Background and Objectives Water shortage and the need for its optimal use in arid and semi-arid regions, including Iran, has led water officials and farmers to use modern irrigation systems, such as drip irrigation with the aim of making optimal use of water resources. Drip irrigation has been welcomed in most parts of the world due to its high efficiency and the possibility of irrigation in different environmental conditions. The most important reason for the superiority of drip irrigation over other irrigation methods is the controllable amount of water for each plant. Drip irrigation is a method in which water is poured out of the net at low pressure through an orifice or device called an emitter and dripped into the bottom of the plant. This irrigation system, like other methods, requires accurate knowledge of the parameters affecting it to achieve the desired efficiency. One of the most important parameters for the irrigation system is the distribution of moisture in the soil and in fact the shape of the moist bulb. Therefore, knowledge of how to distribute water in the soil is essential for the proper design and management of subsurface drip irrigation systems. Since testing is very difficult and time consuming to detect the shape of moisture distribution in the soil, the use of numerical and analytical simulation can be an effective and efficient way to design these systems. Methodology In order to determine the progress of the moisture front in drip irrigation, first the soil texture type and physical properties of the soil were determined. It should be noted that the emitter flow rate was measured and adjusted in volume at the beginning of the test to minimize the difference between the emitter flows along the three side tubes. Evaluation experiments were performed with three outflows of 2, 4 and 6 liters per hour. With the start of the system, the progress of the moisture front at different times was measured by digging a trench using a scale. Numerical simulation of moisture front progress was performed using HYDRUS model based on Richard equation and analytical simulation was performed using Moment Analysis. HYDRUS software was used to numerically simulate the progress of the moisture front. The simulation range was considered to be 100 cm by 100 cm on the two-dimensional plane. In these simulations, 3956 nodes are used to represent the entire simulation range and also, relevant equations were used to calculate the two-dimensional spatial Moment of the wetting pattern . Findings The simulations show that the initial volumetric moisture content is 0.11 and the saturation volumetric moisture content is 0.380 and the water dispersion rate increases over time on the x and z axes. With increasing flow, the maximum dispersion is in the x-axis, which occurs in flow of 6 liters per hour. The result for flow of 6 liters per hour based on the data used is slightly higher than the desired value. The reason why the value of M00 in the flow rate of 6 liters per hour is higher than expected, is that in the simulation flow rate of 6 liters per hour change in the size of the inlet diameter and the amount of flux changes the amount of water entering the soil and moistens a large volume of soil. Due to the different amount of moisture applied to the area at different times, the value of 𝑧𝑐, 𝜎𝑥 2, 𝜎𝑧 2 is different and has caused a change in the size of the oval in different flows. The increase in the size of the ovals indicates the high dispersion of water in that area. The results showed that the Moment analysis was able to express the position of the center of mass of water distributed in the soil with correlation coefficient of 0.986 in linear mode and 0.982 in logarithmic mode. By comparing the values of diameter and depth obtained from the HYDRUS and the drawn ovals, it can be concluded that both methods provide close results. The accuracy of the Moment analysis method in simulating different types of moisture patterns resulting from drip irrigation under different flows with the use of different volumes of water is similar to the HYDRUS model and therefore it is possible to use this feature to predict the pattern of moisture from a certain flow using a specific volume of water . Conclusion In this study, the accuracy of Moment analysis in simulating various moisture patterns resulting from drip irrigation under different flows with the use of different volumes of water was investigated and the possibility of using this feature to predict the pattern of moisture from a given flow using a specific volume of water checked. In order to investigate the Moment of the amount of water distributed in the soil by subsurface drip irrigation, simulation was performed by two-dimensional HYDRUS software for three discharges of 2, 4 and 6 liters per hour with an inlet water volume of 12 liters. Then, using the results of simulation of moisture distribution range by a programming language including MATLAB software, and by calculating the Moments, it was determined that the Moments are able to express the position of the center of mass of water distributed in the soil and how it is distributed relative to x and z axes. The increase in the size of the ovals indicates that more water is distributed in that area. Comparing the diameters and depths of the moisture front between the simulated HYDRUS model and the Moment analysis model, it was found that the Moment analysis is an efficient way to study the distribution of water moisture by drip irrigation and this method can be used as an alternative input to estimate parameters. Moreover 31 urine samples were collected from healthy individuals and patients with other diseases such as malaria, brucellosis, hydatidosis and cutaneous leishmaniasis. Collected samples were examined to detect anti-Leishmania antibod-ies in urine, using ELISA and DAT."nResults: Anti-Leishmania antibody was detected in urine of 18 out of 30 (60%) VL patients by DAT while ELISA detected anti-Leishmania antibodies in urine of 28 out of 30 (93.3%) of VL cases. Sensitivity and specificity of urine-based DAT was 60% and 83.9%, respectively while sensitivity and specificity of urine-based ELISA were 93.3% and 93.5%, corre-spondingly. "nConclusion: Urine-based DAT and ELISA have a reasonable specificity and sensitivity in diagnosis of VL. Human toxocariasis, a worldwide parasitic disease, is caused by the larval stage of intestinal nematodes of dogs and cats, namely Toxocara canis and Toxocara cati. Human infection occurs by the accidental ingestion of embryonated eggs present in the soil, vegetables or on other contaminated surfaces, as well as via consumption of uncooked paratenic hosts, such as bird meat and giblets. The objective of this study was to evaluate the contamination of soil in public parks and playgrounds in Shiraz using microscopy and molecular methods. A total of 150 soil samples were collected from public parks and playgrounds in various areas of Shiraz, southern Iran. The samples were treated with saturated zinc sulphate solution, and Toxocara spp. eggs were detected by microscopic observation followed by nested polymerase chain reaction (PCR). To differentiate T. canis and T. cati eggs from each other, PCR restriction fragment length polymorphism (RFLP) of the internal transcribed spacer (ITS)-rDNA region by Sal I endonuclease enzyme was used. PCR-sequencing was performed to confirm the results of the PCR-RFLP method. Based on the flotation results of the 150 soil samples, six (4%) were found to be positive for Toxocara spp. eggs, whereas nested-PCR showed 24 samples to be