This article discusses the revision of the chemical structures of a group of bacterial sulfur-containing aminolipids called cysteinolides. The study used various analytical techniques to determine the absolute stereochemistry of these lipids and found that they have an unusual N,O-acylated cysteic acid head group. The research also mapped the distribution and structural diversity of cysteinolides in different bacteria. The article also discusses the synthesis and structural analysis of sulfonolipids, the purification and analysis of lipid extracts, and the evaluation of the bioactivity of aminosulfonolipids. Summary The significant cultural achievement of the development of the cultivation of Coca (Erythroxylum coca) by the indigenous South Americans was never adequately recognised by the European Conquerors. The aim of this paper is to demonstrate its importance for Western science by summarising its perception during the colonial period and the 19th century and reviewing the isolation of cocaine, its use as local anaesthetic, determination of its chemical structure and synthesis of substitutes as milestones of Western chemistry, pharmacy and medicine. Wir präsentieren die erste Totalsynthese der Thiamyxine A–C und des jetzt vollständig charakterisierten Thiamyxins E, einer interessanten Klasse thiazol‐ und thiazolinreicher Depsipeptide mit vielfältiger antiviraler Aktivität. Die Synthese zeichnet sich aus durch einen parallelen Ringschluss zu zwei Methylthiazolin‐Einheiten unter geringer Epimerisierung des sehr labilen benachbarten stereogenen Zentrums. Des Weiteren wird die dreistufige Synthese einer ungewöhnlichen Hydroxysäure sowie die oxidationsfreie Eliminierung eines Phenylselenids zum Dehydroalanin gezeigt. Die Ausnutzung des säurelabilen Stereozentrums am Isoleucin‐Rest und die erneute Öffnung der Makrolactone ermöglichen den Zugang zu den vier Thiamyxinen in guten Ausbeuten und Diastereomerenverhältnissen aus einem einzigen Vorläufer. In diesem Bericht beschreiben wir eine milde, effiziente und epimerisierungsfreie Methode für die Synthese 2‐thiazolin‐ und 5,6‐dihydro‐4H‐1,3‐thiazinhaltiger Peptide durch Cyclodesulfhydrierung entsprechender N‐thioacylierter 2‐Mercaptoethylamin‐ oder 3‐Mercaptopropylaminderivate. Die beschriebene Reaktion wird durch eine Änderung des pH‐Wertes ausgelöst, kann in wässrigen Lösungen und bei Raumtemperatur durchgeführt werden und führt in hervorragenden bis quantitativen Ausbeuten zu komplexen Thiazolin‐ und Dihydrothiazinderivaten. In den letzten Jahrzehnten ist die Entwicklungspipeline neuer Antibiotika gegen Gram‐negative Bakterien nahezu zum Erliegen gekommen, da die meisten entdeckten Kandidaten Defizite im Bereich der Wirksamkeit, ihrer pharmakokinetischen Eigenschaften oder eine zu hohe Toxizität aufwiesen. Die Darobactine, eine vielversprechende neue Peptid‐Klasse, binden an BamA, ein zuvor von Antibiotika nicht adressiertes Zielprotein der äußeren Membran. Wir haben bereits gezeigt, dass durch Biosynthese‐Modifikationen in einem heterologen Wirt neuartige Darobactine mit erhöhter antibakterieller Aktivität erzeugt werden können. Nun verwenden wir eine optimierte Reinigungsmethode und präsentieren Kryo‐EM‐Strukturen des Bam‐Komplexes mit Darobactin 9 (D9), welche als Basis für die biotechnologische Herstellung von zwanzig neuen Darobactinen, einschließlich halogenierter Analoga, dient. Das neu entwickelte Darobactin 22 bindet enger an BamA und übertrifft das bereits hohe Aktivitätsprofil von D9 gegen klinisch relevante Krankheitserreger, wie Carbapenem‐resistente Acinetobacter baumannii, um das bis zu 32‐fache, ohne erkennbare Anzeichen von Toxizität. Unsere Suche nach neuen myxobakteriellen Naturstoffen führte zur Entdeckung der Thiamyxine, thiazol‐ und thiazolinreichen nichtribosomalen Peptid‐Polyketid‐Hybriden mit potenter antiviraler Aktivität. Vier Mitglieder dieser noch unbeschriebenen Naturstofffamilie wurden isoliert. Darunter auch ein offenkettiges Derivat, welches eine Glycerin‐Einheit am C‐Terminus trägt. Basierend auf der Analyse des Thiamyxin‐Biosythesegenclusters und unterstützt durch Fütterungsexperimente mit isotopenmarkierten Vorläufersubstanzen, beschreiben wir hier neben der Strukturaufklärung der Thiamyxine ein Biosynthesemodell. Als Besonderheit ist der Einbau eines 2‐(Hydroxymethyl)‐4‐methylpent‐3‐ensäure‐Bausteins durch die Polyketidsynthase zu nennen, die auch eine GCN5‐verwandte N‐Acetyltransferase‐Decarboxylase‐Domäne beinhaltet. Die Thiamyxine zeigen in mit Corona‐, Zika‐ und Dengue‐Virus infizierten Zellkulturmodellen eine deutliche Hemmung von RNA‐Viren. Ihre Potenz, bis zu einer halb‐maximalen Hemmkonzentration von 560 nM, in Verbindung mit milderen zytotoxischen Wirkungen auf menschliche Zelllinien, indizieren