Purpose: To examine the remodeling process of both the soft and hard tissue components of the postextraction socket around immediately loaded dental implants after tooth extraction in maxillary esthetic areas. Materials and Methods: Subjects underwent immediate placement of single implants in postextraction sockets without bone grafting, and their immediate provisionalization with custom tooth-like interim crowns were fabricated using digital diagnostic impressions and a dental milling machine. Intraoperative and 1-year follow-up layered scans of the postextraction sockets after implantation were acquired using a 3D optical system. In the short term, subjects underwent computed tomographic scans. Digital impressions for gingival contours, originally stored as STL (standard tessellation language) files, were converted to DICOM (Digital Imaging and Communications in Medicine) files with the implant shoulder working as a referral point, which were then superimposed to 3D radiologic images. The observed volumetric and linear outcomes were measured using a program known as DentaScan. The width of the alveolar crest at the level of the implant shoulder and marginal bone levels were acquired. Nonparametric tests were applied with a level of significance set at P < .01. Results: No failure was reported after a follow-up of 1 year. Little or no inflammation of the treated areas was registered, and there were practically no signs of suppuration. The areas showed a significant reduction in the overall volumes for both soft and bone tissue, with a P value < .0001 from the baseline (0.983 ± 0.172 cm3) to the 1-year survey (0.865 ± 0.156 cm3). If the soft and bone tissue changes were separately evaluated, a significant loss (with a P value < .0001) was registered for only the bone tissues (from 0.434 ± 0.075 to 0.355 ± 0.061 cm3). Moreover, changes in gingival tissue from baseline to the 1-year survey (-0.040 ± 0.067) appeared to be significantly different from the overall volume loss (-0.118 ± 0.083 cm3). A shrinkage in width (-0.5 ± 0.7 mm) was found from baseline (12.6 ± 0.6 mm) to the 1-year followup (12.1 ± 0.9 mm). Marginal bone levels were 0.97 ± 0.70 mm and 0.39 ± 0.78 mm, respectively, at the mesial and distal aspects of the implants. Conclusions: The present analysis suggested that immediately customized provisionalization was effective enough to prevent both volume loss and linear shrinkage at the layers of the treated areas. Moreover, the buccal aspects seemed to be the areas most affected by the loss of volume. Background: Early-stage breast cancer (BC) presents a certain risk of recurrence, leading to variable prognoses and complicating individualized management. Yet, preoperative noninvasive tools for accurate prediction of disease-free survival (DFS) are lacking. This study assessed the potential of strain elastography (SE) and diffuse optical tomography (DOT) for non-invasive preoperative prediction of recurrence in T1 BC and developed a prediction model for estimating the probability of DFS. Methods: A total of 565 eligible patients with T1 invasive BC were enrolled prospectively and followed to investigate the recurrence. The associations between imaging features and DFS were evaluated and a best-prediction model for DFS was developed and validated. Results: During the median follow-up period of 10.8 years, 77 patients (13.6%) developed recurrences. The fully adjusted Cox proportional hazards model showed a significant trend between an increasing strain ratio (SR) (P < 0.001 for trend) and the total hemoglobin concentration (TTHC) (P = 0.001 for trend) and DFS. In the subgroup analysis, an intensified association between SR and DFS was observed among women who were progesterone receptor (PR)-positive, lower Ki-67 expression, HER2 negative, and without adjuvant chemotherapy and without Herceptin treatment (all P < 0.05 for interaction). Significant interactions between TTHC status and the lymphovascular invasion, estrogen receptor (ER) status, PR status, HER2 status, and Herceptin treatment were found for DFS(P < 0.05).The imaging–clinical combined model (TTHC + SR + clinicopathological variables) proved to be the best prediction model (AUC = 0.829, 95% CI = 0.786–0.872) and was identified as a potential risk stratification tool to discriminate the risk probability of recurrence. Conclusion: The combined imaging-clinical model we developed outperformed traditional clinical prognostic indicators, providing a non-invasive, reliable tool for preoperative DFS risk stratification and personalized therapeutic strategies in T1 BC. These findings underscore the importance of integrating advanced imaging techniques into clinical practice and offer support for future research to validate and expand on these predictive methodologies. Background: Autism spectrum disorder (ASD), a neurodevelopmental disorder defined by social communication deficits plus repetitive behaviors and restricted interests, currently affects 1/36 children in the general population. Recent advances in functional brain imaging show promise to provide useful biomarkers of ASD diagnostic likelihood, behavioral trait severity, and even response to therapeutic intervention. However, current gold-standard neuroimaging methods (e.g., functional magnetic resonance imaging, fMRI) are limited in naturalistic studies of brain function underlying ASD-associated behaviors due to the constrained imaging environment. Compared to fMRI, high-density diffuse optical tomography (HD-DOT), a non-invasive and minimally constraining optical neuroimaging modality, can overcome these limitations. Herein, we aimed to establish HD-DOT to evaluate brain function in autistic and non-autistic school-age children as they performed a biological motion perception task previously shown to yield results related to both ASD diagnosis and behavioral traits. Methods: We used HD-DOT to image brain function in 46 ASD school-age participants and 49 non-autistic individuals (NAI) as they viewed dynamic point-light displays of coherent biological and scrambled motion. We assessed group-level cortical brain function with statistical parametric mapping. Additionally, we tested for brain-behavior associations with dimensional metrics of autism traits, as measured with the Social Responsiveness Scale-2, with hierarchical regression models. Results: We found that NAI participants presented stronger brain activity contrast (coherent > scrambled) than ASD children in cortical regions related to visual, motor, and social processing. Additionally, regression models revealed multiple cortical regions in autistic participants where brain function is significantly associated with dimensional measures of ASD traits. Limitations: Optical imaging methods are limited in depth sensitivity and so cannot measure brain activity within deep subcortical regions. However, the field of view of this HD-DOT system includes multiple brain regions previously implicated in both task-based and task-free studies on autism. Conclusions: This study demonstrates that HD-DOT is sensitive to brain function that both differentiates between NAI and ASD groups and correlates with dimensional measures of ASD traits. These findings establish HD-DOT as an effective tool for investigating brain function in autistic and non-autistic children. When undertaking optical sparse projection reconstruction, the reconstruction of the tested field often requires the utilization of a priori knowledge to compensate for the lack of information due to the sparse projection angle. In order to reconstruct the radiation field of unknown materials or in situations where a priori knowledge cannot be obtained, this paper proposes an extremely sparse tomography multispectral temperature field reconstruction algorithm that analyzes the similarity (the similarity here compares and calculates the Euclidean distance of the spectral emissivity values at various wavelengths between different spectral curves) of radiation characteristics of materials under the same pressure and concentration but different temperature, describes the similarity between the radiation information of the tested field using the dynamic time warping (DTW) algorithm, and uses the similarity sum of the radiation information among the subregions of the temperature