The article deals with teaching Russian intonation of declarative sentences to foreign students. The emphasis is placed on the way teaching materials are presented. In particular, the variable rows for intonation patterns in declarative sentences have been developed, as well as the teaching of syntagmatic segmentation and intonation patterns in complex utterances. This method of working with foreigners is efficient for teaching foreign students the Russian declarative intonation which manifests in verbal communication and reading.

The ribonuclease III enzyme Dicer is essential for the processing of micro-RNAs (miRNAs) and small interfering RNAs (siRNAs) from double-stranded RNA precursors. miRNAs and siRNAs regulate chromatin structure, gene transcription, mRNA stability, and translation in a wide range of organisms. To provide a model system to explore the role of Dicer-generated RNAs in the differentiation of mammalian cells in vivo, we have generated a conditional Dicer allele. Deletion of Dicer at an early stage of T cell development compromised the survival of alphabeta lineage cells, whereas the numbers of gammadelta-expressing thymocytes were not affected. In developing thymocytes, Dicer was not required for the maintenance of transcriptional silencing at pericentromeric satellite sequences (constitutive heterochromatin), the maintenance of DNA methylation and X chromosome inactivation in female cells (facultative heterochromatin), and the stable shutdown of a developmentally regulated gene (developmentally regulated gene silencing). Most remarkably, given that one third of mammalian mRNAs are putative miRNA targets, Dicer seems to be dispensable for CD4/8 lineage commitment, a process in which epigenetic regulation of lineage choice has been well documented. Thus, although Dicer seems to be critical for the development of the early embryo, it may have limited impact on the implementation of some lineage-specific gene expression programs.

The paper reveals the future teachers’ attitude towards freedom and responsibility in the context of sustainable development of education. Sustainable development seems to be impossible without the new generation of free, dignified, respectful, and responsible individuals. Freedom and responsibility are outlined from a pedagogical point of view. The concepts under study are considered to be of great significance for child’s development. The information obtained during the survey and questionnaire conducted with first-year students of the pedagogical university indicates that future teachers do not yet have sufficient experience to correlate adequately the possibility of revealing school students’ freedom and the need to be in charge of the behavior, actions and thoughts of their own. University students believe that school students generally take responsibility for completing individual assignments rather than common ones. Future teachers do not think that school students’ attitude towards education is responsible and trustworthy, would-be teachers doubt that school students are able to manifest freedom and responsibility to full extent. The empirical data drive to the conclusion that it is necessary to change future teachers’ attitude towards freedom and responsibility in their pedagogical activities. Future teachers are advised to change their attitude to freedom and responsibility in their activities.

Age-related changes in human cardiomyocytes are closely related to cardiac diseases, especially atrial fibrillation. Restricted availability of biological preparations from the human atrial myocardium complicates experimental studies on the aging processes in cardiomyocytes. In this preliminary study, we used available experimental data on the age-related changes in ionic conductances in canine atrial cardiomyocytes to predict possible consequences of similar remodeling in humans using two mathematical models (Courtemanche98 and Maleckar09) of human atrial cardiomyocytes. The study was performed using the model population approach, allowing one to assess variability in the cellular response to different interventions affecting model parameters. Here, this approach was used to evaluate the effects of age-related parameter modulation on action potential biomarkers in the two models. Simulation results show a significant decrease in the action potential duration and membrane potential at 20% of the action potential duration in aging. These model predictions are consistent with experimental data from mammalians. The action potential characteristics are shown to serve as notable biomarkers of age-related electrophysiological remodeling in human atrial cardiomyocytes. A comparison of the two models shows different behavior in the prediction of repolarization abnormalities.

Electrophysiology of cardiomyocytes changes with aging. Agerelated ionic remodeling in cardiomyocytes may increase the incidence and prevalence of atrial fibrillation (AF) in the elderly and affect the efficiency of antiarrhythmic drugs. There is the deep lack of experimental data on an action potential and transmembrane currents recorded in the healthy human cardiomyocytes of different age. Experimental data in mammals is also incomplete and often contradicting depending on the experimental conditions. In this in-silico study, we used a population of ionic models of human atrial cardiomyocytes to transfer data on the age- related ionic remodeling in atrial cardiomyocytes from canines and mice to predict possible consequences for human cardiomyocyte activity. Based on experimental data, we analyzes two hypotheses on the aging effect on the ionic currents using two age-related sets of varied model parameters and evaluated corresponding changes in action potential morphology with aging. Using the two populations of aging models, we analyzed the agedependent sensitivity of atrial cardiomyocytes to Dofetilide which is one of the antiarrhythmic drugs widely used in patients with atrial fibrillation.