ein Potenzial für die weitere Entwicklung der Thiamyxine. Im Folgenden beschreiben wir das aus dem Myxobakterium Corallococcus coralloides isolierte Antibiotikum Corramycin. Die lineare Peptidstruktur enthält eine bislang unbeschriebene (2R,3S)‐γ‐N‐Methyl‐β‐hydroxyhistidin‐Einheit. Corramycin weist eine anti‐Gram‐negative Aktivität gegen Escherichia coli (E. coli) auf und wird über zwei Tansportersysteme, SbmA und YejABEF, in die Zelle aufgenommen. Darüber hinaus wurde das Corramycin Biosynthese‐Gencluster (BGC) identifiziert und ein Biosyntheseweg bestimmt, welcher auf einer 12‐modularen nicht‐ribosomale Peptidsynthetase/Polyketidsynthase beruht. Die bioinformatische Analyse des BGC in Verbindung mit der Entwicklung einer Totalsynthese ermöglichte die Aufklärung der absoluten Konfiguration des Moleküls. Außerdem führte die intravenöse Verabreichung von 20–30 mg Corramycin pro kg Mausgewicht in einem E. coli‐Infektionsmodell zu einer 100 %igen Überlebensrate der Tiere ohne toxische Nebenwirkungen. Somit ist Corramycin ein vielversprechender Ausgangspunkt für die Entwicklung eines wirksamen antibakteriellen Arzneimittels gegen Krankenhausinfektionen. [ABSTRACT FROM AUTHOR]

Die Kristallstrukturen der Cattleyen‐Synthase (apo‐CyS) und der mit Geranylgeranylpyrophosphat (GGPP) komplexierten CyS wurden gelöst. Die CySC59A‐Variante zeigte eine erhöhte Produktion von Cattleyen und weiteren Diterpenen mit unterschiedlichen Skeletten. Ihre Struktur zeigte ein vergrößertes aktives Zentrum, was die relaxierte Selektivität erklärt. Isotopenmarkierungsexperimente offenbarten einen bemerkenswerten Cyclisierungsmechanismus, der mehrere Gerüstumlagerungen für eines der neuen Diterpene umfasst. [ABSTRACT FROM AUTHOR]

Die Zunahme von antimikrobiellen Resistenzen stellt eine ernsthafte Bedrohung für die öffentliche Gesundheit dar. Der Naturstoff Chlorotonil wurde als neues Antibiotikum identifiziert, das gegen multiresistente Gram‐positive Pathogene und Plasmodium falciparum wirkt. Obwohl Chlorotonil vielversprechende Aktivitäten zeigt, ist das Substanzgerüst hoch lipophil und weist mögliche biologische Labilitäten auf. Deshalb bemühten wir uns um Semisynthesen, um die pharmazeutischen Eigenschaften zu verbessern. Wir konnten die stereoselektive Epoxidierung und Epoxidringöffnung mit moderaten bis guten Ausbeuten demonstrieren, wobei Derivative mit signifikant erhöhter Löslichkeit erhalten wurden. Weiterhin wurde die in vivo‐Stabilität der Derivate verbessert, während deren nanomolare Aktivität gegen wichtige Humanpathogene (z. B. Methicillin‐resistente Staphylococcus aureus und P. falciparum) beibehalten wurde. Die enantioselektive Synthese bioaktiver Verbindungen, die ein Pyrroloindolgerüst tragen, ist oft aufwändig. Im Gegensatz dazu sind mehrere S‐Adenosylmethionin (SAM)‐abhängige Methyltransferasen bekannt, die stereo‐ und regioselektive Methylierungen an der C3‐Position verschiedener Indole durchführen und damit eine direkte Bildung des gewünschten Pyrroloindolgerüsts ermöglichen. Hier wird die SAM‐abhängige Methyltransferase PsmD aus Streptomyces griseofuscus, ein Schlüsselenzym in der Biosynthese von Physostigmin, im Detail charakterisiert. Die biochemischen Eigenschaften von PsmD und ihr Substratspektrum werden aufgezeigt. Die enzymatische Methylierung im präparativen Maßstab, einschließlich der SAM‐Regeneration, wurde für drei ausgewählte Substrate nach einer Design‐of‐Experiment‐Optimierung erreicht. Das kürzlich beschriebene Rhizolutin und Collinolacton, das aus Streptomyces Gö 40/10 isoliert wurde, besitzen dasselbe neuartige Kohlenstoffgerüst. Analysen mittels NMR und Kristallstrukturanalyse belegen die Struktur von Collinolacton und machen eine Korrektur der Stereochemie von Rhizolutin erforderlich. Fütterungsexperimente mit isotopenmarkierten Vorstufen zeigen, dass Collinolacton über eine Typ‐I‐Polyketidsynthase mit anschließender Baeyer‐Villiger‐Oxidation synthetisiert wird. CRISPR‐basierte genetische Arbeiten führten zur Entdeckung des Genclusters und einem optimierten Produktionsstamm. Chemische Semisynthesen lieferten Derivate von Collinolacton, welche die Viabilität von L929‐Zellen reduzieren. Fluoreszenzmikroskopie zeigte, dass diese Derivate während der Zellteilung monopolare Spindeln induzieren. Dem beschriebenen Schutz von Rhizolutin gegen Alzheimer folgend studierten wir die neuroprotektiven Eigenschaften von Collinolacton und dessen Derivaten an Glutamat‐sensitiven Zellen (HT22). Tatsächlich zeigt Collinolacton eine Schutzwirkung gegen intrazellulären oxidativen Stress. [ABSTRACT FROM AUTHOR]