field as the optimization objective. This is combined with the equation-constrained optimization algorithm and multispectral thermometry to establish the statistical law between the missing information and finally realize the reconstruction of the temperature field. Simulation experiments show that, without any a priori knowledge, the method in this paper can realize reconstruction of the temperature field with an accuracy of 1.53–12.05% under two projection angles and has fewer projection angles and stronger robustness than other methods. A lensless holographic microscope based on in-line holograms and optical diffraction tomography is an ideal imaging system for label-free 3D biological samples and can achieve large-volume imaging with single-cell resolution in a convenient way. However, due to the phase information loss and the missing cone problem, the imaging quality is significantly degraded by the reconstructed artifacts of twin images and out-of-focus images, which severely hinders the identification and interpretation of the objects. We propose an artifacts-free lensless on-chip tomography certified by three-dimensional deconvolution, which facilitates the extraction of real object morphology through straightforward yet effective computation. Initially, a globally valid systemic point spread function (PSF) is generated by simulating the imaging output of an ideal point light source positioned at the origin of the object space coordinate. Subsequently, an iterative three-dimensional deconvolution process is applied to the primitive imaging outcome of the lensless on-chip tomography using this PSF. Through rapid iterations, the optimized imaging result is swiftly obtained. Both the simulated and experimental results indicate that the artifacts-free lensless on-chip tomography can effectively circumvent the reconstructed artifacts and retrieve the real object morphology, which is critical for detailed observation and further quantitative analysis. In addition, we anticipate that the proposed approach has the potential to be transferred to other 3D imaging systems in systemic artifacts removal after corresponding modifications. Macrophages are employed as targets for delivering genes, drugs, or lipid nanoparticles into tumors or other specific sites. Studying the interaction between solid lipid nanoparticles (SLNs) and macrophages is essential for assessing nanotoxicity and advancing the development of nanomedicines. However, limited data are currently available on the membrane microstructure and biochemical changes that occur when macrophages interact with SLNs. We conducted a label-free morphological and biochemical investigation of NR8383 macrophages using optical diffraction tomography (ODT), which validated the efficiency of the SLNs as a drug delivery system. ODT provided intracellular holotomography to characterize the macrophages and fluorescence imaging to analyze delivery efficiency. ODT analysis revealed the responses of phagocytic macrophages. Additionally, a quantitative analysis of lipid droplets using refractive indices revealed that, compared with incubation with normal cells, incubation with SLNs significantly increased the lipid droplet volume and surface area. The uptake of SLNs into macrophages resulted in increased cell volume, surface area, and concentration, which indicated greater morphological and biochemical variability in the treated cells than in the control cells. The results suggest that ODT imaging is promising for understanding the intracellular distribution of SLNs and useful for validating the efficacy of delivery of SLNs to macrophages. Three critical problems, including high-cost instrumentations, time-consuming image recovery, and low image quality, limit clinical applications of diffuse optical tomography (DOT). Image reconstruction based on deep learning can enhance the image quality of DOT, especially in the case of low number of measurements where image reconstruction becomes a more ill-posed and underdetermined problem. Here, we present a sparse-view image reconstruction based on deep learning to recover the absorption coefficient of a phantom. The presented neural network enhances image quality and the results show that deep learning can enhance the contrast to noise-ratio effectively (more than 80%). The presented method provides recovered images with more accurate localization capabilities (an increase in localization metric by more than 30% compared with the classic method). Moreover, the time of image reconstruction is reduced by three orders of magnitude. This technology utilizes the principles of multi-spectral temperature measurement technology, takes the correlation of radiation information between sub-regions of the temperature field as the optimization objective, and establishes statistical rules between the missing information by combining the equation constraint optimization algorithm and multi-spectral temperature measurement technology. Finally, the temperature field to be measured is reconstructed. The simulation and experimental tests show that, without any prior knowledge, the proposed method can reconstruct the temperature field under two projection angles, with an accuracy of 1.64~12.25%. Laser drilling is widely used for fabricating holes in the semiconductor industry due to high throughput and a small heat-affected zone. However, it produces varying depths owing to uncertain external conditions and requires live control at the rate of a few tens of kHz to handle the fast material removal rate. Optical coherent tomography is capable of in situ acquiring a raw interferogram at a high rate (>80 kHz), but the depth extraction is slow due to the involved heavy Fast Fourier Transform (FFT). To address this, an efficient depth-tracking algorithm is proposed to save the FFT. It searches the depth in the raw interferogram locally with a known last depth given the two truths that only one depth exists and the adjacent depths do not change significantly. The proposed algorithm was proven to expedite the measuring rate six times with sub-pixel tracking precision. To further secure the rate against the interrupting of the system, the tracking process is parallelly implemented in a field-programmable gate array. The closed-loop control tests were conducted on probe cards with depth variations introduced by offsetting laser focus. Spectral-domain optical coherence tomography is a pervasive, non-invasive, in vivo biomedical imaging platform that currently utilizes incoherent broadband superluminescent diodes to generate interferograms from which depth and structural information are extracted. Advancements in laser frequency microcombs have enabled the chip-scale broadband generation of discrete frequency sources, with prior soliton and chaotic comb states examined in discrete spectral-domain optical coherence tomography at 1.3 μm. In this work, we demonstrate coherence tomography through Si3N4 microresonator laser frequency microcombs at 1 μm, achieving imaging qualities on-par with or exceeding the equivalent commercial optical coherence tomography system. We characterize the noise performance of our frequency comb states and additionally show that inherent comb line amplitude fluctuations in a chaotic state and the resultant tomograms can be compensated via multi-scan averaging. Background: The emergence and spread of multidrug-resistant Staphylococcus aureus strains demonstrates the urgent need for new antimicrobials. Xanthorrhizol, a plant-derived sesquiterpenoid compound, has a rapid killing effect on methicillin-susceptible strains and methicillin-resistant strains of S. aureus achieving the complete killing of staphylococcal cells within 2 min using 64 µg/mL xanthorrhizol. However, the mechanism of its action is not yet fully understood. Methods: The S. aureus cells treated with xanthorrhizol were studied using optical diffraction tomography. Activity of xanthorrhizol against the wild-type and mscL null mutant of S. aureus ATCC 29213 strain was evaluated in the time-kill assay. Molecular docking was conducted to predict the binding of xanthorrhizol to the SaMscL protein. Results: Xanthorrhizol treatment of S. aureus cells revealed a decrease in cell volume, dry weight, and refractive index (RI), indicating efflux of the cell