At 6 post-hatching weeks, the ovary differentiated with organization of the ovarian lamellae and the appearance of oocytes in meiotic prophase (4 out of 10 trout). The gonads of the other fish stayed at the indifferent stage. The number of germ cells was significantly higher in female-type gonads (6 335 +/- 3 558) than in indifferent gonads (1 696 +/- 467). The situation was the same at 8 to 10 weeks.

International audience; Two-year old male rainbow trout, Salmo gairdnerii, were exposed to the following photoperiods at 80 or 16 °C from February 24 to June 19 : constant short photoperiod (8 L +16 D), constant long photoperiod (16 L + 8 D), and gradually decreasing photoperiod (from 16 L to 8 L). Control fish were exposed to natural photoperiod and temperature. Plasma and pituitary gonadotropin levels were determined by radioimmunoassay and correlated with spermatogenic response in testes. Control fish did not show advancement of spermatogenesis, whereas those exposed to decreasing photoperiod at 80 or 16 °C showed complete induction of spermatogenesis. While cooler temperature favoured production of spermatogonia and spermatocytes, the warmer temperature stimulated formation of spermatids and spermatozoa. On the other hand, in trout exposed to constant short or constant long photoperiod, spermatogenic response was weak at both temperatures. Plasma GtH levels increased in groups kept under decreasing photoperiod at both 8 °C as well as 16 °C ; the magnitude of rise of GtH was greater at 16 °C than at 8 °C. GtH levels were significantly higher in trout exposed to gradually decreasing photoperiod than in animals kept under constant short or constant long photoperiod at 80 or 16 °C. The data suggest that the optimum photothermal combination for GtH release and induction of spermatogenesis in the rainbow trout is a gradually decreasing photoperiod (from 16 L to 8 L) at 16 °C.; Les effets de la température et de la photopériode sur le cycle reproducteurde la truite Arc-en-ciel mâle sont appréciés par l’analyse quantitative de la spermatogenèse et la dynamique de sécrétion d’hormone gonadotrope (GtH) mesurée par radioimmunologie. Seule une photopériode décroissante de 16 à 8 h de lumière par jour, appliquée dès le mois de février en fin de période de spermiation naturelle est susceptible d’induire un nouveau cycle spermatogénétique dans les 4 mois qui suivent. Dans ces conditions, une élévation de la température stimule la spermatogenèse qui est complète (formation de spermatozoïdes) à 16 °C. Sous photopériode constante longue ou courte, l’activité spermatogénétique est faible, une légère stimulation des divisions spermatogoniales et une augmentation du nombre de spermatocytes sont enregistrées sous photopériode constante 16 L - 8 N, spécialement à 8°C. Quel que soit le rythme photopériodique, le taux d’hormone gonadotrope circulante est plus élevé à 16

In winther, long photoperiod stimulated gonad growth. When fish were kept at a constant high temperature for 9 consecutive months from September to January, the gonads remained regressive during the first 4 months and then developed normally.; Nous avons testé les effets de la température et de la photopériode sur la gamétogenèse du Poisson rouge (Carassius auratus) à trois saisons différentes (l’automne, l’hiver et le printemps). Dans tous les cas, les températures élevées inhibent l’ovogenèse malgré une augmentation du taux de gonadotropine plasmatique. Les photopériodes longues stimulent la croissance des gonades en hiver seulement. Lorsque les poissons sont élevésà 30 OC pendant une période prolongée (de septembre à juin), leurs gonades demeurent régressées jusqu’au mois de janvier. Après cette période, les gonades se développent normalement.

International audience; Steroid hormones are administrated in food to young fishes during gonad morphogenesis (1 to 6 months) in order to induce preferentially artificial sex reversal to either the male or the female side. The results which follow are evaluated after two years, when most of the fishes reach sexual maturity.