Chlorophyll und Häm sind sogenannte "Pigmente des Lebens", tetrapyrrolische Strukturen, ohne die das Leben auf der Erde nicht möglich wäre. Ihre Abbauprodukte, die Phyllobiline bzw. die Biline, teilen nicht nur strukturelle Merkmale, sondern auch eine ähnliche Geschichte: Lange Zeit wurden sie nur als Abfallprodukte von Entgiftungsprozessen angesehen; inzwischen konnten für beide Klassen wichtige Bioaktivitäten nachgewiesen werden. Für die Phyllobiline gibt es jedoch nur wenige Erkenntnisse über physiologische Rollen. Hier stellen wir mit Aktin, dem Hauptbestandteil des Zytoskeletts, die erste entdeckte Zielstruktur von Phyllobilinen und eine neue Zielstruktur von Bilinen vor. Wir zeigen die Hemmung der Aktindynamik in vitro und Auswirkungen auf Aktin und Aktin‐abhängige Prozesse in Krebszellen. Eine direkte Interaktion mit G‐Aktin wird durch in silico‐Studien dargelegt und durch Affinitätschromatographie bestätigt. Unsere Ergebnisse eröffnen ein neues Kapitel über Bioaktivitäten von Tetrapyrrolen – insbesondere der Phyllobiline – und bilden die Grundlage für weitreichende Anwendungen in der Pflanzenwissenschaft, Ökologie und Physiologie. Eine durch Bioassay geleitete Fraktionierung führte zur Isolierung von Hangtaimycin (HTM) aus Streptomyces spectabilis CCTCC M2017417 und zur Entdeckung seiner hepatoprotektiven Eigenschaften. Die Strukturaufklärung per NMR zeigte die Notwendigkeit einer Strukturrevision. Ein putatives Abbauprodukt von HTM wurde ebenfalls isoliert und dessen Struktur wurde durch Totalsynthese bestätigt. Der Biosynthesegencluster wurde identifiziert und entspricht einer hybriden trans‐AT‐PKS/NRPS Biosynthesemaschinerie, dessen erstes PKS‐Enzym ein internes dehydatisierendes Bimodul enthält, welche in anderen trans‐AT PKSs üblicherweise gespalten vorliegen. Die Mechanismen solcher dehydatisierender Bimodule wurden oft vorgeschlagen, aber niemals genau untersucht. Hier präsentieren wir in vivo‐Mutationen und enzymatische in vitro‐Experimente, die erste und detaillierte Einblicke in die Katalyse durch dehydatisierende Bimodule geben. [ABSTRACT FROM AUTHOR]

Die diversen Mikroorganismen, die Naturstoffe produzieren, sind eine wichtige Quelle für neuartige Therapeutika, Arzneimittelkandidaten und wissenschaftliche Hilfsmittel. Der allergrößte Teil der mikrobiellen Vielfalt konnte jedoch nicht axenisch kultiviert werden und gehört komplexen Lebensgemeinschaften an. Während meta′omische Methoden wie Metagenomik, ‐transkriptomik und ‐proteomik kollektive molekulare Merkmale der "mikrobiellen dunklen Materie" identifizieren, kann die Untersuchung einzelner Mikrobiom‐Bakterien eine Herausforderung sein. Um diese Limitierungen zu überwinden, wurden in den letzten eineinhalb Jahrzehnten mehrere Techniken für das Studium einzelner Bakterienzellen entwickelt. Während verschiedene dieser Methoden in der mikrobiellen Ökologie Verbreitung finden, werden sie bisher weniger häufig in der Naturstoffforschung eingesetzt. In diesem Aufsatz stellen wir die verfügbaren und neu aufkommenden Techniken für die gezielte Einzelzellanalyse vor, mit einem besonderen Schwerpunkt auf Anwendungen in der Entdeckung und Untersuchung von Naturstoffen. Um die Selektivität potenter und zugleich toxischer Antibiotika zu erhöhen, wurde eine Konjugationsstrategie entwickelt, die eine Anreicherung des Wirkstoffs am Infektionsort und seine anschließende infektionsvermittelte Freisetzung beinhaltet. Es wurde ein modifiziertes Fragment des humanen antimikrobiellen Peptids Ubiquicidin eingesetzt, dessen Akkumulation am Bakterium durch Fluorophor‐markierte Varianten visualisiert werden konnte. Um die Freisetzung von Colistin räumlich auf den Ort der Infektion zu begrenzen, wurde ein optimierter Linker verwendet, der mit hoher Effizienz durch das Enzym neutrophile Elastase (NE) gespalten wird. Die NE wird von humanen Immunzellen (neutrophilen Granulozyten) direkt am Infektionsherd freigesetzt. Die beste Anbindung an Colistin wurde anhand von fünf regioisomeren Konjugaten, die durch Totalsynthese hergestellt wurden, ermittelt. Die antibakterielle Aktivität der Konjugate war zunächst maskiert, konnte aber durch rekombinante NE freigesetzt werden, wenn sich der Linker an den Aminogruppen in 1‐ oder 3‐ Position befand. Eine Validierung des Konzepts gelang in Cokulturen von primären humanen Neutrophilen und Escherichia coli. [ABSTRACT FROM AUTHOR]

