Processing is required to render the products of some fish species edible, thus enhancing their economic value. The aim of the present study was to evaluate the nutritional value of goldfish (Carassius auratus L.) products from Lake Trasimeno (Umbria, Italy). The same number of fillets (n=20), raw and pre-cooked pulp samples, as well as of croquettes were used to evaluate the proximate composition, total lipid fatty acid profile and the protein and lipid oxidative stability. The water holding capacity (WHC) and the percentage of cooking loss (CL) were also assessed. Despite the low amount of lipids (1.85%), fresh goldfish fillets were characterized by a high PUFA level (54.30%), with DHA being the most abundant long-chain fatty acid (15.60%) followed by EPA (9.45%). Fillet processing reduced both WHC and CL of the final products, while the production of TBARS and carbonylated proteins increased, especially in croquettes (+ 59 and + 77%, respectively). Thrombogenic and atherogenic indexes also increased due to the PUFA reduction, reaching statistical significance only for the croquettes (0.53 vs 0.21 and 0.46 vs 0.34, respectively). These results suggest that goldfish fillets are characterized by a high nutritional value and a good oxidative stability. Forty rainbow trout (Oncorhynchus mykiss L.) reared in Valnerina (umbria, italy) were studied to determine the oxidative stability of fillets during a 5-day storage period at 2°C under continuous cool white fluorescent illumination. Chemical, physical and microbiological analyses were carried out. High quality fresh trout fillets were characterized by a low lipid and cholesterol content (4.2% and 37 mg/100 g, respectively), a high hypo-cholesterolemic/ hyper-cholesterolemic fatty acid ratio (2.9) and low atherogenic and thrombogenic index values (0.45 and 0.21, respectively). After three days of display, the oxidative stability of the fillets decreased (2.7 vs 0.8 mg MDA/ kg, P<0.05) compromising the quality of the product. The lipid profile of the fillets also changed: the SFA and MUFA increased due to PUFA oxidation and/ or drip loss, while EPA and DHA tended to decrease. Nevertheless the levels of both long-chain PUFA were higher with respect to the baseline values, indicating that drip loss, more than oxidation, affected the amount of these compounds in fillets at the end of the experimental storage. Abstract Background The prostate gland of rabbits produces numerous granules, which are specifically implicated in the inhibition of sperm capacitation during the first hours after mating. These granules are rich in vitamin E, but their role in the antioxidant protection of rabbit sperm has not been studied. Aim of study The objectives of this study were to investigate whether the prostatic secretory granules (PSGs) could prevent sperm induced-lipid peroxidation and to verify a potential involvement of tocopherols from the PSGs to the sperm. Methods Washed sperm samples taken from eighteen White New Zealand rabbits were either incubated with tert-butyl-hydroperoxide (t-BHP, an oxidative stressor) or with buffered Tyrode's medium for 1 hour. The same number of sperm samples that contained PSGs were subjected to the previously mentioned treatments and thiobarbituric acid reactive substances (TBARS), vitamin E compounds and the acrosome status were assessed. Results The incubation of the sperm with t-BHP resulted in a noticeable production of TBARS (0.38 vs. 0.22 nmol/10(7) cells) and an associated decrease of alpha-tocopherol (alpha-T, 72.3 vs. 103.2 nmol/10(8) cells) with respect to the sperm samples containing PSGs. The sperm incubated with the PSGs had a higher amount of alpha-T compared to the control (292.2 vs. 251.0 nmol/10(8) cells). The acrosome status was not affected by the occurrence of the organic peroxide in the medium and the amount of capacitated sperm was lower when the PSGs were also present. Conclusions Overall, these results suggest that the PSGs may represent a source of protection for rabbit sperm against in vitro oxidative stress by supplying the sperm with endogenous alpha-T. This mechanism could be in part involved in the inhibition of sperm capacitation by the granules.

This study compared dietary isoflavone aglycones with the glycoside conjugates in a novel model of postmenopausal status, the aging domestic hen ( Gallus gallus domesticus ), to determine the effects on reproductive performance, cholesterol levels, and nutritional quality of eggs laid. Hens, 18 mo old, were randomized into four groups ( n = 10/group) and fed for 28 d a conventional poultry corn/soymeal diet (Control), or diets supplemented with isoflavone glycosides from soy germ (diet A), isoflavone aglycons from a soy germ pasta (diet B), or conventional pasta lacking isoflavones (diet C). The egg-laying rate was recorded daily, plasma isoflavones and cholesterol were measured, and the nutritional composition of the eggs was determined. Egg-laying declined over a 4-week period in hens in the Control group and those fed isoflavone glycosides (diets A and C), whereas hens fed isoflavone aglycons (diet B) significantly increased their egg-laying efficiency. The total egg count and egg yield were significantly higher in hens fed isoflavone aglycons, and their plasma cholesterol concentrations were lower and the eggs laid had a 30% lower yolk cholesterol content. None of these effects were observed with diets containing similar levels of isoflavone glycosides. These studies recapitulate the clinical effects of soy germ pasta enriched with isoflavone aglycons and lend support to the greater efficacy of a diet rich in isoflavone aglycons.