International audience; Les cellules de Sertoli localisées autour des cystes subissent une évolution marquée à mesure que progresse la spermatogenèse. Plusieurs rôles peuvent leur être attribués à partir d'arguments morphologiques. Rôle de soutien : la présence d'engrenage entre les cellules de Sertoli au début de la spermatogenèse fait place, lorsque la taille des cystes augmente, à des liaisons plus élaborées, telles que zona occdudens et surtout desmosomes, associés à des faisceaux de tonofilaments, ce qui constitue une armature qui intervient vraisemblablement dans la consolidation des cystes. Rôle de nutrition : toutes les cellules germinales sont séparées de la membrane basale par une couche de cellules de Sertoli, de sorte que ces dernières sont obligatoirement impliquées dans le transport des métabolites destinés aux cellules germinales. Les éléments figurés du cytoplasme sertolien sont peu nombreux au début dc la spermatogenèse, mais le réticulum endoplasmique (essentiellement du type granulaire), prend une grande importance au cours de la spermiogenèse au moment où toutes les spermatides sont en contact avec les cellules de Sertoli. Rôle de phagocytose : La présence d'inclusions cytoplasmiques (corps résiduels et lysosomes) fait supposer que les cellules de Sertoli interviennent dans la phagocytose des cellules germinales en dégénérescence et des résidus cytoplasmiques éliminés au cours de la spermiogenèse. Les cellules de Sertoli sont également susceptibles d'intervenir dans la morphogenèse des spermatozeugmes.

International audience; The influence of the gonads and/or of LHRH on gonadotropic function, and possibly on gonadal development, was studied in juvenile rainbow trout treated with low doses of testosterone or untreated. Whatever the treatment, all fish of both sexes remained sexually immature, although large individual variations in plasma GTH were observed in all experimental and control groups. This dispersion in the individual values of plasma GTH seemed to be the first sign of an increase in the previously low gonadotropic activity and might announce the beginning of a pubertal period several months before the onset of meiosis in males and of vitellogenesis in females. Castration did not change the plasma and pituitary GTH values, suggesting that control of the gonadotropic function did not depend mainly on sexual steroids during this period. In our experimental conditions, LHRH administration did not lead to any change in plasma or pituitary levels of GTH. Only the testosterone treatment (20 μg) had an effect, causing a higher pituitary GTH load in some fish which was not modified either by castration or by LHRH and decreased with time ; neither of these treatments changed the plasma GTH levels. Thus, the fact that the required amount of GTH was not released to initiate early onset of gametogenesis did not seem to be due to a repressive control by the gonads or to the absence of stimulation by an LHRH-like hypothalamic factor. The quantity of GTH accumulated in the pituitary gland and the dose of steroid used have been discussed.

International audience; During a 9-month period (corresponding to a sexual cycle), the adult male tench, Tinca tinca, was kept in fish farm ponds receiving heated water from a power plant. In 1974, the males were submitted to three different temperature regimes : group I : ambiant temperature ; group II : ambiant temperature + 3 °C ; group III : ambiant temperature + 6 °C. The experiment was repeated in 1975, but only using groups I and Ill. The reproductive cycle and thermal treatment were studied from a quantitative analysis of spermatogenesis, the duration of the spawning cycle and radioimmunoassay (RIA) measurement of gonadotropin (GTH) in plasma and pituitary, using a carp RIA system turned out to be sensitive enough to assay tench GTH, which was expressed in a c-GTH equivalent. Spermatogenesis in the tench was a discontinuous process, starting in the spring and finishing in the summer. It began earlier in heated water in which the spawning period was also considerably longer (3 months in group III against 1 month in group I). At the beginning of spermatogenesis, pituitary and plasma GTH was low, but rose rapidly when spermatogenesis was initiated (appearance of type B spermatogonial cysts and meiosis). The highest GTH levels in the blood were recorded during the spawning period, with important fluctuations probably due to discharges from the pituitary.