Pactamycin und Jogyamycin sind Aminocyclopentitol‐Naturstoffe, deren Kohlenstoffkern stereochemisch definierte Alkohol‐ oder Amineinheiten trägt. Aufgrund ihrer strukturellen Komplexität sowie der Vielfältigkeit der funktionellen Gruppen auf engem Raum sind sie faszinierende Syntheseziele. Pactamycin und dessen Derivate binden zur ribosomalen 30S‐Untereinheit und zeigen Wirkung gegen Parasiten, die arzneimittelresistente Malaria und afrikanische Schlafkrankheit verursachen; jedoch wird die Weiterentwicklung ihres therapeutischen Potenzials durch zelluläre Toxizität gebremst. Interessanterweise zeigen biotechnologisch erzeugte Analoga Unterschiede in der Selektivität und Toxizität in Säugerzellen, was die Entwicklung flexibler Strategien zur genauen Untersuchung der Struktur‐Wirkungs‐Beziehungen (SARs), insbesondere von Pactamycinanaloga ohne C7‐Hydroxygruppe, anspornt. In diesem Aufsatz werden Ansätze zur Synthese von Pactamycin und Jogyamycin gegenübergestellt, einschließlich zweier Totalsynthesen von Pactamycin. Wir diskutieren kurz die Bedeutung jeder Syntheseroute zu Analoga für die Untersuchung von SARs und die Entwicklung von Verbindungen mit erhöhter Selektivität für die Bindung an Ribosomen des Malariaerregers anstelle menschlicher Ribosomen. Summary Myxobacteria ‐ survivalists in soil Myxobacteria like Myxococccus xanthus are soil‐living microorganisms featuring a complex lifestyle, including movement by coordinated swarming on surfaces, predatory feeding on other microorganisms, and the formation of multicellular fruiting bodies when unfavorable environmental conditions are encountered. Bioinformatic analysis of the large myxobacterial genomes has enabled fascinating insights into the molecular basis for the biosynthesis of complex secondary metabolite structures by myxobacteria, and has set the stage for the discovery of novel natural products. Moreover, well‐characterized myxobacteria like M. xanthus increasingly play a role as "biochemical factories" for the biotechnological production of bioactive molecules using synthetic biology approaches. Eine übergangsmetallvermittelte Spaltung von C‐C‐Einfachbindungen kann eine völlig neue Retrosynthesestrategie in der Naturstoffsynthese ermöglichen. Da eine C‐C‐Bindungsspaltung das Kohlenstoffgerüst einer Verbindung stets ändert und das unter Übergangsmetallkatalyse erzeugte metallorganische oder radikalische Intermediat für den Aufbau größerer Komplexität geeignet ist, eröffnen sich durch diese C‐C‐Bindungsspaltungen neue Möglichkeiten zum drastischen und schnellen Umbau von Gerüstsystemen. Die jüngste Zunahme bei der Zahl der Anwendungen übergangsmetallvermittelter Spaltungen von C‐C‐Einfachbindungen in der Totalsynthese kann auf die allgemeine Anerkennung dieser Tatsache zurückgeführt werden. In diesem Aufsatz werden zehn Totalsynthesen von 2014 bis 2019 beleuchtet, in denen gezeigt wird, wie die übergangsmetallvermittelte C‐C‐Einfachbindungsspaltung am Kern oder an der Peripherie synthetischer Intermediate die Synthesebemühungen beschleunigen kann. Während des letzten Jahrzehnts wurde die mehrfach katalytische C‐H‐Funktionalisierung erfolgreich in der Synthese von Naturstoffen angewandt, um die Anzahl der Synthesestufen zu reduzieren, die Gesamtausbeute zu erhöhen, sowie leichter zugängliche Ausgangsverbindungen zu verwenden. In diesem Kurzaufsatz werden ausgewählte Beispiele gezeigt, in welchen die mehrfach C‐H‐Funktionalisierung auf verschiedene konzeptuelle Weisen eingesetzt wird. Zuerst werden lineare Synthesen besprochen, in welchen die mehrfache C‐H‐Funktionalisierung entweder an einfachen (hetero)zyklischen Strukturen, als späte Modifikationen oder zum Aufbauen von polyzyklischen Systemen Verwendung findet. Anschließend wird demonstriert, wie der Gebrauch von mehrfachen C‐H‐Funktionalisierungen als strategisches Werkzeug in konvergenten Synthesen angewandt werden kann, um komplexe Fragmente zu erhalten und zu kuppeln. Informationen über die Skalierbarkeit der verwendeten Methoden ist gegeben, wenn verfügbar. Die ausgewählten Beispiele zeigen, dass die Verwendung von mehrfachen C‐H‐Funktionalisierungsstrategien eine wichtige Rolle in der Gestaltung von zukünftigen Synthesen funktioneller und komplexer Moleküle mit verbesserter Nachhaltigkeit sein sollten. Wir beschreiben die Totalsynthese des potenten neuen Antibiotikums Disciformycin B (2), das eine ausgeprägte Aktivität gegenüber Methicillin‐ und Vancomycin‐resistenten Stämmen von Staphylococcus aureus (MRSA/VRSA) aufweist. Der zentrale Syntheseschritt besteht in der Makrozyklisierung eines Tetraens zum 12‐gliedrigen Makrolactonring des Naturstoffs durch eine ringschließende Olefinmetathese. Zwar wurde neben dem Makrozyklus über einen alternativen Ringschlussmetatheseweg auch ein Cyclopentenderivat gebildet, es konnten jedoch Bedingungen gefunden werden, die das gewünschte Makrolacton als Hauptprodukt lieferten. Die Schlüsselschritte bei der Herstellung des Metathesesubstrats waren eine hocheffiziente Evans‐syn‐Aldolreaktion, die asymmetrische Brown‐Allylierung des Angelikaaldehyds, sowie die stereoselektive 1,2‐Reduktion eines Enons mittels Zn(BH4)2. Die Synthese wurde nach der Makrozyklisierung über eine dehydratative Glykosylierung zur Einführung des d‐Arabinofuranosylrests und schließlich