positive (16%). Based on the PCR-RFLP method and the sequence of the ITS-rDNA region, a total of 23 out of 24 isolates were confirmed as T. cati and one out of 24 as T. canis. The results showed a higher number of soil samples to be positive for Toxocara by the molecular method than microscopy, and higher T. cati infection in soil samples, which could have an important role in human infection with toxocariasis in this region. The study was conducted to determine the sequence variation in two mitochondrial genes, namely cytochrome c oxidase 1 (pcox1) and NADH dehydrogenase 1 (pnad1) within and among isolates of Toxocara cati, Toxocara canis and Toxascaris leonina. Genomic DNA was extracted from 32 isolates of T. cati, 9 isolates of T. canis and 19 isolates of T. leonina collected from cats and dogs in different geographical areas of Iran. Mitochondrial genes were amplified by polymerase chain reaction (PCR) and sequenced. Sequence data were aligned using the BioEdit software and compared with published sequences in GenBank. Phylogenetic analysis was performed using Bayesian inference and maximum likelihood methods. Based on pairwise comparison, intra-species genetic diversity within Iranian isolates of T. cati, T. canis and T. leonina amounted to 0–2.3%, 0–1.3% and 0–1.0% for pcox1 and 0–2.0%, 0–1.7% and 0–2.6% for pnad1, respectively. Inter-species sequence variation among the three ascaridoid nematodes was significantly higher, being 9.5–16.6% for pcox1 and 11.9–26.7% for pnad1. Sequence and phylogenetic analysis of the pcox1 and pnad1 genes indicated that there is significant genetic diversity within and among isolates of T. cati, T. canis and T. leonina from different areas of Iran, and these genes can be used for studying genetic variation of ascaridoid nematodes. Background: The sensitivity of parasitological and molecular methods is unsatisfactory for the diagnosis of strongyloidiasis, and serological techniques are remaining as the most effective diagnostic approach. The present study aimed to design and produce a chimeric recombinant antigen from Strongyloides stercoralis immunoreactive antigen (SsIR) and Ss1a antigens, using immune-informatics approaches, and evaluated its diagnostic performance in an ELISA system for the diagnosis of human strongyloidiasis., Methodology/principal Findings: The coding sequences for SsIR and Ss1a were selected from GenBank and were gene-optimized. Using bioinformatics analysis, the regions with the highest antigenicity that did not overlap with other parasite antigens were selected. The chimeric recombinant antigen SsIR- Ss1a, was constructed. The solubility and physicochemical properties of the designed construct were analyzed and its tertiary structures were built and evaluated. The construct was expressed into the pET-23a (+) expression vector and the optimized DNA sequences of SsIR-Ss1a (873 bp) were cloned into competent E. coli DH5α cells. Diagnostic performances of the produced recombinant antigen, along with a commercial kit were evaluated in an indirect ELISA system, using a panel of sera from strongyloidiasis patients and controls. The physicochemical and bioinformatics evaluations revealed that the designed chimeric construct is soluble, has a molecular with of 35 KDa, and is antigenic. Western blotting confirmed the immunoreactivity of the produced chimeric recombinant antigen with the sera of strongyloidiasis patients. The sensitivity and specificity of the indirect ELISA system, using the produced SsIR-Ss1a chimeric antigen, were found to be 93.94% (95% CI, 0.803 to 0.989) and 97.22% (95% CI, 0.921 to 0.992) respectively., Conclusions/significance: The preliminary findings of this study suggest that the produced SsIR-Ss1a chimeric antigen shows promise in the diagnosis of human strongyloidiasis. However, these results are based on a limited panel of samples, and further research with a larger sample size is necessary to confirm its accuracy. Cystic echinococcosis (CE), is one of the common parasitic infections of domestic animals and humans caused by the larval stages of the tapeworm Echinococcus granulosus . This research aimed to identify of CE cysts in cattle isolates from Shiraz slaughterhouse using molecular technique. Thirty CE cysts isolates were collected and total DNA was extracted from protoscolices or the germinal layer of each cyst followed by polymerase chain reaction (PCR) using cytochrome c oxidase I (cox1) and NADH dehydrogenase subunit 1 (NADH-1) mitochondrial genes (~ 450 bp) and sequencing. The sequence results were compared with available reference sequences in GenBank by BLAST system. Phylogenetic analysis was performed using MEGA 5.0 software and the maximum likelihood method. Background: This study aimed to determine the therapeutic efficacy of curcumin nanoemulsion (CUR-NE) in mice infected with Echinococcus granulosus sensu stricto protoscoleces., Methods: Forty-two inbred BALB/c mice were divided into seven groups of six animals each. Six groups were inoculated intra-peritoneally with 1500 viable E. granulosus protoscoleces, followed for six months and used as infected groups. The infected groups were named as: CEI1 to CEI6 accordingly. The 7th group was not inoculated and was named cystic echinococcosis noninfected group (CENI7). CEI1 and CEI2 groups received 40 mg/kg/day and 20 mg/kg/day curcumin nanoemulsion (CUR-NE), respectively. CEI3 received nanoemulsion without curcumin (NE-no CUR), CEI4 received curcumin suspension (CUR-S) 40 mg/kg/day, CEI5 received albendazole 150 mg/kg/day and CEI6 received sterile phosphate-buffered saline (PBS). CENI7 group received CUR-NE 40 mg/kg/day. Drugs administration was started after six months post-inoculations of protoscoleces and continued for 60 days in all groups. The secondary CE cyst area was evaluated by computed tomography (CT) scan for each mouse before treatment and on the days 30 and 60 post-treatment. The CT scan measurement results were compared before and after treatment. After the euthanasia of the mice on the 60th day, the cyst area was also measured after autopsy and, the histopathological changes of the secondary cysts for each group were observed. The therapeutic efficacy of CUR-NE in infected groups was evaluated by two methods: CT scan and autopsied cyst measurements., Results: Septal calcification in three groups of infected mice (CEI1, CEI2, and CEI4) was revealed by CT scan. The therapeutic efficacy of CUR-NE 40 mg/kg/day (CEI1 group) was 24.6 ± 26.89% by CT scan measurement and 55.16 ± 32.37% by autopsied cysts measurements. The extensive destructive effects of CUR-NE 40 mg/kg/day (CEI1 group) on the wall layers of secondary CE cysts were confirmed by histopathology., Conclusion: The current study demonstrated a significant therapeutic effect of CUR-NE (40 mg/kg/day) on secondary CE cysts in BALB/c mice. An apparent septal calcification of several cysts revealed by CT scan and the destructive effect on CE cysts observed in histopathology are two critical key factors that suggest curcumin nanoemulsion could be a potential treatment for cystic echinococcosis., (© 2024. Background: Toxocariasis is a zoonotic disease caused by the larvae of Toxocara canis or Toxocara cati . Patients with schizophrenia may be at higher risk of infection, possibly due to their cognitive and personal self-care impairments. We aimed to assess the association between Toxocara spp. exposure and schizophrenia., Methods: This case-control study was conducted on 109 patients with schizophrenia admitted to Ibn Sina Hospital, Shiraz, Iran, and 104 age- and gender-matched healthy controls