Abstract

Resumen La industria vitivinícola y de destilación de bebidas espirituosas generan una notable cantidad de subproductos y residuos, cuya gestión inadecuada puede generar problemas socioeconómicos y riesgos ambientales debido a algunas características de disponibilidad estacional y elementos contaminantes. Tradicionalmente, las estrategias de valorización se han enfocado en el compostaje, para uso directo como mejorador de suelos en el viñedo. Sin embargo, este proceso requiere un tiempo prolongado y gran disposición de espacio libre (aeróbico), pudiendo causar fitotoxicidad y un efecto antimicrobiano. Una estrategia alternativa de tratamiento de estos residuos orgánicos es la digestión anaeróbica, después de la cual, en un periodo menor a 25 días y en una reducida área de trabajo, se obtienen dos digestatos: un líquido y un sólido, aptos para el uso agrícola en el viñedo. El objetivo del presente estudio fue evaluar la factibilidad de aprovechar las lías de vinificación y las vinazas de destilación para la elaboración de biofertilizantes y enmiendas orgánicas mediante digestión anaeróbica usando consorcios microbianos y estimar su valorización económica a mediana escala para una bodega vitivinícola del valle de Cañete (Perú). Se monitoreó la evolución del proceso de digestión (pH, Conductividad Eléctrica y Temperatura) para finalmente realizar una caracterización fisicoquímica y microbiológica de los digestatos obtenidos. Los resultados muestran que los productos elaborados presentan una significativa concentración de materia orgánica y fitonutrientes (Nitrógeno, Fosforo, Potasio, etc.) en un medio ligeramente ácido (pH entre 3.8 y 5.8), debido a la presencia de algunas especies de bacterias acido-lácticas y Bacillus, lo que podría conferirles propiedades como bioestimulantes y agentes de biocontrol. La estabilidad de los productos (pH, color y olor) fue evaluada durante 30 días más, mostrando excelentes condiciones higiénicas, debido a la ausencia de microorganismos patógenos, posibilitando su incorporación junto a otras enmiendas durante la mitad de la temporada vitícola. Por lo tanto, la valorización de estos subproductos y residuos vitivinícolas en la producción de biofertilizantes y enmiendas orgánicas para uso agrícola, podría suplir en gran parte el uso de fertilización sintética y recuperar la fertilidad del suelo. Los beneficios socio-económicos y ambientales serían múltiples, dentro de un enfoque de transición agroecológica y de economía circular.

Abstract

La vid (Vitis vinifera L.) fue introducida en Sudamérica durante el siglo XVI. En los valles de Ica y Cañete, los nombres "Quebranta Mollar", "Mollar" y "Prieta Mollar" son comúnmente utilizados en referencia a los diferentes colores de las bayas de estas variedades dentro de un mismo racimo. Esta heterogeneidad en cuanto al color de las bayas, sumada a los antecedentes históricos, sugieren que otras variedades, además de Quebranta y Mollar Cano, podrían estar implicadas en esta denominación genérica "Mollar". En este trabajo se identificaron diferentes fenotipos correspondientes a algunas variedades utilizadas en la elaboración de vino y Pisco. Para ello, 10 accesiones fueron recolectadas en 9 viñedos diferentes (valles de Ica y Cañete), y analizadas utilizando 20 marcadores moleculares y 25 descriptores morfológicos según la OIV. Nuestros resultados mostraron que las muestras recolectadas correspondieron a 5 genotipos, tres cultivares tradicionales y dos genotipos desconocidos no registrados previamente. Los tres cultivares conocidos fueron Listan Prieto, Quebranta y Mollar Cano. Las dos accesiones correspondientes a variedades no conocidas previamente fueron Mollar de Ica y Prieta Mollar, generadas a partir de retrocruzamientos entre Quebranta y sus progenitores. También se identificaron diferentes variaciones fenotípicas de Quebranta, lo cual enriquece la diversidad vitícola peruana.