The aim of this study was to investigate the presence of the different isoforms of tocopherol (T) in seminal plasma (P) and in the sperm fractions of individuals with abnormal (group 1) and normal (group 2) sperm parameters; the relationships between these isoforms and conventional sperm parameters were also explored. Two vitamin E homologues, α-T and δ-T, were identified in the semen of all participants. Although α-T and δ-T concentrations were similar in the semen of the 2 groups, group 1 showed a lower α-T ratio (S/P) (0.90 vs. 1.20, P < .001) and δ-T ratio (0.86 vs 1.13, P = .007) than group 2. In addition, both T ratios were correlated with the percentage of viable cells, detected by eosin staining. These results suggested that α-T and δ-T are not homogeneously distributed in the semen fractions; in normal semen they are more concentrated in the sperm membrane, whereas in abnormal semen the damaged sperm cells may release both Ts in the plasma. To verify whether sperm membrane breakage could alter α-T and δ-T distribution between the seminal plasma and the spermatozoa, normal sperm samples were sonicated; after sonication a consistent sperm plasma membrane fragmentation, highlighted by transmission electron microscopy, and a concomitant release of α-T and δ-T were observed. In conclusion, the Ts coupled directly with the sperm membrane seem to play the main protective role in the semen, and the release of α-T and δ-T in the P fraction is probably an index of lower antioxidant power and sperm quality.

Sterols are essential components of the cell membrane lipid bilayer that include molecules such as cholesterol and desmosterol, which are significantly found in the spermatozoa of several animal species. However, the presence of desmosterol in rabbit semen has never been investigated. The aims of this study were to characterize the sterol composition of subfractions of ejaculated rabbit semen and evaluate the in vitro effects of sterol on the spermatozoa acrosome reaction and motility. Two sterols, occurring prevalently in the free form (94.3%), were identified in whole semen collected from 10 fertile New Zealand White rabbits, specifically desmosterol (58.5% of total sterols) and cholesterol (35.9% of total sterols). Desmosterol was the predominant sterol found in all subfractions of rabbit semen, varying from 56.7% (in the prostatic secretory granules, PSGs) to 63.8% (in the seminal plasma). Spermatozoa contained an intermediate proportion of desmosterol (59.8%), which was asymmetrically distributed between the heads (52.0% of the total content of sterols) and the tails (81.8%). Results showed that both desmosterol and cholesterol can be transferred from the PSGs to the spermatozoa and are equally effective in inhibiting in vitro spermatozoa capacitation at a concentration higher than 1 mg L(-1). In contrast, neither desmosterol nor cholesterol had a significant effect on spermatozoa motility. Thus, it was concluded that, the various fractions of rabbit seminal fluid differ from each other in sterol composition and quantity, probably due to their different functional properties, and these fractions may undergo significant sterol changes depending on the stage of spermatozoa capacitation.

The dose of warfarin needed to obtain a therapeutic anticoagulation level varies widely among patients and can undergo abrupt changes for unknown reasons. Drug interactions and genetic factors may partially explain these differences. Intestinal flora produces vitamin K2 (VK2) and patients with small intestinal bacterial overgrowth (SIBO) rarely present reduced INR values due to insufficient dietary vitamin K. The present study was undertaken to investigate whether SIBO occurrence may affect warfarin dose requirements in anticoagulated patients. Based on their mean weekly dose of warfarin while on stable anticoagulation, 3 groups of 10 patients each were defined: low dose (LD, or=70 mg/wk). Each patient underwent a lactulose breath test to diagnose SIBO. Plasma levels of warfarin and vitamin K-analogues were also assessed. Patients with an altered breath test were 50% in the VHD group, 10% in the HD group, and none in the LD group (P=0.01). Predisposing factors to SIBO were more frequent in the VHD group, while warfarin interfering variables were not. VHD patients were younger and had a higher plasma vitamin K1 (VK1) concentration (P>0.05). On the contrary, the plasma VK2 levels tended to be lower. This pilot study suggests that SIBO may increase a patient's warfarin dose requirement by increasing dietary VK1 absorption through the potentially damaged intestinal mucosa rather than increasing intestinal VK2 biosynthesis. The effect of Outdoor or Indoor housing systems on the growth, welfare and carcass and meat quality of a local rabbit population was investigated. The slaughter age was 103±2days. Open-field tests showed an effective capacity of the Outdoor group to combat stressors. Compared to Indoor rabbits, Outdoor rabbits showed better growth performance and higher slaughter weight (SW) (2535 vs 2137g; P<0.01). Outdoor housing conditions increased the physical activity of rabbits and their hind legs were more developed (36.1% vs 34.9%; P<0.01). Slaughter yield was lower in Outdoor rabbits (57.8% vs 58.4% SW; P<0.05) due to the higher skin proportion (17.2% vs 15.6% SW; P<0.05). Outdoor rabbit meat showed lower L∗ value (L. lumborum: 55.6 vs 59.2; P<0.01; B. femoris: 53.0 vs 55.5; P<0.01) and cooking loss (L. lumborum: 15.9% vs 18.1%; P<0.05). Outdoor rabbit hind leg meat was characterized by lower water (74.5% vs 75.1%; P<0.01) and higher protein (22.9% vs 22.6%; P<0.01) and fat (1.4% vs 1.1%; P<0.01) contents; lipids were lower in SFA and higher in MUFA. Outdoor rearing seems to be a possible alternative housing system that allays the ethical concerns of modern consumers while also providing good meat quality.

