Plastic pollution is now considered globally ubiquitous, irreversible, and a planetary boundary threat. Solutions are urgently needed but their development and application are hampered by the complexity and scale of the issue. System dynamics is a technique used to understand complex behaviours of systems through model building and is useful for conceptualising the relationships between various interacting, dynamic factors, and identifying potential intervention points within the system where specific policies or innovations might have the greatest impact or meet with the greatest resistance. Here, twenty-five participants (all scientific researchers of various career stages, disciplines and nationalities working on plastic pollution) completed a series of exercises through an interactive, iterative group model building exercise during a one-day workshop. The process culminated in the generation of a causal loop diagram, based on participants' perspectives, illustrating the dynamic factors relating to the constraints and enablers of solutions to plastic pollution. A total of 18 factors and seven feedback loops were identified. Key factors influencing the system were Effective legislation, Funding, Public education and awareness, Behaviour change, Innovation, and Effective waste management. Although the twenty-first century has seen major advances in evidence-based medicine to improve health, athletic performance, and injury prevention, our inability to implement these best practices across underserved American communities has limited the impact of these breakthroughs in sports medicine. Rochester, NY is stereotypical of American communities in which an economically challenged racially diverse urban center with grossly underperforming public schools is surrounded by adequately resourced predominantly Caucasian state-of-the-art education systems. Large filter-feeding animals are potential sentinels for understanding the extent of microplastic pollution, as their mode of foraging and prey mean they are continuously sampling the environment. However, there is considerable uncertainty about the total and mode of exposure (environmental vs trophic). Here, we explore microplastic exposure and ingestion by baleen whales feeding year-round in coastal Auckland waters, New Zealand. Plastic and DNA were extracted concurrently from whale scat, with 32 ± 24 (mean ± SD, n = 21) microplastics per 6 g scat sample detected. Using a novel stochastic simulation modeling incorporating new and previously published DNA diet information, we extrapolate this to total microplastic exposure levels of 24,028 (95% CI: 2119, 69,270) microplastics per mouthful of prey, or 3,408,002 microplastics (95% CI: 295,810, 10,031,370) per day, substantially higher than previous estimates for large filter-feeding animals. Critically, we find that the total exposure is four orders of magnitude more than expected from microplastic measurements of local coastal surface waters. This suggests that trophic transfer, rather than environmental exposure, is the predominant mode of exposure of large filter feeders for microplastic pollution. Efforts to understand macroplastic pollution have primarily focused on coastal and marine environments to the exclusion of freshwater, terrestrial, and urban ecosystems. To better understand macroplastics in the environment and their sources, a dual approach examining plastic input and leakage can be used. In this study, litter aggregation pathways at 40 survey sites with varying ambient population counts in the Ganges River Basin were surveyed in pre- and postmonsoon seasons. We examine active litter leakage using transect surveys of on-the-ground items, in conjunction with assessments of single-use plastic consumer products at the point of sale. We find that sites with low populations have a significantly higher number of littered items per 1,000 people than those with mid to high populations. Over 75% of litter items were plastics or multimaterial items containing plastic, and tobacco products and plastic food wrappers were the most recorded items. There was no significant variation of litter densities pre- and postmonsoon. Most single-use plastic consumer products were manufactured in-country, but approximately 40% of brands were owned by international companies. Stratified sampling of active litter input and consumer products provides a rapid, replicable snapshot of plastic use and leakage.