Abstract

The denomination of “Uva de Italia” or “Italia” is commonly used by growers to refer to a group of varieties used from the colonial times to making a brandy of Pisco with muscat aroma. Previous work have demonstrated that Uva de Italia corresponds in fact to the variety Muscat of Alexandria, a widely spread variety around the world and in South America. However, the distinction between “Italia Dorada” and “Italia Rosada” is usually made, in allusion to the color variations observed in the berries. Our aim was to characterize 5 samples collected in vineyards from the valleys of Ica and Cañete. The genotypes were identified using 13 molecular markers of nuclear simple sequence repeat, and 23 morphological descriptors according to OIV. Our results showed that four genotypes were identified as Muscat of Alexandria while the other corresponded to variety well-known in Argentina as Moscatel Rosado or Uva Pastilla in Chile respectively. Moscatel Rosado showed functionally female flowers, with variable berries size and color in the range from greenish yellow through pink. This is the first identification of Moscatel Rosado as a variety present in the Peruvian vineyards and would allow its use in the pisco industry with distinctive aromatic characteristics. Abstract

A set of granular SmFeAsO1-xFx films were synthesized on LaAlO3 substrates with different percentage of the coverage of the substrate surface, and then their electrical properties were studied. At high temperatures, as the substrate coverage is increased, the films go from an insulating to a metallic behavior; and then, at low temperatures, the films that present a resistance RN of few kOhms in the normal state show a transition to the superconducting state with a width in the R(T) curves that changes with the coverage of the substrate surface. The line shape of these curves can be explained by the Ambegaokar-Halperin (AH) model of the resistance produced by the thermal activated phase slippage at the intergranular junctions if the dimensionality for the films is presumed appropriately. For the thicker films, that can be considered as a 3D network of the Josephson junctions, a temperature dependence of the AH parameter γ given by ∼ A(1 − t)3/2, fits the R(T) curve satisfactorily. However, for the films that cover the surface of the substrates only partially, the AH model fits the R(T) curves only if γ ~ A´(1 − t2)2, which is suitable for a 2D network of the Josephson junctions. 17. GrayC: Greybox fuzzing of compilers and analysers for C

Abstract

Fuzzing of compilers and code analysers has led to a large number of bugs being found and fixed in widely-used frameworks such as LLVM, GCC and Frama-C. Most such fuzzing techniques have taken a blackbox approach, with compilers and code analysers starting to become relatively immune to such fuzzers. We propose a coverage-directed, mutation-based approach for fuzzing C compilers and code analysers, inspired by the success of this type of greybox fuzzing in other application domains. The main challenge of applying mutation-based fuzzing in this context is that naive mutations are likely to generate programs that do not compile. Such programs are not useful for finding deep bugs that affect optimisation, analysis, and code generation routines. We have designed a novel greybox fuzzer for C compilers and analysers by developing a new set of mutations to target common C constructs, and transforming fuzzed programs so that they produce meaningful output, allowing differential testing to be used as a test oracle, and paving the way for fuzzer-generated programs to be integrated into compiler and code analyser regression test suites. We have implemented our approach in GrayC, a new open-source LibFuzzer-based tool, and present experiments showing that it provides more coverage on the middle- and back-end stages of compilers and analysers compared to other mutation-based approaches, including Clang-Fuzzer, PolyGlot, and a technique similar to LangFuzz. We have used GrayC to identify 30 confirmed compiler and code analyser bugs: 25 previously unknown bugs (with 22 of them already fixed in response to our reports) and 5 confirmed bugs reported independently shortly before we found them. A further 3 bug reports are under investigation. Apart from the results above, we have contributed 24 simplified versions of coverage-enhancing test cases produced by GrayC to the Clang/LLVM test suite, targeting 78 previously uncovered functions in the LLVM codebase.

