Abstract

Mass mortality events in wildlife can be indications of an emerging infectious disease. During the spring and summer of 2021, hundreds of dead passerines were reported across the eastern US. Birds exhibited a range of clinical signs including swollen conjunctiva, ocular discharge, ataxia, and nystagmus. As part of the diagnostic investigation, high-throughput metagenomic next-generation sequencing was performed across three molecular laboratories on samples from affected birds. Many potentially pathogenic microbes were detected, with bacteria forming the largest proportion; however, no singular agent was consistently identified, with many of the detected microbes also found in unaffected (control) birds and thus considered to be subclinical infections. Congruent results across laboratories have helped drive further investigation into alternative causes, including environmental contaminants and nutritional deficiencies. This work highlights the utility of metagenomic approaches in investigations of emerging diseases and provides a framework for future wildlife mortality events. [ABSTRACT FROM AUTHOR]

Abstract

Ischemic myelomalacia secondary to fibrocartilaginous emboli (FCE) is an idiopathic disease in humans and animals. On the other hand, congenital spinal cord malformations result from neural tube defects in fetal development (ie, spinal dysraphism), with structural anomalies referred to collectively as myelodysplasia. Spinal dysraphisms are frequently accompanied by skin and vertebral abnormalities because of the embryogenic relationship. In this observational case study, we report the pathologic findings of 13, 18- to 24-weeks-old pigs from a large conventional operation that presented with acute paraparesis. Ischemic myelomalacia secondary to FCE was observed in 5 of 13 examined pigs. Congenital spinal cord malformations located between the caudal thoracic and sacral spinal cord were identified in 7 pigs, with structural abnormalities that ranged from diplomyelia/split cord malformation to segmental spinal dysgenesis (myelodysplasia) to caudal agenesis. Concurrent myelomalacia and congenital spinal cord malformations in the same or different sites were noted in 2 pigs. No spinal lesion was observed in 3 pigs. Although gross vertebral abnormalities were not observed herein, intervertebral instability due to minor defects in the articular facets, as well as other unidentified factors, is suspected to contribute high incidence of FCE. It is likely that these congenital malformations were previously underdiagnosed or are possibly new conditions associated with continuous inbreeding and genetic improvement in the modern swine industry. Abstract

The 72 kDa heat shock protein (HSP72) is a molecular chaperone that binds native protein with low affinity. These interactions can alter function of the substrate, a property known as HSP-mediated activity control. In the present work, BIAcore instrumentation was used to monitor binding reactions between HSP72 and naturally occurring sequence variants of the measles virus (MV) nucleocapsid protein (N), a structural protein regulating transcription/replication of the viral genome. Binding reactions employed synthetic peptides mimicking a putative HSP72 binding motif of N. Sequences were identified that bound HSP72 with affinities comparable to well-characterized activity control reactions. These sequences, but not those binding with lesser affinity, supported HSP72 activity control of MV transcription/replication. BIAcore instrumentation thus provides an effective way to measure biologically relevant low affinity interactions with structural variants of viral proteins.

Feline immunodeficiency virus (FIV) infection in cats, a disease similar to HIV infection, causes progressive neurological disease as the ratio of CD4/CD8 lymphocytes falls. CD4 lymphocyte depletion with progressive disease may cause the onset of encephalopathy and neurological abnormalities.

Abstract

Feline immunodeficiency virus Maryland isolate (FIV-MD) may cause neurologic abnormalities similar to AIDS-associated encephalopathy in children. AIDS-associated encephalopathy is a degenerative disease of the brain. Among 12 eight-week-old kittens who were evaluated over a 16-month period, six were injected with FIV-MD and six (in the control group) were not. The infected kittens developed immunodeficiency and their growth was delayed. They also developed neurologic abnormalities characterized by repetitive and compulsive behavior as early as four weeks after infection. Three cats displayed this behavior throughout the 16-month follow-up period. Twelve months after infection, a magnetic resonance imaging (MRI) scan of the brains of infected cats revealed wasting of brain tissue and white matter lesions. Electroencephalography (EEG) and evoked potential tests were also performed to evaluate brain function. These electrodiagnostic tests were found to be more sensitive than MRI in detecting neurologic abnormalities caused by FIV-MD.

Abstract

A 2.5-year-old, female llama with weight loss and lethargy had a packed cell volume (PCV) of 45% which increased to 57% over 3 wk. Transrectal ultrasonography revealed a mass of mixed echogenicity involving the right ovary, which was removed. A histopathological diagnosis of granulosa theca cell tumor was made. This is the first report of its kind in a llama.
