This article discusses a partnership with two different civic education organizations to study three different student-centered discussion designs. In one study, the authors worked with the Close Up Foundation. Close Up is a non-profit that annually brings 20,000 high school and middle school students from all 50 states and U.S. territories to Washington, D.C., for a weeklong, place-based study of the federal government. During a program week, students are put into geographically diverse groups of 20 and participate in two discussions of public policy issues. The second study was done in partnership with Street Law, Inc., using a deliberation strategy. Street Law is a non-profit that produces free law and public policy related classroom materials. Deliberations follow the format for Structured Academic Controversy (SAC), originally designed by Johnson and Johnson. In a SAC, students work in groups of four to learn about competing positions on a policy issue. Using the provided texts, one pair in the group presents the best reasons for their assigned side while the opposite team takes notes. This process is then repeated by the other team. Upon completion, pairs reverse positions and present new reasons for each side. Finally, the two teams drop their roles and discuss possible consensus positions within their group of four. For both studies, the authors were interested in how the structure of the discussion affected students' opinions about the issues being discussed and whether the students experienced the discussion as inclusive and fair to competing points of view. In this article, the authors discuss the findings as they relate to some common concerns that teachers have about bringing politics into the classroom.

Sedentary occupation of the southern Levantine coast spans from the Pre-Pottery Neolithic C to the Early Bronze Age Ib phase (c. 7000-3100 BC). Sites dating to the Early Pottery Neolithic (c. 6400-5500 BC) are scarce, however, potentially reflecting the effects of the 8.2ka climatic event. Here, the authors present the investigations at the submerged site of Habonim North off the Carmel Coast. Typological and radiocarbon dating indicate an Early Pottery Neolithic occupation and evidence for continuity of subsistence and economic strategies with both earlier and later Neolithic cultures. The results indicate the resilience of coastal communities in the face of significant climatic uncertainty and contribute to understanding human responses to environmental change. Photogrammetry and LiDAR have become increasingly accessible methods for documentation of Cultural Heritage sites. Academic and government agencies recognize the utility of high-resolution 3D models supporting long-term asset management through visualization, conservation planning, and change detection. Though detailed models can be created with increasing ease, their potential for future use can be constrained by a lack of accompanying topographic data, data collector skill level, and incomplete recording of the key metadata and paradata which make such survey data useful to future endeavors. In this paper, informed by various international survey organizations and data archives, we present a framework to record and communicate Cultural Heritage - focusing on architectures based on 3D metric survey - to first describe the data and metadata which should be included by surveyors to enable data usage and to communicate the expected utility of this data.

Underwater photogrammetry presents unique challenges due to the optical properties of water that, if not correctly taken into account, might affect the quality of the survey and the related 2D and 3D products. It is recognized nowadays the importance to train newcomers to underwater surveying, and extend and consolidate the knowledge of best practices for underwater data acquisition. Starting from this consideration, we propose the development of POSER, a 3D simulation framework designed to facilitate the teaching of underwater imaging principles. The project, an ISPRS Educational and Capacity Building Initiative, is built upon the open-source platform Blender, incorporating realistic modelling of the physical properties of water, including light refraction, scattering, and absorption phenomena, to simulate underwater surveying conditions. We foster a learning-by-doing approach, providing users with ready-to-use application scenarios inspired by real-life case studies. They will cover a range of application fields, from marine ecology to archaeology and subsea metrology, and allow users to address the complexities of underwater surveying practices. This paper introduces POSER to the community, presenting its educational vocation and describing its constituent components.

Clearing and construction activities related to the Maya Train (Tren Maya) project resulted in potential and inevitable impacts to archaeological caves sites in largely undeveloped areas of Quintana Roo. An effort coordinated by Mexico’s National Institute of Anthropology and History (INAH) involved accelerated digital documentation of two caves – via SLAM-enabled mobile LiDAR scanning and targeted photogrammetry – to facilitate prompt visualization and evaluation of terrestrial and subterranean geospatial relationships. Mobile LiDAR is well suited to the challenges of capturing the complex, multilevel morphology of caves and was readily deployed across and through priority environments. Specific archaeological features – such as ancient Maya rock art and masonry shrines – were documented via photogrammetry, and the resulting higher-resolution models co-referenced with the georeferenced mobile LiDAR-generated point clouds of each cave and the surrounding topographic context. This integrative approach contributed to a more informed decision-making process, with respect to conservation and construction, and provided baseline data for future monitoring of the affected cave sites.