from May to September 2021. A questionnaire was obtained and serum samples were tested for IgG antibodies to Toxocara excretory/secretory (TES) antigens using an enzyme-linked immunosorbent assay (ELISA)., Results: Anti- Toxocara IgG was detected in 12 schizophrenic patients and 10 control subjects, giving respective seroprevalences of 11.0% (95% confidence interval [95% CI]=5.8-18.4%) and 9.6% (95% CI=4.7-17.0%). Univariate logistic analyses estimated an odds ratio (OR) of 1.16 (95% CI=0.44-3.16); however, it was not statistically significant ( P= 0.915). Individuals with a history of eating unwashed vegetables or fruits (23.1%, 95% CI=9.0-43.6, crude odds ratio [COR]=3.21, 95% CI=1.13-9.13) and rural residency (19.5%, 95% CI=8.8-34.9, COR=2.74, 95% CI=1.06-7.05) had significantly higher rates of seropositivity using the univariate logistic analyses. After multivariate logistic analyses, the differences were not statistically significant., Conclusion: The toxocariasis seroprevalence among schizophrenic and healthy participants was not significantly different (11% vs. 9.6%). Background: Toxocara canis is one of the most important causes of animal toxocariasis with global distribution. We aimed to find out the seroprevalence of toxocariasis in dogs in a rural area in Fars Province, south of Iran., Methods: Dogs blood samples were collected from 60 dogs in three rural areas in the Sar Mashhad region, Fars Province. Dog sera were evaluated for anti- Toxocara antibodies by an indirect ELISA method. The association between the seropositivity and age, gender, and the sampling location were statistically evaluated., Results: Serological assay detected anti- Toxocara antibodies in sera of 32 out of 60 dogs, corresponding to a seroprevalence of 53.3%. The rate of seropositivity was higher in the male dogs. The rate of seropositivity was higher in old dogs. This rate increased with increasing age, however, the association between age and Toxocara seropositivity was not statistically significant., Conclusion: The high prevalence of Toxocara infection in dogs in the current study area confirms that infected dogs are an important source of Toxocara infection for their owners and people who are in close contact with these animals, especially children., Competing Interests: Conflicts of interest None to declare., (Copyright© Pouryousef et al. Background: Toxocariasis is an important zoonotic infection, especially in tropical areas. One of the significant challenges in the serodiagnosis of human toxocariasis is the cross-reaction of Toxocara antigens with other parasites due to their relatively similar glycan structures. Removing the glycan structure from Toxocara excretory-secretory (TES) antigens may increase the efficacy of these antigens in the diagnosis of toxocariasis. The current study aimed to assess the efficacy of deglycosylated Toxocara cati excretory-secretory (dTES) antigens for the serodiagnosis of human toxocariasis., Methods: Toxocara ES antigens were prepared from T. cati second-stage larvae and deglycosylated using sodium hydroxide (NaOH). The TES antigens, along with the dTES antigens, were used in an ELISA as well as a western blotting system for the detection of anti- Toxocara antibodies. Sera samples collected from 30 confirmed cases of toxocariasis, 30 patients with other diseases, and 30 healthy subjects were evaluated by both systems., Results: The sensitivity of TES and dTES ELISA for the diagnosis of human toxocariasis was 96.67% (95% CI = 82.78-99.92) and 93.33% (95% CI = 77.93-99.18), respectively, while the specificity of dTES (88.33%; 95% CI = 77.43-95.18) increased significantly compared to the TES (80.00%; 95% CI = 67.67-89.22). The sensitivity of both antigens was 100% (95% CI = 88.43-100) by the western blotting system. Moreover, the specificity of TES and dTES antigens was 95% (95% CI = 86.08-98.96) and 98.33% (95% CI = 91.06-99.96), respectively, when using the western blotting system., Conclusion: Results of the current study indicate that the chemical removal of the glycan epitopes of T. cati ES antigens significantly reduces cross-reactivity rates with other parasitic infections. Although the appendix is the most commonly resected and examined intra-abdominal organ, the pathogenesis and etiology of acute nonspecific appendicitis remain enigmatic. This retrospective study aimed to assess the prevalence of parasitic infection in surgically removed appendices and to evaluate the probable associations between the presence of parasites and the occurrence of appendicitis through parasitological and histopathological examinations of the appendectomy specimens., Materials and Methods: This retrospective study was carried out from April 2016 to March 2021 among all patients referred to hospitals affiliated with Shiraz University of Medical Sciences, Fars Province, Iran, for appendectomy. Patient information, including age, sex, year of appendectomy, and type of appendicitis, was collected from the available data in the hospital information system database. In positive cases, all pathology reports were retrospectively evaluated for the presence of the parasite and its type, and analytical and descriptive statistics were carried out using SPSS software version 22., Results: A total of 7628 appendectomy materials were evaluated in the present study. Of the total participants, 4528 (59.4%, 95% CI: 58.2-60.5) were males, and 3100 (40.6%, 95% CI: 39.5-41.8) were females. The mean age of participants was 23.87±14.28 years. Overall, Enterobius vermicularis was observed in 20 appendectomy specimens. Fourteen of these patients (70%) were less than 20 years old., Conclusion: This study indicated that E. vermicularis is one of the common infectious agents that could be found in the appendix and may increase the risk of appendicitis. Toxocariasis as a common neglected disease is the culprit of infecting all age groups. The current cross-sectional study was designed to assess the seroprevalence of toxocariasis and risk factors associated with seropositivity of Toxocara infection among the general population of adults in the Kavar district, south of Iran. A total of 1060 participants with an age range of 35 to 70 years from the Kavar region entered the study. Manual ELISA was utilized to detect anti- Toxocara -specific antibodies in their serum samples. In addition, demographic information as well as risk factors related to toxocariasis was collected from individuals who participated in the survey. The mean age of the participants was 48.9 (±7.9) years old. Out of 1060 subjects, 532 (50.2%) were men, and 528 (49.8%) were women. The overall Toxocara seroprevalence was 5.8% (61/1060). The prevalence of Toxocara seropositive cases was significantly different between males and females ( p  = 0.023). The seropositive rate for Toxocara infection was also significantly higher in housewives ( p  = 0.003) and subjects with learning disabilities ( p  = 0.008). Multivariable logistic regression revealed that housewives (OR = 2.04, 95% CI: 1.18-3.51, p  = 0.010) and subjects with learning disability (OR = 3.32, 95% CI: 1.29-8.52, p  = 0.013) were at increased risk of Toxocara infection. The findings of the current study depicted a noticeable seroprevalence of Toxocara infection in the general population in the Kavar district, southern Iran. An increased risk of toxocariasis has been associated with learning disabilities and being a housewife. All of the toxocariasis-positive cases had contact with animals, at some point in their life. Cutaneous leishmaniasis (CL) is one of the most important parasitic