cytoplasm, which is consistent with the spontaneous activation of the mechanosensitive MscL channel. S. aureus ATCC 29213mscL was significantly more resistant to xanthorrhizol than was the wild-type strain. Xanthorrhizol had an enhanced inhibitory effect on the growth and viability of exponentially growing S. aureus ATCC 29213mscL cells overexpressing the SaMscL protein and led to a noticeable decrease in their viability in the stationary growth phase. The amino acid residues F5, V14, M23, A79, and V84 were predicted to be the residues of the binding pocket for xanthorrhizol. We also showed that xanthorrhizol increased the efflux of solutes such as K+ and glutamate from S. aureus ATCC 29213mscL cells overexpressing SaMscL. Xanthorrhizol enhanced the antibacterial activity of the antibiotic dihydrostreptomycin, which targets the MscL protein. Conclusion: Our findings indicate that xanthorrhizol targets the SaMscL protein in S. aureus cells and may have important implications for the development of a safe antimicrobial agent. Introduction: Sildenafil citrate is an effective treatment for erectile dysfunction (ED). Despite established safety profile, its long-term ocular implications remain unclear. We evaluate the relationship between the duration of use with retinal nerve fibre layer (RNFL), macular and choroidal thickness. Materials and methods: A cross-sectional study was done between July 2020 and June 2021, among 47 ED patients on sildenafil. The subjects fulfilling the inclusion criteria underwent optical coherent tomography (OCT) to evaluate RNFL, macular and choroidal thickness. Linear regression analysis was done to assess the relationship between duration of use with OCT parameters. Other possible associated factors were evaluated. Results: Forty-seven patients with the mean age of 54.30±8.41 years old recruited. These patients had not experienced visual disturbance on each sildenafil use. There were significant correlations between diabetes mellitus (DM) (r=0.330, P=0.023), erection hardness score (EHS) (r=-0.469, P=0.001) and total cumulative dose (r=0.806, P=<0.001) with duration of use. Duration of use had significant negative linear relationship with the average RFNL (b = -0.284, P=<0.001), superior RNFL (b = -0.195, P=0.018), and inferior RNFL (b = -0.887, P=<0.001). Multiple linear regression (MLR) reveals average RNFL was also influenced by total cumulative dose (b = -0.003, P = 0.029). No significant relationship observed to the macular thickness. Significant linear relationship observed between duration of use with sub-foveal choroidal thickness (b = 0.640, P<0.001). Conclusion: Sildenafil in general does not cause visual symptoms, however subclinical ocular changes; RNFL thinning and choroidal thickening may be influenced by its duration of use. In this paper, we consider inverse shape problems coming from diffuse optical tomography and the Helmholtz equation. In both problems, our goal is to reconstruct small volume interior regions from measured data on the exterior surface of an object. In order to achieve this, we will derive an asymptotic expansion of the reciprocity gap functional associated with each problem. The reciprocity gap functional takes in the measured Cauchy data on the exterior surface of the object. In diffuse optical tomography, we prove that a MUSIC-type algorithm that does not require evaluating the Green's function can be used to recover the unknown subregions. This gives an analytically rigorous and computationally simple method for recovering the small volume regions. For the problem coming from inverse scattering, we recover the subregions of interest via a direct sampling method. The direct sampling method presented here allows us to accurately recover the small volume region from one pair of Cauchy data, requiring less data than many direct sampling methods. We also prove that the direct sampling method is stable with respect to noisy data. The aim of our study is to investigate the feasibility and outcomes of using a femtosecond laser (FSL) platform (Ziemer LDV Z8) for deep anterior lamellar keratoplasty (DALK), enabling the creation of mushroom-shaped graft–host junctions, lamellar cuts, and intrastromal tunnels, to facilitate the big bubble, in one step. We included wet lab experiments on nine porcine eyes to assess the laser accuracy and cuts depth using an anterior segment (AS) OCT. This was followed by an interventional prospective case series on 10 eyes with variant corneal pathologies. The Z8 system, with in-built intraoperative optical coherence tomography (iOCT), guided corneal scans and directed the cuts. ASOCT showed visible mushroom configurations, lamellar cuts, and tunnels. Deviations from the target were 1.6%, 2.6%, and 3.5%. Anterior lamellar removal was easy in all clinical cases, including corneal scarring. The intrastromal tunnel was found at the preset location and the mushroom configuration was acquired. A big bubble was achieved in all cases. Type 1, 2, and 3 bubbles were formed in eight, one, and one case, respectively. We describe a new approach to DALK in which the in-built iOCT-guided FSL enables safe, precise, controlled, and reproducible desired cuts in one step. Touch is an essential form of mother-child interaction, instigating better social bonding and emotional stability. We used diffuse optical tomography to explore the relationship between total haemoglobin (HbT) responses to affective touch in the child's brain at two years of age and maternal self-reported prenatal depressive symptoms (EPDS). Affective touch was implemented via slow brushing of the child's right forearm at 3 cm/s and non-affective touch via fast brushing at 30 cm/s and HbT responses were recorded on the left hemisphere. We discovered a cluster in the postcentral gyrus exhibiting a negative correlation (Pearson's r = −0.84, p = 0.015 corrected for multiple comparisons) between child HbT response to affective touch and EPDS at gestational week 34. Based on region of interest (ROI) analysis, we found negative correlations between child responses to affective touch and maternal prenatal EPDS at gestational week 14 in the precentral gyrus, Rolandic operculum and secondary somatosensory cortex. The responses to non-affective touch did not correlate with EPDS in these regions. The number of mother-child dyads was 16. However, by utilising high-density optode arrangements, individualised anatomical models, and video and accelerometry to monitor movement, we were able to minimize methodological sources of variability in the data. The results show that maternal depressive symptoms during pregnancy may be associated with reduced child responses to affective touch in the temporoparietal cortex. Responses to affective touch may be considered as potential biomarkers for psychosocial development in children. An isooctane spray from a high-pressure multihole GDI injector (Bosch HDEV6) was characterised by means of optical extinction tomography, relying on collimated illumination by a focused shadowgraph setup. The tests were carried out in air under ambient conditions at an injection pressure of 300 bar. Spray images were acquired over a 180-degree angular range in 1-degree increments. The critical issues of optical extinction tomography of sprays, related to the strong light extinction by the dense liquid core of fuel jets, were addressed. To mitigate artefacts arising from the reconstruction process, the extinction data were subjected to spatially-variant filtering steps for both raw and post-log data before being analytically inverted through the inverse Radon transform. This approach made it possible to process extinction data at very large optical depths. A nearly complete three-dimensional reconstruction of the spray was obtained, providing significant details of the spray morphology and the internal structure of the jets throughout spray development. As a high-sensitivity and non-invasive technique, optical molecular tomography (OMT) can locate and visualize tissue quantitatively in three dimensions by collecting surface photons generated from luminescent biomarkers. It has great potential for tumor detection, surgery guidance, and pharmacokinetics research studies. However, due to the limited measurable surface photons and the highly scattered