eine chemoselektive Oxidation eines allylischen Alkohols zu Ende geführt. Non‐natural terpenoids offer potential as pharmaceuticals and agrochemicals. However, their chemical syntheses are often long, complex, and not easily amenable to large‐scale production. Herein, we report a modular chemoenzymatic approach to synthesize terpene analogues from diphosphorylated precursors produced in quantitative yields. Through the addition of prenyl transferases, farnesyl diphosphates, (2E,6E)‐FDP and (2Z,6Z)‐FDP, were isolated in greater than 80 % yields. The synthesis of 14,15‐dimethyl‐FDP, 12‐methyl‐FDP, 12‐hydroxy‐FDP, homo‐FDP, and 15‐methyl‐FDP was also achieved. These modified diphosphates were used with terpene synthases to produce the unnatural sesquiterpenoid semiochemicals (S)‐14,15‐dimethylgermacrene D and (S)‐12‐methylgermacrene D as well as dihydroartemisinic aldehyde. This approach is applicable to the synthesis of many non‐natural terpenoids, offering a scalable route free from repeated chain extensions and capricious chemical phosphorylation reactions. Tapinarof is a stilbene drug that is used to treat psoriasis and atopic dermatitis, and is thought to function through regulation of the AhR and Nrf2 signaling pathways, which have also been linked to inflammatory bowel diseases. It is produced by the gammaproteobacterial Photorhabdus genus, which thus represents a model to probe tapinarof structural and functional transformations. We show that Photorhabdus transforms tapinarof into novel drug metabolism products that kill inflammatory bacteria, and that a cupin enzyme contributes to the conversion of tapinarof and related dietary stilbenes into novel dimers. One dimer has activity against methicillin‐resistant Staphylococcus aureus (MRSA) and vancomycin‐resistant Enterococcus faecalis (VRE), and another undergoes spontaneous cyclizations to a cyclopropane‐bridge‐containing hexacyclic framework that exhibits activity against Mycobacterium. A spider‐transmitted fungus (Rhizopus microsporus) that was isolated from necrotic human tissue was found to harbor endofungal bacteria (Burkholderia sp.). Metabolic profiling of the symbionts revealed a complex of cytotoxic agents (necroximes). Their structures were characterized as oxime‐substituted benzolactone enamides with a peptidic side chain. The potently cytotoxic necroximes are also formed in symbiosis with the fungal host and could have contributed to the necrosis. Genome sequencing and computational analyses revealed a novel modular PKS/NRPS assembly line equipped with several non‐canonical domains. Based on gene‐deletion mutants, we propose a biosynthetic model for bacterial benzolactones. We identified specific traits that serve as genetic handles to find related salicylate macrolide pathways (lobatamide, oximidine, apicularen) in various other bacterial genera. A divergent synthetic approach to six Ganoderma meroterpenoids, namely ganocins A–C, ganocochlearins C and D, and cochlearol T, has been developed for the first time. This synthetic route features a two‐phase strategy which includes early‐stage rapid construction of a common planar tricyclic intermediate followed by highly selective late‐stage transformations into various Ganoderma meroterpenoids. Key to the strategy are a bioinspired intramolecular hetero‐Diels–Alder reaction and Stahl‐type oxidative aromatization, allowing efficient formation of the common tricyclic phenol intermediate. A nucleophilic dearomatization of the phenol unit, combined with a regioselective 1,4‐reduction of the resulting dienone, enabled rapid access to ganocins B and C. Additionally, site‐selective Mukaiyama hydration, followed by an intramolecular oxa‐Michael addition/triflation cascade, served as a key strategic element in the chemical synthesis of ganocin A. Vibralactone is isolated from the basidiomycete fungus Boreostereum vibrans as one of the strongest lipase inhibitors. Its unusual β‐lactone‐fused bicycle is derived from an aryl ring moiety by an oxidative ring‐expansion prior to an intramolecular cyclization. Herein, we report the discovery of the cyclase VibC which belongs to the α/β‐hydrolase superfamily and is involved in the vibralactone biosynthesis. Biochemical and crystal studies suggest that VibC may catalyze an aldol or an electrocyclic reaction initiated by the Ser‐His‐Asp catalytic triad. For the aldol and pericyclic chemistry in living cells, VibC is a unique hydrolase performing the carbocycle formation of an oxepinone to a fused bicyclic β‐lactone. This presents a naturally occurring, new enzymatic reaction in both aldol and hydrolase (bio)chemistry that will guide future exploitation of these enzymes in synthetic biology for chemical‐diversity expansion of natural