Abstract

Interest in ancient and autochthonous cultivars has increased in recent years since they are directly related to the historical and cultural values of a region. Ica and Cañete valleys (Peru) have a long history of grapevine cultivation and the aim of this study was to identify phenotypes corresponding to the most used varieties for the local production of Pisco and wine. The 29 samples were collected in 17 vineyards in the Ica and Cañete valleys, and were analyzed using 20 molecular markers and 5 morphological descriptors according to the OIV. Results showed that the 29 collected samples corresponded to 11 genotypes: seven traditional cultivars and four unknown genotypes not registered previously. The known cultivars were 'Muscat of Alexandria', 'Listán Prieto', 'Quebranta', 'Moscatel Rosado', 'Pedro Giménez', 'Muscat Hamburg' and 'Palomino Fino'. The four not registered genotypes are locally known as 'Mollar de Ica', 'Moscatel Rosada de Cañete', 'Prieta Mollar' and 'Torontel'. All of them correspond to offspring of traditional-heritage Peruvian cultivars. We also found a phenotypic variation of 'Listán Prieto' with muscat flavor and identified phenotypic berry color variations in 'Quebranta'. Study questionWhat is the genetic basis of preeclampsia in Andean families residing at high altitudes?Summary answerA top candidate region associated with preeclampsia containing clotting factor genesPROZ, F7andF10was found on chromosome 13 of the fetal genome in affected Andean families.What is known alreadyPreeclampsia, a multi-organ complication of pregnancy, is a leading cause of maternal morbidity and mortality worldwide. Diagnosed by the onset of maternal hypertension and proteinuria after 20 weeks of gestation, this disorder is a common cause of preterm delivery and affects approximately 5-7% of global pregnancies. The heterogeneity of preeclampsia has posed a challenge in understanding its etiology and molecular basis. However, risk for the condition is known to increase in high altitude regions such as the Peruvian Andes.Study design, size, durationTo investigate the genetic basis of preeclampsia in a high-altitude resident population, we characterized genetic diversity in a cohort of Andean families (N=883) from Puno, Peru, a high-altitude city above 3,500 meters. Our study collected DNA samples and medical records from case-control trios and duos between 2011-2016, thus allowing for measurement of maternal, paternal, and fetal genetic factors influencing preeclampsia risk.Participants/materials, setting, methodsWe generated high-density genotype data for 439,314 positions across the genome, determined ancestry patterns and mapped associations between genetic variants and preeclampsia phenotype. We also conducted fine mapping of potential causal variants in a subset of family participants and tested ProZ protein levels in post-partum maternal and cord blood plasma by ELISA.Main results and the role of chanceA transmission disequilibrium test (TDT) revealed variants near genes of biological importance in pregnancy physiology for placental and blood vessel function. The most significant SNP in this cluster, rs5960 (p−6) is a synonymous variant in the clotting factorF10. Two other members of the coagulation cascade,F7andPROZ, are also in the top associated region. However, we detected no difference of PROZ levels in maternal or umbilical cord plasma.Limitations, reasons for cautionOur genome-wide association analysis (GWAS) was limited by a small sample size and lack of functional follow up. Our ELISA was limited to post-natal blood sampling (only samples collected immediately after birth). But, despite a small sample size, our family based GWAS design permits identification of novel significant and suggestive associations with preeclampsia. Further longitudinal studies could analyze clotting factor levels and activity in other pregnant cohorts in Peru to assess the impact of thrombosis in preeclampsia risk among Andean highlanders.Wider implications of the findingsThese findings support previous evidence suggesting that coagulation plays an important role in the pathology of preeclampsia and potentially underlies susceptibility to other pregnancy disorders exacerbated at high altitudes. 22. Saturación de oxígeno en pacientes durante hemodiálisis a diferentes altitudes

