International collaborative research projects conducted at academic research institutions, including complex basic science, clinical, and translational HIV/AIDS research, require intensive communication, coordination, and thoughtful relationship-building at all institutional levels and especially require the support of experienced and well-trained Research Administrators. To be successful, global research teams must be not only scientifically adept, but supported by a staff skilled in identifying opportunities, submitting proposals, and managing all aspects of award administration and reporting. Using a National Institutes of Health, Fogarty International Center funded training grant, the project team aimed to develop a comprehensive Research administration capacity-building program that would improve the support provided to investigators at Mbarara University of Science and Technology in Uganda through collaboration with the University of Virginia in the United States and expand future funding success for innovative HIV/AIDS and HIV-related research. The experiences and achievements of conducting a comprehensive, primarily remote training program are documented for other global partnerships to highlight successes and uncover opportunities to ensure sustainability. Despite the hurdles of the COVID-19 pandemic, this program was able to succeed through a long-term relationship of mutual respect between the two institutions. In rural Uganda where birth dose vaccination for hepatitis B is not routine, we implemented a pilot program for preventing mother-to-child transmission that effectively identified women with high-risk hepatitis B virus (HBV) infection and started antiviral treatment during pregnancy. Sea cucumber tendon, one of the main parts of sea cucumber viscera, is widely accepted as a waste residue. In this study, a sea cucumber tendon polysaccharide (SCTPII) was purified from sea cucumber tendons and its primary structures and immunomodulatory activity were investigated. SCTPII is a triple-helix conformation homogeneous polysaccharide with a molecular weight of 3.97 × 10 6 Da that consists of glucose and fucose with molar ratios of 92.09% and 7.91% with high thermostability. In vivo tests on immunosuppressed Balb/c mice revealed that compared with the model group, the proliferation of T cells and B cells in splenic lymphocytes of mice in the high-dose group was significantly improved by 0.92 times and 5.14 times, respectively. Additionally, SCTPII could improve the proliferation ability and phagocytosis of macrophages, as well as promoting the expression of IL-6, TNF-α and IFN-γ and enhancing the intestinal physical barrier function by increasing the protein expression of claudin-1, occludin, ZO-1 and MUC2. Furthermore, the 16S rRNA sequencing of fecal samples was performed, and gene count and α-diversity analysis revealed that SCTPs could improve the microbial community richness. In particular, SCTPs could increase the relative abundance of Lactobacillus , Bacteroides and Akkermansia and reduce the relative abundance of Lachnospiraceae_NK4A136_group and Rikenellaceae_RC9_gut_group. These results demonstrate that SCPII possesses potential immunoregulatory activities in cyclophosphamide-induced mice by regulating intestinal microbiota diversity and improving immune organs, enhancing the proliferation ability of macrophages and splenocyte proliferation, and enhancing intestinal physical barrier function, which might provide important evidence for the development and utilization of the viscera of sea cucumber.

Background: Progress against malaria has stalled and may even be slipping backwards in high-burden countries. This is due to a range of factors including insecticide resistance and mosquito feeding behaviours that limit contact with widely-employed interventions including long-lasting insecticidal nets and indoor-residual spraying. Thus, further innovations in malaria control are urgently needed., Methods: The pilot was a randomized, placebo-controlled pilot study of permethrin-treated baby wraps-known locally as lesus-in children 6-18 months of age at a single site in rural western Uganda. Fifty mother-infant pairs were assigned to permethrin-treated or untreated lesus in a 1:1 allocation. Participants and clinical staff were blinded to group assignments through use of sham treatment and re-treatment of lesus. Participants attended scheduled clinic visits every 2 weeks for a total 12 weeks. The primary outcome of interest was the safety of the intervention, assessed as changes in the frequency of use, rates of discontinuation, and incidence of adverse events, such as skin rash. Secondary outcomes included acceptability and feasibility of the intervention as measured through participant satisfaction and completion of study activities, respectively., Results: Overall, rates of retention and participation were relatively high with 86.0% (43 of 50) of participants completing all scheduled visits, including 18 (75.0%) and 25 (96.2%) in the intervention and control arms respectively. By the conclusion of the 12-week follow-up period, one adverse event (0.35 events per 100 person-weeks, one-sided 95% CI 0.0-1.65) was reported. Satisfaction with the lesu was high in both groups. In each study arm, there were five incident RDT positive results, but the only PCR-positive results were observed in the control group (n = 2)., Conclusions: Permethrin-treated baby wraps were well-tolerated and broadly acceptable. Adverse events were infrequent and mild. Herpesviruses are thought to have evolved in very close association with their hosts. This is notably the case for cytomegaloviruses (CMVs; genus Cytomegalovirus ) infecting primates, which exhibit a strong signal of co-divergence with their hosts. Some herpesviruses are however known to have crossed species barriers. Based on a limited sampling of CMV diversity in the hominine (African great ape and human) lineage, we hypothesized that chimpanzees and gorillas might have mutually exchanged CMVs in the past. Here, we performed a comprehensive molecular screening of all 9 African great ape species/subspecies, using 675 fecal samples collected from wild animals. We identified CMVs in eight species/subspecies, notably generating the first CMV sequences from bonobos. We used this extended dataset to test competing hypotheses with various degrees of co-divergence/number of host switches while simultaneously estimating the dates of these events in a Bayesian framework. The model best supported by the data involved the transmission of a gorilla CMV to the panine (chimpanzee and bonobo) lineage and the transmission of a panine CMV to the gorilla lineage prior to the divergence of chimpanzees and bonobos, more than 800,000 years ago. Panine CMVs then co-diverged with their hosts. These results add to a growing body of evidence suggesting that viruses with a double-stranded DNA genome (including other herpesviruses, adenoviruses, and papillomaviruses) often jumped between hominine lineages over the last few million years.

Batrachochytrium dendrobatidis (Bd), the cause of chytridiomycosis, is a pathogenic fungus that is found worldwide and is a major contributor to amphibian declines and extinctions. We report results of a comprehensive effort to assess the distribution and threat of Bd in one of the Earth's most important biodiversity hotspots, the Albertine Rift in central Africa. In herpetological surveys conducted between 2010 and 2014, 1018 skin swabs from 17 amphibian genera in 39 sites across the Albertine Rift were tested for Bd by PCR. Overall, 19.5% of amphibians tested positive from all sites combined. Skin tissue samples from 163 amphibians were examined histologically; of these two had superficial epidermal intracorneal fungal colonization and lesions consistent with the disease chytridiomycosis. One amphibian was found dead during the surveys, and all others encountered appeared healthy. We found no evidence for Bd-induced mortality events, a finding consistent with other studies. To gain a historical perspective about Bd in the Albertine Rift, skin swabs from 232 museum-archived amphibians collected as voucher specimens from 1925-1994 were tested for Bd. Of these, one sample was positive; an Itombwe River frog (Phrynobatrachus asper) collected in 1950 in the Itombwe highlands. This finding represents the earliest record of Bd in the Democratic Republic of Congo. We modeled the distribution of Bd in the Albertine Rift using MaxEnt software, and trained our model for improved predictability. Our model predicts that Bd is currently widespread across the Albertine Rift, with moderate habitat suitability extending into the lowlands. Under climatic modeling scenarios our model predicts that optimal habitat suitability of Bd will decrease causing a major range contraction of the fungus by 2080. Our baseline data and modeling predictions are important for comparative studies, especially if significant changes in amphibian health status or climactic conditions are encountered in the future.