products. A total synthesis of tiacumicin B, a natural macrolide whose remarkable antibiotic properties are used to treat severe intestinal infections, is reported. The strategy is in part based on the prior synthesis of the tiacumicin B aglycone, and on the decisive use of sulfoxides as anomeric leaving groups in hydrogen‐bond‐mediated aglycone delivery (HAD). This new HAD variant permitted highly β‐selective rhamnosylation and noviosylation. To increase convergence, the rhamnosylated C1–C3 fragment thus obtained was anchored to the C4–C19 aglycone fragment by adapting the Suzuki–Miyaura cross‐coupling used for the aglycone synthesis. Ring‐size‐selective macrolactonization provided a compound engaged directly in the noviolysation step with virtually total β selectivity. A total synthesis of tetrodotoxin was accomplished. A Diels–Alder reaction between a known enone and a siloxy diene gave a tricyclic product, the steric bias of which was used to construct the remaining stereogenic centers. A nitrogen atom was introduced either by a four‐step sequence involving a Curtius rearrangement, or a three‐step sequence featuring a newly developed transformation of a terminal alkyne into a nitrile. LuxR‐type transcriptional activator proteins frequently regulate the expression of biosynthetic gene clusters (BGCs). With only a fraction of bacterial BGCs being expressed under standard culturing conditions, modulation of LuxRs would provide a powerful approach to activate silent clusters. We show that by exploiting the modular nature of LuxR proteins, it is possible to construct functional chimeric LuxRs, which enables both the rewiring of quorum sensing systems and the activation of silent BGCs. Butenolides are well‐known signaling molecules in Gram‐positive bacteria. Here, we describe a novel class of butenolides isolated from a Gram‐negative Pseudomonas strain, the styrolides. Structure elucidation was aided by the total synthesis of styrolide A. Transposon mutagenesis enabled us to identify the styrolide biosynthetic gene cluster, and by using a homology search, we discovered the related and previously unknown acaterin biosynthetic gene cluster in another Pseudomonas species. Mutagenesis, heterologous expression, and identification of key shunt and intermediate products were crucial to propose a biosynthetic pathway for both Pseudomonas‐derived butenolides. A highly convergent approach was developed to achieve the first asymmetric and scalable total synthesis of FD‐594, a complex polycyclic xanthone natural product from Streptomyces sp. TA‐0256, in a longest linear sequence (LLS) of 20 steps. The trans‐9,10‐dihydrophenanthrene‐9,10‐diol fragment (B‐C‐D ring) was generated through a new strategy involving asymmetric dihydroxylation followed by Cu‐mediated oxidative cyclization. C–S bond formation reactions are widely distributed in the biosynthesis of biologically active molecules, and thus have received much attention over the past decades. Herein, we report intramolecular C–S bond formation by a P450 monooxygenase, TleB, which normally catalyzes a C−N bond formation in teleocidin biosynthesis. Based on the proposed reaction mechanism of TleB, a thiol‐substituted substrate analogue was synthesized and tested in the enzyme reaction, which afforded the unprecedented sulfur‐containing thio‐indolactam V, in addition to an unusual indole‐fused 6/5/8‐tricyclic product whose structure was determined by the crystalline sponge method. Interestingly, conformational analysis revealed that the SOFA conformation is stable in thio‐indolactam V, in sharp contrast to the major TWIST form in indolactam V, resulting in differences in their biological activities. The first total synthesis of the Euphorbia diterpenoid pepluanol B in both racemic and enantioenriched form involves 20 steps from a known bicyclic diol. This synthesis features an unprecedented bromo‐epoxidation to control the eight‐membered‐ring conformation. In addition, salient reactions for the construction of the tetracyclic backbone include a sterically challenging aldol reaction to establish the quaternary center, a ring closing metathesis (RCM) to forge the eight‐membered ring, and a diastereoselective cyclopropanation to assemble the embedded cyclopropane motif. We present the first asymmetric total synthesis and absolute configuration determination of (+)‐tronocarpine. The [6.5.7.6.6] pentacyclic core was constructed at an early stage by using a sequential cyclization strategy through a newly developed catalytic asymmetric Michael/aldol cascade to build the aza[3.3.1]‐bridged cycle and a tandem reduction/hemiamidation procedure to assemble the seven‐membered lactam. The side‐chain functionalities were incorporated at a late stage by several