Three research trials were performed over two spring seasons to assess the performance of bell pepper (Capsicum annuum) cultivars in southern Georgia, USA. Studies were conducted in commercial production fields in Echols and Lowndes counties, which are major bell pepper production areas in Georgia, USA. Eleven cultivars were evaluated, and 10 newer bell pepper cultivars with improved disease resistances for bacterial leaf spot races 1 to 10, tomato spotted wilt virus, and/or phytophthora were compared with ‘Aristotle’, which historically was the most widely grown bell pepper hybrid in the southeastern United States. Our goal was to determine how the new and improved cultivars compared with ‘Aristotle’ for total yield, fruit size distribution, and unmarketable defects in the absence of disease pressure. There were significant differences in fruit size, marketable fruit, and culls between cultivars contrasting their adaptability for production in southern Georgia. The top performers for yield and preferred fruit size for fresh market bell pepper spring production in southern Georgia were ‘Autry’, ‘Standout’, and ‘Antebellum’. Yields and fruit size distribution of these cultivars were similar to ‘Aristotle’ but may be preferred in certain situations due to additional disease resistance. The cultivar, SV 3255 also had higher yields and low defects but had smaller average fruit sizes.

Objective: The Exercise is Medicine® On Campus (EIM-OC) international campaign leverages university resources (e.g., health centers, recreation, and kinesiology departments) to encourage students, faculty, and staff to integrate physical activity into campus culture. This involves evaluating student physical activity levels during health visits and establishing referral systems for exercise prescriptions. EIM-OC allows universities to earn tiered recognition (Gold, Silver, or Bronze) based on their on-campus physical activity promotion and integration. For Gold recognition, schools must incorporate routine physical activity assessments into their health system, ultimately connecting healthcare providers with health/fitness professionals (HFPs, e.g., campus recreation professionals, kinesiology professors). This research worked to uncover pivotal factors driving EIM-OC on-campus collaborations through HFPs’ perspectives. Methods: HFPs (n = 11) working full-time at a Gold-level institution (n = 10 in United States) participated. Semi-structured, Zoom-recorded interviews with a generic qualitative research design were completed between June and September 2022. Results: Major thematic findings included the importance of tangible support (e.g., personnel), encounters with both trust and tension cross-campus, positive student development opportunities, and variations in outcome reporting and program evaluation. Faculty and staff emphasized the need for methods to obtain and sustain program funding. Participants also expressed the importance of interdisciplinary collaboration to increase the collective impact of EIM-OC on student health and overall collegiate success. Conclusion: HFPs expanded on their EIM-OC experiences and program sustainment or growth requirements. With increased interdisciplinary collaboration, rigor in outcome reporting, and tangible resources, the collective impact of EIM-OC on student health outcomes and overall collegiate success could be greatly perpetuated.

Phytophthora capsici (PCap), which causes Phytophthora root rot, is the most destructive soilborne pathogen for bell pepper (Capsicum annuum L.) production in Georgia, USA. Extensive host range, persistence of inoculum in the soil, and lack of effective chemical control methods make this disease particularly difficult to manage. Resistant cultivars offer a practical solution to manage PCap in affected bell pepper fields. However, most commercial cultivars resistant to PCap are predominantly grown in the northeastern United States. This research aimed to screen commercial bell pepper cultivars for resistance to PCap, marketable yield, fruit size distribution, and overall adaptability for production within the largest growing areas in southern Georgia. Field trials were conducted during Spring 2022 and 2023 in commercial growers’ fields with a known history of PCap infestation to evaluate PCap-resistant bell pepper cultivars across four trials conducted in the three counties (Colquitt, Echols, and Lowndes) responsible for ∼78% of bell pepper production in Georgia. Eleven cultivars were evaluated: nine that claimed PCap resistance and two widely grown PCap susceptible cultivars included for comparison. Phytophthora root rot incidence was very low in these research trials. However, there were significant differences in total yields, marketable yields, fruit size, and unmarketable yields. On the basis of marketable yields for jumbo, extra large, and large-sized fruit, we recommend the PCap-resistant cultivars PS 0994-1819, Paladin, and Mercer for fresh market bell pepper growers. Furthermore, ‘Revolution’ is recommended for processor growers due to higher jumbo-sized fruit yields and lower quality and higher pancaking for the spring season in southern Georgia, USA. Newer cultivars Tarpon and Nitro have a more desirable disease-resistance package; however, ‘Nitro’ had small-sized fruit, and ‘Tarpon’ tended to have lower total yields than current commercial standards.