diseases in the world. Despite the existence of many therapeutic strategies, the treatment of this infection still faces problems. Sodium chlorite as an antimicrobial agent has been shown to have acceptable tissue regenerative and wound healing properties. Therefore, the present study aimed to analyze the in vitro effects of different concentrations of sodium chlorite on Leishmania major promastigotes and macrophage cells. The inhibitory and toxicity effect of various concentrations (0.0035, - 1.8 mg/ml) of sodium chlorite on the standard Iranian strain of L. major promastigotes were evaluated via counting the cells and flow cytometry. Furthermore, cytotoxicity on promastigotes and J774 macrophage cell line were performed by MTT assay. The results of the inhibitory test demonstrated that sodium chlorite had dose-dependent, anti-leishmanial activities. The half-maximal inhibitory concentration (IC 50 ) for promastigotes and J774 cells by cytotoxicity test was detected at 0.17 mg/ml and 0.08 mg/ml after 48 h respectively. Flow cytometry results showed that 27.34% death of promastigotes was observed in 0.0035 mg/ml of sodium chlorite and 78.12% in 1.8 mg/ml. The results of the present study showed that sodium chlorite could be used as an effective treatment for CL, especially in cases resistant to treatment with pentavalent compounds. Toxocariasis is caused by infection with the nematode species Toxocara canis and Toxocara cati. Serological methods using eggs, larvae and adult worms of Toxocara spp. as antigen have been used for the diagnosis of human toxocariasis. The current study aimed to evaluate indirect immunofluorescence assay (IFA) using embryonated eggs of Toxocara for diagnosis of human toxocariasis. A total of 58 sera including twenty sera from patients with toxocariasis, 20 from healthy persons and 18 from patients with other parasitic infections were collected and used for the study. The embryonated eggs of Toxocara were prepared as antigen. Indirect immunofluorescence assay was performed using the frozen section of uterus containing embryonated T. canis eggs and unembryonated T. cati eggs. All serum samples had a positive reaction using IFA. The eggs of Toxocara as antigen exposed to the serum samples of toxocariasis, other parasitic infections and healthy persons, followed by IFA gave a bright greenish-yellow fluorescence. A number of samples such as eggs of Toxocara, Toxascaris, Trichuris and strongyloides larvae, and adult worm of Ancylostoma exhibited the bright greenish-yellow autofluorescence under fluorescent microscope. Intestinal parasitic infections (IPIs) are still a serious public health problem worldwide, particularly in developing countries. The COVID-19 pandemic has affected people's lifestyles and quality of life. Besides, the prevalence of IPIs is directly associated with environmental sanitation, overcrowding, and personal hygiene. Therefore, the current study aimed to determine the prevalence of IPIs among patients referred to hospitals affiliated to Shiraz University of Medical Sciences by reviewing the available data of hospital information system database in the pre- and post-COVID-19 pandemic. The total data of 13,686 patients referred to hospitals affiliated to Shiraz University of Medical Sciences, southern Iran were collected from March 2019 to March 2021. The overall prevalence of IPIs was found to be 4.4%, and Blastocystis spp. was the most common parasite. The prevalence of protozoan parasites (4.4%) was significantly higher than helminthic parasites (0.04%, P < 0.001). A significant association was observed between IPIs with age, gender, and year (P < 0.05). The prevalence of IPIs among those referred to hospitals before the COVID-19 pandemic was higher than those referred to hospitals during the COVID-19 pandemic (5.8% vs 2.8%), and the differences were statistically significant (P < 0.001). The odds of infection among people investigated during the COVID-19 pandemic was about 40% lower than those investigated before the COVID-19 pandemic (AOR: 0.60, 95% CI: 0.49-0.73, P < 0.001). Our results showed that the prevalence of IPIs has decreased during the COVID-19 pandemic. Background: Parkinson's disease (PD) has been described in dopamine brain level reductions. Conversely, several studies have shown that Toxoplasma parasite can increase the level of dopamine in an infected host. This study was conducted to assess the serum, cerebral dopamine levels, and downregulation of Parkinson's disease manifestations in mice with chronic toxoplasmosis., Methods: PD induction was done by oral inoculation of rotenone into BALB/c mice. To induce the chronic infection, cysts of T. gondii Prugniaud strain (genotype II) were injected intraperitoneally into the mice. The rotarod test was used for the evaluation of functional motor disorders in experimental mice. The serum and cerebral dopamine levels of the mice were also measured by using high-performance liquid chromatography (HPLC) on consecutive periods (10 days)., Results: Findings of the rotarod test showed the highest and lowest average of running duration belonged to the uninfected mice and PD mice, respectively. Remarkably, the running duration of infected mice with PD was higher than PD mice. As well, the level of serum and cerebral dopamine increased in mice with PD and toxoplasmosis in comparison with PD mice., Conclusion: Increasing the serum and cerebral dopamine levels in mice infected with toxoplasmosis is related to the presence of the parasite. Background and Aim: Parasitology course is one of the basic science courses in the educational curriculum for medical students. Since the integration of basic and clinical sciences has helped students better understand the basic science course content, the aim of the present study was to determine the effect of integration of basic and clinical aspects of a specific topic in a parasitology course on medical students learning., Materials and Methods: A randomized controlled trial was conducted on 110 undergraduate fifth-semester medical students from April to July 2018. The students were randomly divided into two groups, based on student identification number: Intervention and control groups. The topic selected for this study from the parasitology course was "cutaneous leishmaniasis." At the beginning of the program, a dermatologist presented the clinical aspects of the topic to the intervention group. Then, a parasitologist delivered a traditional lecture about the basic aspects of the topic to both groups. The students' scores on questions related to cutaneous leishmaniasis in the final exam were used as a measure of learning and teaching outcomes. A questionnaire that consisted of seven items and three open-ended questions was used based on the objectives of the randomized controlled trial. Statistical analysis was performed by SPSS software., Results: Based on the result of the final examination, there was no significant difference in the learning rate of students between the intervention and control groups ( P ≥ 0.05). According to students' comments, the teaching of clinical science alongside basic science increased the importance of the topic and the students' interest in basic science. Most students agreed that this method prepares them for their clinical years., Conclusion: Many medical students believe that the content of many basic science courses are forgotten in the future. Background: Human toxocariasis is caused by Toxocara canis and T. cati , the nematodes in the intestine of dogs and cats, respectively. Since the association between asthma and