feature of photons, the reconstruction of OMT is highly ill-posed and ill-conditioned, which limits the performance in practice. To improve the accuracy of OMT, plenty of studies focus on precisely modeling photon propagation and accurately reconstructing light source. Since these methods are carried out based on different theories, we review famous solutions for the reconstruction in terms of the photon propagation models and inverse reconstruction algorithms to make an overall understanding for the methods of OMT reconstruction. We introduce a new scanning probe microscopy (SPM) concept called reverse tip sample scanning probe microscopy (RTS SPM), where the tip and sample positions are reversed as compared to traditional SPM. The main benefit of RTS SPM over the standard SPM configuration is that it allows for simple and fast tip changes. This overcomes two major limitations of SPM which are slow data acquisition and a strong dependency of the data on the tip condition. A probe chip with thousands of sharp integrated tips is the basis of our concept. We have developed a nanofabrication protocol for Si based probe chips and their functionalization with metal and diamond coatings, evaluated our probe chips for various RTS SPM applications (multi-tip imaging, SPM tomography, and correlative SPM), and showed the high potential of the RTS SPM concept. It is characterized by retinal degeneration, short stature, skeletal anomalies, and neurological defects. We described the clinical features of a Chinese patient with CWC27-related spliceosomeopathy and identified the pathogenic variant.The affected subject underwent detailed ophthalmic examinations. Systemic abnormalities were assessed, including body height, craniofacial morphology, oral cavity, hands, feet, hair and skin. Genomic DNA was isolated from peripheral blood and sequenced by next-generation sequencing. Sanger sequencing was performed for validation and segregation.The patient had poor vision, nyctalopia and nystagmus from childhood. Fundoscopy revealed extensive chorioretinal atrophy with numerous scattered greyish pigmentation. Severe circular areas of macular atrophy were observed. Optical coherent tomography showed reduced retinal thickness with nearly absent ellipsoid zone and retinal pigment epithelium. In addition, craniofacial abnormalities, short statue, brachydactyly, dental anomalies, cafe-au-lait spots, scant hair, absent eyebrows and thin eyelashes were documented. Genetic analysis revealed a novel homozygous novel small deletion c.1133delG(p.G378Efs*12) in CWC27 (NM_005869.2).We present a patient with early-onset retinitis pigmentosa and marked syndromic features. A novel CWC27 pathogenic variant was identified. Our findings broaden the clinical and mutation spectrum of CWC27-related spliceosomeopathy, and could be helpful in diagnosis of this rare disease. A novel high‐speed, high‐resolution 3D microscopy technique named BP‐TIDT is presented that quantifies the refractive index (RI) distribution of label‐free, transparent samples. This method combines a bi‐plane detection scheme (BP) with the transport of intensity diffraction tomography (TIDT), effectively circumventing the need for matched illumination conditions under high numerical aperture (NA) objectives, which enables 15 fps volume rates and 326 nm lateral resolution. The effectiveness and accuracy of the proposed approach are validated through high‐resolution imaging of polystyrene microspheres and HepG2 cells. Moreover, the wide‐ranging applicability of BP‐TIDT is demonstrated by investigating subcellular organelle motion, including mitochondria and lipid droplets, as well as the macroscopic apoptosis process in living COS‐7 cells. To the best of current knowledge, this is the first time that high spatial‐temporal resolution dynamic ODT results are obtained in a non‐interferometric and motion‐free manner, highlighting the potential of BP‐TIDT in advancing research on dynamic cellular processes. Ultrasound-modulated optical tomography (UOT) leverages the strengths of light and sound, enabling deep-tissue imaging with optical absorption contrast and acoustic resolution. Camera-based UOT, with its parallel detection capability, excels at handling weak light–sound interactions. However, the limited frame rate of the camera typically results in poor axial resolution and poses challenges for holographic measurements. In this study, we introduced intersected ultrasonic modulation to address these limitations, thereby achieving equal resolution in both lateral and axial dimensions through referenceless measurements. As a proof of concept, we constructed an imaging system and demonstrated two-dimensional imaging for absorptive targets buried inside a scattering medium. Significance: Scanning laser optical tomography (SLOT) is a volumetric multimodal imaging technique that is comparable to optical projection tomography and computer tomography. Image quality is crucially dependent on matching the refractive indexes (RIs) of the sample and surrounding medium, but RI matching often requires some effort and is never perfect. Aim: Reducing the burden of RI matching between the immersion medium and sample in biomedical imaging is a challenging and interesting task. We aim at implementing a post processing strategy for correcting SLOT measurements that have errors caused by RI mismatch. Approach: To better understand the problems with poorly matched Ris, simulated SLOT measurements with imperfect RI matching of the sample and medium are performed and presented here. A method to correct distorted measurements was developed and is presented and evaluated. This method is then applied to a sample containing fluorescent polystyrene beads and a sample made of olydimethylsiloxane with embedded fluorescent nanoparticles. Results: From the simulations, it is evident that measurements with an RI mismatch larger than 0.02 and no correction yield considerably worse results compared to perfectly matched measurements. RI mismatches larger than 0.05 make it almost impossible to resolve finer details and structures. By contrast, the simulations imply that a measurement with an RI mismatch of up to 0.1 can still yield reasonable results if the presented correction method is applied. The experiments validate the simulated results for an RI mismatch of about 0.09. Conclusions: The method significantly improves the SLOT image quality for samples with imperfectly matched Ris. Although the absolutely best imaging quality will be achieved with perfect RI matching, these results pave the way for imaging in SLOT with RI mismatches while maintaining high image quality. The remarkable damage-tolerance of enamel has been attributed to its hierarchical microstructure and the organized bands of decussated rods. A thorough characterization of the microscale rod evolution within the enamel is needed to elucidate this complex structure. While prior efforts in this area have made use of single particle tracking to track a single rod evolution to various degrees of success, such a process can be both computationally and labor intensive, limited to the evolution path of a single rod, and is therefore prone to error from potentially tracking outliers. Particle image velocimetry (PIV) is a well-established algorithm to derive field information from image sequences for processes that are time-dependent, such as fluid flows and structural deformation. In this work, we demonstrate the use of PIV in extracting the full-field microstructural distribution of rods within the enamel. Enamel samples from a wild African lion were analyzed using high-energy synchrotron X-ray micro-tomography. Results from the PIV analysis provide sufficient full-field information to reconstruct the growth of individual rods that can potentially enable rapid analysis of complex microstructures from high resolution synchrotron datasets. Such information can serve as a template for designing damage-tolerant bioinspired structures for advanced manufacturing. Thorough characterization and analysis of biological microstructures (viz. dental enamel) allows us to understand the basis of their excellent mechanical properties. Prior efforts have successfully replicated these microstructures via single particle tracking, but the process is computationally