Study region:: The Ohio River in the northeast United States (US). Study focus:: Low streamflows are critical for urban water security, agricultural irrigation, water quality regulatory thresholds, navigation passage, and ecological well-being. However, there is insufficient understanding of the natural low flow conditions in rivers containing dams and artificial reservoirs, in part because we have inadequate records of natural flows prior to the introduction of the water control infrastructure. We demonstrate an improved technique for estimation of human impact on low flow, and describe the analytical innovations necessary to apply the technique. The primary innovation necessary was the development of a parsimonious streamflow routing algorithm to aggregate naturalized flow from reservoirs operated by the US Army Corps of Engineers (USACE) to locations of concern along the Ohio River mainstem. New hydrological insights for the region:: This study shows that, in dry years, releases from USACE reservoirs during autumn months account for up to approximately half of the mainstem flow, and the influence of USACE water control infrastructure is more pronounced on many of the Ohio River’s tributaries. The Flow Duration Curves also show significant differences in low flow throughout the mainstem. This has implications for dry season river functionality in all the categories listed above; if the infrastructure were to fail, or lose effectiveness due to climate change, these river functions would be threatened.

PurposeSustained minimal residual disease (MRD) negativity is associated with long-term survival in multiple myeloma. The gut microbiome is affected by diet, and in turn can modulate host immunity, for example through production of short-chain fatty acids including butyrate. We hypothesized that dietary factors affect the microbiome (abundance of butyrate-producing bacteria or stool butyrate concentration) and may be associated with multiple myeloma outcomes.Experimental designWe examined the relationship of dietary factors (via a food frequency questionnaire), stool metabolites (via gas chromatography-mass spectrometry), and the stool microbiome (via 16S sequencing - α-diversity and relative abundance of butyrate-producing bacteria) with sustained MRD negativity (via flow cytometry at two timepoints 1 year apart) in myeloma patients on lenalidomide maintenance. The Healthy Eating Index 2015 score and flavonoid nutrient values were calculated from the food frequency questionnaire. The Wilcoxon rank sum test was used to evaluate associations with two-sided P < 0.05 considered significant.ResultsAt 3 months, higher stool butyrate concentration (P = 0.037), butyrate producers (P = 0.025), and α-diversity (P = 0.0035) were associated with sustained MRD negativity. Healthier dietary proteins, (from seafood and plants), correlated with butyrate at 3 months (P = 0.009) and sustained MRD negativity (P = 0.05). Consumption of dietary flavonoids, plant nutrients with antioxidant effects, correlated with stool butyrate concentration (anthocyanidins P = 0.01, flavones P = 0.01, and flavanols P = 0.02).ConclusionsThis is the first study to demonstrate an association between a plant-based dietary pattern, stool butyrate production, and sustained MRD negativity in multiple myeloma, providing rationale to evaluate a prospective dietary intervention.

Urbanization in South and Southeast Asia is accelerating due to economic growth, industrialization, and rural-to-urban migration, with megacities like Mumbai, Delhi, and Jakarta leading the trend. By analyzing VIIRS nighttime satellite data from 323 cities across 17 countries, we investigated the relationship between nighttime light (NTL) brightness and population density at varying distances from city centers. Our findings reveal a significant distance-decay effect, with both the intensity of NTL brightness and the strength of the NTL-population density relationship decreasing as the distance from city centers increases. A clear negative exponential relationship with the highest R2 was observed between NTL brightness and the distance from the city center. Our analysis indicates that a 105 km radius most effectively captures the extent of major metropolitan areas, showing a peak correlation between NTL brightness and population density. Cities like Delhi and Bangkok exhibit high NTL brightness, reflecting advanced infrastructure, while mountainous or desert cities such as Kabul and Thimphu show lower brightness due to geographical constraints. These results highlight the importance of adaptive urban planning, infrastructure development, and sustainability practices in managing urbanization challenges in South and Southeast Asia.

Background CD33 is a tractable target in acute myeloid leukemia (AML) for chimeric antigen receptor (CAR) T cell therapy, but clinical success is lacking.Methods We developed 3P14HLh28Z, a novel CD33-directed CD28/CD3Z-based CAR T cell derived from a high-affinity binder obtained through membrane-proximal fragment immunization in humanized mice.Results We found that immunization exclusively with the membrane-proximal domain of CD33 is necessary for identification of membrane-proximal binders in humanized mice. Compared with clinically validated lintuzumab-based CAR T cells targeting distal CD33 epitopes, 3P14HLh28Z showed enhanced in vitro functionality as well as superior tumor control and increased overall survival in both low antigen density and clinically relevant patient-derived xenograft models. Increased activation and enhanced polyfunctionality led to enhanced efficacy.Conclusions Showing for the first time that a membrane-proximal CAR is superior to a membrane-distal one in the setting of CD33 targeting, our results demonstrate the rationale for targeting membrane-proximal epitopes with high-affinity binders. We also demonstrate the importance of optimizing CAR T cells for functionality in settings of both low antigen density and clinically relevant patient-derived models.
