Abstract

The role of polarity signaling in cancer metastasis is ill defined. Using two three-dimensional culture models of mammary epithehal cells and an orthotopic mouse model of breast cancer, we reveal that Par6 signaling, which is regulated directly by TGFβ, plays a role in breast cancer metastasis. Interference with Par6 signaling blocked TGFβ-dependent loss of polarity in acini-like structures formed by non-transformed mammary cells grown in three-dimensional structures and suppressed the protrusive morphology of mesenchymal-like invasive mammary tumor cells without rescuing E-cadherin expression. Moreover, blockade of Par6 signaling in an in vivo orthotopic model of metastatic breast cancer induced the formation of ZO-1-positive epithehium-like structures in the primary tumor and suppressed metastasis to the lungs. Analysis of the pathway in tissue microarrays of human breast tumors further revealed that Par6 activation correlated with markers of the basal carcinoma subtype in BRCA 1-associated tumors. Abstract

Background: Porphyromonas gingivalis is found in subgingival biofilm and is associated with periodontal disease. Bacteria in biofilms are able to resist higher antimicrobial concentrations than in suspension. Little is known about the susceptibility of P. gingivalis in biofilms to antimicrobial agents. The effects of chlorhexidine gluconate, minocycline hydrochloride, and metronidazole on P. gingivalis biofilms were examined in vitro. Methods: P. gingivalis strain 381 biofilms were prepared on 32 hydroxyapatite disks. At 0, 24, 72, and 144 hours after perfusion of the three antimicrobial agents, two disks from each device were used to assess the antimicrobial effects by adenosine triphosphate (ATP) bioluminescence, and for morphological investigation by scanning electron microscopy (SEM). Results: Close relationships were found between the results of the ATP analyses and the SEM observations in all groups examined. A significant decrease (P <0.001) in ATP content was found between the chlorhexidine-treated and control groups. The extra-cellular matrix structure and P. gingivalis cells were altered in the presence of chlorhexidine. Minocycline hydrochloride also caused a decrease (P <0.05) in the ATP content and morphological change on P. gingivalis biofilms. Metronidazole showed no significant efficacy against P. gingivalis biofilms. Conclusion: Chlorhexidine gluconate was effective at reducing the viability of P. gingivalis 381 cells in biofilms, while minocycline hydrochloride and metronidazole exhibited weaker effects. Abstract

ABSTRACTPorphyromonas gingivalis, an oral bacterium, might play a role in the pathogenesis or progression of adult periodontitis. In this study, we isolated from P. gingivalisa putative glycosyltransferase gene, designated gtfA, which had a consensus domain for glycosyltransferase in its N terminus. GtfA consisted of 248 amino acids and its predicted molecular mass was 28 kDa; however, as the molecular mass of endogenous GtfA protein was around 40 kDa, this suggested that GtfA had undergone some posttranslational modifications. To reveal the role of the gtfAgene in P. gingivalis, we established gtfA-deficient strains by allelic replacement. Morphologically, gtfA-deficient P. gingivalislacked mature fimbriae. gtfA-deficient P. gingivalisalso showed a very low ability for autoaggregation, and its ability to attach to epithelial cells was severely impaired. Thus, the results indicate that the gtfAgene is required for P. gingivalisautoaggregation as well as attachment to epithelial cells. These results suggest that GtfA might have an important role in the pathogenicity of P. gingivalisby regulating adhesion.

Signal transducer and activator of transcription 3 (STAT3) mediates signals of various growth factors and cytokines, including interleukin-6 (IL-6). In certain IL-6-responsive cell lines, the stat3gene is autoregulated by STAT3 through a composite IL-6 response element in its promoter that contains a STAT3-binding element (SBE) and a cyclic AMP-responsive element. To reveal the nature and roles of the stat3autoregulation in vivo, we generated mice that harbor a mutation in the SBE (stat3mSBE). The intact SBE was crucial for IL-6-induced stat3gene activation in the spleen, especially in the red pulp region, the kidney, and both mature and immature T lymphocytes. The SBE was not required, however, for IL-6-induced stat3gene activation in hepatocytes. T lymphocytes from the stat3mSBE/mSBEmice were more susceptible to apoptosis despite the presence of IL-6 than those from wild-type mice. Consistent with this, IL-6-dependent activation of the Pim-1and junBgenes, direct target genes for STAT3, was attenuated in T lymphocytes of the stat3mSBE/mSBEmice. Thus, the tissue-specific autoregulation of the stat3gene operates in vivo and plays a role in IL-6-induced antiapoptotic signaling in T cells.

Abstract

This in vitro study aimed to design a method for the objective evaluation of carious dentin using numerical values. This study also investigated the relationship between the color of carious dentin stained with a caries detector dye using this objective method and the rate of bacterial detection as detected by a polymerase chain reaction (PCR). In 15 molars with occlusal dentin caries and three extracted sound molars, dentin was removed in multiple steps with 300 microm removed each step. Before and after every removal, images of a color-matching sticker and carious surfaces stained with a caries detector dye were acquired simultaneously using a CCD camera and dentinal tissue samples were removed with a round bur. Next, corrected L*, a* and b* values of the carious surfaces (CIE 1976 L*a*b* color system) were calculated from the color changes of the stickers in the images. In addition, bacterial DNA in the dentinal tissue was detected by PCR. From evaluations of the receiver operating characteristic curves for the L*, a* and b* values, the L* value was determined to be a more useful parameter than a* or b* for detecting bacterial infection using the caries detector dye. The bacterial detection rates of carious dentin decreased as the L* values of carious dentin stained with the dye increased. When the L* values were more than 60, the dentin had no bacterial infection. This study clarified the relationship between the colors of lesions stained with a caries detector dye and the rates of bacterial detection.