toxocariasis is controversial, the aim of the present study was to investigate the seroprevalence of Toxocara infection among asthmatic children in comparison with healthy children., Methods: This case-control study was conducted on 92 asthmatic and 91 healthy children aged 1-16 years old in Shiraz City, Southern Iran in 2019-2020. The serum samples were tested for IgG anti- Toxocara antibodies by ELISA method using the T. canis larval excretory-secretory (E/S) antigens. The collected data were analyzed using SPSS software., Results: The seroprevalence of toxocariasis in asthmatic patients was higher than the healthy children with no significant difference in Toxocara seropositivity between two groups (9.8% vs 8.8%, P = 0.817). The association between Toxocara infection and variables such as gender and age were not statistically significant., Conclusion: There was no significant association between toxocariasis and childhood asthma. Background: Toxoplasma parasite alters the transduction of neurotransmitter signals and leads to changes in the level of brain neurotransmitters including tyrosine and dopamine, so behavior changes can occur in infected hosts. Based on this concept, this study was conducted for evaluation of the tyrosine and dopamine serum levels in infected mice with chronic toxoplasmosis., Materials and Methods: Toxoplasma gondii (Prugniaud strain II) was injected intraperitoneally into BALB/c mice to induce chronic toxoplasmosis. Modified agglutination test (MAT), polymerase chain reaction (PCR), and microscopic methods were conducted to confirm the induction of chronic toxoplasmosis. The infected mouse sera were separated at days 40, 50, 60, 70, and 80 for evaluation of tyrosine and dopamine serum levels using high-performance liquid chromatography (HPLC) ., Results: Microscopic methods confirmed the formation of the Toxoplasma cysts in mouse tissues. Inducing chronic toxoplasmosis is also confirmed by MAT, PCR, and histological methods. HPLC results indicated a decrease in serum tyrosine level at day 40 in infected mice in comparison to control, and the levels were too low to be measured at other times. However, a significantly high serum dopamine level was observed that gradually increased after parasite inoculation., Conclusions: No detection of tyrosine level in most of the sample groups is probably related to the very low concentration of tyrosine in sera. Mentally retarded individuals are more likely to become infected with soil-transmitted infections including toxocariasis. The current study aimed to determine the serostatus of toxocariasis among institutionalized mentally retarded individuals in Hormozgan Province, in the south of Iran. Subjects of the study were 117 mentally disabled individuals, including children and adults, maintained in a charity-based institution. Three millilitres of venous blood was taken from each subject. While sampling, demographic features of the subject were documented in a questionnaire. An ELISA system based on Toxocara larvae excretory-secretory antigens (TES) was utilized to detect anti- Toxocara IgG antibodies in the sera of the patients. The mean age of the subject was 27.6 (±12.31) years and consisted of 55 (47%) males and 62 (53%) females. Out of 117 cases, 33 (28.2%) were seropositive for toxocariasis. The seroprevalence was higher in females (37.1%) than males (18.2%), and the difference was statistically significant ( p  < 0.05). Out of 117 subjects, 76 (64.9%) had severe and 41 (35.1%) had profound mental retardation. Anti- Toxocara antibodies were detected in 18 (23.7%) patients with severe and 15 (35.6%) patients with profound mental retardation. The highest seroprevalence rate of toxocariasis (44.4%) was observed in the age group of ≤10 years followed by 21-30 years old (36.7%). No statistically significant association was seen between the age and also the duration of residency in the care center and seropositivity of toxocariasis ( p  > 0.05). The current study represents the high prevalence of toxocariasis in mentally retarded patients in Hormozgan Province, southern Iran. Regarding preparing and serving foods, food handlers have an influential role in the spreading of foodborne infections. Due to the great potential to cause foodborne infections, intestinal parasites (IPs) are considered a significant public health problem in today's societies. In Iran, despite several regional reports, national data on IPs prevalence in food handlers are lacking. The aim of the present study, therefore, is to estimate the pooled prevalence of IPs infections and associated pooled odds ratio of educational level among food handlers in Iran. PubMed, Web of Science, Scopus, Embase, and Google Scholar databases (international) and SID and Magiran databases (national) were systematically searched for studies that reporting the prevalence of IPs in food handlers in Iran, published between 2000 and 2020. Pooled prevalence was estimated using a random-effects model with a 95% confidence interval (CI) and depicted as a forest plot, while heterogeneity was evaluated using Cochran's Q-test. The overall pooled prevalence estimate for IPs was 19.3% (95% CI = 14.9-23.7%). Prevalence of the protozoan parasites (20%, 95% CI = 13.7-26.3%) was significantly higher than that of the helminthic parasites (1.6%, 95% CI = 1.1-2.0%). Giardia lamblia (5.2%), Entamoeba coli (5.0%), and Blastocystis spp. (4.4%), as protozoan parasites, and Ascaris lumbricoides (1.4%), Enterobius vermicularis (0.9%), and Hymenolepis nana (0.5%), as helminthic parasites, were the most commonly reported species. Food handlers with low educational levels were 20% more exposed to IPs infection, compared to those with high educational levels (OR = 1.21, 95% CI = 0.88 to 1.65). The prevalence of IPs infections among food handlers in Iran is significant. Background: Baylisascaris devosi is an intestinal nematode found in several carnivores including fisher, wolverine, Beech marten, American marten and sable in different parts of the world, but this nematode has not been reported from Pine marten. Therefore, this study aimed to identify Baylisascaris isolated from a Pine marten in Iran using morphological and molecular approaches., Methods: Specimens of B. devosi were collected from one road-killed Pine marten in northern Iran. Morphological features were evaluated using scanning electron microscopy, energy dispersive x-ray analysis and ion sectioning. The molecular characterization was carried out using partial Cox1, LSU rDNA and ITS-rDNA genes., Results: The nematodes isolated from the Pine marten were confirmed to be B. devosi based on the morphological features and the sequence of ribosomal and mitochondrial loci. X-ray scans (EDAX) were completed on gallium cut structures (papillae, eggs, male spike and mouth denticles) of B. devosi using a dual-beam scanning electron microscope. The male spike and mouth denticles had a high level of hardening elements (Ca, P, S), helping to explain the chemical nature and morphology of the worm. Based on these genetic marker analyses, our sequence had the greatest similarity with Russian B. devosi isolated from sable., Conclusions: In this study, to our knowledge, the occurrence of B. devosi infection in Pine marten is reported for the first time. Molecular analysis showed that these three genes are suitable molecular markers for identification and inferring phylogenetic relationships of Baylisascaris species. Furthermore, the high divergence of Cox1 between Baylisascaris species indicates that Cox1 could be used for their phylogenetic and taxonomic studies.