and labor intensive. In this work, optical flow imaging algorithms were used to extract full-field microstructural distribution of enamel rods from synchrotron X-ray computed tomography datasets, and a field method was used to reconstruct the growth of individual rods. Such high throughput information allows for the rapid production/prototyping and advanced manufacturing of damage-tolerant bioinspired structures for specific engineering applications. Upcoming heliophysics missions utilize state-of-the-art wide field-of-view (FOV) imaging technology to measure and investigate the space plasma environment on a global scale. At Earth, remote sensing of soft X-ray emissions, which are generated via the charge exchange interaction between heavy solar wind ions and exospheric neutral atoms, is a promising means to investigate the global magnetosheath structure, its response to varying solar wind conditions, and the spatiotemporal properties of the dayside magnetic reconnection. Data analysis techniques such as optical tomography can provide additional structural and time-varying information from the observed target and thus enhance the mission's scientific return. In this work, we simulate multiple and simultaneous observations of the dayside magnetosphere using soft X-ray imagers located at long-distance vantage points to reconstruct the time-dependent, three-dimensional (3-D) structure of the magnetosheath using a dynamic tomographic approach. The OpenGCCM MHD model is used to simulate the time-varying response of the magnetosheath to solar wind conditions and, subsequently, generate synthetic soft X-ray images from multiple spacecraft vantage points separated along a common orbit. A detailed analysis is then performed to identify the nominal set of spacecraft that produces the highest fidelity tomographic reconstruction of the magnetopause. This work aims to (i) demonstrate, for the first time, the use of dynamic tomography to retrieve the time-varying magnetosheath structure and (ii) identify a nominal mission design for multi-spacecraft configurations aiming for optical tomography. Human studies of early brain development have been limited by extant neuroimaging methods. MRI scanners present logistical challenges for imaging young children, while alternative modalities like functional near‐infrared spectroscopy have traditionally been limited by image quality due to sparse sampling. In addition, conventional tasks for brain mapping elicit low task engagement, high head motion, and considerable participant attrition in pediatric populations. As a result, typical and atypical developmental trajectories of processes such as language acquisition remain understudied during sensitive periods over the first years of life. We evaluate high‐density diffuse optical tomography (HD‐DOT) imaging combined with movie stimuli for high resolution optical neuroimaging in awake children ranging from 1 to 7 years of age. We built an HD‐DOT system with design features geared towards enhancing both image quality and child comfort. Furthermore, we characterized a library of animated movie clips as a stimulus set for brain mapping and we optimized associated data analysis pipelines. Together, these tools could map cortical responses to movies and contained features such as speech in both adults and awake young children. This study lays the groundwork for future research to investigate response variability in larger pediatric samples and atypical trajectories of early brain development in clinical populations. Optical molecular tomography (OMT) is a potential pre-clinical molecular imaging technique with applications in a variety of biomedical areas, which can provide non-invasive quantitative three-dimensional (3D) information regarding tumor distribution in living animals. The construction of optical transmission models and the application of reconstruction algorithms in traditional model-based reconstruction processes have affected the reconstruction results, resulting in problems such as low accuracy, poor robustness, and long-time consumption. Here, a gates joint locally connected network (GLCN) method is proposed by establishing the mapping relationship between the inside source distribution and the photon density on surface directly, thus avoiding the extra time consumption caused by iteration and the reconstruction errors caused by model inaccuracy. Moreover, gates module was composed of the concatenation and multiplication operators of three different gates. It was embedded into the network aiming at remembering input surface photon density over a period and allowing the network to capture neurons connected to the true source selectively by controlling three different gates. To evaluate the performance of the proposed method, numerical simulations were conducted, whose results demonstrated good performance in terms of reconstruction positioning accuracy and robustness. Non-invasive techniques, such as close-range photogrammetry (CRP) and 3D ultrasonic tomography complemented with optical and scanning electron microscopy and mercury porosimetry, were applied to characterize the carbonate rock samples of the Calcari di Cagliari formation. The integrated approach started with the computation of high-resolution 3D models of the carbonate samples using the CRP technique to produce 3D high-resolution models texturized both with natural colors and intensity. Starting from the 3D models from previous techniques, a 3D ultrasonic tomography on each rock sample was accurately planned and carried out in order to detect the elastic properties of such rocks and relate them to textural heterogeneity or internal defects. The results indicate that the relationship between longitudinal velocity and rock properties is complex even in the same carbonate formation. Understanding the relationship between the geomatic and geophysical responses in the investigated rock properties, such as textural characteristics and especially structure and geometry of pores, type of pores, tortuosity and cementing material, is important for many practical applications and especially in the diagnostic process of the conservation state of monumental structures. The integration of the above non-invasive techniques complemented by petrographical–petrophysical data proved to be a powerful method to associate each lithotype with a different susceptibility to degradation. The results presented in this paper demonstrate that the proposed integrated use of complementary methodologies would guarantee the reproducibility of the measurements both at the laboratory and field scale for the monitoring in time of the rock condition while giving a useful contribution in making decisions on an appropriate remedial strategy. Background and Objective: This study compared the surgeon experience between conventional microscope-integrated intraoperative optical coherence tomography (iOCT) and digitally enabled microscope-integrated iOCT in vitreoretinal surgery. Patients and Methods: This is a post hoc case-control analysis of the DISCOVER study. Conventional microscope-integrated iOCT (Rescan 700, Zeiss) was compared with digitally enabled iOCT (Artevo 800, Zeiss). Compared variables included surgical field–based visualization (ie, ocular heads-up display in the conventional group; three-dimensional screen-based visualization in the digital iOCT group) and non-surgical field–based visualization (ie, review on the external two-dimensional monitor). Results: A total of 200 patients were included. Surgical field–based visualization of iOCT was significantly higher in the digitally enabled group (P < 0.0001). Required endoillumination level was significantly lower in the digital iOCT group (P < 0.0001). Surgeons reported "significant" back discomfort and headache more frequently when using conventional iOCT (P = 0.003 and P = 0.001, respectively). Conclusions: Digitally enabled iOCT resulted in greater surgical visualization efficiency, appeared to require a lower illumination level, and may provide advantages for ergonomic-related discomfort. Optical diffraction tomography can be performed with low phototoxicity and photobleaching to analyze 3D cells and tissues. It is desired to develop high throughput and powerful data processing capabilities. We propose high