International audience; After their first reproductive period, adult male and female rainbow trout were put under three different photoperiodic regimes between January and June : (1) short daylength (6 h of light/day), (2) decreasing daylength (16L : 8D -> 10L : 14D), (3) skeleton photoperiod consisting of short day-length (6L : 18D) with a one-hour flash at various times during the dark cycle in a decreasing daylength fashion (between 16L and 10L), i.e. at the end of the daylight period of the preceding group. The rearing temperature was 15°C. After the fish were killed in June, it was found that decreasing daylength (group 2) had stimulated vitellogenesis. In that group, spermatogenesis was also highly stimulated in all the males, and some were in spermiation. No significant stimulation of vitellogenesis was observed in any of the groups of females which showed a follicular diameter of less than 1 mm. Spermatogenesis had not begun in any males of group 1 and 3 but thetesticular cycle was slightly advanced in group 3 in which residual spermatozoa had disappeared from the lobules, while they still subsisted in group 2. The hypothesis proposing the present pattern of photosensitive phase, shifting between 16L and 10L (group 3) during the gametogenetic cycle, cannot explain the stimulatory effect of the decreasing photoperiod (group 1).

International audience; Cryopreservation trials on rainbow trout (Salmo gairdneri) sperm were carried out using two basic extenders : Mounib's medium (M) and Ménézo's medium (Me) to which were added bovine serum albumin (BSA) and tellurite egg yolk (Institut Pasteur). After 10 p. 100 of DMSO was added to these different deep-freeze diluents (DD), they were mixed with the sperm and then deep-frozen into 100 !Ll pellets on dry ice. The pellets were stored in liquid nitrogen for periods lasting from 3 days to 6 months. The intensity of sperm motility and fertilizing ability were measured before and after cryopreservation. After the sperm was diluted in Ménézo's medium, slight spermatozoon motility was noticed, which probably caused their early exhaustion and would explain the lower fertilizing ability observed after thawing. Mounib's medium gave better results, especially after 10 p. 100 of egg yolk was added. The optimal deep-freeze conditions were : 1/3 dilution, no equilibration after dilution but immediate deep-freezing at a rate of 10 to 40 °C¡min. Thawing had to be carried out rapidly in 10 sec. However, the spermatozoa were altered during the freezing-thawing process, and during insemination more frozen spermatozoa had to be used to equal the fertilization rate obtained with non-frozen sperm. However, the fertile spermatozoa gave normal embryogenesis and no abnormal development was seen up to the vesicle resorption stage. At the end of spermiation, sperm fitness for deep-freezing decreased, perhaps due to sperm senescence. Pooling the sperm of several males partially compensated for the loss of fertilizing ability seen at the end of the reproductive period.

International audience; A stereotaxic apparatus and technique for electrode placement in diencephalic nuclei of sexually immature rainbow trout of 120-140 g body weight is described. An atlas of nuclei in the diencephalon is also presented.

International audience; Juvenile rainbow trout (weighing 10 or 20 g) were treated thrice weekly for 10 weeks with salmon-gonadotropin (S-GTH), salmon pituitary extract (S-PE), S-GTH plus estradiol-17β, and estradiol-17β alone. The effects of these treatments on the oocytes were studied at ultrastructural level. Saline-injected control fish contained oocytes at previtellogenic stage of development. S-GTH (0.1 or 0.5 µg/g) induced a synthesis of endogenous yolk in the oocyte cytoplasm but failed to initiate incorporation of exogenous yolk or vitellogenin into the oocytes. S-GTH (0.1 µg/g) in combination with estradiol-17β (1 µg)g) produced similar results to those with S-GTH alone. Estradiol-17β alone failed to induce vitellogenesis. In contrast, S-PE (10 µkg/g) induced the incorporation of exogenous yolk into the oocyte indicated by an extensive pinocytotic activity at the oocyte membrane. It is suggested that gonadotropin is necessary for the induction of endogenous yolk synthesis and that some other pituitary hormone(s) is involved in the process of incorporation of exogenous yolk or vitellogenin into the oocyte.