Abstract

Introduction: Patients undergoing hemodialysis may develop hypoxemia during the procedure, and hypoxemia may become more intense if hemodialysis is performed in a high altitude environment. Objectives: To assess variations in oxygen saturation in patients undergoing hemodialysis at different altitude levels. Material and methods: This is a prospective case series, where patients from hemodialysis programs in three cities were assessed. These places were at 101-, 2335-, and 3825- meters above sea level. Patients had their oxygen saturation measured in a semi-sitting position with a pulse oxymeter device. Measurements were taken before, during, and at the end of hemodialysis sessions. Results: Sixty-five patients undergoing hemodialysis were included. They were from three cities at different altitude above sea level. Oxygen saturation significantly decreased as long as altitude increased (97,32 ± 1,10 vs. 94,75 ± 2,17 vs. 84,85 ± 5,86 p= 0,00). Oxygen saturation showed changes before, during, and at the end of hemodialysis procedures at different altitude. Ferritin levels were lower in high altitude patients compared with those at sea level and at moderate altitude (131,75 [106,04 – 157,45] vs. 493,38 [273,19 – 713,56] vs. 550,66 [329,75-771,57] p=0,01). Hemoglobin values were lower in patients from middle altitude levels (10,8 [10,12-11,47] vs 11,32 [10,79-11,85] vs 11,84 [11,54-12,14] p=0,03). Conclusions: In patients undergoing chronic hemodialysis oxygen saturation is reduced as long as altitude increases, and significant variations were found during hemodialysis procedures., Introducción: Los pacientes en hemodiálisis pueden presentar hipoxemia durante este procedimiento, la cual puede ser mayor si la hemodiálisis se realiza en ciudades de gran altitud. Objetivos: Evaluar las variaciones de la saturación de oxígeno en pacientes en hemodiálisis a diferentes altitudes. Material y métodos: Es una serie de casos prospectivos, donde se evaluaron pacientes de un programa de hemodiálisis crónica en tres ciudades, localizadas a diferentes altitudes:101, 2 335 y3 825 metros sobre el nivel del mar, en los que se evaluó la saturación de oxígeno en posición semisentada con pulsoxímetro: antes, durante y al finalizar el procedimiento de hemodiálisis. Resultados: Se incluyeron 65 pacientes en hemodiálisis, de 3 ciudades de diferente altitud, con características similares, la saturación de oxígeno disminuyó significativamente a medida que aumentaba la altitud (97,32 ± 1,10 vs 94,75 ± 2,17 vs 84,85 ± 5,86 p=0,00), la saturación de oxígeno mostró cambios antes, durante y al final del procedimiento de hemodiálisis en las diferentes altitudes. Los niveles de ferritina fueron menores en pacientes de gran altitud comparado con los de nivel del mar y altitud moderada (131,75 [106,04 – 157,45] vs 493,38 [273,19 – 713,56] vs 550,66 [329,75-771,57] p=0,01). Los valores de hemoglobina fueron menores en los pacientes de mediana altitud (10,8 [10,12-11,47] vs 11,32 [10,79-11,85] vs 11,84 [11,54-12,14] p=0,03). Artículo digital., The malaria is an infectious disease caused by Plasmodium parasites. Plasmodium falciparum is the responsibleof the most severe form of the disease. The report of cases of malaria in Sucre (Colombia) increased of 371 in 2000to 611 in 2004, according to data of the epidemiology department of DASSALUD; in spite of this, studies publishedabout of causes of this increase do not exist.One of the factors related to the increase of the morbidity and mortality by malaria at world is the resistancethat P. falciparum has developed towards antimalarials drugs . The resistance to pirimetamina has been relatedto point mutations in the gene that codifies for the Dihydrofolate reductase (DHFR), component of the biosyntheticroute of folate of the parasite. The objective of the present study was to detect the presence in Sucre, of P.falciparum strain with the Asn 108 DHFR mutation, associated with in vitro pyrimethamine resistance.The investigation was carried out between October of 2002 and March of 2004. The detection of the wildgenotype Ser- 108 and the mutant genotype Asn -108 was made by allele specific PCR. During the study 122 casesof falciparum malaria were reported, of which 6.6 % were native infections and 93.4% were acquired in endemicdepartments. Of 36 analyzed samples by PCR, 27.7 % displayed the genotype Asn -108, the rest of the samplesdisplayed the genotype Ser -108. These results indicate that one of the main causes of the increase of registeredmalaria cases in Sucre, are the migrations and frequent flow of infected people, originating of endemic region.This is an important factor by the appearance of native infection and favors the introduction of P. falciparumstrain with resistance to pirimetamina., La malaria es una enfermedad infecciosa causada por parásitos del género Plasmodium, de las cuales P. falciparumes el causante de la forma más severa de la enfermedad. En Sucre (Colombia), el reporte de casos de malaria incrementó de 371 en el año 2000 a 611 para el año 2004, según datos del departamento de epidemiología de DASSALUD; a pesar de esto, no existen estudios publicados sobre las causas de este incremento. Uno de los factores relacionado con el aumento de la morbilidad y la mortalidad por malaria a nivel mundial es la resistencia que el parásito P. falciparum ha desarrollado hacia los antimalaricos. La resistencia a pirimetaminaha sido asociada con la presencia de mutaciones puntuales en el gen que codifica para la enzima Dihidrofolatoreductasa (DHFR), componente de la ruta biosintética de folatos del parásito. El propósito del presente estudio fuedetectar la presencia en Sucre, de cepas de P. falciparum con la mutación Asn 108 en DHFR, asociada con resistencia In Vitro a la pirimetamina. La investigación se llevó a cabo entre octubre del 2002 y marzo del 2004. La detección del genotipo silvestre Ser-108 y del genotipo mutante Asn –108 se realizó mediante un ensayo de PCR alelo específico. Durante el estudio se reportaron 122 casos de malaria por P. falciparum, de los cuales 6,6 % eran infecciones autóctonas y 93,4%adquiridas en departamentos vecinos endémicos. De 36 muestras analizadas por PCR, el 27,7 % presentó el genotipo Asn –108, el resto de las muestras presentó el genotipo Ser- 108.Estos resultados indican que una de las principales causas del aumento en los casos de malaria registrados en Sucre, son las migraciones y flujo frecuente de personas infectadas, provenientes de municipios de alta en demicidad. Este es un factor importante en la aparición de brotes de malaria autóctonos y favorece la introducción de cepas de P. falciparum resistentes a la pirimetamina. (Aljure S., Mendoza K., Mendoza DL. Abstract