bandwidth holographic microscopy (HBHM). Based on the analyticity of complex amplitudes, the unified holographic multiplexing transfer function is established. A high bandwidth scattering field is achieved via the k-space optical origami of two 2D wavefronts from one interferogram. Scanning illumination modulates the high-horizontal and axial k-space to endow synthetic-aperture from 2D high space-bandwidth product (SBP) scattering fields. The bright-field counterpart SBP of a single scattering field from HBHM is 14.6 megapixels, while the number of pixels is only 13.7 megapixels. It achieves an eight-fold SBP enhancement under the same number of pixels and diffraction limit. Introduction: Optical Projection Tomography (OPT) and light sheet fluorescence microscopy (LSFM) are high resolution optical imaging techniques, ideally suited for ex vivo 3D whole mouse brain imaging. Although they exhibit high specificity for their targets, the anatomical detail provided by tissue autofluorescence remains limited. Methods: T1-weighted images were acquired from 19 BABB or DBE cleared brains to create an MR template using serial longitudinal registration. Afterwards, fluorescent OPT and LSFM images were coregistered/normalized to the MR template to create fusion images. Results: Volumetric calculations revealed a significant difference between BABB and DBE cleared brains, leading to develop two optimized templates, with associated tissue priors and brain atlas, for BABB (OCUM) and DBE (iOCUM). By creating fusion images, we identified virus infected brain regions, mapped dopamine transporter and translocator protein expression, and traced innervation from the eye along the optic tract to the thalamus and superior colliculus using cholera toxin B. Fusion images allowed for precise anatomical identification of fluorescent signal in the detailed anatomical context provided by MR. Discussion: The possibility to anatomically map fluorescent signals on magnetic resonance (MR) images, widely used in clinical and preclinical neuroscience, would greatly benefit applications of optical imaging of mouse brain. Granular media constitute the most abundant form of solid matter on Earth and beyond. When external forces are applied to a granular medium, the forces are transmitted through it via chains of contacts among grains-force chains. Understanding the spatial structure and temporal evolution of force chains constitutes a fundamental goal of granular mechanics. Here, we introduce an experimental technique, interference optical projection tomography, to study force chains in threedimensional (3D) granular packs under triaxial shear loads and illustrate the technique with random assemblies of spheres and icosahedra. We find that, in response to an increasing vertical load, the pack of spheres forms intensifying vertical force chains, while the pack of icosahedra forms more interconnected force-chain networks. This provides microscopic insights into why particles with more angularity are more resistant to shear failure-the interconnected force-chain network is stronger (that is, more resilient to topological collapse) than the isolated force chains in round particles. The longer force chains with less branching in the pack of round particles are more likely to buckle, which leads to the macroscopic failure of the pack. This work paves the way for understanding the grain-scale underpinning of localized failure of 3D granular media, such as shear localization in landslides and stick-slip frictional motion in tectonic and induced earthquakes. Significance: Optical imaging is a non-invasive imaging technology that utilizes near-infrared light, allows for the image reconstruction of optical properties like diffuse and absorption coefficients within the tissue. A recent trend is to use signal processing techniques or new light sources and expanding its application. Aim: We aim to develop the reflective optical imaging using the chaotic correlation technology with chaotic laser and optimize the quality and spatial resolution of reflective optical imaging. Approach: Scattering medium was measured using reflective configuration in different inhomogeneous regions to evaluate the performance of the imaging system. The accuracy of the recovered optical properties was investigated. The reconstruction errors of absorption coefficients and geometric centers were analyzed, and the feature metrics of the reconstructed images were evaluated. Results: We showed how chaotic correlation technology can be utilized for information extraction and image reconstruction. This means that a higher signal-tonoise ratio and image reconstruction of inhomogeneous phantoms under different scenarios successfully were achieved. Conclusions: This work highlights that the peak values of correlation of chaotic exhibit smaller reconstruction error and better reconstruction performance in optical imaging compared with reflective optical imaging with the continuous wave laser. In this study, a set of 316 L stainless steel test specimens was additively manufactured by laser-based Powder Bed Fusion. The process parameters were varied for each specimen in terms of laser scan speed and laser power. The objective was to use a narrow band of parameters well inside the process window, demonstrating detailed parameter engineering for specialized additive manufacturing cases. The process variation was monitored using Optical Tomography to capture light emissions from the layer surfaces. Process emission values were stored in a statistical form. Micrographs were prepared and analyzed for defects using optical microscopy and image manipulation. The results of two data sources were compared to find correlations between lack of fusion, porosity, and layer-based energy emissions. A data comparison of Optical Tomography data and micrograph analyses shows that Optical Tomography can partially be used independently to develop new process parameters. The data show that the number of critical defects increases when the average Optical Tomography grey value passes a certain threshold. High spatial resolution imaging over the entire volume for intravital imaging of biological specimens has long been vital. However, the depth‐of‐field (DOF) and spatial resolution are intrinsically interdependent. Here, a new extended DOF (EDOF) microscopy technique based on partially coherent annular illumination is proposed, termed AI‐EDOF, by combining an electrically tunable lens with a conventional microscope with modified illumination, for motion‐free, high spatial resolution real‐time all‐in‐focus imaging over thick volumes. EDOF imaging is obtained with spatial resolution up to near the incoherent diffraction limit (∼$\sim $388 nm, 20×$\times$/ 0.8NA) and temporal resolution ∼$\sim $30 fps. Moreover, it is demonstrated that the coupled phase and absorption components of the complex refractive index in optical diffraction tomography microscopy can be solved by focus scanning. The Richardson‐Lucy deconvolution with total variation regularization is adopted for deblurring and suppressing noise under low light efficiency high‐speed exposure. To demonstrate the capabilities of the proposed method, experiments are conducted using fixed transgenic zebrafish larvae, Drosophila larvae and dynamic Caenorhabditis elegans under both transmissive and fluorescent imaging modalities, revealing the proposed approach is prospective to be adopted by broad applications such as pharmacokinetics and tumor immunology. Purpose: To determine the role of Optical Coherence Tomography Angiography (OCTA) in predicting risk of progression of diabetic retinopathy. Study Design: Descriptive observational study. Place and Duration of Study: Jinnah Post graduate Medical Center, Karachi, from June 2022 to June 2023. Methods: Patients with type 1 or 2 diabetes were included. Base line investigations were done including Central subfield thickness and OCTA at first visit. Second visit was conducted at 6th month and patients were followed for 24 months. OCT images of poor quality, motion artifact, inaccurate partition of tissue layers and blurry images were excluded. Results: Among 97 enrolled patients, 88 cases finished the complete 24-months follow up. Out of 88 patients, 16 (18.18%) patients showed progression of diabetic