appropriately orchestrated manipulations under mild conditions. The rhytidenone family comprises spirobisnaphthalene natural products isolated from the mangrove endophytic fungus Rhytidhysteron rufulum AS21B. The biomimetic synthesis of rhytidenone A was achieved by a Michael reaction/aldol/lactonization cascade in a single step from the proposed biosynthetic precursor rhytidenone F. Moreover, the mode of action of the highly cytotoxic rhytidenone F was investigated. The pulldown assay coupled with mass spectrometry analysis revealed the target protein PA28γ is covalently attached to rhytidenone F at the Cys92 residue. The interactions of rhytidenone F with PA28γ lead to the accumulation of p53, which is an essential tumor suppressor in humans. Consequently, the Fas‐dependent signaling pathway is activated to initiate cellular apoptosis. These studies have identified the first small‐molecule inhibitor targeting PA28γ, suggesting rhytidenone F may serve as a promising natural product lead for future anticancer drug development. A catalytic dynamic kinetic resolution and asymmetric acylation reaction of hydroxyphthalides is developed. The reaction involves formation of a carbene catalyst derived chiral acyl azolium intermediate that effectively differentiates the two enantiomers of racemic hydroxyphthalides. The method allows quick access to enantiomerically enriched phthalidyl esters with proven applications in medicine. Herein, we report an engineered enzyme that can monooxygenate unprotected tryptophan into the corresponding 3a‐hydroxyhexahydropyrrolo[2,3‐b]indole‐2‐carboxylic acid (HPIC) in a single, scalable step with excellent turnover number and diastereoselectivity. Taking advantage of directed evolution, we analyzed the stepwise oxygen‐insertion mechanism of tryptophan 2,3‐dioxygenases, and transformed tryptophan 2,3‐dioxygenase from Xanthomonas campestris into a monooxygenase for oxidative cyclization of tryptophans. It was revealed that residue F51 is vital in determining the product ratio of HPIC to N′‐formylkynurenine. A novel palladium‐catalyzed Heck‐type reaction of thiocarbamates has been designed to construct bridged seven‐membered‐ring systems that are otherwise challenging to prepare. Taking advantage of this newly developed method, enantioselective syntheses of lyconadins A–E (1–5), lycopecurine (6), and dehydrolycopecurine (7) have been realized in a divergent fashion. Our synthetic strategy also features an intramolecular cyclization of a N‐chloroamine to forge the C6−N bond, a transannular Mannich‐type reaction of a cyclic nitrone to stitch the C4 and C13 together, and a cyclocondensation to deliver the (dihydro‐)pyridone motif. The ophiobolin sesterterpenes are notable plant pathogens which have recently elicited significant chemical and biological attention because of their intriguing carbogenic frameworks, reactive functionalities, and emerging anticancer profiles. Reported herein is a total synthesis of (+)‐6‐epi‐ophiobolin A in 14 steps, a task which addresses construction of the synthetically challenging spirocyclic tetrahydrofuran motif as well as several other key stereochemical problems. Für die Behandlung schwerer Krankheiten, wie z. B. Krebs, sind neue Angriffspunkte für die Therapie von großer Bedeutung. Seit Jahrzehnten treiben Naturstoffe die Entwicklung innovativer Therapieansätze voran, da diese in zellbiologische Prozesse eingreifen. Von der potenten zytotoxischen Aktivität der myxobakteriellen Vioprolide A–D inspiriert, wurden hier umfassende Studien bezüglich ihrer Wirkmechanismen durchgeführt. Auf Grundlage der potenten Aktivität gegen akute lymphoblastische Leukämie(ALL)‐Zellen, wurde zur Identifikation der Angriffsziele des aktivsten Derivats, Vioprolid A (VioA), ein thermisches Protein‐Profiling (TPP) in Jurkat‐Zellen durchgeführt. Durch eine Vielzahl daran anschließender proteomischer und biologischer Experimente konnte das nukleoläre Protein 14 (NOP14), welches eine essentielle Rolle in der Ribosomen‐Biogenese einnimmt, als Angriffsziel von VioA validiert werden. Aufgrund der erhöhten Aktivität gegen ALL‐Krebszellen im Vergleich zu gesunden Lymphozyten weist VioA ein einzigartiges therapeutisches Potenzial über einen neuen Wirkmechanismus auf. Upon injury, psychotropic psilocybin‐producing mushrooms instantly develop an intense blue color, the chemical basis and mode of formation of which has remained elusive. We report two enzymes from Psilocybe cubensis that carry out a two‐step cascade to prepare psilocybin for oxidative oligomerization that leads to blue products. The phosphatase PsiP removes the 4‐O‐phosphate group to yield psilocin, while PsiL oxidizes its 4‐hydroxy group. The PsiL reaction was monitored by in situ 13C NMR