Growing evidence suggests that anthropogenic litter, particularly plastic, represents a highly pervasive and persistent threat to global marine ecosystems. Multinational research is progressing to characterise its sources, distribution and abundance so that interventions aimed at reducing future inputs and clearing extant litter can be developed. Citizen science projects, whereby members of the public gather information, offer a low-cost method of collecting large volumes of data with considerable temporal and spatial coverage. Furthermore, such projects raise awareness of environmental issues and can lead to positive changes in behaviours and attitudes. We present data collected over a decade (2005-2014 inclusive) by Marine Conservation Society (MCS) volunteers during beach litter surveys carried along the British coastline, with the aim of increasing knowledge on the composition, spatial distribution and temporal trends of coastal debris. Unlike many citizen science projects, the MCS beach litter survey programme gathers information on the number of volunteers, duration of surveys and distances covered. This comprehensive information provides an opportunity to standardise data for variation in sampling effort among surveys, enhancing the value of outputs and robustness of findings. We found that plastic is the main constituent of anthropogenic litter on British beaches and the majority of traceable items originate from land-based sources, such as public littering. We identify the coast of the Western English Channel and Celtic Sea as experiencing the highest relative litter levels. Increasing trends over the 10-year time period were detected for a number of individual item categories, yet no statistically significant change in total (effort-corrected) litter was detected. We discuss the limitations of the dataset and make recommendations for future work. Proteome modifications downstream of monogenic or polygenic disorders have the potential to uncover novel molecular mechanisms participating in pathogenesis and/or extragenic modification of phenotypic expression. We tested this idea by determining the proteome sensitive to genetic defects in a locus encoding dysbindin, a protein required for synapse biology and implicated in schizophrenia risk. We applied quantitative mass spectrometry to identify proteins expressed in neuronal cells the abundance of which was altered after downregulation of the schizophrenia susceptibility factor dysbindin (Bloc1s8) or two other dysbindin-interacting polypeptides, which assemble into the octameric biogenesis of lysosome-related organelles complex 1 (BLOC-1). We found 491 proteins sensitive to dysbindin and BLOC-1 loss of function. Gene ontology of these 491 proteins singled out the actin cytoskeleton and the actin polymerization factor, the Arp2/3 complex, as top statistical molecular pathways contained within the BLOC-1-sensitive proteome. Subunits of the Arp2/3 complex were downregulated by BLOC-1 loss of function, thus affecting actin dynamics in early endosomes of BLOC-1-deficient cells. Furthermore, we demonstrated that Arp2/3, dysbindin, and subunits of the BLOC-1 complex biochemically and genetically interact, modulating Drosophila melanogaster synapse morphology and homeostatic synaptic plasticity. Our results indicate that ontologically prioritized proteomics identifies novel pathways that modify synaptic phenotypes associated with neurodevelopmental disorder gene defects., Significance Statement: The mechanisms associated with schizophrenia are mostly unknown despite the increasing number of genetic loci identified that increase disease risk. We present an experimental strategy that impartially and comprehensively interrogates the proteome of neurons to identify effects of genetic mutations in a schizophrenia risk factor, dysbindin. We find that the expression of the actin polymerization complex Arp2/3 is reduced in dysbindin-deficient cells, thus affecting actin-dependent phenotypes in two cellular compartments where dysbindin resides, endosomes and presynapses. Background: Measurement of selenium in serum is an important clinical biomarker of nutritional status. The presence of gadolinium (Gd) in samples following administration of the contrast agents used for magnetic resonance imaging (MRI) results in a significant positive bias when using quadrupole inductively coupled plasma mass spectrometry (Q-ICP-MS)., Methods: Three instrumental set-ups were assessed: standard mode with no collision gas and collision cell mode with either a hydrogen:helium mixture or hydrogen. The effect of Gd on the selenium (Se) signal was assessed using external quality assurance (EQA) specimens and internal quality control (IQC) materials, both unspiked and spiked with Gd. Serum previously shown to contain high concentrations of Gd-containing contrast agents were also analysed., Results: Recoveries of Se in the spiked compared to the unspiked samples were: between 500% and 1300% using standard mode; 100% and 29,000% using collision cell mode with hydrogen:helium mixture; and between 99% and 103% using hydrogen. The use of H2 in the collision cell provided accurate results, indicating that the charge exchange reaction (CER) of Gd(2+) with H2 removes this interference. Analysis of patient serum known to contain the Gd contrast agent using the method gave results within the selenium reference range (adults 0.89-1.65 µmol/L). The presence of Gd, as low as 0.2 mg/L, in serum samples causes a positive interference on the measurement of Se by ICP-MS., Conclusions: Using a CER mode with pure H2 in the collision cell it was possible to fully remove the interference due to Gd(2+) from the signal for Se.

Background: Manganese (Mn) toxicity has been reported in patients receiving total parenteral nutrition. To avoid unnecessary exposure it is recommended by NICE (National Institute for Clinical Excellence) that blood Mn concentrations are monitored. The aim of the study was to develop a method using inductively coupled plasma mass spectrometry (ICP-MS) for the reliable determination of Mn in plasma and whole blood, as indices of acute and chronic exposure., Methods: Whole blood and plasma samples were prepared by appropriate dilution (diluent containing 0.005% Triton X-100, 0.2% propan-2-ol, 0.2% butan-1-ol and 1% nitric acid) addition of an internal standard gallium, followed by centrifugation to remove cell debris. Thermo Fisher Scientific ExCell and X Series ICP-MS instruments were used to define and correct for polyatomic interference on Mn assay., Results: Mn was quantified at mass 55 using aqueous calibration and the polyatomic interference from FeH was successfully eliminated by modified (Xt) skimmer cones but not with the collision cell (collision gas 7% H2 in He, flow rate 4-7 mL/min). The assay was validated showing good precision, limit of detection and percentage recovery. Good agreement was observed with the All Laboratory Trimmed Mean of External Quality Assurance samples y (in house)=1.1 (ALTM)-45.0 between values of 250 and 750 nmol/L., Conclusions: A method has been developed using ICP-MS for the analysis of whole blood and plasma Mn incorporating a novel method of eliminating interference by utilizing the different geometries of the Xt interface cones. The procedure is simple and robust with good precision and recovery over a wide dynamic range.

Selenium is an important clinical biomarker of nutritional status; however, the occurrence of gadolinium in a patient's serum as a result of the contrast agents used during magnetic resonance imaging investigations, results in a significant positive bias in its measurement by inductively coupled plasma-quadrupole mass spectrometry.

We evaluated the medical records of 17 dogs with hyphema of presumed retinal origin to evaluate the clinical, laboratory, ultrasonographic, and histologic features as well as known complications. The mean age of the dogs was 11.5 years. Routine hematologic and biochemical evaluation failed to identify an underlying cause in any dog. Retinal detachments, however, were identified in 10 of 13 dogs evaluated by ultrasonography and 5 of 6 globes evaluated histologically. In 1 dog, hyphema was associated with retinal vascular disease, presumed to be caused by hypertension. The prognosis for vision in geriatric dogs with hyphema, secondary to retinal disease, was found to be grave, as 10 dogs developed secondary glaucoma. The outcome for all dogs was loss of vision.

Ulcerative keratitis is one of the most common vision-threatening diseases of the horse. Successful therapy, however, can be achieved in the majority of cases provided the diagnosis is made early and the treatment is specific for the disease. This article reviews the pathophysiology, clinical signs, and contemporary medical and surgical approaches to bacterial and fungal ulcerative keratitis in the horse.