The article discusses the possibility of the development of axiological competences in university students in the process of teaching language-related disciplines within the world educational space. The problem of the development of communicative competences is relevant in the modern educational environment; it determines the purpose of the study: to analyze the survey results illustrating the attitude of students to the value of communication in society. During the research, linguistic and axiological parameters, defined by students as linguistic and communicative values, were identified, which makes it possible to talk about modern trends in education. Particular attention is paid to the value categories of the Russian language as the basis for national and cultural orientation, successful communication, intellectual development of people and formation of their linguistic personalities. The authors regard the axiological approach in the development of the student’s communicative competences as a fundamentally structured vision of the world through the prism of value orientation, which in the practice of cognition allows developing competences based on the cognitive needs and communicative intentions of students.

In the paper the concept MONEY is considered to be one in a special class of concepts which we propose to call prothteconomic concepts, as they combine prothtologic (everyday) and academic (economic) elements. The prothtologic element of the concept reveals itself in the common national collective conscience, while the academic component manifests itself in the scientific conscience. The vocabulary used by the common national collective conscience describes the concept under study with different semantic attributes. Proverbs as parameological units fully illustrate the way the common national collective conscience expresses ideas and phenomena. The scientific conscience deals with terminological word stock. The comparison of the prothtologic components of the concept with the economic ones finds out coincidence in the plane of contents. As regards the plane of expression, it differs in a considerable number of instances. So the common national collective conscience has worked out its folk economics which has much in common with scientific economics and is stored in proverbs.

Developmentally regulated changes in stability of Xist RNA can be attributed to a promoter switch prompting stable and unstable RNA isoforms. Stable Xist RNA comes from the previously identified promoter or from a unique promoter situated 1.5 kb further downstream, while unstable Xist RNA comes from a second novel promoter situated 6.5 kb upstream. It has been shown that promoter switching controls Xist RNA stability at the onset of random and imprinted X inactivation. It has also been shown that imprinted Xist expression takes place as a result of activation of P1/P2 on the Xp allele.

Modulation of Xist RNA was studied to identify its effects on the developmental up-regulation of the X transcript. Results show that up-regulation of Xist RNA marks the beginning of X inactivation due to the increased stability of the transcript it has caused. However, an unstable transcript is caused by the inhibition of the Xist allele such as in the XX ES cells and in XX embryos which also occurs at the onset of X inactivation.

Abstract Background Expression of Xist, the master regulator of X chromosome inactivation, is extinguished in pluripotent cells, a process that has been linked to programmed X chromosome reactivation. The key pluripotency transcription factors Nanog, Oct4 and Sox2 are implicated in Xist gene extinction, at least in part through binding to an element located in Xist intron 1. Other pathways, notably repression by the antisense RNA Tsix, may also be involved. Results Here we employ a transgene strategy to test the role of the intron 1 element and Tsix in repressing Xist in ES cells. We find that deletion of the intron 1 element causes a small increase in Xist expression and that simultaneous deletion of the antisense regulator Tsix enhances this effect. Conclusion We conclude that Tsix and pluripotency factors act synergistically to repress Xist in undifferentiated embryonic stem cells. Double mutants do not exhibit maximal levels of Xist expression, indicating that other pathways also play a role.

Abstract Background X chromosome inactivation, the mechanism used by mammals to equalise dosage of X-linked genes in XX females relative to XY males, is triggered by chromosome-wide localisation of a cis-acting non-coding RNA, Xist. The mechanism of Xist RNA spreading and Xist-dependent silencing is poorly understood. A large body of evidence indicates that silencing is more efficient on the X chromosome than on autosomes, leading to the idea that the X chromosome has acquired sequences that facilitate propagation of silencing. LINE-1 (L1) repeats are relatively enriched on the X chromosome and have been proposed as candidates for these sequences. To determine the requirements for efficient silencing we have analysed the relationship of chromosome features, including L1 repeats, and the extent of silencing in cell lines carrying inducible Xist transgenes located on one of three different autosomes. Results Our results show that the organisation of the chromosome into large gene-rich and L1-rich domains is a key determinant of silencing efficiency. Specifically genes located in large gene-rich domains with low L1 density are relatively resistant to Xist-mediated silencing whereas genes located in gene-poor domains with high L1 density are silenced more efficiently. These effects are observed shortly after induction of Xist RNA expression, suggesting that chromosomal domain organisation influences establishment rather than long-term maintenance of silencing. The X chromosome and some autosomes have only small gene-rich L1-depleted domains and we suggest that this could confer the capacity for relatively efficient chromosome-wide silencing. Conclusions This study provides insight into the requirements for efficient Xist mediated silencing and specifically identifies organisation of the chromosome into gene-rich L1-depleted and gene-poor L1-dense domains as a major influence on the ability of Xist-mediated silencing to be propagated in a continuous manner in cis.