retinopathy (DR), 9 (10.22%) patients developed DME, two showed both DR and DME. In univariate analysis, greater FAZ area, reduced FAZ circularity, Fractal dimension (FD) and vessel density (VD) of the deep capillary plexus (DCP) were significantly related to the advancement in DR. Among these, FAZ area, VD, and FD remained significant in the multivariate analysis. Whereas, only decreased VD and FD of superficial capillary plexus (SCP) were significantly related to advancement in DR and DME. Conclusion: Changes in OCTA parameters is significantly associated with progression of diabetic ocular complications including diabetic retinopathy and diabetic macular edema. Significance: Accurate spatial registration of probes (e.g., optodes and electrodes) for measurement of brain activity is a crucial aspect in many neuroimaging modalities. It may increase measurement precision and enable the transition from channel-based calculations to volumetric representations. Aim: This technical note evaluates the efficacy of a commercially available infrared three-dimensional (3D) scanner under actual experimental (or clinical) conditions and provides guidelines for its use. Method: We registered probe positions using an infrared 3D scanner and validated them against magnetic resonance imaging (MRI) scans on five volunteer participants. Results: Our analysis showed that with standard cap fixation, the average Euclidean distance of probe position among subjects could reach up to 43 mm, with an average distance of 15.25 mm [standard deviation (SD) = 8.0]. By contrast, the average distance between the infrared 3D scanner and the MRI-acquired positions was 5.69 mm (SD = 1.73), while the average difference between consecutive infrared 3D scans was 3.43 mm (SD = 1.62). The inter-optode distance, which was fixed at 30 mm, was measured as 29.28 mm (SD = 1.12) on the MRI and 29.43 mm (SD = 1.96) on infrared 3D scans. Our results demonstrate the high accuracy and reproducibility of the proposed spatial registration method, making it suitable for both functional near-infrared spectroscopy and electroencephalogram studies. Conclusions: The 3D infrared scanning technique for spatial registration of probes provides economic efficiency, simplicity, practicality, repeatability, and high accuracy, with potential benefits for a range of neuroimaging applications. Label-free thermometry is a pivotal tool for many disciplines. However, most current approaches are only suitable for planar heat sources in steady state, thereby restricting the range of systems that can be reliably studied. Here, we introduce pump probe-based optical diffraction tomography (ODT) as a method to map temperature precisely and accurately in three dimensions (3D) at the single-particle level. To do so, we first systematically characterize the thermal landscape in a model system consisting of gold nanorods in a microchamber and then benchmark the results against simulations and quantitative phase imaging thermometry. We then apply ODT thermometry to resolve thermal landscapes inaccessible to other label-free approaches in the form of nonplanar heat sources embedded in complex environments and freely diffusing gold nanorods in a microchamber. Last, we foresee that our approach will find many applications where routine thermal characterization of heterogeneous nanoparticles samples in 3D or in non-steady state is required. A technique for the design of a hybrid tooth and bone-supported implant drilling guide is described. The patient was scanned using cone beam computed tomography and an optical intraoral scanner. The dicom file was segmented according to the area of interest composed of bone and the remaining natural teeth. The Standard Tessellation Language (STL) file was trimmed to only the teeth providing support, followed by merging between the bone and teeth files in one STL. The implant drilling guide was designed with the Real Guide software program, and the file was 3-dimensionally printed in clear surgical guide resin. This technique offers an accurate, cost-effective digitally designed implant placement guide for patients with long-span distal extension edentulous areas and few remaining natural dentitions providing distal bone support. It can also be used in patients with hemi maxillectomy for zygomatic implant placement. Quantum optical technologies promise advances in sensing, computing, and communication. A key resource is squeezed light, where quantum noise is redistributed between optical quadratures. We introduce a monolithic, chip-scale platform that exploits the (2) nonlinearity of a thin-film lithium niobate (TFLN) resonator device to efficiently generate squeezed states of light. Our system integrates all essential components--except for the laser and two detectors--on a single chip with an area of one square centimeter, reducing the size, operational complexity, and power consumption associated with conventional setups. Using the balanced homodyne measurement subsystem that we implemented on the same chip, we measure a squeezing of 0.55 decibels and an anti-squeezing of 1.55 decibels. We use 20 milliwatts of input power to generate the parametric oscillator pump field by using second harmonic generation on the same chip. Dynamic contrast optical coherence tomography (DyC-OCT), an emerging imaging method, utilizes fluctuation patterns in OCT signals to enhance contrast, thereby enabling non-invasive label-free volumetric live cell imaging. In this mini review, we explain the core concepts behind DyC-OCT image formation and its system configurations, serving as practical guidance for future DyC-OCT users. Subsequently, we explore its applications in delivering high-quality, contrast-enhanced images of cellular morphology, as well as in monitoring changes in cellular activity/viability assay experiments. Significance: Diabetes can lead to the glycation of proteins and dysfunction of skin collagen. Skin lesions are a prevalent clinical symptom of diabetes mellitus (DM). Early diagnosis and assessing the efficacy of treatment for DM are crucial for patient health management. However, performing a non-invasive skin assessment in the early stages of DM is challenging. Aim: By using the polarization-sensitive optical coherent tomography (PS-OCT) imaging technique, it is possible to noninvasively assess the skin changes caused by diabetes. Approach: The PS-OCT was used to monitor the polarization characteristics of mouse skin at different stages of diabetes. Results: Based on a multi-layered adhesive tape model, we found that the polarization characteristics (retardation, optic axis, and polarization uniformity) were sensitive to the microstructure changes in the samples. Through this method, we observed significant changes in the polarization states of the skin as diabetes progressed. This was in line with the detected microstructure changes in skin collagen fibers using scanning electron microscopy. The method of common lines is a well-established reconstruction technique in cryogenic electron microscopy (cryo-EM), which can be used to extract the relative orientations of an object given tomographic projection images from different directions. In this paper, we deal with an analogous problem in optical diffraction tomography. Based on the Fourier diffraction theorem, we show that rigid motions of the object, i.e., rotations and translations, can be determined by detecting common circles in the Fourier-transformed data. We introduce two methods to identify common circles. The first one is motivated by the common line approach for projection images and detects the relative orientation by parameterizing the common circles in the two images. The second one assumes a smooth motion over time and calculates the angular velocity of the rotational motion via an infinitesimal version of the common circle method. Interestingly, using the stereographic projection, both methods can be reformulated as common line methods, but these lines are, in contrast to those used in cryo-EM, not confined to pass through the origin and allow for a full reconstruction of the relative orientations. The present in vitro study focuses on the filling ability of three different bioceramic cements with or without the addition of a bioceramic sealer in an open apex model