The most important pathogenesis factor in the Apicomplexa parasites is invasion to the host cell. Given the inhibitory role of Butanedione Monoxime (BDM) on myosin-actin interaction, this study aimed to investigate the effects of this molecule on the vitality and infectivity of Toxoplasma tachyzoites in order to provide a new option for vaccine development. The tachyzoites of the RH strain of Toxoplasma gondii were exposed to different concentrations (1, 2, 4, 8, 16, 32, 64, and 128 μg/mL) of BDM, and mortality effect was assessed by flow cytometry. Then, the penetration ability of the tachyzoites was investigated in HeLa and macrophage cell lines. The infectivity of exposed tachyzoites to BDM were also investigated in mice through following up and detecting the etiological factor. The highest percentage of mortality (72.69%) was seen in the tachyzoites exposed to 128 μg/mL of the compound. The tachyzoites exposed to 32, 64, and 128 μg/mL of BDM began the proliferation in HeLa cells after 48 h, while this proliferation was initiated within 24 h in macrophage cells. All the mice inoculated with the BDM-treated tachyzoites died after 13 days. The mean survival time of the mice receiving tachyzoites exposed to 128 μg/mL of BDM was 12.4 days, which was significantly different from the negative control group ( p  = 0.001). BDM, as the inhibitor of myosin-actin interaction, and other substances that block the entry of parasites into cells may be suitable candidates for vaccine production against Toxoplasma . Background: Blastocystis is a unicellular protozoan and one of the most common parasites found in humans and many animals' intestinal tract. The present study aimed to compare the genotypes of Blastocystis infecting cattle and humans in the south of Iran., Methods: A total of 100 human stool samples and 75 cattle stool samples were microscopically examined for Blastocystis infection. DNA was extracted from thirty-eight microscopically positive samples (13 humans and 25 cattle). PCR was performed on positive samples targeting the Blastocystis-specific SSU rDNA gene. PCR products of eight humans and eleven cattle samples were sequenced and compared with available reference sequences in GenBank by BLAST queries. Genetic diversity was measured for Blastocystis subtypes in human and cattle, based on haplotype and nucleotide diversities., Results: The PCR detected Blastocystis in ten humans and twenty-four cattle samples. Blastocystis subtypes 1, 2, and 6 were found in humans whereas subtypes 5 and 10 were found in cattle. Subtype (ST) 2 was the most predominant subtypes in humans whereas, in cattle specimens, the ST5 was the most dominant subtype. Based on the Blastocystis sequences of SSU rDNA, 68 sites were polymorphic and 49 sites were parsimony informative, resulting in the identification of 15 haplotypes, 10 haplotypes in the cattle and 5 in humans. No haplotype was shared between cattle and human parasites., Conclusion: Human-derived Blastocystis subtypes were different from cattle subtypes in southern Iran. Background: Human toxocariasis is caused by the larval stage of Toxocara canis and Toxocara cati. This cross-sectional study was conducted to evaluate the seroprevalence and the risk factors associated with Toxocara infection in nomads of Boyer-Ahmad County in the southwest of Iran., Methods: A total of 968 blood samples were collected from nomads. A structured questionnaire, containing sociodemographic data, was completed for each participant. Sera were evaluated for anti-Toxocara antibodies, using the T. canis excretory-secretory antigens prepared from the second stage larvae, in an ELISA system., Results: Among the 968 participants, 220 (22.7%) were males and 748 (77.3%) were females; the mean age was 40.71 y. Anti-Toxocara IgG antibodies were detected in 14 (1.4%) of the 968 nomads. The association between Toxocara infection and gender, age, contact with dogs, residential area and level of education were not statistically significant (p>0.05)., Conclusions: The findings of the study revealed a low prevalence rate of toxocariasis in nomadic communities in southwest Iran. Balantidium coli is a common parasite of pig and wild boars (Sus scrofa) which can infect humans and several species of mammals. This study aimed to determine the genotype of Balantidium isolated from Eurasian wild boars in Bushehr province, Southwestern Iran. Twenty-five faecal samples, originating from 25 wild boars captivated in our previous study, were processed. DNA was extracted from the faecal samples and PCR-amplified, targeting an ITS1-5.8s-rRNA-ITS2 region of Balantidium genome. PCR product was purified from the gel, and sequenced. BLAST analysis was performed in order to compare our isolates with other previously reported ones. A phylogenetic tree was constructed, using MegaX software, to find out the phylogenetic diversity of the isolates. With PCR it was possible to detect Balantidium DNA in the faecal samples of 13 out of 25 (52%) of the wild boars. BLAST analysis of seven isolates revealed that the isolates belong to the newly introduced genus Neobalantidium coli. Sequences of three isolates were deposited in the GenBank. Moreover, molecular analysis revealed six areas of nucleotide differences within the isolates and nine areas of difference between the sequences obtained in this study and those available in the GenBank. Phylogenetic analysis revealed that the sequences of isolates of this study have up to 2.2% dissimilarity from those published in the GenBank. Background: Given the lack of routine screening and the high prevalence of toxoplasmosis in pregnant women in Iran, the current study aimed to find out the rate and features of Toxoplasma gondii infection in the spontaneously aborted human fetuses in Kohgiluyeh and Boyer-Ahmad Province, Southwestern Iran., Methods: This cross-sectional study was performed on 100 spontaneously aborted fetuses' tissues and their mother blood samples. The mothers' sera were evaluated for anti-Toxoplasma antibodies while their buffy coat and aborted fetuses tissues were evaluated for Toxoplasma DNA. PCR product at GRA6 locus was sequenced and phylogenetic analysis was done. Likewise, quantitative Real-Time PCR was performed to find out the parasite burdens in mothers buffy coat and fetuses tissues., Results: Using serological method, anti-Toxoplasma IgG and IgM antibodies were detected in 7 (7%) and 3 (3%) out of 100 sera from women with spontaneous abortion. Real-time PCR method detected T. gondii DNA in the buffy coat of one seronegative and 2 (out of 3) IgM seropositive cases. None of the samples from aborted fetuses were infected with T. gondii. BLAST and phylogenetic analysis showed that the sequenced isolates belonged to type I of T. gondii and two identified T. gondii isolates were taxonomically grouped into one clade., Conclusion: Our findings revealed type I genotype of T. gondii in two mothers with spontaneous abortion, without fetus involvement. Background: The prevalence of fascioliasis in a population of cattle in a new-emerging focus of human fascioliasis in Boyer-Ahmad District, in the southwest of Iran, was investigated, using an ELISA system and coprological methods., Methods: Blood, as well as stool samples, were collected from 150 dairy cattle consisted of 82 (54.7%) males and 68 (45.3%) females, aged from 1 to 8 years, selected from different pastures. The stool samples were evaluated by direct wet mount microscopic examination as well as sedimentation methods. Moreover, modified Telemann was used for detecting of Fasciola eggs. Anti-Fasciola antibodies in the cattle sera samples were determined, using a Fasciola excretory-secretory antigen-specific ELISA., Results: Anti-Fasciola antibodies were detected in 64 (42.4%) of the subjects by ELISA. Out of 64 seropositive cases, 29 (45.3%) were female and 35 (54.6%) were male. No significant association was found between sex and seropositivity to fascioliasis. Considering the age of the cattle, the highest prevalence of fascioliasis (23%) was found in the 5-year old age group. However, the difference between the age of the cattle and seropositivity to fascioliasis was not statistically significant (P > 0.05). Fasciola eggs were not detected in fecal samples of any of the studied cattle, while Paramphistomide, as well as Dicrocoelium dendriticum eggs, were detected in 6 (4%) and 17 (11.3%) of the subjects, respectively., Conclusion: Findings of the current study provide basic information about bovine fascioliasis in a new focus of human fascioliasis in Iran, which is needed for effective control of this parasitic infection in such areas., (Copyright © 2019 Elsevier Ltd. Objective: Echinococcus granulosus contains a complex of different strains that represent diversity in the pattern of the life cycle and also their host types. So far 10 genotypes of this parasite have been identified, using molecular methods. The current study aimed to evaluate and compare the genotypic diversity of E. granulosus metacestodes from livestock of Turkey and Iran., Methods: A total of 90 livestock liver and lung organs infected with hydatid cyst from industrial slaughterhouses of Bonab Province in the East Azerbaijan Province in Iran (60 samples, including 30 sheep and 30 cattle) and Van Province in Turkey (30 samples, including 15 sheep and 15 cattle) were collected. DNA was extracted from the protoscolices or germinal layers and polymerase chain reaction (PCR) were utilized, targeting the partial mitochondrial cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase 1 (nad1) genes. PCR products were isolated from the electrophoresis gels and sequenced. The sequences were compared with each other, as well as with those related available sequences in the GenBank, using the BioEdit software and the BLAST algorithm. Finally, the phylogenetic trees were constructed by comparing sequences of cox1 and nad1 fragments, using the MEGA7 software and the maximum likelihood method., Results: All samples sequenced from Iran corresponded to the genotype G1 (100%). Among the samples from Turkey, 15 samples (78.9%) were identified as G1 while only one sample (5.3%) corresponded to the genotype G3 and 3 isolates (15.8%) were defined as genotypes G1/G3. Five distinct haplotypes were determined within the examined isolates from sheep and cattle in both countries and all isolates clustered in one group. Phylogenetic analysis revealed that the intra-species genetic variations were 0.0-0.6% and 0.0-1.4% for cox1 and nad1, respectively., Conclusion: The dominant genotype of E. granulosus sensu stricto of livestock in both countries was the G1 (sheep strain) genotype. Our findings indicate that the sheep-dog cycle is the leading cycle of E. granulosus in these two areas. Hence, adopting regional common policies and bilateral cooperation helps to control the disease in livestock as well as in human in these two regions. Further study is required to compare the genetic diversity of human isolates of E. granulosus in these two countries.