spectroscopy, which indicated that oxidative coupling of psilocyl residues occurs primarily via C‐5. MS and IR spectroscopy indicated the formation of a heterogeneous mixture of preferentially psilocyl 3‐ to 13‐mers and suggest multiple oligomerization routes, depending on oxidative power and substrate concentration. The first total synthesis of nominal harziane diterpenoid 1 is disclosed, whose spectral characteristics did not match those of the reported natural product. Stereochemical analysis and subsequent synthesis of the epimeric tertiary alcohol led to reassignment of configuration of the natural product as shown for 2. At the heart of the synthesis is an enyne cycloisomerization that sets a key quaternary stereocenter within a cyclobutane with high diastereocontrol. The discovery of biologically active small molecules requires sifting through large amounts of data to identify unique or unusual arrangements of atoms. Here, we develop, test and evaluate an atom‐based sort to identify novel features of secondary metabolites and demonstrate its use to evaluate novelty in marine microbial and sponge extracts. This study outlines an important ongoing advance towards the translation of autonomous systems to identify, and ultimately elucidate, atomic novelty within a complex mixture of small molecules. Bioactive natural products underpin the intriguing pollination strategy used by sexually deceptive orchids. These compounds, which mimic the sex pheromones of the female insect, are emitted in particular blends to lure male insect pollinators of specific species. By combining methods from field biology, analytical chemistry, electrophysiology, crystallography, and organic synthesis, we report that an undescribed β‐hydroxylactone, in combination with two specific hydroxymethylpyrazines, act as pollinator attractants in the rare hammer orchid Drakaea micrantha. This discovery represents an unusual case of chemically unrelated compounds being used together as a sexual attractant. Furthermore, this is the first example of the identification of pollinator attractants in an endangered orchid, enabling the use of chemistry in orchid conservation. Thiostrepton is a potent antibiotic against a broad range of Gram‐positive bacteria, but its medical applications have been limited by its poor aqueous solubility. In this work, the first C(sp2)−H amidation of dehydroalanine (Dha) residues was applied to the site selective modification of thiostrepton to prepare a variety of derivatives. Unlike all prior methods for the modification of thiostrepton, the alkene framework of the Dha residue is preserved and with complete selectivity for the Z‐stereoisomer. Additionally, an aldehyde group was introduced by C−H amidation, enabling oxime ligation for the installation of an even greater range of functionality. The thiostrepton derivatives generally maintain antimicrobial activity, and importantly, eight of the derivatives displayed improved aqueous solubility (up to 28‐fold), thereby addressing a key shortcoming of this antibiotic. The exceptional functional group compatibility and site selectivity of CoIII‐catalyzed C(sp2)−H Dha amidation suggests that this approach could be generalized to other natural products and biopolymers containing Dha residues. MtmOIV and MtmW catalyze the final two reactions in the mithramycin (MTM) biosynthetic pathway, the Baeyer–Villiger opening of the fourth ring of premithramycin B (PMB), creating the C3 pentyl side chain, strictly followed by reduction of the distal keto group on the new side chain. Unexpectedly this results in a C2 stereoisomer of mithramycin, iso‐mithramycin (iso‐MTM). Iso‐MTM undergoes a non‐enzymatic isomerization to MTM catalyzed by Mg2+ ions. Crystal structures of MtmW and its complexes with co‐substrate NADPH and PEG, suggest a catalytic mechanism of MtmW. The structures also show that a tetrameric assembly of this enzyme strikingly resembles the ring‐shaped β subunit of a vertebrate ion channel. We show that MtmW and MtmOIV form a complex in the presence of PMB and NADPH, presumably to hand over the unstable MtmOIV product to MtmW, yielding iso‐MTM, as a potential self‐resistance mechanism against MTM toxicity. The synthesis of structurally complex and highly strained natural products provides unique challenges and unexpected opportunities for the development of new reactions and strategies. Herein, the synthesis of (+)‐[5]‐ladderanoic acid is reported. En route to the target, unusual and unexpected strain release driven transformations were uncovered. This occurrence required a drastic revision of the synthetic design that ultimately led to the development of a novel stepwise cyclobutane assembly by an allylboration/Zweifel olefination sequence.