International audience; Le phénomène de maturation in vitro des ovocytes de Truite a été utilisé pour mettre au point une méthode de dosage biologique de l’hormone gonadotrope hypophysaire. Chaque extrait à doser a subi une série de dilutions dont l’effet a été testé sur des lots de 25 ovocytes au stadede la vésicule germinale périphérique incubés 4 jours à 10 C°. La dose théorique provoquant 50 p.1 00 de maturation (DE50) a été déterminée avec précision pour chaque extrait. Les DE50 relatives à différents extraits ou hormones ont pu être comparées directement ; ainsi c-GtH(hormone gonadotrope de Carpe) et SG-G100 (extrait gonadotrope de Saumon) se sont révélés environ 1000 fois plus efficaces que o-LH.Malgré la variabilité de sensibilité observée pour un même extrait entre les ovocytes de femelles différentes, la conservation des rapports d’efficacité entre couples d’extraits identiques permet de comparer toutes les séries de dosages, par référence à l’efficacité d’une préparationgonadotrope standard testée sur les ovocytes de chaque femelle. La sensibilité du dosage est suffisante pour déterminer l’activité gonadotrope de l’hypophyse d’une truitelle immature. Elle peut être doublée par l’adjonction d’hydrocortisone (0,5 µg/ml) au milieu d’incubation. Dans l’ensemble la méthode de dosage biologique proposée apparaît sensible et précise, spécifique et représentative de l’activité gonadotrope des Salmonidés, applicable aussi au dosage de l’hormone gonadotrope de Carpe et éventuellement d’autres poissons. Elle ne peut cependant être employée directement pour le dosage plasmatique et son utilisation paraît devoir être limitée au dosage d’extraits hypophysaires. spécifique et représentative de l’activité gonadotrope des Salmonidés, applicable aussi au dosage de l’hormone gonadotrope de Carpe et éventuellement d’autres poissons. Elle ne peut cependant être employée directement pour le dosage plasmatique et son utilisation paraît devoir être limitée au dosage d’extraits hypophysaires.

International audience; Le méthallibure administré en bains à la dose de 2 p.p.m. entraîne chez le poisson rouge (Carassius auratus) une inhibition de la spermatogenèse qui intéresse toutes les cellules germinales à l'exception des spermatogonies A. Lorsque les mâles soumis à ce traitement reçoivent en même temps des injections de poudre hypophysaire brute de Carpe, la spermatogenèse est maintenue et même fortement stimulée. Après la suppression du traitement au méthallibure, la spermatogenèse se rétablit complètement en 20 jours, mais la restauration est plus importante si les animaux reçoivent des injections de poudre hypophysaire brute de Carpe.

International audience; L'administration de méthyltestostérone à raison de 4 0 wg par gramme d'aliment à des alevins de T. nilotica et T. macvochir pendant deux mois à partir de la fin de l'incubation buccale par la mère, a conduit aux résultats suivants :- Chez T. nilotica, nous avons obtenu l'induction complète et fonctionnelle du sexe mâle chez tous les animaux traités. Certains d'entre eux ont fourni une descendance entièrement femelle, révélant un génotype sexuel femelle, ainsi qu'une homogamétie femelle.- Chez T. macrochir, nous avons observé l'induction de la papille génitale de type mâle, et des modifications anormales dans la morphologie de la tête ; à une exception près, tous les animaux expérimentaux étaient stériles, avec disparition complète de la lignée germinale chez la plupart d'entre eux.

International audience; Une étude ultrastructurale de la spermiogenèse et du spermatozoïde d’Anguille dont la maturité a été induite expérimentalement par traitement gonadotrope a mis en évidence un certain nombre de particularités.

International audience; La production spermatogénétique de Poecilia reticulata est de 36 spermatozeugmes par jour soit environ 750 000 spermatozoïdes. Dans cette espèce, le canal testiculaire apparaît comme un véritable organe de stockage desspermatozoïdes qui n'assure pas une élimination régulière des spermatozeugmes ce qui conduit à une grande hétérogénéité dans l'âge des spermatozoïdes et pose le problème du vieillissement des gamètes et de ses conséquences.

La 17alpha-hydroxy-20 beta-dihydroprogestérone (pregn-4-ene-17 alpha, 20 beta-diol-3-one) induit la maturation in vitro d'ovocytes de truites arc-en-ciel, Salmo gairdnerii, entourés de leur follicule. Avec 97 % de maturations vitellines à la concentration de 22 ng/ml, il représente le stéroïde le plus efficace éprouvé jusqu'à présent. « Progestagène » connu chez les salmonidés, son rôle physiologique possible est discuté.