Metal compounds have been associated with male reproductive toxicity in vivo. The aim of the present study was to investigate the in vitro effects of 20 metal compounds using rabbit ejaculated spermatozoa as a study model for spermiotoxicity. Five of the metals tested (arsenic, cadmium, chromium, mercury and vanadium) reduced sperm motility and curvilinear velocity. Ultrastructural analyses revealed three types of damage to sperm head membranes in relation to the metal used: acrosome breakage with formation of various sized microvesicles (arsenic, cadmium, mercury and platinum); a large round hole (arsenic, cadmium and chromium), and numerous folds in the acrosome membrane (vanadium). The vanadium compound, followed by chromium and mercury compounds, determined a higher number of damaged spermatozoa. In conclusion, all the studied metal compounds, at levels higher than 1microM, may reduce sperm kinetic characteristics and probably fertilizing capacity by triggering specific morphological damages to the head and/or by inhibiting motility.

Review of literature has shown an increased rate of thrombotic complications in diabetic patients with frequent episodes of hyperketonemia. However, the mechanisms by which ketosis promotes vascular disease in diabetic patients are unclear. It was the aim of this study to investigate early changes in haemostatic parameters and oxidative stress markers during the hyperketonemic status which follows the interruption of continuous subcutaneous insulin infusion (CSII) in type I diabetic patients. Eight CSII-treated type I diabetic patients underwent a 4-hour pump arrest. Blood glucose, insulin and 3-hydroxybutirate were measured to verify the metabolic response. A vein-occlusive (VO) test was performed for the determination of tPA and PAI-1 activities and their antigen levels before and after the CSII arrest. Coagulation factor VII and VIII were evaluated by one-stage PT and PTT method, respectively. TF, vWF, tPA and PAI-1 antigens were determined by ELISA, whereas tPA and PAI-1 activities using chromogenic methods. Plasma malondialdehyde (MDA) and protein carbonyl groups (PCG) levels were determined by HPLC and spectrophotometry, respectively. After the insulin deprivation phase, post-VO tPA antigen level significantly decreased (P = 0.0391), whereas TF and post-VO PAI-1 activity and antigen levels significantly increased (P = 0.0156 and P = 0.0234, respectively). Plasma MDA and PCG levels were 1.88-fold and 1.74-fold higher than baseline values, respectively. In conclusion, the impairment of the fibrinolytic potential and the increases in TF, MDA and PCG levels may enhance the risk of both arterial and venous thrombosis during ketosis. Thus, early detection of hyperketonemia in DM patients could contribute to the prevention of life-threatening vascular events.

Several methods have been developed for measurement of plasma total homocysteine (tHcy), a proven graded risk factor for cardiovascular diseases. The aim of the study was to compare 3 commonly used clinical methods (enzyme immunoassay, Axis Biochemicals, Oslo, Norway; and fluorescence polarization immunoassay [FPIA], Axis, on the Abbott IMx System, Abbott Laboratories, Abbott Park, IL; and BioRad high-performance liquid chromatography [HPLC], Laboratories, Munich, Germany) for tHcy determination in samples from cardiovascular patients also undergoing methionine load measurement. Only HPLC reached the required coefficient of variation (<6%) for desirable performance for tHcy concentrations ranging between 0.68 and 5.41 mg/L (5 and 40 micromol/L). A higher variability was observed for postmethionine load vs fasting samples, which reached statistical significance for FPIA (P = .0013). The closest agreement between methods was observed between HPLC and FPIA for fasting tHcy concentrations (-0.23 mg/L [-1.69 micromol/L], -30% to 8%).Both immunoassays could be suitable alternatives for laboratories with high workloads when HPLC is not available. Differences among tHcy values must be taken into account when these methods will be used interchangeably for diagnosis and monitoring of treatment.