on the marginal apical adaptation, tubule infiltrations, and void distributions as well as the interface between the cement and the sealer materials. To this end, sixty mandibular premolars were used. MTA-Biorep (BR), Biodentine (BD), and Well-Root Putty (WR) were used to obturate the open apex model with or without the addition of a bioceramic sealer, namely TotalFill® BC sealer™ (TF). A digital optical microscope and scanning electron microscope (SEM) were used to investigate the cement–dentin interface, marginal apical adaptation, and the material infiltration into the dentinal tubules. Micro-computed X-ray tomography and digital optical microscopy were used to investigate the cement–sealer interface. The results were analyzed by using the Kruskal–Wallis test. No significant difference was found between the groups for the marginal apical adaptation quality (p > 0.05). Good adaptation of the dentin–cement interface was found for all tested groups and the sealer was placed between the cement material and dentinal walls. All the groups demonstrated some infiltrations into the dentinal tubules at the coronal part except for the BR group. A good internal interface was found between the cement and the sealer with the presence of voids at the external interface. A larger number of voids were found in the case of the BD-TF group compared to each of the other two groups (p < 0.05). Within the limitations of the present in vitro study, all the groups demonstrated good marginal apical adaptation. The use of a sealer in an open apex does not guarantee good filling and, in addition, creates voids at the external interfaces with the dental walls when the premixed sealer is used with powder–liquid cement systems. The use of a premixed bioceramic cement could offer fewer complications than when a powder–liquid cement system is used. [ABSTRACT FROM AUTHOR]

This paper proposes an experimental investigation of a commercial photopolymer resin followed by material modelling and manufacturing system characterization. We focus on the effect of the degree of cure and temperature on the material properties of the photopolymer materials. UV curing properties of the liquid resin are assessed with the thickness measurement by optical tomography. Besides, the specific heat capacity is determined for the almost completely cured and uncured samples with DSC measurements. Photo-DSC experiments are performed to investigate the curing reaction and modelling of the evolution of the degree of cure depending on the light intensity and temperature. In addition, chemical shrinkage behaviour is captured as function of the degree of cure by the high-precision balance set-up. Functional neuroimaging studies of neuropsychiatric disorders and cognitive impairment are commonly conducted in the clinic setting but less so in the acutely medically ill while hospitalized. This is largely due to technical and logistical limitations, given the lack of portable devices with high spatial and temporal resolutions. This exploratory study reports on the development and implementation of a novel diffuse optical tomography (DOT) system that can be employed for bedside three-dimensional functional neuroimaging. To test this portable DOT system, our protocol included a task-based sequence involving the Months Backwards Test with imaging centered on the bilateral prefrontal cortex. Fifteen subjects were recruited from intensive care units and the general wards of a single tertiary academic hospital and included in our final analysis. Volumetric hemoglobin analyses of the dorsolateral prefrontal cortex (DLPFC) and dorsomedial prefrontal cortex (DMPFC) were reliably captured in all our subjects. The peak value was calculated to be 3.36 µM and 0.74 µM for oxygenated-hemoglobin (HbO) and total-hemoglobin (HbT) (p < 0.042, [HbT]), respectively. The standard error was calculated to be 4.58 uM and 3.68 uM for (HbO) and (HbT). We additionally developed a seed-based correlation analysis to demonstrate the capability of DOT in studying functional connectivity. The right DLPFC was found to be moderately associated with the left DLPFC in all our subjects (r = 0.656). The DMPFC was observed to be associated with the left DLPFC but less so (r = 0.273) at the group level. Overall, the contribution of left-to-right DLPFC connectivity was significantly higher than left DLPFC to DMPFC in our group (p = 0.012). Future studies should investigate the potential of such a DOT system in the research of neuropsychiatric and neurocognitive disorders within the hospital to study different types of mechanisms, pathophysiology, and interventions that occur acutely and can advance our knowledge of these disorders. Recent studies in transillumination imaging for developing an optical computed tomography device for small animal and human body parts have used deep learning networks to suppress the scattering effect, estimate depth information of light-absorbing structures, and reconstruct three-dimensional images of de-blurred structures. However, they still have limitations, such as knowing the information of the structure in advance, only processing simple structures, limited effectiveness for structures with a depth of about 15 mm, and the need to use separated deep learning networks for de-blurring and estimating information. Furthermore, the current technique cannot handle multiple structures distributed at different depths next to each other in the same image. To overcome the mentioned limitations in transillumination imaging, this study proposed a pixel-by-pixel scanning technique in combination with deep learning networks (Attention Res-UNet for scattering suppression and DenseNet-169 for depth estimation) to estimate the existence of each pixel and the relative structural depth information. The efficacy of the proposed method was evaluated through experiments that involved a complex model within a tissue-equivalent phantom and a mouse, achieving a reconstruction error of 2.18 % compared to the dimensions of the ground truth when using the fully convolutional network. Furthermore, we could use the depth matrix obtained from the convolutional neural network (DenseNet-169) to reconstruct the absorbing structures using a binary thresholding method, which produced a reconstruction error of 6.82 %. Therefore, only one convolutional neural network (DenseNet-169) must be used for depth estimation and explicit image reconstruction. Therefore, it reduces time and computational resources. With depth information at each pixel, reconstruction of 3D image of the de-blurred structures could be performed even from a single blurred image. These results confirm the feasibility and robustness of the proposed pixel-by-pixel scanning technique to restore the internal structure of the body, including intricate networks such as blood vessels or abnormal tissues. During orthodontic treatment, the early diagnosis of microscopic changes in soft and hard tissues, including periodontal tissue, is very important to prevent iatrogenic side effects like root resorption and periodontal diseases. Cervical periodontal tissue is the most critical area that reacts first to mal-habits or orthodontic forces, and it is also the place where bacteria deposits in the early stage of periodontal diseases. The early diagnosis of hard tissue changes, such as demineralization, is also very important in maintaining a patient's health during orthodontic treatment. Many diagnostic devices, including radiographic equipment and intra-oral scanners, are helpful in diagnosing these problems, but have certain limitations in invasiveness and precision. The purpose of this study is to verify the possible utilities of non-invasive diagnostic devices in the orthodontic field that can compensate for these limitations. For this, non-invasive optical diagnostic devices, including optical coherence tomography and optical Doppler tomography, were used in vivo with animal and human examination for hard and soft tissues. These devices can provide real-time three-dimensional images at the histological scale. The results of this study verified these devices can be used in clinical practice during orthodontic treatment and introduced a new diagnostic paradigm differentiating microstructural changes in tissues in orthodontic diagnosis.