International audience; Grâce à un protocole expérimental standard suivant lequel le sperme ou les ovules sont mélangés à différents milieux (tabl.1i) avant insémination, il a été possible de montrer qu'une solution minérale reconstituant la composition minérale du liquide séminal de Truite (dite milieu minéral de liquide séminal - MMLS) permet de conserver la fécondabilité des ovules pendant 2 heures et du sperme pendant 30 minutes. Le milieu constitue donc un dilueur de conservation dans lequel les gamètes peuvent être maintenus, sans affecter leur fertilité, pendant des temps relativement longs, par opposition au dilueur d'insémination antérieurement défini (Petit et al., 1974) impropre à la conservation des gamètes mais favorable à l'efficacité de l'insémination (solution de NaCl posm 2,50 pH 9,5). Une méthode d'insémination par double dilution peut être proposée en mettant à profit les propriétés de ces 2 dilueurs pour augmenter le rendement de l'insémination : 1ere dilution dans le dilueur de conservation qui maintient l'immobilité des spermatozoïdes et prolonge leur survie puis 2eme dilution avec le dilueur d'insémination qui provoque la mise en mouvement des spermatozoïdes.

International audience; Des expériences ont été conduites dans le but de définir la nature et la molarité du tampon à utiliser avec les deux dilueurs d'insémination et de conservation définis précédemment. Les effets des différents tampons ont été testés d'une part sur la stabilité du pH du dilueur après introduction de quantités croissantes de NaOH et HCI, de liquide coelomique et d'ovules et d'autre part sur le pourcentage de fécondation établi sur de petits lots de 200 ovules. Les conclusions sont les suivantes : 1) Le dilueur d'insémination (solution de NaCI à 250 mosm) peut être avantageusement tamponné avec un tampon carbonate-bicarbonate 0 ,02 M, pH de 9,2 a 9,5 Ce tampon se révèle plus stable après introduction de grande quantité d'ovules que les autres tampons testés (Tvis- HCl, Glycocolle-NaOH), mais le pourcentage de fécondation reste le même avec chacun de ces tampons. 2) Lorsque le milieu minéral de liquide séminal (MMLS) est utilisé comme dilueur de conservation, la conservation du pouvoir fécondant du sperme est meilleure si le tampon utilisé est le Tris-HC1 ; le pH optimum est voisin de 9,0.

International audience; La spermiogenèse de Poecilia veticulata est caractérisée par un allongement important du noyau, accompagné de mouvements complexes de la chromatine en même temps que se manifestent des structures transitoires (formation intercentriolaire et faisceau de microtubules). Le noyau est successivement occupé par trois types de chromatine. Les mitochondries sont peu modifiées ; elles subissent seulement un aplatissement et une élongation et se disposent autour du flagelle duquel elles restent séparées par le canal cytoplasmique. Il subsiste du cytoplasme dans la pièce intermédiaire. Le spermatozoïde mûr conserve la symétrie bilatérale acquise par les jeunes spermatides, il ne comporte pas d'acrosome et ne conserve que le centriole distal modifié.

International audience; Une technique de mesure spectrophotométrique de la concentration en spermatozoïdes du sperme de truite a été mise en oeuvre et un certain nombre de mesures pratiquées pendant la période de reproduction ont montré que :- la concentration varie entre 10.109 et 25.109 spermatozoïdes par ml au début de la fraie (fig. 2) et diminue ensuite ;- la concentration du sperme dans le canal déférent n'est pas constante et diminue dans le sens antéro-postérieur (fig. 4) ;- la quantité de spermatozoïdes par gramme de testicule est très élevée au début de la fraie et diminue ensuite (fig.5 ) ;- quelle que soit la fréquence des prélèvements, la quantité de spermatozoïdes récoltés dans les conditions de la pratique piscicole reste toujours faible par rapport à la quantité totale de spermatozoïdes présents dans les testicules au moment de la fraie : 22 p.100 dans le cas le plus favorable c'est-à-dire lorsque les spermatozoïdes sont collectés tous les 2 jours (tabl. I).