INTRODUCTION: Turner syndrome(TS) is a chromosomal disorder that occurs in one of every 2500 live births, characterized by the complete or partial absence of a chromosome X (the complete monosomy [45,X] accounts for more than 50% of cases), although some studies have found evidence of a high prevalence of mosaicism. It is associated with a range of phenotypic characteristics, chief of which are short stature, gonadal dysgenesis, hand and foot lymphoedema, pterygium colli, cubitus valgus and cardiovascular malformations, among others. Growth failure is a cardinal feature of TS, which may impair their quality of life, and adult patients have a mean height approximately 20 cm lower than that of unaffected women of the same ethnic group. METHODOLOGY: A study performed in the outpatient clinic of pediatric endocrinology of the National Institute for Pediatrics, descriptive, retrospective in 19 patients with Turner syndrome confirmed with karyotype, was followed up for 1 year in growth hormone treatment, size, weight was determined , Body mass index, growth rate at the beginning and at the end of the treatment. In addition, serum glucose, glycosylated hemoglobin was determined at the beginning and end of treatment. Statistical analysis was performed in the SPSS version program. RESULTS: We evaluated 19 patients with mean age 9 years +3.51, the average of the dose of growth hormone was of 1.43mg / day the size before treatment was an average of 114.73 with a SD:-3.06, showing improvement in the posterior size to treatment with an average of 119.25 SD:-2.7. In addition, the Body Mass Index was assessed with an average of 24.63 + 9.9 and at the end of treatment 27.98 +10.4. Regarding the growth rate, it is observed prior to the treatment Media 4.3 DE: 1.66 and the growth rate after the average treatment: 17.5 DE: 3.42 Levels of glycosylated hemoglobin (HBA1C) were assessed at the start of treatment with a mean of 5.3 (4.2- 5.7) and at the end of treatment the average of HBA1C 5.59 (4-6). DISCUSSION AND CONCLUSIONS: The short stature in TS is the fundamental finding and may be present in the absence of other clinical manifestations.The use of rhGH to increase the speed of growth and final height in patients with TS is an accepted and endorsed by different study. It has been observed in international studies that gains of 5-10cm are obtained with respect to the final size. It should start when the size is below -2 SDS with respect to the population mean or when the growth rate is lower than the 10th percentile for their bone age.The study that is presented is 12 months after the start of the treatment, an increase in the speed of growth as well.

