- Abstract
Background In recent years, positron emission tomography (PET)-adapted protocols and new agents such as brentuximab vedotin are increasingly used as part of first-line protocols of advanced-stage Hodgkin lymphoma (HL). Objectives This manuscript reviews guideline-based therapy and available trial data in advanced-stage HL. Materials and methods This is a narrative review. Results The German HL guideline recommends interim PET2-guided (PET after 2 cycles) chemotherapy using 4 cycles of eBEACOPP (bleomycin, etoposide, doxorubicin, cyclophosphamide, vincristine, procarbazine and prednisone) for PET2-negative patients up to 60 years of age with advanced HL and 6 cycles eBEACOPP for PET2-positive patients. Furthermore, patients with PET-positive residual lymphoma should receive consolidation radiotherapy with 30 Gy. This is based on effective tumor control and short duration of therapy of 3 months in most patients. For elderly patients, treatment with 2 cycles of ABVD (adriamycin, bleomycin, vinblastine and dacarbazine) followed by 4 cycles of AVD and irradiation of PET-positive residuals is recommended. Conclusions Advanced-stage HL is curable in more than 90% of cases. The goal of current scientific efforts is to reduce toxicity while maintaining efficacy by including immune checkpoint inhibitors.

Background: Antibacterial agents play important roles in the treatment of bacterial infections. However, the development of antimicrobial resistance (AMR) and carry-over of substances into the environment are several problems arising during oral treatment of bacterial infections.We assessed AMR development in commensal Escherichia coli (E. coli) in enrofloxacin treated and untreated animals. In addition, we examined fluoroquinolone in the plasma and urine of treated and untreated animals, and in sedimentation dust and aerosol. Methods: In each trial, six pigs were treated with enrofloxacin via powder, granulate or pellet forms in two time periods (days 1-5 and 22-26). Four pigs served as untreated controls. The minimum inhibitory concentration (MIC) was determined to evaluate AMR development. Analysis of enro- and ciprofloxacin was performed with high performance liquid chromatography. Results: Non-wildtype E. coli (MIC > 0.125 µg/mL) was detected in the pellet treated group after the first treatment period, whereas in the other groups, non-wildtype isolates were found after the second treatment period. E. coli with MIC > 4 µg/mL was found in only the pellet trial. Untreated animals showed similar susceptibility shifts several days later. Bioavailability differed among the treatment forms (granulate > pellet > powder). Enro- and ciprofloxacin were detected in aerosols and sedimentation dust (granulate, powder > pellet). Conclusions: This study indicates that the kind of the oral dosage form of antibiotics affects environmental contamination and AMR development in commensal E. coli in treated and untreated pigs.

Background H1 receptor antagonists are commonly used for the treatment of allergic diseases. The aim of this study was to find out, if antihistaminic compounds like mepyramine have the ability to influence the activity of antibacterials. Therefore, the checkerboard method was chosen to detect these possible effects in vitro. Studies were performed with two different Escherichia coli (E. coli) strains as test microbes, treated with antibacterials in combination with mepyramine. Results The minimum inhibitory concentration (MIC) of E. coli ATCC® 25922™ and E. coli PIG 01 was reduced by combinations of the tested antibacterials with mepyramine. Conclusions These results have to be confirmed in vivo, before the use of antihistamines should be considered as potential way to minimize the amount of used antibacterials for treatment of E. coli infections.

- Abstract
We consider the online traveling salesperson problem (TSP), where requests appear online over time on the real line and need to be visited by a server initially located at the origin. We distinguish between closed and open online TSP, depending on whether the server eventually needs to return to the origin or not. While online TSP on the line is a very natural online problem that was introduced more than two decades ago, no tight competitive analysis was known to date. We settle this problem by providing tight bounds on the competitive ratios for both the closed and the open variant of the problem. In particular, for closed online TSP, we provide a 1.64-competitive algorithm, thus matching a known lower bound. For open online TSP, we give a new upper bound as well as a matching lower bound that establish the remarkable competitive ratio of 2.04. Additionally, we consider the online DIAL-A-RIDE problem on the line, where each request needs to be transported to a specified destination. We provide an improved non-preemptive lower bound of 1.75 for this setting, as well as an improved preemptive algorithm with competitive ratio 2.41. Finally, we generalize known and give new complexity results for the underlying offline problems. In particular, we give an algorithm with running time O(n) for closed offline TSP on the line with release dates and show that both variants of offline DIAL-A-RIDE on the line are NP-hard for any capacity c≥ 2 of the server.

- Abstract
Background Chemical exchange saturation transfer (CEST) is a novel MRI technique applied to brain tumor patients. Purpose To investigate the anatomic location dependence of CEST MRI obtained at 7T and histopathological/molecular parameters in WHO IV° glioma patients. Study Type Analytic prospective study. Population Twenty‐one patients with newly diagnosed WHO IV° gliomas were studied prior to surgery; 11 healthy volunteers were investigated. Field Strength/Sequence Conventional MRI (contrast‐enhanced, T2w and diffusion‐weighted imaging) at 3T and T2w and CEST MRI at 7T was performed for patients and both patients and volunteers. Assessment Mean CEST signal intensities (nuclear‐Overhauser‐enhancement [NOE], amide‐proton‐transfer [APT], downfield NOE‐suppressed APT [dns‐APT]), ADC values, and histopathological/molecular parameters were evaluated with regard to hemisphere location and contact with the subventricular zone. CEST signal intensities of cerebral tissue of healthy volunteers were evaluated with regard to hemisphere discrimination. Statistical Tests Spearman correlation, Mann–Whitney U‐test, Wilcoxon signed‐rank‐test, Fisher's exact test, and area under the receiver operating curve. Results Maximum APT and dns‐APT signal intensities were significantly different in right vs. left hemisphere gliomas (P = 0.037 and P = 0.007), but not in right vs. left hemisphere cerebral tissue of healthy subjects (P = 0.062‐0.859). Mean ADC values were significantly decreased in right vs. left hemisphere gliomas (P = 0.044). Mean NOE signal intensity did not differ significantly between gliomas of either hemisphere (P = 0.820), but in case of subventricular zone contact (P = 0.047). A significant correlation was observed between APT and dns‐APT and ADC signal intensities (rs = –0.627, P = 0.004 and rs = –0.534, P = 0.019), but not between NOE and ADC (rs = –0.341, P = 0.154). Histopathological/molecular parameters were not significantly different concerning the tumor location (P = 0.104–1.000, P = 0.286–0.696). Data Conclusion APT, dns‐APT, and ADC were inversely correlated and depended on the gliomas' hemisphere location. NOE showed significant dependence on subventricular zone contact. Location dependency of APT‐ and NOE‐mediated CEST effects should be considered in clinical investigations of CEST MRI.