International audience; Des particules de glycogène [3 et ce ont été observées en grande quantité dès le début de la spermiogenèse dans les cellules de Sertoli et les spermatides au cours de la spermiogenèse. Ce glycogène se localise autour des mitochondries de la pièce intermédiaire. La présence de ce glycogène peut être mise en relation avec le type de spermiogenèse qui se déroule dans des cystes, ce qui interdit toute libération de polysaccharides dans le fluide testiculaire. La forte diminution des quantités de glycogène observée dans certains spermatozeugmes du tractus génital mâle et dans les spermatozoïdes stockés dans l’ovaire peut être interprétée comme résultant, soit de processus de vieillissement, soit de l’épuisement des réserves de glycogène au cours de la nage. Dans ces conditions, la présence de glycogène dans le spermatozoïde d’une espèce de Poisson àfécondation interne, pourrait être considérée, du fait de l’absence de liquide séminal, comme une source endogène de matériel énergétique.

International audience; La lumière ovarienne du Guppy présente, dans la partie antéro-dorsale, un ou plusieurs diverticules alvéolés qui forment un réceptacle séminal assurant une longue survie des spermatozoïdes. Ceux-ci sont profondément enfoncés dans le cytoplasme des cellules épithéliales des alvéoles du réceptacle ; l'aspect ultrastructural de ces « cellules hôtes u ainsi que le mode d'insertion des spermatozoïdes permettent de les comparer aux cellules de Sertoli, dans le testicule. Leur dégénérescence, spontanée ou induite, pourrait être l'un des mécanismes responsables de la libération des spermatozoïdes fécondants.

International audience; Dans le testicule d'un certain nombre de Poissons Téléostéens les cellules somatiques intralobulaires peuvent être considérées comme des cellules de Sertoli qui interviennent dans la contention et la nutrition des cellules germinales et dans leur phagocytose si elles dégénèrent. Les cellules de Sertoli assurent en outre, à la fin de la spermatogenèse, un rôle dans l'accumulation et l'excrétion de lipides qui participent vraisemblablement à la formation du liquide séminal. Malgré la présence passagère de réticulum agranulaire, cette étude morphologique ne permet pas de conclure définitivement au caractère stéroïdogène de ces cellules de Sertoli.

International audience; Il existe une confusion importante dans la notion de spécificité d’action des hormones gonadotropes de Poissons. Cette notion a été précisée en utilisant deux méthodes différentes : d’une part la comparaison des activités biologiques de différents extraits hypophysaires de Poissonssur un même récepteur testiculaire, d’autre part la détermination de leur comportement immunologique vis-à-vis d’anticorps hautement spécifiques dirigés contre l’hormone gonadotrope de Carpe c-HG et o-LH. La spermatogenèse du Carassin, Carassius auratus et du Guppy, Poecilia reticutata hypophysectomisés ne peut être regénérée quantitativement qu’avec des hormones gonadotropes ou extraits hypophysaires autologues d’une espèce donneuse très proche de l’espèce receveuse. La technique de compétition isotopique vis-à-vis d’anticorps spécifiques d’une entité moléculaire définie, donne des résultats comparables. Seuls des extraits hypophysaires de Carpe et d’autres Cyprinidés peuvent déplacer de l’hormone de Carpe marquée à l’iode 131 I (c-HGI 131) du complexe c-HGI131, anti c-HG. Les hormones mammaliennes o-LH, o-FSH et HCG n’ont en aucun cas montré d’activité sur la spermatogenèse du Carassin et du Guppy ni d’affinité vis-à-vis d’anticorps dirigés spécifiquement contre une hormone gonadotrope de Poisson. Ces résultats confirment l’existence d’une spécificité zoologique d’action des hormones gonadotropes hypophysaires entre les Mammifères et les poissons teléostéens et à l'intérieur même des différentes familles de téléostéens

International audience; L'utilisation du méthallibure, en bain, aux doses de I à 2 p.p.m., entraîne une inhibition de la spermatogenèse du Guppy, qui est plus lente que celle obtenue après hypophysectomie chirurgicale ; après 3 0 jours de traitement l'mhibition de la spermatogenèse n'est pas totale. Le taux de mortalité reste élevé (20 % au 30e jour à la dose de I p.p.m.) mais il est dû en partie au solvant utilisé.