This study used quantitative research method as a tool in conducting survey questionnaire to 100 respondents that composed of student, workers, professional in brgy Gulod Quirino Highway novaliches Q.C to determine its marketing viability. This study also utilized financial statement to know its financial growth in the future. The resultof the survey based on the research is that 85% of the respondents agree to have computer shop near in the area becuase it will be useful as a medium for their study and entertainment. Also. 55% of the respondents are willing to spend money for renting a computer shop to cater their needs. 95% of the people agreed on the location because it is very accessible and convenient to them for the promotion, survey shows 90% agreed that advertisement is very useful in making name in the industry. As the years past, and the rapid changing of the technology, computer shops are still existing in the market because many students still need this equipment for their study and entertainment. Jakes cafe will cater the needs of every student and gamers that cannot afford to have a laptop. Abstract

En este estudio se describió el cultivo y producción de Pleurotusostreatus (Hongo ostra) utilizando mazorcas de cacao como sustrato. El cultivo de P. ostreatus se realizó en ocho etapas, donde se evaluó el efecto del tiempo defermentación (1, 3 y 7 días) y el tratamiento térmico del sustrato (pasteurización y esterilización) sobre la producción de P.ostreatus. Se evaluó el peso de carpóforos, % Eficiencia biológica (E.B) y % Rendimiento (R), en 6 tratamientos con cinco repeticiones. Para el experimento se utilizó un diseño completamente al azar (DCA)mediante un análisis de varianza (ANOVA). Además, se realizó la prueba de rangos múltiples de Tukey a los tratamientos.El tratamiento que resultó significativamente diferente y con mejores resultados en comparación a los demás, fue el tratamiento con3 días de fermentación tratado por esterilización (T5), que obtuvo una producción de283g de hongos recolectados en dos cosechas, 31,22% de Eficiencia biológica y 28,30% de Rendimiento.

Abstract

El artículo presenta la actividad científica desarrollada en los congresos internacionales de ingeniería de proyectos organizados por AEIPRO. Analizando y visualizando la información a través del análisis de dominios científicos y del análisis de redes de la literatura científica desarrollada desde el II Congreso Internacional del 1998 hasta el XVI Congreso Internacional del 2012. Los resultados permiten identificar los frentes de investigación y la base de conocimientos científica en Ingeniería de Proyectos desarrollada en los congresos internacionales de AEIPRO, proporcionando resultados estadísticos sobre la distribución del aporte internacional, el grado de integración de la investigación y la colaboración científica entre universidades, instituciones científicas y profesionales. Finalmente, se realiza una comparación entre la distribución de la investigación según la temática actual de los congresos y las áreas de conocimientos que gestionan el ciclo de vida del proyecto, alcance, tiempo, costes, calidad, recursos humanos, comunicación, riesgos y adquisiciones.

Abstract

The major histocompatibility complex (MHC) is a highly polymorphic region of the genome essential to immune responses and animal health. In galliforms, the MHC is divided into 2 genetically unlinked regions (MHC-B and MHC-Y). Many MHC-B genes are involved in adaptive or innate immunity, yet others have nonimmune or unknown functions. The sequenced MHC-B region of the turkey (Meleagris gallopavo) contains 40 genes, the majority of which are predicted transcripts based on comparison with the chicken or quail, without direct evidence for expression. This study was designed to test for the presence of MHC-B gene transcripts in a panel of immune and nonimmune system tissues from domestic turkeys. This analysis provides the first locus-wide examination of MHC-B gene expression in any avian species. Most MHC-B genes were broadly expressed across tissues. Expression of all predicted genes was verified by reverse-transcription PCR, including ß-butyrophilin 2 (BTN2), a predicted gene with no previous evidence for expression in any species. Previously undescribed splice variants were also detected and sequenced from 3 genes. Characterization of MHC-B expression patterns helps elucidate unknown gene functions and potential gene coregulation. Determining turkey MHC-B expression profiles increases our overall understanding of the avian MHC and provides a necessary resource for future research on the immunological response of these genes. Abstract