The current study was performed to find out the seroprevalence of Toxocara infection in children living in a rural community in Fars province, southern Iran. Venous blood was taken from 617 children and evaluated for anti-Toxocara antibodies, using an ELISA system. Of the 617 studied children, 318 (51.5%) were boys and 299 (48.5%) were girls. Mean age of the participants was 9.2 (±10.7) years. Most cases of the recruited subjects (37.4%) were in the age group of 0-5 years. Anti-Toxocara antibodies were detected in sera of 39 (6.3%) of children. From these, 23 (62.2%) were boys and 14 (37.8%) were girls. The infection rate was almost the same in different age groups. No statistically significant differences were seen between seropositivity to Toxocara infection and gender or age of the participants. Dogs or cats ownership was not associated with Toxocara seropositivity. The adjusted associations of the study variables suggested that the visceral leishmaniasis (VL) seropositivity is significantly associated with the seroprevalence of Toxocara infection (P < 0.001). Findings of the current study revealed that Toxocara infection is a common infection among children in the studied rural community in Iran. Preventive measurements are necessary to minimize the rate of Toxocara infection in children in such communities., (© 2018 John Wiley & Sons Ltd.)

A single-step synthesis of super-water-repellent oil sorbents based on cellulose acetate (CA) mats is reported in this paper. Key phenomenological mechanisms involving roughness and changes in chemistry are used to describe the change in hydrophobic behavior of the CA mats. Contact angle calculations followed by Cassie's model apparent contact angle prediction have shown roughness alone is not capable of producing the super-hydrophobicity exhibited by as-spun mats. Fourier transform infrared spectroscopy of spin coated and electrospun mats shows a significant difference in the stretching of the hydroxyl bonds of the two materials. As it is this hydroxyl group which adds to the overall polarity of surface thus hydrophilicity of the material, we propose that the electrospinning process not only creates a rougher surface but also alters the chemistry of the electrospun cellulose acetate mats which ultimately gives rise to the reported hydrophobicity. Finally, due to their water repellent nature, and oleophilicity of the as-spun mats were tested as oil sorbent mats. The as-spun mats were capable of absorbing thirty times their weight in oil demonstrating their application for oil-water remediation.

Background: Cystic echinococcosis (CE), known as hydatid cyst, is a zoonotic parasitic infection caused by the larval stage of Echinococcus granulosus (E. granulosus) . Antigen B, the major component of hydatid cyst fluid, is encoded by members of a multigene family. The present study aimed to evaluate the genetic diversity of the gene encoding antigen B8/1 ( EgAgB8/1 ) among the main intermediate hosts of E. granulosus ., Methods: Twenty-eight hydatid cyst isolates (10 sheep, 9 human, and 9 cattle) were collected in Fars province, Iran. DNA was extracted from each cyst and PCR, followed by DNA sequencing was used to identify potential EgAgB8/1 sequence variation and polymorphism. A phylogenetic tree was constructed using MEGA 7.0 software and the maximum likelihood method., Results: Using EgAgB8/1 primers, an approximately 315 bp band was amplified from all the isolates. The PCR products were sequenced, and the sequences were deposited in GenBank (accession numbers, KY709266-KY709293). The polymorphism variation among the isolates was 0.0, while intra-species variation within the isolates and related sequences in GenBank was 0.5-1%. Analysis of the phylogenetic tree revealed that the isolates from humans, sheep, and cattle all cluster in one group and are homologous to the EgAgB8/1 M1 allele., Conclusion: Findings of this study revealed close similarity between the EgAgB8/1 of human, sheep, and cattle E. granulosus isolates. However, differences were found between the EgAgB8/1 sequences in our study and those reported from other CE endemic areas. Whether such similarities and differences exist in other subunits AgB subunits require further study.

The definitive genetic identification of Toxocara species is currently based on PCR/sequencing. The objectives of the present study were to design and conduct an in silico polymerase chain reaction-restriction fragment length polymorphism method for identification of Toxocara species. In silico analyses using the DNASIS and NEBcutter softwares were performed with rDNA internal transcribed spacers, and mitochondrial cox1 and nad1 sequences obtained in our previous studies along with relevant sequences deposited in GenBank. Consequently, RFLP profiles were designed and all isolates of T. canis and T. cati collected from dogs and cats in different geographical areas of Iran were investigated with the RFLP method using some of the identified suitable enzymes. The findings of in silico analyses predicted that on the cox1 gene only the MboII enzyme is appropriate for PCR-RFLP to reliably distinguish the two species. No suitable enzyme for PCR-RFLP on the nad1 gene was identified that yields the same pattern for all isolates of a species. DNASIS software showed that there are 241 suitable restriction enzymes for the differentiation of T. canis from T. cati based on ITS sequences. RsaI, MvaI and SalI enzymes were selected to evaluate the reliability of the in silico PCR-RFLP. The sizes of restriction fragments obtained by PCR-RFLP of all samples consistently matched the expected RFLP patterns. The ITS sequences are usually conserved and the PCR-RFLP approach targeting the ITS sequence is recommended for the molecular differentiation of Toxocara species and can provide a reliable tool for identification purposes particularly at the larval and egg stages.