Cytochromes P450 (P450 for protein; CYP for gene) are a superfamily of membrane-bound hemoproteins that oxidize a large number of endogenous and exogenous compounds. Through oxidation reactions, these enzymes are often responsible for the toxic and carcinogenic effects of natural food-borne toxicants, such as the mycotoxin aflatoxin B1 (AFB1). Previous studies in our laboratory have shown that the extreme sensitivity of turkeys to AFB1 is in part explained by efficient hepatic P450-mediated epoxidation to the toxic and reactive metabolite the exo-AFB1-8,9-epoxide (AFBO). Using 3′-5′-rapid amplification of cDNA ends (RACE), we amplified CYP3A37 from turkey liver RNA, the E. coli-expressed protein which efficiently epoxidates AFB1. Turkey CYP3A37 has an ORF of 1512 bp, and the protein is predicted to be 504 amino acids with 97% homology to chicken CYP3A37. The turkey gene is organized into 13 exons and 12 introns. A single nucleotide polymorphism in the 11th intron was used to assign CYP3A37 to turkey linkage group 10 (corresponding to chicken chromosome 14, GGA14). Because of the important role of P450s in the extreme sensitivity of turkeys to the toxic effects of AFB1, this study will contribute to the identifying allelic variants of this important gene in poultry. Abstract

Although the domestic turkey (Meleagris gallopavo) is a valuable agricultural commodity, genetic studies on this species lag behind those of other agricultural species. In this study, we examined expressed sequence tags (EST) from a turkey cardiac cDNA library constructed from 4 birds representing 2 developmental stages. A collection of 3,937 EST sequences were sequenced and analyzed for gene annotation and sequence variation. Clustering of sequences resulted in 353 contigs and 874 singletons (1,227 putative transcripts). All EST sequences were compared by BLASTN to the chicken whole genome sequence and to Ensembl and National Center for Biotechnology Information databases. The majority of significant matches correspond to genes found in the chicken. Sequence polymorphisms were identified in 310 contigs, 64 where the minor allele was observed to be present in more than 1 sequence. This study gives species-specific insight into the cardiac transcriptome of turkeys and provides resources for future studies of cardiac function. [ABSTRACT FROM AUTHOR]

Abstract

An integrated genetic linkage map was developed for the turkey (Meleagris gallopavo) that combines the genetic markers from the three previous mapping efforts. The UMN integrated map includes 613 loci arranged into 41 linkage groups. An additional 105 markers are tentatively placed within linkage groups based on two-point LOD scores and 19 markers remain unlinked. A total of 210 previously unmapped markers has been added to the UMN turkey genetic map. Markers from each of the 20 linkage groups identified in the Roslin map and the 22 linkage groups of the Nte map are incorporated into the new integrated map. Overall map distance contained within the 41 linkage groups is 3,365 cM (sex-averaged) with the largest linkage group (94 loci) measuring 533.1 cM. Average marker interval for the map was 7.86 cM. Sequences of markers included in the new map were compared to the chicken genome sequence by ‘BLASTN’. Significant similarity scores were obtained for 95.6% of the turkey sequences encompassing an estimated 91% of the chicken genome. A physical map of the chicken genome based on positions of the turkey sequences was built and 36 of the 41 turkey linkage groups were aligned with the physical map, five linkage groups remain unassigned. Given the close similarities between the turkey and chicken genomes, the chicken genome sequence could serve as a scaffold for a genome sequencing effort in the turkey. Copyright © 2007 S.