Leishmaniasis is a parasitic disease caused by different species of protozoan parasites belonging to the genus Leishmania . In this study, Leishmania major (MRHO/IR/75/ER) promastigotes were cultured at 23-25 °C in brain heart infusion ( BHI ) medium supplemented with 10 % heat-inactivated fetal bovine serum (FBS) and penicillin and streptomycin. Antileishmanial effects of Lowsonia inermis and Cedrus libani methanolic extracts (0.07, 0.15, 0.31, 0.62, 1.25, 2.5, 5, 10 mg/mL) on Leishmania major promastigotes were evaluated using the MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. All experiments were repeated at least three times. Cedrus libani methanolic extract did not show any activity while Lowsonia inermis methanolic extract inhibited the growth of promastigote forms of L. major in vitro after 72 h of incubation and had a 50 % inhibitory concentration (IC50) of 1.25 mg/mL. The methanolic extract of Lawsonia inermis (henna) can be a promising antileishmanial agent in the future. Further experiments are needed for isolation of active fractions and identification of the active components of methanolic extract.

Dirofilaria immitis is an important filarial nematode in dogs. In this study, age and sex distribution of this zoonotic nematode among dogs were investigated in northwest of Iran in Meshkin-Shahr city. Molecular characteristics of the isolates, based on cytochrome oxidase subunit 1 (COX1) gene were compared to the isolates from other areas of the world.Blood samples were collected from 91 dogs which were selected by simple classified accidental sampling. Thin and thick blood smear examinations were used to find out infectivity with D. immitis . DNA extraction was performed from adult D. immitis recovered from heart of infected dogs. The COX1 gene was amplified and sequenced. Phylogenetic analysis was carried out using sequences obtained in this study along with relevant sequences deposited in the GenBank. Phylogenetic analysis and sequence variation was performed using MEGA software in comparison with those COX1 sequences deposited in GenBank. Out of 91 dogs, 19 (20.87%) were found positive for infection with D. immitis . There was no statistically significant difference between males and females of dogs in terms of D. immitis infection. However, the rate of infection in dogs more than 2 years old was significantly higher than those with lower age. Both sequences analyzed in this study showed 100% homology to each other. Intra-species variation of these isolates with those from other areas of the world amounted to 0 to 0.50%. Phylogenetic analysis of the COX1 gene suggested that it is conserved, and can be used for study on genetic diversity and classification of filarial nematodes.

Background: Toxocara is a common nematode of cats in different parts of Iran. Despite the close association of cats with human, no attempt has been done so far for molecular identification of this nematode in the country. Therefore, current study was performed on identification of some isolates of Toxocara from stray cats in Shiraz, Fars Province, Southern Iran, based on morphological and molecular approaches, and also determination of intensity of infection., Methods: This cross-sectional study was carried out on 30 stray cats trapped from different geographical areas of Shiraz in 2011. Adult male and female worms were recovered from digestive tract after dissection of cats. Morphological features using existing keys and PCR-sequencing of ITS-rDNA region and pcox1 mitochondrial l gene were applied for the delineating the species of the parasites., Results: Eight out of 30 cats (26.7%) were found infected with Toxocara nematodes. All the isolates were confirmed as Toxocara cati based on morphological features and the sequence of ribosomal and mitochondrial targets. Intensity of infection ranged from one to a maximum of 39 worms per cat, with a mean of 10.25±12.36, and higher abundance of female nematodes., Conclusion: The most prevalent ascaridoid nematode of stray cats in the study area was T. cati and female nematodes were more abundant than that of males. This issue has important role in spreading of eggs in the environment and impact on human toxocariasis.

An antigen-based ELISA system was evaluated for diagnosis of visceral leishmaniasis (VL). Urine samples from confirmed VL cases were tested by the system in comparison with urine samples from patients with non-VL infectious disease and patients with non-infectious diseases. Antigen was detected in urine of 21 out of 35 (60%) of VL cases. No cross reaction was found with samples from healthy individuals except in 3 samples from non-VL infectious diseases. Two samples from cutaneous leishmaniasis patient and one from patient with toxoplasmosis. The results obtained with the antigen-based ELISA were compared to those obtained with direct agglutination test (DAT), an antibody-based ELISA and indirect immunofluorescent antibody (IFA) revealed that the antigen-based ELISA is comparable in terms of specificity (91.2%; 95% CI=75.2-97.7%) but with a lower sensitivity (60%; 95% CI=42.2-75.6%). These results suggest that the antigen detection in urine by the noninvasive antigen-based ELISA system might offer a useful method for diagnosis of VL.

In the Mediterranean basin and Middle East, including Iran, visceral leishmaniasis (VL), also known as kala-azar, is caused by Leishmania donovani infantum. For the first time, the use of urine samples for the diagnosis of VL in immunocompetent patients has been used in this study. Based on its high sensitivity and specificity, as well as simplicity, this approach can serve as a valuable tool in the diagnosis of VL. We studied 60 urine samples from 60 individuals, 30 of which were patients with VL confirmed by parasitology, serology, or molecular methods, 5 were from healthy individuals, and 25 were from patients with cutaneous leishmaniasis, malaria, brucellosis, and hydatid cyst. Out of 30 samples from confirmed VL immunocompetent patients, 29 were positive (sensitivity, 96.8%) by polymerase chain reaction (RV1 and RV2 primers), and all the remaining 30 samples either from healthy individuals or patients with other diseases were negative (specificity, 100%). High sensitivity, specificity, and simplicity of the test can serve as a valuable tool in the diagnosis of VL.

Background: The causative agent of visceral leishmaniasis (VL) in Iran is Leishmania infantum (L. infantum) (Mediterranean type) and its major reservoir host is the dog., Objective: To compare the serological methods including direct agglutination test (DAT), indirect immunofluorescent-antibody test (IFA) and enzyme-linked immunosorbent assay (ELISA) for serodiagnosis of endemic strain of L. infantum., Methods: 61 blood samples from VL patients referred to Shiraz hospitals and 49 blood samples from control group were collected. Native strain of the parasite isolated from a VL patient from the region was cultured and characterized. Antigens from this L. infantum parasite were used in ELISA and IFA system., Results: Anti-Leishmania antibody was detected in 43 (70.5%), 49 (80.3%) and 51(83.6%) cases using DAT, IFA and ELISA, respectively. Based on these results, sensitivity and specificity of DAT was found to be 70.5% and 100%, respectively. Sensitivities of IFA and ELISA in diagnosis of VL were 80.3% and 83.6% and their specificity was 90.5%., Conclusion: Results of this study showed that DAT and ELISA have the highest specificity and sensitivity in diagnosis of VL. DAT is a simple, cost-effective and field applicable test. Thus, it can be recommended for early and accurate diagnosis of VL, especially in regions where malaria, brucellosis and tuberculosis are prevalent.
